Phytochelatin synthase (PCS) is a key enzyme that catalyzes the synthesis of phytochelatin (PC). PC can reduce the toxicity of heavy metals to plants through complexation. Heavy metal pollution in soil increases the risk of cassava food safety. It is of great significance to study the reduction and control of heavy metals in cassava by analyzing the function of MePCS1 gene. In this study, the coding region of MePCS1 gene in cassava variety SC8 was cloned by PCR. The full length was 1512 bp, encoding 503 amino acids. Protein sequence analysis showed that MePCS1 protein was a hydrophilic protein, containing 41 phosphorylation sites and 2 glycosylation sites, without signal peptide. The phylogenetic tree showed that the cassava MePCS1 protein had the closest relationship with the PCS protein of the rubber tree. The qPCR analysis showed that the MePCS1 gene had the highest expression level in the stem. During the development of cassava roots, the expression level of MePCS1 gene was the highest in the root enlargement period. Lead (Pb) stress induced MePCS1 gene expression. MePCS1 gene can improve the tolerance of BY4741 yeast to Pb. The results of this study provide new information for further analysis of the functional characteristics of the MePCS1 gene and its mechanism for reducing Pb in cassava tuber roots.

植物络合素合酶(PCS)是催化植物络合素(PC)合成的关键酶,PC可通过络合作用减轻重金属对植物的毒害。土壤重金属污染增加木薯食用安全风险,对木薯络合素合酶基因MePCS1进行功能分析,对木薯重金属减控研究具有重要意义。本研究通过PCR技术克隆了SC8木薯品种的MePCS1基因的编码区,全长为1512 bp,共编码503个氨基酸。蛋白序列分析表明:MePCS1蛋白为亲水性蛋白,含有41个磷酸化位点和2个糖基化位点,不含有信号肽。构建系统进化树发现,木薯MePCS1蛋白与橡胶树的PCS蛋白亲缘关系最近。利用qPCR技术分析表明,MePCS1基因在茎中表达量最高;在木薯块根发育过程中,MePCS1基因在块根膨大期的表达量最高;铅(Pb)胁迫诱导MePCS1基因表达。MePCS1基因能提高BY4741酵母对Pb的耐受能力。本研究结果为进一步解析MePCS1基因功能特性及其对木薯的Pb减控机制提供新的信息。

植物络合素合酶(PCS)是催化植物络合素(PC)合成的关键酶,PC可通过络合作用减轻重金属对植物的毒害。土壤重金属污染增加木薯食用安全风险,对木薯络合素合酶基因MePCS1进行功能分析,对木薯重金属减控研究具有重要意义。本研究通过PCR技术克隆了SC8木薯品种的MePCS1基因的编码区,全长为1512 bp,共编码503个氨基酸。蛋白序列分析表明:MePCS1蛋白为亲水性蛋白,含有41个磷酸化位点和2个糖基化位点,不含有信号肽。构建系统进化树发现,木薯MePCS1蛋白与橡胶树的PCS蛋白亲缘关系最近。利用qPCR技术分析表明,MePCS1基因在茎中表达量最高;在木薯块根发育过程中,MePCS1基因在块根膨大期的表达量最高;铅(Pb)胁迫诱导MePCS1基因表达。MePCS1基因能提高BY4741酵母对Pb的耐受能力。本研究结果为进一步解析MePCS1基因功能特性及其对木薯的Pb减控机制提供新的信息。

Phytochelatin synthase (PCS) is a key enzyme that catalyzes the synthesis of phytochelatin (PC). PC can reduce the toxicity of heavy metals to plants through complexation. Heavy metal pollution in soil increases the risk of cassava food safety. It is of great significance to study the reduction and control of heavy metals in cassava by analyzing the function of MePCS1 gene. In this study, the coding region of MePCS1 gene in cassava variety SC8 was cloned by PCR. The full length was 1512 bp, encoding 503 amino acids. Protein sequence analysis showed that MePCS1 protein was a hydrophilic protein, containing 41 phosphorylation sites and 2 glycosylation sites, without signal peptide. The phylogenetic tree showed that the cassava MePCS1 protein had the closest relationship with the PCS protein of the rubber tree. The qPCR analysis showed that the MePCS1 gene had the highest expression level in the stem. During the development of cassava roots, the expression level of MePCS1 gene was the highest in the root enlargement period. Lead (Pb) stress induced MePCS1 gene expression. MePCS1 gene can improve the tolerance of BY4741 yeast to Pb. The results of this study provide new information for further analysis of the functional characteristics of the MePCS1 gene and its mechanism for reducing Pb in cassava tuber roots.

