Samples of Jumbo flying squid (Dosidicus gigas) were collected from Chinese squid jigging vessels in the high seas of Ecuador, Peru and Chile in the southeast Pacific Ocean in 2015 and 2017. The trace elements in the eye lenses of D. gigas were measured to study the geographical differences and their relationship with sea surface temperature. The results showed that elements such as Mg25, Ni60, Cu63, Sr88 and Ba137 in the peripheral part of eye lenses of D. gigas in the high seas of Ecuador, Peru and Chile were significant difference, while there were no significant differences between each two sea areas in the elements of Na23, Al27, Si29, P31, Ca43, Mn55, Zn66 and Pb. Except for Sr88, Ba137, Fe57 and Ni60, which showed significantly linear negative correlations with SST (p<0.05), there were no significant correlations with SST in the other elements (p>0.05). Sr88, Ba137, Fe57 and Ni60 could be regarded as indicator elements of the ambient temperature of D. gigas, and Ba137 could also be regarded as indicator element of the water depth and upwelling. Consequently, trace elements in eye lenses could be used to reconstruct the habitat environment of cephalopod.

本研究以2015年和2017年我国远洋鱿钓渔船在东南太平洋的厄瓜多尔、秘鲁和智利公海生产时采集的茎柔鱼眼睛晶体为研究材料,采用LA-ICPMS测定了眼睛晶体外缘的微量元素,比较微量元素地理区域间差异,分析微量元素浓度与茎柔鱼栖息水温的关系。结果显示,厄瓜多尔、秘鲁和智利公海的茎柔鱼眼睛晶体外缘Mg25、Ni60 、Cu63、Sr88和Ba137等元素在3个海区之间存在显著差异(p<0.05),Na23、Al27、Si29、P31、Ca43、Mn55、Zn66和Pb等元素在两两海区之间均不存在显著差异(p>0.05)。除了Sr88、Ba137、Fe57和Ni60与海表面温度呈显著的线性负相关,其他元素与海表面温度均无显著的相关关系。分析认为,Sr88、Ba137、Fe57和Ni60均可以看作茎柔鱼生活水温的指示元素,Ba137元素还可以看作茎柔鱼生活水深以及上升流的指示元素。研究认为,眼睛晶体微量元素可用于头足类栖息环境的重建。

本研究以2015年和2017年我国远洋鱿钓渔船在东南太平洋的厄瓜多尔、秘鲁和智利公海生产时采集的茎柔鱼眼睛晶体为研究材料,采用LA-ICPMS测定了眼睛晶体外缘的微量元素,比较微量元素地理区域间差异,分析微量元素浓度与茎柔鱼栖息水温的关系。结果显示,厄瓜多尔、秘鲁和智利公海的茎柔鱼眼睛晶体外缘Mg25、Ni60 、Cu63、Sr88和Ba137等元素在3个海区之间存在显著差异(p<0.05),Na23、Al27、Si29、P31、Ca43、Mn55、Zn66和Pb等元素在两两海区之间均不存在显著差异(p>0.05)。除了Sr88、Ba137、Fe57和Ni60与海表面温度呈显著的线性负相关,其他元素与海表面温度均无显著的相关关系。分析认为,Sr88、Ba137、Fe57和Ni60均可以看作茎柔鱼生活水温的指示元素,Ba137元素还可以看作茎柔鱼生活水深以及上升流的指示元素。研究认为,眼睛晶体微量元素可用于头足类栖息环境的重建。

Samples of Jumbo flying squid (Dosidicus gigas) were collected from Chinese squid jigging vessels in the high seas of Ecuador, Peru and Chile in the southeast Pacific Ocean in 2015 and 2017. The trace elements in the eye lenses of D. gigas were measured to study the geographical differences and their relationship with sea surface temperature. The results showed that elements such as Mg25, Ni60, Cu63, Sr88 and Ba137 in the peripheral part of eye lenses of D. gigas in the high seas of Ecuador, Peru and Chile were significant difference, while there were no significant differences between each two sea areas in the elements of Na23, Al27, Si29, P31, Ca43, Mn55, Zn66 and Pb. Except for Sr88, Ba137, Fe57 and Ni60, which showed significantly linear negative correlations with SST (p<0.05), there were no significant correlations with SST in the other elements (p>0.05). Sr88, Ba137, Fe57 and Ni60 could be regarded as indicator elements of the ambient temperature of D. gigas, and Ba137 could also be regarded as indicator element of the water depth and upwelling. Consequently, trace elements in eye lenses could be used to reconstruct the habitat environment of cephalopod.

