Drought is one of the most serious abiotic stresses to land plants. Environmental water deficiency triggers an osmotic stress signaling cascade. OSCA1 was described as a hyperosmolarity-gated calcium-permeable channel and its function in osmosensing in plants, homologues of OSCA genes are found throughout eukaryotes. Water is crucial to plant growth and development. As one of the primary horticulture commodities, tomato plays an important role in vegetable research. To enhance the survival and yield of tomatoes in drought conditions, it is essential to thoroughly study the function of SlOSCA genes and their drought-resistant mechanisms. Based on reverse genetics, we selected SlOSCA1.1/1.2/1.6/2.3/3.1 as our target genes through bioinformatics analysis, the slosca mutant of tomato was made by CRISPR/cas9-mediated gene editing directly in AC (Ailsa Craig). To explore the role of tomato SlOSCA protein family members in the response to osmotic stress. Here, we found impaired guard cell osmotic stress signaling and attenuated plant responses to osmotic stress in slosca3.1/1.1 plants, impaired osmotic stress signaling and attenuated plant responses to osmotic stress in slosca1.6/1.2/2.3 plants, the slosca1.6/1.2/2.3 plants showed lower water use efficiency and poorer drought resistance, there is genetic redundancy in OSCA. In this study, slosca mutants were constructed and then the physiological phenotypes of the mutants were investigated, which revealed that multiple genes of the SlOSCA family cooperatively regulate responses to osmotic stress, this study provides a reference for subsequent analysis of the biological functions of OSCA gene family under different stress conditions and different physiological processes.