木薯(Manihot esculenta Crantz),又称树薯、南洋薯、木番薯,为大戟科多年生作物,是世界上10亿人口的主要食物[1]。随着人类工业化进程的发展,土壤重金属污染已经成为全球的重大问题,危害着人类的身体健康和作物的粮食安全。木薯作为热带地区重要的经济作物与粮食作物,具有淀粉含量高、耐贫瘠等特点。块根为木薯淀粉的主要贮藏器官,木薯受到重金属污染后,金属离子容易在木薯块根中积累,导致木薯生长受限,产量降低。因此解析木薯响应重金属关键的基因,减少重金属在木薯块根的积累,对创制木薯新种质、提高木薯食用的安全性具有十分重要的意义。
植物受到金属胁迫后,植物络合素(phytochelatins, PC)可与金属结合形成较稳定的复合物,减轻金属的毒害。植物络合素的结构一般为(γ-Glu-Cys)n-X(n=2~11,X为Gly、Clu、Cln、Ser和β-Ala的形式存在),以(γ-Glu-Cys)n-Gly的结构较为常见,其中半胱氨酸Cys中(-SH)巯基可以与重金属结合具有解毒功能。植物络合素合酶(phytochelatin synthases, PCS)可催化GSH合成PC。目前对拟南芥[2]、豆类[3]、烟草[4]、水稻[5]的PCS研究较为深入,过表达PCS基因可增强植物对重金属的耐受性。通过分析不同物种中PCS基因的启动子,发现其含有多个顺式作用的元件,说明PCS基因的表达受多种因素调控[6]。如转录因子MYB40可以诱导AtPCS1的表达,在As2+胁迫条件下拟南芥中AtMYB40被诱导表达,AtMYB40正调控AtPCS1表达[7];水稻中Hg2+和Pb2+金属离子可以诱导OsPCS7表达,Cd2+与Zn2+金属离子可以诱导OsPCS9表达[8];番茄叶片在Pb胁迫下,SlPCS1基因表现出较高的表达水平[9]
为了研究木薯MePCS1基因在金属胁迫下的功能,本研究克隆了木薯MePCS1基因,并对其进行生物信息学分析、基因表达的组织特异性分析、不同发育时期的块根表达模式分析、重金属Pb胁迫条件下的表达分析以及在酵母中的功能验证分析。为进一步研究MePCS1基因参与金属Pb胁迫的耐受性,减少重金属胁迫对木薯的危害提供新的依据。
供试材料华南8号木薯(SC8)由中国热带农业科学院提供;酵母感受态BY4741、大肠杆菌感受态DH5α均购自上海唯地生物技术有限公司;RNA提取试剂盒购自成都福际生物技术有限公司;MonScript™ RTIII Super Mix with dsDNase(Two-Step)反转录试剂盒购自莫纳生物科技有限公司;PrimeSTAR HS(Premix)DNA Polymerase高保真酶购自TaKaRa公司。
提取SC8木薯总RNA,利用反转录试剂盒将总RNA反转录成cDNA。根据木薯MePCS1基因CDS区设计特异性引物(表1),以SC8木薯cDNA为模板,利用PrimeSTAR HS(Premix)DNA Polymerase高保真酶扩增MePCS1基因。反应体系为:PrimeSTAR HS(Premix)DNA Polymerase 25 μL,MePCS1-F/R引物1 μL,cDNA模板2 μL,ddH2O补充至总体积为50 μL。反应程序为:94 ℃预变性4 min;98 ℃变性20 s,61 ℃退火30 s,72 ℃延伸1 min 40 s,35个循环。PCR产物于4 ℃下保存。
对MePCS1的蛋白序列进行生物信息学分析,所用软件见表2
以田间种植6个月的SC8木薯块根、须根、茎、叶柄、成熟叶、嫩叶、顶芽、腋芽等8个组织部位为材料,提取其总RNA,反转录成cDNA,荧光定量PCR(qRT-PCR)分析MePCS1基因的表达模式。基于本实验室前期的SC8木薯块根形成期(种植后的80 d)、块根膨大期(种植后的130、180 d)、块根成熟期(种植后的230、280 d)的转录组数据,对MePCS1基因在块根不同发育时期的表达模式进行分析。
利用1.2 mmol/L Pb(NO3)2处理种植60 d的SC8木薯组培苗移栽幼苗,分别胁迫处理0、3、6、12、24、48 h。提取处理后材料的总RNA反转录成cDNA,荧光定量PCR(qRT-PCR)分析MePCS1基因的表达模式,反应体系和程序参照LU等[10]的方法。使用SPSS 26软件进行邓肯单因素多样本差异显著性方差分析。
