To maintain the storage quality and prolong the storage period of postharvest bananas, the present study used ‘Guijiao No.6’ as the test material to investigate the 1-hexanol effect on the fruit quality and storability of the postharvest bananas under room temperature storage conditions. Banana fruits with intact fruit appearance and similar ripeness and size were selected and randomly divided into control and treatment groups. The control and treatment groups were treated with water and 0.1% (V/V) aqueous solution of 1-hexanol, respectively. Then, the bananas were removed to air-dry and packaged in polyethylene bags with a zipper. Finally, the samples were stored at 25 ℃ for 15 days. Changes in firmness, soluble solids, respiration intensity, condition index, malondialdehyde (MDA), membrane permeability, and PLD activity were measured every 3 days, and changes in PLDγ and PLDζ expression during storage were analyzed by quantitative real-time PCR (qRT-PCR). The results showed that 0.1% (V/V) 1-hexanol significantly (P<0.05) delayed the process of postharvest banana fruit softening, and the delaying effect was highly (P<0.01) significant on days 9 and 12. From day 9, 1-hexanol highly significantly inhibited the rise in total soluble solids and condition index. From day 6, 1-hexanol significantly inhibited respiration, where the peak respiration was reached on day 9 and was highly significantly reduced by 33.93% compared to the control. After day 12, compared with the control group, 1-hexanol highly significantly reduced the rise of MDA content and membrane permeability, by 26.70% and 62.20% on day 15, respectively. In addition, 1-hexanol significantly inhibited PLD activity, especially from 6 to 12 days, and the inhibition was highly significant (P<0.01). At day 15, compared with the control group, 1-hexanol highly significantly reduced the banana expression of PLDγ and PLDζ by 72.06% and 33.08%, respectively. In conclusion, 1-hexanol can effectively delay postharvest banana fruit softening, inhibit the respiration and PLD activity, and reduce postharvest fruit disease, thereby inhibiting total soluble solids, MDA production, and increased membrane permeability, and inhibiting PLDγ and PLDζ expression, thereby maintaining postharvest fruit storage quality and extending storage life. Therefore, 1-Hexanol can be used as a fruit preservative, which is not only important for maintaining the post-harvest fruit quality and the development of the banana industry, but also has good application prospects during post-harvest storage.

为保持采后香蕉贮藏品质,延长贮藏期限,以桂蕉6号香蕉为实验材料,探究正己醇在常温贮藏条件下对采后香蕉果实品质和耐贮性的影响。挑选果实外观完好及成熟度、大小相近的香蕉果实,随机分成对照组和处理组2组,分别用蒸馏水和0.1%浓度正己醇处理,取出晾干后置于PE保鲜袋中扎口包装,于25 ℃下贮藏15 d,每3 d取样观察测定香蕉果实的硬度、可溶性固形物(TSS)含量、呼吸强度、病情指数、丙二醛(MDA)含量、膜透性和PLD活性的变化,利用实时荧光定量PCR(qRT-PCR)分析贮藏期间PLDγPLDζ表达量的变化。研究结果显示:正己醇处理可显著延缓采后香蕉果实软化的进程(P<0.05),其中,9、12 d延缓效果极显著(P<0.01);在9 d后,正己醇处理极显著抑制TSS含量和病情指数的上升;6 d起,正己醇处理显著抑制呼吸强度,其中,9 d达到呼吸峰值,较对照组极显著降低33.93%;12 d后,与对照组相比,正己醇处理极显著降低MDA含量和膜透性的上升,分别于15 d降低了26.70%和62.20%。此外,正己醇处理显著抑制采后香蕉果实PLD活性,尤其是6~12 d,抑制效果极显著(P<0.01);在15 d,正己醇处理后香蕉PLDγPLDζ表达量较对照组分别极显著降低72.06%和33.08%。综合分析可知,正己醇处理可有效延缓采后香蕉果实软化,抑制果实呼吸强度和PLD活性,减轻采后果实病害发生,从而抑制TSS、MDA的产生和膜透性的增大,并下调PLDγPLDζ表达,进而维持采后果实贮藏品质和延长贮藏期限。因此,正己醇可作为一种水果保鲜剂,不仅对维持采后果实品质及香蕉产业的发展具有重要意义,且在采后贮藏期间具有良好的应用前景。

