10 inflorescence traits of 200 domestic and international Macadamia germplasms in the Jinghong Macadamia Germplasm Repository of the Ministry of Agriculture and Rural Affairs were evaluated using QGA Station combined with different genetic distance metrics, clustering methods, sampling strategies, and sampling proportions to conduct a core macadamia collection construction. The optimal methodology for core collection development was identified as “Mahalanobis distance+multi-clustering deviation sampling+centroid clustering method+15% sampling proportion”, resulting in a core collection comprising 30 accessions. The mean difference percentage, variance difference percentage, range coincidence rate and variation coefficient change of core collection and original collection was 0.00%, 90.00%, 99.31% and 150.08%, respectively. No significant differences in trait means were detected between the core and original collections (T-test). The variances of all traits in the core collection exceeded those of the original collection, with 9 traits showing statistically significant differences. While genetic diversity indices slightly decreased, the conformity rate for all traits remained above 80%. Principal component analysis (PCA) demonstrated that the cumulative variance contribution rate of the first three principal components was 74.118% and 84.350% for the original and core collections, respectively, with congruent geometric structures in their PCA scatterplots. Correlation analysis further confirmed that trait relationships in the core collection closely mirrored those of the original collection. The core collection comprised 8 Australian accessions, 10 Hawaiian accessions, 9 China Yunnan accessions, and 3 accessions of special types, exhibiting a degree of geographic representativeness. Analyses using both cluster analysis and PCA on the core collection revealed that domestic and foreign accessions were distinguishable to a certain extent based on the inflorescence phenotypes. The Macadamia core collection developed in this study, based on inflorescence phenotypic traits, effectively represents the genetic diversity of the original collection and would provide a robust foundation for future research on Macadamia genetic resources.

