The study was aimed to explore the resistance to Colletotrichum siamense, dominant specie of rubber tree induced by Burkholderia arboris DHR18. The enzyme activity changes on the treatments of different concentrations of DHR18 fermentation broth and co-treatments of DHR18 with C. siamense CH-1 using rubber clone GT1 were studied. Five defensive enzymes including catalase (CAT), peroxidase (POD), phenylalanin ammonialyase (PAL), and superoxide dismutase (SOD) were assayed. Results indicated that treatments with 1×108 CFU/mL, 1×107 CFU/mL, 1×106 CFU/mL of DHR18 fermentation broth showed significant higher activities of CAT, POD, PAL, SOD and PPO in rubber leaves compared to the control group. The enzyme activities with broth concentration 1×107 CFU/mL were the highest. The activity peak of the five enzymes was 1 009.14, 29 138.67, 110.24, 902.42 and 148.00 U/g, respectively, which was 2.68, 3.35, 1.88, 3.97 and 4.51 times of the control group, respectively. Under the co-treatments of B. arboris DHR18 and C. siamense CH-1, treatment group inoculated with DHR18 fermentation broth before inoculated with CH-1, the enzyme activities were higher than other co-treatment groups, and significantly higher than that of the control group. Activity peak of CAT, POD, PAL, SOD and PPO was 1 230.85, 45 504.67, 117.53, 1 342.17 and 134.40 U/g, respectively. They were 4.56, 3.10, 2.04, 3.28 and 5.98 times that of the control group, respectively; The plate inhibition rate of strain DHR18 against C. siamense was 71.56%, the preventive effect against C. siamense on rubber saplings was 81.79%, and the control effect was 44.35%. The results showed that Burkholderia arboris DHR18 could promote the activity of defense-related enzymes in rubber trees and induce systemic resistance in rubber trees. The induced resistance may be one of the reasons for the control of C. siamense by B. arboris DHR18.

为探究树木伯克霍尔德氏菌(Burkholderia arboris)DHR18诱导橡胶树对胶孢炭疽菌优势种(Colletotrichum siamense)的抗性,以橡胶树苗GT1为材料,检测不同浓度的树木伯克霍尔德氏菌DHR18发酵液处理以及DHR18与胶孢炭疽菌CH-1协同处理下橡胶树叶片中多酚氧化酶(polyphenol oxidase,PPO)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)、苯丙氨酸解氨酶(phenylalanin ammonialyase,PAL)和超氧化物歧化酶(superoxide dismutase,SOD)5种防御相关酶的活性变化。