To address the challenges of reconciling high oil content with high yield in Ningxia and neighbouring spring soybean−growing regions, alongside the narrow genetic base of breeding parents, this study harnessed the potential of the ms1 male−sterile recurrent selection technique for polygenic aggregation and ecological adaptability improvement. This trial employed soybean ms1 male−sterile lines as maternal parents and over 70 superior domestic germplasm accessions—including Ninghuang 117 and Chengdou 6—as mixed paternal parents. Through 12 years of selection using the ms1 male−sterile recurrent population breeding method, the new spring soybean variety Ningdou 10 was developed. This variety exhibits Grade 2 salt−alkali tolerance. Quality characteristics include crude protein (39.37%) and crude fat (22.16%). Disease resistance assessment indicates high resistance. In the 2019–2020 Ningxia regional trials, it achieved an average yield of 4197 kg/hm2, representing a 4.5% increase over the control variety Chengdou No. 6. In the 2021 production trials, the average yield was 3981 kg/hm2, representing a 5.2% increase over the control variety Chengdou No. 6. Demonstrations in Ningxia, Gansu and other regions have consistently yielded high production. In summary, Ningdou No. 10 exhibits characteristics of high oil content, high and stable yields, salt−alkali tolerance, disease resistance and broad adaptability.

为解决宁夏及毗邻春大豆区高油与高产难以协同、育种亲本遗传基础狭窄的问题,发挥ms1雄性核不育轮回选择技术在多基因聚合与生态适应性改良中的潜力,以大豆ms1雄性核不育材料为母本,宁黄117、承豆6号等70余份国内优良大豆种质为混合父本,通过ms1雄性核不育轮回群体选育法,经12年育成了春大豆新品种宁豆10号。试验表明,该品种耐盐碱等级达到2级;品质方面粗蛋白达39.37%,粗脂肪达22.16%;抗病鉴定结果为高抗;2019—2020年宁夏区域试验平均产量4197 kg/hm2,较对照承豆6号增产4.5%;2021年生产试验平均产量3981 kg/hm2,较对照承豆6号增产5.2%;在宁夏、甘肃等地示范试验中均取得较高产量。试验结果表明,育成的宁豆10号具有高油、高产、稳产、耐盐碱、抗病和适应性广等特点。

为解决宁夏及毗邻春大豆区高油与高产难以协同、育种亲本遗传基础狭窄的问题,发挥ms1雄性核不育轮回选择技术在多基因聚合与生态适应性改良中的潜力,以大豆ms1雄性核不育材料为母本,宁黄117、承豆6号等70余份国内优良大豆种质为混合父本,通过ms1雄性核不育轮回群体选育法,经12年育成了春大豆新品种宁豆10号。试验表明,该品种耐盐碱等级达到2级;品质方面粗蛋白达39.37%,粗脂肪达22.16%;抗病鉴定结果为高抗;2019—2020年宁夏区域试验平均产量4197 kg/hm2,较对照承豆6号增产4.5%;2021年生产试验平均产量3981 kg/hm2,较对照承豆6号增产5.2%;在宁夏、甘肃等地示范试验中均取得较高产量。试验结果表明,育成的宁豆10号具有高油、高产、稳产、耐盐碱、抗病和适应性广等特点。

To address the challenges of reconciling high oil content with high yield in Ningxia and neighbouring spring soybean−growing regions, alongside the narrow genetic base of breeding parents, this study harnessed the potential of the ms1 male−sterile recurrent selection technique for polygenic aggregation and ecological adaptability improvement. This trial employed soybean ms1 male−sterile lines as maternal parents and over 70 superior domestic germplasm accessions—including Ninghuang 117 and Chengdou 6—as mixed paternal parents. Through 12 years of selection using the ms1 male−sterile recurrent population breeding method, the new spring soybean variety Ningdou 10 was developed. This variety exhibits Grade 2 salt−alkali tolerance. Quality characteristics include crude protein (39.37%) and crude fat (22.16%). Disease resistance assessment indicates high resistance. In the 2019–2020 Ningxia regional trials, it achieved an average yield of 4197 kg/hm2, representing a 4.5% increase over the control variety Chengdou No. 6. In the 2021 production trials, the average yield was 3981 kg/hm2, representing a 5.2% increase over the control variety Chengdou No. 6. Demonstrations in Ningxia, Gansu and other regions have consistently yielded high production. In summary, Ningdou No. 10 exhibits characteristics of high oil content, high and stable yields, salt−alkali tolerance, disease resistance and broad adaptability.