以农业农村部景洪澳洲坚果种质资源圃内200份国内外澳洲坚果种质的10个花序性状,使用QGA Station软件组合筛选不同遗传距离、聚类方法、取样策略和取样比例,进行核心种质构建探究。澳洲坚果基于花序表型的核心种质最佳构建方法为“马氏距离+多次聚类偏离度取样法+重心法聚类+15%取样比例”,构建了包含30份资源的核心种质。核心种质和原始种质的均值差异百分率、方差差异百分率、极差符合率和变异系数变化率分别为0.00%、90%、99.31%和150.08%。核心种质与原始种质各性状均值T检验无显著差异,方差均大于原始种质,其中9个性状的方差达到显著水平;遗传多样性指数较原始种质有所减小,但符合率均大于80%。主成分分析显示,原始种质和核心种质前3个主成分累计方差贡献率分别为74.118%和84.350%,核心种质的主成分得分图几何结构与原始种质一致。相关性分析显示核心种质各性状相关关系与原始种质较为一致。核心种质由8份澳大利亚种质、10份夏威夷种质、9份中国云南种质和3份特殊种质构成,存在一定地域性。核心种质的聚类和主成分分析显示,国内和国外种质可依据花序表型进行一定程度的区分。本研究基于花序表型构建的澳洲坚果核心种质能较好代表原始种质,可为澳洲坚果种质资源研究提供依据。

以农业农村部景洪澳洲坚果种质资源圃内200份国内外澳洲坚果种质的10个花序性状,使用QGA Station软件组合筛选不同遗传距离、聚类方法、取样策略和取样比例,进行核心种质构建探究。澳洲坚果基于花序表型的核心种质最佳构建方法为“马氏距离+多次聚类偏离度取样法+重心法聚类+15%取样比例”,构建了包含30份资源的核心种质。核心种质和原始种质的均值差异百分率、方差差异百分率、极差符合率和变异系数变化率分别为0.00%、90%、99.31%和150.08%。核心种质与原始种质各性状均值T检验无显著差异,方差均大于原始种质,其中9个性状的方差达到显著水平;遗传多样性指数较原始种质有所减小,但符合率均大于80%。主成分分析显示,原始种质和核心种质前3个主成分累计方差贡献率分别为74.118%和84.350%,核心种质的主成分得分图几何结构与原始种质一致。相关性分析显示核心种质各性状相关关系与原始种质较为一致。核心种质由8份澳大利亚种质、10份夏威夷种质、9份中国云南种质和3份特殊种质构成,存在一定地域性。核心种质的聚类和主成分分析显示,国内和国外种质可依据花序表型进行一定程度的区分。本研究基于花序表型构建的澳洲坚果核心种质能较好代表原始种质,可为澳洲坚果种质资源研究提供依据。

10 inflorescence traits of 200 domestic and international Macadamia germplasms in the Jinghong Macadamia Germplasm Repository of the Ministry of Agriculture and Rural Affairs were evaluated using QGA Station combined with different genetic distance metrics, clustering methods, sampling strategies, and sampling proportions to conduct a core macadamia collection construction. The optimal methodology for core collection development was identified as “Mahalanobis distance+multi-clustering deviation sampling+centroid clustering method+15% sampling proportion”, resulting in a core collection comprising 30 accessions. The mean difference percentage, variance difference percentage, range coincidence rate and variation coefficient change of core collection and original collection was 0.00%, 90.00%, 99.31% and 150.08%, respectively. No significant differences in trait means were detected between the core and original collections (T-test). The variances of all