The community structure and distribution patterns of endophytic fungi in wild apricot (Prunus armeniaca), an endemic species exclusively distributed in Xinjiang Uygur Autonomous Region, northwestern China was investigated. During 2023-2024, healthy plant tissues (leaves, buds, branches, bark) were collected from Turgun Town of Xinyuan County, Xinyuan County Botanical Garden, Fushou Mountain of Huocheng County, and Liuhuanggou of Changji Hui Autonomous Prefecture. Endophytic fungi were isolated using tissue culture methods and identified through morphological analysis and ITS sequencing. The obtained 2 749 isolates were classified into 6 classes, 20 orders, 38 families, and 64 genera, with Dothideomycetes dominating at the class level (isolation frequency, IF=88.7%), Pleosporales at the ordinal level (IF=83.41%), Dothideaceae at the family level (69.33%), and Alternaria at the generic level (IF=67.01%). Ten genera were common to all sampling sites, while endemic genera numbered 8 in Turgun, 14 in Botanical Garden, 12 in Fushou Mountain, and 6 in Liuhuanggou. α-Diversity was the highest in Turgun (Shannon Hʹ=1.206±0.111 9; Simpson D=0.531±0.050) and lowest in Liuhuanggou (Hʹ=0.983±0.128; D=0.468±0.055). β-Diversity analysis revealed the most significant community divergence between Liuhuanggou and Turgun (Bray-Curtis P=0.006 4). These results demonstrate rich taxonomic diversity (64 genera) and distinct regional variations in community composition, relative abundance, and dominant taxa among wild apricot endophytes in Xinjiang.

为探究分析我国仅分布在新疆维吾尔自治区的野杏Prunus armeniaca内生真菌的群落和分布特征,于2023年至2024年在伊犁哈萨克自治州新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉回族自治州的硫磺沟等地区采集健康野杏的叶、芽、枝、树皮等部位,采用组织分离法进行内生真菌的分离,结合形态学和ITS序列进行菌株的种类鉴定,同时分析不同地区野杏内生真菌的群落多样性。共分离得到2 749株内生真菌,鉴定为6纲、20目、38科、64属,其中以座囊菌纲Dothideomycetes为优势菌纲(IF=88.7%),优势目为格孢腔菌目Pleosporales (IF=83.41%),优势科为座囊菌科Dothideaceae (69.33%),以链格孢属Alternaria为优势属(IF=67.01%);新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉硫磺沟4个不同样地之间的共有属有10个,特有属依次为8、14、12和6个。α多样性指数分析表明物种多样性最高的样地是吐尔根乡(Hʹ=1.206±0.111 9,D=0.531±0.050),最低的是昌吉硫磺沟样地(Hʹ=0.983±0.128,D=0.468±0.055)。基于β多样性分析不同样地间的Bray-Curtis距离的P值显示昌吉硫磺沟与新源县吐尔根乡之间野杏内生真菌群落差异最显著(0.006 4)。本研究结果揭示了野杏内生真菌菌群具有丰富的多样性,4个不同样地的野杏内生真菌群落组成、相对多度及优势类群皆存在差异。

