This study aims to screen the microbial strains capable of producing β-D-glucosidase from fermented vanilla pods and apply the screened strains to the fermentation process of vanilla with the goal of enhancing vanillin production and improving the quality of vanilla bean. Septenary selective medium was utilized to isolate β-D-glucosidase producing bacteria, identify the homology of each bacterial strain and measure the production of β-D-glucosidase activity of each strain on two strains with higher enzyme production. The phylogenetic tree was constructed. The two strains of bacteria with the highest enzyme production in different combinations were sprayed onto the fermented vanilla pods. The content of vanillin, vanillin glucoside, and volatile components in different treatments of β-D-glucosidase producing bacteria were determined. Nine strains were successfully screened from the vanilla pods originating in Madagascar (M), and twelve strains were isolated from the vanilla pods produced by Spice and Beverage Research Institute, Chinese Academy of Tropical Agricultural Sciences (S). The fifteen strains were detected as Bacillus. M3 and S7 had higher enzyme activities than other isolates. Vanilla pods treated with M3 obtained the highest vanillin content, which reached to 2.25% and possed the highest aroma components. The maximum decrease in vanillin glucoside content was 80.7% when sprayed with M3 and S7 in a volume ratio of 1:1. There were 34 volatile substances in the fermented pods sprayed with M3, making it the most volatile combination of spraying bacteria. In the present study, we successfully isolated β-D-glucosidase producing bacteria for improving the quality of vanilla pods fermentation.

