In order to investigate the genetic diversity of phenotypes and improve the utilization efficiency of different germplasm resources in Phalaenopsis, 76 germplasm resources of Phalaenopsis were investigated on the phenotype diversity by differential analysis, correlation analysis, principal component analysis and cluster analysis. The ornamental value of big-flower, medium-flower and small-flower cultivars were evaluated comprehensively. The results showed that plant height was the most significant difference among the cultivars, and the flower number per plant was the largest coefficient of variation. Most phenotypes showed significant or extremely significant positive correlation. Flower width and length were the main factors leading to the genetic diversity of phenotypes in Phalaenopsis. The 11 phenotypes were divided into three categories by the R-type cluster analysis, among which flower length and width were clustered firstly when the Euclidean distance was 1.1, indicating the highest degree of correlation between them. 76 germplasm resources could be divided into three categories by Q-type cluster analysis, big flower, medium flower and small flower cultivars. In the comprehensive evaluation of ornamental value, the big flower Baihuahonxin, Huanying and V3, the medium flower Chulian, Huangpao, Jixiang, Memphis, 79, Ama and the small flower Auspicious, Xiaomeihua, Christmas Tree, LL29 and Queen Beer obtained higher scores. The results would provide a reference for the efficient utilization of germplasm resources and parent selection of crossbreeding in Phalaenopsis.

为探究不同蝴蝶兰种质资源表型性状的遗传多样性特征,提高种质资源利用效率,本研究以76份蝴蝶兰种质资源为研究对象,通过表型性状的差异性分析、相关性分析、主成分分析以及聚类分析进行表型遗传多样性研究,并分别对大花型、中花型和小花型蝴蝶兰进行观赏性综合评价。结果显示:蝴蝶兰品种间差异性最显著的是株高,变异系数最大的是单株花朵数,大部分性状之间呈现显著或极显著正相关,花朵宽度和花朵长度是导致蝴蝶兰表型遗传多样性的主要因素。R型聚类分析将11个表型性状分为3个大类,其中花朵长度和花朵宽度在欧式距离为1.1时最先聚为1类,表明其相关程度最高;Q型聚类分析显示76份种质资源可划分大花型、中花型和小花型3个类别。在观赏性综合评价中,大花型包括白花红心、幻影、V3,中花型:初恋、黄袍、吉祥、孟菲斯、79、阿玛等,小花型包括吉祥如意、小梅花、宝贝、圣诞树、LL29、满天红等品种综合评分较高。本研究结果将为蝴蝶兰种质资源的高效利用以及杂交亲本选择提供参考依据。

为探究不同蝴蝶兰种质资源表型性状的遗传多样性特征,提高种质资源利用效率,本研究以76份蝴蝶兰种质资源为研究对象,通过表型性状的差异性分析、相关性分析、主成分分析以及聚类分析进行表型遗传多样性研究,并分别对大花型、中花型和小花型蝴蝶兰进行观赏性综合评价。结果显示:蝴蝶兰品种间差异性最显著的是株高,变异系数最大的是单株花朵数,大部分性状之间呈现显著或极显著正相关,花朵宽度和花朵长度是导致蝴蝶兰表型遗传多样性的主要因素。R型聚类分析将11个表型性状分为3个大类,其中花朵长度和花朵宽度在欧式距离为1.1时最先聚为1类,表明其相关程度最高;Q型聚类分析显示76份种质资源可划分大花型、中花型和小花型3个类别。在观赏性综合评价中,大花型包括白花红心、幻影、V3,中花型:初恋、黄袍、吉祥、孟菲斯、79、阿玛等,小花型包括吉祥如意、小梅花、宝贝、圣诞树、LL29、满天红等品种综合评分较高。本研究结果将为蝴蝶兰种质资源的高效利用以及杂交亲本选择提供参考依据。

In order to investigate the genetic diversity of phenotypes and improve the utilization efficiency of different germplasm resources in Phalaenopsis, 76 germplasm resources of Phalaenopsis were investigated on the phenotype diversity by differential analysis, correlation analysis, principal component analysis and cluster analysis. The ornamental value of big-flower, medium-flower and small-flower cultivars were evaluated comprehensively. The results showed that plant height was the most significant difference among the cultivars, and the flower number per plant was the largest coefficient of variation. Most phenotypes showed significant or extremely significant positive correlation. Flower width and length were the main factors leading to the genetic diversity of phenotypes in Phalaenopsis. The 11 phenotypes were divided into three categories by the R-type cluster analysis, among which flower length and width were clustered firstly when the Euclidean distance was 1.1, indicating the highest degree of correlation between them. 76 germplasm resources could be divided into three categories by Q-type cluster analysis, big flower, medium flower and small flower cultivars. In the comprehensive evaluation of ornamental value, the big flower Baihuahonxin, Huanying and V3, the medium flower Chulian, Huangpao, Jixiang, Memphis, 79, Ama and the small flower Auspicious, Xiaomeihua, Christmas Tree, LL29 and Queen Beer obtained higher scores. The results would provide a reference for the efficient utilization of germplasm resources and parent selection of crossbreeding in Phalaenopsis.

