The level of cyanogenic glucosides content is an important trait for the edible quality of cassava, and Me-CYP79D2 is one of the key enzyme genes regulating cassava cyanogenic glucosides synthesis. This study used the edible variety SC9 as the material, cloned the MeCYP79D2 gene in the cytochrome P450 enzyme family of cassava, analyzed it using bioinformatics software, and detected its expression characteristics in different cassava germplasm tubers using real-time quantitative PCR. The cyanogenic glucosides content was measured using HPLC, and its correlation with the expression level of MeCYP79D2 gene was analyzed. The results showed that the length of the coding region sequence of MeCYP79D2 gene was 1625 bp, encoding 528 amino acids. It belonged to an unstable lipophilic hydrophilic protein with 38 phosphorylation sites. Sequence alignment indicated that the gene was highly conserved, and phylogenetic analysis showed that MeCYP79D2 was closely related to rubber and leprosy trees. There was a significant positive correlation between the expression of MeCYPD2 gene and the content of cyanogen glycosides in cassava roots of germplasms with different levels of cyanogen glycosides. After treatment with methyl jasmonate, the expression of MeCYP79D2 gene in roots was significantly upregulated and the content of cyanogenic glucosides was significantly increased. The results of this study indicates that MeCYP79D2 is involved in cyanogenic glucosides synthesis, providing a theoretical basis for studying the molecular mechanism of cassava cyanogenic glucosides biosynthesis.

氰苷含量是木薯改良的重要靶标性状,MeCYP79D2是木薯氰苷合成的限速酶基因之一。本研究以食用木薯品种SC9为材料,克隆木薯细胞色素P450酶家族中的MeCYP79D2基因,通过生物信息学软件对其进行分析,利用实时荧光定量PCR检测其在不同木薯种质块根中的表达特性,采用高效液相色谱仪测定氰化物的含量,并分析其与MeCYP79D2基因表达量的相关性。研究结果显示:MeCYP79D2基因编码区序列长度为1625 bp,编码528个氨基酸,属于不稳定脂溶性亲水蛋白,具有38个磷酸化位点;序列比对表明该基因保守性较高;系统进化分析显示MeCYP79D2与橡胶及麻风树关系较近;氰苷含量不同的木薯种质资源块根中MeCYP79D2基因表达量与氰苷含量存在极显著正相关;茉莉酸甲酯处理后,根中MeCYP79D2基因的表达显著上调且氰苷含量显著升高。本研究结果表明MeCYP79D2参与氰苷合成,为解析木薯氰苷生物合成的分子机制研究提供理论基础。

