Meloidogyne enterolobii is a soil-borne disease that seriously harms crop production in tropical areas, and the agricultural losses caused by it are incalculable every year. For the control of root-knot nematodes, the traditional physical and chemical control methods have some disadvantages, while biological control is environmentally friendly and safe for people and livestock, so it has gradually become a research hotspot. This study used the impregnation method to screen strains with killing activity against the second instar larvae of M. enterolobii. The fermentation conditions and treatment time of the screened highly active strains were optimized. The strains were identified using 16S rRNA sequencing as well as physiological and biochemical methods, and growth promoting characteristics were analyzed. The research results indicated that the nematicidal activity of the fermentation supernatant of BWLY1PSB-1, BWLY2X-4 and BWLY3X-11 were more than 65%. Through morphological, physiological and biochemical characteristics, and molecular biology identification, BWLY1PSB-1, BWLY2X-4 and BWLY3X-11 were identified as Staphylococcus pateuri, Bacillus methylotrophicus and Bacillus sp., respectively; The nematicidal activity of the fermentation supernatant of each strain could reach the maximum after 24 h treatment of nematodes, and the optimal fermentation medium was LB medium, and the nematicidal activity of the supernatant reached relatively high when cultured for five days. The research results on the growth promoting characteristics of thee strains indicated that BWLY1PSB-1 had the ability to produce iron carriers and IAA, while BWLY2X-4 and BWLY3X-11 had the ability to produce proteases, cellulases, iron carriers, and IAA. The preliminary screening of excellent strains that can effectively control plant root-knot nematodes and have rowth promoting characteristics would enrich the microbial resources for the biological control of plant root-knot nematodes.

