The 16S rRNA high-throughput sequencing analysis was conducted on the rhizosphere soil of Cassava South China No.12 under treatment without fertilization (CK), organic fertilizer (T1), conventional fertilization applied with organic fertilizer (T2), and reduced chemical fertilizer applied with organic fertilizer (T3) by using the Illumina Nova 6000 sequencing platform to study the effects of chemical fertilizer application combined with organic fertilizer on the growth of cassava and the bacterial diversity and community structure in cassava rhizospheric soil. T3 not only significantly increased the height and stem thickness of cassava, but also improved the yield of cassava, compared to CK. Cluster analysis was performed on the sequences based on 97% similarity, the OTUs numbers of CK, T1, T2 and T3 are 3603, 1688, 1276 and 3317 respectively. Different fertilization methods altered the diversity and richness of soil bacteria, with the species diversity ranked from highest to lowest as CK>T3>T1>T2. Based on PCoA and cluster analysis, the community composition of CK and T3 was similar, the community composition of T1 and T2 was similar. The dominant phylum in cassava rhizospheric soil under different fertilization treatments were Proteobacteria, Bacteroidetes, Acidobacteria, Firmicutes, Actinobacteria and Chloroflexi. The relative abundance of Proteobacteria, Bacteroidetes and Actinobacteria increased under T3 treatment, while the relative abundance of Firmicutes and Chloroflexi decreased. Redundancy analysis results indicated that available potassium was the main factor affecting the bacterial community in the cassava rhizosphere soil. In summary, the study demonstrates that reduced chemical fertilizer application combined with organic fertilizer not only increases the yield of cassava, but also changes the community structure and diversity of the bacterial in the rhizosphere soil. The results would provide a solid theoretical foundation for the development of a green, efficient, and sustainable cassava industry.