结果表明:经3个浓度(1×108、1×107、1×106 CFU/mL)的DHR18发酵液处理后,橡胶树叶片中CAT、POD、PAL、SOD和PPO活性均高于对照,其中1×107 CFU/mL浓度处理的5种防御酶活性最高,防御酶活峰值分别为1 009.14、29 138.67、110.24、902.42、148.00 U/g,分别是对照的2.68倍、3.35倍、1.88倍、3.97倍、4.51倍。树木伯克霍尔德氏菌DHR18与胶孢炭疽菌CH-1协同处理下,先喷施DHR18处理组的5种防御酶活性均高于其他处理组,显著高于对照,其CAT、POD、PAL、SOD和PPO活性峰值分别为1 230.85、45 504.67、117.53、1342.17、134.40 U/g,分别是对照的4.56倍、3.10倍、2.04倍、3.28倍、5.98倍;菌株DHR18对胶孢炭疽菌的平板抑制率为71.56%,在橡胶树苗上对胶孢炭疽病预防效果为81.79%,防治效果为44.35%。研究结果表明,树木伯克霍尔德氏菌DHR18可促使橡胶树防御相关酶的活性提高,诱导橡胶树产生系统抗性,诱导抗病性可能是树木伯克霍尔德氏菌DHR18拮抗胶孢炭疽病的原因之一。

为探究树木伯克霍尔德氏菌(Burkholderia arboris)DHR18诱导橡胶树对胶孢炭疽菌优势种(Colletotrichum siamense)的抗性,以橡胶树苗GT1为材料,检测不同浓度的树木伯克霍尔德氏菌DHR18发酵液处理以及DHR18与胶孢炭疽菌CH-1协同处理下橡胶树叶片中多酚氧化酶(polyphenol oxidase,PPO)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)、苯丙氨酸解氨酶(phenylalanin ammonialyase,PAL)和超氧化物歧化酶(superoxide dismutase,SOD)5种防御相关酶的活性变化。结果表明:经3个浓度(1×108、1×107、1×106 CFU/mL)的DHR18发酵液处理后,橡胶树叶片中CAT、POD、PAL、SOD和PPO活性均高于对照,其中1×107 CFU/mL浓度处理的5种防御酶活性最高,防御酶活峰值分别为1 009.14、29 138.67、110.24、902.42、148.00 U/g,分别是对照的2.68倍、3.35倍、1.88倍、3.97倍、4.51倍。树木伯克霍尔德氏菌DHR18与胶孢炭疽菌CH-1协同处理下,先喷施DHR18处理组的5种防御酶活性均高于其他处理组,显著高于对照,其CAT、POD、PAL、SOD和PPO活性峰值分别为1 230.85、45 504.67、117.53、1342.17、134.40 U/g,分别是对照的4.56倍、3.10倍、2.04倍、3.28倍、5.98倍;菌株DHR18对胶孢炭疽菌的平板抑制率为71.56%,在橡胶树苗上对胶孢炭疽病预防效果为81.79%,防治效果为44.35%。研究结果表明,树木伯克霍尔德氏菌DHR18可促使橡胶树防御相关酶的活性提高,诱导橡胶树产生系统抗性,诱导抗病性可能是树木伯克霍尔德氏菌DHR18拮抗胶孢炭疽病的原因之一。

The study was aimed to explore the resistance to Colletotrichum siamense, dominant specie of rubber tree induced by Burkholderia arboris DHR18. The enzyme activity changes on the treatments of different concentrations of DHR18 fermentation broth and co-treatments of DHR18 with C. siamense CH-1 using rubber clone GT1 were studied. Five defensive enzymes including catalase (CAT), peroxidase (POD), phenylalanin ammonialyase (PAL), and superoxide dismutase (SOD) were assayed. Results indicated that treatments with 1×108 CFU/mL, 1×107 CFU/mL, 1×106 CFU/mL of DHR18 fermentation broth showed significant higher activities of CAT, POD, PAL, SOD and PPO in rubber leaves compared to the control group. The enzyme activities with broth concentration 1×107 CFU/mL were the highest. The activity peak of the five enzymes was 1 009.14, 29 138.67, 110.24, 902.42 and 148.00 U/g, respectively, which was 2.68, 3.35, 1.88, 3.97 and 