OBJECTIVE

To establish the quality standards for Jianpixiaoxian spray and provide a scientific basis for the optimization of its preparation process and quality control.

METHODS

Jianpixiaoxian spray was prepared using a combination of steam distillation and water decoction methods. The quality of the product was preliminarily evaluated through pH value, filling volume variation, and spray volume. High-performance liquid chromatography (HPLC) was used to establish characteristic fingerprints for different batches, with neochlorogenic acid, chlorogenic acid, and 1, 3-dicaffeoylquinic acid used as marker components for multi-component content determination.

RESULTS

The prepared Jianpixiaoxian spray was a dark brown, clear, and transparent liquid with a fragrant aroma and a bitter taste. The pH value ranged from 6.2 to 7.4, with each bottle containing 9.5 to 10.5 mL and an average spray volume per actuation of 90.7 to 95.4 mg. The similarity of the HPLC fingerprints of 10 batches was above 0.90, with 13 common peaks identified. Among which Peaks 2, 4, and 8 were identified as neochlorogenic acid, chlorogenic acid, and 1, 3-dicaffeoylquinic acid, respectively. The contents of these three components were (0.314 7±0.0057) mg·mL-1, (0.626 3±0.009 1) mg·mL-1, and (0.170 9±0.003 2) mg·mL-1.

CONCLUSION

The optimized preparation process for Jianpixiaoxian spray is feasible. The established HPLC fingerprint and multi-component content determination methods are specific, repeatable, and stable, providing an effective means for quality control and evaluation of this preparation and laying a foundation for its clinical application and industrial production.

目的

建立健脾消腺喷雾剂的质量标准,为其制备工艺的优化和质量控制提供科学依据。

方法

采用水蒸气蒸馏法和水煎煮法制备健脾消腺喷雾剂,通过pH值、装量差异和喷射量对其质量进行初步评价。基于高效液相色谱法(HPLC)构建特征性指纹图谱,并以新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸为指标成分,建立多组分含量测定方法,对不同批次样品的质量进行标准化研究。

结果

所制备的健脾消腺喷雾剂为深褐色澄清透明液体,气芳香,味苦,pH值在6.2~7.4之间,每瓶装量在9.5~10.5 mL范围内,每揿平均喷射量为90.7~95.4 mg。10批样品的HPLC指纹图谱相似度> 0.90,标定了13个共有峰,其中2号峰为新绿原酸,4号峰为绿原酸,8号峰为1,3-二咖啡酰奎宁酸。含量测定结果显示,3种成分的含量分别为(0.314 7±0.005 7)mg·mL-1、(0.626 3±0.009 1)mg·mL-1和(0.170 9±0.003 2)mg·mL-1

结论

本研究优化的健脾消腺喷雾剂制备工艺可行,基于HPLC技术建立的指纹图谱及多组分同步定量分析方法专属性强、重复性高、稳定性好,可有效用于该制剂的质量控制和评价,为其临床应用和工业化生产奠定了基础。

目的

建立健脾消腺喷雾剂的质量标准,为其制备工艺的优化和质量控制提供科学依据。

方法

采用水蒸气蒸馏法和水煎煮法制备健脾消腺喷雾剂,通过pH值、装量差异和喷射量对其质量进行初步评价。基于高效液相色谱法(HPLC)构建特征性指纹图谱,并以新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸为指标成分,建立多组分含量测定方法,对不同批次样品的质量进行标准化研究。

结果

所制备的健脾消腺喷雾剂为深褐色澄清透明液体,气芳香,味苦,pH值在6.2~7.4之间,每瓶装量在9.5~10.5 mL范围内,每揿平均喷射量为90.7~95.4 mg。10批样品的HPLC指纹图谱相似度> 0.90,标定了13个共有峰,其中2号峰为新绿原酸,4号峰为绿原酸,8号峰为1,3-二咖啡酰奎宁酸。含量测定结果显示,3种成分的含量分别为(0.314 7±0.005 7)mg·mL-1、(0.626 3±0.009 1)mg·mL-1和(0.170 9±0.003 2)mg·mL-1

结论

本研究优化的健脾消腺喷雾剂制备工艺可行,基于HPLC技术建立的指纹图谱及多组分同步定量分析方法专属性强、重复性高、稳定性好,可有效用于该制剂的质量控制和评价,为其临床应用和工业化生产奠定了基础。

OBJECTIVE

To establish the quality standards for Jianpixiaoxian spray and provide a scientific basis for the optimization of its preparation process and quality control.

