Diaphus is one of the genera with the highest species abundance and the most abundant population in the family Myctophidae, and it is also the dominant group of deep sea fishes in the ocean. However, due to the high similarity of external morphology, it is difficult to identify the lanternfish species. In order to explore the feasibility of otoliths morphological analysis in identifying the otolith differences between Diaphus species with similar otoliths and large overlapping distribution areas, the otoliths of three Diaphus species were collected from the Indian Ocean and the South China Sea for morphometrics analysis with landmark method. The results showed that the morphological variation of sagittal otoliths in three lanternfishes were mainly in the internal main sulcus, and the internal punctuation contribution rate reached 62%. The grid difference visualization showed that the wing length of sagittal otoliths in D. thiollierei was longer than that of the other two species, while the width was slightly narrower. The Bayes function was used for classification and discrimination, and the results showed that the success rate of D. thiollierei was 100%, while the other two species had a few misjudgments. In addition, taking into account the habitat environment and the genetic relationship between these three species, we supposed that the main reason for the morphological differences in the main sulcus of the three spescies was the huge salinity difference in the habitat sea area. However, the impact and physiological mechanism of salinity on the morphology of the main sulcus of sagittal otoliths needs to be further studied.

眶灯鱼属(Diaphus)是灯笼鱼科中物种丰度最高,种群数量最丰富的属之一,也是大洋中深层鱼类中的优势类群,但由于外部形态的高度相似,目前对眶灯鱼属的种类鉴定存在较多困难。为探究耳石地标点形态分析法(简称地标点法)在识别耳石形态相似、分布海域有较大重叠的眶灯鱼种间矢耳石差异的可行性,本文采集了分布于印度洋的西氏眶灯鱼(Diaphus thiollierei)和分布于南海的喀什眶灯鱼(D. garmani)和马来眶灯鱼(D. malayanus)的耳石应用地标点法进行形态分析。分析结果显示,3种眶灯鱼的矢耳石形态差异主要集中于内部听沟,内部标点贡献率达62%。网格差异可视化表明,西氏眶灯鱼矢耳石翼叶长度长于其他两种,而宽度则略窄。利用贝叶斯函数进行分类判别,结果表明西氏眶灯鱼判别成功率达到100%,而其余两种眶灯鱼则存在少量误判。此外,本研究结合样本栖息环境差异及相互间的亲缘关系,发现造成3种眶灯鱼矢耳石听沟形态差异的主要原因可能来源于所栖息海域的巨大盐度差异,但盐度影响矢耳石听沟形态的内在生理机制还需进一步研究。

眶灯鱼属(Diaphus)是灯笼鱼科中物种丰度最高,种群数量最丰富的属之一,也是大洋中深层鱼类中的优势类群,但由于外部形态的高度相似,目前对眶灯鱼属的种类鉴定存在较多困难。为探究耳石地标点形态分析法(简称地标点法)在识别耳石形态相似、分布海域有较大重叠的眶灯鱼种间矢耳石差异的可行性,本文采集了分布于印度洋的西氏眶灯鱼(Diaphus thiollierei)和分布于南海的喀什眶灯鱼(D. garmani)和马来眶灯鱼(D. malayanus)的耳石应用地标点法进行形态分析。分析结果显示,3种眶灯鱼的矢耳石形态差异主要集中于内部听沟,内部标点贡献率达62%。网格差异可视化表明,西氏眶灯鱼矢耳石翼叶长度长于其他两种,而宽度则略窄。利用贝叶斯函数进行分类判别,结果表明西氏眶灯鱼判别成功率达到100%,而其余两种眶灯鱼则存在少量误判。此外,本研究结合样本栖息环境差异及相互间的亲缘关系,发现造成3种眶灯鱼矢耳石听沟形态差异的主要原因可能来源于所栖息海域的巨大盐度差异,但盐度影响矢耳石听沟形态的内在生理机制还需进一步研究。

Diaphus is one of the genera with the highest species abundance and the most abundant population in the family Myctophidae, and it is also the dominant group of deep sea fishes in the ocean. However, due to the high similarity of external morphology, it is difficult to identify the lanternfish species. In order to explore the feasibility of otoliths morphological analysis in identifying the otolith differences between Diaphus species with similar otoliths and large overlapping distribution areas, the otoliths of three Diaphus species were collected from the Indian Ocean and the South China Sea for morphometrics analysis with landmark method. The results showed that the morphological variation of sagittal otoliths in three lanternfishes were mainly in the internal main sulcus, and the internal punctuation contribution rate reached 62%. The grid difference visualization showed that the wing length of sagittal otoliths in D. thiollierei was longer than that of the other two species, while the width was slightly narrower. The Bayes function was used for classification and discrimination, and the results showed that the success rate of D. thiollierei was 100%, while the other two species had a few misjudgments. In addition, taking into account the habitat environment and the genetic relationship between these three species, we supposed that the main reason for the morphological differences in the main sulcus of the three spescies was the huge salinity difference in the habitat sea area. However, the impact and physiological mechanism of salinity on the morphology of the main sulcus of sagittal otoliths needs to be further studied.