氰苷含量是木薯改良的重要靶标性状,MeCYP79D2是木薯氰苷合成的限速酶基因之一。本研究以食用木薯品种SC9为材料,克隆木薯细胞色素P450酶家族中的MeCYP79D2基因,通过生物信息学软件对其进行分析,利用实时荧光定量PCR检测其在不同木薯种质块根中的表达特性,采用高效液相色谱仪测定氰化物的含量,并分析其与MeCYP79D2基因表达量的相关性。研究结果显示:MeCYP79D2基因编码区序列长度为1625 bp,编码528个氨基酸,属于不稳定脂溶性亲水蛋白,具有38个磷酸化位点;序列比对表明该基因保守性较高;系统进化分析显示MeCYP79D2与橡胶及麻风树关系较近;氰苷含量不同的木薯种质资源块根中MeCYP79D2基因表达量与氰苷含量存在极显著正相关;茉莉酸甲酯处理后,根中MeCYP79D2基因的表达显著上调且氰苷含量显著升高。本研究结果表明MeCYP79D2参与氰苷合成,为解析木薯氰苷生物合成的分子机制研究提供理论基础。

The level of cyanogenic glucosides content is an important trait for the edible quality of cassava, and Me-CYP79D2 is one of the key enzyme genes regulating cassava cyanogenic glucosides synthesis. This study used the edible variety SC9 as the material, cloned the MeCYP79D2 gene in the cytochrome P450 enzyme family of cassava, analyzed it using bioinformatics software, and detected its expression characteristics in different cassava germplasm tubers using real-time quantitative PCR. The cyanogenic glucosides content was measured using HPLC, and its correlation with the expression level of MeCYP79D2 gene was analyzed. The results showed that the length of the coding region sequence of MeCYP79D2 gene was 1625 bp, encoding 528 amino acids. It belonged to an unstable lipophilic hydrophilic protein with 38 phosphorylation sites. Sequence alignment indicated that the gene was highly conserved, and phylogenetic analysis showed that MeCYP79D2 was closely related to rubber and leprosy trees. There was a significant positive correlation between the expression of MeCYPD2 gene and the content of cyanogen glycosides in cassava roots of germplasms with different levels of cyanogen glycosides. After treatment with methyl jasmonate, the expression of MeCYP79D2 gene in roots was significantly upregulated and the content of cyanogenic glucosides was significantly increased. The results of this study indicates that MeCYP79D2 is involved in cyanogenic glucosides synthesis, providing a theoretical basis for studying the molecular mechanism of cassava cyanogenic glucosides biosynthesis.

