OBJECTIVE

To establish a size exclusion high-performance liquid chromatography (SEC-HPLC) method for the determination of the purity of recombinant human basic fibroblast growth factor (rh-bFGF).

METHODS

The purity of rh-bFGF solution was determined using TSK G2000SWXL column and high-performance liquid chromatography, and the specificity, repeatability, precision, detection limit, stability, and durability of the method were evaluated.

RESULTS

The blank solvent has no interference with the rh-bFGF main peak, and the degradation impurities under various destruction conditions didn't interfere with the determination of the rh-bFGF peak, and the purity index of the rh-bFGF peak was greater than 990. The repeatability and intermediate precision experimental results showed that the separation degree of impurities and the rh-bFGF main peak was greater than 1.5, and the RSD was 0.36% and 0.64%, respectively. The detection limit was 0.185 μg·mL-1. The durability experiment results show that fluctuations in flow rate, column temperature, and wavelength had no effect on the detection results, and the RSD of each condition was less than 2.0%.

CONCLUSION

The SEC-HPLC method developed has the advantages of simple operation, accurate results, strong specificity, high stability and good applicability, and can be used for rh-bFGF purity determination.

目的

建立测定重组人碱性成纤维细胞生长因子(recombinant human fibroblast growth factor-basic,rh-bFGF)纯度的分子排阻高效液相色谱法(SEC-HPLC)。

方法

采用TSK G2000SWXL色谱柱和高效液相色谱法,测定rh-bFGF原液的纯度,并对方法的专属性、重复性、精密度、检测限、稳定性及耐用性等指标进行评估。

结果

空白溶剂对rh-bFGF的检测无干扰,各破坏条件下的降解杂质不干扰rh-bFGF的测定,且rh-bFGF峰纯度均大于990;重复性及中间精密度实验结果显示杂质与rh-bFGF峰的分离度大于1.5,RSD分别为0.36%、0.64%;检测限为0.185 μg·mL-1;耐用性实验结果表明,流速、柱温、波长的波动,对检测结果无影响,各条件下的RSD小于2.0%。

结论

所开发的SEC-HPLC方法操作简便,结果准确,专属性强,稳定性高,适用性良好,可用于rh-bFGF纯度测定。

目的

建立测定重组人碱性成纤维细胞生长因子(recombinant human fibroblast growth factor-basic,rh-bFGF)纯度的分子排阻高效液相色谱法(SEC-HPLC)。

方法

采用TSK G2000SWXL色谱柱和高效液相色谱法,测定rh-bFGF原液的纯度,并对方法的专属性、重复性、精密度、检测限、稳定性及耐用性等指标进行评估。

结果

空白溶剂对rh-bFGF的检测无干扰,各破坏条件下的降解杂质不干扰rh-bFGF的测定,且rh-bFGF峰纯度均大于990;重复性及中间精密度实验结果显示杂质与rh-bFGF峰的分离度大于1.5,RSD分别为0.36%、0.64%;检测限为0.185 μg·mL-1;耐用性实验结果表明,流速、柱温、波长的波动,对检测结果无影响,各条件下的RSD小于2.0%。

结论

所开发的SEC-HPLC方法操作简便,结果准确,专属性强,稳定性高,适用性良好,可用于rh-bFGF纯度测定。

OBJECTIVE

To establish a size exclusion high-performance liquid chromatography (SEC-HPLC) method for the determination of the purity of recombinant human basic fibroblast growth factor (rh-bFGF).

METHODS

The purity of rh-bFGF solution was determined using TSK G2000SWXL column and high-performance liquid chromatography, and the specificity, repeatability, precision, detection limit, stability, and durability of the method were evaluated.

RESULTS

The blank solvent has no interference with the rh-bFGF main peak, and the degradation impurities under various destruction conditions didn't interfere with the determination of the rh-bFGF peak, and the purity index of the rh-bFGF peak was greater than 990. The repeatability and intermediate precision experimental results showed that the separation degree of impurities and the rh-bFGF main peak was greater than 1.5, and the RSD was 0.36% and 0.64%, respectively. The detection limit was 0.185 μg·mL-1. The durability experiment results show that fluctuations in flow rate, column temperature, and wavelength had no effect on the detection results, and the RSD of each condition was less than 2.0%.

CONCLUSION

The SEC-HPLC method developed has the advantages of simple operation, accurate results, strong specificity, high stability and good applicability, and can be used for rh-bFGF purity determination.

碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)是一种多功能的多肽生长因子,也是FGF家族的原型成员之一。生物学效应非常广泛,其在血管形成、促进创伤愈合与组织修复、促进组织再生和神经组织生长发育过程中起着十分重要的作用[1],适用于烧伤创面(包括浅、深Ⅱ度、肉芽创面)、慢性创面(包括慢性肉芽创面、溃疡和褥疮等)、新鲜创面(包括创伤、手术创伤等)以及难愈性创面愈合等[2,3],具有重要的临床研究价值。目前rh-bFGF纯度测定方法采用《生物制品检验技术操作规范》中重组细胞因子产品蛋白质纯度测定方法(分子排阻高效液相色谱法)测定[4],该方法针对性不强,对rh-bFGF纯度测定存在分离度差、耐用性差等问题。因此建立针对rh-bFGF纯度测定的方法具有十分重要的意义。
高效液相色谱仪(安捷伦,型号:1260 Infinity Ⅱ)、电子天平(梅特勒托利多,型号:ME204/02)、pH计(梅特勒托利多,型号:S220)、超声波清洗仪(深圳市福洋科技集团有限公司,型号:F-100S)、TSK-gel 3000SWXL色谱柱(日本东曹,规格:7.5 mm×30 cm)、TSKgel G2000SWXL色谱柱(日本东曹,规格:7.8 mm×30 cm)、TSKgel guardcolumn SWXL保护柱(日本东曹,规格:6.0 mm×4 cm)。
重组人碱性成纤维细胞生长因子原液(朗肽生物制药股份有限公司,批号:C202204010)、磷酸二氢钠(湖南九典宏阳制药公司,批号:T202110I01)、磷酸氢二钠(湖南九典宏阳制药公司,批号:TF22220103)、L-精氨酸单盐酸盐(Sigma,批号:WXBD6298V)、EDTA-Na2(湖南尔康制药公司,批号:102320210901)、氯化钠(广州试剂厂,批号:2022041221)、磷酸二氢钾(湖南九典宏阳制药公司,批号:TF05210101)。
采用《生物制品检验技术操作规范》中重组细胞因子产品蛋白质纯度测定方法(分子排阻高效液相色谱法)对rh-bFGF进行纯度测定,发现其对rh-bFGF纯度测定存在分离度差,峰对称性等问题,适用性较差。故为了建立一种适用于rh-bFGF纯度测定的方法,本研究对色谱柱、流动相、流速、柱温、进样量等色谱条件进行了选择及优化[5-9],并确定最终的色谱条件,色谱条件见表1
表2结果可知,采用2种方法,对同一rh-bFGF原液进行纯度测定,2种方法有显著的差异;新开发的方法能很好地将目的主峰与杂质峰分开,分离度为2.23,拖尾因子更趋近于1,说明峰型较好,而《生物制品检验技术操作规范》的检测方法分离度仅为1.26,分离度较差,且拖尾因子偏离1较大。可见,新开发的纯度测定方法,在分离度、拖尾因子、测定结果的准确性等方面均优于《生物制品检验技术操作规范》的检测方法,为进一步验证新开发方法在测定rh-bFGF纯度方面的适用性,笔者对该方法从专属性、重复性、精密度、溶液稳定性、耐用性等方面进行了验证。
色谱条件见表1
取空白溶剂(50 mmol·L-1 PB)和rh-bFGF原液按照“3.1”项下色谱条件进行测定,空白溶剂在目的峰积分范围内应无干扰峰。结果见图1~2,结果显示空白溶剂保留时间不在rh-bFGF主峰和杂质峰积分范围内,说明空白溶剂对rh-bFGF主峰和杂质峰的测定无干扰。
取rh-bFGF原液适量,分别放置于高温、光照、氧化条件下进行强制破坏,制成适当的溶液并进样,按建立的方法进行分析,结果表明本品在高温、光照、氧化破坏性条件下,降解杂质峰与主峰的分离度均大于1.5,且主峰UV纯度均大于990,符合要求,说明降解杂质不干扰主成分的测定。
取6份供试品溶液,每份供试品溶液按照建立的方法进行分析,结果显示,连续进样6次的供试品溶液纯度均大于95.0%,纯度RSD为0.36%,小于2.0%,符合要求,说明该方法重复性良好。
由不同的分析人员在不同的日期取6份相同批次的供试品溶液按照建立的方法进行测定,与重复性纯度数据进行RSD计算,结果显示12份供试品纯度均大于95.0%,纯度的RSD为0.64%,小于2.0%,符合要求,说明该方法精密度良好。
供试品溶液在2 ℃~8 ℃条件下放置0、1、2、3、5、7、9 h后取样按照建立的方法进行检测,结果显示各时间点供试品溶液纯度均大于95.0%,各时间点纯度RSD为0.47%,小于2.0%,符合要求,表明供试品溶液在2 ℃~8 ℃条件下放置9 h内溶液稳定性良好。
分别在流速为0.48、0.50、0.52 mL·min-1对同一样品进行测定,主成分峰与相邻杂质峰分离度>1.5,峰纯度RSD为0.45%;分别在柱温为18 ℃、20 ℃、22 ℃,对同一样品进行测定,主成分峰与相邻杂质峰分离度>1.5,峰纯度RSD为0.54%;分别在检测波长278、280、282 nm,对同一样品进行测定,主成分峰与相邻杂质峰分离度>1.5,峰纯度RSD为0.65%;说明该方法耐用性良好,适用性强。
取rh-bFGF原液逐级稀释后进样,以3倍信噪比为检出限最低响应值,计算结果检测限为0.185 μg·mL-1
本文按照2020年版中国药典四部通则9101分析方法验证指导原则要求,对新开发的方法从专属性、精密度、溶液稳定性、耐用性、检测限进行了方法学验证,结果表明,该方法具有良好的适用性和专属性,目的蛋白色谱峰峰型良好,无杂质峰干扰,精密度和耐用性良好,RSD均小于2.0%,各项验证结果均符合判定标准,适用于rh-bFGF纯度的检测。
本方法与《生物制品检验技术操作规范》中重组细胞因子产品蛋白质纯度测定方法相比,测定rh-bFGF原液纯度,分离度更好,准确度更好,更加具有针对性,且本方法具有操作简便、耗时短等优点,非常适合rh-bFGF产品过程控制,对中间样品的快速检测能及时反映生产过程中出现的问题,能够更好地控制中间产品质量,亦可以准确评价rh-bFGF终产品的质量。本方法的成功开发及验证,对于生物制品rh-bFGF的质量控制具有十分重要的意义。
