In this study, four samples of Gymnothorax mucifer were collected from the fishery market of Xiamen City, Fujian Province during the year 2020 to 2021, which were newly recorded in the coastal waters of China. Previously, the species had only been recorded in Australia and Hawaii, and was considered to be a synonym of Gymnothorax kidako. Detailed morphological characteristics of four G. mucifer species were analyzed, and the molecular identifications as well as phylogenetic constructions were also carried out basing on DNA barcode COI gene in this study. The main distinguishing characteristics of G. mucifer were as follow: the colour of the body was yellowish brown, the front of the head was slightly purple, the whole body was covered with slender, sparse, irregular branch-liked brown marking and the markings became darker and thicker near the tail, forming clear net-liked patterns; the margin on the anal fin was white, and became serial pale blotches on posterior part of the tail; total length was 1.01 times of standard length and 8.00−8.39 times of head length; the maxillary teeth were 8−10 and dentary teeth were 14−20 on each side, both teeth were uniserial; the median inter maxillary teeth were slender and uniserial; the total vertebrae were 117−139 and mean vertebral formula was 6-47-130. Basing on the COI gene analysis, the genetic distance between G. mucifer and G. kidako was 0.074, which was greater than the value 2% (0.020) suggested by Herbert as minimum genetic distance value to distinguishing different species, revealing that the two species might be two independent species. Morphologically, G. mucifer could also be distinguished from G. kidako by certain external characteristic: the body markings of G. mucifer were slender, sparse and inconspicuous, the front of the head was slightly purple, and the white margin of the anal fin broke into series of pale blotches on posterior part of the tail; the markings of G. kidako were obvious and thick with darker colour, the front of the head was yellow-white, the white margin on the anal fin continuous to the tip of the tail. The results of the study provided a taxonomic basis for the systematic classification and the species list revision of the Gymnothorax fish in our country.

