Michelia odora is a rare and endangered species unique to China. In recent years, it has been discovered that this species has been suffering from leaf blight. In this study, the symptoms were observed to reveal the pathogen of M.odora leaf blight. A fungal strain Fg-1 with high pathogenicity was isolated from the diseased leaves of M.odora by conventional tissue isolation. The pathogenic strain Fg-1 was identified as Colletotrichum gloeosporioides by morphological observation and multi-gene sequence analysis of internal transcribed spacer(ITS), ribosomal large subunit(LSU), and ribosomal small subunit(SSU). The results showed that the optimum growth temperature for the pathogen was 25 ℃, and the optimal light condition was 12 h light/12 h darkness. The pathogen could grow in the potato dextrose agar with pH 4-12, with the optimal pH 7. The colonies grew on all six types of culture media tested, namely water agar, synthetic low-nutrient agar, potato sucrose agar(PSA), mung bean agar, wheat agar, and corn agar. The colonies demonstrated the best growth on PSA. The most suitable carbon source was sorbitol, and the most suitable nitrogen sources were beef extract.

观光木是中国特有的珍稀濒危树种, 近年发现观光木遭受叶枯病危害, 为明确其病因, 对观光木叶枯病的发病症状进行调查, 通过组织分离法从观光木病叶中分离到具有强致病力的菌株Fg-1。采用形态学观察与内转录间隔区(ITS)、核糖体大亚基(LSU)、核糖体小亚基(SSU)多基因序列分析相结合的方法对病原菌Fg-1进行鉴定, 确定其为胶孢炭疽菌。生物学特性研究表明, 该病原菌菌落的最适生长温度为25 ℃; 最适光照条件是12 h光暗交替; 菌落可在pH值为4~12的马铃薯葡萄糖琼脂(PDA)培养基中生长, 最适培养基pH值为7;菌落在水琼脂(WA)、合成低营养琼脂(SNA)、马铃薯蔗糖(PSA)、绿豆琼脂(MMA)、小麦琼脂(WMA)及玉米琼脂(CMA)6种培养基上均有生长, 其在马铃薯蔗糖(PSA)培养基上生长最好; 菌落生长最适碳源为山梨醇, 最适氮源为牛肉浸膏。

观光木是中国特有的珍稀濒危树种, 近年发现观光木遭受叶枯病危害, 为明确其病因, 对观光木叶枯病的发病症状进行调查, 通过组织分离法从观光木病叶中分离到具有强致病力的菌株Fg-1。采用形态学观察与内转录间隔区(ITS)、核糖体大亚基(LSU)、核糖体小亚基(SSU)多基因序列分析相结合的方法对病原菌Fg-1进行鉴定, 确定其为胶孢炭疽菌。生物学特性研究表明, 该病原菌菌落的最适生长温度为25 ℃; 最适光照条件是12 h光暗交替; 菌落可在pH值为4~12的马铃薯葡萄糖琼脂(PDA)培养基中生长, 最适培养基pH值为7;菌落在水琼脂(WA)、合成低营养琼脂(SNA)、马铃薯蔗糖(PSA)、绿豆琼脂(MMA)、小麦琼脂(WMA)及玉米琼脂(CMA)6种培养基上均有生长, 其在马铃薯蔗糖(PSA)培养基上生长最好; 菌落生长最适碳源为山梨醇, 最适氮源为牛肉浸膏。

Michelia odora is a rare and endangered species unique to China. In recent years, it has been discovered that this species has been suffering from leaf blight. In this study, the symptoms were observed to reveal the pathogen of M.odora leaf blight. A fungal strain Fg-1 with high pathogenicity was isolated from the diseased leaves of M.odora by conventional tissue isolation. The pathogenic strain Fg-1 was identified as Colletotrichum gloeosporioides by morphological observation and multi-gene sequence analysis of internal transcribed spacer(ITS), ribosomal large subunit(LSU), and ribosomal small subunit(SSU). The results showed that the optimum growth temperature for the pathogen was 25 ℃, and the optimal light condition was 12 h light/12 h darkness. The pathogen could grow in the potato dextrose agar with pH 4-12, with the optimal pH 7. The colonies grew on all six types of culture media tested, namely water agar, synthetic low-nutrient agar, potato sucrose agar(PSA), mung bean agar, wheat agar, and corn agar. The colonies demonstrated the best growth on PSA. The most suitable carbon source was sorbitol, and the most suitable nitrogen sources were beef extract.

