With the long-term operation of transformers, microorganisms will be produced in transformer oil, and the microbial metabolites can affect the quality of transformer oil. In this paper, the changes of the content, composition, and structure of extracellular polymeric substances (EPS) produced by microorganisms in the transformer oil were analyzed by using three-dimensional fluorescence spectroscopy (3D-EEM), ultraviolet spectroscopy, and Fourier transform infrared spectroscopy (FTIR). The types of organic compounds in new oil and fault oil were compared and analyzed, and the effect of EPS on the quality of transformer oil was investigated. The results show that C=C and C=O characteristic functional groups generate in the transformer oil after long-term operation. The color of the oil in darkened, and the absorbance increases significantly in the near ultraviolet region with the wave length of 190 nm-400 nm. The content of protein (PN) in new oil, running oil, and fault oil is always higher than that of polysaccharide (PS), and PN is the main component of EPS. The number of fluorescent emission contour rings of EPS in fault oil decreases, mainly including O-H, C-H, C=O and unsaturated bond. Compared with new oil, the water content of faulty oil increases by up to 5 times, and the acid value is 40 times higher than that of new oil.

随着变压器的长期运行,变压器油中会产生微生物,微生物代谢产物会影响变压器油的品质。本文结合三维荧光光谱(3D-EEM)、紫外光谱和红外光谱(FTIR)分析变压器油中微生物产生的胞外聚合物(EPS)含量、组成及结构的变化,对比分析新油及故障油中有机物的种类,考察EPS对变压器油品质的影响。结果表明:变压器油长期运行会生成C=C和C=O特征官能团,颜色加深,在波长为190~400 nm的近紫外区吸光度明显增大。新油、运行油及故障油中蛋白质(PN)含量始终高于多糖(PS)含量,PN是EPS的主要成分,故障油EPS的荧光发射等高圈的数量减少,主要存在O-H、C-H、C=O和不饱和键。与新油相比,故障油中含水量最多增大5倍,酸值提高40倍。

随着变压器的长期运行,变压器油中会产生微生物,微生物代谢产物会影响变压器油的品质。本文结合三维荧光光谱(3D-EEM)、紫外光谱和红外光谱(FTIR)分析变压器油中微生物产生的胞外聚合物(EPS)含量、组成及结构的变化,对比分析新油及故障油中有机物的种类,考察EPS对变压器油品质的影响。结果表明:变压器油长期运行会生成C=C和C=O特征官能团,颜色加深,在波长为190~400 nm的近紫外区吸光度明显增大。新油、运行油及故障油中蛋白质(PN)含量始终高于多糖(PS)含量,PN是EPS的主要成分,故障油EPS的荧光发射等高圈的数量减少,主要存在O-H、C-H、C=O和不饱和键。与新油相比,故障油中含水量最多增大5倍,酸值提高40倍。

With the long-term operation of transformers, microorganisms will be produced in transformer oil, and the microbial metabolites can affect the quality of transformer oil. In this paper, the changes of the content, composition, and structure of extracellular polymeric substances (EPS) produced by microorganisms in the transformer oil were analyzed by using three-dimensional fluorescence spectroscopy (3D-EEM), ultraviolet spectroscopy, and Fourier transform infrared spectroscopy (FTIR). The types of organic compounds in new oil and fault oil were compared and analyzed, and the effect of EPS on the quality of transformer oil was investigated. The results show that C=C and C=O characteristic functional groups generate in the transformer oil after long-term operation. The color of the oil in darkened, and the absorbance increases significantly in the near ultraviolet region with the wave length of 190 nm-400 nm. The content of protein (PN) in new oil, running oil, and fault oil is always higher than that of polysaccharide (PS), and PN is the main component of EPS. The number of fluorescent emission contour rings of EPS in fault oil decreases, mainly including O-H, C-H, C=O and unsaturated bond. Compared with new oil, the water content of faulty oil increases by up to 5 times, and the acid value is 40 times higher than that of new oil.