traits in the core collection exceeded those of the original collection, with 9 traits showing statistically significant differences. While genetic diversity indices slightly decreased, the conformity rate for all traits remained above 80%. Principal component analysis (PCA) demonstrated that the cumulative variance contribution rate of the first three principal components was 74.118% and 84.350% for the original and core collections, respectively, with congruent geometric structures in their PCA scatterplots. Correlation analysis further confirmed that trait relationships in the core collection closely mirrored those of the original collection. The core collection comprised 8 Australian accessions, 10 Hawaiian accessions, 9 China Yunnan accessions, and 3 accessions of special types, exhibiting a degree of geographic representativeness. Analyses using both cluster analysis and PCA on the core collection revealed that domestic and foreign accessions were distinguishable to a certain extent based on the inflorescence phenotypes. The Macadamia core collection developed in this study, based on inflorescence phenotypic traits, effectively represents the genetic diversity of the original collection and would provide a robust foundation for future research on Macadamia genetic resources.

澳洲坚果(Macadamia spp.)属山龙眼科(Proteaceae)澳洲坚果属(Macadamia)常绿乔木果树,又称夏威夷果,食用部分为果仁,富含不饱和脂肪酸、蛋白质和多种矿质元素及维生素[1],营养价值丰富。中国为澳洲坚果种植大国,截至2023年,我国澳洲坚果种植面积达35.94万hm2,占全球澳洲坚果种植面积的61.85%[2]。丰富的种质资源是育种的基础[3],目前澳洲坚果种质资源研究多集中于形态特征鉴定评价[4-7]及亲缘关系鉴定[8-10]等方面,尚无基于表型性状构建澳洲坚果核心种质的报道。
核心种质指整个种质资源群体中,能够以最小的资源数量和遗传重复保存整个群体遗传多样性的一小部分资源,该概念由FRANKEL[11]提出并由BROWN[12]进一步完善。核心种质的个体数量少,有利于种质资源的保存利用,并且构成核心种质的个体特异性强,更易找到群体内所需的特殊资源,能有效提高育种效率[13]。核心种质可基于作物的表型性状及分子标记等进行构建[14],其中表型数据直观且容易获得,是种质整体遗传多样性的重要组成部分[15]。目前核桃[16]、芝麻[17]、花生[18]和大豆[19]等油料作物已基于表型构建了有效核心种质,能较大程度保留原始群体遗传多样性。在花器官表型中,花的大小往往与传粉者造访传粉的概率密切相关[20],澳洲坚果存在自交不亲性,自然授粉条件下座果率为0.1%~0.3%[21],生产中需由虫媒传粉以保证产量[22]。不同澳洲坚果品种的小花数量和结实率存在显著差异[23],花序表型可能与座果率存在一定相关性。进行花序表型性状测定,并构建核心种质,对依据花序性状快速筛选种质具有重要意义。
为探究澳洲坚果核心种质构建策略,本研究使用200份澳洲坚果种质的花序数量性状,通过不同聚类距离、取样策略及取样比例,筛选构建澳洲坚果核心种质的最佳方法,并构建澳洲坚果基于花序表型的核心种质库,以期为澳洲坚果种质资源研究提供参考。
