Using 77 Cymbidium ensifolium germplasms as the test materials, the phenotypic diversity of 21 quantitative traits and 12 qualitative traits was analyzed. The results showed that the coefficient of variation (CV) for quantitative traits ranged from 14.72% to 42.79%, indicating a significant degree of variation. Among these, the CV value for flower length was the lowest (14.72%), suggesting that this trait was relatively stable across different germplasms. The CV value for the number of flower stalks was the highest (42.79%), followed by the number of flowers (41.67%). Additionally, the Shannon-Wiener index (I) was close to 5, indicating a high level of diversity in quantitative traits among the germplasms. In the binary analysis of qualitative traits, the distribution of sepal spots or stripes (colors) among the germplasms was similar, with a rich variety of expressions for the features on the flower sepals. The CV for qualitative traits ranged from 9.86% to 83.81%, also showing a significant degree of variation. Among these, the CV value for leaf posture was the lowest (9.86%), while the CV values for lip shape (83.81%) and sepal color (70.69%) were the highest. Furthermore, the I index values for qualitative traits ranged between 4 and 5, indicating a high level of diversity in qualitative traits among the germplasms. The Simpson index (D) for qualitative traits ranged from 0.985 to 0.991, close to the value of 1, suggesting a relatively uniform distribution of qualitative traits and a high degree of diversity among the germplasms. Principal component analysis was conducted on 33 phenotypic traits of the germplasms, and the first 10 principal components were extracted based on the criterion that the characteristic root value is greater than 1. The characteristic value of the first 10 principal components was 6.406, 4.475, 2.840, 2.425, 2.139, 1.619, 1.517, 1.430, 1.163, and 1.038, with a cumulative contribution rate of 75.917%. The number of flower parts among the phenotypic traits had a significant weight in the principal components, indicating it is a major phenotypic trait of C. ensifolium. Hierarchical clustering analysis was performed on the germplasms using Euclidean distance, resulting in a dendrogram. At the 15th Euclidean