干旱是陆地植物面临的严重非生物胁迫之一,干旱环境会造成植物水分缺失从而产生高渗胁迫,OSCA1是植物中第一个被鉴定的渗透感受器,OSCA基因在不同物种中广泛存在。植物对干旱胁迫的感受和响应对其生长发育至关重要,番茄作为经济作物和模式植物在蔬菜研究中具有重要作用,番茄抗旱性研究是保障蔬菜产量的重要途径,因此,研究番茄SlOSCA基因家族在番茄应对干旱胁迫过程中的生理功能方面具有重要意义。本研究以反向遗传学为基础,通过SlOSCA基因家族在叶片中的表达量分析,选定SlOSCA1.1/1.2/1.6/2.3/3.1共5个基因作为番茄抗旱研究的目标基因,利用CRISPR/Cas9定点基因编辑技术构建番茄slosca突变体,以探索番茄SlOSCA蛋白家族成员在番茄响应渗透胁迫过程中的作用。研究结果显示,番茄slosca3.1/1.1突变体在细胞水平对渗透胁迫的响应减弱,SlOSCA3.1/1.1参与了番茄保卫细胞对渗透胁迫的响应过程;番茄slosca1.6/1.2/2.3突变体整株水平对渗透胁迫的响应能力减弱,SlOSCA1.6/1.2/2.3参与了番茄对PEG渗透胁迫以及土壤干旱胁迫的响应过程,再次证明OSCA蛋白家族成员之间存在功能冗余。本研究通过构建slosca突变体,研究slosca突变体的生理表型,揭示SlOSCA蛋白家族成员协调作用于番茄抗旱过程,为后续解析OSCA基因家族成员在不同胁迫条件和不同生理过程中的生物学功能提供参考。

干旱是陆地植物面临的严重非生物胁迫之一,干旱环境会造成植物水分缺失从而产生高渗胁迫,OSCA1是植物中第一个被鉴定的渗透感受器,OSCA基因在不同物种中广泛存在。植物对干旱胁迫的感受和响应对其生长发育至关重要,番茄作为经济作物和模式植物在蔬菜研究中具有重要作用,番茄抗旱性研究是保障蔬菜产量的重要途径,因此,研究番茄SlOSCA基因家族在番茄应对干旱胁迫过程中的生理功能方面具有重要意义。本研究以反向遗传学为基础,通过SlOSCA基因家族在叶片中的表达量分析,选定SlOSCA1.1/1.2/1.6/2.3/3.1共5个基因作为番茄抗旱研究的目标基因,利用CRISPR/Cas9定点基因编辑技术构建番茄slosca突变体,以探索番茄SlOSCA蛋白家族成员在番茄响应渗透胁迫过程中的作用。研究结果显示,番茄slosca3.1/1.1突变体在细胞水平对渗透胁迫的响应减弱,SlOSCA3.1/1.1参与了番茄保卫细胞对渗透胁迫的响应过程;番茄slosca1.6/1.2/2.3突变体整株水平对渗透胁迫的响应能力减弱,SlOSCA1.6/1.2/2.3参与了番茄对PEG渗透胁迫以及土壤干旱胁迫的响应过程,再次证明OSCA蛋白家族成员之间存在功能冗余。本研究通过构建slosca突变体,研究slosca突变体的生理表型,揭示SlOSCA蛋白家族成员协调作用于番茄抗旱过程,为后续解析OSCA基因家族成员在不同胁迫条件和不同生理过程中的生物学功能提供参考。

Drought is one of the most serious abiotic stresses to land plants. Environmental water deficiency triggers an osmotic stress signaling cascade. OSCA1 was described as a hyperosmolarity-gated calcium-permeable channel and its function in osmosensing in plants, homologues of OSCA genes are found throughout eukaryotes. Water is crucial to plant growth and development. As one of the primary horticulture commodities, tomato plays an important role in vegetable research. To enhance the survival and yield of tomatoes in drought conditions, it is essential to thoroughly study the function of SlOSCA genes and their drought-resistant mechanisms. Based on reverse genetics, we selected SlOSCA1.1/1.2/1.6/2.3/3.1 as our target genes through bioinformatics analysis, the slosca mutant