为探究分析我国仅分布在新疆维吾尔自治区的野杏Prunus armeniaca内生真菌的群落和分布特征,于2023年至2024年在伊犁哈萨克自治州新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉回族自治州的硫磺沟等地区采集健康野杏的叶、芽、枝、树皮等部位,采用组织分离法进行内生真菌的分离,结合形态学和ITS序列进行菌株的种类鉴定,同时分析不同地区野杏内生真菌的群落多样性。共分离得到2 749株内生真菌,鉴定为6纲、20目、38科、64属,其中以座囊菌纲Dothideomycetes为优势菌纲(IF=88.7%),优势目为格孢腔菌目Pleosporales (IF=83.41%),优势科为座囊菌科Dothideaceae (69.33%),以链格孢属Alternaria为优势属(IF=67.01%);新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉硫磺沟4个不同样地之间的共有属有10个,特有属依次为8、14、12和6个。α多样性指数分析表明物种多样性最高的样地是吐尔根乡(Hʹ=1.206±0.111 9,D=0.531±0.050),最低的是昌吉硫磺沟样地(Hʹ=0.983±0.128,D=0.468±0.055)。基于β多样性分析不同样地间的Bray-Curtis距离的P值显示昌吉硫磺沟与新源县吐尔根乡之间野杏内生真菌群落差异最显著(0.006 4)。本研究结果揭示了野杏内生真菌菌群具有丰富的多样性,4个不同样地的野杏内生真菌群落组成、相对多度及优势类群皆存在差异。

The community structure and distribution patterns of endophytic fungi in wild apricot (Prunus armeniaca), an endemic species exclusively distributed in Xinjiang Uygur Autonomous Region, northwestern China was investigated. During 2023-2024, healthy plant tissues (leaves, buds, branches, bark) were collected from Turgun Town of Xinyuan County, Xinyuan County Botanical Garden, Fushou Mountain of Huocheng County, and Liuhuanggou of Changji Hui Autonomous Prefecture. Endophytic fungi were isolated using tissue culture methods and identified through morphological analysis and ITS sequencing. The obtained 2 749 isolates were classified into 6 classes, 20 orders, 38 families, and 64 genera, with Dothideomycetes dominating at the class level (isolation frequency, IF=88.7%), Pleosporales at the ordinal level (IF=83.41%), Dothideaceae at the family level (69.33%), and Alternaria at the generic level (IF=67.01%). Ten genera were common to all sampling sites, while endemic genera numbered 8 in Turgun, 14 in Botanical Garden, 12 in Fushou Mountain, and 6 in Liuhuanggou. α-Diversity was the highest in Turgun (Shannon