Based on morphological, chemical and molecular phylogenetic methods, a taxonomical study on the lichen family Ramalinaceae in Anhui Province has been carried out. Bacidia areolata, Bacidia suffusa, Biatora appalachensis and Ramalina sphaerophora are new to China. Bacidia schweinitzii, Biatora printzenii, Phyllopsora porphyromelaena and Ramalina conduplicans are found from Anhui Province for the first time. The systematic positions of the above species are detailed, and the species new to China are morphologically and anatomically described.

综合形态、化学和分子系统发育研究,对采自安徽省的树花科地衣开展分类学研究,发现了4个中国新记录种,即网状杆孢衣Bacidia areolata、覆粉杆孢衣Bacidia suffusa、阿巴拉契亚蜡盘衣Biatora appalachensis和球顶树花Ramalina sphaerophora。施氏杆孢衣Bacidia schweinitzii、普氏蜡盘衣Biatora printzenii、鳞片树痂衣Phyllopsora porphyromelaena和对折树花Ramalina conduplicans在安徽省首次发现。本文阐明了上述物种的系统位置,并对4个中国新记录种进行了详细的描述,提供了形态和解剖图片。本研究丰富了我国树花科地衣的物种多样性。

Based on morphological, chemical and molecular phylogenetic methods, a taxonomical study on the lichen family Ramalinaceae in Anhui Province has been carried out. Bacidia areolata, Bacidia suffusa, Biatora appalachensis and Ramalina sphaerophora are new to China. Bacidia schweinitzii, Biatora printzenii, Phyllopsora porphyromelaena and Ramalina conduplicans are found from Anhui Province for the first time. The systematic positions of the above species are detailed, and the species new to China are morphologically and anatomically described.

