Untargeted metabolomics analysis using a UHPLC-Exploris240 system was used to analyze intracellular metabolites and regulatory mechanisms between the high erythritol-producing mutant and wild-type strain of Yarrowia lipolytica during fermentation. A total of 482 significantly differential metabolites were identified. KEGG pathway enrichment analysis revealed that the pathways related to erythritol biosynthesis of this strain were significantly enriched, involving the pathways related to carbon flux utilization, which also preliminarily revealed that carbon flux redistribution may be a key regulatory mechanism affecting the increase of erythritol production of this mutant strain. In addition, the study also showed that the enhancement of the cofactor had a synergistic effect on erythritol synthesis in this mutant strain. Further analysis revealed that the intracellular content of glycerol, which can serve as a non-oxidative carbon source, significantly increased in the mutant strain. Subsequent fermentation experiments further demonstrated that under mixed carbon source (glycerol/glucose) conditions, the mutant strain achieved a higher erythritol yield of 73.67 g/L. This indicates that appropriate exogenous glycerol has a certain promoting effect on erythritol synthesis in the mutant strain. Thus, this study provides insights into the metabolic regulation of erythritol synthesis in Y. lipolytica at the metabolite level through metabolomic analysis, and also offers a basis for the rational strain modification via metabolic engineering strategies to enhance erythritol production.

为探究解脂耶氏酵母（Yarrowia lipolytica）高产赤藓糖醇突变菌株与原始菌株胞内代谢产物差异及调控机制，非靶向代谢组学采用超高效液相色谱串联傅里叶变换质谱 UHPLC-Exploris240 系统，对突变菌株及其原始菌株发酵过程中的代谢产物进行差异性分析，共鉴定出482个差异代谢产物。随后通过KEGG通路富集分析发现，与该菌株赤藓糖醇合成相关的通路被显著富集，其中涉及与碳通量利用相关的通路，初步揭示了碳通量重新分配可能是影响该突变菌株赤藓糖醇产量提高的关键调控机制。同时，研究还表明辅助因子的强化对该突变菌株赤藓糖醇的合成具有协同增效作用。进一步分析发现，作为Y. lipolytica非氧化途径碳源的甘油在突变菌株胞内的含量升高，随后的发酵实验进一步证明，该突变菌株在混合碳源（甘油/葡萄糖）条件下赤藓糖醇产量可达73.67 g/L，表明适当的外源甘油对突变菌株的赤藓糖醇合成具有一定的促进效果。本研究通过代谢组学分析，从代谢产物层面为解析Y. lipolytica赤藓糖醇合成过程中的代谢调控提供了思路，也为后续通过代谢工程策略进行菌株的理性改造从而提高赤藓糖醇合成能力提供了一定的依据。