为保持采后香蕉贮藏品质,延长贮藏期限,以桂蕉6号香蕉为实验材料,探究正己醇在常温贮藏条件下对采后香蕉果实品质和耐贮性的影响。挑选果实外观完好及成熟度、大小相近的香蕉果实,随机分成对照组和处理组2组,分别用蒸馏水和0.1%浓度正己醇处理,取出晾干后置于PE保鲜袋中扎口包装,于25 ℃下贮藏15 d,每3 d取样观察测定香蕉果实的硬度、可溶性固形物(TSS)含量、呼吸强度、病情指数、丙二醛(MDA)含量、膜透性和PLD活性的变化,利用实时荧光定量PCR(qRT-PCR)分析贮藏期间PLDγPLDζ表达量的变化。研究结果显示:正己醇处理可显著延缓采后香蕉果实软化的进程(P<0.05),其中,9、12 d延缓效果极显著(P<0.01);在9 d后,正己醇处理极显著抑制TSS含量和病情指数的上升;6 d起,正己醇处理显著抑制呼吸强度,其中,9 d达到呼吸峰值,较对照组极显著降低33.93%;12 d后,与对照组相比,正己醇处理极显著降低MDA含量和膜透性的上升,分别于15 d降低了26.70%和62.20%。此外,正己醇处理显著抑制采后香蕉果实PLD活性,尤其是6~12 d,抑制效果极显著(P<0.01);在15 d,正己醇处理后香蕉PLDγPLDζ表达量较对照组分别极显著降低72.06%和33.08%。综合分析可知,正己醇处理可有效延缓采后香蕉果实软化,抑制果实呼吸强度和PLD活性,减轻采后果实病害发生,从而抑制TSS、MDA的产生和膜透性的增大,并下调PLDγPLDζ表达,进而维持采后果实贮藏品质和延长贮藏期限。因此,正己醇可作为一种水果保鲜剂,不仅对维持采后果实品质及香蕉产业的发展具有重要意义,且在采后贮藏期间具有良好的应用前景。

To maintain the storage quality and prolong the storage period of postharvest bananas, the present study used ‘Guijiao No.6’ as the test material to investigate the 1-hexanol effect on the fruit quality and storability of the postharvest bananas under room temperature storage conditions. Banana fruits with intact fruit appearance and similar ripeness and size were selected and randomly divided into control and treatment groups. The control and treatment groups were treated with water and 0.1% (V/V) aqueous solution of 1-hexanol, respectively. Then, the bananas were removed to air-dry and packaged in polyethylene bags with a zipper. Finally, the samples were stored at 25 ℃ for 15 days. Changes in firmness, soluble solids, respiration intensity, condition index, malondialdehyde (MDA), membrane permeability, and PLD activity were measured every 3 days, and changes in PLDγ and PLDζ expression during storage were analyzed by quantitative real-time PCR (qRT-PCR). The results showed that 0.1% (V/V) 1-hexanol significantly (P<0.05) delayed the process of postharvest banana fruit softening, and the delaying effect was highly (P<0.01) significant on days 9 and 12. From day 9, 1-hexanol highly significantly inhibited the rise in total soluble solids and condition index. From day 6, 1-hexanol significantly inhibited respiration, where the peak respiration was reached on day 9 and was highly significantly reduced by 33.93% compared to the control. After day 12, compared with the control group, 1-hexanol highly significantly reduced the rise of MDA content and membrane permeability, by 26.70% and 62.20% on day 15, respectively. In addition, 1-hexanol significantly inhibited PLD activity, especially from 6 to 12 days, and the inhibition was highly significant (P<0.01). At day 15, compared with the control group, 1-hexanol highly significantly reduced the banana expression of PLDγ and PLDζ by 72.06% and 33.08%, respectively. In conclusion, 1-hexanol can effectively delay postharvest banana fruit softening, inhibit the respiration and PLD activity, and reduce postharvest fruit disease, thereby inhibiting total soluble solids, MDA production, and increased membrane permeability, and inhibiting PLDγ and PLDζ expression, thereby maintaining postharvest fruit storage quality and extending storage life. Therefore, 1-Hexanol can be used as a fruit preservative, which is not only important for maintaining the post-harvest fruit quality and the development of the banana industry, but also has good application prospects during post-harvest storage.

