Fujian Shuixian belongs to the semi-tree form of tea plant (Camellia sinensis), which is the main variety for processing Wuyi Rock Tea (WRT). At present, there is a widely phenomenon that Shuixian grows naturally (no-pruning) in the management of tea gardens of Wuyishan, and its quality is obviously different from that of pruned Shuixian. In order to clarify the basis and differences of aroma and flavor substances WRT under natural growth and pruning conditions, this study used gas chromatography-mass spectrometry (GC-MS) and high performance liquid chromatography (HPLC) technology, combined with multivariate statistical analysis, to explore the internal relationship among aroma substances, flavor components and quality of WRT before baking (raw tea) and after baking (finished tea) under natural growth and pruning conditions. The results showed that 261 volatile compounds were identified by GC-MS technology. It reveals that there are obvious differences in aroma substances between raw tea and finished tea of the natural growth Shuixian and pruned Shuixian with the help of principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). Compared with the raw tea of pruning Shuixian, the raw tea of natural growth Shuixian showed significant differences in 41 volatile substances (Selection standard: VIP>1, and the FC≥1.5 or the FC≤0.67), in which the relative content of hexyl hexanoate, plant alcohol, isophytol, trans-nerolidol are higher, however, the relative content of β-cedrene, γ-junitene and 2-methoxyfuran are lower. Compared with the finished tea of pruning Shuixian, the finished tea of natural growth Shuixian showed significant differences in 71 volatile substances, such as hexyl hexanoate, methyl caproate, trans-nerolidol, (Z)-β-farnesene, 1-heptanol, 2-heptanone with fruity aroma, and 3-ethyl-1H-pyrrole, 2-formyl-4with baking aroma, whose relative content are higher. There are also great differences in aroma substances between natural growth Shuixian and pruned Shuixian before and after