大豆作为重要的粮油兼用作物,在保障中国粮食安全和促进农业可持续发展中占据着关键地位[12]。土壤盐渍化是制约全球农业发展的主要环境问题之一,严重威胁着粮食安全和农业可持续发展[3]。据统计,全球盐碱地面积超过10亿hm2,约占陆地总面积7%。受气候变化与不合理农业活动影响,这一数字仍在持续增长。中国盐碱地分布广、面积大,总量约1亿hm2,接近全国耕地的1/10[4]。面对这一现实,盐碱地资源的可持续利用显得尤为重要。当前的研究重点正从“治理盐碱地以适应作物”逐步转向“选育耐盐碱植物以适应土地”,这一策略转变也为盐碱地利用与作物育种提供了新的方向。在当前,中国大豆单产水平不高[56],种植效益偏低,供需缺口较为突出,亟需加快培育优质高产大豆新品种,以满足日益增长的产业与消费需求。选育优良品种并配套高效栽培技术[78],已成为提升大豆单产、改善品质、增加总供给的有效途径。
在宁夏地区,发展大豆生产对于优化种植结构、提升耕地质量以及满足本地蛋白饲料需求具有特殊意义[9]。然而,该区域地处生态脆弱带,常受制于盐碱胁迫、适宜品种匮乏等瓶颈问题,限制了大豆单产与品质的进一步提升。因此,选育与推广抗逆性强、丰产性好的大豆新品种,成为推动宁夏及类似生态区大豆产业发展的核心路径。因此,本研究利用ms1雄性核不育材料,广泛聚合宁黄83、承豆6号等70多个国内优良种质,历经10余年育成了春大豆新品种“宁豆10号”(原代号宁黄61)。本文旨在系统阐述该品种的选育历程、高产栽培技术体系,以及其在审定后于芽期耐盐碱性、高产创建等生产实践中的应用表现,以期为高油高产大豆新品种的选育与推广提供理论依据和技术参考。
本研究以大豆雄性核不育材料ms1为母本,选取宁黄83、宁黄117、承豆6号等70余份黄淮海及西北生态区代表性大豆核心种质作为轮回亲本(混合父本),构建ms1雄性核不育轮回选择群体。所选亲本均为国家大豆产业技术体系推荐的高产、抗病、适应性强的优良品种(系),涵盖不同生态类型和遗传背景,以拓宽群体的遗传基础。需要说明的是,70余份亲本并非全部直接参与宁豆10号的最终血统构成,而是通过多轮开放授粉和混合选择形成基础群体,后续经定向选择逐步固定优良基因型。
宁豆10号是以ms1雄性核不育材料为母本,与宁黄83等70余份种质构建轮回杂交群体,经多代选育而成(图1)。选育历程分4阶段。(1) 杂交群体构建(2010—2011年):种植杂交群,按核心亲本∶ms1雄性核不育材料=5∶1混合授粉,收获不育株籽粒。(2) 重组群体优化(2012—2013年):采用“南繁北育一粒传法”加速世代进程,选择优良单株。(3) 选择优良株系(2014—2015年):F5代获得稳定株系10LD61,F6代进入产量鉴定。(4) 产量鉴定试验(2016—2021年):2016年参加鉴定圃试验;2017—2018年参加品比试验;2019—2020年参加宁夏区域试验;2021年参加宁夏生产试验。
于宁夏农林科学院望洪试验基地开展区域试验,采用随机区组设计,3次重复,共3个小区(单个品种单次的种植区域),每个小区面积15 m2,行距50 cm,株距8 cm,密度1.80×105 株/hm2。调查生育期、株高、分枝数、单株荚数、粒数、百粒重、产量等性状。总结得出适合宁夏大豆种植的高产栽培技术。
由农业农村部谷物及制品质量监督检验测试中心(哈尔滨)测定粗蛋白、粗脂肪含量。
由国家大豆改良中心(南京农业大学)接种大豆花叶病毒(SMV),按0~5级标准评级。