香蕉(Musa nana L.)是世界四大水果之一,原产于东南亚,目前国内主产于广西、云南、海南及台湾等地[1]。成熟香蕉具有气味芬芳、果肉细腻的特点。其次,香蕉因富含蛋白质、VC、矿物质K等营养元素和多酚、胺类等活性成分,且具有抗氧化、预防心血管等保健功能,因此深受消费者喜爱[2]。另外,香蕉属于呼吸跃变型果实,采后出现呼吸跃变,乙烯释放量迅速上升,从而促进香蕉成熟和衰老。但由于香蕉在贮运、销售等过程中易产生微生物、机械伤、冷害等采后病害,尤其在贮运过程中常因振动产生的碰撞和挤压,使组织细胞膜系统和细胞结构遭到破坏,造成果实采后品质劣变,加速果实成熟和衰老,严重影响果实贮藏期限,降低商品价值[3]。因此,有效延长具有机械伤香蕉的贮藏保鲜期,保持其在贮藏期间的品质对香蕉产业的发展具有重要意义。
磷脂酶D(phospholipase D,PLD),全称磷脂酰胆碱磷脂水解酶,是催化生物膜主要成分磷脂水解的关键酶[4]。由于机械伤可使植物细胞膜完整性丧失,激活并促进PLD与膜结合而提高PLD活性,导致磷脂降解,膜通透性改变,最终造成植物伤害甚至死亡[5]。目前,随着人们生活水平提高,越来越注重果蔬采后品质,因此对果蔬采后保鲜技术的要求也不断提高。正己醇是植物脂氧合酶代谢途径中形成的代谢产物,也是细胞膜衰老代谢关键酶磷脂酶D的抑制剂,可用于果蔬采后保鲜[6-8]。李英华等[9-10]研究了不同浓度正己醇对草莓的保鲜效果,发现0.05%正己醇处理可有效抑制采后草莓呼吸作用和果实软化,降低果实采后腐烂率,较好地维持草莓VC含量;0.1%正己醇显著提高草莓在贮藏期间的抑菌能力,减少微生物病害,此外,正己醇处理还有效抑制草莓LOX活性,降低丙二醛(MDA)含量,提高POD活性。正己醇处理在桃[11]、桑葚[12]等作物中均较好地维持果实品质,延缓果实成熟衰老进程。
目前,正己醇对香蕉在机械伤胁迫后其生理特性的影响尚无报道。因此,研究正己醇对机械伤香蕉采后贮藏期间生理特性的影响,为维持机械伤香蕉采后品质和延长贮藏期限提供理论依据。
本试验所用香蕉品种为桂蕉6号,成熟度为7~8成熟,采自广西南宁市坛洛镇果园。采后立即运回实验室,将香蕉分成单根果指,挑选果实外观完好、成熟度一致、大小相近、无病虫害的果实为材料。用0.1%次氯酸钠溶液浸泡清洗10 min,取出晾干备用。
正己醇(1-hexanol)购自中和化学(山东)有限公司;磷酸氢二钠、磷酸二氢钠、乙酸乙酯、NaOH等试剂均为国产分析纯。
本试验所用仪器设备:BSA124s型电子天平、GY-4型果实硬度计、UV-3200 PCS型紫外可见分光光度计、3-18ks型高速冷冻离心机。
将预处理的香蕉果实进行模拟机械损伤处理,采用不锈钢打孔器(直径为2 mm)在香蕉中轴部位戳伤。随后将处理好的香蕉随机分成2组,每组60个香蕉果实,分别用蒸馏水(对照组)和(正己醇处理组)0.1%浓度正己醇浸泡处理果实10 min,然后自然晾干。晾干后的香蕉置于PE保鲜袋中扎口包装,于25 ℃室温下贮藏15 d,每3 d取样观察并测定相关指标,试验重复3次。
使用GY-4型果实硬度计测定(探头直径为5 mm),每次随机取6~8个香蕉,每个果面测2个点,最终取平均值为该果实的硬度。
使用MZB-53(N)便携式数显糖度计测定,取去皮香蕉的中部果肉进行切块打浆,重复测6次,最终取平均值为果实的TSS含量。
呼吸强度测定参照曹健康等[13]的方法,采用静置法进行测定。
