Mango (Mangifera indica L.) is an important tropical fruit in Hainan province. Ethylene plays a vital role in the ripening process of mango. ERF transcription factors are key downstream components of the ethylene signaling pathway, and as one of the largest families of transcription factors, they play a role in regulating signal transduction and physiological responses. To investigate the impact of ERF transcription factors on the coloration of postharvest mango peels, this study focused on the Guifei mango as the research subject. Through PCR cloning, the MiERF3 gene was obtained, and bioinformatics methods were used to analyze its basic physicochemical characteristics, conserved domains, protein structure, and evolutionary relationships. Real-time quantitative PCR (qRT-PCR) technology was then employed to analyze the specific expression of the MiERF3 gene in different tissues of mango and during postharvest storage periods. The results indicated that MiERF3 was 717 bp in length, encoding 238 amino acids. The protein had a molecular formula of C1130H1763N339O355S9, a molecular weight of 26.066 kDa, and a theoretical isoelectric point of 8.62, with 30 positively charged residues and 27 negatively charged residues. The protein exhibited hydrophilic properties and a structurally unstable nature. MiERF3 protein had no a signal peptide and transmembrane region, with threonine being the primary site for phosphorylation modification. It contained an AP2 conserved domain, and its secondary structure was primarily composed of 61.67% random curl, along with 27.75% α-helix, 3.96% β-fold, and 6.61% extended chain. The three-dimensional structural model of MiERF3 contained three β-fold and one α-helix, consistent with the characteristics of the AP2 structural domain. Multi-sequence alignment and motif analysis suggested that MiERF3 belonged to the ERF subfamily. Based on phylogenetic analysis, the protein most closely related to MiERF3 was PvERF3-like from Pistacia vera, a plant belonging to the same family as mango. Onion subcellular localization and transcriptional self-activation assays showed that MiERF3 was localized in the nucleus and had transcriptional self-activating activity. Quantitative real-time fluorescence analysis showed that the expression of MiERF3 varied among different mango tissues, with the highest expression observed in the sunny side peel of mango fruit. During storage, the expression of MiERF3 showed a clear trend of initial increase followed by decrease, suspecting that MiERF3 may play a positive regulatory role in the process of fruit ripening and color change. This study would provide a theoretical basis for revealing the regulatory role of ERF transcription factors in postharvest ripening and senescence of mango.

芒果(Mangifera indica L.)是海南省重要的热带水果之一,是典型的呼吸跃变型水果,乙烯在芒果后熟过程中扮演着至关重要的角色。ERF转录因子是乙烯信号通路的关键下游成分,作为最大的转录因子家族之一,起到调节信号传导和生理反应的作用。为了探究ERF转录因子对采后芒果果皮色泽的影响,本研究以贵妃芒果为研究对象,通过PCR克隆得到MiERF3基因,利用生物信息学方法分析MiERF3的基本理化特征、保守结构域、蛋白质结构、进化关系等,利用实时荧光定量PCR(qRT-PCR)技术分别对MiERF3基因在芒果不同组织部位和采后贮藏时期的特异性表达进行分析。结果表明:MiERF3基因全长717 bp,编码238个氨基酸,蛋白分子式为C1130H1763N339O355S9,分子量为26.066 kDa,理论等电点为8.62,带30个正电残基和27个负电残基,蛋白具有亲水性,结构不稳定;MiERF3蛋白不含信号肽,不含跨膜区域,磷酸化修饰以苏氨酸为主,包含1个AP2保守结构域,二级结构以61.67%的无规卷曲为主,还含有27.75%的α-螺旋、3.96%的β-折叠、6.61%的延长链,其三维结构模型包含有3个β-折叠和1个α-螺旋,符合AP2结构域的特征;多序列比对和motif分析结果表明,MiERF3蛋白属于ERF亚家族成员;系统进化树分析结果表明,与MiERF3蛋白亲缘关系最近的是同为漆树科植物的阿月浑子PvERF3-like;洋葱亚细胞定位及转录自激活检测结果显示,MiERF3蛋白定位于细胞核且具有转录自激活活性;实时荧光定量分析表明,MiERF3基因的表达量在不同芒果部位表达情况不同,在芒果果实阳面果皮的表达量最高,且在芒果贮藏期间,其表达量呈明显的先增后减的趋势,故而推测MiERF3在果实成熟转色过程中起正调控作用。本研究为揭示ERF转录因子在芒果采后成熟衰老过程中的调控作用提供理论依据。

