Cassava (Manihot esculenta Crantz) has excellent characteristics such as barren resistance, drought resistance and strong stress resistance. Its roots are rich in carbohydrates, which can provide food rations for 1 billion people around the world, and plays an important role in consumption. The glycemic index (GI) is an indicator of how fast blood sugar rises after eating food containing carbohydrates for a certain period of time. Edible cassava with low glycemic index can improve blood sugar level, control body weight, and promote the edible and diversified development of cassava. Resource evaluation can provide a basis for the improvement of cassava varieties with low glycemic index. With three domestic popular edible cassava varieties for the test materials (SC9, SC12, SC6068), the determination of the important agronomic traits and cooking before and after the total starch, fast digest starch, slow digestion starch, resistant starch, amylose, amylopectin, crude protein, crude fiber and other quality traits and in vitro sugar rise index were evaluated. The average weight of single potato was 3.31-4.13 kg, the starch rate of fresh potato was 25.07%-27.53%, and the dry rate was 37.07%-38.91%. GI of SC9, SC12 and SC6068 was 42.6, 43.5 and 43.4, respectively, which was low glycemic index, 69.4, 66.9 and 64.0, respectively, all belonging to medium glycemic index food. In addition, the nutrient components changed significantly before and after cooking, in which the rapidly digested starch content and crude fiber content increased, the resistant starch content and soluble sugar content decreased significantly. Correlation analysis of each index found that the correlation between the glycemic index and the rapidly digested starch content and crude fiber content, the correlation coefficient was 0.95, with the resistant starch content and soluble sugar content, the correlation coefficient was -0.99 and -0.88 respectively, and the glycemic index and gelatinization characteristics. The differences of cassava varieties, processing methods, starch content and fiber content have a great influence on the glycemic index. Before cooking, SC9 has the lowest glycemic index, the highest taste evaluation, and good stability in processing, so it has the most potential for development among the three varieties.

木薯(Manihot esculenta Crantz)具有耐贫瘠、耐干旱、抗逆性强等优良特性,其块根富含碳水化合物,可为全球10亿人提供口粮,在食用方面发挥着重要作用。升糖指数(glycemic index,GI)是表示在一定时间内,进食含有碳水化合物的食物后血糖升高快慢的指标。升糖指数低的食用木薯,可以改善血糖水平,控制体重,推进木薯食用化和多元化发展,资源评价可为实现低升糖指数木薯品种选育和生物育种改良提供依据。以国内3个受欢迎的食用木薯品种华南9号(SC9)、华南12号(SC12)、华南6068(SC6068)为试验材料,测定其块根的重要农艺性状及蒸煮前后的总淀粉、快速消化淀粉、慢速消化淀粉、抗性淀粉、直链淀粉、支链淀粉、粗蛋白、粗纤维等品质性状和体外升糖指数。结果表明:3个品种平均单株薯质量为3.31~4.13 kg,鲜薯淀粉率为25.07%~27.53%,干物率为37.07%~38.91%;蒸煮前SC9、SC12和SC6068的GI分别为42.6、43.5和43.4,均属于低升糖指数食物,蒸煮加工后升糖指数分别为69.4、66.9和64.0,均属于中升糖指数食物。另外,各营养成分蒸煮前后变化显著,其中快速消化淀粉含量和粗纤维含量上升,抗性淀粉含量和可溶性糖含量显著下降;对各指标进行相关性分析发现,升糖指数与快速消化淀粉含量和粗纤维含量呈正相关,相关系数均为0.95,与抗性淀粉含量和可溶性糖含量呈负相关,相关系数分别为-0.99和-0.88,且升糖指数与糊化特性各值均存在相关性。木薯品种差异、加工处理方式、淀粉含量、纤维含量等对升糖指数具有较大影响,蒸煮前SC9升糖指数最低、食味评价最高,且在加工方面具有较好的稳定性,因此在3个品种中最具开发潜力。