Hʹ=1.206±0.111 9; Simpson D=0.531±0.050) and lowest in Liuhuanggou (Hʹ=0.983±0.128; D=0.468±0.055). β-Diversity analysis revealed the most significant community divergence between Liuhuanggou and Turgun (Bray-Curtis P=0.006 4). These results demonstrate rich taxonomic diversity (64 genera) and distinct regional variations in community composition, relative abundance, and dominant taxa among wild apricot endophytes in Xinjiang.

野杏Prunus armeniaca属于蔷薇科杏属植物,是世界栽培杏的原生起源物种、第三纪孑遗植物(张加延和张钊 2003),新疆野杏仅分布于新疆伊犁地区的天山野果林内。近年来,因环境变化、人为干扰以及野杏的天然更新,野杏种群繁衍受阻、分布范围内数量减少、病害严重、退化趋势显著,在伊犁调查各野生果树居群的种群非空间结构,综合评定目前新疆野杏种群的健康状况为中等偏下,其整个种群的生存状况较差,急需开展保育恢复(杨其享等 2022)。
植物内生真菌生长在植物体内的特殊环境中,在与植物的长期共同进化中寄主与内生菌间形成了互惠的共生关系(Tan & Zou 2001;Clay & Schardl et al. 2002;李聪等2023;Giger et al. 2024),解析植物内生真菌的种群分布特征对保护植物起着重要的作用(臧威等 2014;唐官美等 2024)。研究发现橡胶树不同组织内生真菌中子囊菌门占绝对优势(93%),核心属为刺盘孢属Colletotrichum、间座壳属Diaporthe、镰孢菌属Fusarium、木霉属Trichoderma和青霉属Penicillium (Kaliane et al. 2020);桫椤叶柄凋落物与活体组织的内生真菌群落存在显著差异,部分内生真菌可能在宿主组织衰老后转为腐生生活,参与生态系统物质循环(臧威等 2020)。对7个不同产地杜仲Eucommia ulmoides的树皮内生真菌群落的研究结果表明,杜仲树皮内生真菌具有丰富多样性且受产地影响显著(张青青等 2021);对比云南、贵州、四川和西藏的48种水生植物内生真菌发现,随着海拔升高,内生真菌分离频率和多样性整体下降(Xu et al. 2025)。关于杏树内生真菌的研究,国内仅有新疆轮台县、乌鲁木齐市、吐鲁番市和伊宁市4个地区的10个杏树品种的花芽、茎、叶和根4种器官中内生真菌群落多样性的研究,发现真菌类群的组成存在较大差别(白周艳等 2011)。
天山腹地生长着一片亚洲面积最大、最密集的原始野生果林,主要分布于新源县、巩留县和塔城地区境内,分布面积约100 km2,对区域生物多样性和生态系统稳定性的维持至关重要(丁吉达等 2023;尹马泉等2025)。野杏作为天山野果林重要的建群树种及我国重要的种质资源,对其健康状况及保护方面的研究至关重要(杨小平 2020)。然而关于野杏的研究主要集中在种质资源开发(董胜君等 2002;刘娟 2015)、病虫害的诊断防控(Ye et al. 2020;陈帅康等 2023;刘楚丽等 2023)等。本研究在新疆野杏主要分布区新疆伊犁哈萨克自治州、昌吉回族自治州收集野杏样本,分析不同地区野杏内生真菌的种类组成及多样性,为深入了解野杏与内生真菌间的共生关系、野杏内生真菌资源的开发利用和保护奠定研究基础。
在野杏的主要分布点新疆昌吉回族自治州硫磺沟和新疆伊犁哈萨克自治州的新源县吐尔根乡、新源县植物园和霍城县福寿山开展样品的采集,样地详细信息见表1
在供试的4个样地中分别设置一个10 m × 10 m的样方,每个样方内通过五点取样法采集样本,每个点分别选取样树间距大于10 m且胸径基本一致的5棵样树,于2023年11月- 2024年12月的每月初采集野杏的一年生侧枝和主干树皮,2023年11月-2024年3月的每月初采集杏芽,2024年4月-2024年10月的每月初采集杏叶,2024年6月-2024年8月的每月初采集杏果,每个组织部位选择20个样本装入无菌保鲜袋,4 ℃条件保存(Ye et al. 2020)。