baking, with 91 and 70 different substances respectively. The analysis of aroma activity value (OAV) showed that (E)-3-hexenol, indole, trans-nerolidol, epoxy-β-ionone, (E)-linalool oxide, 2-phenylethanol and methyl salicylate may be important substances for the aroma formation of natural growth Shuixian. Analysis of the main flavor substances showed that there were obvious differences in catechin components and caffeine between natural growth Shuixian and pruned Shuixian. The results of sensory evaluation are consistent with the results of physical and chemical test, showing that natural growth Shuixian with rich fruit fragrance, mellow taste and better quality. This study has a guiding role in scientific management of tea gardens and tea quality control.

福建水仙属于小乔木型茶树,是加工武夷岩茶的主要品种之一。目前,武夷山茶园管理过程中存在水仙茶树自然生长的现象(不修剪),其制茶品质与修剪水仙有明显区别。为明确自然生长和修剪条件下,武夷水仙岩茶的香气和呈味物质基础及其差异,本研究采用气相色谱-质谱联用(gas chromatography-mass spectrometry,GC-MS)和高效液相色谱(high performance liquid chromatography,HPLC)技术,结合多元统计分析,探寻自然生长和修剪下武夷水仙岩茶在焙火前(毛茶)和焙火后(成品茶)香气物质、滋味成分与品质的内在联系。结果表明:利用GC-MS技术共鉴定出261种挥发物;通过主成分分析(principal component analysis,PCA)与正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)可知,自然生长水仙茶树和修剪水仙茶树制成的毛茶和成品茶的香气物质差异明显。与修剪水仙毛茶相比,自然生长水仙毛茶有41种挥发性物质存在显著差异(筛选差异标准:VIP>1,且FC≥1.5或FC≤0.67),己酸己酯、植物醇、异植醇、反式-橙花叔醇等花果香物质的相对含量更高;而β-雪松烯、γ-杜松烯、2-甲氧基呋喃等相对含量更低。与修剪水仙成品茶相比,自然生长水仙成品茶有71种挥发性物质存在显著差异,呈果香型的己酸己酯、己酸甲酯、反式-橙花叔醇、(Z)-β-法呢烯、1-庚醇、2-庚酮等挥发物,呈烘烤香型的3-乙基-1H-吡咯、2-甲酰基-4,5-二甲基吡咯、乙基甲基吡嗪等挥发物的相对含量更高。修剪水仙和自然生长水仙茶焙火前后的香气物质也存在很大差异,分别有91和70种差异物质。通过香气活性值(aroma activity value,OAV)分析表明,(E)-3-己烯醇、吲哚、反式-橙花叔醇、环氧-β-紫罗兰酮、(E)-芳樟醇氧化物、2-苯乙醇、水杨酸甲酯可能是自然生长水仙香气形成的重要物质。主要呈味物质分析表明,自然生长和修剪水仙茶的儿茶素组分和咖啡碱存在明显差异。感官审评结果与理化检测结果一致,表现为自然生长水仙茶花果香浓郁,滋味醇厚,品质更佳。本研究对科学管理茶园和茶叶品质调控有指导作用。

福建水仙属于小乔木型茶树,是加工武夷岩茶的主要品种之一。目前,武夷山茶园管理过程中存在水仙茶树自然生长的现象(不修剪),其制茶品质与修剪水仙有明显区别。为明确自然生长和修剪条件下,武夷水仙岩茶的香气和呈味物质基础及其差异,本研究采用气相色谱-质谱联用(gas chromatography-mass spectrometry,GC-MS)和高效液相色谱(high performance liquid chromatography,HPLC)技术,结合多元统计分析,探寻自然生长和修剪下武夷水仙岩茶在焙火前(毛茶)和焙火后(成品茶)香气物质、滋味成分与品质的内在联系。