木薯(Manihot esculenta Crantz)具有耐贫瘠、耐干旱、抗逆性强等优良特性,其块根富含碳水化合物,可为全球10亿人提供口粮,在食用方面发挥着重要作用。升糖指数(glycemic index,GI)是表示在一定时间内,进食含有碳水化合物的食物后血糖升高快慢的指标。升糖指数低的食用木薯,可以改善血糖水平,控制体重,推进木薯食用化和多元化发展,资源评价可为实现低升糖指数木薯品种选育和生物育种改良提供依据。以国内3个受欢迎的食用木薯品种华南9号(SC9)、华南12号(SC12)、华南6068(SC6068)为试验材料,测定其块根的重要农艺性状及蒸煮前后的总淀粉、快速消化淀粉、慢速消化淀粉、抗性淀粉、直链淀粉、支链淀粉、粗蛋白、粗纤维等品质性状和体外升糖指数。结果表明:3个品种平均单株薯质量为3.31~4.13 kg,鲜薯淀粉率为25.07%~27.53%,干物率为37.07%~38.91%;蒸煮前SC9、SC12和SC6068的GI分别为42.6、43.5和43.4,均属于低升糖指数食物,蒸煮加工后升糖指数分别为69.4、66.9和64.0,均属于中升糖指数食物。另外,各营养成分蒸煮前后变化显著,其中快速消化淀粉含量和粗纤维含量上升,抗性淀粉含量和可溶性糖含量显著下降;对各指标进行相关性分析发现,升糖指数与快速消化淀粉含量和粗纤维含量呈正相关,相关系数均为0.95,与抗性淀粉含量和可溶性糖含量呈负相关,相关系数分别为-0.99和-0.88,且升糖指数与糊化特性各值均存在相关性。木薯品种差异、加工处理方式、淀粉含量、纤维含量等对升糖指数具有较大影响,蒸煮前SC9升糖指数最低、食味评价最高,且在加工方面具有较好的稳定性,因此在3个品种中最具开发潜力。

Cassava (Manihot esculenta Crantz) has excellent characteristics such as barren resistance, drought resistance and strong stress resistance. Its roots are rich in carbohydrates, which can provide food rations for 1 billion people around the world, and plays an important role in consumption. The glycemic index (GI) is an indicator of how fast blood sugar rises after eating food containing carbohydrates for a certain period of time. Edible cassava with low glycemic index can improve blood sugar level, control body weight, and promote the edible and diversified development of cassava. Resource evaluation can provide a basis for the improvement of cassava varieties with low glycemic index. With three domestic popular edible cassava varieties for the test materials (SC9, SC12, SC6068), the determination of the important agronomic traits and cooking before and after the total starch, fast digest starch, slow digestion starch, resistant starch, amylose, amylopectin, crude protein, crude fiber and other quality traits and in vitro sugar rise index were evaluated. The average weight of single potato was 3.31-4.13 kg, the starch rate of fresh potato was 25.07%-27.53%, and the dry rate was 37.07%-38.91%. GI of SC9, SC12 and SC6068 was 42.6, 43.5 and 43.4, respectively, which was low glycemic index, 69.4, 66.9 and 64.0, respectively, all belonging to medium glycemic index food. In addition, the nutrient components changed significantly before and after cooking, in which the rapidly digested starch content and crude fiber content increased, the resistant starch content and soluble sugar content decreased significantly. Correlation analysis of each index found that the correlation between the glycemic index and the rapidly digested starch content and crude fiber content, the correlation coefficient was 0.95, with the resistant starch content and soluble sugar content, the correlation coefficient was -0.99 and -0.88 respectively, and the glycemic index and gelatinization characteristics. The differences of cassava varieties, processing methods, starch content and fiber content have a great influence on the glycemic index. Before cooking, SC9 has the lowest glycemic index, the highest taste evaluation, and good stability in processing, so it has the most potential for development among the three varieties.