变压器油作为变压器的绝缘介质,可以增强变压器内各部件的绝缘强度,在运行过程中起散热和冷却作用[1-2]。变压器油经过压力过滤和蒸馏处理后,品质纯净,介质损耗小[3],但是在储存、运输和检修过程中容易被微生物污染。变压器油的主要成分是烷烃、环烷烃和芳香族化合物,其中碳氢元素的质量分数为95%~99%,在适宜条件下,这些化合物可作为碳源供微生物自身生长[4-5]。微生物本身及其代谢产物含有的蛋白质具有胶体性质,会使变压器油的电导系数升高,缩短变压器的使用寿命,甚至导致线圈烧毁,造成巨大的经济损失。韦强等[6]研究发现,变压器新油中主要含有球菌,污油中含有大量的球菌、杆菌和放线菌,细菌量越多,变压器油的介质损耗因数越大。姬晓川等[7]研究发现,变压器油中存在以油作为唯一碳源的芽孢杆菌微生物,能够长期存活。但是,关于微生物的代谢产物对变压器油性能的影响研究较少。
胞外聚合物(extracellular polymeric substances,EPS)是微生物分泌的高分子聚合物,主要由多糖(polysaccharides,PS)和蛋白质(protein,PN)组成[8-9]。胞外多糖是带负电的黏性物质,可以聚集细菌细胞,在饥饿环境中EPS在胞外酶的作用下会水解生成小分子物质,为微生物提供碳源,还可以形成保护层,抵抗外部环境中有害物质的入侵,维持微生物的结构和功能完整性[10]。目前,尚不清楚变压器油中EPS的组成和结构变化,以及EPS对变压器油品质的影响。
吉林地区气温多变和干湿交替的环境易于微生物的滋生、侵入和生长演替。本研究分析变压器油中微生物代谢产物的性质,考察新油及故障油中有机物的种类,研究微生物代谢产物含量、组成及结构的变化,分析EPS对变压器油性能的影响,以期为后续变压器油的处理提供理论基础。
采用66 kV变电站的油样作为研究对象,其中新油、运行油和故障油分别取自通化市城东、干沟、柳河和太王66 kV变电站,具体信息如表1所示。
利用热处理法提取油中EPS,流程如下:取变压器油30 mL,在4 000 r/min的转速下离心10 min,取上层清液在20 Hz超声频率下处理20 min,再在8 000 r/min的转速下离心10 min,将所得混合液在60℃下水浴加热10 min,在10 000 r/min的转速下离心15 min,取上层清液利用孔径为0.45 µm的滤膜过滤,所得滤液即为EPS。
采用紫外可见分光光度计(UV-2700型,日本岛津公司)进行全光谱扫描,检测变压器油组分及溶解产物的光谱信号,波长范围为190~600 nm。采用考马斯亮蓝法测定EPS各层PN含量,采用蒽酮-硫酸法测定PS含量,采用三维荧光光谱(3D-EEM,RF-6000型,日本岛津公司)分析EPS中有机物种类的变化,激发/发射波长为220~550 nm,波长间隔和狭缝宽度均为5 nm,扫描速度设置为6 000 nm/min。利用傅里叶变换红外光谱(FTIR,Tensor 27型,德国布鲁克公司)对EPS的官能团进行测定,扫描范围为500~4 000 cm-1,扫描16次,分辨率为0.4 cm-1,根据吸收峰位置和强度分析官能团结构。
采用卡尔费休库仑法水分测试仪(MiniHYD型,上海双旭电子有限公司)测定含水量;根据GB/T 28552—2012《变压器油、汽轮机油酸值测定法(BTB法)》使用全自动酸值测定仪(HK-3160SZ型,北京华科仪科技股份有限公司)检测酸值;采用击穿电压测试仪(ZJC-50kV型,北京中航时代仪器设备有限公司)测定油样击穿电压;采用介质损耗仪(2821型,瑞士TETTEX公司)测定油样介质损耗因数,频率和电压分别设置为50 Hz和1 kV。
变压器油品的颜色和透明度取决于油中树脂物质及其他染色化合物的含量[11],变压器油的外观如图1所示。纯净的变压器新油不适合微生物生长,只有当变压器中含有水分、杂质,且在合适的温度环境下,微生物才会大量繁殖。从图1可以看出,变压器新油无气味、透明;运行油由于自身氧化的影响,颜色呈淡黄色,但油样色泽均匀,无可见的杂质和沉淀。变压器油经过长期运行,油样受到氧化作用而形成树脂质氧化产物,同时油样放电会产生碳粒,导致故障油颜色较深,产生棕黑色沉淀,使得变压器油黏度增加,电气性能下降,影响变压器的运行。
变压器油样品C4、Y4、G4的紫外光谱测试结果如图2所示。从图2可以看出,在波长为400~600 nm的可见光部分,样品吸光度接近0,3条曲线重合在一起,说明在可见光区域变压器油不受老化的影响[12]。在波长为190~400 nm的近紫外区,3份样品的吸光度明显增大,受变压器油老化故障的影响较大,这是因为变压器油在老化过程中产生的C=C和C=O特征基团会产生紫外吸收[13]。在近紫外区故障油的吸光度最大,且在205 nm处吸光度出现突增。因此,在变压器油的故障老化监测过程中,可以选择在205 nm处进行测定。
