SOC1 (SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1) is a flowering time regulator in plants. It belongs to the MADS-box transcription factor family and plays a crucial role in the transition from vegetative to reproductive growth in plants. SOC1 is involved in various biological processes, including flower organ development, flowering time control, and response to environmental stimuli and hormone signals. In this study, shading and application of Lorsben were used for flower induction in wax apple, and the number of leaf buds, flowers, and the rate of flowering shoots were recorded after treatment. Additionally, the existing transcriptome data of wax apple in our research group were analyzed to identify the candidate gene SsSOC1 involved in regulating flowering in wax apple. The differential expression of SsSOC1 in different stages and tissues of wax apple under shading and Lorsben treatment was studied using qPCR. The results showed that the expression of the SsSOC1 gene significantly increased in the buds, leaves, leaf tips, and petioles of wax apple treated with shading, with the highest relative expression in the leaf tips. In this study, the SsSOC1 gene from Hei Tang Ba Bi wax apple was successfully cloned, and its promoter analysis, physicochemical properties of the protein, prediction of conserved functional domains, and sequence alignment were performed using bioinformatics methods. The open reading frame of the SsSOC1 gene in wax apple was found to be 654 bp, encoding a protein of 217 amino acids. The theoretical isoelectric point of the protein was 9.37, with a molecular formula of C1076H1794N322O334S10 and a molecular weight of 24.91 kDa. The protein contained 40 positively charged amino acid residues (Lys+Arg) and 32 negatively charged amino acid residues (Asp+Glu). The protein had a fat coefficient of 80.64, a grand average of hydropathicity (GRAVY) of -0.729, and an instability coefficient of 66.17. The protein structure lacked β-folds and did not belong to secretory or membrane proteins. Subcellular localization analysis indicated that the protein was mainly present in the nucleus and belonged to an unstable hydrophilic basic protein. The SsSOC1 protein in wax apple possessed conserved domains including MADS_MEF2_like and K-Box and belonged to the MADS superfamily