Transcriptional regulation is one of the pivotal mechanisms governing biological activities. Natural rubber (NR), a crucial industrial raw material, is predominantly derived from rubber trees. The transcriptional regulation of NR biosynthesis has emerged as a focal point in the fundamental research of rubber tree in recent years. Mevalonate diphosphate decarboxylase (MVD) is a key enzyme in the biosynthetic pathway of NR, yet its transcriptional regulation remains poorly understood. In this study, a 1500 bp promoter region of the HbMVD1 gene from rubber tree was cloned. Sequence analysis revealed the presence of various cis-acting elements, including the responsive to jasmonic acid, ethylene, mechanical wounding, light, and MYC binding. A gradient assay for autoactivation suppression demonstrated that the transcriptional autoactivation of the pAbAi-pHbMVD1 bait vector was effectively inhibited at an AbA concentration of 100 ng/mL. 13 non-redundant proteins from a rubber tree latex yeast cDNA library that interact with the HbMVD1 promoter, including one WRKY transcription factor (HbWRKY1), one heat shock transcription factor, and one G-box binding factor were identified utilizing yeast one-hybrid screening. Tissue-specific expression analysis indicated that the genes encoding the proteins exhibited distinct expression patterns across different tissues. Transcriptional regulation analysis showed that HbWRKY1 activated the expression of HbMVD1 in vivo. Molecular docking revealed that HbWRKY1 bound to the mechanical wounding element of the HbMVD1 promoter, and 11 potential amino acid binding sites were identified. This study would lay the groundwork for further elucidating the transcriptional regulatory network of HbMVD1 in rubber tree.

转录调控是生命活动的重要调控方式之一。天然橡胶是一种重要的工业原材料,主要来源于橡胶树。天然橡胶生物合成的转录调控是近年来橡胶树基础研究领域的热点之一。焦磷酸甲羟戊酸脱羧酶(MVD)是天然橡胶生物合成途径的关键酶,目前对其转录调控尚不清楚。本研究克隆了长度为1500 bp的橡胶树HbMVD1启动子,序列分析显示其含有茉莉酸响应、乙烯响应、机械伤害响应、光响应、MYC结合等顺式作用元件。自激活抑制梯度试验显示,AbA浓度为100 ng/mL时,pAbAi-pHbMVD1诱饵载体的转录自激活得到有效抑制。利用酵母单杂交技术从橡胶树胶乳酵母cDNA文库共筛选到13个与HbMVD1启动子结合的非冗余蛋白,包括1个WRKY转录因子(HbWRKY1)、1个热激转录因子、1个G-box结合因子。组织特异性分析显示这些蛋白的编码基因在不同组织间存在不同的表达模式。转录调控分析显示HbWRKY1在体内可以激活HbMVD1基因的表达。分子对接发现,HbWRKY1与HbMVD1启动子的机械伤害响应元件结合,并鉴定到11个潜在的氨基酸结合位点。本研究为进一步揭示橡胶树HbMVD1转录调控网络奠定基础。