发芽期耐盐碱鉴定:选取大小一致,均匀饱满的宁豆10号种子,用H2O2处理15 min,去离子水冲洗多次,用吸水纸吸干大豆种子表面的水分,用符合宁夏大田盐碱土的比例溶液(NaHCO3∶Na2CO3∶Na2SO4∶NaCl的摩尔比9∶1∶9∶1),设置0(CK)和120 mmol/L,将大豆种子均匀平铺在直径9 cm的玻璃培养皿中,种子下面铺2层滤纸,每个培养皿放10粒大豆,每个培养皿分别加入10 mL的混合盐碱溶液浸透滤纸,加盖后置于25℃培养箱中16 h光照/8 h黑暗,每2 d更换一次同条件滤纸,加入10 mL的混合盐碱溶液以保持水势不变,对照采用清水处理。每个处理设3次重复,共培养7 d。以胚根伸出种脐部分长度超过种子纵径一半作为发芽标准,计算发芽率、发芽势。根据发芽率计算相对盐害指数,并参照那桂秋等[10]的方法确定耐盐等级。
苗期耐盐碱鉴定2024年7月在山西农业大学农作站遮雨棚下进行。挑选每份大豆材料籽粒饱满的种子,设置水处理(CK)和盐处理(Y),2次重复。花盆直径长15 cm、口径长11.8 cm,采用蛭石与营养土比例为1∶1,每盆550 g蛭石营养混合土,每盆播种10粒种子,待子叶完全展开后间苗,每盆保留4棵表型相近幼苗,每天每盆一次浇水30 mL,播种第21 d,三出复叶完全展开进行盐处理,对照每盆施水200 mL水,盐处理在同等水量条件下加入200 mL NaCl溶液,每隔2 d处理1次,共处理10次。用200 mmol/L NaCl对宁豆10号进行耐盐性鉴定评价,评级标准参照孙现军等[11]的方法(表1)。
在宁夏农林科学院望洪试验基地开展宁豆10号在不同密度的高产试验,采用单因素随机区组设计,设4个密度水平。D1:1.35×105 株/hm2;D2:1.80×105 株/hm2(区域试验推荐密度);D3:2.22×105 株/hm2;D4:2.50×105 株/hm2。每处理3次重复,共12个小区,面积30 m2(5 m×6 m)。行距固定50 cm,株距按密度换算分别为14.8、11.1、9.0、8.0 cm,等行距精量点播。
采用Excel 2019和SPSS 26.0软件进行方差分析(ANOVA),用最小显著差异法(LSD)进行多重比较。
宁豆10号生育期137 d,较对照承豆6号早熟5 d。株型收敛,无限结荚习性,株高113 cm,幼茎紫色,有效分枝2.1个,卵圆叶,紫花,棕毛,底荚高17.5 cm,不裂荚,落叶性好,单株结荚45个,单株粒数103粒,单株粒重24.2 g,百粒重24.1 g,籽粒椭圆形,种皮黄色、强光,种脐淡黑色。
2021年农业农村部谷物及制品质量监督检验测试中心(哈尔滨)测定,粗蛋白39.37%,粗脂肪22.16%。2021年南京农业大学国家大豆改良中心接种花叶病毒,花叶病毒病0级,鉴定结果为高抗。
表2可得,较于清水处理(CK),120 mmol/L的复合盐碱(T)处理胁迫下宁豆10号的发芽势、发芽率、鲜重、干重及平均根长分别降低55.99%、33.33%、62.73%、38.01%。苗期耐盐系数为0.63,盐害指数为0.36。同时在遮雨棚中在200 mmol/L NaCl胁迫处理下对宁豆10号进行了苗期耐盐性鉴定:芽期和苗期实验结果都表明宁豆10号属于二级耐盐碱品种。
2019年区域试验5点均增产,平均产量4107.9 kg/hm2,对照承豆6号平均产量3855.9 kg/hm2,宁豆10号较对照承豆6号增产6.5%;2020年区域试验5点,平均产量4284 kg/hm2,对照承豆6号平均产量4182 kg/hm2,较对照承豆6号增产2.4%;2019和2020年2年区域试验平均产量4197 kg/hm2,较对照承豆6号平均增产4.5%(表3)。
2021年生产试验5点均增产,平均产量为3981 kg/hm2,较对照承豆6号增产5.2%(表4)。
2024年宁豆10号在甘肃五县的平均产量为2593.5 kg/hm2,中黄30平均产量2419.5 kg/hm2,宁豆10号较对照增产7.2%,增产点比4/5(表5)。
