在免疫学和肿瘤学的研究领域,1986年发现的NF-κB和2013年发现的cGAS-STING 2条信号通路一直是研究热点。其中,cGAS-STING信号通路是天然免疫系统中关键的DNA“识别器”。
研究团队发现,cGAS-STING在流感病毒感染过程中可被激活,并通过NF-κB信号通路抑制病毒复制,构成人类抵御流感病毒的重要先天免疫屏障。
通过构建并系统筛选cGASSTING点突变体库,研究团队首次鉴定出cGAS-STING蛋白中一个重要结构基础,也是开启NF-κB信号通路的“开关”——第90位氨基酸(Gly90)。当发生G90A突变后,cGAS-STING激活NF-κB并发挥抗病毒作用的能力明显受损,证明了Gly90是激活NF-κB信号并限制流感病毒复制的关键一环。
在此基础上,研究团队比较了野生型cGAS-STING与G90A突变体所诱导的基因表达差异,继而鉴定出一类由NF-κB调控、但不同于传统干扰素刺激基因(ISGs)的抗病毒效应分子——NF-κB刺激基因(NSGs)。
通过功能筛选,研究人员发现GADD34是其中在抑制流感病毒复制中的关键“卫士”。当上游的cGAS-STING通路被激活后,“开关”G90会上调GADD34的表达,大量的抗病毒“卫士” GADD34就迅速出击,破坏流感病毒的“心脏”。
研究人员进一步发现,GADD34在人体呼吸系统中高度表达,能够通过抑制流感病毒聚合酶活性限制病毒复制,且这一抗病毒作用不依赖其经典的磷酸酶功能,揭示了其在先天免疫防御中的全新角色。
既然如此,这些病毒究竟是通过哪个“漏洞”实现由动物向人的跨物种传播?通过分析传染病数据库中十几种跨物种传播的流感病毒,研究团队发现,人源流感病毒编码的M1蛋白能够有效拮抗人源STING-NF-κB-GADD34抗病毒通路,而禽源流感病毒的M1蛋白在这一方面能力明显不足。
通过后续研究,研究团队揭示了流感病毒应对人体免疫屏障的进化策略:M1蛋白第115位氨基酸是由缬氨酸(Val)向异亮氨酸(Ile)的进化突变,这一突变显著增强了病毒对人源cGAS-STING通路,尤其是STING G90A的拮抗能力,从而提高其在人体细胞中的复制适应性。
该研究不仅加深了对人类先天免疫系统在阻断禽流感跨物种传播中作用机制的认识,还为流感大流行风险监测和防控策略提供了重要理论依据。