转录调控是生命活动的重要调控方式之一。天然橡胶是一种重要的工业原材料,主要来源于橡胶树。天然橡胶生物合成的转录调控是近年来橡胶树基础研究领域的热点之一。焦磷酸甲羟戊酸脱羧酶(MVD)是天然橡胶生物合成途径的关键酶,目前对其转录调控尚不清楚。本研究克隆了长度为1500 bp的橡胶树HbMVD1启动子,序列分析显示其含有茉莉酸响应、乙烯响应、机械伤害响应、光响应、MYC结合等顺式作用元件。自激活抑制梯度试验显示,AbA浓度为100 ng/mL时,pAbAi-pHbMVD1诱饵载体的转录自激活得到有效抑制。利用酵母单杂交技术从橡胶树胶乳酵母cDNA文库共筛选到13个与HbMVD1启动子结合的非冗余蛋白,包括1个WRKY转录因子(HbWRKY1)、1个热激转录因子、1个G-box结合因子。组织特异性分析显示这些蛋白的编码基因在不同组织间存在不同的表达模式。转录调控分析显示HbWRKY1在体内可以激活HbMVD1基因的表达。分子对接发现,HbWRKY1与HbMVD1启动子的机械伤害响应元件结合,并鉴定到11个潜在的氨基酸结合位点。本研究为进一步揭示橡胶树HbMVD1转录调控网络奠定基础。

Transcriptional regulation is one of the pivotal mechanisms governing biological activities. Natural rubber (NR), a crucial industrial raw material, is predominantly derived from rubber trees. The transcriptional regulation of NR biosynthesis has emerged as a focal point in the fundamental research of rubber tree in recent years. Mevalonate diphosphate decarboxylase (MVD) is a key enzyme in the biosynthetic pathway of NR, yet its transcriptional regulation remains poorly understood. In this study, a 1500 bp promoter region of the HbMVD1 gene from rubber tree was cloned. Sequence analysis revealed the presence of various cis-acting elements, including the responsive to jasmonic acid, ethylene, mechanical wounding, light, and MYC binding. A gradient assay for autoactivation suppression demonstrated that the transcriptional autoactivation of the pAbAi-pHbMVD1 bait vector was effectively inhibited at an AbA concentration of 100 ng/mL. 13 non-redundant proteins from a rubber tree latex yeast cDNA library that interact with the HbMVD1 promoter, including one WRKY transcription factor (HbWRKY1), one heat shock transcription factor, and one G-box binding factor were identified utilizing yeast one-hybrid screening. Tissue-specific expression analysis indicated that the genes encoding the proteins exhibited distinct expression patterns across different tissues. Transcriptional regulation analysis showed that HbWRKY1 activated the expression of HbMVD1 in vivo. Molecular docking revealed that HbWRKY1 bound to the mechanical wounding element of the HbMVD1 promoter, and 11 potential amino acid binding sites were identified. This study would lay the groundwork for further elucidating the transcriptional regulatory network of HbMVD1 in rubber tree.

