The objective of this study is to elucidate the floral characteristics and flowering dynamics of Bougainvillea spp., investigate the variations in pollen viability, stigma acceptability, and cross-compatibility among different varieties, and provide theoretical and technical guidance for the crossbreeding in Bougainvillea spp.. Nine varieties of Bougainvillea were chosen to examine the floral traits and flowering patterns, also to assess pollen viability and stigma acceptability. Suitable parents were selected for cross-pollination, and the germination of pollen tubes along with the seed-setting rate of hybridization were observed. The pollen viability of the six male varieties, measured by the ex vivo germination method was high. Among which, three varieties exhibited high pollen viability and could be used as the male parents. The results from the stigma detection of three varieties by benzidine-hydrogen peroxide method showed that the stigma had the strongest deliverability in the V. period, it was concluded that the low seed-setting rate was due to the substantial amount of callose produced in the stigma and the columella following pollination, which distorted and deformed the pollen tube, resulting in only a limited number of pollen tubes reaching the ovule to complete fertilization. Additionally, significant differences were observed in the seed-setting rates among various hybrid combinations. Incompatibility was noted across various cross combinations, predominantly attributed to pre-fertilization disorders.

本研究旨在揭示三角梅花部特征和开花动态,探究花粉活力、柱头可授性的变化和不同品种间的杂交亲和性,为三角梅杂交育种提供理论和技术指导。以9个三角梅品种为研究对象,观测其花部特征和开花动态,检测花粉活力及柱头可授性;筛选出适合的亲本进行杂交授粉,观测花粉管萌发情况和杂交结实率。采用离体萌发法测定的6个父本品种中有3个品种花粉活力较高,可以作为父本。采用联苯胺-过氧化氢法检测3个母本的柱头可授性,结果均显示在第Ⅴ时期的柱头可授性最强。通过对结实率最低的2个杂交组合授粉后的花粉管生长情况进行荧光显微镜观察,发现导致结实率低的原因是授粉后柱头和花柱内产生大量胼胝质并持续堆积,使花粉管发生扭曲变形,从而导致只有少量花粉管伸入胚珠完成受精,不同杂交组合之间结实率差异较大。不同杂交组合均存在不同程度不亲和性,受精前障碍是三角梅杂交不亲和的主要原因。

本研究旨在揭示三角梅花部特征和开花动态,探究花粉活力、柱头可授性的变化和不同品种间的杂交亲和性,为三角梅杂交育种提供理论和技术指导。以9个三角梅品种为研究对象,观测其花部特征和开花动态,检测花粉活力及柱头可授性;筛选出适合的亲本进行杂交授粉,观测花粉管萌发情况和杂交结实率。采用离体萌发法测定的6个父本品种中有3个品种花粉活力较高,可以作为父本。采用联苯胺-过氧化氢法检测3个母本的柱头可授性,结果均显示在第Ⅴ时期的柱头可授性最强。通过对结实率最低的2个杂交组合授粉后的花粉管生长情况进行荧光显微镜观察,发现导致结实率低的原因是授粉后柱头和花柱内产生大量胼胝质并持续堆积,使花粉管发生扭曲变形,从而导致只有少量花粉管伸入胚珠完成受精,不同杂交组合之间结实率差异较大。不同杂交组合均存在不同程度不亲和性,受精前障碍是三角梅杂交不亲和的主要原因。

The objective of this study is to elucidate the floral characteristics and flowering dynamics of Bougainvillea spp., investigate the variations in pollen viability, stigma acceptability, and cross-compatibility among different varieties, and provide theoretical and technical guidance for the crossbreeding in Bougainvillea spp.. Nine varieties of Bougainvillea were chosen to examine the floral traits and flowering patterns, also to assess pollen viability and stigma acceptability. Suitable parents were selected for cross-pollination, and the germination of pollen tubes along with the seed-setting rate of hybridization were observed. The pollen viability of the six male varieties, measured by the ex vivo germination method was high. Among which, three varieties exhibited high pollen viability and could be used as the male parents. The results from the stigma detection of three varieties by benzidine-hydrogen peroxide method showed that the stigma had the strongest deliverability in the V. period, it was concluded that the low seed-setting rate was due to the substantial amount of callose produced in the stigma and the columella following pollination, which distorted and deformed the pollen tube, resulting in only a limited number of pollen tubes reaching the ovule to complete fertilization. Additionally, significant differences were observed in the seed-setting rates among various hybrid combinations. Incompatibility was noted across various cross combinations, predominantly attributed to pre-fertilization disorders.