METHODS

Jianpixiaoxian spray was prepared using a combination of steam distillation and water decoction methods. The quality of the product was preliminarily evaluated through pH value, filling volume variation, and spray volume. High-performance liquid chromatography (HPLC) was used to establish characteristic fingerprints for different batches, with neochlorogenic acid, chlorogenic acid, and 1, 3-dicaffeoylquinic acid used as marker components for multi-component content determination.

RESULTS

The prepared Jianpixiaoxian spray was a dark brown, clear, and transparent liquid with a fragrant aroma and a bitter taste. The pH value ranged from 6.2 to 7.4, with each bottle containing 9.5 to 10.5 mL and an average spray volume per actuation of 90.7 to 95.4 mg. The similarity of the HPLC fingerprints of 10 batches was above 0.90, with 13 common peaks identified. Among which Peaks 2, 4, and 8 were identified as neochlorogenic acid, chlorogenic acid, and 1, 3-dicaffeoylquinic acid, respectively. The contents of these three components were (0.314 7±0.0057) mg·mL-1, (0.626 3±0.009 1) mg·mL-1, and (0.170 9±0.003 2) mg·mL-1.

CONCLUSION

The optimized preparation process for Jianpixiaoxian spray is feasible. The established HPLC fingerprint and multi-component content determination methods are specific, repeatable, and stable, providing an effective means for quality control and evaluation of this preparation and laying a foundation for its clinical application and industrial production.

腺样体/扁桃体病理性肥大是儿童期常见的上呼吸道疾病,其持续性肥大导致张口呼吸,进而引发特征性腺样体面容,严重时可发展为阻塞性睡眠呼吸暂停低通气综合征(OASHS),主要临床特征包括呼吸睡眠异常,认知功能障碍及行为异常等,未经及时干预的重度OASHS患者进一步可导致心血管系统并发症,甚至危及生命[1]。近年来,儿童呼吸系统健康问题日益受到学界重视。腺样体和扁桃体作为呼吸道的重要免疫屏障,长期暴露于炎性刺激下易发生病理性肥大[2]。然而,由于儿童手术麻醉存在潜在风险[3]、术后容易复发[4]以及目前关于儿童腺样体扁桃体切除术对免疫功能远期效应的循证医学证据仍显不足[5],开发安全有效的药物治疗策略已成为临床医师的重点研究方向。近年来,中药鼻腔给药系统的研究取得了重要突破,其独特的治疗价值在鼻部疾病领域日益凸显。研究表明,该给药途径具有高靶向性、高安全性以及药物依从性好的特点,特别适合儿童等特殊人群的长期治疗。这些优势使得中药鼻腔给药在治疗鼻部疾病中展现出独特的价值[6]。健脾消腺喷雾剂(又可叫鼻炎通窍喷雾剂)是一种基于传统中医理论和临床经验研发的中药复方鼻腔给药制剂[7,8],由苍耳子、辛夷、白芷、薄荷、石菖蒲和路路通六味中药组成。方中苍耳子为君药,发挥祛风通窍之功效[9];配伍辛夷、白芷为臣药,协同增强疏风通鼻的作用;薄荷辛散上行,能疏风清利头目[10,11];石菖蒲和路路通则发挥祛风通窍的协同作用[12,13]。诸药合用,共奏散风消炎、宣通鼻窍之功,主要用于治疗慢性鼻炎、过敏性鼻炎及鼻窦炎等鼻部疾病[14]。吴超霞等[15]观察发现由苍耳子,辛夷等中药组成的鼻鼽通窍方鼻喷雾剂可有效改善鼻部症状、鼻体征用于治疗儿童过敏性鼻炎。
然而,中药复方制剂的质量控制一直是中药现代化研究中的关键问题。中药复方多组分、多靶点的特性决定了单一指标成分分析无法完整表征其整体质量特征[16,17]。因此,中药复方制剂的质量控制研究中,指纹图谱技术与多组分定量分析相结合已成为关键评价方法。本研究旨在通过优化健脾消腺喷雾剂的制备工艺,建立其高效液相色谱(HPLC)指纹图谱及多组分含量测定方法,以新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸为指标成分,构建科学完善的制剂质量标准体系,为其工业化生产提供关键技术支撑,同时也为临床疗效评价奠定物质基础。