眶灯鱼类隶属于灯笼鱼目(Myctophiformes)灯笼鱼科(Myctophidae),通常生活在大洋200~1000 m深度,鱼体呈长梭形,嘴大且口裂长。眶灯鱼属(Diaphus)是灯笼鱼科中物种丰度最高,种群数量最丰富的属之一,因头部具有突出明显的次级发光器,也被称为头灯鱼[1]。目前对于眶灯鱼属鱼类的分类鉴定工作存在许多困难,主要有以下几个原因:首先属内存在许多近缘种,部分近缘种在分布上还存在较大重叠,这些外部形态高度近似的近缘种在外观上很难进行有效分辨,其次,发光器是眶灯鱼种类鉴定的主要依据之一,但由于灯笼鱼一般属于兼捕物种,在捕捞过程鱼体容易受到不同程度的损坏导致发光器不完整从而影响鉴定结果的准确性;最后,现阶段针对难以辨别的灯笼鱼物种往往利用传统鱼类形态学和新兴的分子鉴定手段相结合的方式,但由于眶灯鱼种类繁多,而当前用于序列比对的参考数据库并不完善[2-4]
在鱼类分类学中,矢耳石早已被证实具有高度的物种特异性。近年来,随着耳石形态学研究的兴起与相关分析技术的成熟,耳石形态学开始广泛用于鱼类种群鉴定[5-7]。但即便如此,也主要应用于耳石外观上形态差异较大的物种类群,如Battaglia等[8]利用传统的形态学分析手段分析鉴别灯笼鱼科里的几种灯笼鱼矢耳石;欧利国和刘必林[9]利用地标点法识别4种耳石差异较大的鲹科(Carangidae)鱼类;Fortunato等[10]则通过识别微耳石形态差异对8种鲻科(Mugilidae)鱼类进行分类。灯笼鱼同样可通过耳石形态进行种间的鉴别[11],但由于耳石形成不仅受到内在遗传因素的影响[12],同样也会受到所处海域的温度、盐度以及物种生活习性等外在环境因素的影响[13-14],因而,目前传统耳石形态学方法针对近缘并栖息于相同或相近海域环境的灯笼鱼种类并未能取得较好的判别效果[15-16]
地标点法是目前较为新兴的耳石形态分析方法,通过获取二维影像中各个特征点的坐标数据,处理后进行耳石形态变异程度分析和判别分析,从而直观显示不同类群鱼类耳石的形态差异[11, 17],此外所需样本量较小也是地标点的一个优势,相较于其他形态学分析方法需要大量数据进行模拟推演,地标点法可有效把控研究对象的形态差异,即使是在样本量较小的情况下也能较为准确地实现种间判别[18-19]。耳石的地标点法在深海鱼类中的应用较少,本次研究对象西氏眶灯鱼(Diaphus thiollierei)、喀什眶灯鱼(D. garmani)及马来眶灯鱼(D. malayanus)三者亲缘关系较近,同时分布纬度一致,后两者生活海域存在较大重叠。因此本文旨在研究利用地标点法识别该类眶灯鱼矢耳石差异的可行性,并对造成耳石形态差异的主要原因进行初步分析,研究结果将为解决某些渔业资源上常见的灯笼鱼物种鉴定难题提供参考。
西氏眶灯鱼采样调查船为顺来发海洋渔业有限公司渔船“福远渔637”,采样工具为灯光敷网,网目规格为35 mm。调查海域为印度洋−阿拉伯海海域(ARA,图1a),调查时间为2020年10−11月。共收集西氏眶灯鱼样本150尾。
喀什眶灯鱼和马来眶灯鱼采样调查船为北海市外海南沙渔业有限公司调查船“桂北渔80208”,采样工具为灯光罩网,网目规格为50 mm,调查海域为南海海域(SCS,图1b),调查时间为2019年6−7月。共收集马来眶灯鱼样本22尾,喀什眶灯鱼36尾。