结果表明:利用GC-MS技术共鉴定出261种挥发物;通过主成分分析(principal component analysis,PCA)与正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)可知,自然生长水仙茶树和修剪水仙茶树制成的毛茶和成品茶的香气物质差异明显。与修剪水仙毛茶相比,自然生长水仙毛茶有41种挥发性物质存在显著差异(筛选差异标准:VIP>1,且FC≥1.5或FC≤0.67),己酸己酯、植物醇、异植醇、反式-橙花叔醇等花果香物质的相对含量更高;而β-雪松烯、γ-杜松烯、2-甲氧基呋喃等相对含量更低。与修剪水仙成品茶相比,自然生长水仙成品茶有71种挥发性物质存在显著差异,呈果香型的己酸己酯、己酸甲酯、反式-橙花叔醇、(Z)-β-法呢烯、1-庚醇、2-庚酮等挥发物,呈烘烤香型的3-乙基-1H-吡咯、2-甲酰基-4,5-二甲基吡咯、乙基甲基吡嗪等挥发物的相对含量更高。修剪水仙和自然生长水仙茶焙火前后的香气物质也存在很大差异,分别有91和70种差异物质。通过香气活性值(aroma activity value,OAV)分析表明,(E)-3-己烯醇、吲哚、反式-橙花叔醇、环氧-β-紫罗兰酮、(E)-芳樟醇氧化物、2-苯乙醇、水杨酸甲酯可能是自然生长水仙香气形成的重要物质。主要呈味物质分析表明,自然生长和修剪水仙茶的儿茶素组分和咖啡碱存在明显差异。感官审评结果与理化检测结果一致,表现为自然生长水仙茶花果香浓郁,滋味醇厚,品质更佳。本研究对科学管理茶园和茶叶品质调控有指导作用。

Fujian Shuixian belongs to the semi-tree form of tea plant (Camellia sinensis), which is the main variety for processing Wuyi Rock Tea (WRT). At present, there is a widely phenomenon that Shuixian grows naturally (no-pruning) in the management of tea gardens of Wuyishan, and its quality is obviously different from that of pruned Shuixian. In order to clarify the basis and differences of aroma and flavor substances WRT under natural growth and pruning conditions, this study used gas chromatography-mass spectrometry (GC-MS) and high performance liquid chromatography (HPLC) technology, combined with multivariate statistical analysis, to explore the internal relationship among aroma substances, flavor components and quality of WRT before baking (raw tea) and after baking (finished tea) under natural growth and pruning conditions. The results showed that 261 volatile compounds were identified by GC-MS technology. It reveals that there are obvious differences in aroma substances between raw tea and finished tea of the natural growth Shuixian and pruned Shuixian with the help of principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). Compared with the raw tea of pruning Shuixian, the raw tea of natural growth Shuixian showed significant differences in 41 volatile substances (Selection standard: VIP>1, and the FC≥1.5 or the FC≤0.67), in which the relative