木薯(Manihot esculenta Crantz)是世界热带地区的主要粮食作物,是重要的淀粉、能源及饲料加工原料,全球以木薯为原料的食品多达上千种[1-2]。因木薯中含有氰苷化合物,人们在食用时要经过一定的处理,费工费时,极大限制了木薯食用化的发展。氰苷是一种广泛存在于高粱、木薯、杂粮杂豆(利马豆和野生荞麦等)、牧草(白三叶和百脉根等)等植物中的代谢产物,其被水解酶水解后能释放有毒化合物氰氢酸,可抵抗病原微生物的侵害和昆虫啃食,也能在植物体内被分解作为氮源[3-4]。木薯中含有1种或2种氰苷化合物,为亚麻苦苷和百脉根苷,其中亚麻苦苷含量占95%以上[5]。木薯分为甜木薯和苦木薯,主要差别在于块根中氰苷类化合物含量的高低,低于50 mg/kg为甜木薯,反之则为苦木薯,甜木薯主要用于食用和饲用,苦木薯主要用于加工利用[6]
细胞色素P450单加氧酶(cytochrome P450, CYP450)是一类普遍存在于动植物和真菌中的超大酶类家族,约占植物基因组编码蛋白质的1%[7]。它作为一种末端加氧酶,可以催化各类羟基化、环氧化、脱烷基化反应,参与植物激素、萜类、生物碱类、黄酮类等次生代谢产物的合成与代谢,在生物过程中发挥重要作用[8-9],广泛分布在玉米[10]、水稻[11]、小麦[12]、番茄[13]、茶树[14]等作物中。氰苷生物合成途径中涉及三大类酶,其中两类均属于细胞色素CYP450单加氧酶家族,为CYP79和CYP71亚族成员,另一类为葡萄糖转移酶。氰苷生成过程中首先通过CYP450,将α-氨基酸羟基化形成N-羟基氨基酸,然后形成醛肟,进一步形成腈,最后一步是由糖基转移酶催化的半氰醇的糖基化反应生成氰苷[15]。CYP450作为氰苷合成途径中的第一步关键酶,在调控植物氰苷含量方面发挥着重要作用。前人研究表明,抑制CYP79D1CYP79D2基因的表达会降低木薯中氰苷含量,从而降低木薯毒性[16]。因此,解析木薯氰苷类化合物代谢的分子机制,选育氰化物含量低的木薯品种,是木薯食用化利用的重要研究方向。
本研究以食用木薯品种SC9为材料,克隆MeCYP79D2基因并进行生物信息学分析,研究其在氰苷含量不同的木薯种质资源块根中的表达情况,并对其在MeJA诱导下的表达量进行分析,初步分析氰苷代谢关键调控酶基因MeCYP79D2的序列及表达差异,以期为后续相关基因功能和调控机理研究奠定基础。
MeCYP79D2基因克隆供试材料为SC9木薯品种的叶片和块根,基因荧光定量检测供试材料为国家木薯种质资源圃保存的16份种质资源。主要试剂:与凝胶电泳有关试剂购自北京擎科生物科技有限公司;乙腈、甲酸购自海南冷港科技有限公司。主要仪器设备:基因扩增仪(TaKaRa,日本)、超微量分光光度计(Implen N60,德国)、实时荧光定量PCR仪(Thermo,美国)和液相色谱仪(Agilent 1260,美国)。PCR引物合成和DNA测序由北京擎科生物科技有限公司完成。
采用RNAprep Pure多糖多酚植物总RNA提取试剂盒(Tiangen)提取样品总RNA,并用核酸检测仪检测其浓度和质量。待RNA质量检测合格后,按照RevertAid RT逆转录试剂盒(Thermo)说明书将总RNA反转录合成cDNA。
在NCBI数据库(https://www.ncbi.nlm.nih.gov/)中下载MeCYP79D2基因序列,利用Primer3Plus(https://www.bioinformatics.nl/cgi-bin/primer3plus/primer3plus.cgi)在线软件设计特异性扩增MeCYP79D2基因编码区(CDS)序列的引物(5′-ATGGCCATGAACGTCTCCAC-3′和5′-TCAAGGTGAAGTGGGGTAGA-3′)。扩增体系为:cDNA模板2 μL、MeCYP79D2-F(10 μmol/L)0.5 μL、MeCYP79D2-R(10 umol/L)0.5 μL、Phanta Max Master Mix(2×)23.5 μL、无菌水补足至50 μL。扩增程序:95 ℃预变性5 min;95 ℃ 30 s,56 ℃ 30 s,72 ℃ 1 min,35个循环;72 ℃ 10 min。PCR产物经1.5%琼脂糖凝胶电泳检测,并送至北京擎科生物科技有限公司测序。