变压器油中EPS组分和含量的变化如表2所示。从表2可以看出,不同变压器油中PS和PN含量的变化趋势相同,EPS含量从大到小的样品顺序为:运行油、故障油、新油。变压器新油中含有较低含量的PN(40.98~50.21 mg/L),而PS含量接近零,油样呈无色透明状态。随着变压器运行时间的增加,变压器油中出现水分及杂质[14],这易于微生物的滋生和富集,运行油的PN和PS含量最高,其中太王变电站Y4油样的EPS总量最高达到约740.45 mg/L。各变电站的故障油中EPS总量约为573.60~594.71 mg/L,低于运行油,这可能是因为故障油中含有大量的杂质,部分成分不适合微生物的生长。3种变压器油样品中的PN含量均高于PS,而且是EPS的主要成分。
变压器油主要由烷烃、环烷烃及芳香烃组成,芳香烃类物质具有荧光特性,不同油类中芳香烃的种类和含量不同,在一定浓度范围内,荧光的强度与油中芳香烃类物质的浓度呈正比[15]图3是不同变压器油样中EPS的三维荧光光谱。从图3可以看出,4个变压器新油、运行油和故障油的EPS光谱相似,新油的激发波长主要出现在200~350 nm,运行油的激发波长主要出现在200~400 nm,故障油的激发波长主要出现在270~400 nm。新油中的EPS主要有3个荧光发射等高圈,其中330、265、240 nm波长处为主要激发中心;随着变压器的运行,运行油的主要激发中心发生偏移,但仍然存在3个荧光发射等高圈;故障油的图谱与新油和运行油存在较大差别,荧光发射等高圈的数量减少,只存在两个荧光发射等高圈。
为了进一步分析EPS的官能团结构,选取具有代表性的太王变压器油中EPS进行红外光谱分析,结果如图4所示。从图4可以看出,新油C4和运行油Y4中EPS的主要吸收峰位置没有明显变化,官能团结构相似,C4和Y4在3 400 cm-1(O-H键)处没有明显吸收峰。然而,故障油G4中在3 400 cm-1出现轻微驼峰,表明EPS中存在微量水分[16]。3种油品在2 937 cm-1和2 860 cm-1附近均存在吸收峰,分别对应于有机物中C-H的不对称和对称伸缩振动[17];1 456 cm-1和1 367 cm-1附近均有强烈的吸收,它们与-CH3、-CH2-官能团的C-H键的红外吸收有关[18],其中Y4和G4此处的吸收峰增强。G4在900~600 cm-1存在不饱和键吸收峰[19],在1 600 cm-1和1 490 cm-1处出现的振动吸收峰是苯环骨架伸缩振动导致的[20]。与新油和运行油相比,故障油EPS出现更多的杂峰,故障油被更多的有机物污染,其中在1 739~1 770 cm-1出现C=O伸缩振动峰,这是油中醛和酸类物质的吸收峰,说明变压器油已经发生氧化降解。
含水量、酸值、击穿电压以及介质损耗因数是评价变压器油品质的主要指标。为了进一步明确微生物代谢产物与变压器油品质的相关性,对不同变压器油样的性能指标进行测定,结果如表3所示。从表3可以看出,4个变压器新油的含水量均较低,运行油的含水量略有升高,故障油G4的含水量变化最大,由6 µg/g升高至31 µg/g,对应EPS红外光谱中3 400 cm-1处的O-H键。含水量增加会使变压器油中的离子浓度和载流子迁移率提高,导致变压器油的力学性能下降,介质损耗因数增大[21-22]。酸值可以有效表征油中有机酸与无机酸的含量[23],4个故障油的酸值分别比新油高85倍、88倍、72倍、64倍,且故障油中的EPS在1 739~1 770 cm-1处出现明显伸缩振动峰,说明长期运行的变压器油吸收了空气中的氧,产生各种有机酸、酚类及胶状油泥(图2(c)[24],生成的极性杂质和带电的胶体物质逐渐增多。从表3还可以看出,随着运行时间增加,变压器油的介质损耗因数不断增大,故障油G4的介质损耗因数最大,达到约6.5%。故障油G1、G2、G3、G4的击穿电压分别下降至28、31、31、35 kV,这一方面是由于故障油中含有的杂质被吸引到电场强度较大的区域,导致变压器油的击穿强度降低[25],另一方面是由于变压器油放电产生的炭粒会使击穿电压大幅下降[26]。与新油相比,故障油G4的击穿电压下降最多,降幅达到约61.7%。
(1)长期运行的变压器油颜色逐渐变深,产生C=C和C=O特征官能团,在波长为190~400 nm的近紫外区吸光度明显增大,可以选择在205 nm处监测变压器油的故障老化。
(2)新油的微生物代谢产物量较少,运行油中EPS总量最高约为740.45 mg/L,故障油中含有大量的杂质,其EPS总量约为579.19~594.71 mg/L;3种变压器油中的PN含量均高于PS,PN是EPS的主要成分。
(3)故障油中EPS的激发波长主要出现在270~400 nm。与新油和运行油相比,故障油中EPS荧光发射等高圈的数量减少,主要存在O-H、C-H、C=O和不饱和键;故障油中EPS出现更多的杂峰,主要是酸醛类物质。
(4)长期运行的变压器油品质变化明显,故障油G4中含水量最多,酸值(0.84 mgKOH/g)比新油C4高64倍,产生了各种有机酸类及胶状油泥,导致其介质损耗因数增大,击穿电压下降。