content of hexyl hexanoate, plant alcohol, isophytol, trans-nerolidol are higher, however, the relative content of β-cedrene, γ-junitene and 2-methoxyfuran are lower. Compared with the finished tea of pruning Shuixian, the finished tea of natural growth Shuixian showed significant differences in 71 volatile substances, such as hexyl hexanoate, methyl caproate, trans-nerolidol, (Z)-β-farnesene, 1-heptanol, 2-heptanone with fruity aroma, and 3-ethyl-1H-pyrrole, 2-formyl-4with baking aroma, whose relative content are higher. There are also great differences in aroma substances between natural growth Shuixian and pruned Shuixian before and after baking, with 91 and 70 different substances respectively. The analysis of aroma activity value (OAV) showed that (E)-3-hexenol, indole, trans-nerolidol, epoxy-β-ionone, (E)-linalool oxide, 2-phenylethanol and methyl salicylate may be important substances for the aroma formation of natural growth Shuixian. Analysis of the main flavor substances showed that there were obvious differences in catechin components and caffeine between natural growth Shuixian and pruned Shuixian. The results of sensory evaluation are consistent with the results of physical and chemical test, showing that natural growth Shuixian with rich fruit fragrance, mellow taste and better quality. This study has a guiding role in scientific management of tea gardens and tea quality control.

武夷岩茶属于半发酵茶类,具有独特的香气、滋味和保健功效,品质表现为“岩骨花香”和“香清甘活”[1-2],深受广大消费者的青睐。一杯香高味醇的好茶需要具备优良的茶树品种、优越的生长环境和精湛的加工工艺,三者缺一不可。香气和滋味是构成茶叶品质的重要因素,与茶叶品种、生长环境及茶园栽培措施和加工工艺密切相关。有研究表明,武夷岩茶香气不仅受生长地域的显著影响,茶园管理措施如施肥、修剪也直接影响茶青基础香气的形成[2-4]。修剪是茶树栽培中常用的栽培方式之一,与未修剪的茶树相比具有代谢差异。目前,茶园管理过程中存在水仙茶树自然生长的现象(不修剪)。传统认为,茶树修剪与不修剪模式影响岩茶香气的形成。
香气和滋味是衡量武夷岩茶品质的重要指标,分别占茶叶感官审评的30%和35%[5]。茶叶香气和滋味取决于内含物的种类和含量。目前,基于GC-MS技术,主要研究了不同地域[2]、栽培措施[6]和加工工艺[7-9]对武夷岩茶产量和品质的影响,及不同品种制成的武夷岩茶香气和滋味物质存在的差异,探寻影响武夷岩茶品质的关键因素,为客观评价和提升茶叶品质提供技术支撑。刘扬等[2]通过对高山、正岩、半岩、洲茶产区的水仙茶树进行研究,采用气相色谱飞行时间质谱仪测定茶青香气代谢物,证实岩茶“地域香”的存在。王瑜等[10]对不同配比施肥的茶青进行香气代谢物研究,表明适当降低施肥过程中磷、钾肥的比例,能显著提高武夷岩茶的产量和品质。