本研究从发酵后的香草兰豆荚中筛选出产β-D-葡萄糖苷酶的微生物菌株,并将筛选出的菌株应用到香草兰发酵过程中,以提高发酵豆荚品质。采用七叶苷选择培养基传统分离培养方法,从发酵后的香草兰豆荚中筛选出可产β-D-葡萄糖苷酶的菌株,鉴定每株菌的菌株同源性,测量每株菌产β-D-葡萄糖苷酶酶活,绘制2株产酶较高的菌株系统发育树,并将产酶最高的2株菌按照不同组合喷洒至发酵中的香草兰豆荚中,测定其发酵豆荚香兰素含量、香兰素葡萄糖苷含量和挥发性成分。结果表明:从产自马达加斯加(M)的香草兰发酵豆荚中共筛选出9株菌、产自中国热带农业科学院香料饮料研究所(S)的香草兰发酵豆荚中筛选到12株菌,检测出的15株菌均为芽孢杆菌,其中M3、S7的β-D-葡萄糖苷酶活力较高。使用M3、S7以不同组合添加喷菌发酵试验的豆荚中,只喷M3的发酵豆荚香兰素含量最高为2.25%,M3与S7按照体积比1∶1喷洒处理的香兰葡萄糖苷含量下降最多,为80.7%,只喷M3的发酵豆荚鉴定出挥发性物质34种。本研究成功筛选出产β-D-葡萄糖苷酶的功能微生物,并利用筛选出的微生物辅助香草兰豆荚发酵生香,可用于提升香草兰豆荚品质。

本研究从发酵后的香草兰豆荚中筛选出产β-D-葡萄糖苷酶的微生物菌株,并将筛选出的菌株应用到香草兰发酵过程中,以提高发酵豆荚品质。采用七叶苷选择培养基传统分离培养方法,从发酵后的香草兰豆荚中筛选出可产β-D-葡萄糖苷酶的菌株,鉴定每株菌的菌株同源性,测量每株菌产β-D-葡萄糖苷酶酶活,绘制2株产酶较高的菌株系统发育树,并将产酶最高的2株菌按照不同组合喷洒至发酵中的香草兰豆荚中,测定其发酵豆荚香兰素含量、香兰素葡萄糖苷含量和挥发性成分。结果表明:从产自马达加斯加(M)的香草兰发酵豆荚中共筛选出9株菌、产自中国热带农业科学院香料饮料研究所(S)的香草兰发酵豆荚中筛选到12株菌,检测出的15株菌均为芽孢杆菌,其中M3、S7的β-D-葡萄糖苷酶活力较高。使用M3、S7以不同组合添加喷菌发酵试验的豆荚中,只喷M3的发酵豆荚香兰素含量最高为2.25%,M3与S7按照体积比1∶1喷洒处理的香兰葡萄糖苷含量下降最多,为80.7%,只喷M3的发酵豆荚鉴定出挥发性物质34种。本研究成功筛选出产β-D-葡萄糖苷酶的功能微生物,并利用筛选出的微生物辅助香草兰豆荚发酵生香,可用于提升香草兰豆荚品质。

This study aims to screen the microbial strains capable of producing β-D-glucosidase from fermented vanilla pods and apply the screened strains to the fermentation process of vanilla with the goal of enhancing vanillin production and improving the quality of vanilla bean. Septenary selective medium was utilized to isolate β-D-glucosidase producing bacteria, identify the homology of each bacterial strain and measure the production of β-D-glucosidase activity of each strain on two strains with higher enzyme production. The phylogenetic tree was constructed. The two strains of bacteria with the highest enzyme production in different combinations were sprayed onto the fermented vanilla pods. The content of vanillin, vanillin glucoside, and volatile components in different treatments of β-D-glucosidase producing bacteria were determined. Nine strains were successfully screened from the vanilla pods originating in Madagascar (M), and twelve strains were isolated from the vanilla pods produced by Spice and Beverage Research Institute, Chinese Academy of Tropical Agricultural