木薯(Manihot esculenta Crantz)为大戟科(Euphorbiaceae)木薯属(Manihot)多年生小灌木,具有耐贫瘠、耐干旱、适应性强等特性[1],木薯在工业方面是生物质能源的重要原料,在食用方面是近10亿人的食粮、第三大能量供给,在巴西、非洲等热带地区,是重要的人体碳水化合物及淀粉来源。全球已有100多个国家种植,且食用占比很大(如尼日利亚),在各国粮食安全中扮演着重要角色[2]。块根是重要食用部位,味觉上有甜和苦之分,具有推广潜力的食用木薯一般是味甜,氢氰酸含量低于50 mg/kg(温和性木薯),而高于100 mg/kg(烈性木薯)一般用于加工木薯淀粉[3]。木薯此前是一种救荒作物,随着生活水平的提高,人们逐渐重视饮食健康,食用木薯不仅能饱腹,而且因其绿色粗粮且富含淀粉、矿物质及纤维等优点重新回到餐桌,多样化食品开发效果显著,人们研制出了木薯羹、木薯蛋糕、木薯饼干、木薯米等一系列美食。
当前高糖、高淀粉、高油脂的不健康饮食正在影响人类健康,截至2022年我国糖尿病患者超过1.18亿,占世界高血糖患者的22%[4],饮食健康始终是广大消费者关心的重大问题,研究发现合理健康的饮食习惯可以改善个人的身体状况及预防许多慢性疾病,健康意识高的消费者趋向于选择具有功能性的食品,因为它能减小患病的风险以及治疗疾病[5]
升糖指数(glycemic index, GI)在1981年由JENKINS等[6]首次提出,是用来表示一定时间内,进食含有碳水化合物的食物后血糖升高快慢的指标。根据数值大小,把食物进行分类:高(GI≥70)、中(55<GI<70)、低(GI≤55)[7]。高升糖指数食物释放葡萄糖速度快,容易导致肥胖和葡萄糖耐受不良等问题;低升糖食物释放葡萄糖缓慢,吸收率低,对稳定胰岛素水平有益,因此对于糖尿病、肥胖超重及具有心血管疾病患者而言,更适合进食低升糖食物。对于低升糖食物的研究,人们发现不同加工方式、蛋白质、脂肪、纤维、淀粉含量等均会影响GI的大小[8],木薯淀粉消化特性与GI联系密切,XU等[9]的研究表明,快速消化淀粉(RDS)和慢速消化淀粉(SDS)与GI表现为正相关,抗性淀粉(RS)与GI表现为负相关,通过酚酸酯化改性木薯淀粉,降低其RDS含量,可以有效调控GI。此外粗纤维含量也与GI呈负相关,它可通过阻止消化酶与碳水化合物充分接触,来减缓淀粉的水解和消化速度[10],而粗脂肪和粗蛋白在消化吸收过程中与粗纤维紧密关联,三者比例的合理搭配有助于营养最大化及降低GI。糊化特性可反应烹饪过程结构变化对消化速率的影响,完全糊化的淀粉可显著提高酶解效率,导致GI升高[11]。目前降低GI较好的方法是糖源替换、向食品中添加纤维或优化食物加工过程等,沈彤等[12]利用3种不同甜味剂来替换蔗糖,制作出的蛋糕GI仅为30。蔡攀福[13]在面条中添加2种膳食纤维(OBDF、KGM),结果使得3种面条的GI值下降了15.31%~20.42%。吴琼等[14]阐明选择合适的原料配比至关重要,再经过超声及微波辅助处理的加工方式,可以提升抗性淀粉来降低GI值。另外,选育低升糖指数品种是实现低GI的第一步,也是获得低升糖指数食物的关键,通过利用杂交育种和人工智能技术锁定低GI的优异特性基因等方法,截至2023年国外已筛选出16种低升糖水稻品种[15],小麦、马铃薯等作物在低GI上也取得了较大进展[16-18]。对于谷物、块根、块茎类含较高淀粉的食物,GI的预测通常是利用模拟人类消化道消化食物过程的方法,人体对淀粉的酶促降解是由唾液淀粉酶开始的,到小肠再由胰酶分解成单糖进行吸收,为人体大脑、红细胞、肾脏、生殖组织等提供能量,该方法在设定的温度和pH范围内测定淀粉转化为葡萄糖的量,这种体外方法与体内方法相比具有简便、价格低、高通量、对未被批准为人类食品的材料也可进行测定等优势[19]