转录调控是生命活动的重要调控方式之一,是指在转录过程中通过一系列复杂的机制对基因表达进行精细调控[1]。转录因子是执行转录调控的重要成员。转录因子通过识别并结合到DNA上的顺式作用元件,激活或抑制相关基因的转录,进而调节生物体的生长发育、响应各种外界刺激以及自身生理反应等生物学过程[2]。酵母单杂交技术(yeast one-hybrid method,Y1H)是将顺式作用元件序列和编码转录因子的序列分别构建到含报告基因及激活结构域的表达载体,共转化酵母观察是否启动下游报告基因的表达来确定转录因子是否和顺式作用元件结合[3]。目前Y1H已是筛选鉴定与顺式作用元件结合转录因子的常用技术,被广泛应用于真核生物的转录调控研究。如余婧等[4]利用Y1H从烟草酵母cDNA文库中筛选到与NtCBT基因启动子结合的转录因子ANL2、ML1及NF-YA3;CAO等[5]利用Y1H从百合酵母cDNA文库中筛选到与LhMYBSPATTER基因启动子结合的转录因子ERF4和MYB4,并利用荧光素酶实验系统证明MYB4对LhMYBSPATTER基因的表达起激活作用而ERF4对LhMYBSPATTER基因的表达起抑制作用。
天然橡胶是一种重要的工业原材料,主要来源于橡胶树。与人工合成橡胶相比,天然橡胶具有良好的绝缘性、耐磨性和抗拉性,被广泛运用于工农业生产、国防建设、交通运输、医药卫生等领域,在国计民生中发挥着不可替代的作用[6-7]。天然橡胶本质是高分子聚异戊二烯链,分子量可达数百万道尔顿。自然界中存在2种主要的异戊二烯焦磷酸(isoprene pyrophosphate, IPP)合成途径:甲羟戊酸(mevalonic acid, MVA)途径和2-C-甲基-D-赤藻糖醇-4-磷酸(2-methyl-D-erythritol-4-phosphate, MEP)途径,其中MVA途径与橡胶树中的天然橡胶生物合成密切相关。MVA途径以乙酰辅酶A为底物,经过6步酶促反应最终形成IPP。焦磷酸甲羟戊酸脱羧酶(meva-lonate-5-pyrophosphate decarboxylase, MVD)催化MVAPP脱去1分子的CO2和H2O形成IPP,是MVA途径关键酶之一[8-11]。1971年SKILLETER等[12]从橡胶树胶乳中成功纯化MVD蛋白,并对其酶学活性进行分析,发现其活性与胶乳代谢存在显著正相关。但有关MVD的转录调控研究尚无报道。本研究克隆了橡胶树HbMVD1的启动子,通过筛选胶乳酵母cDNA文库,得到13个与HbMVD1启动子结合的蛋白,包括3个转录因子。本研究为进一步揭示天然橡胶生物合成的转录调控机制奠定基础。
橡胶树无性系热研8-79定植于中国热带农业科学院儋州基地。橡胶树热研8-79胶乳酵母cDNA文库为本课题组保存。多糖多酚植物基因组DNA提取试剂盒购自天根生化科技(北京)有限公司;平末端克隆试剂盒(pEASY®-Blunt Cloning Kit)购自北京全式金生物技术股份有限公司;胶回收试剂盒(FastPureGel DNA Extraction Mini Kit)购自南京诺唯赞生物科技股份有限公司;同源重组试剂盒(ClonExpress Ultra One Step Cloning Kit)购自南京诺唯赞生物科技股份有限公司;酵母菌株Y1H Gold、农杆菌GV3101、大肠杆菌DH10B感受态细胞购自上海唯地生物技术有限公司;其他常规试剂及生化试剂均为国产超纯和分析纯。
取橡胶树热研8-79幼嫩叶片,液氮研磨后根据多糖多酚植物基因组DNA提取试剂盒说明书进行基因组DNA提取,并通过1%琼脂糖凝胶电泳进行完整度检测。
从橡胶树热研8-79基因组数据中获取HbMVD1基因的启动子序列,利用Primer Premier 5.0软件设计克隆HbMVD1启动子的上游引物(proHbMVD1-F:TGTAGCATTGATGCAGCTT)和下游引物(proHbMVD1-R:TACAGCAAGCAACACACCA)。以橡胶树热研8-79基因组DNA为模板进行PCR扩增,反应体系为:DNA模板1 μL,2×PrimeSTAR mix 25 μL,10 μmol/L上、下游引物各1 μL,超纯水22 μL。反应程序为:95 ℃3 min;95 ℃ 30 s,56 ℃ 30 s,72 ℃ 30 s,35个循环;72 ℃ 10 min。PCR产物经1%琼脂糖凝胶电泳分离后,切胶回收连接至pEASY-Blunt克隆载体并转化大肠杆菌DH10B,过夜培养后挑选阳性克隆送至生工生物工程(上海)股份有限公司测序验证。将测序正确的HbMVD1启动子(proHbMVD1)序列提交至PlantCARE(http://bioinformatics.psb.ugent.be/webtools/plantcare/html/)在线网站进行顺式作用元件分析。