and K-Box superfamily. The analysis identified the amino acid positions 3 to 75 and 88 to 171 as the start and end sites of the conserved domains, respectively, conforming to the typical structural features of the MADS protein family and the K-Box family. The evolutionary tree showed the highest homology between the SsSOC1 sequence in wax apple and Syzygium oleosum. By floral dip transformation, the SsSOC1 gene was introduced into Arabidopsis, and it was found that the 35S::SsSOC1 line exhibited an early flowering phenotype and a reduced number of rosette leaves compared to the wild type. This study preliminarily elucidated the important role of the SsSOC1 gene in flowering in wax apple and attempted to reveal the intrinsic regulatory mechanism of shading-induced flowering, providing guidance for wax apple production and theoretical basis for improving out-of-season cultivation techniques.

SOC1SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1)是植物中的一个开花时间调节因子,属于MADS-box转录因子家族,在植物从营养生长向生殖生长的转变中发挥关键作用。SOC1参与多种生物过程,包括花器官发育、开花时间控制以及对环境刺激和激素信号的响应。本研究通过遮荫和喷施乐斯本的方式对莲雾进行催花处理,统计处理后的叶芽和花芽数量以及成花枝率,同时,利用课题组现有的莲雾转录组进行分析,筛选出调控莲雾开花的候选基因SsSOC1,并通过实时荧光定量PCR技术研究遮荫处理和喷施乐斯本药剂处理下莲雾不同时期和组织中SsSOC1表达量的差异。结果显示:经过遮荫处理的莲雾树体芽点、叶片、叶尖以及叶柄中的SsSOC1基因表达量显著增加,其中叶尖的相对表达量最高。本研究成功克隆了莲雾黑糖芭比品种的SsSOC1基因,并利用生物信息学方法对其进行启动子分析、蛋白理化性质分析、保守功能域的预测和序列比对。结果表明:SsSOC1基因的开放阅读框长度为654 bp,共编码217个氨基酸,其编码蛋白的理论等电点为9.37,分子式为C1076H1794N322O334S10,分子量为24.91 kDa,带正电荷氨基酸残基总数(Lys+Arg)为40,带负电荷氨基酸残基总数(Asp+Glu)为32,脂肪系数为80.64,总平均亲水性系数(GRAVY)为-0.729,不稳定系数为66.17;莲雾SsSOC1蛋白结构不含β-折叠,不属于分泌蛋白与膜结构蛋白,亚细胞定位显示其主要存在于细胞核中,属于不稳定的亲水性碱性蛋白,该蛋白具有MADS_MEF2_like和K-Box保守结构域,同时隶属于MADS超级家族和K-Box超级家族,该结构域的起止氨基酸位点为第3~75位和第88~171位,符合MADS蛋白家族与K-Box家族的典型结构特征;系统进化树分析显示,莲雾SsSOC1序列与油叶蒲桃的同源性最高;通过花序侵染法将SsSOC1基因转化拟南芥,发现35S::SsSOC1表现出早花表型,莲座叶数量比野生型要少。本研究初步确定了SsSOC1基因在成花方面的重要作用,并揭示莲雾遮荫与成花相关的内在调控机理,为莲雾生产提供指导意义,以及为莲雾反季节栽培技术改良提供理论依据。

SOC1SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1)是植物中的一个开花时间调节因子,属于MADS-box转录因子家族,在植物从营养生长向生殖生长的转变中发挥关键作用。SOC1参与多种生物过程,包括花器官发育、开花时间控制以及对环境刺激和激素信号的响应。本研究通过遮荫和喷施乐斯本的方式对莲雾进行催花处理,统计处理后的叶芽和花芽数量以及成花枝率,同时,利用课题组现有的莲雾转录组进行分析,筛选出调控莲雾开花的候选基因SsSOC1,并通过实时荧光定量PCR技术研究遮荫处理和喷施乐斯本药剂处理下莲雾不同时期和组织中SsSOC1表达量的差异。结果显示:经过遮荫处理的莲雾树体芽点、叶片、叶尖以及叶柄中的SsSOC1基因表达量显著增加,其中叶尖的相对表达量最高。本研究成功克隆了莲雾黑糖芭比品种的SsSOC1基因,并利用生物信息学方法对其进行启动子分析、蛋白理化性质分析、保守功能域的预测和序列比对。