芒果(Mangifera indica L.)是海南省重要的热带水果之一,是典型的呼吸跃变型水果,乙烯在芒果后熟过程中扮演着至关重要的角色。ERF转录因子是乙烯信号通路的关键下游成分,作为最大的转录因子家族之一,起到调节信号传导和生理反应的作用。为了探究ERF转录因子对采后芒果果皮色泽的影响,本研究以贵妃芒果为研究对象,通过PCR克隆得到MiERF3基因,利用生物信息学方法分析MiERF3的基本理化特征、保守结构域、蛋白质结构、进化关系等,利用实时荧光定量PCR(qRT-PCR)技术分别对MiERF3基因在芒果不同组织部位和采后贮藏时期的特异性表达进行分析。结果表明:MiERF3基因全长717 bp,编码238个氨基酸,蛋白分子式为C1130H1763N339O355S9,分子量为26.066 kDa,理论等电点为8.62,带30个正电残基和27个负电残基,蛋白具有亲水性,结构不稳定;MiERF3蛋白不含信号肽,不含跨膜区域,磷酸化修饰以苏氨酸为主,包含1个AP2保守结构域,二级结构以61.67%的无规卷曲为主,还含有27.75%的α-螺旋、3.96%的β-折叠、6.61%的延长链,其三维结构模型包含有3个β-折叠和1个α-螺旋,符合AP2结构域的特征;多序列比对和motif分析结果表明,MiERF3蛋白属于ERF亚家族成员;系统进化树分析结果表明,与MiERF3蛋白亲缘关系最近的是同为漆树科植物的阿月浑子PvERF3-like;洋葱亚细胞定位及转录自激活检测结果显示,MiERF3蛋白定位于细胞核且具有转录自激活活性;实时荧光定量分析表明,MiERF3基因的表达量在不同芒果部位表达情况不同,在芒果果实阳面果皮的表达量最高,且在芒果贮藏期间,其表达量呈明显的先增后减的趋势,故而推测MiERF3在果实成熟转色过程中起正调控作用。本研究为揭示ERF转录因子在芒果采后成熟衰老过程中的调控作用提供理论依据。

Mango (Mangifera indica L.) is an important tropical fruit in Hainan province. Ethylene plays a vital role in the ripening process of mango. ERF transcription factors are key downstream components of the ethylene signaling pathway, and as one of the largest families of transcription factors, they play a role in regulating signal transduction and physiological responses. To investigate the impact of ERF transcription factors on the coloration of postharvest mango peels, this study focused on the Guifei mango as the research subject. Through PCR cloning, the MiERF3 gene was obtained, and bioinformatics methods were used to analyze its basic physicochemical characteristics, conserved domains, protein structure, and evolutionary relationships. Real-time quantitative PCR (qRT-PCR) technology was then employed to analyze the specific expression of the MiERF3 gene in different tissues of mango and during postharvest storage periods. The results indicated that MiERF3 was 717 bp in length, encoding 238 amino acids. The protein had a molecular formula of C1130H1763N339O355S9, a molecular weight of 26.066 kDa, and a theoretical isoelectric point of 8.62, with 30 positively charged residues and 27 negatively charged residues. The protein exhibited hydrophilic properties and a structurally unstable nature. MiERF3 protein had no a signal peptide and transmembrane region, with threonine being the primary site for phosphorylation modification. It contained an AP2 conserved domain, and its secondary structure was primarily composed of 61.67% random curl, along with 27.75% α-helix, 3.96% β-fold, and 6.61% extended chain. The three-dimensional structural model of MiERF3 contained three β-fold and one α-helix, consistent with the characteristics of the AP2 structural domain. Multi-sequence alignment and motif analysis suggested that MiERF3 belonged to the ERF subfamily. Based on phylogenetic analysis, the protein most closely related to MiERF3 was PvERF3-like from Pistacia vera, a plant belonging to the same family as mango. Onion subcellular localization and transcriptional self-activation assays showed that MiERF3 was localized in the nucleus and had transcriptional self-activating activity. Quantitative real-time fluorescence analysis showed that the expression of MiERF3 varied among different mango tissues, with the highest expression observed in the sunny side peel of mango fruit. During storage, the expression of MiERF3 showed a clear trend of initial increase followed by decrease, suspecting that MiERF3 may play a positive regulatory role in the process of fruit ripening and color change. This study would provide a theoretical basis for revealing the regulatory role of ERF transcription factors in postharvest ripening and senescence of mango.