蝴蝶兰(Phalaenopsis spp.)为兰科(Orchidaceae)蝴蝶兰属(Phalaenopsis)附生常绿草本植物,因花型奇特、花色繁多、花期较长,故素有“洋兰王后”之称,是中国销量最大的年宵花,也是世界范围内销量第一的盆栽观赏植物[1-2]。2022年,中国蝴蝶兰年产量达1.3亿株,其中8000万株供应2023年的年宵花市场,具有重要的经济价值。
表型性状多样性在一定程度上可反映遗传的异质性,是遗传多样性的具体表现,利用表型性状分析种质资源的遗传多样性直观易行,有助于种质资源挖掘和利用[3]。基于主成分分析法、聚类分析和层次分析法的表型性状遗传多样性评价已经应用于春兰(Cymbidium goeringii[4]、文冠果(Xanthoceras sorbifolium[5-6]、杜鹃(Rhododendron spp.)[7-8]、苹果(Malus pumila[9]、朱槿(Hibiscus rosa-sinensis[10]、火龙果(Hylocereus undulatus[11]等物种中。
自1887年英国皇家园艺学会(Royal Horticultural Society, RHS)登录第1个蝴蝶兰属杂交品种,蝴蝶兰经过长达130余年的育种形成了数量庞大的栽培品种[3]。截至2024年7月,据国际兰花新杂交种登录机构RHS统计,蝴蝶兰杂交种数达31 818个[12]。尽管部分蝴蝶兰品种已进行表型性状遗传多样性的相关研究[13-16],但涉及的品种较为有限,尚有大量的蝴蝶兰品种未进行表型性状遗传多样性分析,这极大阻碍了对蝴蝶兰种质资源的充分利用。
本研究以76份蝴蝶兰种质资源为研究对象,对11个关键表型性状进行差异性分析、相关性分析、主成分分析以及聚类分析,并分别对大花型、中花型和小花型蝴蝶兰进行观赏性评价,进而探究蝴蝶兰不同表型性状的遗传多样性,建立蝴蝶兰种质资源表型性状的综合评价体系,研究结果将为蝴蝶兰种质资源的高效利用以及杂交亲本的选择提供重要参考。
试验材料为种植于云南省农业科学院花卉研究所九溪基地的76份蝴蝶兰种质资源,详见图1。按照不同花型蝴蝶兰的划分标准[17],将76份蝴蝶兰品种划分为大花型24份,中花型30份,小花型22份。
76份蝴蝶兰资源种植于云南省农业科学院花卉研究所九溪基地,进行常规管理,试验于2021—2023年连续3 a在资源圃内开展,以《植物新品种特异性、一致性和稳定性测试指南 蝴蝶兰》(NY/T 2230—2012)[18]为依据,于盛花期对供试种质资源均随机选取3株植株进行株高、叶长、叶宽、叶片姿态、花梗数、花序梗长、花序长、花序梗粗、花朵长、花朵宽、单株花梗数和花香等性状测定,其中花朵长、花朵宽和花香等性状每株选取3朵花进行测量,并对花梗数和花香分别进行编码,单梗(1分)、1~2梗或双梗(2分)、3梗(3分)、≥4梗(4分),有香(2分)、无香(1分),详见图2
结合表型性状,使用Excel 2021软件对数据初步整理,并进行性状的最大值、最小值、平均值、标准差、方差及变异系数的计算,对其株高、叶长、叶宽、花序梗数、花序梗长、花序长、花序梗粗、花朵长、花朵宽、单株花朵数和花香11个性状进行相关性分析,并使用Origin 2021软件绘制相关性热图。运用SPSS 27软件进行主成分分析,采用组内联接的聚类方法进行11个性状的R型聚类分析和76个品种间的Q型聚类分析,最后输出聚类树形图。
参考袁闯等[19]的方法计算不同花型蝴蝶兰的11个性状的综合评价值(D),并运用K-Means聚类进行分级。如表1所示,按照层次分析法构造出A(目标层)、C(约束层)、P(指标层)3个分层结构模型,指标层P共有11个指标,分别为:株高(P1)、叶长(P2)、叶宽(P3)、花梗数(P4)、花序梗长(P5)、花序梗粗(P6)、花序长(P7)、花朵长度(P8)、花朵宽度(P9)、单株花朵数(P10)、花香(P11)。约束层C对指标层进行数据的分类(植株C1、叶部性状C2、花梗性状C3、花朵性状C4)。