参考吴小建等[14]的方法并加以改进,根据香蕉果实表面腐烂面积大小分为0~4级。分级标准为:0级果(无病斑)、1级果(<1/10病斑)、2级果(1/10~1/4病斑)、3级果(1/4~1/2病斑)、4级果(>1/2病斑)。每次取样测量香蕉表面病斑的大小。

病情指数=Σ(腐烂级别×该级别样品数量)/(最高级别×样品总数量)×100%

参照刘瑶等[15]的方法,采用硫代巴比妥酸法测定450、532、600 nm处的吸光值。
参考SONG等[16]的方法,用相对电导率表示膜透性,并稍作修改。将样品煮沸10 min并冷却至室温,测定相对电导率。
参照SUN等[17]和CHEN等[18]的方法,取磨样后的香蕉果肉组织1 g,测定PLD活性及蛋白质含量,单位用U/g表示。
按照RNAprep Pure Plant Kit试剂盒说明书,提取香蕉果皮中轴组织总RNA。取总RNA 1 μg使用HiFiScript cDNA试剂盒反转录合成cDNA第一链,用于后续实时荧光定量分析[17]。根据PLD基因序列,使用Premier 5.0软件设计基因特异性引物(表1),由生工生物工程(上海)股份有限公司合成引物。根据Real time PCR Master Mix(SYBR Green)试剂盒说明书进行RT-qPCR。以香蕉Actin基因为内参基因[19-20],采用2–ΔΔCT法计算PLDγPLDζ基因的相对表达量。
采用Excel 2010软件对实验数据进行统计;利用SPSS 26.0软件Duncan’s法对数据进行差异显著性分析;使用Origin 2018软件绘图。
硬度是反映果实采后软化程度的重要指标[21]。如图1所示,采后贮藏过程中,香蕉硬度整体呈下降趋势。贮藏0 d,果实硬度最大,为378.89 N。至贮藏3 d,2组硬度变化不明显。随贮藏时间增加,果实硬度逐渐降低。贮藏6 d时,处理组硬度为262.38 N,在贮藏15 d降至最低值165.25 N,较对照组显著提高71%(P<0.05)。贮藏15 d,对照组和处理组较贮藏6 d分别降低65.5%和37%,表明正己醇处理可延缓香蕉果实软化的进程。
可溶性固形物(TSS)可反映果蔬的成熟度和品质状况。如图2所示,采后贮藏过程中,香蕉TSS含量整体呈上升趋势。贮藏6 d前,2组的TSS含量无显著差异。贮藏9 d,对照组和处理组的TSS含量分别为9.35%和6.07%。贮藏15 d,香蕉TSS含量积累至最大值,处理组为16.27%,较对照组降低20%(相对值)。贮藏9 d后,处理组TSS含量极显著低于对照组(P<0.01),表明PLD抑制剂正己醇在一定程度上降低了香蕉TSS含量,维持果实品质,延缓采后贮藏期限。
图3所示,采后贮藏过程中,香蕉呼吸强度整体呈先上升后下降的趋势。贮藏0 d,香蕉呼吸强度为2.17 mg/(kg·h),随后逐渐上升。贮藏6 d,处理组呼吸强度为7.74 mg/(kg·h),显著低于对照组。对照组和处理组均于9 d出现呼吸峰,处理组呼吸强度较对照组极显著降低33.93%,随后下降。贮藏9~15 d,对照组呼吸强度由14.62降至8.27 mg/(kg·h),处理组由9.66降至6.56 mg/(kg·h),处理组显著低于对照组(P<0.05)。采后贮藏期间,处理组呼吸强度均低于对照组,其中6~15 d降低效果显著,表明正己醇处理可有效抑制采后香蕉贮藏期间呼吸强度上升,延缓果实成熟和衰老的进程。