观光木(Michelia odora) 又称香木楠或宿轴木兰,为木兰科(Magnoliaceae) 含笑属(Michelia) 常绿阔叶乔木,是我国特有的古老珍稀树种。由于生境退化、自然更新困难,观光木自然种群数量不断减少,被列为国家二级重点保护野生植物[1],主要分布于我国南方热带和南亚热带常绿阔叶林中[2]。观光木树形高大秀丽,枝叶茂密,在园林绿化中具有良好的应用潜力[3];其根与树皮中含萜类、生物碱等活性成分,具有显著的抗肿瘤活性,为抗癌药物研发提供重要物质基础[4];其干燥果实常用于治疗消化不良及慢性鼻炎等症;其枝条及叶片富含挥发性芳香物质,可提取用于香料工业、化妆品配方及医药中间体合成,具有很大的经济价值和发展潜力[5-7]。目前关于观光木的研究主要在人工繁育、群落生态、物理性质及化学成分等方面[8-11],有关病虫害的研究报道较少。1985年, 谢宝多等[12]首次在广西发现观光木青枯病病害,经鉴定,该病害是由青枯假单胞杆菌(Pseudomonas solanacearum) 亚洲变种引起的。观光木栽培主要受苗期病害(如猝倒病、根腐病、立枯病) 和鳞翅目幼虫(如潜叶蛾、卷叶蛾) 影响。国内外暂未发现关于观光木叶枯病及其病原菌的研究报道。
2023年,在福建省林业科学研究院的观光木培育苗圃中发现叶枯病危害,受害叶片边缘出现轮状病斑,导致叶片大面积黄化坏死,严重影响观光木生长。目前,产生该病害的病原尚不明确,为了有效防止病害发生,对受害病叶病原菌进行分离纯化,基于柯赫氏法则开展致病性试验,结合该致病菌在培养基中的形态特征和分子鉴定结果,明确了其分类地位,同时系统测定了该致病菌在不同培养条件下的生物学特性,为确定观光木叶枯病的发生规律提供理论依据,从而制定有效的防治措施。
2023年12月3日,从福建省林业科学研究院观光木培育苗圃(26°15′N,119°29′E) 采集观光木叶枯病病叶样本。
采用组织分离法从病叶样本中获取致病菌株,具体操作步骤如下:将病叶样本用流水冲洗3 min,清除叶片表面杂质后,用无菌纸吸干残留水分,依次进行消毒处理(75%C2H6OH,1 min;5%NaClO,1.5 min;5%C2H6OH,30 s),最后使用无菌水漂洗数次,将叶片放置在无菌纸上晾干。为验证灭菌效果,取适量末次漂洗无菌水涂布于马铃薯葡萄糖琼脂(potato dextrose agar,PDA) 培养基平板,25 ℃恒温培养,定期观察培养基中是否有菌落长出,若无,则说明组织完成消毒。于叶片病健交接处取2 mm×2 mm的组织块,每个培养基平板放置5块,25 ℃恒温培养,若培养基中组织块上有菌丝长出,将菌丝转至新的培养基中继续培养,编号并保存于4 ℃冰箱。
基于柯赫氏法则开展离体组织致病性验证:选取观光木上生长状态相同的新鲜健康叶片,对叶片进行清洗处理(清水冲洗→75%乙醇消毒→无菌水漂洗) 后,在刺伤的创口处接种5 mm的新鲜病原菌菌饼,对照组在创口处接种空白PDA。接种叶片放入托盘内,用无菌水润湿的无菌棉包裹叶柄保湿,置于25 ℃培养箱恒温培养。对发病的叶片进行病原菌分离处理,将再次分离出的菌株与原致病菌株进行比对,观察二者的培养特性,确定形态特征是否一致。
将病原菌Fg-1在PDA培养基中25 ℃恒温培养,定期观察,记录菌株在培养基中的形态特征,在显微镜下观察并测量菌丝、孢子、子实体等结构。