4.51 times of the control group, respectively. Under the co-treatments of B. arboris DHR18 and C. siamense CH-1, treatment group inoculated with DHR18 fermentation broth before inoculated with CH-1, the enzyme activities were higher than other co-treatment groups, and significantly higher than that of the control group. Activity peak of CAT, POD, PAL, SOD and PPO was 1 230.85, 45 504.67, 117.53, 1 342.17 and 134.40 U/g, respectively. They were 4.56, 3.10, 2.04, 3.28 and 5.98 times that of the control group, respectively; The plate inhibition rate of strain DHR18 against C. siamense was 71.56%, the preventive effect against C. siamense on rubber saplings was 81.79%, and the control effect was 44.35%. The results showed that Burkholderia arboris DHR18 could promote the activity of defense-related enzymes in rubber trees and induce systemic resistance in rubber trees. The induced resistance may be one of the reasons for the control of C. siamense by B. arboris DHR18.

天然橡胶是一个国家至关重要的战略资源以及不可替代的工业原料,而全世界98%的天然橡胶是由橡胶树(Hevea brasiliensis Muell. Arg.)生产[1]。橡胶树种植受到独特的地域和气候限制,其中影响橡胶产量的重要因素之一是生物胁迫[2]。炭疽病是橡胶树生产中的重要病害,胶孢炭疽菌复合群中的Colletotrichum siamense是我国植胶区的田间优势致病种,通过侵染橡胶树的枝条、花、叶等组织,造成叶片枯萎和落叶,最终导致严重减产[3]。目前,主要防治方法是使用化学药剂,由于橡胶树属高大乔木,在防治过程中使用药剂比较单一、药量大、易发生漂移,且化学药剂在防治时存在抗药性、防治成本高和环境污染等问题[4-5]。因此,急需一种绿色环保、经济安全的手段来防治橡胶树炭疽病。
生物防治是通过利用有益微生物或其代谢物来防治植物病害,已经成为病害防控中一种可持续和有效的方法[6-7]。生物防治剂不仅可以直接抑制植物病原体的生长来减少对植物的侵染,还能诱导植物的系统抗性,引发植物广泛的防御反应,对多种病原体有着长期的防治效果[8]。众多研究表明,生防菌可以通过诱导植物相关防御酶的升高,从而增强植物的抗性[9]。内生耐盐芽孢杆菌Cal.l.30和Cal.f.4可以诱导番茄对灰葡萄孢的系统抗性,从而有效防治番茄灰霉病[10]。罗氏假单胞菌GC-7处理的植物根系中过氧化氢酶(catalase,CAT)、多酚氧化酶(polyphenol oxidase,PPO)、苯丙氨酸解氨酶(phenylalanin ammonialyase,PAL)等防御相关酶的活性显著增加,从而增强植物抗性[11]
伯克霍尔德氏菌(Burkholderia sp.)是一种能有效防治多种植物病害的生防菌,与植物有着密切的共生关系,而且伯克霍尔德氏菌产生的一些次级代谢产物,对大部分植物病原真菌有明显抑制作用。此外,可以通过诱导植物防御相关酶活性的升高来增强对植物病原菌的抗性[12]。已有研究发现,越南伯克霍尔德氏菌C12通过诱导水稻防御相关酶活性(CAT、PAL、SOD、PPO和POD)的升高,从而增强水稻的抗病性[13];吡咯伯克霍尔德氏菌S17-377可诱导水稻防御相关酶活性变化,有3种防御相关酶的活性增强,说明伯克霍尔德氏菌可以增强水稻对水稻纹枯病的抗性[14]。本实验室前期分离得到1株对橡胶树胶孢炭疽菌有较强拮抗作用的树木伯克霍尔德氏菌(B. arboris),本研究系统测定了不同浓度树木伯克霍尔德氏菌DHR18发酵液以及DHR18与胶孢炭疽菌协同处理下橡胶树叶片中CAT、POD、PAL、SOD和PPO活性变化,初步探索树木伯克霍尔德氏菌诱导橡胶树对胶孢炭疽菌的抗性机理,以期为橡胶树炭疽病的绿色防控提供科学依据。
树木伯克霍尔德氏菌株DHR18及橡胶树胶孢炭疽菌CH-1均由中国热带农业科学院橡胶研究所种质资源实验室分离、鉴定及保存,其中树木伯克霍尔德氏菌DHR18分离自中国热带农业科学院试验场(海南儋州)橡胶树根病区;橡胶树胶孢炭疽菌CH-1分离自国家橡胶种质资源圃(儋州)。
