During an investigation of marine diatom flora in the sand samples from Guangxi, three species were identified as new records in China, which were Halamphora woelfeliae Stachura-Suchoples, Enke, Schlie, Schaub, Karsten & R.Jahn, Psammodiscus calceatus Tsuy.Watanabe, Nagumo & Ji.Tanaka and Ehrenbergiulva granulosa (Grunow) A.Witkowski, Lange-Bertalot & Metzeltin. Their morphological features, ecological habitats and geographical distribution were described in detail. This study enriched the biodiversity of marine diatoms in China.

一般认为,由于沙质海岸具有底质不稳定、营养物质缺乏和生态环境恶劣等特点,导致其中分布的生物(如大型海藻和底栖生物)种类相对较少[12]。然而,底栖硅藻却能够较好适应这一特殊生境[34],它们甚至可以成为沙质海岸带中一个十分重要的生物类群[56]。为了应对复杂的沙质环境,生活在其中的大多数硅藻种类通常个体较小且偏好附着在沙粒的凹陷处和裂缝处,此类独特的生存方式可以帮助硅藻减小受到沙粒之间摩擦的影响,但也使得该生境中的硅藻常在多样性研究中被忽略[2]。沙质环境中的硅藻在国外较早受到关注,并取得了一系列的研究成果。Round曾在一个小型淡水湖泊中发现了附着有硅藻的沙粒,由此首次提出了沙表生硅藻(epipsammic diatom)的概念[7];其后,他又在美国马萨诸塞州巴恩斯特布尔湾的表层沙子中观察到大量附着生长的硅藻[8];Jewson等人甚至在0.5 m深的沙子中也发现了高密度的硅藻,并说明了两种不同的硅藻生态策略[9];Krejci和Lowe的研究则表明了沙表生硅藻对不同材质的沙粒具有一定的选择性[10];此外,以Witkowski和Park等人为代表的学者还对沙质环境中的硅藻进行了分类学研究,并发现了一些新属和新种[1112]。与国外相比,虽然我国对该硅藻类群的认识较晚,但在多样性研究方面进展较快。例如,赵龙首次对我国沿海沙质环境中的硅藻进行了初步调查,一共记录了81种硅藻[13],发现了1个新种和3个中国新记录种;陈长平等人则系统地针对福建沿海沙表生硅藻开展了详细的种类鉴定和形态描述,总计观察到了161种硅藻,包含9个新种、6个中国新记录属和40个中国新记录种[2]。尽管如此,我国针对沙质环境中的硅藻研究仍有很大的探索空间,已被描述的分布区域还十分有限,可能还存在着许多未知的硅藻种类。
作者于2021年在对广西沿海主要优质沙滩的潮间带进行硅藻多样性调查时,发现了3个我国首次记录的硅藻种类,分别为沃氏海生双眉藻(Halamphora woelfeliae Stachura-Suchoples, Enke, Schlie, Schaub, Karsten & R.Jahn)、履沙盘藻(Psammodiscus calceatus Tsuy.Watanabe, Nagumo & Ji.Tanaka)和颗粒爱氏藻(Ehrenbergiulva granulosa (Grunow) A.Witkowski, Lange-Bertalot & Metzeltin)。海生双眉藻属(Halamphora (Cleve) Levkov)原是双眉藻属(Amphora Ehrenberg ex Kützing)下的一个亚属[14],Levkov则将其修订为独立的硅藻属[15]。此属种类大多生活在海洋或半咸水中,仅有少数种类可发现于淡水中[16]。与双眉藻属相比,海生双眉藻属的种类仅可能在背侧具有壳缝突起(raphe ledge),且线纹由单列或双列的点纹组成[15]。沙盘藻属(Psammodiscus Round & Mann)是一个寡种硅藻属,目前仅包含3个种类,细胞全部呈圆盘形。该属模式种为光亮沙盘藻(P. nitidus (Gregory) Round & Mann),其基名为光亮圆筛藻(Coscinodiscus nitidus Gregory),在我国已有报道,但该种主要营沙表生生活,壳面点纹由无网孔(slits)的筛板(rotae)覆盖,且具有较小的无柄唇形突(sessile rimoportulae)。因此,Round和Mann将其从圆筛藻属(Coscinodiscus Ehrenberg)中分离出来并独立为沙盘藻属[17]。爱氏藻属(Ehrenbergiulva A.Witkowski, Lange-Bertalot & Metzeltin)的主要形态特征包括细胞呈圆盘形、壳面中部无纹、壳缘处可见一圈很短的线纹、壳缘内侧分布有等距的壳面支持突[11]。此属也是一个寡种硅藻属,目前只有2个种类,其模式种为颗粒爱氏藻[11]
本文通过扫描电子显微镜对3个硅藻新记录种开展了形态学观察和分类学研究,并分别与各自的相似种进行了对比和区分。此外,作者还详细说明了它们的生态习性和分布情况。
本文中所描述的3种硅藻的采集日期、地点和坐标如表1所示。每次采样均在最低潮时的前后半小时内,通过使用不锈钢铲子刮取沙滩低潮区的表层沙完成。为减小运输过程中产生的不利影响,所采样品均置于自封袋中,并保存在放有冰袋的冰盒内,带回实验室做进一步处理。
实验室内,首先取适量的表层沙样品转移至干净的烧杯中,加入大量的过滤后海水清洗样品3次,以去除杂质;其后再次向清洗后的样品中加入适量的过滤海水,置于超声波清洗机中进行超声处理,以分离得到附着在沙粒表面的硅藻样品;然后将分离下来的硅藻样品分成2份,其中1份样品直接加入鲁格试剂进行固定,另1份则利用稀释法分离得到纯培养的硅藻单克隆株系;接着取10 mL固定后的硅藻样品和10 mL扩大培养后的纯种硅藻培养液转移至干净的试管中,分别加入等体积的浓硝酸(65%~68%),置于沸水浴中加热20 min,待样品冷却后,加入蒸馏水清洗硅藻样品多次直至中性,以去除样品中有机质和盐分;最后,利用移液枪吸取20 μL清洗后的硅藻样品,均匀地滴加在5 mm × 5 mm的单晶硅片上,自然晾干后,喷金,使用Hitachi S-3400N扫描电子显微镜进行观察和拍照。