芒果,素有“热带水果之王”之美誉,具有很高的营养和经济价值。其中贵妃芒果以其黄、红相间的果皮以及核薄肉厚的特点深受人们喜欢[1]。果皮色泽是最直观的果实品质性状,花色苷是贵妃芒果最重要的色素物质[2],使果实展现为与其他芒果品种不同的红色。乙烯是一种重要的植物激素,对果实的后熟生理变化和次生代谢具有重要的调控作用[3]。伴随生物学技术的发展,关于乙烯对果实后熟生理变化的影响及次生代谢的调控研究取得了显著的进展[4]
研究指出,乙烯可以影响苯丙氨酸转氨酶(PAL)和查尔酮异构酶(CHI)的活性,从而实现对花青素生成的促进或抑制[5]。ERF转录因子是乙烯信号通路的关键下游成分,作为最大的转录因子家族之一,起到调节信号传导和生理反应的作用[6]。AP2/ERF转录因子超家族存在于众多植物中,该家族蛋白特征明显,含有典型的AP2/ERF结构域,具有结合DNA的功能[7]。AP2/ERF最早从烟草和拟南芥中分离发现,功能域主要由转录调控功能域、DNA结合功能域、寡聚化位点功能域和核定位信号功能域4个部分构成[8]。AP2/ERF转录因子家族成员包含AP2/ERF结构域,该结构域是ERF行使转录调控的关键,也是与其他转录因子区分的主要依据[9-10]。AP2/ERF家族转录因子根据其含有的结构域数量和是否含有其他类型结构域为依据,将其分为4个亚族(AP2、DREB、ERF、RAV)和单独成员(Soloist)[1113]
大量文献表明,在许多水果中已经发现ERF转录因子作为与果实色泽转变相关的激活因子或抑制因子,在果实成熟的过程中,ERF转录因子因其多样性,可能发挥多种功能[1417]。研究表明,番茄果实中的SiERF6能够参与调控类胡萝卜素合成途径和乙烯代谢途径,在成熟过程中发挥重要的调控作用[18]。经乙烯处理后,SlERF-E1、SlERF-E2和SlERF-E4能够有效调控番茄果实成熟转色[19]。WANG等[20]的研究结果表明,6个ERF基因在桃子成熟过程中显著上调,表现出与成熟相关的表达模式。有研究表明,乙烯信号转导过程中,AP2/ERF转录因子能够结合乙烯合成相关基因启动子从而调控果实成熟与发育,也可以激活新受体的合成或受体的降解来调控乙烯反应的转导[6]。如,在梨中,转录因子PbERF24可以结合PbACO54的启动子参与梨果实成熟[21];在苹果中,MdERF2/3可以通过结合MdACS1启动子中的DRE结合位点参与果实成熟[22-23];CpERF9作为转录抑制因子,通过结合CpPME1/2和CpPG5启动子中的GCC box来控制木瓜果实成熟[24-25];在香蕉中,MaERF9和MaERF11也可作为转录激活物或抑制物,在转录过程中结合乙烯合成相关基因启动子达到调控果实成熟的作用[26]。因此,本研究结合前期报道及本课题组转录组测序数据,以采后贵妃芒果为材料,通过PCR扩增技术,获得1个AP2/ERF转录因子MiERF3,并进行不同层面的生物信息学分析,随后对其在芒果不同组织部位以及芒果贮藏阶段的基因表达量进行检测。本研究旨在进一步探讨ERF在芒果乙烯信号传导过程中的作用,以及其在芒果采后贮藏保鲜应用中的功能,为相关领域提供理论依据和参考。
本研究采用产自海南省陵水县英州镇芒果基地(18°25′N,109°51′E)的贵妃芒果(Mangifera indica L. var.)作为试材。在果实达到八成熟阶段时,立即进行采摘并运送至实验室。在培养箱恒温恒湿条件下,将采摘后的芒果分别贮藏0(即采摘当天)、3、6、9、12 d,温度25 ℃,湿度为95%。随后,采集芒果阴面的果皮样本,经过液氮速冻处理后,保存于-80 ℃的冰箱,以备后续的基因克隆及qRT-PCR分析使用。