以测序正确的pEASY-proHbMVD1质粒为模板,用携带pAbAi载体同源臂的引物pHbMVD1-F(AAGC TTGAATTCGAGCTGTAGCATTGATGCAGCTT)和pHbMVD1-R(ACAGATCCCCGGGTACCGAT ACAGCAAGCAACACAC)进行PCR扩增并回收目的片段,通过ClonExpress Ultra One Step Cloning Kit同源重组方法将片段构建至pAbAi载体获得pAbAi-pHbMVD1诱饵载体,并以pHbMVD1-F端引物和pAbAi载体通用R端引物(CCATCTCGAA AAAGGGTTTGCC)对重组载体进行PCR验证。
将pAbAi-pHbMVD1诱饵载体和pAbAi-p53阳性对照载体线性化,分别转化酵母Y1H Gold感受态细胞并涂布于SD/-Ura固体培养基,30 ℃恒温倒置培养3 d。分别挑选阳性菌落,经无菌水稀释后涂布到SD/-Ura、SD/-Ura/AbAi(100 ng/mL)、SD/-Ura/AbAi(150 ng/mL)、SD/-Ura/AbAi(200 ng/mL)的固体培养基上,30 ℃倒置培养3 d,观察平板上菌落的生长情况。
从SD/-Ura平板上挑取含pAbAi-pHbMVD1诱饵载体的Y1H Gold单菌落,接种于5 mL SD/-Ura液体培养基中,30 ℃、250 r/min振荡培养16~18 h至饱和。取1 mL过夜培养物转接至50 mL YPDA液体培养基中,30 ℃振荡培养至OD600为0.5。室温下1000×g离心5 min收集菌体,用25 mL无菌水洗涤并重悬于1 mL 1.1×TE/LiAc缓冲液(10 mmol/L Tris-HCl、1 mmol/L EDTA、100 mmol/L LiAc, pH 7.5)。取50 μL含pAbAi-pHbMVD1诱饵载体的Y1H Gold感受态细胞,依次加入:2 μg胶乳酵母cDNA文库质粒DNA、10 μL鲑精DNA(10 mg/mL,预先煮沸5 min后冰浴)、500 μL PEG/LiAc溶液(40% PEG4000、100 mmol/L LiAc、10 mmol/L Tris-HCl、1 mmol/L EDTA, pH 7.5)轻柔混匀后于30 ℃水浴孵育30 min。加入20 μL DMSO(终浓度为7%),42 ℃热激15 min。室温下1000×g离心5 min,弃上清,菌体用1 mL YPDA重悬。30 ℃、250 r/min复苏培养1 h。将复苏培养物1000×g离心5 min,菌体用100 μL无菌水重悬。取10 μL涂布于SD/-Ura-Leu/AbA(100 ng/mL)筛选平板,30 ℃倒置培养3~5 d。待阳性克隆菌斑直径达1~2 mm时,挑取单菌落至新鲜SD/-Ura-Leu/AbA平板进行二次筛选。以含pAbAi-p53与pGADT53m的酵母菌株为阳性对照,含pAbAi-pHbMVD1与pGADT53m的酵母菌株为阴性对照。pGADT53m载体为课题组保存。
将阳性菌落接种于SD/-Ura-Leu/AbAi的液体培养基,30 ℃摇床震荡培养。以pGADT7载体测序引物(F: TAATACGAC TCACTATAGGGC; R: TAAGAGTCACTTTAAAA TTTGTAT)进行菌液PCR,取5 μL的PCR产物进行电泳分析,挑选条带清晰的PCR产物送至生工生物工程(上海)股份有限公司测序。
从国家基因组数据中心下载橡胶树形成层、树皮内层、初生胶乳、次生胶乳、雌花、雄花、叶片等7个组织的转录组数据,数据序列号为PRJCA004986。利用Tophat程序,将所下载数据与橡胶树热研8-79基因组序列进行比对,根据Cufflink程序完成基因的相对表达(FPKM)值计算。提取目的基因对应的FPKM值,经log2转换后通过Genesis软件进行表达热图绘制。