为了研究化肥减量配施有机肥对木薯根际土壤细菌多样性和群落结构及产量的影响,采用Illumina Nova 6000测序平台,对未施肥(CK)、有机肥(T1)、常规施肥+有机肥(T2)和化肥减施+有机肥(T3)等施肥方式处理下的华南12号木薯根际土壤进行16S rRNA高通量测序分析。结果表明,与CK相比,T3处理不仅能显著增加木薯株高和茎粗,还能提高木薯产量。在相似水平为97%下聚类分析得到木薯根际土壤OTUs,分别为3603个(CK)、1688个(T1)、1276个(T2)和3317个(T3)。不同施肥方式改变土壤细菌的多样性和丰度,其物种多样性由高到低依次为:CK>T3>T1>T2。基于土壤细菌群落的PCoA和聚类分析,CK和T3细菌群落结构组成较为相似,T1与T2细菌群落结构组成相似。不同施肥处理下木薯根际土壤中优势菌门为变形菌门、拟杆菌门、酸杆菌门、厚壁菌门、放线菌门和绿弯菌门。化肥减施配施有机肥增加了变形菌门、拟杆菌门和放线菌门的相对丰度,降低了厚壁菌门和绿弯菌门的相对丰度。冗余性分析结果表明速效钾是影响木薯根际土壤菌群的主要效应因子。研究表明,化肥减量配施有机肥不仅能增加木薯的产量,还能改变木薯根际土壤细菌群落结构和多样性,为发展绿色、高效、可持续的木薯产业提供坚实的理论支撑。

为了研究化肥减量配施有机肥对木薯根际土壤细菌多样性和群落结构及产量的影响,采用Illumina Nova 6000测序平台,对未施肥(CK)、有机肥(T1)、常规施肥+有机肥(T2)和化肥减施+有机肥(T3)等施肥方式处理下的华南12号木薯根际土壤进行16S rRNA高通量测序分析。结果表明,与CK相比,T3处理不仅能显著增加木薯株高和茎粗,还能提高木薯产量。在相似水平为97%下聚类分析得到木薯根际土壤OTUs,分别为3603个(CK)、1688个(T1)、1276个(T2)和3317个(T3)。不同施肥方式改变土壤细菌的多样性和丰度,其物种多样性由高到低依次为:CK>T3>T1>T2。基于土壤细菌群落的PCoA和聚类分析,CK和T3细菌群落结构组成较为相似,T1与T2细菌群落结构组成相似。不同施肥处理下木薯根际土壤中优势菌门为变形菌门、拟杆菌门、酸杆菌门、厚壁菌门、放线菌门和绿弯菌门。化肥减施配施有机肥增加了变形菌门、拟杆菌门和放线菌门的相对丰度,降低了厚壁菌门和绿弯菌门的相对丰度。冗余性分析结果表明速效钾是影响木薯根际土壤菌群的主要效应因子。研究表明,化肥减量配施有机肥不仅能增加木薯的产量,还能改变木薯根际土壤细菌群落结构和多样性,为发展绿色、高效、可持续的木薯产业提供坚实的理论支撑。

The 16S rRNA high-throughput sequencing analysis was conducted on the rhizosphere soil of Cassava South China No.12 under treatment without fertilization (CK), organic fertilizer (T1), conventional fertilization applied with organic fertilizer (T2), and reduced chemical fertilizer applied with organic fertilizer (T3) by using the Illumina Nova 6000 sequencing platform to study the effects of chemical fertilizer application combined with organic fertilizer on the growth of cassava and the bacterial diversity and community structure in cassava rhizospheric soil. T3 not only significantly increased the height and stem thickness of cassava, but also improved the yield of cassava, compared to CK. Cluster analysis was performed on the sequences based on 97% similarity, the OTUs numbers of CK, T1, T2 and T3 are 3603, 1688, 1276 and 3317 respectively. Different fertilization methods altered the diversity and richness of soil bacteria, with the species diversity ranked from highest to lowest as CK>T3>T1>T2. Based on PCoA and cluster analysis, the community composition of CK and T3 was similar, the community composition of T1 and T2 was similar. The dominant phylum in cassava rhizospheric soil under different fertilization treatments were Proteobacteria, Bacteroidetes, Acidobacteria, Firmicutes, Actinobacteria and