Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV) are common viruses in Phalaenopsis production, and monitoring CymMV and ORSV of in vitro seedlings is the foundation of high-quality and healthy seedling production in Phalaenopsis. In this study, 20 Phalaenopsis cultivars were used as the research objects. Two batches of in vitro seedlings selected from the production line with 10-month intervals between the subculture proliferation cycles, were investigated for CymMV and ORSV through RT-PCR (reverse transcription polymerase chain reaction) and RT-qPCR (real time quantitative polymerase chain reaction) at the same time. RT-PCR results showed that the positive rate of CymMV in the first and second batch was 25% and 20%, respectively, and the positive rate of ORSV was both 15% in the two batches. RT-qPCR results showed that the positive rate of CymMV in the first and second batch was 100% and 91.67%, respectively, and the positive rate of ORSV in the two batches was 50% and 25%, respectively. Comparing with the first batch, CymMV content of 16 cultivars and ORSV content of 9 cultivars decreased in the second batch, but ORSV content of Phalaenopsis Sogo Yukidian V3 increased significantly, which increased from negative to 6.85×105-8.50×105 copies/μL. The results indicated that CymMV and ORSV widely existed in in vitro seedlings of Phalaenopsis, and the content varied with different batches. The cultivars detected as the negative also faced the risk of virus content increase. Thus, CymMV and ORSV should be investigated regularly in the production of in vitro Phalaenopsis seedlings to ensure the high-quality seedling source for Phalaenopsis industry.

建兰花叶病毒(Cymbidium mosaic virus,CymMV)和齿兰环斑病毒(Odontoglossum ringspot virus,ORSV)是蝴蝶兰生产中的常见病毒,对组培苗CymMV和ORSV的携带情况进行监测是生产蝴蝶兰优质健康种苗的前提。本研究以20个蝴蝶兰品种为研究对象,从组培生产线上选取继代增殖周期间隔10个月的2个批次组培苗样品,同时采用RT-PCR和RT-qPCR进行CymMV和ORSV检测。RT-PCR检测结果显示,第1批次和第2批次样品的CymMV阳性率分别为25%和20%,ORSV阳性率均为15%。RT-qPCR检测结果显示,第1批次和第2批次样品的CymMV阳性率分别为100%和91.67%,ORSV阳性率分别为50%和25%。与第1批次相比,第2批次组培苗分别有16个和9个品种的CymMV和ORSV含量减少,但V3的ORSV含量增加明显,由阴性增加至6.85×105~8.50×105 copies/μL。研究结果表明,CymMV和ORSV广泛存在于蝴蝶兰组培苗中,且病毒含量随批次存在差异,检测为阴性的品种在继代增殖过程中亦存在病毒量增加的风险。因此,在蝴蝶兰组培苗生产过程中应定期进行CymMV和ORSV监测,以保证蝴蝶兰产业的优质种源。

建兰花叶病毒(Cymbidium mosaic virus,CymMV)和齿兰环斑病毒(Odontoglossum ringspot virus,ORSV)是蝴蝶兰生产中的常见病毒,对组培苗CymMV和ORSV的携带情况进行监测是生产蝴蝶兰优质健康种苗的前提。本研究以20个蝴蝶兰品种为研究对象,从组培生产线上选取继代增殖周期间隔10个月的2个批次组培苗样品,同时采用RT-PCR和RT-qPCR进行CymMV和ORSV检测。RT-PCR检测结果显示,第1批次和第2批次样品的CymMV阳性率分别为25%和20%,ORSV阳性率均为15%。RT-qPCR检测结果显示,第1批次和第2批次样品的CymMV阳性率分别为100%和91.67%,ORSV阳性率分别为50%和25%。与第1批次相比,第2批次组培苗分别有16个和9个品种的CymMV和ORSV含量减少,但V3的ORSV含量增加明显,由阴性增加至6.85×105~8.50×105 copies/μL。研究结果表明,CymMV和ORSV广泛存在于蝴蝶兰组培苗中,且病毒含量随批次存在差异,检测为阴性的品种在继代增殖过程中亦存在病毒量增加的风险。因此,在蝴蝶兰组培苗生产过程中应定期进行CymMV和ORSV监测,以保证蝴蝶兰产业的优质种源。

Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV) are common viruses in Phalaenopsis production, and monitoring CymMV and ORSV of in vitro seedlings is the foundation of high-quality and healthy seedling production in Phalaenopsis. In this study, 20 Phalaenopsis cultivars were used as the research objects. Two batches of in vitro seedlings selected from the production line with 10-month intervals between the subculture proliferation cycles, were investigated for CymMV and ORSV through RT-PCR (reverse transcription polymerase chain reaction) and RT-qPCR (real time quantitative polymerase chain reaction) at the same time. RT-PCR results showed that the positive rate of CymMV in the first and second batch was 25% and 20%, respectively, and the positive rate of ORSV was both 15% in the two batches. RT-qPCR results showed that the positive rate of CymMV in the first and second batch was 100% and 91.67%, respectively, and the positive rate of ORSV in the two batches was 50% and 25%, respectively. Comparing with the first batch, CymMV content of 16 cultivars and ORSV content of 9 cultivars decreased in the second batch, but ORSV content of Phalaenopsis Sogo Yukidian V3 increased significantly, which increased from negative to 6.85×105-8.50×105 copies/μL. The results indicated that CymMV and ORSV widely existed in in vitro seedlings of Phalaenopsis, and the content varied with different batches. The cultivars detected as the negative also faced the risk of virus content increase. Thus, CymMV and ORSV should be investigated regularly in the production of in vitro Phalaenopsis seedlings to ensure the high-quality seedling source for Phalaenopsis industry.