茎柔鱼(Dosidicus gigas)是广泛洄游的大洋性浅海种,主要分布于东太平洋30°N~40°S的海域,近年来,其分布区域逐步向两极扩散,最北可达到阿拉斯加60°N,最南可达到智利南部50°S[1]。茎柔鱼资源丰富,其丰度与补充量和环境息息相关[2-4],在秘鲁已形成了重要的近岸手工性渔业和公海工业性渔业[56]。20世纪90年代初,日本和韩国开始对秘鲁海域的茎柔鱼进行大规模商业性捕捞[7]。2001年我国渔船在秘鲁公海开始捕捞[7],目前已开发厄瓜多尔、秘鲁和智利公海3大渔场,近年来最高年产量超过30万t。
鱼类眼睛晶体的生长是上皮细胞由“前极”开始不断纤维化并延长至“后极”以及一层层累积的过程[8]。由于仅上皮细胞层和新形成的纤维层存在蛋白质合成的细胞器以及其他代谢和分解过程[9],眼睛晶体内部成熟纤维细胞层的蛋白质代谢不活跃,其中的微量元素一旦沉积将不会被替换,因此可反映鱼类所对应成长阶段的生活史信息[1011]
目前,耳石[1215]、内壳[1617]、角质颚[1819]等硬组织中的微量元素已被广泛用于头足类种群结构[16, 20]、栖息环境[21]和迁徙路线[22]等方面的研究。有研究报道[10, 23-25]称,由于鱼类眼睛晶体记录了其整个生命周期内的生活史信息,因此可视为判定鱼类种群的硬组织材料的有力补充。然而,截至目前关于头足类眼睛晶体微量元素的报道见到的仅有1篇[26]。本研究采用LA-ICPMS测定厄瓜多尔、秘鲁和智利公海茎柔鱼眼睛晶体外缘的微量元素,比较微量元素地理区域间差异,分析微量元素浓度与茎柔鱼栖息水温的关系。
本文从2015年和2017我国鱿钓生产船在东南太平洋公海(图1)生产时捕捞的渔获物中随机选取28尾茎柔鱼作为实验样本,其中厄瓜多尔公海10尾(3°21′~8°26′S,84°07′~91°52′W,胴长范围为210~357 mm,体质量为264~1 295 g),秘鲁公海13尾(9°16′~15°22′S,79°45'~85°03'W,胴长范围为226~352 mm,体质量为280~1 301 g),智利公海5尾(37°06′~40°00′S,79°00'~83°00'W,胴长范围为301~470 mm,体质量为538~3 012 g)(表1)。所有样品在船上冷冻保存,在实验室解冻后进行生物学测定、性别鉴定和性成熟划分。测定胴长精确至1 mm,体质量精确至1 g。用手术刀将眼睛角膜切口,用镊子取出眼睛晶体,然后用去离子水冲洗、去污之后置于75%乙醇中保存。
取出保存于75%酒精中的眼睛晶体,用游标卡尺测定晶体最大直径,精确至0.01 mm。由于眼睛晶体是由前后两个部分组成(图2),取出后将两者分离,保存体积较大的后半部分(图3a),使其平放于塑料模具中(图3b),倒入配置好的冷埋树脂溶液进行包埋(图3c),在常温避光处放置待其硬化;将硬化后的树脂块用强力热熔胶黏于载玻片上,先将底部磨平(图3d),然后倒转180°研磨另一面(图3e),研磨时先后以120目、600目、1200目、2 500目水磨砂纸研磨成厚度约为0.5 mm的薄片,然后再用0.05 μm氧化铝剂对薄片进行抛光(图3f)。
为减少污染物对元素测试带来的干扰,研磨后的晶体切片在电阻率大于18 Ω的去离子水中清洗5 min后在Class-100层流柱中晾干。在晶体的外缘选取1个取样点(图4)。