of tomato was made by CRISPR/cas9-mediated gene editing directly in AC (Ailsa Craig). To explore the role of tomato SlOSCA protein family members in the response to osmotic stress. Here, we found impaired guard cell osmotic stress signaling and attenuated plant responses to osmotic stress in slosca3.1/1.1 plants, impaired osmotic stress signaling and attenuated plant responses to osmotic stress in slosca1.6/1.2/2.3 plants, the slosca1.6/1.2/2.3 plants showed lower water use efficiency and poorer drought resistance, there is genetic redundancy in OSCA. In this study, slosca mutants were constructed and then the physiological phenotypes of the mutants were investigated, which revealed that multiple genes of the SlOSCA family cooperatively regulate responses to osmotic stress, this study provides a reference for subsequent analysis of the biological functions of OSCA gene family under different stress conditions and different physiological processes.

植物作为固着生物,整个生长发育周期需要应对持续变化的环境。环境中常见的非生物胁迫包括干旱、高温、低温、涝害以及高盐和重金属等。干旱环境会对植物造成渗透胁迫,影响植物地理分布并限制农业作物生长发育[1-2]。为了减少干旱胁迫引起的损害,植物进化出了不同的抵抗机制[3]
植物应对渗透胁迫的信号传导途径包括胁迫感知、信号传导和对胁迫做出应答3个阶段[4-6]。首先,植物通过细胞膜上特定的受体感受器感知渗透胁迫。植物感知到渗透胁迫后,胞外Ca2+快速通过钙离子通道进入细胞内,引起胞内Ca2+浓度的变化[1]。胞浆内Ca2+浓度变化引发植物渗透胁迫信号级联反应,从而导致植物体内活性氧和脱落酸的积累[7]。最后,这些胁迫产生的物质有助于调节植物适应不利环境。迄今为止,植物遭受渗透胁迫时的信号传导过程已被深入研究,渗透胁迫响应途径主要包含ABA依赖和非ABA依赖[8],但植物感知渗透胁迫的分子机制尚不清晰。
在拟南芥中,OSCA1是一种高渗门控钙离子通道,定位在细胞质膜上第一个被发现的植物渗透感受器,其功能缺失的osca1突变体表现为高渗条件下对Ca2+的调节能力减弱,从而导致植株根系生长减少和叶片蒸腾作用下降[7]。拟南芥中OSCA1包含15个OSCA家族成员,AtOSCA1.2和AtOSCA1.1具有高同源性,是渗透诱导激活的钙离子通道蛋白[9];AtOSCA1.3和AtOSCA1.7的作用已在病原体相关分子模式(PAMP)触发的气孔免疫中确立[10-11];AtOSCA2.1和AtOSCA2.2属于低渗传感器[12]。OSCA1作为第一个报道的渗透感受器,已在多种植物中鉴定出OSCA基因,如籼稻和短梗稻(11个基因)、光稃稻(12个基因)、梨(16个基因)、绿豆(13个基因)、玉米(12个基因)、陆地棉(35个基因)、植物棉(21个基因)和瑞蒙棉(22个基因)等[13-18]。研究表明OSCA基因家族在植物不同物种的发育和应激反应中发挥作用,如玉米12个OSCA成员中的ZmOSCA4.1对干旱胁迫有反应[16],但是只有少数的OSCA家族成员的功能得到验证。此外,目前还没有关于番茄OSCA家族成员生理功能的研究。