詹宝珍等[11]研究炭焙时间对武夷肉桂岩茶香气的影响,表明炭焙前后的肉桂挥发性香气成分差异大;随焙火时间延长,多数醇类的相对含量有所降低,而多数酯类的相对含量则增加;具有焙火香的吡嗪类和吡咯类化合物在炭焙12 h后出现。GUO等[12]通过GC-MS技术测定了武夷岩茶加工过程中的挥发性化合物,发现2-乙基-3,5-二甲基吡嗪是岩茶中具有烘烤香气的芳香活性化合物。林燕萍等[5]以大红袍为材料,研究焙火程度对武夷岩茶品质、茶汤呈味物质以及香气成分的影响,轻火产品的香气表现为碳氢类、醇类、含氮类的相对含量高于中火产品。此外,研究者们采用GC-MS技术研究了焙火对武夷岩茶水仙[13]、白鸡冠[14]、瑞香[15]的香气及其主要呈味物质的影响。
福建水仙属于小乔木型、大叶种茶树,是加工武夷岩茶的主要品种,其种植和推广面积大,香气清雅,似天然兰花香。目前,茶园管理过程中有2种水仙生长形态,一种是自然生长成长为小乔木型,一种是常年修剪矮化的类型。水仙生长到30 a以上,被称为高枞,有独特的气息。根据制茶师多年的审评经验表明,自然生长高枞的品质更佳,香气更加清幽,水感更绵柔,同时形成与修剪水仙茶树不同的品质特征,推测水仙香气可能受修剪模式的影响较大。因此,本研究以相同环境和栽培条件下自然生长和修剪的水仙茶树品种加工的武夷岩茶作为研究对象,对毛茶和成品茶的挥发性物质和理化成分进行检测,并结合感官审评,解析自然生长和修剪水仙茶树的品质特征,探究二者在理化和品质上的差异,为科学管理茶园、茶叶品质调控提供理论依据,对武夷岩茶的栽培、加工和品鉴具有重要意义。
供试材料由武夷山市明慧岩茶厂提供。于2021年5月采摘水仙茶树鲜叶,采摘标准为中开面二、三叶,并按武夷岩茶加工工艺进行制作。材料为20世纪90年代种植的水仙茶树,在相同的生长环境和栽培条件下,自然生长成小乔木的水仙茶树加工的毛茶(G1)和焙火后的成品茶(G2),以及每年修剪的矮化水仙茶树加工的毛茶(D1)和焙火后的成品茶(D2)。样品分为3份,分别用于挥发性物质检测、茶汤呈味物质检测和茶叶感官审评。
武夷水仙茶加工工艺流程:鲜叶→晒青→做青→炒青→揉捻→毛火→足火→毛茶。毛茶制作完成,经手工拣剔后,进行焙火处理并取样。焙火工艺:毛茶经110~120 ℃焙火7 h制作成轻火型水仙。
实验试剂主要包括氯化钠(分析纯,国药)、正己烷(色谱纯,Merck)。标准品用正己烷进行配制,于–20 ℃保存。儿茶素标准品:表没食子儿茶素没食子酸酯(epigallocatechin gallate, EGCG)、表儿茶素没食子酸酯(epicatechin gallate, ECG)、表儿茶素(epicatechin, EC)、表没食子儿茶素(epigallocatechin, EGC)、儿茶素(catechin, C)、没食子酸(gallic acid, GA)和咖啡碱,上述标准品均购自Sigma公司。
实验仪器主要包括气相色谱质谱联用仪(Agilent, 8890-5977B),球磨仪(Retsch, MM400),电子天平(METTLER TOLEDO, MS105DU),高效液相色谱仪(Water Alliance, E2695)。萃取头(Agilent, 120 μm DVB/CAR/PDMS),固相微萃取装置(CTC Analytics, AG SPME AllowCond)。
挥发性物质的检测参照文献[16-17]的方法。采用顶空-固相微萃取(headspace-solid phase microextraction,HS-SPME)法富集香气物质,运用GC-MS进行香气图谱采集和成分分析。茶叶样品研磨成粉末。称取1 g粉末立即转移到20 mL顶空小瓶中,加入NaCl饱和溶液,加入10 μL内标溶液;全自动顶空固相微萃取HS-SPME进行样本萃取,以供GC-MS分析。以正己烷为内标,配制浓度为50 μg/mL。HS-SPME萃取条件:采样前萃取头在250 ℃下老化5 min。在100 ℃恒温条件下,顶空萃取15 min,于250 ℃下解析5 min,然后进行GC-MS分离鉴定。
GC条件:色谱柱DB-5MS毛细管柱(30 m×0.25 mm×0.25 μm,Agilent J&W Scientific,Folsom,CA,USA),载气为高纯氦气(纯度不小于99.999%),恒流流速为1.2 mL/min,进样口温度为250 ℃,不分流进样,溶剂延迟3.5 min。程序升温:40 ℃保持3.5 min,以10 ℃/min升至100 ℃,再以7 ℃/min升至180 ℃,最后以25 ℃/min升至280 ℃,保持5 min。MS条件:参照王莎等[16]的方法进行。