晶体微量元素浓度在武汉上谱分析科技有限责任公司利用LA-ICPMS完成。详细的仪器参数和分析流程见文献[27]。GeolasPro激光剥蚀系统由COMPexPro 102 ArF 193 nm准分子激光器和MicroLas光学系统组成,ICPMS型号为Agilent 7700 e。激光剥蚀过程中采用氦气(流速:0.7 L/min)作载气,氩气(流速:0.8 L/min)为补偿气以调节灵敏度,二者在进入ICPMS之前通过一个T型接头混合,激光剥蚀系统配置有信号平滑装置。本次分析的激光束斑直径和频率分别为44 μm和5 Hz。微量元素浓度处理中采用玻璃标准物质BHVO-2G、BCR-2G、BIR-1G和SRM610进行多外标无内标校正。每个时间分辨分析数据包括20~30 s的空白信号和50 s的样品信号。对分析数据的离线处理(包括对样品和空白信号的选择、仪器灵敏度漂移校正以及元素浓度计算)采用软件ICPMSDataCal完成。
在http://iridl.ldeo.columbia.edu下载各采样站点1周前海表面温度(Sea Surface Temperature, SST)均值数据,空间分辨率为0.1°×0.1°。
采用ANOVA分析各元素海区间的差异,利用线性回归方程拟合各元素与SST关系。
茎柔鱼眼睛晶体全元素测定实验结果显示,在仪器有效检测范围内的元素有Na23、Mg25、Al27、Si29、P31、K39、Ca43、Mn55、Fe57、Ni60、Cu63、Zn66、Sr88 、Ba137和Pb共15种,其中P31浓度最高,之后从高到低依次为Al27、Ca43、Si29、Na23、K39、Mg25、Zn66、Cu63、Fe57、Mn55、Ni60、Sr88、Ba137和Pb(表2)。按海区分析显示(表2),厄瓜多尔公海P31浓度最高,之后依次为Al27、Ca43、Si29、Na23、K39、Mg25、Cu63、Zn66、Fe57、Ni60、Mn55、Sr88、Ba137和Pb;秘鲁公海P31浓度最高,之后由高到低依次为Al27、Ca43、Si29、Na23、K39、Mg25、Zn66、Fe57、Cu63、Mn55、Sr88、Ni60、Pb和Ba137;智利公海P31浓度最高,之后由高到低依次为Ca43、Al27、Na23、Si29、K39、Mg25、Zn66、Fe57、Cu63、Mn55、Ni60、Sr88、Ba137和Pb。
厄瓜多尔、智利和秘鲁公海茎柔鱼眼睛晶体浓度前5的微量元素均为P31、Al27、Ca43、Si29、Na23,其中P31在3海区中浓度均为最高,其余4种元素在3海区中的浓度高低排序有所不同。ANOVA分析显示,Mg25、Ni60、Cu63、Sr88和Ba137等元素在3海区之间存在显著差异(p<0.05,表3),然而在这些显著差异的元素中至少在某两个海区之间无差异(p>0.05,表2);Na23、Al27、 Si29、P31、Ca43、Mn55、Zn66和Pb等元素在两两海区之间均不存在显著差异(p>0.05,表3)。
线性回归拟合分析显示,Sr88、Ba137、Fe57和Ni60浓度与SST呈显著的线性递减关系(p<0.05),除此之外,其他元素与SST均无显著的线性关系(图5),其关系式分别如下:
Sr88=−22.242 SST+570.87 (R2 = 0.234 9, p=0.009),
Ba137=−29.787 SST+658.92 (R2 = 0.312 5, p=0.002),
Fe57=−205.49 SST+5 181.8 (R2 = 0.190 3, p=0.020 3),
Ni60=−68.867 SST+1 608.7 (R2 = 0.271 5, p=0.004 5).