形态特征:壳面窄,半披针形。长8.3~9.7 μm,宽2.0~2.4 μm。两端略延伸,呈轻微头状。腹背分明。腹缘直,或略微凹陷;背缘平滑,呈弓形。背侧线纹呈轻微放射状,10 μm内40条,线纹由单列的长室孔组成,被一条纵向肋纹分隔呈两部分;腹侧线纹由单个小的长室孔组成(图1a)。中轴区狭窄,背侧无中心区。外壳面,近缝端相距较近且向腹侧弯曲(图1a)。内壳面,近缝端之间具硅质肋,远缝端终止于小的喇叭舌上(图1b)。
生态习性:海水种,生活在沉积物中。
地理分布:首次记录于挪威的斯瓦尔巴群岛,本文中的样品采自于广西防城港市白龙尾沙滩,为我国首次记录。

形态特征:壳面呈圆形,直径22.0~39.8 μm。线纹从壳面中部至壳套呈放射状排列,10 μm内约有8条(图2a)。点纹10 μm内4~5个,呈圆形或椭圆形,内有筛板覆盖。每个筛板有5~9个辐条(spokes)与外壳面相连(图2c)。壳套上的点纹明显小于壳面上的点纹。壳面中部最多分布有1个较小的无柄唇形突(图2b),其在外壳面偶见一个开口。内壳面平坦,筛板的辐条不明显。本种在壳面中部无小孔。
生态习性:海水种,营沙表生生活。
地理分布:首次记录于日本福井县敦贺市,本文中的样品采自于广西防城港市白沙湾沙滩、北海市银滩和侨港沙滩,为我国首次记录。