由76份蝴蝶兰种质资源盛花期植株的11个表型性状统计数据可知,其中,花朵宽度、花朵长度、花序长度、单株花朵数属于花部性状,叶长和叶宽属于叶部性状,花序梗长、花序梗粗、花梗数等梗部性状,株高属于植株整体性状(图2)。
表2所示,76份蝴蝶兰种质资源的株高、花序梗长、单株花朵数、花序长等性状的方差值较大,不同品种的差异性较大,其中,株高的品种间差异最大,其最小值为26.00 cm,最大值为90.00 cm,方差值为223.44。11个表型性状的变异系数在0.14~0.57之间,平均变异系数为0.30,变异系数最大的是单株花朵数为0.57,其次是花序梗长为0.39,最小的是叶宽为0.14。
76份蝴蝶兰的11个表型性状的相关性分析显示,大部分花部性状之间具有显著或者极显著相关性,其中,花朵长度和花朵宽度呈极显著正相关,相关系数达到了0.90。单株花朵数与花梗数呈显著正相关,与花序梗长、花朵宽度和花朵长度呈极显著负相关,与叶宽相关性不显著,与其他性状呈负相关。花序梗长与株高、叶长、叶宽呈显著正相关,与花梗数呈极显著负相关(图3)。
在11个表型性状进行主成分分析中,根据特征值大于1的原则筛选出3个主成分,3个主成分的累计贡献率为72.89%(表3)。主成分1的特征值为5.85,方差贡献率为53.21%,载荷值较大的是花朵宽度(0.93)、花朵长度(0.90)、花序梗长(0.86)和株高(0.86);主要成分2的特征值为1.15,方差贡献率为10.46%,载荷值较大的是花香(0.75)和花梗数(0.47);主要成分3的特征值为1.01,方差贡献率为9.16%,载荷值较大的是单株花朵数(0.54)和叶宽(0.43)。11个表型性状中,花朵宽度(0.93)和花朵长度(0.90)的载荷值均在0.90及以上,是导致蝴蝶兰表型性状遗传多样性的主要因素。
11个表型性状的R型聚类结果显示,在欧氏距离为10时,11个性状聚为2个大类,第Ⅰ大类包含株高、花序梗长、单株花朵数和花序长4个性状,第Ⅱ大类包含花朵长度、花朵宽度、叶长、花梗数、叶宽、花香和花序梗粗7种性状。在欧式距离为5时,第Ⅱ类又分为了2类,花朵长度、花朵宽度、叶长和花梗数聚为1类;叶宽、花香和花梗粗聚为1类。第Ⅱ大类中,花朵长度和花朵宽度2种性状在欧式距离为1.1时最先聚为1类(图4),表明其相关程度最高,与相关性分析结果一致。在R型聚类结果中,大多数性状分布均比较分散,说明蝴蝶兰的表型性状是独自进化的,性状相似度较小。
图5所示,以花朵长度和花朵宽度2个表型性状对76份蝴蝶兰种质资源绘制Q型聚类树状图。当欧式距离为15时,76个蝴蝶兰品种聚成2大类,第Ⅰ大类包含55个品种,主要包括小花型和中花型蝴蝶兰品种。第Ⅱ大类有大辣椒、大如意、九五至尊、V3等23个品种,为大花型蝴蝶兰品种。当欧式距离为10时,第Ⅰ大类被分为2大类,第Ⅰ-1类有杭州美女、安娜、阿玛、中国红等33个品种,主要为中花型蝴蝶兰品种;第Ⅰ-2类有彼自豪、金蝶、满天红、吉祥如意等20个品种,均为小花型蝴蝶兰品种。
本研究分别对大花型、中花型和小花型的蝴蝶兰种质资源构建综合评价体系,通过专家对蝴蝶兰性状重要性进行问卷调查,赋予因子两两间比较关键性的评估,建立判断4个矩阵约束层相对于目标层的比较判断矩阵(A~C)和指标层相对于约束层的比较判断矩阵(C~P),并进行一致性检验。如表4~表6所示,所有矩阵的一致性比率(CR)均低于0.1,均通过一致性检验,说明本研究建立的3大花型蝴蝶兰品种观赏性状综合评价判断矩阵是合理的。
将指标层中Pi权重(Wi)与对应约束层C的权重(Wi)相乘得到各指标层对于目标层的权值即为绝对权重。如图6所示,在大花型、中花型以及小花型蝴蝶兰中,花部性状均是权重值最高的约束层。
表7~表9所示,大花型、中花型以及小花型的不同性状权重值排序存在差异。依据权重值从大到小排序,大花型权重值排名前3的性状为花朵宽度(P9)、花朵长度(P8)、花序长(P7),权重值最小的为花梗数(P4)(表8);中花型权重值排名前3的性状为花序长(P7)、单株花朵数(P10)、花朵宽度(P9),权重最小值为叶宽(P3)(表9);小花型权重排名前3的性状为花序长(P7)、单株花朵数(P10)、花梗数(P4),权重最小值为株高(P1)(表10)。