(1)MePCS1基因酵母表达载体构建。以扩增出MePCS1基因PCR产物为模板,根据pYES2载体多克隆位点与MePCS1基因CDS序列设计含有同源重组接头引物PYES2-MePCS1-F/R(表1),通过PCR将重组接头引入该基因CDS区的两端,纯化回收目的片段。利用同源重组方法将纯化MePCS1目的片段与线性化的酵母表达载体pYES2进行重组,利用pYES2载体检测引物PYES2-检测-F/R进行菌液PCR鉴定,选取阳性的大肠菌液进行测序。将测序比对正确的阳性克隆菌液进行扩大培养提取重组质粒,新质粒命名pYES2-MePCS1。将重组质粒转化到酵母感受态BY4741中,PCR鉴定选出阳性菌斑,用于后续实验。
将含有pYES2-MePCS1酵母菌液与空载pYES2酵母菌液分别在SD-Ura液体培养基中扩大培养后,将初始OD600调至0.4,并逐级稀释5个浓度梯度(10-1、10-2、10-3、10-4、10-5);各取4 μL在含有50 mmol/L Pb2+的SD-Ura(含有2%葡萄糖)固体培养基上进行点板,30 ℃倒置培养3 d,观察含pYES2-MePCS1与空载pYES2酵母菌的生长情况。
通过Blast搜索木薯基因组数据库,获得木薯MePCS1基因序列,设计特异性引物克隆该基因的编码区(CDS)。以SC8木薯组培苗的cDNA为模板,PCR扩增获得预期大小目的条带(图1)。将目的条带进行Sanger测序,发现MePCS1基因的CDS序列全长为1512 bp,共编码503个氨基酸。
对木薯MePCS1蛋白的基本理化性质分析发现,其原子组成为C2456H3890N670O730S33,理论等电点为6.04,分子质量为(Mw)55.54 kDa。该蛋白含有20种氨基酸残基,Leu(L)的含量最高(58个),占11.5%;Trp(W)的含量最低(8个),占1.6%。在氨基酸正/负总残基中,带负电荷残基(Asp+Glu)总数为61个,带正电荷残基(Arg+Lys)总数为53个。
MePCS1蛋白的总亲疏水性平均值为0.136,推测该蛋白为亲水性蛋白。磷酸化位点预测分析结果发现,该蛋白含有41个磷酸化位点,其中Ser位点含量较高,为27个。糖基化位点预测发现,MePCS1蛋白可能含有2个糖基化位点。信号肽分析发现,MePCS1含有信号肽的可能性为0.0021,因此推测该蛋白不含有信号肽。
为了分析MePCS1蛋白与不同物种的PCS蛋白的亲缘关系,使用DNAMAN软件分析MePCS1与橡胶树(Hevea brasiliensis, XP_021678313.2)、麻疯树(Jatropha curcas, XP_012085275.1)、蓖麻(Ricinus communis, XP_015581239.2)、乌桕(Triadica sebifera, ARV78463.1)、南欧大戟(Euphorbia peplus, WCJ38289.1)、杨柳(Salix brachista, kAB5527907.1)、毛杨果(Populus trichocarpak, AI5566116.1)、毛白杨(Populus tomentosa, kAG6750280.1)、大枣(Ziziphus jujuba, XP_015891301.1)、苦楝(Melia azedarach, kAJ4726927.1)、柑橘(Citrus sinensisk, DO60421.1)、芒果(Mangifera indica, XP_044491381.1)、长叶黄麻(Corchorus olitorius, OMP11136.1)、伞花草(Herrania umbratica, XP_021281249.1)、木槿(Hibiscus syriacus, XP_039005780.1)、棉花(Gossypium barbadense, kAB2057009.1)等其他16个物种的PCS蛋白序列进行比对,其氨基酸序列一致性为90.1%~78.9%,其中N端区域高度保守(图2)。MePCS1蛋白与橡胶树PCS蛋白的同源性最高,达到90.1%。使用MEGE软件进行1000次的统计检验,构建系统进化树。结果表明,木薯MePCS1蛋白与橡胶树的PCS蛋白亲缘关系较近,其次为麻疯树、蓖麻、乌桕等植物,同棉花、木槿等植物的PCS蛋白亲缘关系较远(图3)。