树花科Ramalinaceae C. Agardh隶属于茶渍纲Lecanoromycetes、茶渍目Lecanorales,包括40属981种(Lücking et al. 2017;Wijayawardene et al. 2022),是地衣型真菌中物种数排名第四的科(Lücking et al. 2017;Kistenich et al. 2018)。目前,我国已报道树花科地衣12属139种(秦起龙 2020;魏江春 2020)。该科的关键形态特征为:子囊盘为蜡盘型或网衣型,子囊孢子透明无色,常有横隔,共生藻为绿球藻,广泛分布于亚洲、美洲、欧洲、非洲等地(Kistenich et al. 2018)。
杆孢衣属Bacidia De Not.和蜡盘衣属Biatora Fr.同为树花科的壳状类群,形态较为相似。杆孢衣属由de Notaris (1846)以Bacidia rosella (Pers.) De Not.为模式种建立,该属子囊盘为网衣型,子囊为Bacidia-type,8孢,子囊孢子无色透明,具3-4或更多横隔(Ekman 1996;Kantvilas 2018;Gerasimova et al. 2021),我国已报道35种(魏江春 2020)。蜡盘衣属是Fries以春蜡盘衣Biatora vernalis (L.) Fr.为模式种建立的,该属的子囊盘为蜡盘型,盘面多为黄棕色,侧丝常黏合,子囊为Bacidia-type或Biatora-type,8孢,子囊孢子无色,窄椭圆形,具3-5横隔(秦起龙 2020),我国有16种(刘玉洁等 2019;秦起龙 2020;魏江春 2020)。
树花属Ramalina Ach.是树花科的枝状类群,小枝实心或中空,皮层下有具支持作用的软骨样菌丝,一般具子囊盘或粉芽(Zhao et al. 2007;付伟 2008;Gasparyan et al. 2017)。1935年以前,我国的树花属地衣主要由国外学者报道,Meyen & Flotow (1843)最早记载了采自广东的掌树花Ramalina digitata Meyen & Flot.。我国最早研究树花属地衣的学者是朱彦承,记载了树花属2种(Tchou 1935)。迄今,中国已知树花属地衣48种(付伟等 2008, 2009;魏江春 2020)。
本课题组于2021-2024年间多次深入安徽省境内的大别山、黄山等区域开展地衣资源调查和标本采集工作,共采集树花科地衣标本65份,基于形态、化学和分子系统发育研究表明,该区域的树花科物种多样性高,其中相当一部分为以往研究不足的新记录物种,包括中国新记录种4个,安徽新记录种4个,极大地丰富了我国该科的物种多样性。
研究标本于2021-2024年采集自安徽省各市县,现存放于安徽师范大学生命科学学院植物标本馆(AHUB)。
地衣体形态特征观察和拍照在体视显微镜OLYMPUS SZ61TR和相机MS60-2下完成,显微特征观察和拍照在正置偏光显微镜OLYMPUS BX43和相机MDX10下完成。
采用显色反应和薄层层析(TLC)方法检测地衣次生代谢产物。通过滴加10% KOH溶液(K)、Ca(ClO)2饱和水溶液(C),观察化学显色反应。薄层层析采用C溶剂系统(甲苯:乙酸=100:15,体积比) (Orange et al. 2001)。
采用Chelex (弱阳离子螯合树脂)法提取地衣体的基因组DNA (殷钰皎等 2025)。PCR扩增以ITS1f (Gardes & Bruns 1993)和ITS4a (Larena et al. 1999)为引物,扩增体系总体积为25 μL (2×Trio Taq Master Mix 12.5 μL、ddH2O 9.5 μL,引物各1 μL,模板DNA 1 μL)。扩增程序:94 ℃预变性5 min;94 ℃变性15 s,53 ℃退火15 s,72 ℃延伸1 min,35个循环;72 ℃延伸10 min,4 ℃保存。扩增产物送至通用生物股份有限公司进行纯化和双向测序。
测序获得的双向序列首先在NCBI上使用BLAST工具进行比较分析(https://blast.ncbi.nlm. nih.gov/Blast.cgi),确定是否为地衣型真菌序列。其后,使用Geneious Prime软件对双向序列进行拼接并去除两侧模糊碱基。在GenBank中下载了31条同科ITS参考序列以及2条外类群[红鳞网衣Psora decipiens (Hedw.) Hoffm.和墙壁鳞网衣P. vallesiaca (Schaer.) Timdal]序列(Kistenich et al. 2018)。使用MAFFT v.7 (Katoh et al. 2019)的E-INS-i策略进行序列比对,最终用于建树的矩阵有587个碱基位点,保守位点(C)占39.0%,简约信息位点(Pi)占47.5%。本研究采用最大似然法(maximum likelihood algorithm, ML),使用IQ-TREE的在线版本(http://iqtree.cibiv.univie.ac. at/)构建系统发育树(Trifinopoulos et al. 2016),将ITS划分为ITS1、5.8S和ITS2这3个分区,IQ-TREE推测的各分区最适模型为ITS1 (TIM2e+G4)、5.8S (TNe+I)、ITS2 (TNe+I+G4),设置1 000次重复计算。采用SH-aLRT (SH-aLRT support) (Guindon et al. 2010)、aBayes (aBayes support) (Anisimova et al. 2011)和UFBoot (Ultrafast bootstrap approximation) (Minh et al. 2013) 3种方法评估各分支节点支持率,SH-aLRT ≥80%、aBayes ≥0.95或UFBoot ≥95%的分支被认为得到良好支持,并以该次序呈现在系统发育树上(图1)。使用MEGA 7可视化系统发育树,并在PowerPoint中进行美化编辑。
本研究新测得树花科ITS序列19条,GenBank检索号及标本采集信息见表1,结合GenBank数据库中筛选的树花科代表物种序列31条和2条外类群序列组成矩阵。系统发育分析显示,本研究报道的新记录种均与其他区域的该种样品形成高支持的单系分支,明确了这些物种的分子鉴定。其中,中国新记录种网状杆孢衣Bacidia areolata Gerasimova & A. Beck (支持率99.3/1.00/100)和覆粉杆孢衣B. suffusa (Fr.) A. Schneid. (100/1.00/100)分别与俄罗斯的该种样品聚为一支,二种互为姐妹类群;阿巴拉契亚蜡盘衣Biatora appalachensis Printzen & Tønsberg (99.8/1.00/100)与美国的该种样品聚为一支,且具有相同的形态和地衣次生代谢产物;球顶树花Ramalina sphaerophora Kashiw. & K.H. Moon (97.5/1.00/100)与韩国的该种样品聚为一支,该分支与同分布区域的安田氏树花R. yasudae Räsänen有较近的亲缘关系。4个安徽新记录种中,施氏杆孢衣Bacidia schweinitzii (Fr. ex E. Michener) A. Schneid. (100/1.00/100)与美国和俄罗斯的该类样品以高支持率聚为一支;普氏蜡盘衣Biatora printzenii Tønsberg (100/1.00/100)与美国的该种样品形成高支持率聚类分支;鳞片树痂衣Phyllopsora porphyromelaena (Vain.) Zahlbr. (99.9/1.00/100)与日本、马来西亚的同种样品聚为一支;对折树花Ramalina conduplicans Vain. (84.2/0.951/98)与日本的同种样品聚为一支。上述系统发育分析结果与基于形态和化学特征的分类结果一致。