大肠杆菌DH5 α感受态、农杆菌GV3101感受态、酵母菌Y2H感受态购自海口基睿生物科技有限公司提供。DL 2000 DNA marker、Taq DNA聚合酶、限制性内切酶SpeⅠ、BglⅡ、琼脂糖、T4 DNA连接酶、氨苄青霉素、卡那霉素等购自海南合辉实业有限公司。引物合成与测序由生工生物工程(上海)股份有限公司完成。
通过CTAB法提取芒果果皮基因组RNA后,采用1%琼脂糖凝胶电泳对RNA质量进行严格检测。使用超微量分光光度计(Implen,Germany)对2 μL的RNA样本进行浓度和纯度的精确测定。参照MonScriptTM RTIII All-in-One Mix with dsDNase试剂盒说明书进行逆转录RNA。所有合格的样品保存于-20 ℃备用。
结合实验室前期结果与对芒果转录组数据的分析,筛选出MiERF3基因。随后,利用NCBI(https://www.ncbi.nlm.nih.gov/)、Expasy(https://web.expasy.org/translate/)、Uniprot(https://www.uniprot.org/blast/)在线工具以及TBtools等专业软件工具,对MiERF3基因序列进行深入分析。使用Primer 5软件设计全长引物(表1),利用PCR技术扩增目标基因。经1%琼脂糖凝胶电泳检测,回收约720 bp大小的目的条带。将回收的片段与pMD19-T载体在4 ℃条件下进行过夜连接,并将连接产物转入DH5α,随后进行菌落PCR验证。将阳性克隆菌液送至生工生物工程(上海)股份有限公司进行测序,测序结果使用DNAMAN 6.0软件进行序列比对。
利用NCBI Blast程序对克隆得到的MiERF3基因的CDS序列进行比对分析。此外,利用在线软件NCBI Conserved Domains(https://www.ncbi.nlm.nih.gov/)预测MiERF3蛋白质的保守结构域。利用ExPASy ProtParam(https://web.expasy.org/protparam/)、NetPhos 3.1(Serverhttps://services.healthtech.dtu.dk/services/NetPhos-3.1/)、Sig nalP-4.1(https://services.healthtech.dtu.dk/services/SignalP-4.1/)和TMHMM Server v 2.0(https://services.healthtech.dtu.dk/services/TMHMM-2.0/)等在线软件进行序列分析。同时,通过SOPMA(https://npsa-prabi.ibcp.fr/cgi-bin/npsa_automat.pl?page=/NPSA/npsa_sopma.html)和SWISS-MODEL(https://swissmodel.expasy.org/interactive)在线软件对MiERF3蛋白的结构进行分析。
为了探究MiERF3在不同物种中的保守性和进化关系,使用DNAMAN 6.0软件进行了多序列比对,并利用MEME 5.5.0软件分析其motif结构,将motif数设为10。使用MEGA-X软件构建系统进化树。