对MeCYP79D2的同源序列、进化关系、蛋白理化性质、结构和功能等方面进行生物信息学分析,使用的数据库和软件见表1
利用RNAprep Pure多糖多酚植物总RNA提取试剂盒(Tiangen)提取16份种质资源的块根RNA。取1 μg RNA,逆转录合成第一链cDNA,稀释10倍后作为模板。使用Primer3Plus(https://www.bioinformatics.nl/cgi-bin/primer3plus/primer3plus.cgi)在线软件设计木薯MeCYP79D2基因的荧光定量PCR引物(正向引物:5'-ACTTCAGAGATCATTTCTCCAGCTA-3',反向引物:5'-ACATTCTTGTTGCTCTTGTACTGGT-3'),选用MeActin为内参基因(正向引物:5'-TGATGAGTCTGGTCCATCCA-3',反向引物:5'-CCTCCTACGACCCAATCTCA-3')。扩增体系(10 μL):cDNA模板1 μL、10 μmol/L荧光定量PCR引物各0.2 μL、ChamQ Universal SYBR qPCR Master Mix(2×)5 μL、ddH2O 3.6 μL。反应程序:95 ℃预变性30 s;95 ℃ 10 s、60 ℃20 s,40个循环。每样品设置3次重复。
参照王琴飞等[17]的方法利用高效液相色谱仪分析样品中氰苷含量。色谱柱:Waters Atlantic C18,4.6 mm×150 mm,粒径5 μm,柱温25 ℃。流动相:A为含0.1%甲酸的80%乙腈/水(V/V),B为0.1%甲酸水溶液。梯度洗脱程序:起始时(0 min),A为10%,B为90%;15 min时,A和B均为50%;18 min时,A为100%,持续2 min;22 min时,A为10%,B为90%;平衡柱子5 min。流速为0.6 mL/min,漂移管温度为95 ℃,雾化温度为80 ℃,气流速度为2.0 L/min,增益为3,进样体积为10 μL。采用外标法计算样品中亚麻苦苷和百脉根苷的含量。
采用Excel 2016和SPSS 20.0软件进行数据处理及分析。应用2-ΔΔCt法计算目的基因的相对表达量,使用Tukey's HSD法进行显著性测验(P<0.05)。使用Graphpad(v8.0.2)绘制相基因对表达量和氰苷含量柱状图。
采用同源克隆策略,在NCBI数据库中搜索获得木薯品种Col22的MeCYP79D2基因cDNA全长1626 bp,编码541个氨基酸。以木薯SC9叶片RNA为模板,扩增MeCYP79D2基因全长,将克隆得到的目的基因纯化、测序后进行序列比对。MeCYP79D2基因编码区长度为1625 bp,编码528个氨基酸(图1)。与Col22的MeCYP79D2基因核苷酸序列比对,在117和1560位置上存在单碱基突变,942位置上存在单碱基缺失,相似性达99.82%。蛋白质序列比对发现有192个位点存在差异,占比38.35%。
在NCBI数据库中对MeCYP79D2氨基酸序列保守结构域进行搜索,结果如图2所示,MeCYP79D2包含细胞色素P450酶(cytochrome_P450)结构域和CYP79结构域非特异性位点,属于细胞色素P450酶超家族(cytochrome_P450 superfamily)。通过Blastp同源比对,发现木薯MeCYP79D2氨基酸序列(XP_021629850.1)与橡胶树(Hevea brasiliensis,XP_021674957.2)、麻风树(Jatropha curcas,AXF53803.1)、蓖麻(Ricinus communis,XP_002534163.3)、水银草(Mercurialis annua,XP_050203649.1)、亚麻(Linum usitatissimum,WAL01197.1)、雷公藤(Tripterygium wilfordii,XP_038697057.1)、大蒲桃(Syzygium grande,KAI6691975.1)、蒜头果(Malania oleifera,XP_057972653.1)、大桉(Eucalyptus grandis,XP_010030374.3)、番木瓜(Carica papaya,XP_021887085.1)等的氨基酸序列相似性较高,分别为88.35%、77.45%、75.60%、70.85%、67.23%、68.74%、60.79%、62.57%、61.19%、59.47%。