番茄是一种重要的经济作物,其生长发育过程易受渗透胁迫影响,导致其产量和品质下降。因此,研究番茄如何响应渗透胁迫具有重要意义。OSCA在植物响应逆境胁迫的信号通路中发挥着重要作用,且OSCA蛋白家族成员之间存在功能冗余。通过番茄多基因敲除手段,可以较大程度降低SlOSCA功能类似蛋白成员共同发挥的生理作用。为了探索番茄SlOSCA家族成员是否响应渗透胁迫,本研究通过预测番茄SlOSCA在叶片中的表达量,利用CRISPR/Cas9基因编辑技术设计番茄SlOSCA3.1/1.1/1.6/1.2/2.3多基因敲除,并通过对突变体进行细胞水平和整株水平的渗透胁迫处理,验证番茄叶片表达的SlOSCA3.1/1.1SlOSCA1.6/1.2/2.3基因是否参与番茄对渗透胁迫的响应过程。本研究为后续深入研究番茄SlOSCA家族的生物学功能奠定基础,为通过基因工程和分子育种提高番茄抗旱性研究提供参考。
本研究所用野生型番茄Ailsa Craig(AC)由湖南农业大学熊程老师课题组提供,本实验室繁殖育种。番茄植株在光照16 h/黑暗8 h,白天26 ℃/夜间18 ℃,相对湿度50%~60%的人工控温生长室中生长。
大肠杆菌DH5α、农杆菌GV3101、KOD高保真酶等购自诺唯赞生物科技股份有限公司;PGR菌液、pTX041载体质粒和Gate8质粒由湖南农业大学熊程老师实验室提供;BsaⅠ和T4 DNA ligase等购自纽英伦生物技术(北京)有限公司;山梨醇、MES、CaCl2、KCl、C4H11NO3、PEG 6000、Trizma、蔗糖和琼脂等购自默克生命科学股份有限公司;MS盐、84消毒液、IBA、玉米素、卡那霉素、特美汀、肌醇、盐酸硫胺素、2,4-D、氯仿、异戊醇、异丙醇、乙醇等购自国产试剂公司。
在植物膜蛋白数据库(https://aramemnon.botanik.uni-koeln.de/index.ep)的拟南芥OSCA基因家族成员与不同物种的OSCA基因家族聚类结果中分析获得番茄OSCA家族基因。下载番茄OSCA蛋白序列和拟南芥OSCA蛋白序列,使用MEGA软件进行序列比对,构建系统发育树,利用AI软件进行颜色标注。通过https://bar.utoronto.ca/eplant_tomato/网站预测番茄OSCA家族基因在不同组织部位的表达量,分析SlOSCA在Heinz和M82两个番茄品种叶片中的表达量以及叶片表达量与不同组织总量的比值,选择叶片表达量高及其与总量比值高的基因作为进一步的研究对象。
利用CRISPR/Cas9构建双基因敲除(SlOSCA3.1/1.1)和3基因敲除(SlOSCA1.6/1.2/2.3)突变体。在http://crispr.hzau.edu.cn/CRISPR2/网站上以中靶率大于50%,脱靶率小于50%为筛选条件在外显子区域选择SlOSCA基因的靶位点,每个目标基因选择2个靶位点。参照XIE等[19]多基因编辑的方法,合成双基因敲除和三基因敲除sgRNA-tRNA表达盒。通过Golden-Gate组装法将sgRNA-tRNA表达盒组装到CRISPR/Cas9二元载体pTX041上[20]。将敲除载体转入GV3101农杆菌后转化AC番茄愈伤组织,待转基因番茄出芽后用卡那霉素进行生根筛选,获得转基因植株。
从转基因植株中剪取新鲜叶片,提取基因组DNA。根据靶序列位置设计特异性引物,以叶片DNA为模板进行PCR扩增,Sanger测序鉴定靶点位置基因是否被编辑,靶点被编辑的植株为阳性植株T1。T2和T3代植株使用上述方法鉴定靶点突变类型,并使用特异性引物Cas9-F和Cas9-R进行PCR扩增,对照组以野生型DNA为模板,将其PCR产物进行核酸电泳,筛选出不含Cas9基因的纯合突变体植株。
待番茄幼苗长有4~6片真叶时,取幼苗自下而上的第2~4片叶片,撕取该叶片表皮置于含有气孔打开溶液(100 μmol/L CaCl2, 5 mmol/L KCl, 10 mmol/L MES-Tris, pH 6.15)的微孔室中,于250 μmol/(m2·s)强光照下孵育2.5 h,待叶表皮上的气孔完全打开,使用蔡司倒置荧光显微镜拍摄表皮光学图像。在微孔室中加入250 mmol/L山梨醇溶液处理叶片表皮3 min,使用蔡司倒置荧光显微镜拍摄处理后的表皮光学图像。使用Image J软件测量表皮光学图像中气孔的宽度。