Chloroflexi. The relative abundance of Proteobacteria, Bacteroidetes and Actinobacteria increased under T3 treatment, while the relative abundance of Firmicutes and Chloroflexi decreased. Redundancy analysis results indicated that available potassium was the main factor affecting the bacterial community in the cassava rhizosphere soil. In summary, the study demonstrates that reduced chemical fertilizer application combined with organic fertilizer not only increases the yield of cassava, but also changes the community structure and diversity of the bacterial in the rhizosphere soil. The results would provide a solid theoretical foundation for the development of a green, efficient, and sustainable cassava industry.

木薯(Manihot esculenta Crantz)块根淀粉含量较高,是世界第六大粮食作物,是全球近10亿人的主要食粮,也是我国工业乙醇、淀粉、变性淀粉及饲料的重要原料[1]。近年来,我国木薯原料大部分依赖进口,是第一大木薯原料进口国和消费国。造成这样的缺口主要是缺乏与高产优质木薯新品种配套的栽培技术,肥料施用不科学,连作时间长,且收获经济效益低,限制了木薯的推广与利用。在农作物生产中,为了获取更高的产量,存在重用化肥,导致土壤板结、肥力下降、土壤微生物的多样性及酶活性降低等诸多负面影响[2-4]。因此,调整与优化木薯施肥方式迫在眉睫,而减少化肥施用量,增加有机肥或添加土壤修复剂将是今后种植木薯的必然趋势。
化肥减施配施生物有机肥模式已广泛地应用在各种农作物的种植端,配施有机肥或生物有机肥可以改善根际土壤微生物群落的多样性和群落结构,提高农作物产量和提升农产品品质,促进农作物生长,改善农业生态环境[5-7]。关于化肥减施配施有机肥研究主要集中在探讨对土壤肥力[8-10]、土壤酶活性[11-12]、作物生长发育及产量[13-15]、土壤微生物多样性及菌群[16-18]的影响等方面。在木薯中关于化肥减量配施有机肥对其生长发育、酶活性、根际土壤微生物多样性及群落结构的研究还未见报道。目前在木薯研究中仅见化肥减施虽能改变木薯根际微生物多样性和群落结构,但会导致木薯根际微生物的多样性和丰度都下降[19-20]的相关报道;而在木薯中化肥减量配施有机肥的根际土壤细菌多样性及群落结构的变化尚未见相关报道。
本研究以木薯品种华南12号(SC12)为研究对象,采用高通量测序平台Illumina Nova 6000对木薯根际土壤进行16S rRNA高通量测序分析,分析未施肥、施有机肥、常规施肥配施有机肥和化肥减施配施有机肥的不同模式对木薯根际土壤细菌多样性和群落结构的影响,结合土壤理化性质,分析木薯生长及产量情况,阐明土壤细菌与环境因子的相互作用对木薯生长发育的影响,为木薯生产科学施肥、改善土壤生态环境、促进木薯提质增效提供理论依据。
试验于2022年在海南省儋州市红星队国家木薯种质资源圃进行,试验地地理位置位于109o30'E,19o30'N。属热带季风气候,年降雨量为900~2000 mm。试验前基地土壤养分含量如下:有机质为10.99 g/kg,速效钾为26.42 mg/kg,有效磷为3.47 mg/kg,碱解氮为21.99 mg/kg。试验所用氮肥为尿素(N 46.2%),磷肥为钙镁磷肥(P2O5 18%),钾肥为硫酸钾(K2O 52%),商品活性有机肥(N+P2O5+K2O≥5%,有机质≥80%,总腐殖酸≥30 g/kg,腐殖质≥300 g/kg,元泰丰(包头)生物科技有限公司生产。供试木薯品种为华南12号。
试验共设4个处理:(1)CK(未施肥);(2)T1(有机肥);(3)T2(常规施肥+有机肥);(4)T3(化肥减施+有机肥)。化肥减施为在常规施肥基础上减氮25%+减磷50%,常规施肥用量:N 90 kg/hm2、P2O5 90 kg/hm2、K2O 90 kg/hm2;化肥减施用量:N 67.5 kg/hm2、P2O5 45 kg/hm2、K2O 90 kg/hm2,有机肥用量均为1500 kg/hm2。木薯种植后2个月离种茎基部15~30 cm处穴施1次肥料。每个试验区面积36 m2,3次重复,共12个试验区。每个小区种植6行,每行6株,36株/小区,株行距1.0 m×1.0 m。小区间隔50 cm,重复间隔100 cm。木薯于2022年5月种植,2023年3月收获。其他管理同一般常规栽培。