distance, the germplasms were clustered into five major groups, with the main features of phenotypic traits concentrated in quantitative traits, especially in the significant weight of the number of flower parts, consistent with the results of the principal component analysis. This study explored the genetic diversity and relationships among C. ensifolium varieties through morphological trait diversity analysis, providing a basis for the scientific management and conservation of C. ensifolium germplasm resources. It also offers valuable genetic materials for future C. ensifolium breeding work.

以77份建兰种质作为供试材料,分析21个数量性状和12个质量性状的表型性状多样性。结果表明:数量性状的变异系数(CV)在14.72%~42.79%范围内变异程度较大。其中,花长度的CV值最低(14.72%),说明此性状在各个种质之间相对稳定。花序梗数量的CV值最高(42.79%),其次是花朵数(41.67%)。同时,Shannon-Wiener指数(I)均接近5,种质数量性状多样性相当高。在质量性状二元分析中,种质的萼片斑点或条纹(颜色)分布比例相近,花部萼片斑点或条纹的表现形式丰富。质量性状CV大小范围为9.86%~83.81%,变异程度也较大。其中,叶姿的CV值最低(9.86%),唇瓣形状的CV值最高(83.81%),萼片颜色居次之(70.69%)。另外,质量性状的I指数数值范围在4~5之间,种质质量性状的多样性相当高。质量性状的辛普森指数(D)范围在0.985~0.991之间,接近数值1,种质的质量性状分布较为均匀,多样性程度较高。对建兰种质33个表型性状进行主成分分析,按照特征根值大于1为原则,提取出前10个主成分。前10个主成分的特征值分别是6.406、4.475、2.840、2.425、2.139、1.619、1.517、1.430、1.163、1.038,累积贡献率为75.917%。花部位数量性状占据的主成分权重较大,是建兰主要的表型性状指标。对77份建兰种质采用欧式距离(Euclidean distance)进行系统聚类分析,得到树状聚类图。在欧氏距离的15处可将种质分为五大群体,群体表型性状主要特征集中在数量性状,尤其在花部位数量性状占据的主成分权重较大,这与主成分分析结果一致。研究通过形态学性状的多样性分析,探讨了建兰品种间的遗传多样性和亲缘关系,为科学管理和保护建兰种质资源提供依据,同时也为未来的建兰育种工作提供宝贵的遗传材料。

Using 77 Cymbidium ensifolium germplasms as the test materials, the phenotypic diversity of 21 quantitative traits and 12 qualitative traits was analyzed. The results showed that the coefficient of variation (CV) for quantitative traits ranged from 14.72% to 42.79%, indicating a significant degree of variation. Among these, the CV value for flower length was the lowest (14.72%), suggesting that this trait was relatively stable across different germplasms. The CV value for the number of flower stalks was the highest (42.79%), followed by the number of flowers (41.67%). Additionally, the Shannon-Wiener index (I) was close to 5, indicating a high level of diversity in quantitative traits among the germplasms. In the binary analysis of qualitative traits, the distribution of sepal spots or stripes (colors) among the germplasms was similar, with a rich variety of expressions for