采用洋葱内表皮转化法[31]进行亚细胞定位分析,在原方法上进行了适度优化。首先,设计特异性引物(表1),并运用双酶切技术,将目的片段构建至pCAMBIA1302-GFP(35S:: GFP)表达载体中,生成35S:: MiERF3-GFP融合表达载体。选用SpeⅠ和BglⅡ作为酶切位点。将重组质粒转化至GV3101农杆菌,并制备OD600为0.8~1.0的悬浮液。将洋葱内表皮(2 cm×2 cm)浸泡于该悬浮液中,减压处理10 min。使用滤纸缓慢吸取洋葱内表皮过多的液体,确保其干燥后,再转移到铺有滤纸的MS固体培养基上培养48 h。培养结束后,制作玻片样本,并通过倒置荧光显微镜观察并记录荧光信号。
运用双酶切构建表达载体,引物序列见表1。利用T4 DNA连接酶将启动子序列插入到载体上,构建重组质粒。将pGBKT7-MiERF3重组质粒、pGADT7空载质粒与Y2H酵母感受态(50 μL)、Carrier DNA(10 μL)以及PEG/LiAc buffer(500 μL)充分混匀。30 ℃水浴30 min后42 ℃水浴15 min,5000 r/min离心40 s弃上清,ddH2O 400 μL重悬,离心30 s弃上清。用ddH2O 50 μL;重悬,涂至DDO培养基上,29 ℃培养48~96 h。挑取正常生长的酵母菌落用0.9% NaCl溶液将其梯度稀释,点至QDO、QDO/X-α-Gal/AbA(1000 ng/mL)培养基上,观察酵母菌生长状况。pGBKT7-Lam、pGADT7-T为阴性对照,pGBKT7-p53、pGADT7-T为阳性对照。
用实时荧光定量PCR(qRT-PCR)技术,分析芒果在采后贮藏期和不同组织部位中MiERF3基因的表达。芒果Actin1基因为内参基因,采用Primer 5软件设计引物(表1)。参照ChamQ Universal SYBR qPCR Master Mix(Vazyme,中国)试剂盒进行qRT-PCR。PCR反应体系10 μL,反应程序为:95 ℃预变性30 s;95 ℃变性5 s,60 ℃退火30 s,72 ℃延伸5 s,进行40次循环。每个样品设置3次生物学重复,基因相对表达量计算采用2-∆∆CT法,以0 d为参照,设其相对表达量为1。数据统计与分析使用WPS Office 2022软件,相对表达量可视化图形使用GraphPad Prism 8软件绘制。
以贵妃芒果阴面果皮cDNA为模板,用MiERF3-F/R引物对其编码区序列进行PCR扩增,扩增产物与目的基因大小一致(图1)。MiERF3(XP_044473973.1)基因CDS序列全长为717 bp,编码238个氨基酸。MiERF3蛋白分子式C1130H1763N339O355S9,相对分子量26.066 kDa,等电点(pI)为8.62,正、负电荷的氨基酸残基总数分别为30个和27个,脂肪系数为51.10,不稳定系数为59.70,总平均亲水性为-0.655,由此推测MiERF3蛋白为不稳定的亲水蛋白。对MiERF3蛋白进行磷酸化位点预测(图2A),MiERF3含有Ser(22个)、Thr(16个)、Tyr(6个)共44个磷酸化位点。MiERF3蛋白无信号肽(图2B)、不含跨膜区域(图2C),在细胞膜内,不属于分泌型蛋白。采用SOPMA软件预测MiERF3蛋白的二级结构,发现无规则卷曲为150个氨基酸(61.67%)、α-螺旋为49个氨基酸(20.9%)、延伸链为19个氨基酸(12.18%)β-折叠为10个氨基酸(4.20%)(图3A)。利用SWISS-MODEL预测该蛋白的三级结构(图3B),MiERF3蛋白含有3个反平行的β-折叠和1个α-螺旋结构,符合AP2结构域典型的结构特征。