Sciences (S). The fifteen strains were detected as Bacillus. M3 and S7 had higher enzyme activities than other isolates. Vanilla pods treated with M3 obtained the highest vanillin content, which reached to 2.25% and possed the highest aroma components. The maximum decrease in vanillin glucoside content was 80.7% when sprayed with M3 and S7 in a volume ratio of 1:1. There were 34 volatile substances in the fermented pods sprayed with M3, making it the most volatile combination of spraying bacteria. In the present study, we successfully isolated β-D-glucosidase producing bacteria for improving the quality of vanilla pods fermentation.

香草兰(Vanilla planifolia Andrews)又名香荚兰、香子兰,属多年生大型肉质藤本香料植物,是兰科植物中最有实用价值的优质食用香料,享有天然食品香料之王的美誉,单价仅次于藏红花[1-3]。新鲜成熟的香草兰豆荚无特殊香气,其豆荚经杀青、发汗、干燥、陈化等工艺后可产生香兰素、香草酸、香草醇等,据报道其芳香成分可达250多种,是各种高端食品饮料及化妆品等优质原料,广泛应用于调配各级高档食品、化妆品等,在国际市场上供不应求[4-10]
香草兰发酵后的众多芳香成分中,最受瞩目及最能反映豆荚品质的香味物质是香兰素[11],其由香草兰豆荚中的β-D-葡萄糖苷酶转化香兰素葡萄糖苷生成[12-14],因此在发酵过程中香兰素葡萄糖苷含量逐渐降低,香兰素含量逐渐提升。香草兰鲜豆荚中香兰素葡萄糖苷含量达10%以上,但加工后豆荚中香兰素含量仅占1%~3%,香兰素葡萄糖苷并不能完全转化为香兰素。浦帆等[15]研究发现添加β-D-葡萄糖苷酶,酶促生香效果明显,可提高豆荚中香兰素含量。莫丽梅[16]在香草兰青豆荚中添加β-D-葡萄糖苷酶、果胶酶、纤维素酶,处理后香兰素含量有明显提升。张彦军等[17]使用冷冻-联合纤维素酶、果胶酶、β-D-葡萄糖苷酶方法处理香草兰鲜豆荚,显著提高香兰素转化率。WALISZEWSKI等[18]使用结晶酶PML-MX、Zymafilt L-300和Novozym 3种纤维素分解酶产物对香草兰豆荚进行预脱水和酶促预处理,结果表明酶促预处理使香兰素含量增加1倍。以上研究均可证明额外添加酶,尤其是β-D-葡萄糖苷酶对提高香兰素含量起着重要作用,但酶价格昂贵,而微生物已被证明参与香草兰发酵过程[19],且微生物来源获得高纯度β-D-葡萄糖苷酶制剂的制备工艺较为简单、酶促效果更佳[20],从香草兰豆荚中筛选的微生物应用到香草兰发酵中也更安全可靠。陈永敢[21]使用产β-D-葡萄糖苷酶的芽孢杆菌辅助豆荚发酵生香,明显提高了香草兰豆荚中香兰素含量。
本研究以产自马达加斯加香草兰发酵豆荚(M)和中国热带农业科学院香料饮料研究所香草兰发酵豆荚(S)为原料,使用七叶苷选择性培养基,筛选产β-D-葡萄糖苷酶的微生物菌株,选择酶活较高的2株菌按照不同比例混合,喷洒到香草兰豆荚表面,使用HPLC技术和GC-MS技术测定发酵豆荚的香兰素、香兰素葡萄糖苷含量和挥发性物质,旨在利用微生物辅助香草兰豆荚发酵生香,为提升豆荚品质提供支撑。
供试香草兰豆荚:产自马达加斯加的香草兰商品豆荚(M)和产自中国热带农业科学院香料饮料研究所的香草兰商品豆荚(S)。
七叶苷麦克林试剂,香兰素标准品,上海源叶生物科技有限公司;香兰素葡萄糖苷标准品,阿拉丁试剂公司;C7~C30烷烃标准样品,美国Sigma公司。