待番茄幼苗在底部有孔的塑料盆中长至4片真叶时,将地下部分浸入40%(W/V)PEG-6000的溶液中,处理1 h。拍摄植物PEG处理前和处理1 h的照片用于进一步的图像分析,使用Image J软件测量植株的叶面积和植株茎秆向下倾斜的角度。
待番茄幼苗长有4~6片真叶时,将番茄幼苗分为实验组和对照组进行干旱胁迫实验。干旱胁迫处理前实验组和对照组浇水至土壤含水量的50%~55%,用土壤水分仪测量土壤含水量。进行干旱胁迫处理时,对照组每天上午9:00浇水至土壤含水量的50%~55%,实验组不浇水。干旱胁迫处理发生12 d后,取实验组和对照组植株自下而上面积相似的叶片,称量并记录鲜重(FW),再将叶片置于5 mmol/L CaCl2溶液中,转入黑暗下孵育8 h,称量膨胀叶片的重量(TW),最后置于70 ℃烘箱干燥48 h称量干重(DW)。根据公式RWC=(FW-DW)/(TW-DW)×100%进行叶片相对含水量的计算。
利用植物膜蛋白数据库(https://aramemnon.botanik.uni-koeln.de/index.ep)筛选番茄OSCA离子通道膜蛋白家族,Cluster结果显示番茄的同源OSCA蛋白家族包含12个成员(图1),将SlOSCA基因分别命名为SlOSCA1.1-1.6SlOSCA2.1-2.4SlOSCA3.1SlOSCA4.1。番茄和拟南芥OSCA基因家族进化树分析结果如图2所示,OSCA基因家族成员可分为4个亚族,番茄OSCA基因家族在4个亚族均有分布。
通过植物生物学BAR数据库(https://bar.utoronto.ca/eplant_tomato/)预测番茄SlOSCA基因家族在番茄不同组织部位的表达量,叶片是植物整个生长周期中应对干旱胁迫的重要组织部位。基因表达量高、低与其发挥功能之间不是线性关系,但是基因时空表达水平是其发挥生理作用功能的前提。关于番茄SlOSCA基因家族参与番茄响应干旱胁迫过程的研究,在选定目标研究基因时,优先选择表达量较高的SlOSCA基因家族成员是常规方法,而随机选择基因或选择表达量较低的基因都是不恰当的。
SlOSCA基因转录水平结果显示,SlOSCA1.1SlOSCA1.6SlOSCA2.3SlOSCA3.1在番茄品种Heinz和M82叶片中的表达量均较高(图3)。另外,把SlOSCA基因在叶片中的表达量与该基因在不同组织中总表达量的比值定义为叶片相对表达量,SlOSCA1.1SlOSCA1.2在番茄Heinz品种和番茄M82品种叶片中的相对表达量均较高(图4)。因此,选择SlOSCA1.1SlOSCA1.2SlOSCA1.6、SlOSCA2.3SlOSCA3.1作为目标基因以探索番茄SlOSCA家族成员在番茄抗旱反应中的作用机理。
根据番茄SlOSCA基因家族表达量分析结果,选定SlOSCA1.1SlOSCA1.2SlOSCA1.6SlOSCA2.3SlOSCA3.1作为目标研究基因,研究SlOSCA蛋白家族参与番茄抗旱响应过程。反向遗传学的研究过程是通过基因工程手段创造突变体,以突变体表现出来的生理表型来解析基因的功能,CRISPR/Cas9基因敲除技术常用于反向遗传学研究中。AtOSCA1.1作为高渗感受器在植物早期响应渗透胁迫的钙信号传导过程中占主要作用,但是植物在干旱胁迫下的生长调控过程是复杂的,为了避免OSCA蛋白家族的功能冗余问题,在利用5个番茄SlOSCA目标研究基因构建单基因敲除突变体(5个)、双基因敲除突变体(10种组合)、三基因敲除突变体(10种组合)、四基因敲除突变体(5种组合)以及五基因敲除突变体的众多选择中,优先设计构建多基因敲除突变体以作为番茄SlOSCA蛋白家族抗旱生理功能研究的开端。通过番茄SlOSCA多基因敲除手段,可以较大程度降低SlOSCA功能类似蛋白成员共同发挥的生理作用,有利于敲除突变体表现出较好的生理表型。
拟南芥AtOSCA1.1[7]和AtOSCA3.1[21]是已知的直接参与植物应对干旱胁迫的响应因子,因此设计番茄SlOSCA1.1/3.1双基因敲除,把余下的3个目标基因设计SlOSCA1.2/1.6/2.3多基因敲除,以开启SlOSCA家族成员参与番茄抗旱生理过程的研究。通过网站(http://crispr.hzau.edu.cn/CRISPR2/)设计基因编辑靶点,根据SlOSCA3.1SlOSCA1.1SlOSCA1.6SlOSCA1.2SlOSCA2.3基因序列信息,在每个目标基因的外显子区域设计2个Target基因编辑靶点以提高基因编辑效率,成功构建SlOSCA3.1/1.1双基因敲除表达载体和SlOSCA1.6/1.2/2.3多基因敲除表达载体(图5)。