以保存于农业农村部景洪澳洲坚果种质资源圃(22°00′50.40″N,100°46′42.24″E,海拔550 m)中的200份澳洲坚果种质为试验对象,试验分别于2022、2023年的3—4月在资源圃进行。试验地点属于北热带湿润季风气候,最高温为38.4 ℃,最低温为10.0 ℃,年均温为21.9 ℃,全年无霜,年降雨量为1196 mm,年日照时数为2428.1 h,海拔为550 m。所有试验材料的种植及管理条件一致。按照种质来源,将200份种质分为国外育成品种及实生选育优株(国外育成及实选),国内育成品种及实生选育优株(国内育成及实选),果壳粗糙及植株形态与其他种质有异的特殊种质,种质分类见表1
每份种质随机调查3株,选择树冠内膛不同部位至少共10个花序进行观测。参考《澳洲坚果种质资源描述规范和数据标准》[24]测定花序梗长。花序梗柄长、花序梗粗度、花序宽度、小花梗长度、小花长度、小花粗度、花冠径、雌蕊长度及花序密集度测量方法见表2。数量性状的等距离分级参考王业社等[25]的方法(表3)。
使用QGA Station软件抽取核心种质。先在25%取样比例下,选择欧氏距离和马氏距离,对多次聚类随机取样法、优先取样法和偏离度取样法3种取样策略,以及最短距离法、最长距离法、中间距离法、重心法、非加权类平均法、可变类平均法及离差平方和法7种聚类方法进行组合对比,筛选较优的聚类取样方法组合,再以5%、10%、15%、20%、25%、30%、40%和50%的取样比例进行取样,筛选最佳取样比例,最终基于核心种质评价参数、核心种质与原始种质各性状TF检验,选出基于花序表型构建澳洲坚果核心种质的最佳策略[26]
核心种质评价参数为均值差异百分率(mean difference percentage,MD)、方差差异百分率(variance difference percentage,VD)、极差符合率(coincidence rate of range,CR)和变异系数变化率(changeable rate of coefficient of variation,VR),参考HU等[27]的方法进行计算。核心种质需满足MD<20%且CR>80%,并且MD越小,VD、CR和VR越大,核心种质越具代表性[27]。使用主成分分析评价核心种质对原始种质遗传变异的保留程度,主成分得分图可视化核心种质在总群体的分布情况。相关性分析判断核心种质性状相关关系保留情况。
使用公式H′=–∑PilnPi计算Shannon-Weaver多样性指数(H′),公式中Pi为某性状第i个代码值的分布频率。使用Excel软件对数据进行处理,用SPSS 26.0软件进行T检验和F检验,用OriginPro 2024软件进行相关性分析、主成分分析及绘图。
在25%取样比例下,使用2种遗传距离,3种取样策略及7种聚类方法组合进行核心种质抽取。结果显示(表4表5),所有取样方法的MD均为0,CR均大于80%,VR均大于100%,所有组合均符合核心种质选择条件。根据VD、CR及VR越大越具代表性的原则对不同组合方法筛选,发现在多次聚类偏离度取样策略下,出现VD和VR的最大值,马氏距离较欧氏距离的CR更大。在马氏距离+多次聚类偏离度取样策略下,最短距离法的VD最高,重心法的CR及VR最高,二者各参数值较为接近,故选出马氏距离+多次聚类偏离度取样策略+最短距离法及重心法2种取样方法,进行下一步取样比例筛选。
以马氏距离为遗传距离,使用最短距离法及重心法进行聚类,取样策略为多次聚类偏离度取样,选择5%、10%、15%、20%、25%、30%、40%和50%共8个取样比例进行核心种质抽取。同样以MD越小,VD、CR及VR越大越具代表性的原则进行筛选,并尽可能平衡各参数水平。结果显示(表6),最短距离法以25%取样比例取样,VD最大,CR>99%,VR为131.33%;重心法选择以15%取样比例取样,VD最大,CR>99%,VR为150.08%。2种方法在5%取样比例下MD已等于0,推测原始种质存在一定遗传冗余,故以核心种质取样比例偏小为佳,最终选择马氏距离+重心法+多次聚类偏离度取样+ 15%取样比例构建核心种质。构成的核心种质包含全部特殊种质、国外和国内育成及实选的部分种质,占原始种质比例分别为100%、20.69%和7.50%。
使用T检验比较核心种质与原始种质性状均值是否存在显著差异,F检验比较性状方差同质性。结果显示(表7),核心种质中10个花序性状与原始种质均值无显著差异;所有性状方差均大于原始种质,其中9个性状的方差显著大于原始种质;多样性指数较原始种质有所减小,但符合率均大于80%,原始种质性状的遗传多样性得到较好保留。
分别对原始种质和核心种质进行主成分分析,结果显示,原始种质前3个主成分累计方差贡献率为74.118%,而核心种质的为84.350%(表8),核心种质累计方差贡献率较原始种质更高,有效减少了遗传冗余度。使用群体内各样本的主成分得分绘制得分图,观察核心种质在原始种质的分布情况(图1),核心种质在几何平面上的分布与原始种质类似,种质重叠分布的情况较少(图1A和图1B);使用随机数从原始种质中抽取样本数为30的随机群体,分析显示随机群体分布在原始种质内部,置信椭圆未包含全部种质,而核心种质的置信椭圆包含全部种质(图1C、图1D),表明核心种质较好地保留了原始种质的遗传结构及遗传多样性。