以特异引物0800-F(GG GCGAATTGGGTACCTGTAGCATTGATGCAGCT T)和0800-R(AGGAATTCGATATCAAGCTTTACAGCAAGCAACACACCA)扩增pEASY-proHbMVD1质粒,以特异引物2300-F(GACAGGGTACCCGGGGATCCATGACTTCCAAAGTTCTC)和2300-R(ACCATGGTACTAGTGTCGACTTAGCTTTGGACAGGCTCTGC)扩增含HbWRKY1基因的阳性酵母菌。参照ClonExpress Ultra One Step Cloning Kit同源重组试剂盒操作说明书,将扩增片段分别连入pGreenⅡ0800-LUC载体和pCAMBIA2300载体,获得pGreenⅡ0800-proHbMVD1: LUC和pCAMBIA2300-HbWRKY1重组载体。将测序正确的pGreenⅡ0800-proHbMVD1-LUC和pCAMBIA2300-HbWRKY1重组载体,以及pCAMBIA2300空载体分别转化农杆菌GV3101感受态,挑选单菌落于YEP液体培养基中过夜培养(30 ℃,220 r/min)。离心收集菌体并用YEP液体培养基重悬,按试验组(pGreenⅡ0800-proHbMVD1-LUC∶pCAMBIA2300-HbWRKY1=1∶1)和对照组(pGreenⅡ0800-proHbMVD1-LUC∶pCAMBIA2300=1∶1)进行菌液混合并调整至OD600=0.6,通过注射法注射烟草叶片。注射后的烟草暗培养48 h后喷施D-荧光素钾盐溶液,置于Princeton公司的活体植物发光成像系统(型号:LumaZonepylon 2048B)中观察。
通过HelixFold3(https://paddlehelix.baidu.com/?redirect=console)在线软件分析HbWRKY1与不同顺式作用元件之间的相互作用,并进一步利用PyMOL3.1软件(Schrödinger,美国)展示互作的氨基酸位点。
提取橡胶树热研8-79基因组DNA作为模板,以HbMVD1启动子引物进行扩增并测序分析,成功获得长度为1500 bp的HbMVD1启动子序列(图1A)。将序列提交至PlantCARE在线网站进行顺式作用元件分析,结果显示,HbMVD1启动子除了常见的TATA-box和CAAT-box等核心元件外,还含有茉莉酸响应元件(CGTCA-motif)、乙烯响应元件(ERE)、器械伤害响应元件(WUN-motif)、MYC结合元件(MYC)、光响应元件(Box 4)等(图1B)。
通过同源重组方式将HbMVD1构建至pAbAi得到pAbAi-pHbMVD1诱饵载体,利用HbMVD1启动子F端引物和pAbAi载体通用R端引物进行PCR扩增验证,结果显示可以扩增出1685 bp目的片段(1500 bp+185 bp),而以空载体为模板未扩增出目的条带(图2)。
取转化pAbAi-pHbMVD1诱饵载体的YIH Gold酵母菌株涂布于不同浓度AbA的SD/-Ura固体培养基上,以含p53-AbAi载体的YIH Gold酵母菌株为阳性对照。结果显示,p53-AbAi阳性对照菌株在AbA浓度为200 ng/mL时生长被抑制,而pAbAi-pHbMVD1测试菌株在AbA浓度为100 ng/mL时生长就被抑制(图3),因此AbA最小浓度为100 ng/mL。
将胶乳cDNA文库转化至含有pAbAi-pHbMVD1的酵母感受态细胞,涂布于多个SD/-Ura-Leu筛选平板进行文库筛选(图4A),并将候选阳性克隆进一步转移至新的SD/-Ura-Leu筛选平板培养,最终得到34个阳性克隆,编号P1~P34(图4B)。
将获得的34个阳性克隆经菌落PCR扩增测序,与NCBI数据库BLAST比对后共得到13个可能与HbMVD1启动子结合的蛋白,包括注释为磷脂酶(Hb15g055630)、细胞分裂周期因子(Hb07g037490)、生长素抑制蛋白(Hb04g-047470)、小分子热休克蛋白(Hb04g052040)、蛋白酶抑制剂蛋白(Hb06g002380)、钙调素结合蛋白(Hb07g028960)、磷脂酸磷酸酶(Hb07 g032180)、铁硫簇组装酶(Hb07g037260)、F-box蛋白(Hb12g001190)、橡胶延伸因子(Hb12 g002580)等10个蛋白。此外,还筛选到3个潜在转录因子,分别是Hb18g010770(WRKY转录因子)、Hb13g050000(G-box结合因子)、Hb16 g003060(热应激转录因子)(表1)。
利用课题组保存的7个橡胶树组织转录组数据,分析13个蛋白编码基因的组织特异性(图5)。结果显示,Hb15g055630Hb07g037260在7个组织中均高丰度表达,Hb06g002380在7个组织中的表达丰度很低甚至不表达,Hb07g037490Hb12g001190在胶乳中的表达量远高于其他组织。此外,Hb13g050000Hb07g028960Hb12g 002580Hb16g003060在开割胶乳中的表达量高于初生胶乳。
Blast比对分析显示,Hb18g010770序列与前人报道的橡胶树HbWRKY1完全一致。为鉴定HbWRKY1对HbMVD1启动子的转录调控,将携带proHbMVD1-0800重组载体(含荧光素酶报告基因)和pCAMBIA2300-HbWRKY1重组载体的农杆菌注射烟草叶片。活体成像分析显示,与空载组相比,HbWRKY1组荧光信号显著增强(图6A),表明HbWRKY1可以在体内激活HbMVD1启动子的表达。