三角梅又称叶子花、三角花、九重葛、勒杜鹃、宝巾等,为紫茉莉科(Nyctaginacea)叶子花属(Bougainvillea)灌木状攀援植物,原产于南美洲,栽培种常见于热带及亚热带地区。三角梅苞片色彩鲜艳、花量大、花期长,具有极高的观赏价值,深受人们的喜爱,被广泛应用于绿化景观或盆栽。同时三角梅在净化空气以及食用、医学、生物农药、纺织染料、高分子燃料合成、天然食品保鲜剂等方面具有多种价值[1-3]。三角梅原种、栽培种、变种、杂交种多达400~500个[4],大部分均引入中国种植。但目前的品种仍然多样化不足,需要培育更多的品种满足消费者的需求。杂交是培育三角梅新品种的重要途径[5],然而三角梅杂交育种存在育性差、杂交困难等难题,因此需要对其花部特征和杂交授粉进行深入研究。了解植物的花部特征是开展杂交育种的前提。国内外关于三角梅繁殖及栽培等方面的研究均有大量报道[6-11],但有关开花生物学方面研究仍显不足。本研究将对三角梅的花部特征以及开花动态进行观测,并测定不同品种的三角梅花粉活力和柱头可授性,在此基础上开展杂交授粉研究,观察花粉管的伸长,统计结实率,然后进行杂种苗培育,旨在探究三角梅杂交的技术体系,为三角梅杂交育种提供借鉴和理论依据。
试验材料为三角梅品种粉红豹(B. spectabilis, Pink Panther)、新加坡宫粉(B. glabra, Singapore Pink)、金边叶浅紫(B. glabra, Golden Dragon)、暗斑叶丽娜(Bougainvillea sp.)、斑叶光叶紫(B. glabr a, Sanderiana Variegata)、安格斯(B. glabra, Elizabeth Angus)、白雪公主(B. glabra, Mrs. Eva White)、浅茄(B. glabra, Mariel Fitz Patrick)、苹果花(B. ougainvillea, Apple Blossom),该9个品种均为嫁接苗,接穗生长5 a左右,栽培于云南农业大学园林园艺学院苗圃中(25°8′13″N,102°45′14″E)。
于2023年4月—2024年4月,昆明年平均气温为17.9 ℃,年平均相对湿度为70%左右,观测粉红豹、浅茄、新加坡宫粉、白雪公主、苹果花和安格斯等6个三角梅品种的花朵结构特征和开花动态,从三角梅花蕾期开始观察苞片的生长、花被管的伸长情况、柱头的形态变化和花被管开裂状态以及花被管的解剖特征,记录其开始生长到完全展开,再到扭曲变形的整个开花动态过程。
选择开放1~2 d的粉红豹、浅茄、新加坡宫粉、金边叶浅紫、暗斑叶丽娜和斑叶光叶紫6个品种,于9:00—11:00采集花粉,采集后立即带回实验室,采用离体萌发法测定活力。以150 g/L蔗糖、50 mg/L CaCl2和150 mg/L H3BO3为基本培养基,采取单因素试验设计,试验在90 mm玻璃培养皿中进行,通过添加HCl和NaOH调节培养基的pH为6±0.1。参照张伟等[12]测试三角梅花粉活力的培养基配方:200 g/L蔗糖+150 mg/L硼酸+30 mg/L CaCl2,琼脂20 g/L(pH 6),将花粉用毛刷均匀地撒在培养基上,(25±0.3)℃条件下恒温培养箱培养4 h,用解剖镜(LEICA DMC 4500)镜检,随机选取6个视野,每个视野不少于40个花粉粒,统计花粉管伸长的花粉数,计算萌发率。每个品种设4个重复。