本研究于2020−2021年在福建省厦门市近海水产市场采集到4尾黏裸胸鳝样品,为中国大陆近海新记录种。此前,该物种仅在澳大利亚、夏威夷等地有分布记录,并被认为是蠕纹裸胸鳝同种异名。本研究对采集的黏裸胸鳝进行详细形态特征分析,结合DNA条形码COI基因进行分子鉴定及系统进化关系分析。黏裸胸鳝的主要鉴别特征:体为黄棕色,头部前端淡紫色,身体布满细长、稀疏、弥散的树枝状不规则棕色斑纹,斑纹颜色靠近尾部加深加粗,形成网格状;臀鳍边缘白色,近尾部变为一列连续白色斑点;全长为体长的1.01倍,为头长的8.00~8.39倍;上颌齿每侧8~10个,下颌齿每侧14~20个,中央齿细长,均为单行;总椎骨数为117~139,平均椎骨式为6-47-130。基于COI基因分析,黏裸胸鳝与蠕纹裸胸鳝的遗传距离为0.074,大于Herbert设定的2%(0.020)作为区分不同物种最小遗传距离,表明两者应为不同的物种。形态上,两种裸胸鳝也存在差异特征,如黏裸胸鳝的斑纹较细长、稀疏、不明显,头部前端偏淡紫色,臀鳍白色边缘近尾部断裂成系列连续白色斑点;蠕纹裸胸鳝的斑纹粗大明显、颜色较深,头部前端黄白色,臀鳍白色边缘连续至尾尖。研究结果可为我国裸胸鳝属鱼类的系统分类及物种名录的修订提供分类基础。

本研究于2020−2021年在福建省厦门市近海水产市场采集到4尾黏裸胸鳝样品,为中国大陆近海新记录种。此前,该物种仅在澳大利亚、夏威夷等地有分布记录,并被认为是蠕纹裸胸鳝同种异名。本研究对采集的黏裸胸鳝进行详细形态特征分析,结合DNA条形码COI基因进行分子鉴定及系统进化关系分析。黏裸胸鳝的主要鉴别特征:体为黄棕色,头部前端淡紫色,身体布满细长、稀疏、弥散的树枝状不规则棕色斑纹,斑纹颜色靠近尾部加深加粗,形成网格状;臀鳍边缘白色,近尾部变为一列连续白色斑点;全长为体长的1.01倍,为头长的8.00~8.39倍;上颌齿每侧8~10个,下颌齿每侧14~20个,中央齿细长,均为单行;总椎骨数为117~139,平均椎骨式为6-47-130。基于COI基因分析,黏裸胸鳝与蠕纹裸胸鳝的遗传距离为0.074,大于Herbert设定的2%(0.020)作为区分不同物种最小遗传距离,表明两者应为不同的物种。形态上,两种裸胸鳝也存在差异特征,如黏裸胸鳝的斑纹较细长、稀疏、不明显,头部前端偏淡紫色,臀鳍白色边缘近尾部断裂成系列连续白色斑点;蠕纹裸胸鳝的斑纹粗大明显、颜色较深,头部前端黄白色,臀鳍白色边缘连续至尾尖。研究结果可为我国裸胸鳝属鱼类的系统分类及物种名录的修订提供分类基础。

In this study, four samples of Gymnothorax mucifer were collected from the fishery market of Xiamen City, Fujian Province during the year 2020 to 2021, which were newly recorded in the coastal waters of China. Previously, the species had only been recorded in Australia and Hawaii, and was considered to be a synonym of Gymnothorax kidako. Detailed morphological characteristics of four G. mucifer species were analyzed, and the molecular identifications as well as phylogenetic constructions were also carried out basing on DNA barcode COI gene in this study. The main distinguishing characteristics of G. mucifer were as follow: the colour of the body was yellowish brown, the front of the head was slightly purple, the whole body was covered with slender, sparse, irregular branch-liked brown marking and the markings became darker and thicker near the tail, forming clear net-liked patterns; the margin on the anal fin was white, and became serial pale blotches on posterior part of the tail; total length was 1.01 times of standard length and 8.00−8.39 times of head length; the maxillary teeth were 8−10 and dentary teeth were 14−20 on each side, both teeth were uniserial; the median inter maxillary