将病原菌Fg-1置于马铃薯葡萄糖液体(potato glucose broth,PDB) 培养基中,放入25 ℃恒温摇床培养3 d后过滤出菌丝,使用十六烷基三甲基溴化铵(hexadecyl trimethyl ammonium bromide,CTAB) 法提取病原菌DNA。分别对内转录间隔区(internal transcribed spacer,ITS)、核糖体大亚基(ribosomal large subunit,LSU)、核糖体小亚基(ribosomal small subunit,SSU) 基因序列进行聚合酶链式反应(polymerase chain reaction,PCR) 扩增。PCR扩增程序为:预变性3 min,变性1 min,温度均为95 ℃;退火50 s,ITS退火温度为55 ℃,LSU、SSU退火温度为52 ℃,延伸72 ℃,30 s,共34个循环,再延伸72 ℃,10 min,产物放入4 ℃冰箱低温保存。
使用DNA纯化试剂盒(FastPure Gel DNA Extraction Mini Kit-DC301) 进行扩增产物的回收。回收片段与载体pMC100-MB酶连,转化进大肠杆菌(Escherichia coli DH5α) 进行扩大培养,将转入目的基因的菌液送至尚亚生物公司,对目的片段进行测序。将所得数据在(national center for biotechnology information,NCBI) 数据库中进行初步比对,用MEGA-11软件构建多基因联合系统发育树,采用最大似然法(maximum-likelihood,ML),重复计算1 000次。
将低温保存的病原菌Fg-1接种于PDA培养基中,25 ℃恒温培养6 d后,使用外径6 mm的无菌打孔器在新鲜菌落边缘打取菌饼备用。
将病原菌菌饼接种于PDA平板中,分别在5~40 ℃ (梯度间隔5 ℃) 下恒温培养6 d,测量并记录菌落直径,试验重复3次。
在25 ℃恒温培养条件中,将病原菌菌饼接种至PDA平板中,分别进行24 h光照、24 h黑暗和12 h光暗交替3种光照条件处理,培养6 d后测量菌落直径,试验重复3次。
用0.1 mol · L-1的HCl和NaOH溶液分别将PDA培养基pH值调整至4~13 (pH值梯度间隔1),将病原菌菌饼分别置于不同pH值的PDA培养基平板中,25 ℃恒温条件下培养6 d,测量并记录菌落直径,试验重复3次。
将病原菌菌饼分别接种于水琼脂(water agar medium,WA)、合成低营养琼脂(synthetic low nutrient agar,SNA)、马铃薯蔗糖(potato sucrose agar,PSA)、绿豆琼脂(mung bean agar medium,MMA)、小麦琼脂(wheat agar medium,WMA) 和玉米琼脂(corn agar medium,CMA) 6种培养基平板中,25 ℃恒温条件下培养6 d,测量并记录菌落直径,试验重复3次。
将固体察氏培养基中的蔗糖分别替换为等质量的淀粉、甘露醇、山梨醇、葡萄糖、果糖、乳糖,将病原菌菌饼分别接种于上述的6种培养基中,对照组为不含碳源的固体察氏培养基,25 ℃恒温下培养6 d,测量并记录菌落直径,试验重复3次。
将固体察氏培养基中的硝酸钠分别替换为等质量的甘氨酸、硫酸铵、氯化铵、牛肉浸膏、硝酸铵、硝酸钾,将病原菌菌饼分别接种于上述的6种培养基中,对照组为不含碳源的固体察氏培养基,25 ℃恒温下培养6 d,测量并记录菌落直径,试验重复3次。
采用IBM SPSS 25.0软件和Excel软件进行数据处理与分析,并运用Duncan新复极差法(P≤0.05) 进行差异显著性检验。该方法通过将数据划分为若干子集,分别计算各子集的均值与方差,进而比较多样本间的统计学差异。