分别对原始种质、核心种质和随机群体的性状进行相关性分析,结果显示(图2),与随机群体相比,核心种质性状间相关性与原始种质更为相似,并保留了相关系数的大小趋势,仅花序梗柄长与花序梗粗度和小花梗长度,花序梗粗度与花序宽度和雌蕊长度,花冠径与小花梗长度和花序密集度的相关显著性未在核心种质中检测到,可能是原始种质中其相关系数较小所致。
构建的核心种质包含了来自澳大利亚、夏威夷、中国云南和特殊种质,占原始种质的比例分别为25.00%、18.52%、8.49%和100.00%(表9),但未包含以色列、中国广东和中国广西种质。使用花序表型对核心种质进行聚类分析,结果显示,30份种质分为3类(图3),Ⅰ类群仅有72号特殊种质,其特征为花序密集度最大;Ⅱ类群包含13份种质,除97号为中国云南种质外,其余均为澳大利亚和夏威夷种质;Ⅲ类群共16份种质,8份中国云南种质及2份粗壳种质均被包含其中,其余分别为3份澳大利亚种质及3份夏威夷种质。
使用聚类分组信息进行主成分得分图绘制,结果显示Ⅲ类群整体主成分1(principal component 1,PC1)和PC2得分较高(图4A),结合载荷图(图4B)和聚类热图(图3)可知,Ⅲ类群除花序密集度外,其他花序性状整体优于Ⅱ类群。基于核心种质的分析显示,可在一定程度上依据花序表型性状区分澳洲坚果国内和国外种质。
核心种质是以最小资源数量和遗传重复,代表整个种质群体的遗传多样性的核心子集。核心种质的构建多基于聚类取样进行,构建时需对聚类遗传距离、聚类方法、聚类取样方法及取样比例进行筛选[14],最终获得具代表性、遗传异质性和多样性的核心种质[28]。对使用不同策略构建的核心种质,以均值差异百分率、方差差异百分率、极差符合率和变异系数变化率对其进行遗传差异保留程度进行评价[27]。本研究对2种遗传距离、7种聚类方法和不同比例的取样策略组合比较,最终确定马氏距离、重心法聚类、多次聚类偏离度取样和15%取样比例组合为澳洲坚果花序表型核心种质库构建的最佳策略,其MD、VD、CR和VR分别为0%、90%、99.31%和150.08%,均值T检验与原始种质无显著差异,所有性状遗传多样性指数符合率均大于80%,符合核心种质的参数要求。
对核心种质库的检验是评价其是否具有代表性和实用性的重要步骤,主成分分析可以比较核心种质与原始种质的空间分布特征,以此判断核心种质对原始种质遗传结构的保留情况[29]。李洪果等[30]的研究结果显示,蒜头果核心种质前6个主成分累计方差贡献率为87.78%,大于原种质的86.64%,核心种质主成分得分图中密集和重叠部分明显减少,保留了遗传结构,降低了遗传冗余;张磊等[31]构建了糜子的骨干种质,其主成分得分图与原种质特征高度相似,并且无重叠现象;曾钦朦等[16]的研究显示,贵州核桃核心种质的置信椭圆包含了整个原种质,有效保留了原种质的遗传信息。在本研究中,核心种质的前3个主成分累计贡献率大于原始种质,主成分得分图在几何平面上的分布与原始种质一致,置信椭圆包含了全部种质,较好地保留了原始种质的遗传多样性和遗传结构。此外,本研究中核心种质的性状相关性与原始种质高度相似,保留了原群体中的相关关系,进一步验证了该核心种质的代表性。
使用不同地域来源分布的种质构建核心种质时,通常按地域进行分组,在组内进行聚类和取样[32]。前人基于澳洲坚果花序表型的聚类分析显示,不同地域来源种质在各类群中均有分布,并未表现出地域性[4,33],故本研究未使用地域分组进行核心种质抽取,但本研究的核心种质由澳大利亚、夏威夷、中国云南和特殊种质构成,存在一定地域性。对核心种质的聚类和主成分分析显示,8份国内种质聚在Ⅲ类群,而12份国外种质聚在Ⅱ类群,Ⅲ类群除花序密集度外整体花序表型优于Ⅱ类群,国内和国外种质在花序表型上存在一定差异。前人的聚类分析未显示地域性的原因可能是参试种质存在遗传冗余[34],而核心种质是以最少样本量最大限度代表原始群体遗传多样性的核心子集[13],去除了遗传冗余,凸显出不同种质的关键特征,进而核心种质内表现出地域性。花序表型的差异分析显示(未公开发表),国内种质在花冠径和小花长度上较国外种质更大,而国内种质中有98份为实选优株,多数因田间果实较大被选育,推测小花大小与果实大小可能存在一定相关性,需进一步研究以明确花序表型的育种价值。
综上,本研究基于200份澳洲坚果种质的花序表型,使用马氏距离、重心法聚类和偏离度取样法,以15%取样比例,构建了含30份资源的核心种质。经评价参数、主成分分析及相关性分析验证,该核心种质具代表性和多样性,并体现了花序表型的地域性,能有效反映澳洲坚果花序表型的整体特征。
本研究为基于表型构建澳洲坚果核心种质的首次尝试,其他表型性状如叶片和果实正陆续收集中。后续研究需精进试验设计,扩大性状选择范围,整合表型和基因型数据,并结合分子标记进一步完善澳洲坚果核心种质库,为澳洲坚果种质资源保护、开发利用及品种选育提供理论依据。