挥发性物质定性分析:基于MWGC数据库(该数据库来源于NIST库,与标准品共同构建而成),对样本的挥发性物质进行质谱定性定量分析。分析时样本的指纹质谱图与数据库中的参考质谱图进行匹配对比,进行定性打分,并且参考保留指数信息,增加定性的准确度,排除假阳性物质的干扰。用MassHunter定量软件处理样本的下机质谱数据,选择定量离子进行色谱峰的积分和校正工作,增强定量的准确性。
挥发性物质定量分析:根据检测出的各挥发性物质的峰面积与内标(正己烷)的峰面积进行相对定量,按文献[18]和公式(1)计算:
公式(1)中,Ci为待测挥发性物质的浓度,单位为μg/g;Cis为内标正己烷的浓度;V为内标的体积,m为茶叶质量,Si为待测组分的色谱峰面积;Sis为内标正己烷的色谱峰面积。其中,V=10 μL,Cis=50 μg/mL,m=1 g。
差异物质筛选:根据OPLS-DA模型的变量重要性投影VIP值(VIP>1),且FC≥1.5或FC≤0.67,筛选出差异化合物[19-20]
OAV计算:在各种挥发性成分定量的基础上,根据参考文献中各挥发性物质在水中的风味阈值和属性描述[21-27],按公式(2)计算各个挥发性成分的OAV[22]
公式(2)中,Ci为根据公式(1)计算出的浓度,单位为μg/g;OTi为该化合物的香气阈值,单位为μg/kg。
茶多酚、儿茶素组分和咖啡碱的测定:参照GB/T 8313—2018茶叶中茶多酚和儿茶素类含量的检测方法。
参照GB/T23776—2018《茶叶感官审评方法》对武夷岩茶水仙进行感官审评。评茶员5人,均是评茶员技师,审评项目分为外形(20%)、汤色(5%)、香气(30%)、滋味(35%)和叶底(10%),按权重法进行打分。
采用R软件进行主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)、对差异挥发性成分进行热图分析,采用SPSS 26.0软件对主要呈味物质和审评结果进行差异显著性分析。
武夷水仙的GC-MS色谱峰基线平稳,且色谱峰被有效分离,保留时间和峰强度一致,表明仪器稳定性高、数据重复性和可靠性有保障。对样本的挥发性物质进行了质谱定性定量分析,鉴定出261个挥发性物质,共分为14类。自然生长和修剪水仙茶,其挥发性成分的种类和数量均保持高度一致,但在相对含量上差异显著。
化合物一级分类结果表明,挥发性成分中,萜类、酯类、烃类数量占绝对优势,分别达37个、34个和29个(未知物除外),3个类别的挥发性物质的个数占总数超过38.12%。而挥发性成分的相对含量分析表明,占比前五的类别分别为杂环化合物、萜类、酯类、酮类和醇类(未知物除外),这5个类别在样品中相对含量的占比均在6%以上(图1)。
表1检测结果表明,通过对G1(自然生长水仙毛茶)的挥发性物质的相对含量由大到小进行排序,占比较高的前60个挥发性物质中,含有吲哚(橙子和茉莉花香)、反式-橙花叔醇(花香)、香叶醇(花香、果香、甜香)、植物醇(花香和香脂香气)、己酸己酯(果香)、月桂烯(甜橘味和香脂气)、2-苯乙醇(蜜香、花香)、芳樟醇(花香、甜香)、(Z)-芳樟醇氧化物(花香)、(E)-芳樟醇氧化物(花香、奶香、焦糖香)、苯甲醛(杏仁香)、环氧-β-紫罗兰酮(果香)等具有花果香的成分,及水杨酸甲酯(青草味、药味)、(Z)-β-法呢烯(青草香)等具有青草香的成分,以及(E)-3-己烯醇等具有清香的成分,这些物质共同构成了水仙茶的基础香型。
主成分分析是一种常用的“线性降维”方法,可初步了解样品间的相似性或差异性。本研究通过对所有样品的香气组分进行主成分分析(PCA),得到拟合了2个主成分的PCA模型。第一主成分(PC1)和第二主成分(PC2)占总方差的66.83%,其中PC1和PC2解释率分别为37.80%和29.03%,自然生长水仙毛茶(G1)和成品茶(G2)均位于PC2正半轴,修剪水仙毛茶(D1)和成品茶(D2)均位于PC2负半轴,二者分离明显,存在可视化差异,说明自然生长水仙和修剪水仙样品在挥发性化合物的组成上有明显差异(图2A)。同时,自然生长水仙毛茶(G1)和修剪水仙毛茶(D1)均在PC1负半轴,自然生长水仙成品茶(G2)和修剪水仙成品茶(D2)均位于PC1正半轴,说明焙火前后在挥发性化合物的组成上有明显差异。三维主成分分析也表明(图2B),自然生长水仙的毛茶和成品茶与修剪水仙相比均存在可视化差异。
从OPLS-DA模型的置换验证结果发现,建立的D1对比G1(D1 vs G1)的OPLS-DA模型共得到2个主成分,R2X=0.716,R2Y=1.000,Q2=0.986,这3个模型参数均大于0.5,OPLS-DA模型拟合准确较好(表2)。R2XR2YQ2三个指标越接近1,模型越稳定可靠。同样地,建立的D2 vs G2、D1 vs D2、G1 vs G2的OPLS-DA模型均是最优,其R2Y分别为0.998、0.999、0.999,均非常接近1,模型的解释能力强,同时其Q2分别为0.981、0.993、0.969,均为有效的模型。因此本研究所建立的OPLS-DA模型构建良好,预测性可靠且有意义,能够有效解释修剪水仙与自然生长水仙之间、焙火前后的挥发性物质差异,并可根据VIP值分析筛选差异挥发性物质。