ERF蛋白关键保守结构域是其发挥功能的基础。使用NCBI网站的CD-Search工具预测MiERF3蛋白功能结构域,结果显示(图4A),MiERF3蛋白N端第39位至98位是1个含59个氨基酸的AP2结构域。
选取与MiERF3同源性较高的9个物种的ERF转录因子进行motif分析,结果显示,这些motif结构相似,有4种motif在所有蛋白中均含有,与MiERF3所含的motif数目和种类一致的有3种,分别为阿月浑子、楝、克莱门柚(图4B)。
经氨基酸同源性比对发现,芒果MiERF3与楝(Melia azedarach,KAJ4725160.1)、克莱门柚(Citrus x clementina,XP_006452450.1)、番木瓜(Carica papaya,XP_021906162.1)、阿月浑子(Pistacia vera,XP_031286524.1)、雷公藤(Tripterygium wilfordii,XP_038689087.1)、水银草(Mercurialis annua,XP_050215426.1)氨基酸序列的相似性分别为68.67%、78.22%、66.54%、92.02%、63.93%和59.76%。将上述氨基酸序列通过DANMAN 5.0软件进行多重比对,结果显示(图4C),MiERF3与其他物种的ERF蛋白序列相似,AP2结构域高度保守,在该结构域中,第14位氨基酸为丙氨酸(A),而天冬氨酸(D)则位于第19位,表明MiERF3蛋白具有ERF亚家族的序列特征,属于AP2/ERF家族中的ERF亚族成员。
选取漆树科植物、模式植物、热带植物等不同物种ERF蛋白构建进化树,结果显示(图4D),芒果MiERF3与同样是漆树科的阿月浑子ERF3-like聚在一起,亲缘关系最近,其次是木薯、甜橙,而热带植物番木瓜、菠萝以及模式植物番茄则与芒果MiERF3亲缘关系较远。
荧光倒置显微镜下观察到,35S: GFP对照洋葱内表皮细胞中的细胞核和细胞壁均检测到了绿色荧光信号,而35S: MiERF3-GFP融合蛋白仅在细胞核中出现绿色荧光信号(图5),这表明芒果MiERF3蛋白定位于洋葱细胞细胞核中,符合转录因子所具有的核定位特征。
将pGBKT7(空载)和pGBKT7-MiERF3重组质粒转入Y2H Gold感受态,酵母菌在SDO培养基上生长正常且与对照菌斑生长大小基本一致(图6A),证明MiERF3蛋白没有毒性。
将pGBKT7-MiERF3重组质粒、pGADT7空载质粒转入Y2H Gold感受态,菌株可在DDO培养基上正常生长。进一步观察发现,实验组(pGBKT7-MiERF3+pGADT7)和阳性对照(pGBKT7-p53+pGADT7-T)在QDO和QDO/X-α-Gal/AbA培养基上的菌斑大小基本一致,且在QDO/X-α-Gal/AbA培养基上的菌斑为蓝色,而阴性对照(pGBKT7-lam+pGADT7-T)在QDO和QDO/X-α-Gal/AbA培养基上均未生长出菌斑(图6B)。表明MiERF3蛋白具有较强转录自激活活性。
为了探究MiERF3基因在芒果不同组织部位表达情况,采用实时荧光定量方法对贵妃芒果同一时期的核、叶、花、茎、果肉、阴面果皮和阳面果皮组织样品进行检测。结果显示(图7A),MiERF3基因在芒果阳面果皮中表达量最高,其次是阴面果皮和茎,在核、叶、花和果肉等组织中的表达量则极低。
