By comparing the methods mentioned in the Chinese Pharmacopoeia and in the literature, we found that there are defects in the extraction method of samples, preparation methods of sample solution, chromatographic conditions, detection methods and determination results in content determination of monoester alkaloids in aconite by HPLC. Therefore, we established a new determination method and compared it with the one stated in the Chinese Pharmacopeia. The established method had better performance than the previous one. 对比《中国药典》与文献中附子单脂型生物碱HPLC 含量测定方法,发现附子HPLC 含量测定中样品的提取方法、供试品溶液的制备方法、色谱条件、检测方法和含量测定结果均欠佳,故建立附子新的含量测定方法并与《中国药典》含量测定方法比较。优化后的附子单脂型生物碱HPLC 含量测定方法优于《中国药典》方法。研究建立的附子HPLC 测定方法简便,可缩短检测时间,降低检测成本,提高了柱效,为附子安全、有效、质量可控提供更科学完善的检测方法。