基于OPLS-DA结果,采用将差异倍数值Fold Change与OPLS-DA模型的变量重要性投影VIP值相结合的方法,筛选修剪和自然生长水仙茶的差异挥发性物质。从D1和G1毛茶中共筛选出71种差异挥发性物质(图3)。其中,有41种挥发性物质在自然生长水仙茶的相对含量高于修剪水仙茶。
前20种差异物质表明(图4A),发现未知物1、4-己酸的相对含量,G1远高于D1,分别高出913.9倍(log2 FC为9.84)和25.1倍(log2 FC为4.65)。另外,己酸己酯(果香)、植物醇、异植醇(花香,药草和青香)、反式-橙花叔醇等具有花果香属性的物质G1也高于D1(图3)。而β-雪松烯、γ-杜松烯、2,5-二甲基嘧啶G1低于D1。
从D2和G2共筛选出71种差异挥发性物质(图5)。其中,G2有41种挥发性物质的相对含量高于D2,30个挥发性物质的相对含量低于D2。通过对比发现,未知物1、4-己酸的相对含量,G2远高于D2,这与2种水仙毛茶的结果一致(图4B)。除萜类外,酯类、酮类化合物大多也具备花果香。呈果香型的己酸己酯、己酸甲酯、反式-橙花叔醇、(Z)-β-法呢烯、2-庚酮(柑橘味)、1-庚醇(果香)等挥发物,呈烘烤香型的2-甲酰基-4,5-二甲基吡咯、乙基甲基吡嗪、3-乙基-1H-吡咯等挥发物,G2也高于D2(图5)。
自然生长水仙和修剪水仙毛茶对比组(D1 vs G1)和成品茶对比组(D2 vsG2)共有差异35个(图6A),其中,反式-橙花叔醇、4-甲基-1-戊醇、己酸己酯、丁酸己酯、紫苏烯、未知物1、4-己酸、丁酸在2个对比组中都是自然生长水仙更高。其中值得关注的是,未知物1和4-己酸在2种水仙焙火前后均有较高含量,同时也是差异比较大的物质。这些物质可能是修剪水仙与自然生长水仙茶叶香气品质差异的重要成分。
在焙火过程中发生氧化、脱水、糖化等作用,提高茶叶香气,降低了苦涩味,使其独具风韵[28]。从修剪水仙毛茶(D1)和成品茶(D2)共筛选出91种差异挥发性物质(图7)。其中,D2有64种挥发性物质的相对含量高于D1。其中,前20种差异物质表明,乙基甲基吡嗪、3-乙基-1H-吡咯、2,5-二甲基嘧啶等香气物质D2远高于D1(图4C);此外,D2相比于D1,呈花香的(Z)-芳樟醇氧化物(花香)、香叶酸、顺式香叶基丙酮(花香、玫瑰),呈烘烤香型的2-甲酰基-4,5-二甲基吡咯等挥发物也较高;β-雪松烯、(Z)-β-法呢烯、γ-杜松烯、癸醛(青气)、庚醛(青味)、己醛(清香、青草香、青气)等物质较低(图7)。
从G1和G2共筛选出70种差异挥发性物质(图8)。其中,有46种挥发性物质在G2的相对含量高于G1。其中,前20种差异物质表明,2,5-二甲基嘧啶、乙基甲基吡嗪、3-乙基-1H-吡咯、己酸甲酯等烘烤和果香型的挥发性物质,G2均远远高于G1,庚醛G2远远低于G1(图4D)。另外(图8),G2相比于G1,2-甲酰基-4,5-二甲基吡咯、乙基甲基吡嗪、2,5二甲基吡嗪、3-乙基-1H-吡咯更高,植物醇、异植醇、香叶醇、紫苏烯、癸醛、己醛等青草香、清香和花香型的成分略有下降。
修剪水仙毛茶与成品茶对比组(D1 vs D2)和自然生长水仙毛茶与成品茶对比组(G2 vs G2)共有差异物质45个(图6B),分别为(Z)-芳樟醇氧化物、2-甲酰基-4,5-二甲基吡咯、3-乙基-1H-吡咯、乙基甲基吡嗪、2,5-二甲基嘧啶等呈果香与烘烤物质均呈上升趋势。癸醛、紫苏烯、庚醛、己醛等物质的相对含量均呈下降趋势,这些物质主要呈现青草气,其含量在水仙成品茶中均下降。
采用OAV值来评价单个香气组分对茶叶整体香气的贡献度。前人研究表明,当OAV>1时,认为该挥发性物质对茶叶香气具有一定的影响,当OAV>10时,认为该挥发性物质对香气贡献极大[21]。通过已报道的挥发性物质的阈值计算OAV,本研究筛选计算出30个重要挥发性成分的OAV(表3),结果表明修剪和自然生长水仙的香气成分OAV值存在较大差异。其中(E)-3-己烯醇(清香)、香豆素(药草香)、吲哚的OAV>10 000,芳樟醇、反式-橙花叔醇、月桂烯的OAV>1000,香叶醇、4-甲基-1戊醇、辛烯-3-醇、癸醛、环氧-β-紫罗兰酮、(E)-芳樟醇氧化物、戊基呋喃的OAV>100,这些成分在相对含量和香气强度上表现都很突出,并且大多呈现出清香和花果香类型的气味。此外,2-苯乙醇、水杨酸甲酯、(+)-柠檬烯、β-环柠檬醛、己醛、顺式香叶基丙酮、3-辛烯-2-酮、2-庚酮、萘的OAV>10。