淀粉占膳食能量摄入的30%~70%,新鲜木薯块根淀粉含量为16.55%~34.00%,干木薯中淀粉含量高达65%以上[20],因此食用木薯能为人体提供大量能量,但目前针对食用木薯的研究,国内外主要围绕获得低氰氢酸、高花青素、高淀粉及食品加工方法等方面,对食用木薯升糖指数及保健食品开发的研究鲜有报道,因此评价木薯的升糖指数对人类健康和木薯综合利用具有重要意义。
本试验在海南省儋州市进行,选择3个木薯品种华南9号(SC9)、华南12号(SC12)、华南6068(SC6068)作为试验材料,材料均取自海南省儋州市国家木薯种质资源圃(109°30ʹE,19°30ʹN),土地平坦,土壤为砖红壤,材料种植期间进行常规田间管理,每年4—5月种植,生育期约16个月。
每个小区选取3~5株长势基本一致、无病虫害的健康植株,将块根挖出,记录鲜薯重要性状,再从每株中选择3~5根薯块进行清洗去皮、切片并充分混匀后分成2份,一份蒸煮,一份不蒸煮,最后将鲜薯和熟薯真空冷冻干燥,打粉过80目筛低温保存待测。每个指标进行3次重复。
总淀粉含量采用Boxbio公司淀粉含量检测试剂盒测定;可溶性糖含量参考蒽酮比色法[21]测定;直链淀粉和支链淀粉含量参考ISO 6647[22]测定;氢氰酸含量采用GB 5009.36的第一法测定;粗纤维含量参考GB/T 8310—2013测定;粗蛋白含量参考GB 5009.5—2016中的凯氏定氮法测定;粗脂肪含量采用GB 5009.6—2016中的第一法测定;干物质含量采用烘干称重法测定。
用泰国进口的鲜薯淀粉测量仪,称5 kg左右的鲜薯在空气中的质量为w1(g)和鲜薯在水中的质量为w2(g),根据国际热带农业中心制定的公式[23-24]计算:
参考段惠敏[25]的方法稍作修改。称取100 mg木薯冻干粉于50 mL离心管中,加入0.5 mol/L乙酸钠缓冲液(pH 5.2)4 mL,混匀后放置在水浴锅保温至37 ℃,加入1 mL提前预热至37 ℃的现配酶液(3260 U/mL糖化酶与290 U/mL胰酶体积比为1∶5),移至200 r/min恒温摇床中,温度设置为37 ℃,准时在水解第0、20、120 min取样200 µL加入至含有200 µL无水乙醇的离心管中,迅速摇匀,常温下4000 r/min离心10 min,离心完成取100 µL上清液稀释待测。
用1 mg/mL葡萄糖标准溶液绘制标准曲线,分别取0、20、40、60、80、100 µL于2 mL离心管中,不足100 µL的用超纯水补齐体积,加入200 µL DNS溶液,沸水浴准确保温2 min,冷却后加超纯水0.9 mL,混合均匀,静置10 min后取200 µL于96孔酶标板上,酶标仪测定540 nm处的吸光值,测定样品含量,并根据以下公式计算快速消化淀粉含量(RDS)、慢速消化淀粉含量(SDS)、抗性淀粉含量(RS)。RDS=(G20-G0)×0.9/TS×100%;SDS=(G120-G20)×0.9/TS×100%;RS=[TS-(RDS+SDS)]/TS×100%。式中,G0为水解0 min产生的葡萄糖质量(mg);G20为水解20 min产生的葡萄糖质量(mg);G120为水解120 min产生的葡萄糖质量(mg);TS为样品总淀粉质量(mg);0.9为淀粉与葡萄糖换算系数。
参考方法及测定步骤同1.2.4,水解至第0、15、30、45、60、90、120 min取样,测定各时间点淀粉水解成葡萄糖的含量,以时间为横坐标,萄糖含量为纵坐标绘制水解曲线,用Origin的积分计算样品及参考食物(无水葡萄糖)的曲线面积(incremental area under the curve, IAUC),规定无水葡萄糖的水解指数为100,根据以下公式求样品的淀粉水解指数(SHI)和升糖指数(GI):