地衣体壳状,连续,龟裂或具不规则疣状突起;表面白色至灰绿色;无光泽;无粉芽、裂芽和粉霜层;前地衣体黑色。
子囊盘贴生,圆形,直径0.2-1 mm;盘面平坦或微凸,淡粉色至淡褐色,无粉霜;盘缘常呈淡粉色,颜色明显浅于盘面。果壳明显,宽40-70 μm,边缘呈淡黄色至淡褐色;子实上层浅黄色,厚3-5 μm;子实层无色或淡黄色,厚度为60-80 μm;囊层基淡黄色,厚度为30-40 μm;侧丝无色,单一不分枝,近棒状或在顶端略膨大,宽约1 μm;子囊呈圆柱状或棒状,40-70 × 6-12 μm;子囊孢子35-40 × 3-4.5 μm,6-9隔。分生孢子器未见。
化学:髓层K-,C-,KC-;TLC未检测出地衣特征化合物。
基物:树皮。
世界分布:俄罗斯(Gerasimova et al. 2018)。中国新记录种。

讨论:本种的关键特征是地衣体龟裂或具不规则疣状突起,前地衣体黑色,子囊盘盘面淡粉色至淡褐色,无粉霜,不含地衣特征化合物。覆粉杆孢衣Bacidia suffusa与本种互为姐妹类群且形态相似,但前者子囊盘具粉霜,子囊孢子较长(40-70 × 2.5-4 μm vs. 35-40 × 3-4.5 μm),含黑茶渍素(atranorin) (Gerasimova et al. 2018)。

地衣体壳状,连续,无裂隙或龟裂状,局部呈颗粒状;表面灰绿色;无光泽;前地衣体未见。
子囊盘贴生,圆形,直径0.25-8 mm;盘面平坦,淡褐色,具有白色粉霜,幼时盘面粉霜较多,成熟后不明显;盘缘淡粉色至淡褐色。果壳宽25-50 μm,边缘呈灰褐色;子实上层褐色至深褐色,厚10-15 μm;子实层无色透明,厚40-70 μm;囊层基淡褐色,厚度为25-40 μm;侧丝无色透明,单一不分枝,宽1-1.5 μm;子囊柱状,50-70 × 10-14 μm;子囊孢子40-70 × 2.5-4 μm,9-14隔;分生孢子器未见。
化学:髓层K+黄色,C-,KC-;TLC检测含有黑茶渍素(atranorin)。
基物:树皮。
世界分布:北美洲、俄罗斯、韩国、巴基斯坦(Ekman 1996;Otte 2007;Gerasimova et al. 2018, 2021;Adrees et al. 2022;Lee & Hur 2022)。中国新记录种。

讨论:覆粉杆孢衣的关键特征是子囊盘褐色,盘面具白色粉霜。本种与Bacidia campalea (Tuck.) S. Ekman & Kalb较为相似,地衣体表面均为无裂隙或龟裂状,具褐色子囊盘、淡褐色囊层基,子囊孢子大小相近,但B. campalea地衣体表面呈浅蓝灰色,果壳中具晶体,除黑茶渍素外还含有泽屋萜(zeorin) (Ekman 1996);本种与B. russeola (Kremp.) Zahlbr.具相同的地衣特征化合物,但B. russeola子囊盘盘面无粉霜,子囊孢子5-9隔,不超过10隔(Ekman 1996;Lee & Hur 2022)。
Biatora appalachensis Printzen & Tønsberg, Symb. bot. upsal. 34(no. 1): 345 (2004)
地衣体壳状,疏松固着于基物,连续或龟裂,外形不规则,蔓延状生长;表面黄绿色,无光泽,具粉芽;粉芽堆浅绿色至黄绿色,不规则或呈球状凸起,宽0.5-1 mm;前地衣体不明显。菌丝透明,光合共生藻为绿藻。子囊盘及分生孢子器未见。
化学:髓层K-,C+粉色,KC+红色;TLC检测含有三苔色酸(gyrophoric acid)。
基物:树皮。
世界分布:美国(Lendemer 2023)。中国新记录种。