健脾消腺喷雾剂(自制,自拟批号:S1~S10);绿原酸标准品(批号:ST02150120,纯度≥98%,上海诗丹德标准服务技术服务有限公司);新绿原酸标准品(批号:MUST-21030108,纯度≥98%,成都曼思特生物科技有限公司)、1,3-二咖啡酰奎宁酸标准品(批号:MUST-17022608,纯度≥98%,成都曼思特生物科技有限公司);色谱级乙腈和甲醇(Fisher Chemical);其余试剂均为分析纯。
该方由临床经验用方化裁而来,结合药材的挥发性成分特性制定提取工艺[18],处方组成及用量见表1。对辛夷、薄荷和石菖蒲三味药材制备芳香水的工艺研究过程中,考虑工艺的便利性选择水蒸气蒸馏法进行提取工艺设计。通过单因素变量控制法,重点考察提取工艺中加水倍数、浸泡时间及提取时间等关键参数对挥发油提取效率的影响。
考察苍耳子、白芷、路路通三味药材采用水提煎煮法,出膏率是中药材提取过程中的关键因素[19],绿原酸为君药苍耳子的指标性成分[20],因此在水提工艺过程中考察加水倍数和提取时间对绿原酸含量及出膏率的影响。试验中,固定了处方比例,选取了8、11、14、17倍的加水量进行试验,每次试验均包括0.5 h的浸泡时间和两次各0.5 h的水提煎煮;对于煎煮时间的选择,在固定处方比例和14倍加水量的基础上,进行了0.5、1、1.5、2 h的煎煮时间试验。
健脾消腺喷雾剂中,绿原酸、新绿原酸和1,3-二咖啡酰奎宁酸3种成分来源于方中的苍耳子、白芷和路路通三味药材。建立健脾消腺喷雾剂中的含量测定方法,能够有效控制制剂质量,确保其疗效稳定,为临床应用提供科学依据。
采用kromasil C18色谱柱(250 mm×4.6 mm,5 μm);检测波长330 nm;流速:1.0 mL·min-1;柱温维持30 ℃;进样体积10 μL;流动相:乙腈(A)-0.2%磷酸溶液(B),梯度洗脱0~5 min 5%A,5~30 min 5%~22%A,30~55 min 22%~30%A,55~56 min 30%~5%A,56~60 min 5%A。
精密称取新绿原酸、绿原酸及1,3-二咖啡酰奎宁酸标准品,以70%甲醇溶解并定容至25 mL,制得浓度分别为0.212 5、0.503 0、0.151 3 mg·mL-1的储备液。精密量取各储备液1 mL,置10 mL容量瓶中,70%甲醇定容,获得终浓度分别为21.25、50.30、15.13 μg·mL-1的标准品工作液。
精密移取新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸标准品储备液各2.5 mL,置25 mL容量瓶中,70%甲醇定容,制得浓度分别为21.25、50.30、15.13 μg·mL-1的混合标准品溶液。
精密移取各批次样品溶液(S1-S10)2 mL,分别置于10 mL具塞锥形瓶中,加入适量70%甲醇后称重,经超声处理(250 W,35 kHZ)30 min,待样品冷却至室温后再次称重,用70%甲醇补足提取过程中损失的溶剂重量,充分混匀得供试品溶液。
在既定色谱条件下,精密移取供试品溶液、混合标准品溶液以及阴性空白溶液进行测定,并记录色谱图。
分别精密移取0.5、0.8、1.0、1.3、1.6、2 mL混合标准品溶液,70%甲醇定容至10 mL,配制6个浓度梯度的标准溶液。按色谱条件下进样测定,以各组分浓度为横坐标(x),相应峰面积为纵坐标(y)进行线性回归,建立标准曲线。
取混合标准品溶液适量,在色谱条件下重复进样测定6次,计算RSD值。
取同批次样品(S1),平行制备6份供试品溶液,分别按照色谱条件进样分析,计算RSD值。
取同一批样品溶液(S1)2 mL,分别在0、2、4、8、12和24 h时间点进行测定,计算RSD值。
精密量取已知含量的S1批次样品溶液9份,每份2 mL,分别加入适量混合标准溶液进行测定,计算回收率。
取10批健脾消腺喷雾剂样品,制备供试品溶液,在既定色谱条件下分别进样测定,外标法计算含量。
采用HPLC对10批健脾消腺喷雾剂(S1~S10)进行指纹图谱分析,基于既定色谱条件获取数据后,导入2012版相似度评价系统进行分析,生成各批次指纹图谱叠加图。在相似度评价系统中,以S1批次样品的色谱图为参照图谱,采用多点校正结合自动匹配算法(时间窗宽度设定为0.3 min)进行谱图对齐,通过中位数法构建对照指纹图谱。