将每次采集到的全部样本进行分离,操作流程依次为:自来水冲洗表面的灰尘,无菌水漂洗1 min,75%乙醇漂洗3 min,3%次氯酸钠溶液漂洗10 min,无菌水漂洗3次,灭菌的滤纸擦净水分后晾干备用。通过无菌水检测法验证表面消毒效果,取最后一次漂洗用无菌水滴涂于PDA培养基上,在25 ℃的恒温培养箱中倒置培养7 d,观察是否有菌落生成,若发现有菌落生成,则证明表面消毒不彻底,不可使用,若无,则证明后续实验所分离得到菌落为内生真菌而非表面附着的真菌(何佳等 2009;吕佳等 2022)。
内生真菌的分离、纯化及保存:将已经进行过表面消毒的组织块用手术刀在无菌条件下切割成0.5 cm × 0.5 cm的组织块,每块PDA培养基平板放置5个组织块,共4个重复,25 ℃恒温培养,每日观察,待能观察到明显菌丝后挑取尖端菌丝到新的PDA培养基上,同时挑取菌丝到含有20%甘油的离心管中,于-80 ℃冰箱冷冻保存(Ye et al. 2020)。
形态学鉴定参照《真菌鉴定手册》(魏景超 1979)和《真菌分类学》(邵力平等 1984),分子生物学鉴定参考范鑫磊(2016)的报道。当特定序列与系统发育相关的参考序列之间的相似性<95%,则被认定为未鉴定菌株,>95%且≤97%则被认定鉴定到属水平,>97%则被认定鉴定到种水平,而≥99%则被鉴定为同种(Zhou et al. 2013),并在MycoBank (http://www.mycobank.org/quicksearch.aspx)网站中查询最新的真菌分类。
对鉴定到种的菌株按照纲、目、科、属依次进行分类,内生真菌(属)数量进行多样性分析。研究中的分离频率、多样性指数、优势度指数和丰富度指数采用张青青等(2021)在杜仲内生真菌多样性研究中的方法。
分离频率(isolation frequency, IF)可比较不同地区野杏内生菌的优势菌群,计算见公式(1)。

利用Shannon-Wiener多样性指数(H′)和Simpson优势度指数(D)评估不同地区野杏内生菌群落多样性。H′指数越大,说明相应的内生真菌群落多样性越大,群落复杂程度越高;D指数正好相反,其指数越大,相应内生真菌群落多样性越小,群落复杂程度越低。公式参考邓文祥等(2019)和张青青等(2021)的报道。

通过软件PAST 5.1统计检验ANOSIM相似性分析吐尔根、霍城、植物园、硫磺沟4个样地之间野杏内生真菌群落Jaccard相似性系数和Bray-Curtis距离。
采用IBM SPSS Statistics Version 22.0统计软件进行单因素方差分析(one-way ANOVA),分析所有野杏内生真菌的Shannon-Wiener多样性指数、Simpson优势度指数、Margalef丰富度指数在P=0.05水平上的显著性。
不同地区野杏分离得到内生真菌群落结构组成、分离频率、丰度均具有一定差异。对稀有类群、常见类群、优势类群的定义:某菌群占整体菌群的百分比≤1%为稀有类群,1%-10%为常见类群,≥10%为优势类群(庞雄飞和尤民生 1996)。
连续14个月从4个样地的野杏树上共收集到3 840份样本,分离得到2 749株内生真菌,鉴定出6纲、20目、38科、64属真菌。隶属于座囊菌纲Dothideomycetes、锤舌菌纲Leotiomycetes、蘑菇纲Agaricomycetes、散囊菌纲Eurotiomycetes、盘菌纲Pezizomycetes、粪壳菌纲Sordariomycetes。其中座囊菌纲为优势菌纲,共分离到2 443株,占菌株总数的88.7%;格孢腔菌目Pleosporales为优势目,占菌株总数的83.41%;座囊菌科Dothideaceae为优势科,占菌株总数的69.33%;优势属为Alternaria,占菌株总数的67.01% (图1)。
从伊犁新源县吐尔根乡野杏样地中共分离得到795株内生真菌,占总分离频率的28.92%,鉴定为28属,Alternaria (65.28%)为优势属(图1);常见属11个:亚隔孢壳属Didymella (6.54%)、附球菌属Epicoccum (5.53%)、枝孢霉属Cladosporium (3.77%)、小穴壳属Dothiorella (2.39%)、镰孢菌属Fusarium (2.26%)、嗜果刀孢菌属Wilsonomyces (1.89%)、曲霉属Aspergillus (1.63%)、壳囊孢属Cytospora (1.51%)、小球藓菌属Microsphaeropsis (1.38%)、假茎点霉属Nothophoma (1.13%)和茎点霉属phoma (1.13%)。其余16个属均为稀有属(图2)。
伊犁新源县植物园样本中共鉴定出32属,共644株菌,占总分离频率的23.43%。Alternaria为优势属,分离频率为66.93% (图1);常见属9个:Epicoccum (7.45%)、Cladosporium (5.43%)、Aspergillus (3.57%)、Monilinia (2.64%)、Didymella (2.48%)、Fusarium (1.40%)、壳二孢属Ascochyta (1.24%)、Nothophoma (1.24%)和Wilsonomyces (1.24%);其余22个属为稀有属(图3)。