蝴蝶兰(Phalaenopsis spp.)为兰科(Orchidaceae)蝴蝶兰属(Phalaenopsis)附生常绿草本植物,因花型奇特、花色繁多、花期较长,故素有“洋兰王后”之称,是中国销量最大的年宵花,也是世界范围内销量第一的盆栽观赏植物,在世界花卉产业中占有极其重要的地位[1-2]
目前蝴蝶兰生产中广泛应用的繁殖方式主要是种子无菌播种及不定芽途径的组培繁殖方法,其中以不定芽途径的组培繁殖方法应用最为广泛[3]。长期无性繁殖会致使病毒逐代积累,同时,病毒也会通过汁液、蚜虫及组培操作时的器械接触等方式不断传递,使得蝴蝶兰商品苗种和盆栽植株中病毒携带率较高,直接影响蝴蝶兰的观赏价值和商品价值[4]。感染病毒的蝴蝶兰植株,其叶片常出现斑驳、皱缩、凹陷环斑等症状,花器受害后表现出花变叶、花穗小、花期短、花色不均匀等现象,给花卉产业造成严重的经济损失[5]
已知能感染兰花的病毒种类有50多种,其中建兰花叶病毒(Cymbidium mosaic virus,CymMV)和齿兰环斑病毒(Odontoglossum ringspot virus,ORSV)是最常见、感染最严重的2种病毒[16]。蝴蝶兰CymMV感病植株会出现叶片黄色环斑以及花朵褪色等病症,ORSV感病植株则会出现叶片条纹花斑以及花朵褪色等病症,严重影响蝴蝶兰盆花产品的观赏价值和商品价值[4]
组培苗是蝴蝶兰盆花产品的种苗来源,对蝴蝶兰盆花产品质量起到关键的决定性作用。目前蝴蝶兰组培苗生产中,通常以花梗为外植体,经花梗腋芽诱导、丛生芽诱导与增殖、组培苗壮苗及生根培养批量获得蝴蝶兰组培苗[7-8]。目前对于蝴蝶兰组培苗研究主要集中于花梗外植体消毒方式[9]、不定芽增殖[10]、抑制组培褐化[11]等繁育技术。同时,蝴蝶兰CymMV和ORSV研究主要集中于病毒检测体系的构建[4]。在现有研究中,仅见采用ELISA方法对国产蝴蝶兰种苗携带CymMV和ORSV的初步探查[12],尚未见针对蝴蝶兰组培苗携带CymMV和ORSV情况的调查。
本研究以云南地区不同企业20个蝴蝶兰品种的组培苗为对象,从组培生产线上选取继代增殖周期间隔10个月的2个批次组培苗样品,采用RT-PCR和RT-qPCR进行CymMV和ORSV检测,并进行病毒含量变化的分析,旨在初步探明蝴蝶兰组培苗携带CymMV和ORSV的情况,研究结果将对蝴蝶兰组培苗生产中的病毒防控提供参考。
从云南蝴蝶兰生产企业的组培生产线中选取20个蝴蝶兰品种,接种培养基为1/2 MS+3.0 mg/L 6-BA+0.2 mg/L NAA+30 g/L土豆+50 ml/L椰子水,培养条件:温度(25±2)℃,湿度(40±2)%,光照强度20 µmol/(m2·s)(16 h光照/8 h黑暗)。每个品种随机选取3瓶组培苗作为第1批次检测样品(图1)。第1批次检测样品取样后,同批次的蝴蝶兰组培苗继续在企业组培生产线上进行继代扩繁,10个月继代增殖扩繁5次后,再对20个蝴蝶兰品种进行取样,每个品种随机选取3瓶组培苗作为第2批次检测样品。选取长势一致的蝴蝶兰组培苗,取相同部位叶片作为CymMV和ORSV检测材料。
使用艾德莱生物科技有限公司的多糖多酚复杂植物RNA快速提取试剂盒进行蝴蝶兰组培苗叶片总RNA提取,对检测合格的RNA采用PrimeScript Ⅳ 1st strand cDNA SynthesisMix试剂盒(TaKaRa)进行反转录以获得cDNA。