用联苯胺-过氧化氢法测定白雪公主、苹果花和浅茄的柱头可授性。于每天上午10:00分别采集微开、半开、全开的母本柱头,每个阶段采集5个柱头,在载玻片凹液面滴入联苯胺-过氧化氢溶液(0.5%联苯胺溶液∶3%过氧化氢溶液∶水=4∶11∶22,体积比),将采集的柱头侵入凹液面溶液里面,染色10 min后在显微镜下观察并拍照记录。若柱头周围反应液呈现蓝色并伴有大量气泡出现,则表示柱头具可授性,气泡越多,可授性越强。柱头周围均匀产生大量气泡并完全包裹住柱头,表示柱头可授性很强;柱头周围产生气泡包住柱头长度的1/3~2/3,表示柱头可授性较强;柱头周围产生气泡包裹柱头的部分小于柱头长度的1/3,表示柱头可授性较弱。
根据花粉活力测定结果,选择花粉活力高的三角梅品种粉红豹、新加坡宫粉作为父本,同时以花粉活力低的三角梅品种安格斯作为对照。选择花朵不易脱落、柱头可授性强的三角梅品种白雪公主、苹果花和浅茄为母本。于早上9:00—11:00,中午13:00—15:00授粉,选择开放当天或开放1 d后的花,先用手术刀沿着花被管的任意一个侧棱,从基部由下向上剖开,取父本花粉均匀涂抹在母本柱头上,直至看到柱头有淡黄色花粉。每个花序选择2朵授粉,去除多余的花朵。授粉后在小花枝上挂上吊牌,注明时间、杂交组合和数量。
分别采集白雪公主×安格斯和白雪公主×粉红豹授粉后4、12、24、48 h的雌蕊,用FAA固定液固定,于4 ℃冰箱中保存备用。用4 mol/L NaOH溶液浸泡,在60 ℃烘箱内软化2 h,蒸馏水冲洗至无色,将材料置于0.1%水溶性苯胺蓝溶液染色48 h后,将材料放置于载玻片中央,并滴加1滴甘油,盖上盖玻片,在荧光显微镜下观察花粉管在花柱中的生长情况并拍照。
授粉15~25 d后观察子房发育情况并记录膨大的果实数量,计算结实率。当花被管枯黄开裂时,种子已成熟,此时及时收集杂交种子,放置于阴凉干燥处备用,结实率=采收种子数/杂交花朵数×100%。
选择发育正常的种子,用5%的高锰酸钾浸泡5 min,再用流水冲洗干净,进行常规播种(基质配比为草炭土∶椰糠∶珍珠岩=7∶2∶1)。暗处理7 d后放到阴凉的苗床上,播种15 d后统计出苗率和生长情况,出苗率=出苗数/播种数×100%。
使用Excel软件进行数据统计及绘图,所有数据为均值±标准差,使用Adobe Photoshop CC 2019软件处理图片。
三角梅是雌雄同体的两性花,花被管细长,中部溢缩,几乎所有雄蕊皆高于雌蕊,雌蕊和雄蕊藏于狭长的花被管中,给人工授粉造成较大困难。三角梅为聚伞花序,每个花序中3个花被管呈不同步开放。如图1所示,从下往上三角梅的整体结构为雌蕊包括子房以及里面包裹的胚珠,往上为着生在子房上面的柱头和附生在柱头上接收花药的柱头毛,往上是从子房基部着生由花丝连接的花药是雄蕊,最后由花被把雌雄蕊包裹起来形成整个花被管,不同品种间花被管颜色有差异。通过观测粉红豹、浅茄、新加坡宫粉、白雪公主、苹果花和安格斯等6个三角梅品种,其单朵的开花动态大致划分为7个时期(图2):幼蕾期(Ⅰ),苞片和花被管不断伸长,雌蕊和雄蕊初具形态;花冠准备绽开期(Ⅱ),苞片和花被管不再伸长,雌蕊和雄蕊继续发育;花冠开裂期(Ⅲ),雌雄蕊发育完成,柱头毛发育完成,花药囊继续膨大;花冠绽开初期(Ⅳ),此时雌蕊柱头不再发育,花药囊成熟开裂开始出现散粉;花冠完全绽开期(Ⅴ),雄蕊花药囊完全成熟开裂,持续散粉达到高峰;花冠闭合期(Ⅵ),雌蕊雄蕊开始变形收缩,花药散粉急剧下降,花瓣开始向内收拢;花冠扭曲变形期(Ⅶ),花被管从中间扭曲变形。三角梅从花蕾期到花被管不再伸长约为10~15 d,花冠开裂到绽开初期约为4 d,绽开初期到完全展开约为2 d,花冠完全展开到开始闭合约为4~12 d,花冠闭合到扭曲变形约为1周,所以开花期约为2周,遇到阴雨天开花时间会有延长,如环境导致温度有较大变化,开花进程会有所提前或推迟。