样本收集后,立即放入酒精中保存,将样品带回实验室后进行基础生物学数据测量,包括标准体长(SL)和体质量(DW)。体长测量工具为游标卡尺(精度为0.1 mm),体质量测量工具为电子天平(精度为0.1 g),并对鱼体进行解剖,辨别雌雄。矢耳石取样使用提前灭菌的镊子和解剖刀,取出矢耳石在解剖镜下洗净,使用去离子水和75%酒精清洗矢耳石。干燥后用精确度为0.000 1 g的电子天平测量左右矢耳石质量,并放置于48孔板记录好编号,通过对西氏眶灯鱼样品日龄鉴定(未发表数据,样品日龄范围113~150 d),并结合Sassa等[20]和Ohshimo等[21]对于眶灯鱼的不同阶段鱼体形态的描述及生物学数据的测量,挑选成年眶灯鱼个体进行后续实验。最终总计获得西氏眶灯鱼矢耳石样本140对,马来眶灯鱼矢耳石样本22对,喀什眶灯鱼矢耳石样本35对。根据地标点软件的使用规则,最终选取西氏眶灯鱼矢耳石样本44对,体长范围为41.2~50.4 mm,体质量范围为1.21~1.64 g,平均体长和体质量分别为45.1 mm和1.51 g;马来眶灯鱼矢耳石样本22对,体长范围为25.4~36.2 mm,体质量范围为1.10~1.34 g,平均体长和体质量为28.3 mm和1.22 g;喀什眶灯鱼样本22对,体长范围为22.6~31.3 mm,体质量范围为1.00~1.29 g,平均体长和体质量分别为26.6 mm和1.20 g。
将干燥后的矢耳石取出,统一选取左矢耳石,将其听沟朝上,矢耳石最长轴线与水平基线平行,而后进行拍照,拍照工具为电子显微镜(QX800HD720P3D,ASONE株式会社,日本)
本研究利用地标点软件Tpsdig软件选取共11个具有结构代表性的点,并建立相应的坐标体系。11个点分别为轮廓上的端点(点1、2、3、4、5、6)以及矢耳石内部听沟纹路拐点(点7、8、9、10)和1个听沟内部纹路交叉点(点11),其中点2、3和4分别为上缘、下缘以及后端最突出的点(图2);利用Tpssmall软件对获得的坐标数据文件进行最小平方和法则计算,检验选取的地标点的合理性;利用Tpsrelw软件对地标点数据进行叠加、放大、扭曲等操作,最终得出3种矢耳石平均形,同时得出相对扭曲得分并记录于EXCEL表格;最后利用Tpsregr软件进行分析,通过放大差异等操作得出3种眶灯鱼矢耳石网格图,并进行差异比较。
(1)观察、描述并记录矢耳石形态特征。
(2)排除放大倍数、矢耳石方向和朝向等外界因素影响,在显微镜下拍照,之后进行地标点选取,最后利用EXCEL和SPSS26.0等软件对3种眶灯鱼相对扭曲得分进行判别分析。
(3)从世界海洋图集(World Ocean Atlas,WOA)网站下载采样区域的温度和盐度数据,其中阿拉伯海数据的空间维度为14.5°~19.5°N,60.5°~69.5°E,南海数据的空间维度为10.5°~15.5°N,110.5°~119.5°E。且两个区域的时间维度都截取WOA上的多年探测的温盐平均数值,时间跨度为1955−2020年。采取此种数据处理方式主要目的是在减少误差的同时,通过多年平均数值来反映本实验采样时间的大洋温盐变化趋势。并利用MATLAB R2019b做出温度和盐度随深度的变化曲线(图3图4),用以分析采样区域温盐差异。
西氏眶灯鱼、马来眶灯鱼和喀什眶灯鱼的矢耳石在形态上无显著差别,均趋近于椭圆形,且一些特征结构如主间沟、听沟等结构都十分相似,肉眼上无法进行有效分辨(图2)。就体积大小而言,3种眶灯鱼的矢耳石均略大于底灯鱼等其他灯笼鱼科的种类,3种眶灯鱼矢耳石的主间沟和听沟都较为明显,且听沟内部纹路交叉位置都有一块凸起结构。矢耳石听沟朝上的一面较为粗糙,均有一些细微纹路分布。整体上看,表面两条凹槽均以主间沟端点和翼叶端点为起点,向矢耳石中心延伸并直至矢耳石后端,但与矢耳石后端之间的交点界限较不清晰,此外3类矢耳石的听沟内部均有纹路交叉点存在。