为进一步解析HbWRKY1与HbMVD1启动子的结合位点,本研究利用HelixFold3分析HbMVD1启动子上不同顺式作用元件与HbWRKY1间的互作关系,发现仅WUN-motif与HbWRKY1存在相互作用,且在2埃距离范围内鉴定到11个与WUN-motif互作的氨基酸:Gln-101、Asn-224、Gly-292、Gly-396、Trp-405、Asn-406、Ser-409、Arg-433、Ser-434、Val-448和Ile-449(图6B)。
天然橡胶的生物合成大致可分为MVA途径的IPP合成(pre-IPP)和橡胶链的聚合延伸(post-IPP)2个阶段[13]。近年来天然橡胶生物合成的转录调控是橡胶树基础研究领域的热点之一,先后有多个转录因子生物学功能得到揭示,如HbMYC2结合并激活HbSRPPHbFPS基因的启动子、HbCZF1结合并激活HbHMGR基因的启动子、HbMADS4结合并抑制HbSRPP基因的启动子等[14-16]。MVD是橡胶生物合成途径关键酶之一,目前对其转录调控尚无报道。本研究克隆了橡胶树HbMVD1启动子序列,并通过酵母单杂交文库筛选获得了3个与其结合的转录因子成员。
编码G-box结合因子的Hb13g050000属于bZIP转录因子家族。bZIP是植物最大的转录因子家族之一,其特征是bZIP结构域高度保守[17]。前人研究显示,bZIP转录因子可以与ABRE、H-box、G-box、GLM、ACE等顺式作用元件相结合,广泛参与植物的生长发育及逆境胁迫等生物学过程[18]。有研究表明bZIP也参与植物次生代谢物的合成调控,刺五加的3个bZIP转录因子可以与刺五加皂苷合成关键基因(EsFPS, EsSS, EsSE)启动子结合并激活其表达[19];拟南芥的bZIP转录因子HY5可以结合萜类合成关键基因AtTPS03的启动子并激活其表达[20]。作为高分子萜类化合物,天然橡胶的合成途径属于典型的次生代谢。G-box结合因子的鉴定为深入研究HbMVD1的转录调控机制奠定基础。
热激转录因子(heat shock transcription factor, HSF)是植物中研究最广泛的转录因子家族之一,其结构可以分为DNA结合结构域、寡聚化结构域、核定位信号、核输出信号和C端转录激活结构域等5个部分,其中DNA结合结构域通过与启动子上的顺式作用元件结合,广泛调控植物对多种非生物胁迫尤其是低温/高温胁迫的响应[21]。在橡胶树中,李言等[22]分析了30个HbHSF家族成员在低温胁迫下抗寒品种和不抗寒品种中的表达模式,发现有5个成员在抗寒品种中的表达量显著高于不抗寒品种。有研究表明HSF也参与植物次生代谢物的合成调控。KUMAR等[23]HvHSFA2e基因在大麦中过表达,发现转基因株系的黄酮和萜类代谢主要基因的表达量显著上调;ZHANG等[24]发现茶树的CsHSFA1b和CsHSFA2可以通过激活茉莉酸途径抑制因子CsJAZ6的表达进而减少儿茶素的积累。本研究鉴定到1个HSF成员与HbMVD1启动子结合,为进一步解析HSF对天然橡胶的调控机制提供新的思路。
WRKY是植物特有的一类转录因子家族,其特征是含有1~2个高度保守的WRKYGQK基序以及1个锌指基序[25]。通过与启动子序列上的顺式作用元件结合,WRKY转录因子广泛参与植物的生长发育以及响应生物/非生物胁迫的生物学过程[26]。前人从橡胶树基因组中鉴定到81个HbWRKY成员,其中HbWRKY27结合并激活HbFPS基因的启动子,HbWRKY1结合并抑制HbSRPP基因的启动子[27-29]。本研究通过酵母单杂交筛库发现HbWRKY1也可以结合HbMVD1启动子,进一步的活体成像试验证明HbWRKY1可以激活HbMVD1启动子的表达。基于MVD是pre-IPP阶段关键酶而SRPP是post-IPP阶段的关键酶,推测HbWRKY1在天然橡胶生物合成的不同阶段起不同的调控作用,这可能是一种精细的调控模式,进而控制天然橡胶的生物合成。
转录因子通过与启动子区域的靶向顺式作用元件结合进而调控功能基因的表达。本研究对HbMVD1启动子序列分析鉴定到多个顺式作用元件,如CGTCA-motif、ERE、WUN-motif、MYC、ARE等。分子对接发现仅WUN-motif与HbWRKY1存在互作,且在2埃范围内鉴定到11个潜在的互作氨基酸。WUN-motif是一种与伤害反应相关的作用元件,通过响应机械伤害来激活目标基因的表达[30]。在生产中,割胶是收获橡胶树天然橡胶的唯一方式。割胶不仅可以提高天然橡胶合成效率,还可以激活橡胶合成相关基因如HbMVD1等的表达[16]。考虑到割胶也是一种严重的机械伤害,推测割胶后HbWRKY1可能通过与HbMVD1启动子上的WUN-motif顺式作用元件结合进而促进天然橡胶的生物合成。