参照宋起萱等[4]所述方法,以水作阴性对照,使用表1引物对蝴蝶兰组培苗分别进行CymMV和ORSV的RT-PCR和RT-qPCR检测。使用EasyPure Quick Gel Extraction试剂盒(全式金生物技术有限公司)对RT-PCR产物目的电泳条带进行切胶回收,回收产物进行测序,测序结果使用DNAMAN软件与CymMV和ORSV的外壳蛋白(coat protein,CP)基因序列进行比对。同时,参考水(对照)的Cq值确定RT-qPCR病毒检测的阳性标准,阴性样品视为病毒含量数值不存在。对检测为阳性的样品计算CymMV和ORSV病毒含量,CymMV病毒含量计算公式:y=–3.459x+43.98,ORSV病毒含量计算公式:y=–3.427x+48.42,其中,y均为RT-qPCR的Cq值,x为模板浓度的对数值(lgC=x)。各品种样品用于病毒检测的cDNA浓度相同。
使用Excel 2021软件计算各品种3个样品重复RT-qPCR中Cq的平均值和病毒含量,并对2个批次的组培苗数据进行比较,若同一个品种2个批次检测结果均为阴性或Cq平均值差异<0.5,则认为病毒含量基本保持不变,若同一个品种2个批次Cq平均值差异≥0.5,则认为病毒含量发生变化。使用GraphPad Prism 10.1.2软件对统计数据进行绘图。
使用RT-PCR对2个批次的蝴蝶兰组培苗样品进行CymMV检测,检测结果显示,在第1批次组培苗中,明日之星、绿闪电、朝日、V3、婚纱5个品种的3个样品重复均为阳性(图2),阳性率为25%。在第2批次组培苗中,明日之星的3个样品重复转为阴性,绿闪电、朝日、V3、婚纱4个品种的3个样品重复仍为阳性,阳性率为20%。对目的电泳条带切胶回收测序后,与CymMV的CP基因序列进行比对,序列相似度为96.62%,说明目的电泳条带确定为CymMV。
同时,使用RT-qPCR再次对2个批次的蝴蝶兰组培苗样品进行CymMV检测,以水(对照)的Cq值为参考,确定Cq≥35为阴性,Cq<35为阳性(表2)。检测结果显示,除第2批次少数样品检测为阴性,2个批次的其他蝴蝶兰样品均不同程度地携带CymMV,说明CymMV在蝴蝶组培苗中广泛存在。第1批次组培苗中,CymMV含量在1.32×103~8.93×1010 copies/μL之间,蓝宝石的病毒含量最低,婚纱的病毒含量最高。第2批次组培苗中,蓝宝石2个样品及宝贝检测为阴性,其他样品CymMV含量在4.36×102~3.87×109 copies/μL之间,病毒含量最高的品种为绿闪电。
使用RT-PCR对2个批次的蝴蝶兰组培苗样品进行ORSV检测,检测结果显示,在第1批次和第2批次组培苗中,明日之星、绿闪电、大财主3个品种的3个样品重复均为阳性(图3),阳性率均为15%。对目的电泳条带切胶回收测序后,与ORSV的CP基因序列进行比对,相似度为96.63%,说明目的电泳条带确定为ORSV。结合CymMV的RT-PCR检测结果可知,在第1批次组培苗中,明日之星和绿闪电复合感染CymMV和ORSV;在第2批次组培苗中,绿闪电复合感染CymMV和ORSV。
同时,使用RT-qPCR再次对2个批次的蝴蝶兰组培苗样品进行ORSV检测,以水(对照)的Cq值为参考,确定Cq≥35为阴性,Cq<35为阳性(表3)。检测结果显示,朝日、金桔、婚纱等9个品种的2个批次样品均检测为阴性,芳美西施、蓝宝石、鑫盛白花等6个品种第1批次样品检测为阳性,第2批次样品检测为阴性,而V3则是第1批次样品检测为阴性,第2批次样品检测为阳性。除检测为阴性的样品外,在第1批次组培苗中,其他样品ORSV含量在1.02×104~5.12×109 copies/μL之间,ORSV含量最高的品种是大财主;在第2批次组培苗中,其他样品的ORSV含量在1.74×104~1.72×109 copies/μL之间,ORSV含量最高的品种是绿闪电(表3)。此外,结合CymMV的RT-qPCR检测结果可知,明日之星、三色鸟、绿闪电、大财主4个蝴蝶兰品种的2个批次组培苗不同程度复合感染CymMV和ORSV(表2表3)。