结果表明:SsSOC1基因的开放阅读框长度为654 bp,共编码217个氨基酸,其编码蛋白的理论等电点为9.37,分子式为C1076H1794N322O334S10,分子量为24.91 kDa,带正电荷氨基酸残基总数(Lys+Arg)为40,带负电荷氨基酸残基总数(Asp+Glu)为32,脂肪系数为80.64,总平均亲水性系数(GRAVY)为-0.729,不稳定系数为66.17;莲雾SsSOC1蛋白结构不含β-折叠,不属于分泌蛋白与膜结构蛋白,亚细胞定位显示其主要存在于细胞核中,属于不稳定的亲水性碱性蛋白,该蛋白具有MADS_MEF2_like和K-Box保守结构域,同时隶属于MADS超级家族和K-Box超级家族,该结构域的起止氨基酸位点为第3~75位和第88~171位,符合MADS蛋白家族与K-Box家族的典型结构特征;系统进化树分析显示,莲雾SsSOC1序列与油叶蒲桃的同源性最高;通过花序侵染法将SsSOC1基因转化拟南芥,发现35S::SsSOC1表现出早花表型,莲座叶数量比野生型要少。本研究初步确定了SsSOC1基因在成花方面的重要作用,并揭示莲雾遮荫与成花相关的内在调控机理,为莲雾生产提供指导意义,以及为莲雾反季节栽培技术改良提供理论依据。

SOC1 (SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1) is a flowering time regulator in plants. It belongs to the MADS-box transcription factor family and plays a crucial role in the transition from vegetative to reproductive growth in plants. SOC1 is involved in various biological processes, including flower organ development, flowering time control, and response to environmental stimuli and hormone signals. In this study, shading and application of Lorsben were used for flower induction in wax apple, and the number of leaf buds, flowers, and the rate of flowering shoots were recorded after treatment. Additionally, the existing transcriptome data of wax apple in our research group were analyzed to identify the candidate gene SsSOC1 involved in regulating flowering in wax apple. The differential expression of SsSOC1 in different stages and tissues of wax apple under shading and Lorsben treatment was studied using qPCR. The results showed that the expression of the SsSOC1 gene significantly increased in the buds, leaves, leaf tips, and petioles of wax apple treated with shading, with the highest relative expression in the leaf tips. In this study, the SsSOC1 gene from Hei Tang Ba Bi wax apple was successfully cloned, and its promoter analysis, physicochemical properties of the protein, prediction of conserved functional domains, and sequence alignment were performed using bioinformatics methods. The open reading frame of the SsSOC1 gene in wax apple was found to be 654 bp, encoding a protein of 217 amino acids. The theoretical isoelectric point of the protein was 9.37, with a molecular formula of C1076H1794N322O334S10 and a molecular weight of 24.91 kDa. The protein contained 40 positively charged amino acid residues (Lys+Arg) and 32 negatively charged amino acid residues (Asp+Glu). The