SlOSCA3.1/1.1双基因敲除表达载体和SlOSCA1.6/1.2/2.3多基因敲除表达载体转入农杆菌,把包含表达载体的农杆菌转化到番茄愈伤组织中,待转基因番茄出芽后,用Kana抗生素进行阳性苗筛选,共获得30株SlOSCA3.1/1.1番茄转基因植株和56株SlOSCA1.6/1.2/2.3番茄转基因植株。利用CTAB法提取番茄转基因植株叶片的DNA,PCR扩增包含基因编辑靶点在内的转基因番茄基因序列,将PCR产物进行测序,以检测靶点基因序列是否被成功编辑。
把包含基因编辑靶点在内的转基因番茄基因序列与对应的野生型番茄SlOSCA基因序列进行比对分析,成功获得16株slosca3.1/1.1突变植株和24株slosca1.6/1.2/2.3突变植株,所获得突变植株均为杂合子,SlOSCA3.1/1.1双基因敲除载体和SlOSCA1.6/1.2/2.3多基因敲除载体的基因编辑效率分别是53%和42%。部分突变植株的基因编辑情况如图6所示,slosca3.1/1.1番茄突变植株(图6A)和slosca1.6/1.2/2.3番茄突变植株(图6B)中存在碱基突变、缺失和插入,且存在大片段碱基缺失和插入情况。
通过繁殖番茄杂合子突变植株,在发生分离的后代中验证获得slosca3.1/1.1slosca1.6/1.2/2.3纯合子突变植株。为了避免后续繁殖过程中,番茄纯合子突变植株中CRISPR/Cas9系统发生额外的基因编辑事件,利用Cas9-F/Cas9-R引物PCR扩增检测slosca3.1/1.1slosca1.6/1.2/2.3纯合子突变株系中是否包含Cas9基因,筛选出不包含Cas9基因的纯合子突变植株(图6C),以开展番茄slosca3.1/1.1slosca1.6/1.2/2.3突变植株的生理表型研究。
植物作为固着生物,整个生长周期需要应对持续的环境变化,其中高渗胁迫会导致植物气孔闭合以及光合作用降低,从而影响生长发育。使用250 mmol/L山梨醇(sorbitol)溶液对番茄叶片表皮进行高渗胁迫处理,在高渗胁迫处理后野生型(WT)和slosca3.1/1.1突变体的气孔均出现闭合情况(图7A)。正常条件下,slosca3.1/1.1突变体气孔与野生型气孔的开度不存在差异;高渗胁迫处理条件下,slosca3.1/1.1突变体气孔比野生型气孔的开度大,存在显著差异(图7B)。结果表明,番茄slosca3.1/1.1突变体对渗透胁迫的响应减弱,SlOSCA3.1/1.1参与了番茄对渗透胁迫的响应过程。
使用40%(W/V)PEG-6000溶液对番茄植株的地下部分进行渗透胁迫处理,在PEG处理1 h后野生型(WT)和slosca1.6/1.2/2.3突变体植株均出现叶片失水、茎秆倾斜等萎蔫情况,番茄slosca1.6/1.2/2.3突变体植株的萎蔫程度较野生型更加明显(图8A)。正常条件下,slosca1.6/1.2/2.3突变体与野生型的叶面积不存在差异;在PEG渗透胁迫处理条件下,slosca1.6/1.2/2.3突变体植株失水速率快致使其叶面积明显小于野生型(图8B)。在渗透胁迫下,植物缺少水分导致细胞膨压降低,植物株型呈萎缩状。用番茄植株主茎干偏离重力方向的倾斜角度作为植物萎蔫的指标,将植物萎蔫程度数据化,正常条件下,slosca1.6/1.2/2.3突变体与野生型主茎干的倾斜角度不存在差异;在PEG渗透胁迫处理条件下,slosca1.6/1.2/2.3突变体植株主茎干的倾斜角度明显大于野生型(图8C),表明slosca1.6/1.2/2.3突变体植株萎蔫程度更严重。
对番茄整株水平进行干旱胁迫处理,在干旱处理12 d后野生型(WT)和slosca1.6/1.2/2.3突变体植株均出现萎蔫情况(图9A)。正常条件下,slosca1.6/1.2/2.3突变体与野生型的叶片含水量不存在差异;在干旱胁迫处理下,slosca1.6/1.2/2.3突变体与野生型的叶片含水量均降低,其中slosca1.6/1.2/2.3突变体的叶片含水量显著低于野生型,说明slosca1.6/1.2/2.3突变体的水分控制能力更差(图9B)。
综上表明,番茄slosca1.6/1.2/2.3突变体对渗透胁迫的响应能力减弱,SlOSCA1.6/1.2/2.3参与了番茄对渗透胁迫的响应过程。