依据2020年版中国药典四部通则0631的检测规范,对健脾消腺喷雾剂3个生产批次(S8~S10)进行了pH值检测。
依据2020年版中国药典四部通则0106鼻用制剂的装量要求,对健脾消腺喷雾剂3个生产批次(S8~S10)进行了装量检测。方法如下:每批样品随机抽取4瓶,充分振摇混匀后,将内容物定量转移至25 mL量筒中。室温条件下测定药液体积。
依据2020年版中国药典四部通则0112喷雾剂检测规范,对健脾消腺喷雾剂3个生产批次(S8~S10)进行喷射性能评价。方法如下:随机抽取样品各4瓶,经试喷预处理后擦净喷头,记录初始重量,连续喷射10次后再次擦净称重,计算单瓶每揿平均喷射量。
图1显示了不同条件对出油量的影响,其中图1A表明加水倍数对出油量有显著影响,10倍加水量时提取率最高,过多或过少的加水量均会导致出油量下降。图1B显示浸泡时间对出油量的影响,其中1 h浸泡时间对应的出油量最高。图1C则显示了提取时间对出油量的影响,2.5 h后出油量趋于稳定,说明此时间为最佳提取时间。综合图1的试验结果,选择10倍加水量、1 h浸泡和2.5 h提取时间作为最优提取条件。
图2A显示,随着加水量的增加,出膏率和绿原酸含量均显著增加,但当加水量达到14倍时,绿原酸含量达到峰值,继续增加反而导致其含量降低。图2B则表明,随着煎煮时间的延长,出膏率和绿原酸含量呈现下降趋势,这可能是因为过长的煎煮时间导致药材中有效成分的损耗。综合图2的试验结果,煎煮工艺选择14倍加水量,每次煎煮0.5 h作为最优提取条件。这些条件不仅能够保证较高的绿原酸含量和出膏率,还能避免因用水量过多或煎煮时间过长而导致的成分损耗,从而提高药材提取的效率和质量。
综合上述试验结果,确定健脾消腺喷雾剂的制备工艺为每剂药材用量为辛夷10 g、薄荷6 g、石菖蒲10 g,加入10倍体积的水,采用水蒸气蒸馏技术提取2.5 h,折合每剂可收集挥发油约0.8 mL;将提取后的药渣与苍耳子(8 g)、白芷(10 g)、路路通(10 g)粗粉混合,加水煎煮两次,第1次加药材与水质量比为14倍体积的水,第2次加12.5倍体积的水,每次煎煮30 min,合并两次煎液,过滤后减压浓缩至60 ℃条件下相对密度达1.1~1.2,离心取上清液,加入预制备的挥发油,加水定容至规定体积,用4% NaOH溶液调节pH至6.5,混匀后静置,分装,灭菌,即得成品。
取上述样品(S8~S10)3批各4瓶,经5秒振荡混匀后,于自然光线下观察。结果显示,样品溶液呈深褐色,澄清透明,无悬浮物或沉淀。开盖后气味芳香,味苦,符合中国药典鼻用喷雾剂的外观、气味及性状要求。
新绿原酸、绿原酸、1,3-二咖啡酰奎宁酸的保留时间分别为14、19、26 min,阴性空白溶液在目标成分保留时间内未检出干扰峰,而标准溶液与供试品溶液在相应保留时间处均呈现出分离良好的色谱峰(图3),结果表明该方法能有效区分各目标成分,阴性样品无干扰,所建立的分析方法专属性良好。
新绿原酸,绿原酸和1,3-二咖啡酰奎宁酸的回归方程如表2所示,3种成分分别在进样量10.63~42.50 μg·mL-1、25.15~100.60 μg·mL-1和7.57~30.27 μg·mL-1浓度范围内与峰面积呈较好的线性关系,可用于后续的含量测定。
新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸3个成分的日内平均浓度RSD值分别为1.17%、0.43%、0.92%;日间平均浓度RSD值分别为1.15%、0.31%、0.55%,均小于2%,表明精密度良好,符合分析方法验证要求。
新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸的含量分别为(24.88±0.11)μg·mL-1、(49.70±0.18)μg·mL-1、(13.90±0.25)μg·mL-1,其中RSD值分别为0.43%、0.35%、1.78%,均小于2%,表明该方法重复性符合分析要求。