象耳豆根结线虫(Meloidogyne enterolobii)引起的作物根结线虫病是一种严重危害热区农作物生产的土传病害,每年造成的农业损失难以估量。对于根结线虫的防治,传统的物理、化学的防治手段存在一些弊端,而生物防治对环境友好、人畜安全,因此逐渐成为研究热点。本研究采用浸渍法筛选出对象耳豆根结线虫2龄幼虫具有抑杀活性的菌株,对筛选出的高活性菌株的发酵条件以及处理时间进行优化,利用16S rRNA序列及生理生化方法对菌株进行鉴定,并分析其促生特性。结果表明:有3株菌的发酵上清液杀线活性高于65%,分别为BWLY1PSB-1、BWLY2X-4、BWLY3X-11。通过形态特征、生理生化特征以及分子生物学鉴定,将BWLY1PSB-1鉴定为巴氏葡萄球菌(Staphylococcus pateuri),BWLY2X-4鉴定为甲基营养芽孢杆菌(Bacillus methylotrophicus),BWLY3X-11鉴定为芽孢杆菌属(Bacillus sp.)细菌;利用各菌株的发酵上清液处理线虫24 h后,其杀线活性即可达到最大,优适发酵培养基均为LB培养基,且培养5 d时上清液对线虫达到较好的致死率。BWLY1PSB-1具有产生铁载体和吲哚乙酸的能力,BWLY2X-4和BWLY3X-11具有产生蛋白酶、纤维素酶、铁载体和IAA的能力。本研究初步筛选出既能高效防控植物根结线虫又具有植物促生特性的优良菌株,丰富了植物根结线虫生物防治的微生物资源。

象耳豆根结线虫(Meloidogyne enterolobii)引起的作物根结线虫病是一种严重危害热区农作物生产的土传病害,每年造成的农业损失难以估量。对于根结线虫的防治,传统的物理、化学的防治手段存在一些弊端,而生物防治对环境友好、人畜安全,因此逐渐成为研究热点。本研究采用浸渍法筛选出对象耳豆根结线虫2龄幼虫具有抑杀活性的菌株,对筛选出的高活性菌株的发酵条件以及处理时间进行优化,利用16S rRNA序列及生理生化方法对菌株进行鉴定,并分析其促生特性。结果表明:有3株菌的发酵上清液杀线活性高于65%,分别为BWLY1PSB-1、BWLY2X-4、BWLY3X-11。通过形态特征、生理生化特征以及分子生物学鉴定,将BWLY1PSB-1鉴定为巴氏葡萄球菌(Staphylococcus pateuri),BWLY2X-4鉴定为甲基营养芽孢杆菌(Bacillus methylotrophicus),BWLY3X-11鉴定为芽孢杆菌属(Bacillus sp.)细菌;利用各菌株的发酵上清液处理线虫24 h后,其杀线活性即可达到最大,优适发酵培养基均为LB培养基,且培养5 d时上清液对线虫达到较好的致死率。BWLY1PSB-1具有产生铁载体和吲哚乙酸的能力,BWLY2X-4和BWLY3X-11具有产生蛋白酶、纤维素酶、铁载体和IAA的能力。本研究初步筛选出既能高效防控植物根结线虫又具有植物促生特性的优良菌株,丰富了植物根结线虫生物防治的微生物资源。

Meloidogyne enterolobii is a soil-borne disease that seriously harms crop production in tropical areas, and the agricultural losses caused by it are incalculable every year. For the control of root-knot nematodes, the traditional physical and chemical control methods have some disadvantages, while biological control is environmentally friendly and safe for people and livestock, so it has gradually become a research hotspot. This study used the impregnation method to screen strains with killing activity against the second instar larvae of M. enterolobii. The fermentation conditions and treatment time of the screened highly active strains were optimized. The strains were identified using 16S rRNA sequencing as well as