teeth were slender and uniserial; the total vertebrae were 117−139 and mean vertebral formula was 6-47-130. Basing on the COI gene analysis, the genetic distance between G. mucifer and G. kidako was 0.074, which was greater than the value 2% (0.020) suggested by Herbert as minimum genetic distance value to distinguishing different species, revealing that the two species might be two independent species. Morphologically, G. mucifer could also be distinguished from G. kidako by certain external characteristic: the body markings of G. mucifer were slender, sparse and inconspicuous, the front of the head was slightly purple, and the white margin of the anal fin broke into series of pale blotches on posterior part of the tail; the markings of G. kidako were obvious and thick with darker colour, the front of the head was yellow-white, the white margin on the anal fin continuous to the tip of the tail. The results of the study provided a taxonomic basis for the systematic classification and the species list revision of the Gymnothorax fish in our country.

裸胸鳝属(Gymnothorax)隶属于硬骨鱼纲(Osteichthyes),鳗鲡总目(Anguillomorpha),鳗鲡目(Anguilliformes),海鳝科(Muraenidae),海鳝亚科(Muraeninae),广泛分布于热带、亚热带及温带海区。裸胸鳝属是海鳝科里面种类最多的一个属,世界上记录有100余种[1-3],我国有20~30种[4-16],不同分类资料收录的种类数目有所不同。其中《中国鱼类系统检索》[4]收录20种、《台湾鱼类志》[6]收录27种、《中国海洋鱼类》[12]与《中国海洋生物名录》[15]收录种类最多,均为37种。主要分布于东海、南海及台湾海峡。
目前,国内有关裸胸鳝属鱼类研究国内报道极少,仅在分子系统发育[17-18]、消化道结构[19]及遗传多样性[20]等方面有少量研究。国外报道相对较多,研究主要集中于裸胸鳝基础生物学及种质资源方面,近些年在裸胸鳝种质分析上有不少新种发现的报道,如Gymnothorax pharaonis[3]Gymnothorax smithi[21]Gymnothorax pseudotile[22]Gymnothorax visakhaensis[23]Gymnothorax pseudome-lanosomatus[24]等。自2015年以来已有5种裸胸鳝属新种被记录,揭示了裸胸鳝属资源还存在巨大的可挖掘空间,物种分类信息有待进一步更新与完善。
黏裸胸鳝(Gymnothorax mucifer)(Snyder, 1904)体色为棕黄色,体表布满细小、弥散的树枝状不规则斑纹,臀鳍边缘为白色。目前我国的分类资料均无黏裸胸鳝的信息,包括比较系统、权威的物种检索资料,如《中国鱼类系统检索》《中国海洋鱼类》《台湾鱼类志》等。国外针对黏裸胸鳝的研究也很少,并且其物种有效性也存在一定争议。有研究认为,黏裸胸鳝其实为蠕纹裸胸鳝(Gymnothorax kidako)的同种异名[25-27],但近些年也有研究揭示两者为独立的物种,两种裸胸鳝在下颌特征、牙齿及脊椎骨数量等形态性状方面存在差异[28-29]。本研究于2020−2021年期间在福建省厦门市近海水产市场采集了4尾黏裸胸鳝的样品,为我国大陆近海一个新记录种。针对其具体的形态特征进行详细测量分析,同时采集蠕纹裸胸鳝、雪花斑裸胸鳝(Gymnothorax niphostigmus)等多种与黏裸胸鳝形态相近的裸胸鳝种类进行比较,结合DNA条形码COI基因开展分子系统分类研究,明确黏裸胸鳝的物种有效性,为我国裸胸鳝属鱼类系统分类及物种名录的修订提供分类基础。