新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸的平均含量分别为(25.30±0.40)μg·mL-1、(50.37±0.52)μg·mL-1、(14.30±0.16)μg·mL-1RSD值分别为1.59%、1.02%、1.12%,表明样品溶液在24 h内稳定性良好。
结果显示,新绿原酸、绿原酸、1,3-二咖啡酰奎宁酸回收率范围分别为(97.82~98.59)%、(98.46~98.67)%、(98.05~98.30)%之间,RSD值均小于2%,该结果证实本方法具有良好的准确度,能够满足定量分析的要求。
结果见表3,10批样品中新绿原酸、绿原酸、1,3-二咖啡酰奎宁酸的含量分别为(0.314 7±0.005 7)mg·mL-1、(0.626 3±0.009 1)mg·mL-1、(0.170 9±0.003 2)mg·mL-1
图4显示了10批样品的指纹图谱叠加图,经分析共识别出13个稳定的共有特征峰,结果如图5。通过将供试品色谱峰与标准品对照,成功鉴定出以下特征成分:2号峰为新绿原酸,4号峰为绿原酸,8号峰为1,3-二咖啡酰奎宁酸,结果如图6。对生成的指纹图谱进行相似度评价,如表4所示,10批样品相似度均在0.90以上,说明健脾消腺喷雾剂不同批次间质量一致性良好,符合中药制剂质量控制要求。
各批次样品pH值在6.2~7.4范围内,符合中国药典对鼻用制剂pH值(5.0~8.0)的质量要求。
通过装量检测方法测得每瓶样品装量在9.5~10.5 mL范围内,均符合鼻用喷雾剂的装量标准要求。
根据喷雾剂的喷射检测方法测得各批次单瓶每揿平均喷射量为90.7~95.4 mg,符合药典对喷雾剂喷射性能的质量要求。
通过查阅、归纳中国药典相关药材的提取方式结合药材的特性,采用水蒸气蒸馏法提取辛夷、薄荷、石菖蒲中的挥发油,以挥发油提取率为指标,确定了最佳的挥发油提取工艺为在处方比例药材中加入10倍的水,浸泡1 h,提取2.5 h。通过水煎煮法提取苍耳子、白芷、路路通的有效成分,以绿原酸和出膏率为指标,确定了最佳提取工艺为在处方比例药材中一煎加入14倍水,二煎加12.5倍水,每次煎煮30 min,合并煎液,滤过,滤液浓缩至相对密度为1.1~1.2(60 ℃)的清膏,离心,取上清液即得。
健脾消腺喷雾剂由6味药材组成,其中苍耳子为君药,酚酸类物质是喷雾剂治疗AR的主要活性成分,在关键成分基础上,确定新绿原酸、绿原酸、1,3-二咖啡酰奎宁酸作为指标性成分。本研究以乙腈-0.2%磷酸溶液为流动相,尝试多个洗脱条件,最终用梯度洗脱程序同时测定这3种目标成分,该方法分离度好,基线平稳,在该方法下3种指标成分的含量测定结果分别为(0.314 7±0.005 7)mg·mL-1、(0.626 3±0.009 1)mg·mL-1、(0.170 9±0.003 2)mg·mL-1,进一步验证了制剂的质量可控性。
本研究围绕健脾消腺喷雾剂的制备工艺优化和质量标准建立开展系统研究,科学的实验设计和系统的质量评价,为制剂产业化转化和临床应用奠定了科学基础。优化后的制备工艺稳定可控,能够有效提取方中各药材的有效成分,制备出的喷雾剂外观性状良好,pH值、装量差异和喷射量等关键指标均符合中国药典相关要求,确保了产品质量的批间一致性。在质量控制方面,本研究采用HPLC建立了健脾消腺喷雾剂的特征指纹图谱确定了13个共有色谱峰,并以新绿原酸、绿原酸和1,3-二咖啡酰奎宁酸作为定量分析的指标成分,所有批次指纹图谱相似度均在0.90以上,充分证实了生产工艺的稳定性和产品质量的可重复性。同时,这些成分主要来源于方中的苍耳子、白芷和路路通,其含量测定不仅反映了药材的质量,也为制剂的整体质量控制提供了重要依据。此外,本研究采用系统化的方法验证方案对所建立的HPLC定量分析方法进行了全面评估,结果表明各项指标均符合分析方法验证的技术要求,能够有效应用于健脾消腺喷雾剂的质量控制。指纹图谱结合多组分含量测定的模式,不仅克服了单一成分评价的局限性,还为中药复方制剂的整体质量评价提供了科学依据。
综上所述,本研究通过科学的工艺参数优化和严格的质量控制体系,为健脾消腺喷雾剂的标准化生产奠定了坚实基础。未来的研究将进一步检测其他药材的特征成分,对指纹图谱中其他特征峰进行准确标定并探索其药效学机制,并通过临床试验验证其疗效和安全性,以期为儿童腺样体/扁桃体肥大、过敏性鼻炎等鼻部疾病的治疗提供一种更为有效、安全的中药复方制剂。
