如今,RNA看似被其耀眼的表亲DNA所掩盖,但许多科学家认为,在生命诞生之初,RNA分子曾是舞台主角。它们既能存储遗传信息,又能自我复制,或曾触发演化进程,催生日益复杂的生命形式。迄今为止,研究者尚未发现能完全自我复制的RNA——这是生命体的关键特征。但他们现已取得接近突破的进展。本周在线发表于《Science》的一项研究显示,研究者创造出可生成自身某种镜像的RNA,并能利用该镜像模板重新合成原始序列。
单一RNA尚无法完成完整循环,但研究者表示,这一进展为“RNA是演化最初推手”的假说注入强劲动力。美国索尔克生物研究所化学家、“RNA世界”假说先驱之一Gerald Joyce评价道:“这是扎实的成果。”
Joyce等关注RNA,是因为即便在今天,该分子仍承担2项对生命至关重要的功能:其化学构件(核苷酸碱基)序列可编码遗传信息;折叠形成的稳定三维结构则能执行第二项关键任务——作为催化剂促进重要化学反应,且自身不被消耗。对许多研究者而言,这种双重角色使RNA成为生命启动器的候选者,既能编码自身组成,又能催化自身繁衍。
1993年,美国芝加哥大学Jack Szostak与麻省理工学院David Bartel团队通过构建并测试数万亿种不同RNA文库,筛选出能执行部分必要功能的分子。2009年,Joyce及其同事分离出两种可相互合成的RNA。
然而它们无法自我复制,且长度达150~200个碱基甚至更长,难以从早期地球化合物中自发产生。Joyce指出:“它们无法从原始汤中诞生。”在此尺度下,它们很可能在完全合成前就已降解。相比之下,新版本长度仅约为前者的1/3,使其自发出现至少具备理论可能。Joyce表示:“这是重大创新。”
为最大限度抑制降解,英国剑桥大学生物化学家Edoardo Gianni与Philipp Holliger团队在冷冻条件下开展实验。他们推测低温不仅能减缓RNA降解反应,还能辅助RNA复制过程。Holliger解释:水开始结冰时,会将其他化合物从生长中的冰晶中挤出,使核苷酸、盐类等RNA合成关键构件在微小溶液通道中浓缩;同时这些通道内液态水浓度较低,可减缓RNA降解。
当RNA处于含单核苷酸混合物的冷冻溶液中时,多数RNA保持折叠状态——这是其发挥催化功能所必需的构象。但要作为复制模板,RNA分子必须解折叠。早期研究曾提出一种策略:引入三联核苷酸,帮助模板RNA链维持开放构象。团队在冷冻混合物中测试约1万亿种随机RNA序列后发现,同时包含单碱基与三联体可使部分RNA链折叠并发挥催化作用,而其他链则保持开放以供复制。
团队筛选出3种长约45个碱基的RNA,它们能将自身复制为互补模板,并进一步以模板为起点重新合成原始序列。目前尚无证据表明单一RNA能完成双向循环——即在合成互补链后,再利用该链合成自身。且冷冻温度下合成新RNA链需耗时约72 d。但生命或始于寒冷:许多生命起源研究者认为,地球天然的冻融循环可能曾助力浓缩生命构件。
Szostak表示:“下一挑战是验证该系统或类似系统能否实现足够高效的重复复制循环。”若能实现,RNA将从生命出现的“可能起点”跃升为“合理起点”。