番茄是广泛栽培的蔬菜作物和模式作物,在研究蔬菜品质和生长发育过程中发挥着重要作用。本研究以反向遗传学为基础,通过基因编辑技术,成功获得番茄slosca3.1/1.1slosca1.6/1.2/2.3多基因敲除突变体。对番茄突变体进行细胞水平或整株水平的渗透胁迫处理,发现SlOSCA3.1/1.1SlOSCA1.6/1.2/2.3参与番茄对渗透胁迫的响应过程,表明OSCA蛋白家族成员在番茄抗旱过程中发挥着重要作用。
OSCA1是植物中第一个被鉴定的渗透感受器,拟南芥AtOSCA家族成员可分为4个亚族,植物膜蛋白数据库信息显示番茄SlOSCA家族包含12个成员,系统进化树结果显示SlOSCA家族同样可分为4个亚族,表明OSCA家族在遗传上具有保守性。已有研究表明,AtOSCA1.1和AtOSCA1.2参与高渗胁迫信号传导[7,9],AtOSCA1.3是干旱胁迫的早期响应因子[10],说明OSCA家族成员之间存在功能冗余。为了较大程度降低SlOSCA功能类似蛋白成员共同发挥的生理作用,本研究利用CRISPR/Cas9技术构建slosca3.1/1.1slosca1.6/1.2/2.3多基因敲除突变体,获得了多种突变类型的突变体,不仅有简单的碱基缺失、插入和替换的突变体,而且有大片段缺失和插入的突变体。在敲除突变体获得的过程中,发现多靶点敲除载体可以提高目的基因被编辑的效率,虽然SlOSCA3.1/1.1敲除载体的靶点2编辑效率较低以及SlOSCA1.6/1.2/2.3敲除载体的靶点5完全脱靶,但在其他靶点的加持下成功获得了目的研究基因的敲除突变体,且目标基因2个靶点相距近的获得了大片段缺失的突变体。
OSCA基因家族在多个物种中均有研究,但只有少数植物的OSCA基因在功能上得到验证。LIU等[1]在大豆基因组中鉴定出20个大豆GmOSCA家族成员,异源表达GmOSCA家族成员进行生物学功能研究,发现GmOSCA1.1GmOSCA1.2GmOSCA1.3GmOSCA1.4GmOSCA1.5可能为大豆中的钙渗透传感器,过表达GmOSCA1.1-1.5GmOSCA3.1GmOSCA3.2增强了转基因拟南芥的抗旱性。玉米ZmOSCA家族基因跨膜结构域和系统进化分析表明,12个成员分为4个亚族且都包含DUF221结构域,对12个家族成员进行PEG、ABA和NaCl处理,大部分基因响应渗透胁迫,在拟南芥中过表达ZmOSCA2.4基因,拟南芥抗旱性显著提高且与抗旱相关基因的表达水平显著上升[22]OsOSCA1.1介导水稻根系中的高渗胁迫和盐胁迫诱导的细胞质内钙离子浓度增加,响应高渗胁迫和盐胁迫[21]。上述研究表明,OSCA作为渗透敏感钙离子通道蛋白,在植物应对渗透胁迫过程中发挥的功能是保守的。
番茄作为重要的经济蔬菜作物,品质和产量易受到干旱胁迫影响。叶片是植物应对干旱胁迫最重要的组织部位,干旱胁迫发生时植物通过调整叶片的形态结构来减少水分散失,从而降低机械损伤,并提高水分利用率和增强耐旱能力[23-24]。在本研究中,通过对slosca3.1/1.1slosca1.6/1.2/2.3突变体进行渗透胁迫处理发现,与野生型相比,slosca3.1/1.1植株气孔关闭程度减弱,slosca1.6/1.2/2.3植株表现出叶面积减小,萎蔫程度更加严重,叶片相对含水量更低;slosca3.1/1.1slosca1.6/1.2/2.3突变体对渗透胁迫的响应能力减弱,表明番茄SlOSCA3.1/1.1SlOSCA1.6/1.2/2.3基因在番茄干旱胁迫应答中起着重要作用。为了更全面地阐明番茄叶片表达的SlOSCA基因在番茄渗透胁迫中的生物学功能,本课题组后续将会从杂合子敲除突变体发生分离的后代中筛选slosca1.1slosca1.2slosca1.6slosca2.3slosca3.1单基因敲除突变体以及slosca1.6/1.2slosca1.6/2.3slosca1.2/2.3双基因敲除突变体,并对其进行保卫细胞渗透胁迫处理、PEG渗透胁迫处理以及干旱胁迫处理,用slosca3.1/1.1slosca1.6/1.2/2.3突变体对渗透敏感的表型作为参照,研究SlOSCA1.1/1.2/1.6/2.3/3.1在番茄抗旱过程中发挥作用功能的强弱。
本研究通过构建番茄slosca突变体,研究SlOSCA蛋白在番茄响应渗透胁迫过程中的功能,证明SlOSCA蛋白家族成员在番茄抗旱中发挥重要作用,有助于推进OSCA基因家族成员在其他蔬菜作物和农作物抗逆生理过程中的功能研究。