表10所示,22份大花型蝴蝶兰品种的分值在2.72~8.92之间,得分在6.50分以上的Ⅰ级品种有6个,分别为白花红心、幻影、V3、绯红之日、梦幻婚礼和2009,这些品种株型高大,花型较大,花朵长在9.5~12.0 cm之间,花朵宽在10.5~12.0 cm之间,花序梗较长,在48.0~65.0 cm之间,这些优良性状使得这6个品种综合得分靠前。
表11所示,30份中花型蝴蝶兰品种的得分在3.29~8.64之间,得分在6.50分以上的Ⅰ级的品种有8个,分别为初恋、黄袍、吉祥、孟菲斯、79、阿玛、福星和金雕,这些品种相对于其他中花型品种而言,株型较大,花序长和花序梗长多在30.0~55.0 cm范围,花型较大,多为双梗,这些优良性状使得这些品种综合得分靠前。
表12所示,22份蝴蝶兰小花品种的分值分布在1.11~9.31之间,得分7.00分以上的Ⅰ级的品种有6个,分别为吉祥如意、小梅花、宝贝、圣诞树、LL29和满天红,这些品种相对于其他小花型品种,花序较长在24.0~30.5 cm之间,花朵繁盛,单株花朵数较多在28~58朵,花梗数为双梗及以上,整体比较丰满,使得综合得分靠前。
遗传和环境共同决定了植物表型性状,植物表型性状具有直观、易调查等特点,可以直观地确定种质多样性的丰富度,并较快地揭示遗传变异的程度,因此研究种质资源的遗传多样性在育种中至关重要,是种质资源鉴定、评价和利用的关键[20-21]
植物表型性状的离散程度和种质间的变异程度可由变异系数体现[22],变异系数越高植物的遗传变异越丰富。本研究中,76份蝴蝶兰种质资源11个性状的变异系数在0.14~0.57之间,该结果与陈剑锋等[15]对135份蝴蝶兰种质资源12个性状的变异系数分析结果相近(0.19~0.52),说明蝴蝶兰不同种质资源表型性状的遗传变异程度相近。与蝴蝶兰不同,朱槿(Hibiscus rosa-sinensis)各表型性状的变异系数在9.49~31.05之间,遗传变异更加丰富[10]。此外,在已有的蝴蝶兰种质资源表型遗传多样性分析中均显示单株花朵数的变异系数最大[14-15],本研究结果亦显示单株花朵数的变异系数最大(0.57),说明在蝴蝶兰的众多表型性状中,单株花朵数的遗传变异性最为丰富。
本研究发现花部性状之间大多数性状存在显著或极显著相关性,其中花朵宽度和花朵长度呈极显著正相关。与之相似,王世尧等[16]对57份蝴蝶兰种质资源表型性状相关性分析显示17个性状间存在不同程度的相关性,且花部性状两两之间呈现极显著相关。此外,本研究对蝴蝶兰表型性状的主成分分析显示,花朵宽度和花朵长度是导致76份蝴蝶兰种质资源表型性状遗传多样性的关键性状,这与王钦等[23]对蝴蝶兰表型性状遗传多样性的分析结果一致。
本研究以花朵长度和花朵宽度等花部性状采用组内联接的方法,进行品种间Q型聚类分析,将76份蝴蝶兰种质资源分为大花型、中花型和小花型3大类群。然而,王钦等[23]采用Q型聚类分析将72份蝴蝶兰品种聚为四大类群,陈和明等[13]通过Q型聚类分析将213份蝴蝶兰品种聚为杂交种和原生种两大类群,这可能与聚类分析时使用的参数和关注的主要性状存在差异有关。
在蝴蝶兰生产销售中,通常根据花型大小,分为大花型、中花型和小花型品种[17]。但在已有蝴蝶兰观赏性综合评价中,通常是对所有花型蝴蝶兰品种一起进行评价,尚未见针对不同花型蝴蝶兰分别建立的评价标准[13-16]。本研究对76份蝴蝶兰种质资源分不同花型(大花型、中花型和小花型)进行了观赏性综合评价,得出大花型蝴蝶兰白花红心、幻影、V3等综合得分较高,中花型初恋、吉祥、阿玛等的综合得分较高,小花型吉祥如意、宝贝、满天红等综合得分较高。相对而言,本研究对不同花型蝴蝶兰进行观赏性综合评价更具有针对性。
本研究对76份蝴蝶兰进行表型性状遗传多样性分析,并分不同花型(大花型、中花型和小花型)进行观赏性综合评价,为蝴蝶兰新品种选育提供重要依据,对蝴蝶兰种质资源的高效利用和杂交亲本的选择均具有重要意义。