通过花粉离体萌发法得出6个三角梅品种的花粉活力,具体数据见表1。花粉活力最高的是粉红豹(43.03%),其次是新加坡宫粉(29.13%);安格斯、金边叶浅紫、暗斑叶丽娜和斑叶光叶紫的花粉活力均较低,分别为9.21%、7.07%、5.75%、3.53%。因此以花粉活力较高的2个品种(粉红豹和新加坡宫粉)和花粉活力较低的品种安格斯作为杂交父本。
图3可知,白雪公主、苹果花和浅茄的柱头可授性在第Ⅲ时期时很微弱,第Ⅳ时期逐渐增强,第Ⅴ时期达最强,所以第Ⅴ时期为授粉最佳时期。通过对比白雪公主、苹果花和浅茄的柱头变化可知,白雪公主、苹果花的柱头明显比浅茄的长,而随着时间推移以及柱头可授性的增强,浅茄的柱头保持直挺,而白雪公主和苹果花的柱头发生一定程度的弯曲,在第Ⅴ期白雪公主的气泡少于苹果花和浅茄。因此推测白雪公主的柱头可授性弱于苹果花和浅茄。
通过观察白雪公主与安格斯杂交授粉后的花粉管生长情况发现,安格斯作为父本授粉4 h后,几乎不萌发;授粉12 h后,只有少量花粉萌发,在柱头表面开始出现胼胝质现象,影响花粉管伸长,随后花粉管穿过绒毛进入中央花柱中,在花柱中出现了胼胝质塞(图4A);24 h后,花粉管到达柱头底部(图4B);由于胚珠附近沉积大量的胼胝质,花粉管未达到胚珠停止生长,未完成受精(图4C)。同时,本研究发现,安格斯在母本柱头存在更多未萌发现象(图4D)。
通过观察白雪公主与粉红豹杂交授粉后的花粉管生长情况发现,粉红豹作为父本授粉4 h,少量花粉萌发;授粉12 h后,大量花粉开始萌发,花粉管开始伸入花柱内,但在花粉萌发的绒毛上以及花柱内出现了大量的胼胝质塞,使得部分花粉管发生扭曲,偏离了花柱通道(图5A);授粉24 h后,随着花粉管继续伸长到达花柱基部,大量胼胝质持续沉积,部分花粉管尖端出现膨胀(图5B);48 h后,胚珠周围沉积了大量胼胝质,少量花粉管伸入胚珠完成受精(图5C),呈现大量花粉管完全到达不了胚珠的现象(图5D)。
表2可知,在所有杂交组合中,苹果花×新加坡宫粉结实率最高,为16.03%,其次是苹果花×粉红豹(16.00%)、浅茄×新加坡宫粉(14.43%)、浅茄×粉红豹(13.45%)、苹果花×安格斯(9.70%)、白雪公主×新加坡宫粉(6.85%)、白雪公主×粉红豹(4.34%);白雪公主×安格斯结实率最低,仅为1.37%。杂交结实种子图片见图6
选取结实多的粉红豹杂交组合进行常规播种,由表3可知,不同杂交组合的成苗率具有明显差异。其中浅茄×粉红豹的成苗率最高,为91.67%;出苗时间也最短。如图7所示,播种17 d后开始出苗,第26天后完全出苗。其次是苹果花×粉红豹,成苗率为58.33%,第19天开始出苗。白雪公主×粉红豹的出苗率最低,为41.67%,第22天开始出苗(注:2024年1月中旬全部杂交组合统计完成,种子常温保存,于2024年4月28日进行播种试验)。