霍城县福寿山野杏样本中共鉴定出31属,共860株菌,占总分离频率的31.28%。其中Alternaria (72.09%)为优势属(图1);常见属7个:Didymella (6.86%)、Cladosporium (4.41%)、Epicoccum (2.67%)、Fusarium (2.33%)、Aspergillus (2.21%)、Microsphaeropsis (1.86%)和Wilsonomyces (1.63%);其余23个属为稀有属(图4)。
昌吉硫磺沟野杏样本中共鉴定出23属,共464株菌,占总分离频率的16.88%。其中Alternaria (58.62%)和Aspergillus (11.85%)为优势属(图1- LHG);常见属7个:火丝菌属Pyronema (8.41%)、Cladosporium (4.74%)、Microsphaeropsis (3.66%)、Nothophoma (1.94%)、Cytospora (1.72%)、斜孢壳菌属Coniothyrium (1.08%)和Fusarium (1.08%);其余14个属为稀有属(图5)。
基于属水平上不同样地野杏样本中内生真菌群落的Upset图分析可知(图6),从新源县吐尔根乡、新源县植物园、霍城县福寿山、昌吉硫磺沟野杏样本中各分离得到内生真菌属一级的数量分别为27、31、30和22个。4个样地同时存在的共有属为10个,即AlternariaAspergillusCladosporiumConiothyriumCytosporaDidymella、小穴壳属DothiorellaFusariumPhoma、光黑壳属Preussia。昌吉硫磺沟与新源县吐尔根乡之间的共有属12个;霍城县福寿山和新源县植物园之间的共有属12个;新源县吐尔根乡与霍城县福寿山之间的共有属16个;昌吉硫磺沟与霍城县福寿山之间的共有属14个。
仅在新源县吐尔根乡、新源县植物园、霍城县福寿山之间分布的共有属有2个,EpicoccumWilsonomyce;仅在吐尔根、霍城、硫磺沟地区之间的有1个共有属,Microsphaeropsis;仅在吐尔根、植物园、硫磺沟地区之间的有1个共有属,假茎点霉属Nothophoma
仅在昌吉硫磺沟与霍城县福寿山分布的共有属有3个,分别为短梗霉属Aureobasidium、毛壳菌属Chaetomium、新刺毛茎点霉属Neosetophoma;仅在吐尔根地区与霍城县福寿山分布的共有属有3个,分别为Paracamarosporium、柄孢壳菌属Podospora、粪壳菌属Sordaria。仅在新源县吐尔根乡与新源县植物园分布的共有属有3个,分别为Chaetopyrena、拟球束孢属Gibellulopsis、青霉属Penicillium;仅在新源县植物园与昌吉硫磺沟分布的共有属有2个,分别为ComoclathrisPyronema
不同地区样地之间特有属与共有属及内生真菌的群落组成与分布存在明显差异(图6)。4个样地中新源县植物园样地的野杏内生真菌群落特有属的种类最多,达14种,分别是Aposphaeria、壳二孢属Ascochyta、烟管菌属Bjerkandera、锥毛壳属Coniochaeta、拟鬼伞属Coprinopsis、弯孢霉属Curvularia、碘光盘菌属IodophanusLasiobolidium、链核盘菌属MoniliniaNeodidymelliopsisQuerciphoma、梭孢壳菌属ThielaviaVacuiphomaXenodidymella
霍城县福寿山样地的野杏内生真菌群落含有12个特有属,分别为多柱壳菌属Camarops、炭疽菌属ColletotrichumCollophorina、圆盘座壳属Discostroma、拟日规壳菌属Gnomoniopsis、黄霉菌属Myceliophthora、炭垫菌属NemaniaNeocamarosporium、拟盘多毛孢属Pestalotiopsis、无柄盘菌属Pezicula、散盘端毛孢Seimatosporium、炭角菌属Xylaria
新源县吐尔根乡样地的野杏内生真菌群落含有8个特有属,分别为细基束梗孢菌属Cephalotrichum、小光壳属Leptosphaerulina、丛赤壳菌属Nectria、黑团孢属PericoniaPhaeoacremoniumPhaeobotryon、拟茎点霉属Phomopsis、毛团状盘菌属Tricharina
特有属最少的是昌吉硫磺沟的野杏内生真菌群落,包含有6个特有属,分别为节菱孢属Arthrinium、葡萄孢属BotrytisConiolariella、座囊菌属DothideaEntoleucaNeomicrosphaeropsis