病情指数可反映采后贮藏期间果蔬的病害情况,是衡量果实品质的重要指标[22-23]。如图4所示,随贮藏时间增加和果实逐渐成熟,香蕉病情指数呈上升的趋势。贮藏0 d,香蕉病情指数为0,贮藏6 d,处理组病情指数为1.22%,显著低于对照组。贮藏9~15 d,处理组病情指数由11.59%升至35.88%,极显著低于对照组。贮藏15 d,香蕉病情指数最大,处理组病情指数为35.88%,较对照组极显著降低54.07%。综上,正己醇处理可有效抑制贮藏期间香蕉果实病斑的扩大和增加,降低采后果实病情指数。
MDA是膜脂过氧化的产物,加速采后果实衰老进程。如图5所示,采后贮藏期间,香蕉MDA含量整体呈上升的趋势。贮藏9 d,对照组MDA含量为2.4 nmol/g,显著低于对照组,随后于15 d迅速增至最大值6.75 nmol/g。采后贮藏期间,处理组MDA含量较对照组推迟3 d上升,贮藏12 d,处理组MDA含量为2.25 nmol/g,较对照组极显著降低55.00%,随后于贮藏15 d迅速增至最大值4.95 nmol/g,较对照组极显著降低26.70%。综上,PLD抑制剂正己醇在采后贮藏过程中可抑制MDA生成,延缓果实衰老。
图6所示,随贮藏时间增加,香蕉的相对电导率整体呈上升趋势。贮藏9 d,对照组和处理组的相对电导率分别为22.88%和15.75%,随后处理组相对电导率于贮藏12 d升至24.56%,较对照组极显著下降(P<0.01)。贮藏15 d,香蕉相对电导率达到最大值,处理组的电导率为33.84%,较对照组极显著降低62.20%(P<0.01)。结果分析表明,贮藏9 d后,处理组的相对电导率和MDA含量均低于同期对照组,说明PLD抑制剂正己醇可有效抑制香蕉膜脂过氧化反应,延缓果实贮藏期限。
磷脂酶D(PLD)是催化磷脂水解的关键酶,在逆境胁迫下被激活的PLD可降解果实的细胞膜,导致膜透性增大,促进果实老化,降低果实品质[24]。如图7所示,采后贮藏过程中,对照组PLD含量呈逐渐上升的趋势,处理组呈先上升后下降再上升的趋势。对照组PLD活性于贮藏15 d增至最大值590.13 U/g。贮藏3 d处理组PLD活性为456.25 U/g,显著低于对照组(P<0.05),随后于6 d降至407 U/g,较对照组极显著降低17.4%(P<0.01)。贮藏15 d,处理组PLD活性增至最大值531.88 U/g,较对照组显著降低10%(P<0.05)。处理组PLD活性在采后贮藏期间显著低于对照组(P<0.05),表明正己醇处理可有效抑制PLD生成,维持果实品质,延缓贮藏期限。
图8所示,香蕉PLDγ表达量整体呈上升的趋势。贮藏0~12 d,对照组和处理组的PLDγ表达量均较低,且两组间的PLDγ表达量无显著差异。贮藏15 d,香蕉PLDγ表达量达到峰值,处理组PLDγ表达量为5.39,较对照组极显著降低72.06%;对照组和处理组PLDγ表达量较12 d上升11.18倍和2.68倍。综上,PLD抑制剂正己醇可抑制采后香蕉PLDγ表达,降低采后果实的膜脂代谢,延长贮藏期限。
图9所示,香蕉PLDζ表达量在贮藏过程中呈先下降后上升的趋势。贮藏第3天,处理组PLDζ表达量降至0.41,较对照组显著降低46.75%(P<0.05),随后上升。随贮藏时间增加,香蕉PLDζ表达量于15 d达到峰值,处理组PLDζ表达量为1.78,较对照组极显著降低33.08%(P<0.01)。综上,PLD抑制剂正己醇可抑制采后香蕉PLDζ表达,降低采后果实的膜脂代谢,延长贮藏期限。