physiological and biochemical methods, and growth promoting characteristics were analyzed. The research results indicated that the nematicidal activity of the fermentation supernatant of BWLY1PSB-1, BWLY2X-4 and BWLY3X-11 were more than 65%. Through morphological, physiological and biochemical characteristics, and molecular biology identification, BWLY1PSB-1, BWLY2X-4 and BWLY3X-11 were identified as Staphylococcus pateuri, Bacillus methylotrophicus and Bacillus sp., respectively; The nematicidal activity of the fermentation supernatant of each strain could reach the maximum after 24 h treatment of nematodes, and the optimal fermentation medium was LB medium, and the nematicidal activity of the supernatant reached relatively high when cultured for five days. The research results on the growth promoting characteristics of thee strains indicated that BWLY1PSB-1 had the ability to produce iron carriers and IAA, while BWLY2X-4 and BWLY3X-11 had the ability to produce proteases, cellulases, iron carriers, and IAA. The preliminary screening of excellent strains that can effectively control plant root-knot nematodes and have rowth promoting characteristics would enrich the microbial resources for the biological control of plant root-knot nematodes.

根结线虫(root-knot nematode)是侧尾腺纲垫刃目异皮总科根结线虫科根结线虫属的动物,是造成植物根结线虫病的罪魁祸首,是危害世界各地粮食、蔬菜生产的主要病原之一,每年都会对世界农作物造成巨大损失[1]。根结线虫生活史包括卵-幼虫-成虫3个阶段,在从卵中孵化出来到重新寄生植物这段时间其一直为2龄幼虫虫态,2龄幼虫也是它们唯一具备迁移能力的阶段,也是从该阶段开始寄生植物,线虫寄生植物后,其吻针会刺伤宿主并吸取寄主营养,并在作物根系上形成根结,影响宿主对营养的吸收而造成植株矮小、发黄、减产甚至绝收;此外,线虫给宿主造成的伤口会增加作物感染其他病害的几率,与其他病害形成复合侵染,加重病害发生,根结线虫种类超过90种,其寄主范围广泛,不同种类之间致病能力差异较大,且分布在各地的优势种群不同,所以在生产上造成的危害也有差别[1-4]。象耳豆根结线虫(Meloidogyne enterolobii)是目前我国记录的根结线虫中值得关注的一种,其对作物的危害与其他根结线虫类似,此外,象耳豆根结线虫繁殖水平高,具有克服MiNRk等抗性基因的能力,易对作物造成严重损害。目前在热带、亚热带和地中海地区已经发现有象耳豆根结线虫危害,且一些国家已将其列为检疫性病原[5-7]。据调查,象耳豆根结线虫在我国海南岛已经广泛存在,并且可以在海南岛的主要栽培作物,如葫芦科作物、茄果类作物、南药植物、蔬菜作物以及一些热带水果上广泛寄生,且有由南向北逐渐扩展的趋势[8-11],因此研究适宜的象耳豆根结线虫防治方法对防止其继续扩散,减轻其对作物威胁意义重大。
