凝血酶作为一种快速止血药，主要从血液中提取，而血液中的血浆蛋白有100多种，因此国内生产的凝血酶不可避免地含有多种血浆杂质蛋白。α-2巨球蛋白（α-2 M）是一种重要的血浆蛋白之一。由于巨球蛋白保护胰蛋白酶，用大豆胰蛋白酶抑制剂灭活未被保护的胰蛋白酶后，利用Na-benzoy-DL-arginine-pnitroanilide（BAPNA）作为底物检测凝血酶中巨球蛋白的活性的方法，证明了从血浆中提取的凝血酶易存在α-2巨球蛋白，从而为凝血酶进一步纯化提供了研究方向。
Thrombin as a hemostatic drug can be extracted from blood. Because there are more than 100 plasma proteins in blood, it is natural for thrombin to contain some plasma proteins. α-2 macroglobulin is one of the important plasma proteins and it can pavilion and protect trypsin.