海洋生物硬组织微量元素的沉积是个复杂的过程,它受海水物理、化学特性,生物体年龄、生长、生理、代谢以及遗传等因素影响[28]。近年来,有关头足类耳石微量元素的报道比较多,研究认为,耳石微量元素的富集与头足类生活的水环境特性,如温度、盐度和食物等有关[13, 29]。本研究发现,茎柔鱼眼睛晶体外缘(捕捞死亡时)部分微量元素存在显著的地理区域差异,类似的研究在茎柔鱼[6]、蓝旗金枪鱼Thunnus orientalis[30]、巴塔哥尼亚犬牙鱼Dissostichus eleginoides[31]和太平洋鲱Clupea pallasii[32]的耳石中也有报道。因此,眼睛晶体中微量元素的变化可以用来指示海洋生物生活水域的物理、化学特性。
海洋生物硬组织中的微量元素被广泛用于重建其生活水温。Swart等[33]报道,环圆菊珊瑚Montastraea annularis骨格中的Sr/Ca与SST关系显著。Zacherl[34]研究发现,克莱特峨螺Kelletia Kelletii耳石中Sr和Ba与生活水温呈反比关系。Campana[35]认为,鱼类耳石的微量元素可用来重建其栖息水温。过去有相关研究是根据耳石中的微量元素浓度,重建了一些头足类的栖息水温[13, 14, 36-38]。本研究对茎柔鱼眼睛晶体外缘的微量元素与捕捞地点的SST进行了相关性分析,结果显示,Sr88、Ba137、Fe57和Ni60浓度与SST呈显著的线性递减关系(p<0.05),这为通过眼睛晶体的微量元素推测茎柔鱼的栖息水温提供了基础。
Sr元素被认为是头足类栖息水温重建的重要元素之一。Ikeda等[13, 36]研究发现,太平洋褶柔鱼Todarodes pacificus和水蛸Octopus dofleini耳石微量元素与栖息水温呈明显的负相关。Arkhipkin等[14]认为,巴塔哥尼亚枪乌贼 Doryteuthis gahi 耳石中的微量元素与历史综合水温呈反比例关系。日本海域长枪乌贼Loligo bleekeri耳石Sr/Ca明显高于安达曼海和泰国湾的热带枪乌贼[38]。本研究中,水温偏高的厄瓜多尔海域茎柔鱼眼睛晶体Sr浓度明显低于水温偏低的秘鲁和智利海域(表2),且线性回归显示,Sr88浓度与SST呈明显的负相关(图5p<0.05),同样的结果在茎柔鱼的耳石中也有报道[6]。然而也有报道[39]称,尽管厄尔尼诺年份水温比正常年份高2.5℃,但是茎柔鱼耳石中的微量元素却没有明显差异。
本研究发现,茎柔鱼眼睛晶体中Ba137浓度与SST呈显著的负相关关系,这在茎柔鱼[12]、巴塔哥尼亚枪乌贼[14]和黵乌贼Gonatus fabricii[37]的耳石也有报道。过去的研究认为,Ba137浓度不仅与水温呈负相关[40-41],而且可反映水体中营养盐的分布,即Ba浓度随着水深的增加而增大[42]。因此,Ba被看作是上升流以及头足类垂直移动的指示元素[14]。Arkhipkin等[14]认为,福克兰群岛大陆架水域巴塔哥尼亚枪乌贼耳石中Ba137浓度高与该海域强烈的上升流有关。然而,Liu等[6]在茎柔鱼耳石中却没有发现与此相匹配的Ba137地理区域间变化,这是因为该研究采用的是耳石整体微量元素,它是茎柔鱼整个生命周期内获取的Ba137浓度的均值。本研究眼睛晶体外缘的微量元素代表其捕捞时的所经历的环境信息,Ba137浓度能够反映其捕捞地点的环境信息。因此,水温偏低、上升流发达的智利海域,茎柔鱼眼睛晶体的Ba137浓度比厄瓜多尔和秘鲁公海明显偏高。研究还发现,茎柔鱼眼睛晶体Fe57和Ni60浓度与SST呈明显的线性负相关,这与在茎柔鱼耳石中的研究结果一致[6]
除此之外,其他元素浓度与SST无显著的相关关系。综合分析认为,Sr88、Ba137、Fe57和Ni60均可以看作茎柔鱼生活水温的指示元素,Ba137元素还可以看作茎柔鱼生活水深以及上升流的指示元素。眼睛晶体微量元素可用来重建头足类栖息环境。