(1)样品收集及处理。木薯根际土壤样品采集于2023年3月在海南省儋州市国家木薯种质资源圃进行,具体参照CAI等[20]的方法。采样时使用的工具、采集袋或其他物品均已灭菌。设置10个采样点,按照“S”型取样,用无菌毛刷将附着在木薯块根表面的土壤刷于报纸上,用1 mm土壤筛去杂质,将所有样品混匀作为该试验小区的木薯根际土样。采集的木薯土壤样品做好标记后,一份用于土壤有机质(OM)、碱解氮(AN)、速效钾(AK)、有效磷(AP)等指标测定;另一份置于干冰中用于细菌16S rRNA高通量测序分析。
(2)农艺性状和产量测定。2023年3月收获木薯,分别用卷尺和游标卡尺测定各个试验小区的木薯株高和茎粗;各小区分别选取木薯10株,统计并记录各小区木薯的薯条数和鲜薯重,以实收的10株木薯鲜薯重换算成每公顷鲜薯产量。
(3)土壤pH和养分测定。测定土壤pH、OM、AN、AK和AP含量,具体参照鲍士旦[21]的方法。
将采集的样品送至广东美格基因科技有限公司进行16S rRNA高通量测序。原始测序数据(raw reads)通过质控得到最后的有效数据(clean tags)。利用美格云平台(http://cloud.magigene.com/)进行细菌Alpha多样性和Beta多样性分析。采用Microsoft Excel 2003处理数据和绘制表格。采用Kruskal-Wallis test软件进行差异显著性分析。
测定各处理下收获期木薯的植物学性状(株高、薯条数、茎粗等)和产量。结果表明,施肥处理后木薯在株高、茎粗、薯条数和鲜薯产量均显著高于CK处理的木薯(表1)。以上结果表明,不同施肥处理在一定程度上促进了木薯的生长,并提高其产量,尤其是在化肥减量(减25% N和减50% P)的基础上配施有机肥不但不会对木薯生长造成不良影响,还能显著增加木薯的产量。
利用高通量测序技术分析12个不同施肥处理的木薯根际土壤样品,统计每个处理(CK、T1、T2、T3)样品通过测序得到的相关指标如表2所示。每个样品测序的覆盖度均大于0.992,结果表明构建的测序文库涵盖了木薯土壤样品中99%以上的细菌菌群。同时,随着测序数据量的增加,样品的稀释曲线也是升高后慢慢趋于平缓(图1),这说明高通量测序的深度和获得的数据量足以覆盖样品中细菌菌群的多样性,可以用于后续的研究。
对各处理木薯根际土壤的OTU和α-多样性指数进行分析(表3)。与CK处理相比,T1、T2、T3处理土壤中OTUs、细菌类群的丰度(Chao1和ACE)均呈现下降的趋势,但T3是施肥处理组中细菌丰度最高的。与CK处理相比,不同处理根际土壤的Simpson指数随着有机肥或化肥的施用呈现出升高的趋势,而Shannon指数随着有机肥或化肥的施用均呈现出下降的趋势。研究结果表明,施有机肥或化肥会在一定程度上改变根际土壤中微生物的丰度和多样性,化肥减施配施有机肥虽然在细菌丰度和多样性上均有所降低,但其丰度和多样性最接近未施肥的土壤。
木薯根际土壤细菌共同的OTUs有885个(图2)。与CK处理相比,不同施肥方式下的每个样本特有OTUs的数量少,分别是1600个(T1)、413个(T2)、1371个(T3)。与CK处理相比,在各施肥处理中特有的OTUs的数量均呈现减少的趋势,与总OTUs数量减少表现一致。结果说明,各施肥处理改变了木薯根际土壤细菌类群的丰度。
对各施肥处理下木薯根际土壤细菌群落进行主坐标分析(PCoA),结果表明主成分1(PCoA1)和主成分2(PCoA2)的贡献率分别为62.10%和25.00%。如图3所示,样本CK和T3距离较近,聚集在一起,说明这2个样本细菌类群组成结构相似;样本T1和T2距离较近,说明这2个样本的细菌类群组成结构相似。
为了进一步了解PCoA的结果,对不同样本进行聚类分析,结果如图4所示,细菌群落结构组成分成两大类,即T1和T2被聚为一个分支,T3和CK被聚为一个分支,说明CK和T3在细菌群落组成结构上有较高相似度。结果表明化肥减施配施有机肥的细菌群落结构和多样性组成介于常规施肥配施有机肥和未施肥之间,不同施肥处理能够显著改变细菌的群落结构和多样性。
细菌16S rRNA高通量测序获得序列经注释后,统计每个处理样本中细菌相对丰度及群落结构组成。在门水平下,细菌菌落相对丰度如图5所示,变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)、拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、绿弯菌门(Chloroflexi)是木薯根际土壤样本的优势细菌菌门。与CK的木薯根际土壤细菌群落相比,变形菌门(Proteobacteria)和拟杆菌门(Bacteroidetes)在施肥组处理中的含量呈现升高的趋势。酸杆菌门(Acidobacteria)在T1和T2施肥处理后的含量明显下降,但在T3中变化不大;厚壁菌门(Firmicutes)在T1处理后的含量有所增加,但在T2和T3处理中呈现下降的趋势;绿弯菌门(Chloroflexi)在T1、T2、T3处理下的含量呈现下降趋势。放线菌门(Actinobacteria)经各施肥处理后(T1、T2、T3)的含量均明显增加(P<0.05)。T1、T2、T3处理的木薯根际土壤中放细菌门分别增加6.3倍、3.5倍、2.4倍。结果表明,有机肥或化肥施用并未改变主要细菌的类群,但提高了木薯根际土壤中有益细菌类群的相对丰度。