目前尚未发现方便、高效的植物根结线虫防治方法。在农业生产中,主要采取物理防治、化学防治、农业防治、生物防治等方法。生物防治相较于传统的防治手段具有巨大的优势,如降低农药残留,减轻环境污染等,此外生物防治还具有持效期长、改善土壤生态等优点[12]。自然界中,尤其是根结线虫病害高发地的土壤中存在着大量具有杀线虫活性的细菌,其中以芽孢杆菌属(Bacillus sp.)细菌的潜力尤为突出。芽孢杆菌属在自然界中广泛存在,在许多土壤生态环境中占据优势地位,也可在植物表面以及水体中存在,可产生芽孢,对高温、辐射化学药品等强烈的物理化学刺激有较强的抵抗能力[13]。芽孢杆菌属种类繁多,生理特性多样可产生多种抗生素,包括脂肽类、肽类、磷脂类、多烯类、氨基酸类、核酸类物质,对多种动、植物及人类病原菌起到很好的抑制作用[14-16]。许多芽孢杆菌属已被报道具有良好的生防效果。2014年至今,我国有116种芽孢杆菌类生物农药在中国农药信息网中登记,其中4种具有杀线虫作用,分别为坚强芽孢杆菌、甲基营养型芽孢杆菌、蜡质芽孢杆菌以及杀线虫芽孢杆菌,芽孢杆菌是一类极具开发潜力的细菌。
因此,为遏制根结线虫对果蔬作物造成的危害,亟需研发新的绿色安全的防治方法。本研究采用浸渍法筛选,从海南霸王岭采集的土壤样品中分离出的细菌中筛选出具有较高杀线活性的菌株并进行种类鉴定,同时开展菌株对象耳豆根结线虫2龄幼虫的致死条件优化筛选,研究结果将丰富植物根结线虫生物防治的微生物资源,并为后续研发高活性的生防杀线虫产品奠定基础。
生防菌株由中国热带农业科学院环境与植物保护研究所微生物资源与利用实验室分离、保存后提供;象耳豆根结线虫由中国热带农业科学院环境与植物保护研究所蔬菜病害与线虫课题组分离、鉴定、扩繁保存后提供。
LB培养基:氯化钠10 g,酵母粉5 g,胰蛋白胨10 g,琼脂15 g,蒸馏水1000 mL(液体培养基不加琼脂)。
PDA培养基:马铃薯200 g,葡萄糖20 g,琼脂15 g,蒸馏水1000 mL。
普通肉汤培养基:氯化钠5 g,蛋白胨20 g,牛肉粉5 g,蒸馏水1000 mL,pH 7.4~7.6。
羧甲基纤维素钠刚果红培养基:硫酸铵0.5 g,羧甲基纤维素钠5 g,磷酸氢二钾0.25 g,七水硫酸镁0.1 g,刚果红0.4 g,琼脂16 g,蒸馏水1000 mL。
酪蛋白培养基:NaCl 2.5 g,Na2HPO4 2 g,干酪素10 g,牛肉膏3 g,琼脂10 g,蒸馏水1000 mL。
以上培养基均在121 ℃条件下灭菌20 min。生理生化鉴定培养基的配制参照《微生物学实验》第2版[17]
(1)象耳豆根结线虫2龄幼虫获取。供试象耳豆根结线虫分离自海南文昌患病番茄和辣椒的病根,经中国热带农业科学院环境与植物保护研究所植物线虫研究实验室鉴定确认种类[9]。将病根洗净后剪碎,用清水浸没,按水∶次氯酸钠溶液=200∶1.5加入次氯酸钠溶液,用绞肉机搅拌2~4下,再手摇5~8 min。将300、400、500目的网筛从上到下依次叠放,将根系倒入筛网之后用水冲洗揉搓,将500目网筛收集的卵于清水中室温孵化,孵化后即为2龄幼虫,每天收集备用。
(2)细菌发酵液制备。将保存的细菌活化后接种于装有50 mL LB液体培养基的150 mL锥形瓶中,在180 r/min,37 ℃条件下培养24 h。将培养液于10 000 r/min,离心4 min,上清液即为细菌发酵液原液,于4 ℃保存,备用。
(3)生防菌发酵上清液对象耳豆根结线虫2龄幼虫的防效试验。吸取900 μL生防菌发酵液的5倍稀释液于24孔板中,再加入100 μL 2龄幼虫悬浮液(约含100头线虫),混匀后常温处理24 h,每处理设置2个重复,以清水组为对照。处理完成后收集处理液以清水定容至8 mL,2000 r/min,离心2 min,去上清,再定容至8 mL重悬4 h以上,然后在解剖镜下观察,统计死亡线虫数,线虫僵直不动且对机械触碰无反应的视为死亡。根据以下公式计算:
(1)形态学鉴定。将生防菌株接种在LB固体培养基上培养24 h后观察其菌落大小、颜色、质地、形状和湿润程度等。
(2)生理生化鉴定。菌株的糖发酵、甲基红、乙酰甲基甲醇、过氧化氢酶、苯丙氨酸脱氢酶试验参照《微生物学实验》第2版[17]
(3)分子生物学鉴定。将菌株用LB液体培养基培养12 h后离心收集菌体,用CTAB法提取DNA,–20 ℃保存备用。使用通用引物27F:5′-AGAGTTTGATCCTGGCTCAG-3′和1492R:5′-GGTTACCTTGTTACGACTT-3′对菌株的16S rRNA序列进行扩增。反应体系为50 μL,2×PCRMix 25 μL,引物各2 μL,DNA模板为2 μL,ddH2O为19 μL。PCR扩增程序如下:95 ℃预变性5 min;94 ℃变性30 s,60 ℃退火50 s,72 ℃延伸1 min,共35个循环;72 ℃延伸10 min。反应结束后将产物回收,送至生工生物工程(上海)股份有限公司进行测序,返回的序列在NCBI上进行序列比对,同时用MEGA 7.0软件以邻接法构建系统发育树,结合形态学观察结果与生理生化鉴定结果确定生防菌株的种属。