采用荧光定量PCR技术分析MePCS1基因在木薯不同组织中的表达情况。结果表明,木薯MePCS1基因在茎中的表达量最高,在块根、须根、叶柄、成熟叶、嫩叶、顶芽、腋芽中的表达量无显著差异(图4)。
基于实验室前期的块根发育不同时期转录组数据,分析MePCS1基因在块根中的表达模式。结果表明,MePCS1基因主要在块根膨大期(种植后的130、180 d)的表达量最高,在成熟块根期(种植后的230、280 d)的表达量最低(图5)。
为了探究MePCS1基因表达对Pb胁迫的响应,本研究采用1.2 mmol/L Pb(NO3)2胁迫处理木薯SC8幼苗,并通过荧光定量PCR分析根系中MePCS1基因在处理0、3、6、12、24、48 h时的表达变化。结果发现,MePCS1基因快速响应在Pb2+胁迫,3 h的表达量显著高于对照组,且在6和12 h的表达量逐步提高,然后24 h和48 h的表达量显著降低(图6)。此结果表明MePCS1基因参与木薯对Pb2+胁迫的响应。
为了验证MePCS1基因的功能,本研究通过PCR将重组接头引入该基因编码区的两端(图7A),并通过同源重组将MePCS1与线性化的酵母表达载体pYES2连接。连接产物转化大肠,菌液PCR筛选阳性克隆,获得正确的pYES2-MePCS1质粒(图7B)。将pYES2-MePCS1质粒转化BY4741酵母,在SD-Ura固体培养基中培养获得单菌落后进行酵母菌斑PCR鉴定。结果发现PCR扩增获得与预期片段大小相同的条带(约1500 bp),表明pYES2-MePCS1质粒成功转化到BY4741酵母中(图7C)。
将含有pYES2-MePCS1、pYES2质粒的酵母菌液,梯度稀释后点在含有Pb2+的SD-Ura(含有2%半乳糖)固体培养基上,分析酵母的生长情况。结果发现,在不含有Pb2+的培养基上,2种酵母菌的长势一致,而在含有50 mmol/L Pb2+的培养基上,含有pYES2-MePCS1的酵母菌株长势优于空载pYES2酵母菌株(图8)。结果表明MePCS1基因可提高酵母对Pb2+的抗性。
Pb是一种重金属污染物,对植物和人体均有严重危害。在植物体内,Pb可以抑制多种酶的活性,干扰光合作用和水分吸收,导致生长受阻和生理功能紊乱[11]。此外,Pb还可以通过干扰矿质营养吸收,影响植物的营养平衡[12]。对人体而言,Pb主要通过食物链累积,长期摄入会对中枢神经系统产生毒性,特别是对儿童可能导致学习障碍和身心活动受限等问题[13]。Pb还可以损害肾脏功能,增加心血管疾病风险[14]PCS1基因是植物体内合成植物螯合素的关键酶,在减轻Pb的毒性效应方面具有重要作用[15]。本研究克隆了木薯MePCS1基因,基因编码区为1512 bp,共编码503个氨基酸。生物信息学分析表明,MePCS1蛋白为亲水性蛋白,无跨膜结构且不含有信号肽。木薯MePCS1蛋白与橡胶树、麻风树PCS蛋白的亲缘关系较近,以上3个物种均为大戟科植物。MePCS1基因主要在木薯茎中表达,以及在块根形成期和块根膨大期表达。植物受到Pb、Cd等重金属胁迫时,PCS1基因的表达通常会上调,以增强植物对重金属的耐受性。Cd胁迫可诱导雀稗PvPCS1PvPCS2[16]、大麻BnPCS1基因表达上升[17]。蓝藻CaPCS基因被AS胁迫诱导[18];马铃薯StPCS受重金属Pb、Cd、Cu、Zn诱导表达[19];Pb胁迫木薯幼苗发现,胁迫6、12 h后MePCS1基因的表达量显著上升,24 h后表达量降低,表明该基因的表达受Pb胁迫的诱导,参与木薯对Pb胁迫的响应。
在酵母细胞中进行异源表达是验证植物重金属胁迫响应相关基因功能的有效手段。水稻金属伴侣OsHIPP17、OsHIPP19可增强酵母对Cd的耐受性[20];苦荞麦FtPCSFtMTPC2FtPCSLFtVCE1aFtNramp3可增强酵母对Pb的耐受性[21]。本研究将MePCS1基因克隆到pYES2酵母表达载体,并转化到酵母菌株BY4741,分析转基因酵母对Pb胁迫(50 mmol/L)的耐受性。结果发现MePCS1基因可提高酵母对金属Pb的耐受能力,因此推测MePCS1基因可提高木薯对Pb胁迫的抗性。在未来的研究工作中,将利用过量表达和基因编辑技术,进一步鉴定MePCS1基因功能特性,并筛选低Pb积累的木薯新种质,以提高木薯食用的安全性。