形态特征:壳面呈圆形,轻微凸起,直径15.1~19.8 μm。壳面中部几乎无纹路(图3a)。壳缘窄,分布有1圈很短的线纹,10 μm内23~28条(图3a)。壳缘内侧可见1圈壳面支持突,10 μm内3个(图3bc)。
生态习性:海水种,常见于潮间带。
地理分布:曾记录于欧洲北海和大西洋沿岸、波罗的海、熊岛、新喀里多尼亚、南非以及塞舌尔群岛,本文中的样品采自于广西北海市银滩以及防城港市金滩、白浪滩和二白龙沙滩,为我国首次记录。
沃氏海生双眉藻最初由Stachura-Suchoples等人发现于挪威斯瓦尔巴群岛孔斯峡湾的沉积物中[18]。本文中,采自广西防城港的沃氏海生双眉藻种群与其挪威模式种群在细胞大小方面较为接近(前者壳面长8.3~9.7 μm,宽2.0~2.4 μm;后者壳面长10~11.5 μm,宽2.0 μm),特别是二者背侧线纹密度均为10 μm内40条。因此,可以确定本文中的标本就是沃氏海生双眉藻。然而,两个种群的壳端形态仍存在细微差异。根据Stachura-Suchoples等人的扫描电子显微镜照片,挪威模式种群的壳端呈现出较为明显的头状特征[18],但在广西种群的个别标本中,这一特征并不明显。在海生双眉藻属较为繁多的种类中,沃氏海生双眉藻与亚盐海生双眉藻(H. subsalina Levkov)和特内海生双眉藻(H. tenerrima (Aleem & Hustedt) Levkov)在光学显微镜下最为相似,但它们之间在形态上仍有较明显的差异:(1)在背侧线纹密度方面,沃氏海生双眉藻的背侧线纹密度较高,而亚盐海生双眉藻和特内海生双眉藻的背侧线纹密度均较低(前者10 μm内24~32条,后者10 μm内22~28条);(2)在近缝端方面,沃氏海生双眉藻外壳面的近缝端相距较近,而亚盐海生双眉藻外壳面的近缝端相距较远;(3)在线纹形态方面,沃氏海生双眉藻的线纹由单列点纹组成,而特内海生双眉藻的线纹由双列点纹组成[15, 18]
沙盘藻属现有的3个种类分别为履沙盘藻、光亮沙盘藻和前光亮沙盘藻(P. praenitidus (Fenner) J.Witkowski),其中后两者均从圆筛藻属中修订而来。在分类系统中,沙盘藻属隶属于缝舟藻目(Rhaphoneidales Round)沙盘藻科(Psammodiscaceae Round & Mann),而圆筛藻属则隶属于圆筛藻目(Coscinodiscales Round & Crawford)圆筛藻科(Coscinodiscaceae Küzing)[19]。此外,沙盘藻属种类均发现于沉积物中[2021]。因此,沙盘藻属和圆筛藻属不仅亲缘关系较远,而且生态习性大不相同。履沙盘藻于2013年由Watanabe等人首次报道[20],该种硅藻首次发现时的沙粒样品采集于日本,本文中记录到该种硅藻的沙粒样品则采自广西防城港和北海等地。两个种群在点纹排列方式、点纹被膜形态以及唇形突的数量和分布等方面保持一致,因此,可以确认本研究中的硅藻样品为履沙盘藻。与属内其他两个种类相比,履沙盘藻的形态特征明显不同:(1)除点纹和唇形突开口外,履沙盘藻的壳面上不存在任何形式的小孔,而光亮沙盘藻和前光亮沙盘藻在近壳面中心处均分布有1个小孔;(2)履沙盘藻的壳面最多有1个分布在中心的唇形突,有些标本中甚至没有该结构,而光亮沙盘藻的壳面最多会有2个分布在近壳缘处的唇形突,前光亮沙盘藻的壳面则始终会存在1个唇形突;(3)履沙盘藻每个点纹的筛板有5~9个辐条与外壳面相连,而光亮沙盘藻每个点纹的筛板仅有2~3个辐条与外壳面相连;(4)履沙盘藻和光亮沙盘藻的线纹在整个壳面都呈明显的放射状排列,而前光亮沙盘藻的点纹在壳面中部的分布较为松散且无规律[2021]
爱氏藻属的拉丁文名称原为Ehrenbergia A.Witkowski, Lange-Bertalot & Metzeltin,Witkowski和 Lange-Bertalot后将其更正为Ehrenbergiulva A.Witkowski, Lange-Bertalot & Metzeltin[22]。目前,该属所包含的2个种类分别为颗粒爱氏藻和豪克爱氏藻(Ehrenbergiulva hauckii (Grunow) A.Witkowski, Lange-Bertalot & Metzeltin)[11]。二者之间的形态差异主要包括:(1)颗粒爱氏藻的直径为15~35 μm,豪克爱氏藻的直径为35~42 μm,后者细胞较大;(2)颗粒爱氏藻壳缘处的线纹密度为10 μm内18~20条,豪克爱氏藻壳缘处的线纹密度为10 μm内18~27条;(3)颗粒爱氏藻的壳面支持突密度为10 μm内3个,豪克爱氏藻的壳面支持突密度为10 μm内2.5~4个[11]。本文中,采自广西的标本的壳缘处线纹密度为10 μm内23~28条,与豪克爱氏藻较为相似。然而,广西种群的细胞直径(15.1~19.8 μm)与颗粒爱氏藻的细胞直径更为接近,且壳面支持突的密度始终保持在10 μm内3个。因此,我们认为广西种群应为颗粒爱氏藻。本研究为爱氏藻属在我国的首次记录,关于该属的详细形态特征描述如下[11]

细胞单个生活。壳面观为圆形并轻微凸起,环面观则为矩形。细胞壳面具有不规则排列的细小孔纹,但中部无任何结构。光镜下,壳缘分布有很短的放射状线纹;扫描电镜下,壳套具有等距排列的狭缝。壳缘可见支持突。
本文详细描述了3种我国首次记录的沙质环境中的海洋硅藻,研究结果不仅扩展了它们的地理分布区域,而且丰富了广西甚至我国海洋硅藻多样性和生态特征资料。