采用RVA-tecMaster快速黏度分析仪测定,在程序中输入样品含水量后自动计算所需加入的样品质量和水质量,准确称取样品和水配成淀粉乳放入仪器中,启动升温程序,样品在50 ℃保持1 min,然后升至95 ℃保持4 min,最后降回50 ℃得到黏度曲线,导出数据获取峰值黏度、最低黏度、最终黏度、崩解值、峰值时间和糊化温度。
将收获的木薯块根清洗去皮,去掉中部纤维硬芯,薯块切成3~5 cm方块放入蒸锅中大火隔水蒸,蒸锅上汽后转中火计时30~40 min,邀请15人以上组成评定小组品尝打分。参考NY/T 2669—2014热带作物品种审定规范进行,评分细则如表1
采用Excel 2016软件计算均值及标准差,利用IBM SPSS Statistics 20软件进行单因素方差分析,利用OriginPro 2024b软件进行绘图和相关性分析,相关性分析采用皮尔逊(Pearson)相关系数。
3个品种的单株薯质量、单株薯数、鲜薯淀粉率和干物率差异显著(表2)。3个品种中SC12的单株薯质量和单株薯数最高,平均单株结薯为8.67个,薯质量为4.13 kg,单株薯质量显著高于SC9和SC6068,SC6068最低,单株结薯仅有5.67个,薯质量为3.31 kg。SC6068的鲜薯淀粉率和干物率最高,分别为27.53%和38.91%,显著高于SC9和SC12,3个品种的干物质含量差异不显著。这表明3个品种中SC12具有较高的产量,而SC6068虽然产量较低但鲜薯淀粉含量高。3个品种的氢氰酸含量均低于50 mg/kg,在安全食用范围之内,其中SC9的最低,为32.20 mg/kg,与SC6068差异不显著,显著低于SC12的48.32 mg/kg。
3个木薯品种的食味评价差异如下(表3)。其中SC9的香度、苦度、甜度、黏度、纤维感的得分均比SC6068和SC12高,但SC6068的粉度比SC9和SC12高,为9.50分,从总分看,SC9得到54.70分,排在第一位,SC6068为50.38分,排在第二位,SC12为47.50分,排在第三位,因此SC9在食味上具有一定优势,而SC12在食味上表现一般。
3个品种的糊化特性差异显著(表4)。SC9的峰值黏度和最低黏度分别为4023 cP和1475 cP,显著高于另外2个品种,崩解值、最终黏度和回生值高于其他2个品种,SC6068的峰值时间和糊化温度最高。3个品种通过升温加热后,淀粉的结构受到破坏,吸水膨胀能力和热稳定性下降,亲水作用增强,更容易与水结合形成淀粉糊。
图1结果所示,3个品种均具有较高的淀粉含量(77.56%~83.79%),不同品种蒸煮后的总淀粉含量和直链淀粉含量与蒸煮前相比仅SC9显著差异,其余2个品种差异均不显著,直链淀粉占比与支链淀粉占比变化也不大,而蒸煮后的RDS含量均表现为显著上升,抗性淀粉(RS)均显著下降,SC12和SC6068的SDS含量显著上升,表明蒸煮处理对不同品种总淀粉含量及直/支链淀粉占比的影响较小,对RDS、SDS和RS含量的影响较大,即蒸煮后的木薯粉更容易被酶解消化。3个品种块根蒸煮后的快速消化淀粉(RDS)和慢速消化淀粉(SDS)含量差异显著。SC9熟薯的RDS含量最高,为50.02%,极显著高于SC12和SC6068,SC6068熟薯的RDS最低,为22.54%。SC6068熟薯的SDS最高,为27.70%,SC12为23.92%,二者显著高于SC9的6.57%。