供试橡胶种苗品种为GT1,挑选叶部生长健康、苗长35 cm,胸径和株高等长势均一的具1篷叶淡绿期叶片的种子苗,设置DHR18发酵液喷施3个浓度处理,以及病原菌与生防菌协同喷施3个处理,每个处理使用种子苗20株,每个处理重复5次,所有种子苗均由国家橡胶种质资源圃提供。
挑取DHR18单菌落于100 mL LB培养基中,在恒温摇床(180 r/min,28 ℃)中培养24 h,将DHR18发酵液浓度调节至1×108 CFU/mL作为原液,用无菌水稀释得到1×107 CFU/mL和1×106 CFU/mL浓度的稀释液。胶孢炭疽菌CH-1接种在PDB培养基中,在恒温摇床(180 r/min,28 ℃)中培养3 d待用。炭疽菌接种:参考蔡志英等[15]的方法,用孢子悬浮液(1×106个/mL)接种,预先用无菌昆虫针刺伤橡胶叶片,孢子悬浮液用喷雾器均匀喷施,直到橡胶叶片挂满水滴为止,每个处理重复3次;空白对照喷施适量无菌水,套袋保湿。
参考唐文等[16]的方法,共设置3个浓度的DHR18发酵液诱导处理(表1),包括1×108、1×107、1×106 CFU/mL处理,以LB培养基为对照,取10 mL不同浓度的DHR18发酵液和对照喷洒植株叶面,每个处理重复5次。分别于处理后的24、48、72、96、120 h,取位置相同的橡胶树叶片,用于防御酶的活性测定。
共设置4个处理,处理方式如表2所示。样品采集和处理与1.2.2相同。
将1.2.2和1.2.3中采集的橡胶树叶片,使用酶活试剂盒(索莱宝科技有限公司)测定POD、SOD、CAT、PAL和PPO活性。按照试剂盒说明书中步骤加入试剂,在相应的波长下测定其吸光值,并按照试剂盒的公式计算其酶活力。
实验数据采用Excel 2016软件进行统计、整理,采用IBM SPSS Statistics 25软件进行统计分析。
经不同浓度DHR18发酵液处理后,橡胶树叶片中CAT活性均高于CK。其中,T2、T3处理的CAT活性呈先升高后降低再升高的趋势,且在48 h时达到峰值,其活性分别为1009.14 U/g和858.79 U/g,为CK的2.68倍和2.29倍,均极显著高于CK(P<0.01);而T1的CAT活性在48 h后下降并趋于稳定,但在同时间的酶活性均高于CK(图1)。
图2所示,经不同浓度DHR18发酵液喷施叶片处理后,处理组叶片POD活性均呈先升高后降低的趋势,并且活性在相同时间均高于CK。在72 h时,每个处理组叶片POD活性达到峰值,此时T2和T3处理的POD酶活性达到29 138.67、27 276.67 U/g,分别是CK的3.35倍和3.13倍,均极显著高于CK(P<0.01)。
图3所示,T2处理的PAL活性呈先升高后降低的趋势,在96 h时,其PAL活性达到峰值110.24 U/g,是CK的1.88倍,显著高于CK(P<0.05)。T1处理的PAL活性呈先升高后降低再升高的趋势。在48 h和96 h时出现峰值,叶片PAL活性分别为92.72 U/g和93.43 U/g,均高于CK。T2处理效果最明显,在相同时间,其处理的叶片PAL活性均高于其他浓度。
图4所示,经不同浓度DHR18发酵液喷施叶片处理后,处理组叶片SOD活性呈先升高后降低的趋势,且其活性在相同时间均高于CK。T2处理的SOD活性在48 h达到峰值1164.89 U/g,是CK的3.83倍,极显著高于CK(P<0.01)。T1和T3处理的SOD活性在72 h达到峰值,分别为774.93 U/g和848.18 U/g,是CK的3.41倍和3.73倍,显著(P<0.05)和极显著高于CK(P<0.01)。
图5可知,T1、T2和T3处理的PPO活性呈先升高后降低的趋势,其活性在相同时间均高于CK。T2、T3处理的PPO活性在72 h时分别达到峰值148.00 U/g和142.80 U/g,是CK的4.51倍和4.35倍。T1处理的PPO活性在96 h时达到峰值(111.60 U/g)。经不同浓度DHR18发酵液处理的叶片PPO活性在峰值时均极显著高于CK(P<0.01)。
图6所示,菌株DHR18在平板上对橡胶树胶孢炭疽菌CH-1表现出明显的抑制效果,抑制率为71.56%。按照表2,使用DHR18与CH-1协同处理橡胶树幼苗,72 h后,CK的叶片未发病,T5处理(先接生防菌)的防治效果为81.79%,T4处理(先接病原菌)的防治效果为44.35%,均显著高于T6处理(只接病原菌)(P<0.0001)。
生防菌DHR18和CH-1共同处理后橡胶树的相关防御酶CAT活性有显著变化,3种不同处理组CAT活性在同时间均高于CK。其中,T5处理的CAT活性呈先升高后降低的趋势,在48 h时达到峰值(1230.85 U/g),极显著高于其他处理(P<0.01),是CK的4.56倍;T4处理的CAT活性呈先降低后升高再降低的趋势,在48 h极显著高于CK(P<0.01)(图7)。
DHR18与CH-1协同处理下,3种处理组的POD活性均高于CK,在同时间均达显著水平(P<0.05),其中,T5的POD活性在48 h时达到峰值(45 504.67 U/g),是CK的3.10倍;T4和T6处理的POD活性在72 h时达到峰值(30 249.33、33 875.33 U/g),分别为CK的2.14倍和2.40倍。3种不同处理在POD活性峰值时均极显著高于CK(P<0.01)(图8)。