(1)种子液制备。用牙签挑取1.2.1筛选获得的3株生防菌株的单菌落,分别接种于10 mL的LB液体培养基中,于37 ℃、180 r/min摇培过夜作为种子液,备用。
(2)优势发酵培养基的筛选。分别吸取100 μL 3株生防菌株的种子液,接种于LB、PDA以及普通肉汤培养基中,于37 ℃、180 r/min培养后对其发酵上清液进行杀线活性测定。
(3)优势发酵时间的筛选。分别吸取100 μL 3株生防菌株的种子液,接种于筛选出的优势培养基中,于37 ℃、180 r/min进行摇培,分别在第3、5、7、10天收集发酵上清液进行杀线活性测定。
(4)对线虫处理时间的筛选。分别吸取100 μL 3株生防菌株的种子液,在筛选出的优势发酵条件下,于37 ℃、180 r/min进行培养,将其发酵上清液分别处理线虫24、48、72 h后,测定发酵上清液在不同处理时间的杀线活性。
(1)分别对菌株的产吲哚乙酸(IAA)[18]、铁载体[19]、蛋白酶[20]以及纤维素酶[21]能力进行测定。
(2)IAA标准曲线绘制。IAA标准曲线浓度依次为0、10、20、30、40、50 mg/mL,各吸取2 mL标准液加入试管中,加入等体积的Salkowski试剂混合均匀,在黑暗中静置30 min,于535 nm波长测定其吸光值(OD值),记录数据并绘制IAA标准曲线(图1),y=0.0338xR2=0.9949)。
采用Excel 2010和SPSS 19.0软件对试验数据进行统计分析。采用LSD法进行多重比较,采用Pearson法进行相关性分析,利用Excel 2010软件制图。
通过对分离后的菌株进行杀线活性评价,筛选到具有杀线活性的菌株:BWLY1PSB-1、BWLY1PSB-4、BWLY1X-9、BWLY2X-4、BWLY3X-11、QHY2X-18(透明),其中BWLY1PSB-1、BWLY3X-11和BWLY2X-4三个菌株的5倍稀释液对象耳豆根结线虫2龄幼虫的校正死亡率较高,分别为73.18%、71.92%和66.13%,因此,选取这3个菌株开展后续试验。
形态学观察表明,菌株BWLY1PSB-1的菌落(图2A)乳白色,不透明,圆形,边缘规则,光滑,表面凸起,湿润,革兰氏染色结果为阳性;菌株BWLY2X-4的菌落(图2B)白色,不透明,圆形,边缘呈光滑齿状,干燥,革兰氏染色结果为阳性;菌株BWLY3X-11的菌落(图2C)白色,不透明,圆形或不规则,干燥,培养3 d后,中间塌陷,革兰氏染色结果为阳性。经形态特征鉴定,可初步判断3株菌均为细菌。
BWLY1PSB-1的甲基红、蔗糖发酵、乳糖发酵、乙酰甲基甲醇(V-P)试验结果均呈阳性,苯丙胺酸脱氢酶与过氧化氢酶试验为阴性。BWLY2X-4与BWLY3X- 11的蔗糖发酵、乳糖发酵、V-P、过氧化氢酶试验结果均呈阳性,甲基红、苯丙胺酸脱氢酶试验为阴性(表1)。
菌株BWLY1PSB-1、BWLY2X-4、BWLY3X-11的16S rRNA长度分别为1449、1451、1448 bp,将所得序列分别在NCBI上进行比对,运用MEGA 7.0软件构建进化树(图3图4)。
综合菌株的菌落特征、生理生化特性与系统发育树,将BWLY1PSB-1鉴定为巴氏葡萄球菌(Staphylococcus pateuri),菌株BWLY2X-4鉴定为甲基营养芽孢杆菌(Bacillus methylotrophicus),BWLY3X-11鉴定为芽孢杆菌属细菌(Bacillus sp.)。
菌株BWLY1PSB-1、BWLY2X-4、BWLY3X-11分别在LB、PDB、普通肉汤培养基中培养后对象耳豆根结线虫2龄幼虫的致死率结果见表2。菌株BWLY1PSB-1、BWLY2X-4的LB、普通肉汤培养基的发酵上清液对线虫的杀线活性显著优于PDB培养基;而BWLY3X-11在LB和PDB培养基中的杀线虫活性显著优于普通肉汤。同时,3株菌株的LB培养基上清液对线虫的致死率无显著性差异,因此,选择LB培养基开展后续的培养时间与处理时间的优化试验。
菌株BWLY1PSB-1、BWLY2X-4、BWLY3X-1在不同培养时间下发酵上清液对象耳豆根结线虫2龄幼虫的致死率见表3。菌株BWLY1PSB-1在第7天达到最大杀线虫活性,并保持平稳;菌株BWLY2X-4在第7天达到峰值,但不同培养时间之间无显著性差异;菌株BWLY3X-11在培养到5 d时达到最大杀线虫活性,但第7天与第10天的发酵上清液的杀线虫活性会有所下降。3个菌株均培养5 d后对线虫达到较好的致死率,因此选择培养5 d的生防菌液开展对线虫处理时间的优化试验。
