裸鼹鼠最长寿命可达近40年,是寿命最长的啮齿动物,且对癌症、神经退行性疾病及关节炎等多种年龄相关疾病具有显著抗性。Chen等比较了培养的裸鼹鼠、小鼠与人类细胞中cGAS活性的差异。研究发现,当DNA受损时,裸鼹鼠cGAS中的氨基酸替换使其相较于小鼠和人类同种型泛素化与降解程度降低。cGAS蛋白丰度的增加进而增强了DNA修复因子FANCI与RAD50之间的相互作用,从而促进了同源重组——该机制对DNA双链断裂修复至关重要。作者还发现,在培养细胞中敲除裸鼹鼠cGAS会导致DNA损伤增加。这些结果表明,裸鼹鼠cGAS中的特定氨基酸差异增强了该蛋白的染色质结合能力,最终提高了DNA修复效率。值得注意的是,cGAS在细胞核内结合的增强预计会抑制炎症反应,这一发现值得后续深入研究。此外,Chen等通过实验证明,表达携带4个裸鼹鼠cGAS突变位点的人类cGAS的果蝇,其寿命比表达未突变人类cGAS的果蝇更长。这一结果进一步证实cGAS在裸鼹鼠的长寿机制中发挥着重要作用。
目前尚不清楚除裸鼹鼠外,其他物种是否能通过cGAS在细胞核内的功能获益。先前对人类和小鼠细胞的研究表明,cGAS会抑制DNA修复与复制,这与cGAS在染色质稳态维持中的作用相符;而cGAS的缺失则可能引发不受控的重组或复制。来自cGAS基因敲除小鼠的初步数据显示,转座因子(即能在基因组一处位置“跳跃”到另一处的“跳跃基因”)的抑制状态被解除,最终导致炎症信号增强与寿命缩短。与Chen等的研究发现相呼应,这些结果共同表明cGAS通过维护基因组完整性,在调控染色质组织与稳态中发挥着重要作用。
若裸鼹鼠的cGAS发生了能使其抵御DNA损伤的变异,那么其他长寿生物是否也会以类似方式改变其cGAS?数种蝙蝠相较于同等体型的哺乳动物同样展现出延长的寿命。此外,与大多数哺乳动物不同,这些蝙蝠的免疫系统能够耐受病毒侵袭——这是其谱系特有的、在数百种蛋白质上发生的进化适应结果。这些适应性变化包括:其一,干扰素基因刺激因子(STING)磷酸化位点发生单残基突变(该突变能编码位于cGAS下游的跨膜蛋白,对启动干扰素生成至关重要);其二,PY‐HIN基因家族完全缺失(该家族编码的胞质传感器可识别与cGAS相同的双链核酸底物)。
那么蝙蝠体内的cGAS又是如何变异的呢?蝙蝠体内确实存在cGAS基因,且相较于PYHIN传感器,该蛋白在进化过程中得以保留,这表明cGAS可能具有免疫感应之外的非典型功能,或者它可能相较于PYHIN传感器发挥着更重要的免疫调控作用。扩展的系统发育分析印证了cGAS的重要性——无脊椎动物乃至细菌中均存在其同源物。然而,通过对比蝙蝠cGAS与其他哺乳动物的氨基酸序列,研究者发现其存在高频率的变异。其中在表现出长寿特征的旧大陆果蝠(狐蝠科)中,其cGAS的N端(该区域对先天免疫激活至关重要)存在一个高度分化区域。值得注意的是,这些蝙蝠在cGAS蛋白的3个关键结构环区中,有2个环区出现了氨基酸突变——这2个环区此前已被证实与核小体(染色质的基本组成部分)结合密切相关。这些突变很可能增强了cGAS与染色质的相互作用。更引人注目的是,研究发现,对裸鼹鼠cGAS增强DNA修复功能不可或缺的4个氨基酸突变之一Glu511→Lys(小鼠和人类cGAS中均不存在该突变),同样存在于狐蝠科物种中。由此可见,通过不同突变组合实现cGAS与染色质结合的进化策略,可能是长寿物种的共同特征。这种机制既能促进DNA修复,又可避免自身免疫反应这一衰老相关炎症加剧的主要诱因。
该项研究揭示了裸鼹鼠cGAS在细胞核内调控寿命的意想不到的作用。虽然仍需通过更多研究来探明cGAS在短寿与长寿生物细胞核中的功能,但答案可能远比最初预测的更为复杂。