植物花部特征与有性繁殖密切相关,其中花部的空间特征是植物繁育系统的重要信息[13]。雌雄同熟和空间分布同位增加了自发授粉的可能性;雌雄异熟和空间分布异位增加了异交的可能性,减少雌雄功能的干扰[14]。本研究中,粉红豹、新加坡宫粉和安格斯都是雌雄同体的两性花,雌雄器官在同一垂直高度,且几乎所有雄蕊高于雌蕊,花药散粉时,花粉活力达到最高值,柱头具有较强的可授性,花粉易自发掉落在柱头上,这种雌雄同熟和雌雄器官结构特征机制有利于自花授粉。然而,在没有人工干预的条件下,极少看到其自然结实,可能存在自交不亲和机制。
雄蕊的花粉质量和花粉密度是决定植物杂交授粉是否成功的前提之一[15-22],而花粉生活力是评估花粉质量的重要参考依据[23-25]。研究表明,相对于染色法,花粉离体培养法能更加准确地反映出三角梅花粉的真实生活力[24]。本研究发现,粉红豹、新加坡宫粉都具有较强的花粉活力,适合作为父本。安格斯的花粉活力较低,不是理想的父本,其杂交结实率仅为1.37%,但是由于其具有较强的耐寒性,仍然值得作为父本进行杂交育种,只要有足够多的杂交样本,仍然可以得到杂交种子。金边叶浅紫、暗斑叶丽娜和斑叶光叶紫的花粉活力很低,不适合做杂交父本。
亲本间的杂交亲和性和花粉活力是决定杂交成功的重要前提,杂交不亲和性主要表现在受精前障碍和受精后障碍,受精前不亲和通常指花粉管无法正常伸长进入花柱或能进入胚珠但无法正常受精[21-22]。本研究中,安格斯和粉红豹的花粉在柱头上萌发时,在花粉粒周围出现大量的胼胝质以及在花粉管内沉积大量胼胝质,使得部分花粉管发生扭曲,伸入花柱异常。在胚珠附近同样有大量胼胝质进一步阻止花粉管完成受精。这说明这些杂交组合存在受精前障碍。安格斯的花粉萌发率很低,仅为(9.21±1.29)%,其杂交结实率也很低,仅为1.37%;而粉红豹的花粉萌发率较高,为(43.03±3.87)%,其杂交结实率为4.34%,高于安格斯的杂交结实率。由此可见,花粉活力与杂交结实率具有正相关性,花粉质量的高低是影响三角梅杂交结实率的重要因素。新加坡宫粉虽然花粉活力较高,但由于其花量较少且持续时间短,给杂交工作造成困难,因此不是理想的父本。
植物柱头可授性在花器官的发育过程中有阶段性的变化[25]。在本研究通过对3个适合做母本的三角梅品种的3个时期开花状态的柱头进行可授性检测,发现从花冠开展的初期到完全展开,柱头可授性逐渐增强,到花冠完全展开的Ⅴ期达到最强,所以人工授粉的最佳时期为花冠完全展开的Ⅴ期,其次为Ⅳ期,Ⅲ期因可授性非常弱不适合授粉。在试验过程中还发现杂交结实成功的种子所在的总花梗更粗,今后杂交育种时应选择母本植株上花梗较粗的花朵进行授粉,有望进一步提高三角梅杂交结实率。
三角梅为雌雄同体两性花,雌雄蕊被狭长的花被管包裹,雄蕊高于雌蕊。6个品种中粉红豹、新加坡宫粉的花粉活力较高,适合作为父本,其次是安格斯。在花冠完全展开的第Ⅴ期,母本的柱头可授性最强,为最佳的授粉时期。8个杂交组合均有结实,其中苹果花×新加坡宫粉的结实率最高(16.03%),苹果花×粉红豹(16.00%)次之;白雪公主×安格斯最低(1.37%),白雪公主×粉红豹次之(4.34%)。通过对结实率最低的2个组合进行花粉管生长的荧光显微观察发现,由于授粉后柱头和花柱内产生大量胼胝质并持续堆积,使花粉管发生扭曲变形,部分花粉管未到达胚珠就停止伸长,且只有少量花粉管伸入胚珠完成受精,这是导致结实率低的直接原因。对父本为粉红豹的3个杂交组合进行常规播种发现,浅茄×粉红豹出苗率最高达91.67%,其他2个组合均较低。
综上所述,8个杂交组合结实率较低,可能存在杂交受精前障碍;粉红豹为最佳父本,苹果花为最佳母本。