观光木叶枯病发病初期,叶片边缘和叶尖出现红褐色轮状病斑,随着病情发展,病斑逐渐扩大,直至大部分组织枯死,叶片脱落(图 1AB)。将病叶带回室内通过组织分离法获得31株真菌,其中相同菌株23株,将其编号为Fg-1 (图 1C),其余菌株按Fg-2~Fg-9依次编号,致病性检测发现菌株Fg-2~Fg-9均无致病性,只有菌株Fg-1回接至健康叶片3 d后叶片出现黑褐色病斑,随着时间延长,接种部位呈现出黑褐色轮状病斑,并出现橙黄色孢子堆,对照组接种7 d后无明显变化(图 1D),试验组7 d后病斑逐渐蔓延至整个叶片(图 1E)。根据柯赫氏法则,二次分离菌株与原菌株一致(图 1F),说明菌株Fg-1为观光木叶枯病的致病菌株。
在PDA培养基上,病原菌Fg-1生长迅速,菌落呈规则圆形,正面灰褐色毛毡状,背面深褐色,向四周辐射状生长(图 2A);后期菌落表面形成橙黄色孢子堆(图 2B);叶片接种15 d左右在表面产生红褐色球形子实体,多单生,直径25~56 μm (图 2C);分生孢子梗圆柱状,光滑有隔膜,产孢细胞来自孢子堆内,细胞透明,不连续(图 2D);分生孢子短棒形或椭圆形,表面光滑无隔膜,内含油滴,大小为(12.58~20.15) μm × (5.83~6.96) μm (图 2E);孢子萌发时常在中部产生1~2个隔膜,每个隔膜处生出1个芽管(图 2F)。结合形态学特征及相关文献报道[13],初步判定病原菌Fg-1为胶孢炭疽菌(Colletotrichum gloeosporioides)。
进一步对病原菌Fg-1的ITS、LSU、SSU基因序列进行PCR扩增,将其结果在NCBI基因数据库进行比对分析,该菌株序列与胶孢炭疽菌相似度较高。将ITS、LSU、SSU三个基因片段串联,利用MEGA-11软件构建多基因串联系统进化树(图 3)。菌株Fg-1与胶孢炭疽菌在自举值99%水平相聚一簇,遗传关系最近。结合菌株在培养基中的形态学特征及培养性状,确定菌株Fg-1为胶孢炭疽菌。
图 4(a) 可见,病原菌Fg-1在培养6 d后,不同温度对菌落直径影响明显。当温度为5 ℃时,菌落生长较慢,菌落直径仅为13.06 mm;温度逐渐升高时,菌落直径逐渐增大,当温度为10、15 ℃时,菌落生长差异性较小,菌落直径分别为30.19、32.31 mm;当温度为25 ℃时,菌落直径达到81.86 mm,显著大于其他温度(P < 0.05);当温度高于25 ℃时,菌落生长受到抑制,菌落直径逐渐减小;当温度达到40 ℃时,菌落停止生长。结果表明,最适合病原菌Fg-1生长的温度为25 ℃。
病原菌Fg-1在不同光照条件下培养6 d的生长状况见图 4(b)。其在24 h光照、24 h黑暗及12 h光暗交替3种光照条件下均表现出生长活性。在不同光照条件下,菌落直径差异显著(P < 0.05),其中,12 h光暗交替的菌落直径最大(78.11 mm),显著高于24 h光照(75.50 mm) 和24 h黑暗(69.10 mm)。结果表明,在12 h光暗交替条件下有利于病原菌Fg-1的生长。
病原菌Fg-1在不同pH值培养基中的生长状况[图 4(c)]表明,不同pH值的培养基对Fg-1菌落的生长有显著影响(P < 0.05)。在酸性至中性(pH值为4~7) 培养基中,随着pH值的升高,菌落直径逐渐增大,当pH值为7时,菌落直径为80.26 mm,显著高于其他pH值(P < 0.05);当培养基从中性变为碱性(pH值>7) 时,菌落直径开始逐渐减小;当培养基为强碱性(pH值为13) 时,菌落停止生长。
图 4(d) 可见,病原菌Fg-1在供试的6种培养基上均有不同程度的生长。在PSA、MMA培养基上生长较好,菌落直径分别为77.62 mm和74.56 mm,在WMA、CMA培养基上生长状况次之,菌落直径分别为70.54 mm和70.93 mm;在WA、SNA培养基上,菌落直径明显小于其他培养基。