黏裸胸鳝样品于2020−2021年在福建省厦门市近海水产市场(30.060 7°N, 122.354 6°E)采集,共获得样品4尾。另外采集了蠕纹裸胸鳝、雪花斑裸胸鳝、埃玛氏裸胸鳝(Gymnothorax emmae)、云纹裸胸鳝(Gymnothorax chilospilus)、白斑裸胸鳝(Gymnothorax prionodon)等与黏裸胸鳝形态相似的种类比较分析。采集样品鲜活状态下进行高清拍摄,标本于仲恺农业工程学院动物科技学院水产养殖系实验室保存(图1图2)。
本研究参考张春光[11]、Böhlke和Randall[28]的测量方法,对采集的4尾黏裸胸鳝样品的形态特征进行描述。利用直尺及游标卡尺准确测量样品全长、体长、体高、肛前体长、肛后体长、头长、头高、吻长、上颌长、下颌长、眼径、眼间距、眼后头长、背鳍前距、臀鳍前距、背鳍基长、臀鳍基长等可量性状,精确到1 mm;采用直接计数方式统计前颌齿、上颌齿、下颌齿、犁骨齿、前颌骨中央齿的数目以及脊椎骨数、背鳍鳍条数、臀鳍鳍条数等可数性状(图3图4)。
(1)DNA提取
取约20 mg样品肌肉组织,使用海洋动物组织基因组DNA提取试剂盒(天根生化科技有限公司),按照说明书进行DNA提取。提取4尾黏裸胸鳝以及蠕纹裸胸鳝、雪花斑裸胸鳝、埃玛氏裸胸鳝、云纹裸胸鳝、白斑裸胸鳝等总基因组DNA,溶解于80 µl灭菌水中。利用1%琼脂糖凝胶对提取的DNA进行电泳检测,−20℃冰箱保存备用。
(2)PCR扩增与测序
PCR扩增引物采用鱼类COI基因序列的通用引物,为COI-F1: 5′-TCAACYAATCAYAAAGATATYGGCAC-3′、COI-R1: 5′-ACTTCYGGGTGRCCRAARAATCA-3′[30]。PCR反应体系为25 µL,包括PCR Mastermix 12.5 µL,灭菌水10.5 µL,上、下游引物各为0.5 µL,DNA模板1 µL。扩增条件为94℃预变性5 min、94℃变性30 s、55℃退火30 s、72℃延伸1 min,35个循环,最后72℃再延伸7 min。PCR产物用1%琼脂糖凝胶电泳检测,选取扩增成功的样品用PCR产物纯化试剂盒纯化回收后送至天一辉远生物技术有限公司进行测序。
(3)分子系统进化分析
将测得的COI基因片段于美国国家生物技术信息中心(National Center for Biotechnology Information, NCBI)进行相似性检索,验证序列的正确性。同时,在GenBank上下载所有学名为Gymnothorax mucifer及其他相关裸胸鳝的COI序列一起分析(表1),利用Clustal W软件对序列进行排列比对[31],去掉两端不规则冗余序列,保留同源序列于MEGA 7.0软件分析碱基组成、遗传距离等信息[32]。以裸海鳝属斑马裸海鳝(Gymnomuraena zebra)为外类群,采用最大似然法与贝叶斯法构建分子系统发育树,最大似然树在RAxML 8.0 软件完成[33],分支置信度利用1 000次重复抽样分析。贝叶斯树在 MrBayes 3.1.2 软件中完成[34],随机选取起始树,计算 1 000 000 代,取样代数为100,节点置信度由后验概率提供。
4尾黏裸胸鳝样品于2020−2021年采自福建省厦门市近海水产市场,编号为GMFJXM1−GMFJXM4,形态测量数据见表2。4尾样品全长范围为515~617 mm,全长为体高(42~56 mm)的10.71~12.79倍,为背鳍前距(48~58 mm)的9.42~10.91倍,为臀鳍前距(245~286 mm)的2.10~2.16倍,为头长(62~75 mm)的8.00~8.39倍。头长范围为上颌长(34~40 mm)的1.82~2.18倍,为下颌长(37~45 mm)的1.67~1.85倍,为眼径(5~7 mm)的10.57~12.40倍,为眼间距(6~11 mm)的6.73~10.67倍。
黏裸胸鳝体为黄棕色,吻部偏紫色。体表布满细小、弥散的树枝状不规则棕色横纹,相连成网状;纹路在尾部加深加粗,使尾部呈现网格状白斑。口角具4~6条纵向黑痕。臀边缘为白色,不延续至尾端,尾部被棕色网纹分隔呈现一列白斑。平均椎骨式为6-47-130。
黏裸胸鳝牙齿尖细呈锥状,前颌齿1行,每侧有4~8个大犬齿。上颌齿1行,齿较前颌齿小且向后倾斜,每侧8~10个;其中有2尾上颌内部每侧均具有4个细小尖形牙。中央齿细长且单行,数量2~4个不等;犁骨齿较小,单行或交错排列,为3~9个。下颌齿1行,每侧有14~20个,前4~5对较大,其余牙齿较小且大小不等。
4尾黏裸胸鳝样品背鳍鳍条数为293~408,臀鳍鳍条数为213~286。总脊椎骨数为117~139,背前脊椎骨数为4~8,肛门前脊椎骨数为43~56,平均椎骨式为6-47-130;椎体呈双凹形,每一椎体的中心均有小孔。
体中等长,头中等大;躯干部呈圆柱形,尾部较侧扁,完全裸露;吻短;前鼻孔位于近吻端,呈管状,中等细长;后鼻孔位于眼前缘上方,圆形;眼中等大,新鲜标本的眼睛虹膜微黄,眼间距稍大于眼径;上下颌基本等长;体为黄棕色至深棕色,头部前端偏紫色,喉部、腹部颜色较浅;眼后头部网纹密集,部分形成细小、淡黄色斑点,斑点分布因物种个体而异;下颌处无斑纹;身体布满细小、稀疏、树枝状不规则褐色横纹,连成网状;纹路在尾部颜色加深加粗,相连围绕,使尾部呈现明显网格状白斑;口角具4~6条纵向黑痕;鳃孔周围颜色呈黑色,较明显;背鳍、臀鳍均与尾鳍相连,无胸鳍;背鳍始于鳃孔前,中等高;肛门位于体中央前方,臀鳍较浅,不如背鳍发达;臀鳍边缘为白色且颜色不连续至尾尖,尾部被棕色纹路分割呈现一列白色斑点。