不同品种间及蒸煮前后的可溶性糖、粗脂肪、粗纤维和粗蛋白含量差异显著(图2)。3个品种中SC9鲜薯的可溶性糖含量最高,为2.11%,显著高于SC12(1.51%)和SC6068(1.45%),而蒸煮后的可溶性糖含量均显著下降。SC9和SC6068品种中的粗脂肪含量蒸煮后有不同程度下降,其中SC9的鲜薯粗脂肪含量最高,为4.97%,蒸煮后降低到4.88%。3个品种鲜薯的粗纤维含量在2.76%~6.77%之间,蒸煮后显著上升至13.41%~17.40%,升高了2~3倍。SC6068蒸煮前后的粗蛋白含量均显著高于SC9和SC12,分别为2.24%和2.14%。
图3结果表明,3个品种蒸煮前后的升糖指数(GI)具有显著差异,蒸煮前SC9、SC12、SC6068的GI分别为42.6、43.5和43.4,均低于55,属于低升糖指数食物,蒸煮后GI分别显著上升至69.4、66.9和64.0,SC6068升糖指数最低,排在第一位,SC9和SC12的升糖指数显著高于SC6068,排在第二和第三位,从蒸煮前和蒸煮后的淀粉消化率曲线可以明显看出,熟木薯比生木薯更容易被消化释放葡萄糖,释放速率由快到慢,而生木薯随时间的延长消化速率基本不变,虽然蒸煮后GI值升高。
木薯块根GI值受到多个指标影响(图4)。在淀粉方面,GI与RDS呈极显著正相关,相关系数为0.95,与RS呈极显著负相关,相关系数为-0.99。在其他营养成分方面,GI与粗纤维含量呈极显著正相关,相关系数为0.95;与可溶性糖呈显著负相关,相关系数为-0.88。在糊化特征中,GI与峰值黏度、崩解值、峰值时间、糊化温度呈极显著负相关,相关系数分别为-0.95、-0.95、-0.94和-0.99;与回生值呈极显著正相关,相关系数为0.96;与最低黏度呈显著负相关,相关系数为-0.91。
木薯升糖指数的评价是筛选低升糖指数食用木薯种质的前提。本研究选取了3种广受欢迎的食用木薯品种,这些品种以其低氢氰酸含量、高淀粉含量和优良的口感(软糯且香甜)而受到市场的青睐,发现未经烹饪处理的木薯粉具有更低的GI值,属于低GI食品,采用蒸煮法作为加工手段,木薯样品的GI值显著增加,这可能是与蒸煮后样品中总碳水化合物含量的增加有关,这一结果与已报道文献[26]相一致。多项研究表明抗性淀粉具有预防糖尿病和调节肠道菌群的作用,与升糖指数间存在负相关关系,本研究结果与此相符[27-29]。此外,传统的高碳水化合物食物食用后会导致血糖快速升高,如煮熟的长粒茉莉香米GI为91.08[30],甘薯干为77.50~99.49[31];而本研究中的木薯与之不同,3个木薯品种蒸煮前GI在42.6~43.5之间,处于低升糖指数范围,蒸煮后升至64.0~69.4,虽大幅增加,但均未超过70,处于中升糖指数范围,说明木薯在低升糖食品开发中相对更具优势。
GI值的确定是一个复杂的过程,受多种因素影响,在评价食用木薯的GI时,除了品种差异、生长周期和栽培条件外,还需关注指标之间的相互作用,本研究对不同品种的淀粉和营养成分进行相关性分析,可知RDS、RS、粗纤维、糊化特性等是影响升糖指数的主要因素。经过热处理后,大部分淀粉解聚成更小的颗粒[32],糊化度增加,淀粉颗粒膨胀、变形、破裂,分子有序性的减弱和双螺旋结构的解离导致更容易被酶水解[33],使得RDS在总淀粉中的比例增加,RS随之减少,GI值升高。同时蒸煮会加速多糖分子的糖苷键断裂,释放寡糖,提高纤维含量[34],蒸煮还能改变纤维的形态,使其更利于消化道中的酶降解。热处理使木薯的峰值黏度明显降低,这与GOMES等[35]的研究一致,而崩解值的显著降低表示木薯粉具有更强的耐高温能力,稳定性增加,本研究中SC9具有最高的峰值黏度和崩解值,说明该品种淀粉机械稳定性更好,在食品加工中能形成更黏稠的食品,在口感和风味上相对其他品种更具优势,另外,SC9有最高的回生值,说明在回生过程中更容易形成抗性淀粉从而增加饱腹感,适合减肥人群食用。