病原菌Fg-1在含不同碳源的固体察氏培养基上生长状况见图 4(e)。在以山梨醇为碳源的培养基上,菌落直径最大,为65.85 mm;在以葡萄糖、果糖、甘露醇、蔗糖、淀粉为碳源的培养基上,菌落生长状况次之;而在无碳源和以乳糖为碳源的的培养基上,菌落直径显著低于其他碳源的培养基(P < 0.05),分别为49.66 mm和44.96 mm。
病原菌Fg-1在含不同氮源的固体察氏培养基上的生长状况见图 4(f)。在以牛肉浸膏、甘氨酸、硝酸钾为氮源的培养基上,菌落生长状况较好,菌落直径分别为66.32、65.19、63.84 mm;在以硝酸铵为氮源的培养基上菌落生长状况次之,菌落直径为50.32 mm;在无氮源和以氯化铵、硝酸钠、硫酸铵为氮源的培养基上菌落生长状况较差,菌落直径明显小于其他培养基。
胶孢炭疽菌是炭疽菌属中的一类重要植物病原真菌[13],多分布于热带和亚热带地区,尤其对亚洲、非洲和美洲的农林业产区造成严重危害[14]。在我国,胶孢炭疽菌分布遍及四川、广西、海南、云南、福建、黑龙江等多个省(区) [15]。胶孢炭疽菌具有极广的宿主范围,可侵染多种植物,包括果树(如芒果、柑橘、榴莲、苹果、梨、荔枝、龙眼)、经济作物(如橡胶树、咖啡)、蔬菜、花卉和林木(如杨树、油茶、杉木) 等[16-19]。该菌通过风雨传播和昆虫媒介(如丽蝇) 扩散,高温高湿易引发病害暴发。胶孢炭疽菌引发的病害可导致叶片坏死、果实脱落甚至植株死亡。在芒果上引发炭疽病,严重时导致果实减产30%~50%[20],橡胶树感染后胶乳产量显著下降[21]。杨树是胶孢炭疽菌的重要寄主之一,该菌通过侵染叶片形成褐色坏死斑,破坏叶肉细胞结构(如叶绿体解体、细胞壁溶解),最终抑制树木生长[22]。杨树感染胶孢炭疽菌后,其免疫应答与生长调控基因(如PagWRKY33) 的失衡可能导致生长受限[23]。此外,树木感染该菌后还削弱其抗逆性,增加次生害虫侵染风险。国内外研究表明,胶胞炭疽菌能够引起多种植物发生病害,对农业和林业经济造成重大损失,而有关胶胞炭疽菌侵染观光木的报道尚未发现。
本研究发现, 观光木叶片从边缘和叶尖出现褐色轮状病斑,后期叶片枯死脱落,具有典型叶枯病症状。采集病叶进行病原菌分离鉴定结果表明, 引起观光木叶枯病的致病菌为胶胞炭疽菌。病原菌Fg-1的生物学特性研究表明,该菌对温度适应范围较宽,在5~35 ℃均能生长,最适生长温度为25 ℃; 可在不同的光照条件下生长,最适光照条件为12 h光暗交替; 在pH值为4~12内均可生长,最佳pH值为7,这与前人研究结果基本一致[24]。并且病原菌Fg-1在不同的培养基中均可生存,说明该菌对外界环境有着较强的适应性。在供试的7种碳源培养基中,最适病原菌Fg-1生长的是山梨醇,葡萄糖、果糖、甘露醇、蔗糖、淀粉次之,这与蒋朝阳等[25]报道的葡萄炭疽病病原菌最适碳源主要为果糖、可溶性淀粉、葡萄糖有所区别。病原菌Fg-1在以氯化铵、硝酸钠、硫酸铵为氮源的培养基中生长状况较差,这与廖旺姣等[26]的研究结果基本一致。但张琳等[27]认为,人参炭疽病菌在含有硝态氮和氯化铵的培养基中生长较好。由此可见,不同寄主的炭疽菌在生长过程中对碳、氮源的需求具有一定的相似性和差异性。
综上所述,引起观光木叶枯病的病原菌Fg-1是胶胞炭疽菌,该菌是一类全球性病原真菌,其遗传多样性和环境适应性增加了病害防控难度。因此, 在观光木苗木繁育过程中应以综合管理为主: 及时摘除病叶,减少侵染源;增强通风透光,避免密植和高温高湿;还可以和非寄主植物混合培育,减少病原菌的积累和传播;病害大爆发时大多采用化学防治手段。今后还应对该致病菌进行室内和田间毒力测定的研究,为观光木苗木繁育提供更多的参考依据。