根据《中国鱼类系统检索》[4]、《中国动物志》[11]、《中国海洋鱼类》[12]、《台湾鱼类志》[6]等形态分类资料,结合在线鱼类数据库[2],汇总出黏裸胸鳝及与其形态相似的5种裸胸鳝可区分彼此的外部特征性状,对黏裸胸鳝及相关裸胸鳝进行比较分析(表3)。
测序获得本研究4尾黏裸胸鳝及蠕纹裸胸鳝、雪花斑裸胸鳝、云纹裸胸鳝、埃玛氏裸胸鳝和白斑裸胸鳝的COI基因序列,结合GenBank下载的裸胸鳝COI基因序列(表1)共同分析,经排列比对,去除两端冗余获得一致序列636 bp。序列中T、C、A、G碱基含量分别为29.6%、27.5%、23.7%、19.2%,A+T含量(53.3%)高于C+G(46.7%)。序列中保守位点482个,变异位点154个,简约信息位点138个,转换颠换比为4.24。基于Kimura-2-parameter模型计算各裸胸鳝种内及种间遗传距离(表4)。6种裸胸鳝种内遗传距离范围为0.006~0.019,平均遗传距离为0.009。种间遗传距离为0.051~0.164,平均遗传距离为0.111。比较黏裸胸鳝与其余5种裸胸鳝遗传距离可知,黏裸胸鳝与雪花斑裸胸鳝之间遗传距离最小(0.051),与云纹裸胸鳝遗传距离最大(0.146)。以斑马裸海鳝为外群,采用最大似然法和叶贝斯法构建的分子系统进化树如图5图6所示,两种方法构建的进化树拓扑结构基本相同,节点均拥有较高支持率。进化树上,同种裸胸鳝不同个体均能紧密聚在一起,形成种内单系分支。本研究的4尾黏裸胸鳝与GenBank中的黏裸胸鳝9条序列一起,形成黏裸胸鳝的单系分支,位于进化树顶部。对于黏裸胸鳝与其他几种裸胸鳝分类关系,黏裸胸鳝与雪花斑裸胸鳝关系最近,两者聚为紧密姐妹分支,随后该分支再与蠕纹裸胸鳝相聚。云纹裸胸鳝与黏裸胸鳝的分类地位最远,位于进化树的基部,其次分别为埃玛氏裸胸鳝与白斑裸胸鳝。
我国记录的裸胸鳝属鱼类有20~30种[4-16],早期1962年编著的《南海鱼类志》[8]收录有18种,后续分类资料收录物种信息逐渐增多,《中国鱼类系统检索》[4]记录20种,《台湾鱼类志》[6]记录27种,数量最多的为2016年陈大刚与张美昭主编的《中国海洋鱼类》[12]及2008年刘瑞玉主编的《中国海洋生物名录》[15],均为37种。而在目前的形态分类资料中,均无黏裸胸鳝(Gymnothorax mucifer )(Snyder, 1904)的物种记录,甚至不曾出现过这一学名。国外关于黏裸胸鳝的研究报道也较少,其为十分罕见的裸胸鳝种类。前期仅在澳大利亚、新喀里多尼亚及夏威夷群岛等海域有黏裸胸鳝记录,但曾一度被误认为是蠕纹裸胸鳝[25-27],直至Böhlke与Randall[28]提出两者为不同物种。Huang等[29]2019年在台湾东北部太平洋海区附近也曾采集过黏裸胸鳝样品,并比较黏裸胸鳝、蠕纹裸胸鳝的特征差异,认为两者应为独立的物种。本研究4尾黏裸胸鳝采集于福建省厦门市近海水产市场,形态特征分析跟Böhlke与Randall[28]、Huang等[29]关于黏裸胸鳝的物种特征描述及图片信息基本一致。本研究中4尾黏裸胸鳝为在我国大陆近海(渤海、黄海、东海、南海)首次记录的种类。针对该物种进行具体形态学及分子DNA条形码鉴定,验证为黏裸胸鳝,可知我国沿海也有该裸胸鳝物种的分布,为我国裸胸鳝属鱼类物种名录修订及分类学研究提供了依据。
黏裸胸鳝体色为黄棕色,体表布满较为弥散的树枝状不规则横纹,臀鳍边缘白色,与蠕纹裸胸鳝的形态十分相似。前期部分形态学研究认为两者为同一物种[25-27]。世界鱼类数据库(Fishebase)目前也将黏裸胸鳝标注为蠕纹裸胸鳝的同种异名[2]。而比较分析黏裸胸鳝与蠕纹裸胸鳝详细形态学特征发现,两者体表也存在较明显的差异。蠕纹裸胸鳝头部黄色,身体环绕较粗大、明显的树枝状多突起不规则褐色横纹,横纹特征在尾部变密,相连围绕,将尾部分隔成许多不规则多突起的白斑,臀鳍白色边缘延续至尾端;黏裸胸鳝头部淡紫色,身体环绕较细小稀疏、弥散的树枝状不规则横纹,横纹在尾部加深加粗,相连围绕,将尾部分隔成网格状白斑,臀鳍白色边缘不延续至尾端。
在DNA条形码COI基因分析方面,本研究中4种黏裸胸鳝COI序列经NCBI在线比对,与在线公布的G. mucifer的同源性最高,达到99%以上,分子水平验证为黏裸胸鳝G. mucifer。通过与蠕纹裸胸鳝、雪花斑裸胸鳝等5种与黏裸胸鳝形态接近的裸胸鳝COI序列比较分析可知,黏裸胸鳝与其他5种裸胸鳝的遗传距离范围为0.051~0.146,其中与雪花斑裸胸鳝遗传距离最小(0.051),与云纹裸胸鳝的遗传距离最大(0.146),均远大于Hebert等[36]推荐的2%(0.020)作为区分不同物种的最小遗传距离,同时远大于本研究6种裸胸鳝种内不同个体的遗传距离范围(0.006~0.019)及目前有关裸胸鳝属鱼类COI基因种内遗传距离变异范围(最小为0,最大为0.037,大部分在0.020以内)[37-39]。可知分子水平上,黏裸胸鳝为区分于其他5种裸胸鳝的独立物种。与黏裸胸鳝形态最为接近且处于同种异名争议的蠕纹裸胸鳝,两者遗传距离高达0.074,在分子水平上存在较大差异。进化树上,黏裸胸鳝优先与雪花斑裸胸鳝聚为一支,该分支再与蠕纹裸胸鳝相聚。可知,相对于雪花斑裸胸鳝,蠕纹裸胸鳝与黏裸胸鳝分子水平的亲缘关系相对较远,结合形态学的特征比较分析可知,两种裸胸鳝应为独立的物种,并非同种异名。