硬度是果实采后品质的重要指标之一,可反映果实采后的耐贮性。本研究结果表明,随贮藏时间增加,香蕉果实硬度不断降低,但与对照组相比,正己醇处理可有效抑制香蕉果实软化。采后贮藏期间,香蕉果实中的淀粉降解转化成葡萄糖,使果实TSS含量累积增大,表现为果实采后成熟度变大而衰老,因此TSS含量可反映果实的营养价值和果实品质[25-26]。本研究结果显示,贮藏期间香蕉果实TSS含量呈上升趋势,正己醇处理在贮藏后期极显著抑制了香蕉可溶性固形物含量上升(P<0.01),从而维持采后香蕉贮藏品质,延长果实货架期。李英华[27]研究发现,正己醇处理显著延缓采后草莓果实软化、抑制MDA累积,但对草莓TSS含量的影响不显著。
呼吸强度和病情指数是衡量果实采后贮藏期间的重要指标[22,28]。本研究结果表明,与对照组相比,正己醇处理在贮藏期间可有效抑制采后香蕉果实呼吸强度和病情指数的上升,维持果实贮藏品质,其中,贮藏后期抑制效果极显著(P<0.01)。霍宪起[12]研究了不同浓度正己醇对采后桑葚的影响,发现1.5 mL/L和1.0 mL/L正己醇抑制采后桑葚呼吸强度的效果最佳。
丙二醛(MDA)是膜脂过氧化的产物,可反映膜脂过氧化的程度[29]。果实采后的成熟衰老与膜脂过氧化反应密切相关,机械伤等非生物性逆境胁迫会促进膜脂过氧化反应,造成膜透性增大,电解质外渗,因此相对电导率可间接反映果实的膜透性[30-32]。本研究结果显示,贮藏0 d时,香蕉MDA含量和相对电导率均较低,说明此时香蕉细胞膜完整性良好,过氧化程度不大。随贮藏时间增加,果实逐渐成熟衰老,香蕉MDA含量和相对电导率呈上升趋势,正己醇处理组MDA含量和相对电导率在贮藏后期显著低于对照组(P<0.05),说明果实的成熟衰老会引起果实膜透性的改变,正己醇处理可有效抑制果实MDA含量上升和膜透性的增大,从而延缓果实衰老的进程。
当果实衰老或遭受机械损伤等逆境胁迫时,会引起细胞膜发生膜脂代谢,促进果实衰老[33]。磷脂是细胞膜的重要组成成分,PLD是膜脂代谢过程中的关键酶,被激活的PLD可催化磷脂水解,进而破坏膜结构和膜功能,因此,PLD的活性可反映磷脂的降解程度。蟠桃[11]、青枣[34]和南果梨[35]的研究中表明,PLD活性上升,膜脂代谢变快,说明PLD在膜脂代谢中有重要作用。
本研究结果显示,正己醇处理显著抑制了PLD活性的上调(P<0.05),与对照组相比,正己醇处理在贮藏结束时极显著抑制了PLDγPLDζ的表达量(P<0.01),表明PLD抑制剂正己醇可有效延缓采后香蕉果实膜脂代谢的进程。李银[11]研究发现,正己醇和正己醛处理均可抑制PLD活性,提高抗氧化酶系活性,延缓果实软化,维持果实采后品质。
综上,正己醇处理可有效抑制采后贮藏期间香蕉果实的呼吸作用,减轻果实病情指数,从而延缓香蕉果实软化的进程和减慢TSS的产生;另一方面,采后正己醇处理能有效抑制PLD活性,从而降低MDA含量和膜透性增大,并下调PLDγPLDζ表达。因此,正己醇处理能够降低机械损伤对采后果实生物膜的影响,保护膜结构和膜功能,延长采后果实的贮藏期限,是维持机械伤香蕉采后贮藏品质的新方法。但正己醇处理延缓果实衰老的机理是否与果实本身抗氧化相关酶活性的提高有关,这有待于进一步研究。