在属水平上,相对丰度大于0.5%的共有14个属。黄杆菌属(Flavobacterium)、不动杆菌属(Acinetobacter)和马赛菌属(Massilia)是木薯根际土壤细菌群落的主要菌属(相对丰度大于10%)(图6)。与CK处理相比,黄杆菌属、不动杆菌属和马赛菌属的含量在T1、T2和T3处理中均呈现显著增加。在各施肥处理中,主要菌属的含量也有显著差异。黄杆菌属在T1、T2处理下的含量相似,但与T3处理下的含量有明显差异,分别是T3处理下的4.3倍和4.9倍。类似的结果也在马赛菌属中观察到,T1、T2处理下的含量是T3处理的19.4倍和5.4倍。然而不动杆菌属在T2和T3处理下的含量显著高于T1处理,分别是T1处理下含量的3.9倍和2.62倍。
利用RDA分析土壤环境因子如何影响木薯根际土壤细菌群落结构。选择pH、SOM、AN、AP和AK等5个环境因子进行冗余性分析,结果如图7所示,在门水平上,RDA1解释了72.40%,RAD2解释了18.50%,共解释了90.90%以上。速效钾、pH、速效磷对土壤细菌落结构有着重要影响,其中速效钾影响最大,且速效钾含量与速效磷、碱解N、有机质、pH均呈正相关。
土壤根际微生物组成及丰度受土壤质量[22-23]、宿主植物[20,24-25]和种植模式[26-27]等因素影响。本研究利用高通量测序分析化肥减量配施有机肥如何影响和改变木薯根际土壤细菌多样性及菌群结构。结果表明化肥减量配施有机肥的木薯根际土壤中,在门水平上,变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、酸杆菌门(Acidobacteria)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、绿弯菌门(Chloroflexi)是木薯根际土壤样本的优势细菌菌门,与前人报道的在间作或轮作下木薯根际土壤的优势菌门一致[9,25,27-28]。因此,推测在南方酸性土壤中宿主植物木薯通过分泌根系代谢物富集特定的微生物,导致南方木薯根际土壤下所富集细菌类群结构组成相似,只是在多样性和丰度上有所不同,这说明木薯根际土壤细菌的类群主要受宿主植物影响和调配,而种植模式(间作或轮作)或施肥方式只能在一定程度上改变其多样性和丰度。
本研究中,化肥减量配施有机肥的木薯根际土壤中,细菌菌群的多样性及丰度均显著高于施有机肥、常规施肥配施有机肥处理,这结果与靳晓拓等[8]、陆海飞[29]、孙家骏等[30]的报道一致。蔡杰等[19]研究发现化肥减施的木薯根际土壤中,细菌多样性和丰度均显著低于未施肥和常规施肥处理,导致这种结果是由于未添加有机肥处理。本研究中,单施有机肥、常规施肥配施有机肥和化肥减施配施有机肥都能提高有益细菌放线菌门的丰度,与王庆等[6]、姜蓉等[7]、蔡杰等[19]的研究结果一致。研究表明,土壤中放线菌数量的增加可以提高对土壤有机质利用率,同时还能抑制一些土传病害,促进土壤健康发展[31]。同时,本研究在属水平上也发现拟杆菌门的黄杆菌属在T1、T2和T3处理下的丰度显著增加。研究表明,黄杆菌属参与土壤中有机物的分解,对维持土壤肥力有积极作用[32]。然而,黄杆菌属的丰度在T3处理中呈现最低,这可能跟减少氮和磷肥施用有关。相关研究表明,黄杆菌属能够通过其代谢活动参与氮、磷循环[33-34]Candidatus Solibacter是分解有机质、利用碳源的细菌属[35]。本研究中,在T1和T2处理下Candidatus Solibacter的丰度显著低于T3处理。综上,合理配施有机肥不仅能为土壤提供大量的有机质及养分,还能在一定程度上还增加土壤有益细菌的数量,改善土壤细菌菌群结构[36],帮助其吸收转化养分,为植物生长创造优良的环境,减轻作物的连作障碍[37]
合理施肥或配施有机肥能够提高农作物产量,提升农产品品质,促进作物生长,改善农业生态环境。本研究中,不同施肥处理均能显著促进木薯生长发育,增加木薯产量。与未施肥处理相比,化肥减施配施有机肥能够显著增加木薯株高和茎粗,获得较高的木薯产量。这可能与配施有机肥增加了土壤中有益微生物多样性有关,从而改变土壤细菌的菌落结构,促进土壤向健康发展。但化肥减量配施不同有机肥对木薯产量及品质的连续影响及其机制有待进一步深入研究。随着化肥减量施肥的倡导,在其基础上合理配施有机肥,在木薯主产区生产中,逐步实现有机肥替代化肥,实现化肥零增长,发展绿色、可持续的木薯产业,促进木薯产业提质增效,具有非常重要的力量意义和应用价值。
与未施肥相比,化肥减量配施有机肥处理不仅能显著增加木薯株高和茎粗,还能提高木薯产量。与单施有机肥和常规施肥配施有机肥的木薯根际土壤相比,化肥减量配施有机肥的木薯根际土壤细菌的多样性和丰度有显著差异。化肥减量配施有机肥处理木薯根际土壤中优势菌门为变形菌门、酸杆菌门、拟杆菌门、厚壁菌门、放线菌门和绿弯菌门,这与已报道的木薯根际土壤细菌优势菌门一致。化肥减施配施有机肥增加了拟杆菌门、变形菌门及放线菌门的相对丰度,降低了绿弯菌门和厚壁菌门的相对丰度。冗余性分析结果表明速效钾是影响木薯根际土壤细菌群落的主要效应因子,与速效磷、碱解N、有机质、pH等指标呈现正相关。