the features on the flower sepals. The CV for qualitative traits ranged from 9.86% to 83.81%, also showing a significant degree of variation. Among these, the CV value for leaf posture was the lowest (9.86%), while the CV values for lip shape (83.81%) and sepal color (70.69%) were the highest. Furthermore, the I index values for qualitative traits ranged between 4 and 5, indicating a high level of diversity in qualitative traits among the germplasms. The Simpson index (D) for qualitative traits ranged from 0.985 to 0.991, close to the value of 1, suggesting a relatively uniform distribution of qualitative traits and a high degree of diversity among the germplasms. Principal component analysis was conducted on 33 phenotypic traits of the germplasms, and the first 10 principal components were extracted based on the criterion that the characteristic root value is greater than 1. The characteristic value of the first 10 principal components was 6.406, 4.475, 2.840, 2.425, 2.139, 1.619, 1.517, 1.430, 1.163, and 1.038, with a cumulative contribution rate of 75.917%. The number of flower parts among the phenotypic traits had a significant weight in the principal components, indicating it is a major phenotypic trait of C. ensifolium. Hierarchical clustering analysis was performed on the germplasms using Euclidean distance, resulting in a dendrogram. At the 15th Euclidean distance, the germplasms were clustered into five major groups, with the main features of phenotypic traits concentrated in quantitative traits, especially in the significant weight of the number of flower parts, consistent with the results of the principal component analysis. This study explored the genetic diversity and relationships among C. ensifolium varieties through morphological trait diversity analysis, providing a basis for the scientific management and conservation of C. ensifolium germplasm resources. It also offers valuable genetic materials for future C. ensifolium breeding work.

建兰(Cymbidium ensifolium)隶属于兰科(Orchidaceae)兰属(Cymbidium),又名四季兰,是一种具有悠久栽培历史的观赏植物[1-2]。自古以来,建兰以其优雅的花姿、淡雅的香气和丰富的文化内涵受到人们的喜爱,尤其在中国,它不仅是园林艺术中的瑰宝,更是文人墨客笔下的常客,中国南宋时期,赵时庚著作的《金璋兰谱》,就详细记录了建兰的品种特性、栽培以及繁殖方法[3]。建兰原产于亚洲热带和亚热带地区,自然分布范围广泛,包括中国南部、印度、东南亚等地。其生长习性多样,既能在山野林间自由生长,也能适应人工栽培环境[4]