为了探究MiERF3基因在贵妃芒果贮藏过程中的表达情况,选择采后贮藏0、3、6、9、12 d的阴面果皮组织,提取总RNA后逆转录,再进行qRT-PCR检测。结果表明(图7B),在果实贮藏过程中,MiERF3基因的表达量整体呈先上升后下降的趋势。在贮藏初期(0 d),该基因的表达量处于最低水平,随着果实的成熟,MiERF3基因的表达量开始上调,并在第6天达到峰值,此时其表达量是初始时期的4倍。这一结果表明MiERF3基因在芒果果实采后贮藏和成熟过程中可能发挥着一定的作用。
芒果果皮颜色直接影响着芒果的外观和品质,与多个色泽相关物质的相对含量密切相关。这些物质的变化决定了芒果的美观度、营养价值以及保健功效[21,27]。果实采后保鲜技术手段的选择对于果皮颜色以及果实风味的影响巨大。由于芒果果实属于典型的呼吸跃变型,生产上往往选择在果实八成熟时采收,随后使用乙烯利进行催熟再销售。乙烯利处理会促进果实成熟,加速果皮转色,使果实硬度降低,快速到达其最佳销售状态。乙烯是ERF的命名来源,展现了其与ERF之间的密不可分关系[28]。乙烯通过信号转导机制,在植物体内诱导ERF参与生长发育及胁迫防御等生理过程。在拟南芥中,乙烯与位于内质网膜上的乙烯受体结合,导致与受体相关联的激酶CTR1失活,进而无法对EIN2的C端进行磷酸化[29]。非磷酸化的EIN2随后进入细胞核,使得转录因子EIN3得以稳定存在并发挥其作用。随后,EIN3激活包括ERF1在内的多个转录因子的转录过程,这些转录因子调控其他乙烯响应基因,完成整个乙烯信号响应过程[30]。因此,本研究通过课题组前期研究结果选择MiERF3作为研究对象,旨在探究其在贵妃芒果中的生理及分子层面的作用,为芒果产业生产实践提供一定的理论依据和参考。
魏玲等[31]在贵妃芒果中成功鉴定出1个MiERF2基因,该基因属于ERF亚家族成员。经过分析发现,信号肽和跨膜结构在该基因编码的蛋白质中并不存在。在亲缘关系上,阿月浑子的PvERF109-like与MiERF2的关系最为接近。这一发现与本研究的结果存在一定的相似性。本研究发现,芒果中的MiERF3属于ERF亚家族,其编码的蛋白质具有亲水性但不够稳定,不含有信号肽,也未发现跨膜结构。在MiERF3蛋白质二级和三级结构预测中,该蛋白质主要由无规则卷曲构成,同时包含α-螺旋、β-折叠和延长链等结构元。MiERF3蛋白质具有典型的AP2结构域三维结构。通过系统进化分析,发现MiERF3与同为漆树科的阿月浑子PvERF3-like蛋白的亲缘关系最为接近。且亚细胞定位和转录自激活分析实验,证明MiERF3是一个定位于细胞核且具有转录自激活活性的转录因子。
为了深入研究MiERF3基因在芒果不同组织部位以及成熟阶段中的表达情况,本研究采用实时荧光定量的方法进行了详细分析。结果显示,MiERF3基因主要在芒果的阳面果皮中表达,其表达量显著高于其他部位。此外,随着果实的逐渐成熟,MiERF3基因的表达量也在不断增加,特别是在采后贮藏的第6天,该基因的表达量达到了最高峰,但随后开始呈现下降趋势。这一发现为进一步了解MiERF3基因在芒果生长发育过程中的作用提供了重要依据。郑珍珍等[32]发现,在草莓果实成熟的过程中,FaERF003的表达水平迅速上升,其变化趋势与乙烯前体ACC的含量变化十分相似。易萍等[33]也观察到MiERF113基因在果皮和果肉中均有表达,并且随着果实的逐渐成熟,其表达量也在不断增加,在果实采后贮藏的第12天,MiERF113基因的表达量达到最高,这与本研究结果具有一定的相似性。因此,推测MiERF3基因具有类似功能,在芒果成熟过程中尤其是果皮转色方面发挥着重要的作用。然而,MiERF3基因在乙烯信号转导通路中的分子机制以及功能验证还有待探索。