2个批次蝴蝶兰组培苗样品间隔10个月,中间经历5次继代增殖扩繁,本研究对2个批次蝴蝶兰组培苗中CymMV和ORSV随组培扩繁周期的变化情况进行监测和比较。RT-PCR和RT-qPCR检测结果显示,在2个批次的蝴蝶兰组培苗中,CymMV阳性率均明显高于ORSV,蝴蝶兰组培苗携带CymMV更为广泛(图4A)。在RT-PCR检测中,2个批组培苗次CymMV的阳性率分别为25%和20%,ORSV的RT-PCR阳性率均为15%。而在RT-qPCR检测中,2个批次组培苗CymMV阳性率分别为100%和91.67%,ORSV阳性率分别为50%和25%,RT-qPCR检测的阳性率均高于RT-PCR,说明RT-qPCR检测的灵敏度更高(图4A图4B)。
对2个批次20个品种各3个样品重复的RT-qPCR的Cq值以及病毒含量的平均值进行比较,分析结果显示,相比第1批次,第2批中16个品种CymMV含量减少(图4C),其中婚纱和朝日的CymMV含量减少较为明显,由1010 copies/μL减少至108 copies/μL。9个品种ORSV含量减少,其中明日之星和大财主的ORSV含量减少较为明显,由109 copies/μL减少至107 copies/μL。2个品种的CymMV含量以及10个品种的ORSV含量在2个批次中未发生明显变化。尽管绝大部分蝴蝶兰品种组培苗的病毒含量减少或无明显变化,但在第2批次组培苗中分别有2个品种的CymMV和1个品种的ORSV含量增加(图4C),其中V3的ORSV含量增加明显,由阴性增加至105 copies/μL(表2),说明检测为阴性的品种在继代增殖过程中亦存在病毒量增加的风险。
根据RT-qPCR检测结果对2个批次蝴蝶兰组培苗进行CymMV和ORSV含量分布情况进行分析(图5),分析结果显示,在2个批次组培苗样本中,CymMV含量均在104 copies/μL区段的样本数最多,ORSV含量均在阴性区段(0 copies/μL)的样本数量最多。整体看,ORSV含量低于CymMV含量,组培苗携带CymMV的情况更为严重。此外,与第1批次相比,由于多数品种样本的病毒含量有所减少,使得低一级别的病毒浓度区段的样本数量增加。第2批次中,CymMV含量在105 copies/μL区段的样本数量明显减少,同时在浓度较低的102、103 copies/μL区段的样本数量明显增加;ORSV含量在104 copies/μL区段的样本数量明显减少,同时在阴性区段(0 copies/μL)的样本数量明显增加。
由上述结果可知,在组培生产线上,蝴蝶兰组培苗广泛携带CymMV和ORSV,且携带CymMV病毒含量普遍高于ORSV。同时,绝大多数蝴蝶兰组培苗的CymMV和ORSV含量会随扩繁次数和组培周期发生变化,在生产上有必要定期对病毒含量进行检测,以保证组培苗整体质量。