初筛培养基:参考高娉娉等[22]的方法,称取氯化钠10 g,胰蛋白胨10 g,酵母提取物5 g,琼脂15 g,七叶苷1 g,柠檬酸高铁氨2.5 g,蒸馏水1000 mL,121 ℃高压蒸汽灭菌20 min,备用。LB培养基:氯化钠10 g,胰蛋白胨10 g,酵母提取物5 g,琼脂15 g,蒸馏水1000 mL,121 ℃高压蒸汽灭菌20 min,备用(发酵条件优化用液体培养基不加琼脂)。发酵培养基:参考崔萍等[23]的方法,酵母浸膏1.0%(氮源),葡萄糖2.0%(碳源),蛋白胨2.0%(氮源和碳源),NH4NO3 0.3%(氮源)、KH2PO4 0.4%,自然pH,115 ℃高压蒸汽灭菌30 min,备用。
分别将M和S 2种香草兰豆荚用无菌剪刀剪短至约0.2 cm长的小段,取剪短后的豆荚5 g置于45 mL无菌水中,180 r/min震荡1 h,静置30 min后,将上清液进行梯度稀释,选取10–5、10–6、10–7稀释液,分别吸取0.2 mL稀释液,均匀涂布至初筛培养皿上,每梯度设置3次重复,在28 ℃的培养箱中倒置培养48 h。待长出菌落后,根据菌落形态、颜色,挑选可产生明显黑色水解圈的单菌落,将单菌落在LB培养皿上划线。连续纯化5次后挑取单菌落接种至LB培养基斜面,置于4 ℃冰箱保存备用。
菌株16S rDNA分子鉴定参考尚方剑等[24]的方法。采用DNA离心柱型试剂盒提取菌株的DNA,将总DNA作为模板,将细菌特异性引物27F(5′-AGAGTTTGATCCTGGCTCAG-3′)和1492R(5′-GGACTACNVGGGTWTCTAAT-3′)对DNA的内转录间隔区进行PCR扩增。将纯化后的产物送至上海派森诺生物技术有限公司进行测序。
酶活测定参考王彩肖等[25]的方法。
将香草兰豆荚置于75 ℃水中杀青5 min,用无菌纱布将豆荚表面水分擦净。将M3、S7菌株接种至LB液体培养基中,将含菌液的培养基在28 ℃和180 r/min的恒温摇床中震荡培养48 h。在无菌环境下将菌液以8000 r/min离心10 min,弃上清液,再加入50 mL无菌水,充分摇匀后再次离心,重复2次以上操作,调整菌液吸光度约为0.5后,作喷菌处理,喷菌比例如表1所示。将菌液均匀喷洒在杀青后的豆荚表面,每组豆荚10根喷菌液10 mL,将豆荚置于干燥阴凉处分别发酵20 d和60 d后取样。测量分析前,将样品低温冻干粉碎并过60目筛。
参考杨峰等[13]的方法,称取0.05 g冻干香草兰豆荚粉末,加入50 mL无水乙醇,充分搅拌后进行超声萃取,设置超声功率100 W,50 ℃条件下萃取10 min;萃取后过滤,重复进行3次萃取,合并滤液,定容至50 mL容量瓶,测豆荚中的香兰素和香兰素葡萄糖苷含量,取萃取液用0.22 μm有机滤膜过滤后进行HPLC分析。分析条件:反相C18柱(Zorbax,4.6 mm×100 mm,3.5 μm,Agilent);流速为0.8 mL/min;进样量为5 μL;柱温为25 ℃;检测波长为280 nm;流动相为0.5%醋酸溶液∶乙腈=85∶15等梯度洗脱。采用香兰素标准品、香兰素葡萄糖苷标准品和出峰保留时间定性,外标法确定物质含量。
称取1 g冻干香草兰豆荚粉末于20 mL顶空进样瓶中,80 ℃振摇SPME萃取40 min,250 ℃解吸5 min。GC-MS采用DB–WAX(J&W Scientific,Folsom,CA,USA)石英毛细管柱(30 m×0.25 mm×0.25 μm),升温程序为:柱温40 ℃保持3 min,以3 ℃/min速率升温到90 ℃,以2 ℃/min速率升温到120 ℃,以3 ℃/min速率升温到240 ℃,保持20 min。样品进样不分流,电子冲击能量为70 eV,离子源和四极杆的温度分别设定在230 ℃和150 ℃。扫描范围为40~450 amu。
采用SPSS 26.0软件对数据进行方差分析;采用Origin 2019软件、MEGA 11.0软件和GenBank中的BLAST软件进行数据处理和绘图。
从发酵后的香草兰豆荚中筛选到21株可在七叶苷筛选培养基上产生黑色水解圈的菌株,各菌株菌落形态特征存在差异(图1),黑色水解圈大小不同,其中菌株M3、M7、S2、S3、S4、S7黑色水解圈较大。从产自马达加斯加香草兰豆荚上筛选到的菌株M3在LB培养基培养24 h后菌落呈圆形,乳白色,表面光滑湿润,边缘整齐。从产自香饮所的香草兰豆荚上筛选到的菌株S7菌落形态与M3相似。
