    抗菌药物耐药性(AMR)已成为全球公共健康的重大威胁。研究估计,仅2019年就有超过495万例死亡与耐药相关。耐药基因之所以能在不同细菌间快速“串门”,很大程度上依赖质粒这类可移动的遗传载体。然而,在抗生素被大规模使用之前,质粒的具体形态及如何被人类“训练”成耐药传播工具,长期缺乏直接证据。
    英国研究团队通过“考古式基因组学”填补了这一空白。他们系统测序了1917—1954年临床样本中的历史质粒(著名的Murray Collection),并与公共数据库中的现代质粒进行比对,重建了跨越100余年的演化轨迹。研究共获得大量历史质粒序列,并与全球范围的现代数据联通,形成了覆盖多国家、多菌属的大型对照网络。2025年12月5日,相关研究成果发表于《Science》。
    结果显示,历史与现代相关质粒可归入3条不同命运:一类是“祖先骨架”,在现代数据库中已不再以原貌出现;另一类几乎“原封不动”地延续至今,保留了稳定的核心基因组;还有一类通过多个祖先质粒的“拼接融合”,演化为体量更大、结构更复杂的现代质粒。换句话说,抗生素时代不仅让耐药基因变得更复杂,也推动了质粒本体的结构重塑。
    更关键的发现是:几乎所有早期质粒都不携带典型耐药基因,但其中一小部分最终“脱颖而出”,成为驱动革兰氏阴性菌对一线乃至“最后防线”抗生素产生耐药的核心载体。现代世界里,耐药基因并非平均分布在所有质粒上,而是高度集中在少数“高风险家族”中。
    研究进一步指出,具备自我传播能力的“共轭型”质粒往往携带更多耐药基因,且体量也更大;不少现代高风险质粒还拥有多个复制起点(多复制子),表明它们经历了频繁的融合事件。
    该研究的第一作者、威康桑格研究所的Adrian Cazares表示:“抗生素的广泛使用从根本上重塑了质粒的遗传格局以及细菌对抗菌治疗的反应,并尖锐地提醒我们,人类行为对细菌进化有着深远而持久的影响。”