随着现代园艺技术的发展,建兰的栽培和繁殖技术得到了极大的提升,市场上建兰品种繁多,种质资源丰富。为了挖掘具有观赏价值与市场前景的优良建兰品种,并对其杂交选育后代拓宽新的市场潜力,建兰种质资源的表型性状研究是最基础和最关键的一个研究方法。表型性状研究作为植物种质资源遗传多样性研究的一种直观且基础的方法,它是植物优良品种选育的基础研究。深入了解和准确掌握种质资源的多样性水平,对于发掘潜在的有益资源、创新种质等方面,具有至关重要的意义[5-7]
迄今为止,我国兰科兰属植物的研究更多集中于品种鉴别分类[8-10]、高效栽培研究[11-15]、组织培养技术[16-17]以及分子调控机理机制[18-20]等方面,而对于兰属植物种质资源表型性状的研究主要集中在春兰[21-23]和蕙兰[24-26],建兰较少。傅巧娟等[27]通过对62份建兰种质的41个表型性状进行遗传多样性分析,主成分分析提取11个主成分,聚类分析划分为两大类,为建兰种质评价与杂交育种中亲本选择提供了主要表型指标。王宏利等[28]研究了30份不同来源的建兰种质资源的25个表型性状并进行主成分分析,提取6个主成分,确定花葶高度、花长和唇瓣长等性状在花部性状中贡献值最大,聚类分析被分别归于4个类群,为建兰的品种改良和遗传育种提供了重要的科学依据。本研究通过对77份不同来源的建兰种质品种进行表型性状多样性分析研究,筛选出重要的遗传表型性状,为未来的建兰育种工作提供宝贵的遗传材料,推动建兰品种的创新和改良,为建兰优良品种的杂交选育提供科学的参考。
77份建兰种质材料来自于福建省林业科技试验中心的中国兰花(建兰、墨兰)省级花卉种质资源库,种质材料来自福建、广东、四川、浙江、台湾等地,种植在种质资源库内,每2 a分盆1次,每隔4个月施放1次缓释肥。77份建兰种质材料详见表1
表型性状的选取主要参考《植物新品种特异性、一致性和稳定性测试指南-兰属(NY/T 2441—2013)》(以下简称《测试指南》),结合艾叶等[29]、王宏利等[28]对建兰表型性状的分析,选取33个表型性状。其中,21个数量性状包括植株高度、叶片数、叶片长度、叶片宽度、花序长、花序梗长、花序梗粗、花序梗数、花朵数、花长度、花宽度、中萼片长度、中萼片宽度、侧萼片长度、侧萼片宽度、花瓣长度、花瓣宽度、唇瓣长度、唇瓣宽度、合蕊柱长度以及合蕊柱宽度,12个质量性状包括叶姿、叶片颜色、叶艺、花型、萼片颜色、萼片姿态、花瓣形状、花瓣颜色、花瓣姿态、唇瓣斑点或条纹、唇瓣形状、唇瓣颜色。
2022—2023年,在建兰盛花期间从供试的建兰种质品种中随机挑选5盆植株,平均每盆5~8苗不等且均有1枝花葶以上,对每盆里开花株最壮的1苗进行数量性状和质量性状的测定与数据收集。数量性状使用直尺、游标卡尺以及卷尺进行测量,质量性状的颜色测定使用英国皇家园艺学会标准比色卡(RHSCC),颜色测量时间选择上午8:30至下午4:30,在自然光条件且光线无遮挡情况下,通过比色卡光孔位置,记录对应的颜色编号,且参照《测试指南》、艾叶等[29]、王宏利等[28]对记录的所有质量性状指标进行赋值转化。其他质量性状通过目测得到并记录。数量性状及测量方法详见表2,质量性状的赋值规则详见表3
以2022—2023年测定数据的平均值为依据。用Excel软件计算77份建兰种质品种数量性状的最大值(Max)、最小值(Min)、平均值(M)、标准差(S)、方差(V)以及变异系数(CV),同时使用Past软件(版本4.09)计算数量性状的Shannon-Wiener指数,分析建兰种质数量性状的表型遗传多样性程度。
用Excel软件对部分质量性状进行二元统计,计算其对应性状材料数和分布比例。同时,对测定的所有质量性状进行赋值后,分析计算其对应性状材料数、频率、MSV、CV。使用Past软件(版本4.09)计算质量性状的Shannon-Wiener指数与Simpson指数,分析建兰种质质量性状的表型遗传多样性程度与丰富程度。
使用SPSS 26软件对建兰种质品种进行主成分分析和系统聚类分析,绘制树状聚类图。
生物群体数量性状的差异性和多样性常用CV和Shannon-Wiener指数(I)来表示。从表4中可以看出,77份建兰种质品种的21个数量性状CV大小范围为14.72%~42.79%,各个数量性状的变异程度较大。其中,花长度的CV值最低(14.72%),说明此性状在各个种质之间相对稳定,变化不大。而花序梗数量的CV值最高(42.79%),其次是花朵数(41.67%),表明种质间的花部数量性状存在丰富的遗传多样性。同时,数量性状的I指数均大于4,且接近5,这一结果与数量性状对应的CV值相互契合。
共有11个质量性状显示为“有”或“无”。从表5可以看出,叶片无叶艺、花瓣有斑点或条纹(颜色)、唇瓣有斑点或条纹(颜色)、花瓣无唇瓣化、萼片无唇瓣化、合蕊柱无斑点或条纹以及有香味的居多,分布比例均在70%以上,其中萼片唇瓣化和香味均达到了100.0%。77份种质品种的萼片斑点或条纹(颜色)的分布比例相近,这说明花部萼片斑点或条纹的表现形式丰富。
表6可以看出,12个质量性状的CV值范围在9.86%~83.81%之间,各质量性状的变异程度较大,其中,叶姿的CV值最低(9.86%),唇瓣形状的CV值最高(83.81%),萼片颜色次之(70.69%)。另外,质量性状的I指数范围在4~5之间,说明建兰种质质量性状的多样性相当高,这一结果与性状对应的CV值情况相互契合。