经16S rRNA序列测定后,用BLAST与GenBank中16S rDNA序列进行同源性比较。发现菌株M1、M3、M6、S1、S3、S7的16S rRNA序列与NCBI数据库中的B. paramycoides MCCC 1A04098、L. macroides LMG 18474、B. subtilis DSM 10、B. altitudinis 41KF2b、B. rugosus SPB7、B. paramycoides MCCC 1A04098相似度分别达到99.79%、99.52%、99.52%、99.93%、99.93%、99.86%(表2)。除6株菌未检测出外,其余15株菌NCBI比对分析结果表明均为芽孢杆菌,其中,M1、M3、M6、S1、S3、S7分别为副蕈状芽孢杆菌(Bacillus paramycoides)、长形氨酸芽孢杆菌(Lysinibacillus macroides)、枯草芽孢杆菌(B. subtilis)、高空芽孢杆菌(B. altitudinis)、胶质芽孢杆菌(B. altitudinis)、副蕈状芽孢杆菌(B. paramycoides)。对菌株M3及S7的测序结果建立系统发育树,结果如图2所示,进一步验证M3为Lysinibacillus macroides,S7为B. paramycoides
鉴定出的15株芽孢杆菌产β-D-葡萄糖苷酶能力如图3所示,菌株产酶活性在0.00294~0.01117 U/mL。分离自马达加斯加香草兰豆荚上的长形赖氨酸芽孢杆菌M3酶活性最高,是副蕈状芽孢杆菌M8酶活力的3.8倍。分离自香饮所香草兰豆荚上的菌株产酶活力在0.00599~0.00736 U/mL,酶活力变化范围低于分离自马达加斯加香草兰豆荚上的产酶菌株,其中菌株副蕈状芽孢杆菌S7酶活力高于本组中其他菌株酶活力。
图4可看出,每组处理喷菌60 d后的香兰素含量均高于20 d时的含量。CK在20、60 d时的香兰素含量分别为0.28%和0.92%。只喷M3菌株的处理在20 d时香兰素含量为0.49%,60 d高达2.25%,显著高于其他处理。只喷S7菌株的处理,在20、60 d时的香兰素含量分别为1.39%、1.52%,上升幅度不大。菌株M3、S7按照1∶1喷菌处理的豆荚20、60 d时香兰素含量分别为0.45%、1.29%,60 d相较于20 d的香兰素含量增加了186.7%。M3与S7按照2∶1喷菌处理的豆荚,在20、60 d时香兰素含量分别为1.57%、1.66%。M3与S7按照1∶2喷菌处理的豆荚发酵20、60 d时香兰素含量分别为1.60%、1.80%。综上,香兰素含量最高的是只喷M3菌株处理的豆荚。在20、60 d时所有喷菌处理的豆荚香兰素含量均高于CK,说明产β-D-葡萄糖苷酶的功能微生物可有效促进香兰素产生,提升发酵豆荚品质。
图5为不同处理香草兰豆荚分别在喷菌20、60 d后的香兰素葡萄糖苷含量。从图5中可得出,喷菌后的20~60 d内,豆荚香兰素葡萄糖苷含量呈下降趋势。喷无菌水豆荚在20、60 d时的香兰素葡萄糖苷含量分别为1.63%、0.61%,60 d相较于20 d的香兰素葡萄糖苷含量下降了62.6%。只喷M3菌株处理的豆荚发酵20、60 d时的香兰素葡萄糖苷含量分别为3.25%、0.93%,60 d相较于20 d的香兰素葡萄糖苷含量下降了71.4%。只喷S7菌株处理的豆荚发酵20、60 d时的香兰素葡萄糖苷含量分别为2.83%、0.88%,60 d相较于20 d的香兰素葡萄糖苷含量下降了68.9%。M3与S7按照体积比1∶1喷洒处理的发酵豆荚20、60 d时的香兰素葡萄糖苷含量分别为3.00%、0.58%,60 d的相较于20 d的香兰素葡萄糖苷含量下降了80.7%;M3与S7按照体积比2∶1喷菌处理的豆荚发酵20、60 d的香兰素葡萄糖苷含量分别为3.31%、0.66%,60 d的相较于20 d的香兰素葡萄糖苷含量下降了80.0%;M3与S7按照体积比1∶2喷菌处理的豆荚发酵20、60 d时香兰素葡萄糖苷含量分别为2.38%、0.79%,60 d的相较于20 d的香兰素葡萄糖苷含量下降了66.8%。由此可见,香兰素葡萄糖苷下降最多的是M3与S7按照体积比为1∶1喷菌处理的豆荚,而下降最少的是CK,这说明产β-D-葡萄糖苷酶的微生物菌株可显著促进香兰素葡萄糖苷水解。