讨论:本种的关键特征是具浅绿色或黄绿色凸起粉芽堆,次生代谢产物为三苔色酸。本种与Biatora pontica Printzen & Tønsberg均具粉芽,但B. pontica髓层遇C试剂和KC试剂呈橙黄色反应,次生代谢产物为asemone和thiophanic acid (Kukwa et al. 2012)。中国分布的蜡盘衣属物种中,仅普氏蜡盘衣和阿巴拉契亚蜡盘衣具粉芽,但普氏蜡盘衣除含三苔色酸外,还含有阿果斯素(argopsin) (Tønsberg 2002)。
Ramalina sphaerophora Kashiw. & K.H. Moon, in Moon, Ahn, Han & Kashiwadani, J. Jap. Bot. 91(Suppl.): 383 (2016)
地衣体枝状至扁枝状,基部以柄固着于基物,灌木状丛生,质硬,高0.5-2 cm;分枝扁平但无背腹性,粗细不均,宽度为0.5-1 mm,局部中空,膨胀,具穿孔,顶端渐尖,侧生有小刺状分枝;表面淡黄绿色,近基部呈淡褐色,光滑;无粉芽,近分枝顶端具皮质的球形结节;子囊盘及分生孢子器未见。
化学:髓层K-,C-,KC-;TLC检测含有树花地衣酸(obtusatic acid)和松萝酸(usnic acid)。
基物:岩石。
世界分布:韩国(Moon et al. 2016)。中国新记录种。

讨论:本种的关键特征是近分枝顶端具皮质的球形结节,无粉芽,含有树花地衣酸和松萝酸。本种与高峰树花Ramalina almquistii Vain.形态相似,地衣体上均有皮质结节,且不含粉芽,但高峰树花含有石花酸(sekikaic acid)和柔扁枝衣酸(divaricatic acid) (付伟等 2008)。本种与安田氏树花R. yasudae具有相同的地衣特征化合物,但安田氏树花分枝较宽(3-5 mm),具粉芽(付伟等 2008;Moon et al. 2016)。

该种的主要特征为:地衣体龟裂,无前地衣体,子囊盘面褐色至黑色,无粉霜,囊层基浅褐色至深褐色,子囊孢子有较多横隔(3-15个);不含地衣特征化合物。

该种的主要特征为:地衣体鳞片状,覆瓦状排列,鳞片较小,长0.5-2 mm,宽0.5-1 mm,多分裂,顶端常翘起;上表面深绿色至灰绿色,无光泽,有粉霜,无粉芽和裂芽;髓层白色;下表面白色,无光泽,无假根;含阿果斯酸(argopsin)。

该种的主要特征为:地衣体分枝圆柱形至扁平状,假杯点表面或边缘生,无粉芽、裂芽,子囊盘常见;含松萝酸(usnic acid)和水杨嗪酸(salazinic acid)。

本研究对采自安徽省的65份树花科地衣标本进行分类学鉴定,发现树花科地衣4属9种,其中4种为中国新记录种,4种为安徽省新记录种。网杆孢衣Bacidia areolata为2018年发表的新种,此前世界范围内发现于俄罗斯大赫黑契尔州自然保护区(Bolshekhekhtsirskiy State Natural Reserve)海拔160 m的青楷槭树干上(Gerasimova et al. 2018),本研究首次报道该种在中国安徽省大别山区有分布,主要生长于海拔382-911 m的山核桃树干表面。覆粉杆孢衣Bacidia suffusa此前报道于北美洲、俄罗斯、韩国、巴基斯坦等国家和地区(Ekman 1996;Otte 2007;Gerasimova et al. 2018, 2021;Adrees et al. 2022;Lee & Hur 2022),本研究进一步扩大了其分布范围,在我国安徽省大别山区海拔380-1 030 m的山核桃或枳椇树干上亦有分布。阿巴拉契亚蜡盘衣Biatora appalachensis此前仅在美国高海拔地区有相关记录(Lendemer 2023),本研究发现该种在中国安徽省海拔511-1 187 m常绿或落叶阔叶林树干表面较为常见,表明此前我国对该属地衣的研究尚不全面。球顶树花Ramalina sphaerophora于2016年首次描述于韩国,其模式标本生境为海拔5-17 m的岩石表面(Moon et al. 2016),中国发现的该物种主要分布在海拔1 000 m左右的岩石表面。安徽省新记录种施氏杆孢衣Bacidia schweinitzii、普氏蜡盘衣Biatora printzenii、鳞片树痂衣Phyllopsora porphyromelaena和对折树花Ramalina conduplicans广布于安徽省海拔549-1 608 m松属或少数阔叶树干及岩石表面。
树花科中国新记录种和安徽省新记录种的发现,填补了该科部分物种在东亚及长江中下游区域演化研究中的空白。该研究进一步丰富了中国树花科地衣的物种多样性,为我国树花科地衣研究夯实基础,并为《中国地衣志》编研提供基础资料。同时,一系列新记录种的发现也反映出该科在我国的调查覆盖范围和研究深度的不足,亟需开展深入的地衣资源调查和系统分类研究。