最小回归平方和分析结果显示,3种眶灯鱼的回归系数为0.997 8,表明所选取的11个地标点均合理且有效。进一步记录各个地标点相对扭曲贡献率,发现外部轮廓的基叶端点5和主间沟端点6的贡献率分别为14.1%和21.8%,贡献率最高的点为听沟之间的交叉点11,贡献率达到30.5%。外部轮廓6个点贡献率总和为38.2%,内部5个点贡献率总和达61.8%(表1),由此可证明本研究在对3种眶灯鱼矢耳石形态差异辨别上内部听沟上的点作用较大,矢耳石形态差异主要集中在听沟的两个拐点(点7、点10)以及听沟中间的交叉点(点11)。
为了更直观的表示3种眶灯鱼矢耳石形态上的差异,首先利用Tpsregr获得3种矢耳石形态平均形,之后对结果进行绝对扭曲等处理,并将差异结果放大3倍,进行网格化和差异矢量化处理。
喀什眶灯鱼和马来眶灯鱼的矢耳石内部听沟轮廓点(7、8、9、10)相对于西氏眶灯鱼而言更向左上方偏移,且喀什眶灯鱼偏移的幅度会略大于马来眶灯鱼,矢耳石上表现为喀什眶灯鱼和马来眶灯鱼的听沟整体上处于矢耳石的左上方位置。马来眶灯鱼和喀什眶灯鱼翼叶端点(5)和主间沟端点(6)之间的距离较短,而西氏眶灯鱼两点之间的距离较长,矢耳石形态上表现为西氏眶灯鱼翼叶长度长于另外两种。此外马来眶灯鱼和喀什眶灯鱼的矢耳石上下端顶点(2、4)相比于西氏眶灯鱼有外扩的趋势,表现为西氏眶灯鱼的矢耳石宽度会略窄于其余两种(图5)。
通过SPSS26.0对18个相对扭曲得分建立贝叶斯判别函数,并作进一步交互验证,所得数据绘制成表格(表2)和判别分析散点图(图6)。经判别分析,3种眶灯鱼共18个矢耳石形态相对扭曲主成分中,有9个主成分被纳入函数判别式,分别是X1、X3、X4、X5、X7、X8、X9、X11和X14。3种眶灯鱼(喀什眶灯鱼G、马来眶灯鱼M和西氏眶灯鱼T)判别分类函数分别为

在预测结果中西氏眶灯鱼和马来眶灯鱼以及喀什眶灯鱼之间的区分较为明显,判别成功率达到100%,而马来眶灯鱼和喀什眶灯鱼之间存在误判个体,判别成功率为86.36%。而在交互验证的结果中显示的判别成功率与预测结果相似,仍旧是西氏眶灯鱼判别成功率最高,为97.73%,马来眶灯鱼和喀什眶灯鱼的判别成功率为81.82%(表2)。利用扭曲得分所建立的判别分析结果显示三者之间组质心有较大的差别,且西氏眶灯鱼与另外两种眶灯鱼的组质心距离会较远,表明差异更大(图6)。
由于矢耳石的生长代谢惰性以及高度物种特异性,其已经成为当前鱼类分类学与生活史研究的重点对象,尤其是在利用矢耳石进行物种鉴定和种群识别方面[22-27]。随着近些年图像处理技术的成熟与应用,矢耳石形态学分析也取得迅速发展,即便用于分析的不同种类矢耳石差异十分细微,但通过网格差异可视化能将肉眼看不见的差异扩大3~10倍,从而提高判别成功率。本文利用较为新兴的地标点分析法对3种不同眶灯鱼进行矢耳石形态学差异分析,结果表明三者之间的判别成功率达到86.36%以上,其中西氏眶灯鱼的判别成功率达到100%,仅有同样采自南海的喀什眶灯鱼和马来眶灯鱼群体之间存在少量的误判,在后续交互验证中也显示了相似的结果,即三者都具有较高判别率的同时,西氏眶灯鱼判别成功率要高于另外两种。在地标点应用于矢耳石差异判别的相关研究表明[827-30],判别成功率低于60%表明模型判别效果不佳,难以准确进行识别;60%~80%表明判别效果一般,能识别大部分差异;80%~100%表明判别效果良好,所拟合的模型能够有效识别差异并作出准确判别。本研究对3种眶灯鱼判别成功率为86.1%~100%,表明地标点在识别矢耳石形态差异较小且分布海域相似的不同种眶灯鱼是可行的。但前提是用于分析的标点必须能较好表达矢耳石形态特异性,本文所选取的11个地标点所计算得出的贡献率表明,所选取的点6和点11的贡献率之和达到52.3%,对3种眶灯鱼矢耳石识别起了显著作用,外部轮廓6个点贡献率总和为38.2%,内部5个点贡献率总和达61.8%,虽然内部标点贡献率要高于外部标点,但并无法否认外部标点在整体识别过程中所起的作用。根据本研究结果并结合前人的研究经验[30-31],我们认为在判别同属且相似度较高的矢耳石中,地标点的采用需遵循两个原则:(1)选取的地标点应具有结构代表性,能较好地反映矢耳石形态特异性;(2)地标点的选取应采取整体形态和听沟形态相结合的方式进行整体分析。
