Through selecting, cutting, testing the sample, it is indicated that the colon has been shaped. The conduction is then followed by PCR detection of plasmid and recombinant T-easy-vectors digested by EcoRI. The equation is y=-0.32x+10.47 and the ratio is 1.00. The constructed plasmid containing only the target gene was used to construct a calibration curve. The absolute standard curve method was shown to be of high linearity, sensitivity and reproducibility. Construction of Senegal Bichir Shh Gene Standard Plasmid DNA and Curve Using Real-time RT-PCR

为了快速、准确定量多鳍鱼Shh基因在机体组织中的表达水平,根据笔者克隆的Shh基因序列,选择其高度保守区域设计一对引物,对多鳍鱼各组织提取mRNA,经RT-PCR扩增,产物纯化后与pGEM-T-easy连接,转化大肠杆菌DH5α。筛选后得到重组标准品质粒,对重组标准品质粒进行酶切、PCR和测序鉴定,表明Shh基因保守区段已经成功克隆。将重组标准品质粒进行倍比稀释,然后以此为模板,进行荧光定量PCR,系统自动分析软件显示Ct值与标准品浓度的对数之间存在良好的线性关系。回归方程为 y=-0.32x+10.47,回归系数为1.00。构建Shh基因表达检测标准品质粒和标准曲线,建立检测多鳍鱼Shh基因荧光定量PCR 方法,为进一步研究Shh基因在组织中的动态分布和研究多鳍鱼系统发育奠定了基础。