不同样地野杏内生真菌群落Shannon-Wiener指数从高到低依次为新源县吐尔根乡(1.206)>新源县植物园(1.015)>霍城县福寿山(0.988)>昌吉硫磺沟(0.983);Simpson指数从高到低依次为新源县吐尔根乡(0.531)>新源县植物园(0.477)>昌吉硫磺沟(0.468)>霍城县福寿山(0.425);丰富度指数从高到低依次为新源县吐尔根乡(1.468)>新源县植物园(1.254)>霍城县福寿山(1.197)>昌吉硫磺沟(1.141)。Shannon-Wiener指数和Simpson指数分析可知,新源县吐尔根乡的内生真菌群落多样性和优势度最大,昌吉硫磺沟的内生真菌群落多样性和优势度最小;Magalef指数分析得出,新源县吐尔根乡的内生真菌群落丰富度最大,昌吉硫磺沟的内生真菌群落丰富度最小。此外,新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉硫磺沟4个样地之间的野杏内生真菌群落的Shannon-Wiener指数(Hʹ)、Simpson指数(D)、Magalef指数(R)未观察到显著性差异(表2)。
对野杏内生真菌群落的分布区新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉硫磺沟进行两两比较发现不同样地之间野杏内生真菌群落均具有差异(表3)。通过Bray-Curtis距离值P值对新源县吐尔根乡、新源县植物园、霍城县福寿山和昌吉硫磺沟的野杏内生真菌群落进行两两比较后发现昌吉硫磺沟样地与新源县吐尔根乡样地的P值为0.006 4、其与霍城县福寿山的P值为0.01,内生真菌群落之间存在显著性差异。其他地区之间的内生真菌群落之间有一定的差异,但不显著,如昌吉硫磺沟与新源县植物园的野杏内生真菌群落的P值(0.138 4)<新源县植物园与霍城县福寿山的P值(0.475)<新源县吐尔根乡与新源县植物园的P值(0.723 8)<新源县吐尔根乡与霍城县福寿山的P值(0.875 8)。
本研究从新疆4个野杏样地共分离获得2 749株内生真菌,分属于6纲20目38科 64属。优势菌纲为Dothideomycetes,优势目为Pleosporales,优势科为座囊菌科Dothideaceae,优势属为链格孢属。链格孢属的分离率最高与前期的研究一致,白周艳等(2011)从新疆10个栽培杏品种中共分离到35属内生真菌,其中链格孢属的分离率最高;此外,链格孢属在新疆盐生植物黑果枸杞不同组织(花、叶、果、茎和根)的内生真菌组成中和黑沙蒿嫩枝与成熟枝内生真菌中均为优势属(顾美英等 2022;乌吉斯古冷等 2022),进一步表明链格孢属在植物内生真菌中占据优势地位。
前人研究表明植物内生真菌的丰度、多样性和物种组成与温度、湿度、光照、地理位置、周围植被等宿主生境条件密切相关(张青青等 2021),真菌群落组成随着水分和生境的变化,真菌丰富度和群落组成与多样性显著正相关(梁钰镁等 2023),与海拔梯度呈负相关(Xu et al. 2025)。本研究中伊犁新源县和霍城县的3个样地位于天山山脉西部的伊犁河谷,气候条件湿润,被誉为“塞外江南”,水汽较北疆其他地区充沛且植物种类丰富(羊海军等 2003;杨其享等 2022)。而昌吉硫磺沟样地则以风蚀地貌为主,属于典型的大陆性干旱气候,植被总体稀疏(金淑聪 2007)。本研究中植物种类丰富且雨水丰沛的伊犁新源县吐尔根乡样地指数最高(Hʹ=1.206±0.1119,D=0.531±0.050),环境相对干旱的昌吉硫磺沟野杏纯林的指数最低,表明优势真菌群落的多样性受所处微环境的影响。
通过β多样性指数能够体现不同地区之间内生菌群落的差异。白周艳等(2011)研究发现气候条件相差较大的北疆伊宁和南疆轮台的杏树内生真菌的相似性最低(Cs=0.44);本研究还发现昌吉硫磺沟与伊犁新源县吐尔根乡的内生真菌群落极不相似且相似性最小(0.008 1),Bray-Curtis距离的P值显示内生真菌群落差异最显著(0.006 4),再次验证不同样地之间的内生真菌群落结构差异与地区间的气候等环境因子密切相关。
此外,本研究也发现受传统分离方法、培养基成分、样本采集方法、培养方式等因素的影响,并不是所有的内生真菌都能人工培养(Amann & Ludwig 1995;Zheng et al. 2021)。后续的研究中可以基于时间序列观察真菌分布的季节动态和年际变化(Giger et al. 2024),探讨不同真菌属在生态系统中的功能角色,全面解析内生真菌与其他微生物群落的相互作用(Clay & Schardl 2002)。为深入解答内生真菌在野杏体内分布特征提供科学依据,助力野杏保护实践。