利用DNAMAN 7.0软件对木薯MeCYP79D2氨基酸序列与上述10个物种进行序列比对,结果表明,不同物种的MeCYP79D2氨基酸序列具有较高的保守性,其中与橡胶树和麻风树相似度较高(图3),说明MeCYP79D2基因的功能与橡胶树和麻风树的CYP79D2功能相似。
利用MEGA7.1[18]软件中neighbor-joining法[19]将木薯MeCYP79D2氨基酸序列与其他10个物种构建同源进化树(图4),结果显示,木薯与橡胶树、麻风树属于同一分支,其中与橡胶树在进化关系上最为接近,与序列比对结果一致。
利用ProtParam在线软件分析预测MeCYP79D2蛋白分子结构式为C2758H4366N742O781S27,相对分子量为61.28 kDa,理论等电点为8.90,预测该蛋白由541个氨基酸残基组成,其中亮氨酸(Leu)比例最高,为9.1%,半胱氨酸(Cys)比例较低,为1.3%;不稳定系数为40.40,脂肪系数为89.78,总平均亲水性为-0.239,依据总平均亲水性小于0为亲水蛋白,推测MeCYP79D2蛋白为不稳定的脂溶性亲水蛋白。
使用ProtScale在线工具对MeCYP79D2蛋白的亲/疏水性进行预测分析,结果如图5所示。第34位氨基酸得分最高,为2.100,疏水性最强,第445位氨基酸得分最低,为-2.063,亲水性最强,第0~50位氨基酸有1个典型的疏水性区域,该区域存在1个跨膜结构。通过TMHMM软件预测发现MeCYP79D2蛋白的第1~18位氨基酸位于细胞膜内,第19~41位氨基酸之间有1个跨膜螺旋区,第42~541位氨基酸位于细胞膜外,推测MeCYP79D2蛋白是一个跨膜蛋白(图6),与ProtScale预测的蛋白跨膜结构结果相符。SignalP预测结果显示,MeCYP79D2蛋白不含信号肽。
采用NetPhos 3.1 Server软件对MeCYP79D2蛋白的磷酸化修饰位点进行分析,结果如图7所示。MeCYP79D2蛋白具有38个潜在磷酸化修饰位点,占氨基酸序列的7.0%,其中丝氨酸位点20个,苏氨酸位点15个,酪氨酸位点3个,主要能被蛋白激酶C(PKC)、蛋白激酶A(PKA)、细胞分裂周期蛋白2(cdc2)和酪蛋白激酶Ⅱ(CKⅡ)等蛋白激酶磷酸化。
利用SOPMA软件对MeCYP79D2蛋白二级结构进行预测,结果如图8所示。MeCYP79D2蛋白主要以α-螺旋和不规则卷曲为主,分别为224个和197个,分别占氨基酸序列的45.10%和36.41%;另外还含有27个β-转角和73个延伸链,分别占氨基酸序列的4.99%和13.49%。基于木薯CYP79D2的晶体结构(Q9M7B7.1A)对MeCYP79D2蛋白进行三级结构建模,结果如图9所示,二者相似度为99.63%。
基于前人转录组数据[20],对MeCYP79D2在木薯叶片、中脉、腋芽、叶柄、茎尖、茎、体胚、脆性愈伤、块根、须根、根尖等11个组织部位的相对表达情况进行分析,结果如图10所示,MeCYP79D2基因在木薯腋芽、叶柄、块根和须根中的表达量显著高于其他组织。
为探究MeCYP79D2在与氰苷含量的关系,取16份氰苷含量的不同的木薯种质资源的块根,检测其MeCYP79D2基因相对表达量和氰苷含量。结果表明,MeCYP79D2基因表达量与氰苷含量存在极显著正相关,相关系数为0.692(图11)。
用100 μmol/L茉莉酸甲酯(MeJA)对SC9木薯种茎进行水培处理,分别检测处理0、3、6、9、12、24 h后MeCYP79D2基因在根、茎、叶中的表达情况及处理0、1、2、3 d后的氰苷含量变化。结果显示,SC9根中MeCYP79D2基因的表达受MeJA诱导显著上调(图12A),且24 h后SC9根中氰苷含量极显著升高,而茎和叶中氰苷含量分别于2~3 d后极显著升高(图12B),推测MeJA调控MeCYP79D2基因的表达来促进氰苷的合成。