protein had a fat coefficient of 80.64, a grand average of hydropathicity (GRAVY) of -0.729, and an instability coefficient of 66.17. The protein structure lacked β-folds and did not belong to secretory or membrane proteins. Subcellular localization analysis indicated that the protein was mainly present in the nucleus and belonged to an unstable hydrophilic basic protein. The SsSOC1 protein in wax apple possessed conserved domains including MADS_MEF2_like and K-Box and belonged to the MADS superfamily and K-Box superfamily. The analysis identified the amino acid positions 3 to 75 and 88 to 171 as the start and end sites of the conserved domains, respectively, conforming to the typical structural features of the MADS protein family and the K-Box family. The evolutionary tree showed the highest homology between the SsSOC1 sequence in wax apple and Syzygium oleosum. By floral dip transformation, the SsSOC1 gene was introduced into Arabidopsis, and it was found that the 35S::SsSOC1 line exhibited an early flowering phenotype and a reduced number of rosette leaves compared to the wild type. This study preliminarily elucidated the important role of the SsSOC1 gene in flowering in wax apple and attempted to reveal the intrinsic regulatory mechanism of shading-induced flowering, providing guidance for wax apple production and theoretical basis for improving out-of-season cultivation techniques.

莲雾[Syzygium samarangense (BI.) Merr.et Perry]是桃金娘科蒲桃属常绿果树,又名洋蒲桃,被誉为“水果皇帝”,以其清甜的口味和高水分含量而深受东南亚地区消费者的喜爱[1]。莲雾最先被引进我国台湾,目前在广东、广西、福建等地大规模种植[2]。莲雾具有一年多次开花结果的特点,但主要开花结果时期在夏季。南方夏季高温多雨,容易遭受到病虫害的侵袭,导致果实品质参差不齐,而且夏季水果大量上市,价格不具优势,因此科研人员对莲雾的产期调节技术开展了较多研究[3]
对于大多数植物而言,遮荫会导致光线不足,限制植株的生长和发育,导致徒长和过度的营养生长,从而引起营养器官与生殖器官之间的竞争,不利于开花。然而,对于莲雾来说,遮荫处理却具有促进开花的作用[4]。遮荫处理可以使莲雾的叶芽和花芽进入休眠状态,控制新梢的生长抽生,减少整个树体养分的消耗,提高莲雾的开花率[5]。早期的研究者对莲雾的印度红品种进行了产期调控实验[6],通过喷施莲雾促花灵、多效唑和催花剂,结合环剥、遮荫、断根等措施,实现了印度红莲雾反季节开花,并获得了比正造果更好的品质和更高的经济效益。研究发现,乐斯本可以促进莲雾提前开花,但其促进莲雾开花的机制并未明晰。最近,一些学者开始对莲雾的基因组学进行研究,挖掘与莲雾生长发育相关的基因,但这些基因大多数与莲雾果实成熟相关[7-8],有关莲雾开花相关基因的研究报道较少。
SOC1SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1)及其同源基因编码MADS-box家族转录因子[9],影响植物的成花过程。SOC1通常作为生殖阶段转换的潜在标记基因[10],在拟南芥中,AtSOC1过表达的植株表现出早花的现象,并且相较于野生型植株,AtSOC1过表达的植株莲座叶数量较少,表明SOC1在调控拟南芥开花中起重要作用。SOC1通过多种途径促进植物开花[11],它整合来自长日照、自主和春化途径的多种开花信号并参与花发育[12-13]。此外,它还整合了来自植物年龄和赤霉素的开花信号[14],是植物开花信号的重要整合子。在光周期途径中,COCONSTANS)并不直接作用于SOC1,而是激活FTFLOWERING LOCUS T)后再调控SOC1[15]