图9可知,T5处理的PAL活性总体呈先升高后降低的趋势,在同时间均高于CK,72 h时与CK差异显著(P<0.05),在48 h时达到峰值(117.53 U/g),是CK的2.04倍,极显著高于CK(P<0.01);T4和T6处理的PAL活性变化不明显,活性保持在85.00 U/g左右,在72 h显著高于CK(P<0.05)。
图10可知,T5处理的SOD活性在72 h极显著高于CK(P<0.01),SOD活性达1342.17 U/g,是CK的3.30倍;T4的SOD活性呈先升高后降低的趋势,在48 h达到峰值,活性为924.49 U/g,是CK的2.47倍,显著高于CK(P<0.05);T6的SOD活性在24 h时低于CK,与CK无显著差异。
图11可知,T5的PPO活性呈先升高后降低的趋势,在同时间均高于CK,且在48 h时达到峰值,为134.40 U/g,是CK的5.6倍;T4的PPO活性在48 h时达到峰值,为134.40 U/g,是CK的5.51倍,均极显著高于对照CK(P<0.01);T6的PPO活性变化不明显,未出现明显的峰值,活性略高于CK。
过氧化氢酶(CAT)[17]、过氧化物酶(POD)和超氧化物歧化酶(SOD)是植物主要的活性氧清除酶,可消除因病害侵染或逆境胁迫产生过多的活性氧,从而避免对自身造成损伤[18];苯丙氨酸解氨酶(PAL)可催化苯丙氨酸合成酚类化合物,而酚类化合物是植物多种次生代谢产物的中间产物,对植物抗病虫害有重要作用[19];多酚氧化酶(PPO)是一类金属蛋白酶,可以催化多种生化反应,调节生理功能,与植物抗逆抗病等息息相关[20]。这些防御相关酶活性对于植物的抗病性具有重要决定作用,也是评价植物抗性水平的重要参考标准[21-22]
研究表明,生防菌通过诱导和增强一系列防御相关酶活性,从而提高植物的系统抗病能力,是其重要的抗病机制之一[23-24]。CHEN等[25]研究发现植物乳杆菌CM-3可以通过增加葡萄POD、PPO、CAT和PAL的活性来增强葡萄的抗病能力。邹强等[26]用生防贝莱斯芽孢杆菌TP-1处理后葡萄的APX、PPO和PAL活性提高,增强了对灰霉菌的抗性。本研究中,生防菌DHR18显著提高橡胶树中CAT、POD、SOD、PAL和PPO五种防御酶活性,增强了橡胶树的抗性,其中多酚氧化酶的变化最为明显,最高可以达到对照的4.51倍。陈秀香等[27]研究发现,经枯草芽孢杆菌FC21的10倍稀释液处理后的植株叶片中CAT、POD、SOD活性最高,生防菌诱导的防御相关酶活性与菌液浓度不呈正相关。本研究发现107 CFU/mL浓度的DHR18发酵液对橡胶树防御诱导效果最好,可作为伯克霍尔德氏菌DHR18防治炭疽病的浓度参考。因此,本研究结果表明,树木伯克霍尔德氏菌DHR18能诱导橡胶树体内CAT、POD和SOD酶活增强来清除过量活性氧;能诱导PAL和PPO酶活性促使橡胶树产生抗菌物质等,从而提高橡胶树系统抗性。
植物在受到病原物侵染后,活性氧作为植物抵御和延缓病原物侵染的第一道物理屏障,在第一时间反应并影响活性氧消除酶的合成酶基因的表达,从而增加CAT、POD和SOD等的活性继而实现解毒作用,更好地应对各种胁迫,其体内防御酶活性也会上升[28]。感染病原物后,活性氧消除酶普遍呈先上升后下降的变化趋势。耿莉娜等[29]的研究发现烟草的抗病品种Beinhart1000-1在接种赤星病菌后,POD、PAL、SOD活性明显高于对照,呈先升高后降低的变化趋势。本研究中,在接种炭疽病CH-1后,橡胶树的SOD和PAL活性无明显变化,可能与植株品种抗病性有关。罗富方等[30]研究发现在青枯病胁迫下,烤烟再接种印度梨形孢,其4种防御酶(PAL、PPO、POD和SOD)活性增加均高于对照,且呈先升高后降低的趋势。本研究中DHR18和CH-1协同处理下,处理组的防御酶(CAT、POD、SOD、PAL和PPO)活性均有提高,主要呈先上升后下降的趋势;协同处理的CAT、SOD和PPO酶活性在24、48、72 h显著高于其只接病原菌的酶活,表明伯克霍尔德氏菌DHR18在橡胶树受到炭疽病菌侵染过程中会增强其活性氧清除系统,并促进合成抗病物质,从而增强植株的抗病能力。唐文等[16]先喷施枯草芽孢杆菌Czk1后接种病原菌处理的5种防御相关酶(CAT、POD、PAL、SOD和PPO)活性升高,显著高于其他处理。本研究橡胶树受到炭疽病侵染前喷施DHR18发酵液,对防御酶活性影响最大,5种防御酶活性高于其他处理组,更有利于诱导抗病性。本研究发现,先喷施生防菌伯克霍尔德氏菌DHR18再接种病原菌处理组的POD活性比只接病原菌和先接病原菌处理组的更快达到峰值,在48 h时显著高于其他处理组,能更快引起植物的防御反应;周东兴等[31]研究发现贝莱斯芽孢杆菌DX-25与番茄枯萎病互作的情况下可以更快地引起植株番茄的抗性反应,与本研究结果相似。
在本研究中,生防菌DHR18发酵液可促使叶片中CAT、POD、PAL、SOD和PPO酶活性上升,诱导橡胶树产生系统抗性,1×107 CFU/mL浓度对橡胶树的诱导效果最好。研究结果初步明确了树木伯克霍尔德氏菌具有诱导抗病性,为探索伯克霍尔德氏菌诱导抗病性提供了部分理论依据。