其中,(E)-3-己烯醇、吲哚、芳樟醇、月桂烯、反式-橙花叔醇、环氧-β-紫罗兰酮、2-苯乙醇、水杨酸甲酯这些挥发性成分的相对含量均排在前60(表1);另外,通过对比发现(表3),自然生长水仙毛茶和成品茶的(E)-3-己烯醇、吲哚、反式-橙花叔醇、环氧-β-紫罗兰酮、(E)-芳樟醇氧化物、2-苯乙醇、水杨酸甲酯均比修剪水仙更高,因此,推测这些物质可能是自然生长水仙香气形成的重要物质,尤其是反式-橙花叔醇在修剪水仙和自然生长水仙的毛茶(D1 vs G1)和成品茶(D2 vs G2)的差异物质分析中均存在较大差异。庚醛只在毛茶中检测到,OAV为0.00~20.00,其对香气呈现青气有较大贡献。乙基甲基吡嗪只存在于水仙成品茶,OAV为0~119.38,推测它对水仙火功香的形成有重要作用。本研究筛选的其中22个挥发性物质(OAV>10)对自然生长水仙茶香气的形成具有较大作用。
通过检测修剪和自然生长水仙毛茶和成品茶的呈味物质,结果表明(表4),G1对比D1,EGCG、酯型儿茶素、咖啡碱(caffeine, CAF)含量显著更高;EGC、EC、ECG含量显著更低,儿茶素总量(total catechin, TC)和茶多酚总量(total tea polyphenols, TP)无显著差异。自然生长水仙成品茶(G2)对比修剪水仙成品茶(D2),EGCG、酯型儿茶素、儿茶素总量显著更高;咖啡碱(CAF)含量显著更低;EGC、EC、简单儿茶素和茶多酚总量无显著差异。
此外,G2对比G1,EGC、EGCG、ECG、简单儿茶素、酯型儿茶素、儿茶素总量、茶多酚总量、咖啡碱含量显著降低。D2对比D1,咖啡碱无显著差异,其他成分均与自然生长水仙成品茶和毛茶对比组(G2 vs G1)呈现相同的变化趋势。自然生长和修剪水仙焙火前后的儿茶素组分和总量、茶多酚变化趋势一致,呈下降趋势。
感官审评结果表明(表5),自然生长水仙毛茶和成品茶的品质均显著优于修剪水仙,其综合评分结果均更高;与修剪水仙毛茶和成品茶相比,自然生长水仙香气均更加馥郁持久、似有花果香,汤感更加醇厚、水中香显。成品茶的品质均显著优于毛茶,其综合评分结果也更高。水仙成品茶与毛茶相比,外形更加油润;汤色由金黄转为橙黄色,这是由于焙火增加引起茶黄素和茶红素进一步氧化聚合,导致茶汤汤色加深[29];修剪水仙香气由毛茶略带青气,转为清香显,滋味浓醇度也进一步加强。
在茶园栽培过程中,茶树管理方式可能通过调节茶鲜叶内一系列的生理生化变化来影响茶树的香气物质。茶青的质量直接影响成品茶香气和滋味的形成。BORA等[30]采用宏基因组学方法,对修剪和非修剪条件下茶树土壤微生物群落结构和功能特征进行了解析和比较,研究表明长期修剪茶园会降低土壤中微生物的丰度。RUBEL MOZUMDER等[31]研究了连续3 a在不同采摘季节和特定采摘时间采集的修剪后和未修剪茶树叶片的代谢特征,其代谢组学结果表明,修剪后和未修剪的茶树具有不同的代谢产物,存在显著的代谢差异。本研究表明,修剪水仙和自然生长水仙的香气物质、儿茶素及其组分等呈味物质、感官品质都存在较大差异,推测可能是由于自然生长水仙的物质代谢区别于修剪水仙。
乌龙茶香气是由茶树品种、加工工艺和生长环境共同决定,是呈香物质对人体产生的生物、物理和心理的刺激[32-33]。武夷水仙的香气物质主要由酯类、萜类、杂环化合物、烃类、芳烃类、酮类、醇类等组成。殷丽琼等[3]研究表明,春茶后轻修剪可提高茶树鲜叶产量,改善春茶品质,显著提升春季烘青和晒青的香气和滋味。本研究发现,感官审评中,自然生长水仙毛茶和成品茶的香气均表现为馥郁的天然花果香型,这与香气差异分析结果一致。对比修剪水仙的毛茶和成品茶,自然生长水仙的毛茶和成品茶均具有多种更高含量的花果香属性的挥发性物质。
茶汤滋味品质是茶汤中茶多酚、儿茶素组分、咖啡碱等呈味物质对人体感官综合作用的结果。本研究结果表明,修剪和自然生长水仙的儿茶素组分、咖啡碱含量存在显著差异。有研究表明,矮化修剪铁观音春茶茶多酚和咖啡碱含量均显著低于对照(对照茶树更高)[4],本研究中咖啡碱的含量,也是自然生长的水仙极显著高于修剪水仙。感官审评结果中,自然生长水仙的滋味多呈醇厚型。修剪水仙茶汤滋味浓厚,其滋味浓度的呈现可能与修剪后茶树的微域环境有很大关系。
综合武夷水仙香气成分、茶汤呈味物质和感官审评结果表明,自然生长水仙产品的品质优于修剪水仙产品。此外,基于OAV值对水仙的香气化合物进行分析,筛选出部分重要的特征香气物质,但由于已鉴定的挥发性物质中有部分化合物阈值缺乏,存在一定局限性;且对主要呈味物质的研究集中于儿茶素和茶多酚;后续研究可结合高灵敏度的电子鼻和高通量的代谢组学等技术对自然生长和修剪水仙的主要呈香和呈味物质进行分析,为进一步探究自然生长水仙的香型和滋味类型提供参考依据。本研究可为武夷岩茶茶园管理和品质调控提供理论和实践指导。