研究发现,即使关闭FTSOC1之间的作用通路,赤霉素途径仍能激活SOC1[16]。目前对于莲雾SOC1基因的功能分析未见报道。为此,本研究开展SsSOC1基因的功能分析,为进一步研究SsSOC1在莲雾成花等方面的功能提供理论依据和参考。
供试材料为8年生黑糖芭比莲雾品种,采自广州市天河区华南农业大学果园(23°9ʹ40ʺN,113°21ʹ18ʺE)。转基因实验用材料为哥伦比亚野生型拟南芥(Columbia-0)。
植物双元表达载体pGreenII-35S-C1由高用顺老师惠赠;pMD18-T购自TaKaRa公司;大肠杆菌菌株DH5α、根癌农杆菌菌株GV3101购自上海唯地生物技术有限公司购。
多糖多酚植物总RNA提取试剂盒(RNA prep Pure)、植物基因组DNA提取试剂盒(离心柱型)购自天根生化科技(北京)有限公司;RNA反转录试剂盒[HiScript® II Q RT SuperMix for qPCR(+gDNA wiper)]、同源重组试剂盒(ClonExpress®Ⅱ One Step Cloning Kit)购自诺唯赞(南京)公司;qPCR试剂(Hieff UNICON® Power qPCR SYBR Green Master Mix)、菌落PCR鉴定试剂(Hieff® PCR Master Mix)购自翌圣生物科技(上海)有限公司;PCR高保真酶(PrimeSTAR® Max DNA Polymerase)购自宝生生物工程(大连)有限公司;限制性内切酶、琼脂糖凝胶回收试剂盒、质粒提取试剂盒、氨苄青霉素(Amp)、卡那霉素(Kan)、利福平(Rif)、LB培养基、Glufosinate Ammonium 10% Solution(Basta)购自生工生物工程(上海)股份有限公司。
内参基因选用β-actin基因,使用Primer Premier 5.0(Premier公司)软件设计引物(表1),委托生工生物工程(上海)股份有限公司合成。qRT-PCR采用LightCycler® 480 Real-Time PCR system仪器(Roche, Basel, Switzerland)。荧光定量采用艾科瑞(湖南)生物的SYBR Green Pro TagHS预混型qPCR试剂盒。
为了探究遮荫处理对黑糖芭比莲雾树体成花的影响,分别记录未经遮荫处理[对照组,光合有效外辐射(PARe):(1448.9±41.3)μmol/(m2·s),0%遮荫率]与经过遮荫处理[处理组,PARe:(145.2±12.1)μmol/(m2·s),90%遮荫率]在解除遮荫处理后21 d时的花芽数、成花枝率(以成花枝的数量除莲雾树体总成熟枝数)以及枝梢长度。
在遮荫期间,每隔3 d采集1次芽点(在对照组中为叶芽,处理组中为花芽)、叶片、叶尖以及叶柄。采集后的各个组织立即用液氮进行冷冻处理,于-80 ℃冰箱保存,之后进行RNA提取再反转录为cDNA。利用qRT-PCR技术进行SsSOC1基因不同组织的表达模式分析。反应体系:cDNA稀释模板3 μL,Primer F(0.25 μmol/L)1 μL,Primer R(0.25 μmol/L)1 μL,SYBR Green PCR Master mix 5 μL。反应程序参照qPCR试剂(Hieff UNICON® Power qPCR SYBR Green Master Mix)说明书。
基于转录组筛选出合适的序列,在NCBI数据库使用ORF finder工具找到最长开放阅读框(ORF),根据最长ORF区设计引物进行克隆测序。利用在线工具(表2)对SsSOC1进行生物信息学分析。通过NCBI数据库的BLAST功能筛选与SsSOC1基因相似的不同于莲雾的其他物种中的SOC1基因,获得其蛋白质序列,利用MEGA 7.0软件中的NJ法对其构建进化树,其中boostrap设置为1000,用以验证可信度。
使用多糖多酚植物总RNA提取试剂盒提取莲雾总RNA,得到RNA样品后,取1 μL样品使用Thermo scientific NanoDrop 2000测定总RNA浓度及纯度。取2 μL样品通过琼脂糖凝胶电泳检查RNA的完整性和浓度。用RNA反转录试剂盒进行反转录获得cDNA,用于基因扩增。利用课题组现有的莲雾转录组查找到SsSOC1序列(Unigene0006265)后,于Primer Premier 5.0软件设计SsSOC1的扩增引物(SsSOC1-pMD18-T-F,SsSOC1-pMD18-T-R,表1)。以反转录后所得cDNA为模板,PCR扩增SsSOC1编码区片段,反应体系:2×PCR buffer for KOD FX 25 μL,2 mmol/L dNTPs 10 μL,10 pmol/μL正/反引物各1.5 μL,Template cDNA 1 μL,KOD FX(1.0 U/μL)1 μL,ddH2O补充至50 μL。反应程序为:94 ℃预变性2 min;98 ℃10 s,58 ℃ 30 s,68 ℃ 2 min,35个循环。4 ℃保存。扩增产物2 μL经1%琼脂糖凝胶电泳检测确定目的片段大小后,采用琼脂糖凝胶回收试剂盒进行回收。目的片段经过回收后采用同源重组试剂盒与pMD18-T载体连接,连接后均匀涂布于含100 mg/mL Amp抗性的LB固体培养基上,平板倒置,37 ℃培养12 h。