目前一些学者有过关于眶灯鱼属矢耳石的简单描述。如Battaglia等[8]在研究灯笼鱼矢耳石形态学参数与体长之间关系的文章中有过对大眼眶灯鱼(D. holti)矢耳石形态的描述,但主要集中于不同属之间的矢耳石比较,并未对相似度较高的同属近缘物种的矢耳石进行相应的讨论,本研究结果将为眶灯鱼属的同一矢耳石类型形态学提供可供参考的研究数据。
从外部轮廓而言,3种眶灯鱼矢耳石在形态上无较大的差异,呈椭圆形。外部轮廓上三者均有基叶、翼叶和主间沟等结构,基叶长于翼叶且较为发达,二者容易分辨。从网格差异可视化中可发现,3类矢耳石外部轮廓上的差异主要集中在点1、点5和点6,即矢耳石基叶、翼叶和听沟的端点。从内部轮廓而言,听沟已经被证实可以用于物种识别,是矢耳石形态差异的一个重要特征。朱元鼎[32],成庆泰和郑葆珊[33]早在20世纪就将石首鱼(Sciaenidae)矢耳石的听沟作为重要的分类依据,之后随着相关技术的发展与应用,实现矢耳石内部差异可视化,使内部听沟等结构能够更加形象体现差异产生的趋向[34-35]。迄今已有多项研究支持耳石听沟可用于鱼类物种鉴定的结论[22-26, 36-40]。本文研究对象喀什眶灯鱼、马来眶灯鱼和西氏眶灯鱼内部轮廓上均有听沟等结构,代表听沟的5个地标点贡献率之和达到61.8%,在整体判别分析中具有显著作用。喀什眶灯鱼和马来眶灯鱼的听沟在位置上与西氏眶灯鱼有较为明显的差别,从网格差异可视化的结果显示,西氏眶灯鱼的听沟位置更加靠右下方,与另外两种眶灯鱼之间的差异较大且差异产生的趋势不一致,而另外两种眶灯鱼之间相比虽然整体差异产生的趋势相似,但也并不完全一致。这一结果也印证了Gaga[11]在文献中所指出的灯笼鱼同属矢耳石形态上存在一定差异的结论。
鱼类耳石听沟的形态特征主要受遗传因素的影响[9, 12, 41],但也有研究表明,鱼类的生活环境以及生活习性也会对其产生一定影响。姜涛[25]在研究不同地理群体凤鲚(Coilia mystus)和七丝鲚(C. gray)时,认为九龙江和珠江两地的盐度不同是导致二者听沟形态产生差异的主要原因。除却温度和盐度条件外,栖息水深所带来的环境变化也有可能会作用于耳石听沟,结合Sassa和Kawaguchi[42]和李励年等[43]的相关研究数据,表明本文3种眶灯鱼在成年时期栖息水深主要集中于水下200~1 000 m的深度范围,其深度范围有较大的重合度。为进一步分析造成听沟产生差异的原因,本课题组基于3种眶灯鱼以及外群七星底灯鱼(Benthosema pterotumCOI基因序列的分析结果发现,无论是碱基组成还是遗传距离均提示马来眶灯鱼和西氏眶灯鱼的亲缘关系更近于马来眶灯鱼和喀什眶灯鱼(未发表数据)。而基于矢耳石的研究结果则表明,喀什眶灯鱼和马来眶灯鱼听沟更为相似,因而我们推测遗传因素可能并不是造成这3种眶灯鱼之间矢耳石听沟差异的主要原因。本文的研究对象采集海域分别为印度洋−阿拉伯海和南海,采样区域的纬度相似,但根据WOA的数据,这两片海域在温度、盐度和深度等环境因素方面存在明显差异,阿拉伯海海域的盐度和温度都明显高于南海,尤其是阿拉伯海海域盐度显著高于南海海域(图3图4)。由此,我们推测听沟产生差异的原因可能是由于两个海区不同的环境所造成,其中盐度是不可忽视的一个因素,但关于盐度造成听沟以及其他矢耳石形态学差异的详细机制还需后续更深入的研究。