病毒病严重影响兰花产业的发展,已知能感染兰花的病毒种类有50多种[1],在蝴蝶兰生产栽培中,感染较多的常见病毒为建兰花叶病毒(CymMV)和齿兰环斑病毒(ORSV)[13]。现有的蝴蝶兰CymMV和ORSV检测主要针对植株[414-16]以及植株不同部位[17],检测发现不同品种的蝴蝶兰植株均不同程度感染CymMV和ORSV,蝴蝶兰唇瓣和气生根的CymMV和ORSV含量高于其他部位。对于蝴蝶兰组培苗的检测CymMV和ORSV则相对较少。丁兰等[12]使用双抗夹心酶联免疫吸附法(DAS-ELISA)对蝴蝶兰17个品系22个样品分生组培苗进行检测,发现除1个样品检测为阴性,其余样品均复合感染CymMV和ORSV。与之相似,本研究使用RT-qPCR对蝴蝶兰20个品种每批次60个组培苗样品进行检测,第1批次样品CymMV和ORSV的阳性率分别为100%和50%,明日之星、三色鸟、绿闪电、大财主4个蝴蝶兰品种的2个批次组培苗不同程度复合感染CymMV和ORSV。上述研究结果说明,CymMV和ORSV的复合感染不仅广泛存在于蝴蝶兰植株以及植株的不同部位,也存在于蝴蝶兰组培苗中。同时,携带病毒的蝴蝶兰组培苗可能是CymMV和ORSV在蝴蝶兰产业链中扩散的源头。
目前,针对蝴蝶兰建兰花叶病毒和齿兰环斑病毒已构建了包括联免疫法(ELISA)、RT-PCR以及RT-qPCR等多种检测方法,但不同方法的灵敏性存在差异[18]。宋起萱等[4]同时采用RT-PCR和RT-qPCR对4个品种的20个样本进行病毒检测,RT-qPCR检测的CymMV和ORSV阳性率分别为100%和90%,显著高于RT-PCR阳性率。本研究亦同时采用RT-PCR和RT-qPCR对2个批次的蝴蝶兰组培苗进行CymMV和ORSV检测,2个批次CymMV的RT-PCR阳性率分别为25%和20%,ORSV的RT-PCR阳性率均为15%,而在RT-qPCR检测中,2个批次CymMV阳性率分别为100%和91.67%,ORSV阳性率分别为50%和25%,RT-qPCR检测的阳性率均高于RT-PCR,说明RT-qPCR检测的灵敏度高于RT-PCR。
目前蝴蝶兰组培苗生产中,通常以花梗为外植体,经花梗腋芽诱导、丛生芽诱导与增殖、组培苗壮苗及生根培养批量获得蝴蝶兰组培苗[7-8]。为了获得大量的蝴蝶兰组培苗,需要经过数轮继代增殖扩繁[19]。本研究对组培生产线上相隔10个月的2个批次的蝴蝶兰组培苗进行CymMV和ORSV检测,RT-qPCR分析结果显示,相比第1批次,第2批次16个品种CymMV含量减少,9个品种ORSV含量减少。有研究表明,病毒浓度呈梯度变化,病毒粒子随植物组织的成熟而增加,旺盛生长的根尖、茎尖很少有病毒分布[20-21]。因此,对茎尖分生组织进行培养有望获得脱毒组培苗[22-23]。蝴蝶兰组培繁殖过程中,丛生芽诱导与增殖时,需要对蝴蝶兰除去芽体的叶片及基部组织,以促进丛生芽的快速诱导和生长[823],该操作虽不如茎尖培养能获得脱毒种苗,但随着丛生芽的快速生长,可能在一定程度上能降低病毒携带,这可能是本研究第2批次中大部分品种病毒含量下降的主要原因。
尽管大部分品种第2批次的病毒含量低于第1批次,但少数品种的病毒含量在第2批次明显增加,如蝴蝶兰V3 2个批次组培苗ORSV的RT-PCR检测均为阴性,第1批次组培苗ORSV的RT-qPCR检测为阴性,而第2批次ORSV病毒含量则上升至8.50×105 copies/μL,这可能与品种生长特性和生产线上潜在的交互感染存在一定的关系[24],应定期对生产线的组培苗进行病毒监测。
蝴蝶兰植株感染CymMV和ORSV后,植株和花朵会出现相应病症,直接影响观赏价值[4]。与之不同,本研究中蝴蝶兰婚纱组培苗的CymMV病毒含量高达8.93×1010 copies/μL,大财主组培苗的ORSV病毒含量可达5.12×109 copies/μL,但这2个品种的组培苗形态正常,未表现出病症。由此可知,在组培生产线上较难通过组培苗形态来判断是否携带病毒,需定期对蝴蝶兰组培苗进行抽样检测。