使用无菌的枪头将单个菌落挑至20 mL灭菌水中混匀,37 ℃孵育30 min。取1 μL作为PCR模板,进行菌液PCR及电泳鉴定,将含有目的基因片段加入含100 mg/mL Amp的LB液体培养基中,37 ℃ 200 r/min振荡培养8 h后,送至生工生物工程(上海)股份有限公司测序。测序验证后用质粒提取试剂盒提取pMD18-T-SsSOC1质粒。以pMD18-T-SsSOC1载体质粒为模板,使用引物(SsSOC1-C1- F/SsSOC1-C1-R)对其进行PCR扩增,使用Hind III和Xba I限制性内切酶对pGreen II-35S-C1载体进行酶切。以未酶切质粒为对照采用1%琼脂糖凝胶进行电泳检测。随后进行切胶回收。纯化后的目的基因片段与线性化功能载体使用同源重组试剂盒将pGreenII-35S-C1载体与SsSOC1基因片段连接。反应体系:SsSOC1基因片段100 ng,pGreenII-35S-C1线性化载体100 ng,5×CE II Buffer 4 μL,ddH2O补充至20 μL。反应程序为:37 ℃ 30 min,0 ℃ 5 min。随后转化至DH5α,挑取单克隆菌落进行菌落PCR鉴定,鉴定完成后送至生工生物工程(上海)股份有限公司测序,测序正确的菌株进行扩大培养,抽提质粒保存。
取100 ng pGreenII-35S-C1-SsSOC1质粒加入50 μL农杆菌感受态(GV3101)中,拨打混匀,冰上静置5 min,液氮5 min,37 ℃水浴5 min,冰浴5 min;加入700 μL无抗生素的LB液体培养基,于28 ℃,200 r/min振荡培养3 h;6000 r/min离心1 min;留取100 μL左右上清液轻轻吹打重悬,将菌块涂布于含Rif 25 mg/L+Kan 50 mg/L的LB平板上倒置,28 ℃培养2 d。挑取单克隆菌落进行菌落PCR鉴定,最后将检测正确的农杆菌保存于10%甘油中,并于-80 ℃保存。采用花序浸染法[17]进行拟南芥的遗传转化,含有重组质粒的上述甘油农杆菌,在含有Rif 25 mg/L+Kan 50 mg/L的LB平板上划线,于28 ℃,200 r/min振荡培养16 h。吸取4 mL摇浓的菌液加入到200 mL含相应抗生素的LB液体培养基中,28 ℃,200 r/min,培养至OD600=1.0;5000 r/min离心10 min,收集细胞并悬浮于0.5%蔗糖渗透培养液(含有0.02% Silwet-77),调整OD600=0.8;将盛花期拟南芥花完全浸入农杆菌悬浮液中侵染2 min,放入黑色托盘中,置于黑暗环境下避光培养24 h后于正常光照下培养。
将收取到的T0代种子经过4 ℃低温浸泡后均匀播撒在花盘中,在生长条件为23 ℃,长日照(16 h/8 h)培养室中培养。培养至1周左右时(子叶展平),喷施0.1% Basta进行筛选。取叶片组织提取DNA和RNA,随后进行1%琼脂糖凝胶电泳和定量PCR的测定,对检测正确的阳性苗和野生型植株进行跟踪观察,待拟南芥抽薹开花长至1 cm时,统计所需天数以及莲座叶数量。
使用Excel、SPSS(v 26.0)软件进行统计分析和差异显著性分析,使用GraphPad Prism(v 8.0.2)软件制图。
遮荫处理对莲雾树体中的花芽数、成花枝率及新梢长度有着显著的影响(图1),经过遮荫处理(T)的花芽数比未经遮荫处理(CK)高123.39%。同时,处理组的成花枝率比对照组高115.06%。而处理组莲雾树体的新梢长度明显短于对照组,表明一定程度的遮荫处理会减短新梢长度。
在对照组的各个组织中,SsSOC1的相对表达量差异不大,且数值较低。但遮荫处理组莲雾的芽点、叶片、叶尖以及叶柄中的SsSOC1相对表达量均增加,其中,叶尖中的SsSOC1相对表达量最高,是对照组的10倍;叶柄中的SsSOC1相对表达量与对照组之间差异最大,是对照组的14倍;处理组花芽中的SsSOC1相对表达量是对照组叶芽的4倍(图2)。
本研究从黑糖芭比莲雾叶片中提取总RNA,并用1%浓度的琼脂糖胶检测其质量,胶图显示质量良好(图3A)。反转录成cDNA后,以其为模板,用PCR扩增出SsSOC1编码区序列,获得全长654 bp的目的基因(图3B)。为验证得到的SsSOC1序列的准确性,将扩增的SsSOC1编码区序列与pMD18-T载体进行连接并测序,测序结果发现扩增获得的基因序列与转录组中对应的基因序列完全一致(Unigene0006265),成功得到目的基因SsSOC1图3C)。
利用相关在线工具对莲雾的SsSOC1蛋白进行生物信息学分析,结果显示,SsSOC1蛋白的理论等电点为9.37,分子式为C1076H1794N322O334S10,分子量为24.91 kDa,带正电荷氨基酸残基总数(Lys+Arg)为40,带负电荷氨基酸残基总数(Asp+Glu)为32,脂肪系数为80.64,总平均亲水性系数(GRAVY)为-0.729,不稳定系数为66.17;SsSOC1蛋白亲水性最低点值为-3.144,疏水性最高点值为2.122(图4A),推测该蛋白为亲水性蛋白;SsSOC1蛋白二级结构中α-螺旋占比最高,为60.83%,延伸链和无规则卷曲占比分别为9.22%、25.81%,无β-折叠(图4B),SsSOC1蛋白质三级结构的预测见图4C;SsSOC1蛋白具有MADS_MEF2_like和K-Box保守结构域,同时隶属于MADS超级家族和K-Box超级家族,该结构域的起止氨基酸位点为第3~75位和第88~171位,符合MADS蛋白家族与K-Box家族的典型结构特征(图4D);SsSOC1蛋白的信号肽预测显示SsSOC1蛋白不含信号肽(图4E);SsSOC1蛋白的跨膜区域检测和亚细胞定位预测结果显示,该蛋白不是分泌蛋白与膜结构蛋白,定位于细胞核中(图4F)。