氰苷不仅是评判木薯能否食用的关键性状,还是其块根还原氮的重要来源,可影响氨基酸和蛋白质合成以及块根组织损伤后,快速引发采后生理腐烂过程[16,21-23]。植物中氰苷的合成、运输和转运是一个高度整合的过程,涉及不同组织和器官中多种基因和酶的差异表达。在模式植物髙粱中发现氰苷的生物合成途径,具体步骤如下:一是L-酪氨酸经CYP79A1催化后氮端羟基化,脱水、脱羧化生成相对应的对羟基苯乙醛肟;二是在CYP71E1的催化下,对羟基苯乙醛肟通过脱水和碳端羟基化,转变为对羟基苯乙醛腈;三是在糖苷转移酶UGT85B1的作用下,对羟基苯乙醛腈糖基化成为蜀黍苷[24]。不同研究表明,在木薯[25-26]、韭菜[27]、百脉根[28]中均发现了上述生物合成途径。在不同种类的植物中,催化氰苷合成的酶不尽相同,但都是由CYP79同源基因编码,且功能都具有相似性,木薯催化氰苷合成第一步的酶是CYP79D1和CYP79D2[22,29],CYP79D1和CYP79D2基因相似性为85%,与高粱CYP79A1序列具有54%的同源性。虽然CYP79D1和CYP79D2的表达相同,并且都可以使用缬氨酸和异亮氨酸作为底物,但缬氨酸的结合亲和力高于异亮氨酸,因此在木薯中,亚麻苦苷是主要的氰苷成分,占95%以上[30]。且CYP79D1和CYP79D2活性被证实高于CYP71E7,在催化氰苷生物合成中具有限速作用[25]。此外参与氰苷运输和代谢途径的基因有多药及毒素外排转运蛋白MATE、硝酸盐/肽转运蛋白NPF、羟基腈裂解酶HNL等,在利用基因工程改造木薯氰苷含量,增强营养等方面具有重要意义[31]
目前国内外尚未培育出叶片或块根中完全不含氰苷的木薯品种[32]。研究表明,亚麻苦苷在叶片或部分在茎中合成,并通过茎韧皮部运输到块根,一部分储存在液泡中,另一部分为氨基酸合成提供还原的氮源,块根则不合成亚麻苦苷。MCMAHON等[31]提出基于库源关系改良木薯品种中氰苷含量的分子策略,即通过阻断亚麻苦苷合成的限速步骤CYP79D1和D2的催化,可将块根中亚麻苦苷降低到安全水平。GOMEZ等[33]使用基因编辑技术对CYP79D1CYP79D2进行敲除,得出CYP79D2的表达在亚麻苦苷合成中作用突出。本研究从食用品种SC9中克隆得到MeCYP79D2基因编码序列,其cDNA全长1625 bp,编码528个氨基酸,分子量为61.28 kDa,理论等电点为8.90。周伟坚等[34]克隆MeCYP79D2基因与数据库中在核苷酸序列和氨基酸序列同源性达99%,本研究与数据库比对,核苷酸序列相似性达99.82%,但氨基酸序列差异达38.35%,可能与克隆基因的品种差异有关。原晓龙等[35]也在蒜头果中克隆出MoCYP79基因cDNA序列,与木薯中AAF27290.1和AAF27289.1蛋白聚为一类。本研究通过系统进化分析表明,MeCYP79D2与包括蒜头果在内的10个物种距离相近,其中和橡胶和麻风树关系最近。结构分析表明,MeCYP79D2基因属于细胞色素P450酶超家族和CYP79结构域非特异性位点,含有一个保守的血红素结合域,序列为FxxGxRxCxG[36]。生信分析结果显示,该蛋白为为不稳定的脂溶性亲水蛋白,存在一个跨膜结构,不含信号肽,具有38个潜在磷酸化修饰位点,二级结构主要以α-螺旋和不规则卷曲为主。MeCYP79D2主要在须根、叶柄、腋芽和块根中表达。通过高氰苷含量与低氰苷含量的木薯种质资源中MeCYP79D2基因表达量与氰苷含量的相关性分析得出二者存在极显著正相关,是否可以作为高氰和低氰品种的功能标记开发,有待进一步研究。
此外,氰苷在植物防御反应中起着重要作用,同时受外界干旱、低温等环境及内源性激素变化的影响。茉莉酸及其衍生物茉莉酸甲酯(MeJA)是植物体内重要的脂质激素,参与调控生物胁迫和非生物胁迫下植物的生长发育过程[37]。茉莉酸含量的增加通常伴随着防御相关基因的上调表达,产生具有防御功能的次级代谢产物,如萜类化合物、葡萄糖苷、黄酮类和花青素等[38-39]。昆虫咬食和茉莉酸及其衍生物均可上调氰苷合成关键基因的表达[40-42],表明氰苷的合成可能与茉莉酸密切相关。在本研究中,MeCYP79D2基因在MeJA作用下,表达量和氰苷含量均呈显著性变化,说明MeCYP79D2基因参与茉莉酸激素的响应影响氰苷合成。以上结果为后续相关基因的功能研究奠定基础,同时为深入研究木薯氰苷合成机制提供理论依据。