采用GC-MS对喷菌处理发酵20 d和60 d时的豆荚进行挥发性成分分析,喷菌发酵20 d时检测出的挥发性物质共47种(表3),其中烷烃类11种、醛类12种、酸类6种、烯烃1种、醇类7种、酚类6种、酮类2种、其他类2种,占比最多的是醛类,占检测出所有物质的28%(图6A)。喷菌发酵60 d时豆荚的挥发性成分共有50种(表4),其中烷烃类有8种、醛类14种、酸类4种、烯烃4种、醇类10种、酚类5种、酯类1种、酮类2种、其他类2种,占比最多的依旧为醛类,占总物质种类数量的25.4%(图6B)。发酵20 d含量最高的物质是香兰素,其中M3与S7按照体积比1∶1喷菌的发酵豆荚香兰素含量占比最高为80.77%,其次是只喷菌M3菌株的豆荚,香兰素含量为78.49%,M3与S7按照体积比1∶2和2∶1喷菌处理的豆荚香兰素含量占比约为77.00%,CK香兰素含量最低。发酵60 d时含量最高的挥发物质也是香兰素,M3与S7按照体积比1∶2喷洒的香兰素含量最高,只喷S7处理与CK差异不大,其他3个喷菌处理香兰素含量均约为88.00%。豆荚发酵20 d时CK和只喷M3菌株的处理均检测到对羟基苯甲醛,且含量仅次于香兰素。对羟基苯甲醛是合成香兰素的重要前体物质,在发酵60 d时每个处理均检测到对羟基苯甲醛,且含量均大于1.00%。豆荚发酵20 d时单独喷菌M3、S7及二者1∶1混合的处理均检测到2,3-丁二醇。2,3-丁二醇是香草兰的特征风味物质,在发酵60 d时不同处理豆荚中均含有2,3-丁二醇,其中喷洒S7菌株处理的豆荚中含量高达2.57%,高于其他处理的豆荚。
图7所示,有18种挥发物在2个时间均有被检出,如香兰素、愈创木酚、乙酸、反-2-癸烯醛、香草乙酮等,且以上物质已被证明大量存在于香草兰豆荚发酵前期[14]。只喷M3菌株处理的豆荚60 d时检测出3-羟基丁酮,该物质是重要食品香料[26],具有独特香味,且其可在无其他碳源的情况下作为B. subtilis的碳源[21]
β-D-葡萄糖苷酶是纤维素酶系中一种重要的水解酶,可以催化风味前体糖苷配基(烃基或芳基)与糖基之间的糖苷键水解,产生芳香物质达到增香效果。微生物来源的β-D-葡萄糖苷酶数量庞大,价格低廉,制备容易且用途广泛[27]。本研究以香草兰豆荚为原料,分离出21株可产β-D-葡萄糖苷酶的菌株,并鉴定出其中15株菌均为芽孢杆菌。从产自马达加斯加的香草兰豆荚中筛选出的菌株中,产β-D-葡萄糖苷酶活力最高的菌株M3为长形赖氨酸芽孢杆菌(L. macroides)。从产自中国热带农业科学院香料饮料研究所的香草兰豆荚中筛选出的菌株中,酶活力最高的菌株S7为副蕈状芽胞杆菌(B. paramycoides)。鲍捷等[28]筛选到1株产β-D-葡萄糖苷酶的植物乳植杆菌c4-3,并使用该菌株用于豆浆的发酵,发现添加该菌株后能够有效提升豆浆的营养。王玲[29]筛选优化了产β-D-葡萄糖苷酶的菌,并将其运用到葡萄酒发酵中,使葡萄酒的香气物质种类和含量相较于未添加的葡萄酒有显著差异,具有开发为酿酒增香酶制剂的潜力。
本研究发现只喷菌M3菌株的香草兰豆荚香兰素含量最高,产生的挥发性成分最多,且只喷M3菌株的豆荚在发酵60 d时产生了3-羟基丁酮。李林海等[30]研究发现,培养条件影响微生物产生3-羟基丁酮,且微生物发酵是生产3-羟基丁酮的主要方法之一。因此,优化微生物培养条件可提升菌株产3-羟基丁酮的能力,增加香草兰发酵豆荚香气。M3与S7按照体积比1∶1喷洒处理的香兰素葡萄糖苷下降最多为80.7%,说明复合菌液可有效转化香兰素葡萄糖苷。傅航等[31]对黄芪渣进行单一菌、2菌剂复合、3菌剂复合发酵,发现3菌剂复合按照1∶1∶1比例的效果最佳。夏欣欣等[32]使用酿酒酵母和芽孢杆菌经混菌固态发酵米糠后,谷维素含量和抗氧化活性均显著提高。焦奕豪等[33]使用酵母菌和枯草芽孢杆菌混合固态发酵沙棘果渣,研究结果发现固态发酵沙棘果渣可改善其营养价值。本研究结果为仅喷M3菌株在香兰素含量、整体挥发性含量等方面效果最佳,M3与S7按照体积比1∶1喷洒处理的香兰素葡萄糖苷下降最多,后续研究可以增加喷菌比例,找出可以提高香草兰品质的最佳组合。
从香草兰豆荚中提取的香兰素由于其纯天然、安全性高、品质优等被广泛应用于调配高端食品和化妆品中,但天然香兰素远远不能满足市场需求[34],采用微生物辅助发酵提升香兰素产量是获得高品质香草兰豆荚,弥补天然香兰素产量难以满足市场需求的重要措施之一。通过测定香兰素含量、香兰素葡萄糖苷含量、挥发成分等方式确定最佳喷菌方式组合,为后续开发微生物辅助豆荚发酵生香,提升品质提供可靠的菌株资源储备和技术参考。未来拟研究不同发酵时期、不同喷洒量等对豆荚香气成分及豆荚表面微生物区系的影响,揭示其提升豆荚品质的微生态机制,研发微生物辅助发酵生香关键技术,为获得高品质香草兰豆荚,开发高附加值产品提供理论与技术支撑。