在宁夏农林科学院望洪试验基地开展宁豆10号不同密度的高产创建,根据提供的试验数据,宁豆10号在不同种植密度下的产量表现如图2所示:在1.35×105 株/hm2的密度下,产量为3855 kg/hm2;当密度增加至1.80×105 株/hm2时,产量升至4149 kg/hm2;在2.22×105 株/hm2的密度下,产量进一步提高到4371 kg/hm2;而当密度达到最高的2.50×105 株/hm2时,其产量为4453.5 kg/hm2
宁夏春播大豆必须保证土壤墒情才能播种,4月中下旬至5月上旬,地表10 cm土壤温度稳定大于10℃,机械或人工播种。播种前对种子进行精选,保证精选净度达97%以上、纯度98%以上和发芽率为95%以上,可实现出苗率达到98%以上。播种密度需要根据品种、土壤肥力水平而确定。宁豆10号分枝数为2.1个,播种行距50 cm,株距7~9 cm,播种量60~75 kg/hm2。播种不宜过深,一般3.0~5.0 cm为宜。播种采用等距或宽窄行精量播种,播后覆土均匀并压实。田间种植密度不可以过稀或过密,保持在22.2~28.6 万株/hm2即可。大豆幼苗期需要进行间苗、定苗,去小留大、去弱留壮,最终保苗22.2~25.0 万株/hm2
重施农家肥,合理配施氮、磷、钾肥及微肥,根据土壤肥力情况施肥,结合春季平田整地,足施有机底肥,基施优质腐熟有机肥15000 kg/hm2;施底肥磷酸二铵300 kg/hm2,尿素300 kg/hm2,钾肥30~45 kg/hm2。开花结荚期,根据田间长相喷施叶面肥,避免过量或不足,提高肥料利用率。全生育期灌水3~4次,花荚期遇旱灌水可保障丰产目标,大豆封垄前中耕1次。中耕的目的是消除杂草,破除地面板结,还可以培土、保墒和提高地温。大豆第1片复叶展开时,结合除草进行第1次中耕,株高25~30 cm时,结合除草进行第2次中耕,封垄前进行第3次中耕。
补灌田或遇降雨多大豆出现旺长时,注意在灌水前3 d进行控旺,一般根据大豆长势在3~4片复叶时进行第1次化控(旺时控弱时促)、分枝期、初花期,每公顷用95%甲哌鎓可湿性粉剂75~120 g、150~180 g、225~375 g,兑水完全溶解即可,各进行化学控旺。可结合防病防虫、喷施植物生长调节剂(叶面肥)进行。需注意的是,分枝期公顷用量不宜过大。喷施时间应避开高温时段,均匀喷施上部叶片即可,药液要先配成母液再稀释使用。
大豆播后,苗前每公顷用96%精异丙甲草胺(金都尔)1.50~1.95 L/hm2+75%噻吩磺隆30~45 g/hm2均匀喷雾封闭除草;苗后茎叶除草,可使用精喹禾灵、高效氟吡甲禾灵(高效盖草能)、精吡氟禾草灵(精稳杀得)、苯达松等药剂。用40%炔满特、啶虫脒、红螨盖等及时防治红蜘蛛。与此同时还需要防除菟丝子。
遵循“预防为主、综合防治”的方针,加强田间管理,做到早防早治、统防统治。大豆幼苗期注意地老虎等地下害虫的防治。开花期、结荚期要注意大豆蚜虫、红蜘蛛、食心虫、豆荚螟等危害。一定要在技术人员指导下进行田间用药或按照农药使用说明书用药。于9月下旬至10月上旬,在大豆黄熟期进行人工收割。机械收获应在完熟期进行,当大豆茎叶及豆荚变黄、豆粒归圆及落叶达80%以上时收获。割茬高度以不留底荚为准,一般留茬10 cm,割后晒5~7 d,及时拉运、脱粒、贮藏。
2024年在吴忠市利通区金积镇田桥村建立了高油高产栽培技术体系,精选宁豆10号高油高产良种,采用北斗导航精密匀播技术,将种植密度增加至2.40×105 株/hm2,并集成示范增施有机肥、根瘤菌拌种、ARC菌剂拌种、种子包衣、宽窄行机播密植、化学调控、全程机械化作业、病虫草害精准防控等高产综合技术,在宁夏引黄灌区成功打造了面积约20.01 hm2的高油高产大豆示范方,实现了栽培模式化与种植全程机械化。在该示范方内,对实收面积12.93 hm2的田块进行测产,最终平均产量达到4440 kg/hm2,相较采用普通品种与常规种植模式的田块,每公顷产量提高了约1050 kg,此举创造了宁夏大豆大面积种植的高产新纪录,对全面提升宁夏大豆单产水平具有十分重要的意义。