通过NCBI数据库中的BLAST功能对SsSOC1的氨基酸序列与油叶蒲桃(Syzygium oleosum)、大桉(Eucalyptus grandis)、银叶玫瑰木(Rhodamnia argentea)、胡桃(Juglans regia)、欧洲榛(Corylus avellana)、小果胡桃与核桃杂交种(Juglans microcarpa × Juglans regia)、枣(Ziziphus jujuba)、欧洲凯木(Alnus glutinosa)、美国山核桃(Carya illinoinensis)、可可(Theobroma cacao)、杨梅(Morella rubra)、石榴(Punica granatum)、白栎(Quercus lobata)、榴莲(Durio zibethinus)、山核桃(Carya cathayensis)、川桑(Morus notabilis)等16种植物的SOC1氨基酸序列进行比对分析,结果显示,莲雾SsSOC1蛋白序列与油叶蒲桃的同源性最高,相似度高达99.08%,而与川桑的蛋白序列同源性最低,相似度只有78.24%(图5A)。利用MEGA 7软件构建SsSOC1氨基酸序列与以上16个物种的SOC1氨基酸序列的系统进化树(图5B),发现莲雾与油叶蒲桃聚于同一分支,亲缘关系相比其他物种最近,而与小果胡桃与核桃杂交种、胡桃的亲缘关系则较远。
为了验证莲雾SsSOC1基因的功能,本研究构建过表达载体35S::SsSOC1,并将其转入哥伦比亚野生型拟南芥中,通过喷施Basta和PCR鉴定筛选转基因植株,观察转基因的开花相关表型,并统计T1和T2代的开花时间和莲座叶数量。通过观察转基因后代表型,并与野生型拟南芥(WT)进行比较,发现35S::SsSOC1株系营养生长期明显短于野生型植株,并且其抽薹时间显著提前(图6A),在抽薹开花时,叶片数量与野生型相比明显减少(图6B),成功转入SsSOC1基因的植株平均提早8 d开花(图6C)。结果表明成功转入SsSOC1基因能够使拟南芥的开花时间提前。
遮荫对莲雾的开花时间已经有相关报道[18]。目前,大多数莲雾种植者使用遮荫技术结合化学调控来调节莲雾的开花,然而这种调控机制的具体原理仍知之甚少。遮荫对植物的生长影响复杂,不同植物受到的影响也各不相同。遮荫对红椿(Toona ciliata)、香樟(Cinnamomum camphora[19]幼苗和拟单性木兰(Parakmeria yunnanensis[20]不利,会显著抑制苗高和地径。然而,对于移栽期的超级水稻,遮荫后植株高度显著增加[21]。此外,遮荫也促进了油茶(Camellia oleifera)的营养生长[22]
本课题组前期已对SsSOC1基因和SsCOP1基因进行了基础的功能验证,并初步探究了遮荫处理对莲雾光合特性的影响[3]。本研究基于前期研究结果展开更为细致的探索,重点分析SsSOC1的结构、特性以及其在莲雾中的具体作用。
SOC1在对植物成花的过程发挥着重要的作用,是花器官分生组织形成过程中的关键基因之一,其能够整合外界以及内在因素等各种信号,从而激活下游花器官发育相关基因[23]。经过遮荫处理莲雾的叶片、花芽、叶尖以及叶柄中SsSOC1相对表达量都增加。其中,叶尖在处理组中SsSOC1相对表达量最高,叶柄中SsSOC1的相对表达量在处理组与对照组之间的差异最大。
SOC1基因编码的转录因子,是典型的MIKC蛋白[24]。本研究克隆了莲雾的SOC1基因并命名为SsSOC1,利用ProtParam、ProtScale以及Prabi等在线工具对SsSOC1进行生物信息学分析发现,SsSOC1中不存在β-折叠,α-螺旋占比最高,其属于不稳定亲水性蛋白质,不含信号肽且无跨膜结构,不属于分泌蛋白和膜结构蛋白,定位于细胞核。保守结构域分析发现SsSOC1中存在MADS_MEF2_like和K-Box保守结构域。通过多重序列比对以及构建进化树发现SsSOC1蛋白与油叶蒲桃SOC1蛋白的同源性最高,与其他植物的SOC1序列同源性均在75%以上,表明SOC1序列在多数物种中具有着极大的保守性,这对于今后分析莲雾中K-box基因家族具有重要的便利性,可通过了解其他物种K-box基因家族中SOC1基因的功能机制,推测莲雾SOC1基因的详细表达模式和作用机制。
据报道,过表达SOC1基因可以促进植株提前开花,而SOC1基因突变体则会导致植株开花延迟[25]。本研究转入SsSOC1基因的植株开花时间早于野生型植株,并且莲座叶数量显著少于野生型植株。这成功验证了SsSOC1基因的转入对提早开花表型的促进作用,与预期效果一致。
果树的产量在一定程度上决定于当年开花的数量以及质量。莲雾是我国新兴果树品种,经济效益较高,但其开花数量及时间的不稳定极大地限制了其经济效益,本研究能够为今后培育一年内多次开花、反季节开花并且高产、稳产的莲雾品种提供一定的指导,同时本研究基于莲雾基因组学,对莲雾中SOC1基因编码的蛋白特征、进化关系以及其在不同时期不同组织的表达差异进行分析,初步鉴定了调控莲雾开花的主要基因之一的SsSOC1,并且对其进行了基因验证得出了较好的实验结果,为深入揭示莲雾开花结实过程中SOC1基因的调控机制奠定基础。