选择培养5 d的生防菌液开展对线虫处理时间的优化试验,3株生防菌发酵上清液不同处理时间对象耳豆根结线虫2龄幼虫的致死率见表4。3株生防菌的杀线虫活性在处理24 h时均达到最大,随着处理时间的延长,3株菌的杀线活性均逐渐下降。
研究表明,3株生防菌均具有产IAA的能力,向菌液中加入Salkowski试剂黑暗静置0.5 h后,与CK相比混合液产生明显的颜色变化,通过测定其吸光度,BWLY1PSB-1、BWLY2X-4和BWLY3X-11的吸光度分别为0.6828、0.4592、0.5586,将数值代入图1中的方程,计算出BWLY1PSB-1、BWLY2X-4和BWLY3X-11菌液中的IAA浓度分别为20.20、13.58、16.53 mg/L,菌株BWLY1PSB-1的产IAA能力最强。
3株生防菌均具有分泌铁载体的能力(图5)。培养1 d后,BWLY2X-4、BWLY3X-11菌落周围出现黄色晕圈;培养时间达到3 d时,BWLY1PSB-1周围也出现黄色晕圈;而BWLY3X-11产生黄色晕圈的速度最快,现象更明显,其分泌铁载体的潜力更强。
研究表明,菌株BWLY2X-4、BWLY3X-11均具有纤维素酶活性(图6)和蛋白酶活性(图7),而BWLY1PSB-1不具有这2种水解酶活性,且不能在酪蛋白培养基上生长。
目前所采取的病虫害防治方法中生物防治具有巨大的优势,其对于环境和人类的损害最小,更加贴合绿色环保的理念。由于成本低,使用简单,具有杀线虫活性的微生物的筛选与应用成为防治植物根结线虫的一大研究热点[22-23]。姚思敏等[24]发现1株具有杀线虫活性的巨大芽孢杆菌(B. megaterium),其盆栽试验和田间试验结果均显示出对拟禾本科根结线虫具有较好的防治效果,田间试验的防治效果最高可达56.3%。NGUYEN等[25]从黑胡椒根际中分离出20株杀线活性较好的生防菌,经鉴定,其中有14株属于芽孢杆菌属细菌,史凤玉等[26]从野生大豆中分离出2株对大豆孢囊线虫具有良好杀线活性的枯草芽孢杆菌和巨大芽孢杆菌,张立强等[27]在海南尖峰岭热带雨林土壤中分离出1株苏云金芽孢杆菌,对松材线虫和南方根结线虫具有良好的杀线活性且效果稳定。中国农药信息网数据显示,近几年也有登记芽孢杆菌类的生物农药,陕西恒田生物农业有限公司2018年登记了一款以甲基营养型芽孢杆菌LW-6为主要成分的生物农药,可用于防治番茄根结线虫,此外还有坚强芽孢杆菌、蜡质芽孢杆菌、杀线虫芽孢杆菌等,说明生防芽孢杆菌在防治线虫上的应用具有巨大的潜力。而有关利用葡萄球菌防治根结线虫的研究较少,袁莉[28]筛选出1株沃氏葡萄球菌,对拟禾本科根结线虫具较好的防效。于丰源[29]筛选出1株杀线活性达91.75%的菌株,经鉴定确定为松鼠葡萄球菌,但葡萄球菌对人体的危害以及是否有应用于实际生产的潜力仍未知,需再进一步研究。
微生物对植物的促生作用包括产铁载体、分泌植物激素、产水解酶(蛋白酶、淀粉酶等)等途径,IAA是调控植物生长的重要信号物质,是促进植物生长的激素,铁载体又称为嗜铁素,是微生物产生的金属螯合物质,在低铁环境中,微生物产生的铁载体能够还原Fe3+为Fe2+供植物吸收[30-31]。通过对3株生防菌的促生特性进行测定,发现3株菌均具有产IAA和铁载体能力,BWLY2X-4和BWLY3X-11还能产生纤维素酶和蛋白酶,具有良好的促生潜力。程鑫宇等[32]通过分离产IAA的根际促生菌并将其用于盐碱胁迫下的玉米和小麦,发现其对盐碱胁迫具有较好的缓解作用。朱森林等[33]发现1株解淀粉芽孢杆菌对香蕉枯萎病4号生理小种具有较好的防效,同时也能促进香蕉的生长发育,通过测定发现该菌株为高产铁载体菌株且能分泌IAA。本研究中的3株生防菌具有杀线活性的同时还具有一定的促生特性,推测其在防治象耳豆根结线虫的同时还能在一定程度上促进作物的生长发育。
本研究通过浸渍法从本实验室分离的一批细菌中筛选出3株杀线活性较好的菌株(BWLY1PSB-1、BWLY2X-4和BWLY3X-11),并对其发酵的培养基、时间以及处理时间进行了优化;通过研究其促生特性发现3株菌均具有产IAA和铁载体的能力,BWLY2X-4、BWLY3X-11还具有产蛋白酶和纤维素酶的能力,具有一定的促生特性。本研究初步筛选出既有高效防控植物根结线虫又具促生特性的优良菌株,丰富了植物根结线虫生物防治的微生物资源,但由于本研究仅进行了室内试验,仍需开展小区与大田试验验证3株生防菌对线虫的防控效果及对作物的促生能力。