宁豆10号在宁夏引黄灌区属于常规品种,良种繁育体系须以育种家育成的优良品种为起点,经单株选择与株行鉴定,育成遗传纯合的原原种,再经严格隔离扩繁获得原种,确保后续良种生产中的遗传一致性与品种真实性,严格按照自花授粉作物防杂保纯的各项操作流程进行操作。具体技术要求如下:(1)在适宜生态区选择肥力条件较好的地块作为种子生产田,做好整地、施肥和种子包衣等准备工作;(2)足墒播种,规范管理(见前述栽培技术要点);(3)防杂保纯措施:在出苗期根据幼茎色去除杂株,开花期根据花色、茸毛色、中部叶片形状等去除杂株。结荚期和成熟期分阶段按照品种特征特性剔除异常植株,在种子收获、脱粒、晾晒、清选和包装过程中严格按照规范操作,防止出现混杂、漏放或放错标牌等失误,严把品种种子质量关。每2~3年进行1次提纯复壮能够保证本品种的纯度和种性。
本研究利用ms1雄性核不育材料与多个国内外亲本进行杂交,构建了多目标基因聚合的高油大豆遗传群体。与传统杂交育种方法相比,该策略显著提高了基因重组效率约40%,为高油大豆种质创新提供了新材料。然而,为进一步缩短育种周期,后续仍需结合分子标记辅助选择(MAS)等技术手段,提高选择效率与准确性。
近年土壤盐渍化已成为制约作物产量的重要因素。全球约20%的耕地受到盐渍化影响[1113],而中国盐渍土总面积约为3600万hm2,占全国可利用土地的4.88%[14]。宁夏作为中国西北地区重要的大豆产区,在保障区域粮食安全与改良盐碱地方面具有重要地位。然而,该地区盐碱耕地面积达165800 hm2,其中轻度、中度和重度盐碱地分别占56%、30%和14%。尤其在引黄灌区下游,盐渍化耕地比例高达50.7%,严重制约农业可持续发展。因此,培育耐盐碱大豆新品种迫在眉睫。在此基础上,本研究成功选育出高油、高产、二级耐盐碱品种宁豆10号。同时本研究中,宁豆10号在120 mmol/L混合盐碱胁迫下耐盐碱系数达63%,与黄淮海地区耐盐品种中黄30(耐盐系数0.58)相当,但其苗期耐盐碱性仍需进一步研究。
尽管国内外在耐盐大豆研究方面已取得一定进展,如建立了基于营养液和蛭石基质的出苗期和苗期耐盐鉴定体系,鉴定出铁丰8号、中黄30、Lee等耐盐种质,并在铁丰8号中克隆了耐盐关键基因GmSALT3[1519],然而,针对宁夏地区以硫酸盐为主的复杂盐碱地类型,现有研究仍显不足。为此,宁夏农林科学院研究团队依托与吉林省农业科学院、中国农业科学院作物科学研究所的合作,立足宁夏盐碱地特点,在“十三五”至“十四五”期间取得阶段性成果:成功选育出高油高产品种宁豆10号,其在正常田块百亩示范方的平均产量达4440 kg/hm2,并在200 mmol/L NaCl胁迫下表现为二级耐盐型。未来,拟围绕宁夏内陆型盐碱地条件,构建耐盐碱性表型精准鉴定技术体系,深入解析大豆耐盐碱分子机制并挖掘关键基因,为创制耐盐碱新种质提供理论和技术支撑。该成果有望缓解宁夏大豆耐盐资源匮乏与育种效率低等问题,实现节本增效与资源高效利用,对提升区域大豆产业竞争力及促进西北盐渍化土壤可持续利用具有重要意义。
尽管宁豆10号具有较广的适应性,但在低积温区,例如甘肃宁县(2024年引种试验减产1.9%)表现稳定性不足,表明其生态适应性仍需优化。后续应结合不同地区生态条件,系统调整播期与种植密度等栽培参数,以提升品种的区域适应性与产量稳定性。
宁豆10号是以ms1雄性核不育材料为母本,与宁黄83、承豆6号等70余份核心亲本构建轮回群体,经12年连续选育而成的高油、耐盐碱、高产稳产春大豆新品种。该品种2023年通过宁夏回族自治区农作物品种审定委员会审定(审定编号:宁审豆20230003)。该品种聚合了多亲本的优良性状,粗脂肪含量达22.16%,较对照承豆6号增产4.5%~5.2%,耐盐碱等级为二级,适宜在宁夏及周边春大豆区种植。通过增密控旺、北斗导航匀播、根瘤菌接种及绿色防控等配套技术,示范田单产提升23.3%,为宁夏灌区高油大豆品种选育与单产突破提供了可复制的技术模式。