辛普森指数(Simpson index,D)范围从0到1,数值越接近1表示物种分布越均匀,多样性越高。反之,数值越接近0表示物种分布越不均匀,可能存在少数物种占主导地位,多样性较低。所以,从表6可以发现,质量性状的D值范围在0.985~0.991之间,接近数值1。因此,从辛普森指数来看,建兰种质品种的质量性状分布较为均匀,多样性程度较高。
对建兰种质33个表型性状进行主成分分析,按照特征根值大于1为原则,提取出前10个主成分。从表7中可以看出,前10个主成分的特征值分别是6.406、4.475、2.840、2.425、2.139、1.619、1.517、1.430、1.163、1.038,累积贡献率为75.917%。
第一主成分的方差贡献率为19.411%,特征值较大的性状有中萼片与侧萼片长度、花长度与花宽度、花瓣长度与唇瓣长度。其中,中萼片长度的特征值最大,其值为0.913。花型的特征值最小,其值为–0.698,这与花的数量性状呈现一定程度的负相关性。第二主成分的方差贡献率为13.559%,特征值较大的性状有中萼片与侧萼片宽度、花瓣宽度与唇瓣宽度,特征值的范围在0.758~0.882之间。第三主成分的方差贡献率为8.607%,特征值较大的性状有植株高度、花序长、花序梗数以及花朵数,特征值的范围在0.577~0.762之间。第四和第五主成分的方差贡献率分别为7.350%和6.482%,特征值较大的性状有合蕊柱长度、萼片与花瓣颜色,特征值分别为0.585、0.844和0.796。
从主成分分析结果可知,建兰种质的花部位数量性状占据的主成分权重较大,是建兰主要的表型性状指标,可作为建兰种质资源收集与杂交选育的参考标准之一。
对77份建兰种质采用欧式距离(euclidean distance)进行系统聚类分析,得到树状聚类图。从图1可以看出,在欧氏距离的15处可将种质分为五大群体。
类群Ⅰ共有13份种质,分别是金皱红荷、红玉、铁骨银针、蝴蝶王、丹霞、荷鼎、连城素荷、东方红荷、赤丹、夏蕊蝶、青龙荷蝶、太白邀月、新品三星蝶。该类群的主要表型性状特征是植株矮、叶片短,萼片颜色以黄绿为主,花瓣性状以椭圆或圆形为主。
类群Ⅱ共有2份种质,分别是大宝岛与芽黄素。该类群的主要表型性状特征是叶片窄、花序短、花序梗长且细、花朵数少、花长度高、侧萼片长、唇瓣与合蕊柱长,花型以竹叶瓣为主。
类群Ⅲ共有2份种质,分别是牙签与仙野佛光。该类群的主要表型性状特征是花序梗数少、唇瓣宽度窄。
类群Ⅳ共有57份种质,分别是吉利三星、银边大贡、金丝马尾、绿鸟嘴、大凤素等。该类群的主要表型性状特征是花序梗粗、花部表型性状丰富多样化。
类群Ⅴ共有3份种质,分别是高山春色、娇鹤与红荷。该类群的主要表型性状特征是植株高、叶片长且宽、花序及花序梗长、花序梗数多、花朵数量多、花朵宽、花瓣长。
从分析结果可以得知,五大类群的主要表型性状区别主要集中在数量性状方面上,尤其在花部位数量性状上的特征区别占据大部分,这与主成分分析结果一致。
通过对77份建兰种质资源的33个表型性状进行多样性分析、质量性状二元分析、主成分分析和聚类分析筛选出重要的遗传表型性状。在表型性状的多样性分析中,变异系数作为衡量性状离散程度的指标,其数值的大小直接反映了性状在不同种质间的差异程度。当CV值超过10%时,即表示该性状在不同种质间的差异较大[30-31]。建兰种质资源的数量性状变异程度范围在14.72%~42.79%之间,说明其性状具有较高水平的多样性与品种创新改良的潜力。Shannon-Wiener指数(I)和辛普森指数(Simpson index,D)用于衡量种质资源多样性的程度[32-33]。77份建兰种质资源的I指数范围在4~5之间,D指数范围在0.9~1之间,这2个指数的结果与性状对应的CV值情况相互契合,说明建兰种质存在其他种/品种间的杂交,亲本来源多样且复杂[34]。同时,本研究还注意到在质量性状中,唇瓣形状的CV值最高(83.81%),这一结果与王宏利等[28]在建兰表型性状的变异分析结果一致。在质量性状的二元分析中,77份种质品种的萼片斑点或条纹(颜色)的分布比例相近,这说明花部萼片斑点或条纹的表现形式丰富。在主成分分析中,建兰种质的花部位数量性状占据的主成分权重较大,主要有中萼片长宽、侧萼片长宽、花长度与花宽度、花瓣长宽、唇瓣长宽、合蕊柱长度、花序长、花序梗数以及花朵数,这些表型性状可以为建兰新品种选育提供科学的依据及参考。在聚类分析中,将建兰种质分为五大群体。类群Ⅰ以植株矮、叶片短、萼片黄绿、花瓣椭圆或圆形为主;类群Ⅱ以叶片窄、花序短、花序梗长且细、花少、花长高、侧萼片长、唇瓣与合蕊柱长、花型竹叶瓣为主;类群Ⅲ以花序梗少、唇瓣宽度窄为主;类群Ⅳ以花序梗粗、花部表型性状丰富多样化为主;类群Ⅴ以植株高、叶片长且宽、花序及花序梗长、花序梗多、花朵多、花朵宽、花瓣长为主。在聚类分析中可知,五大类群的表型性状特征主要集中在数量性状方面,尤其在花部位数量性状上占据大部分,这与主成分分析结果相互呼应。这一结论与王宏利等[28]在建兰表型性状的主成分分析与聚类分析结果相契合。但是结论与傅巧娟等[27]并不一致,主要原因可能是研究中所选取的建兰种质品种不一致,且植物表型性状的多样性受到当地物候条件与空间差异的多方面因素所影响[35]。因此,本研究通过对77份建兰种质资源进行表型性状多样性分析,得出种质间存在较大的表型差异,且通过主成分分析和聚类分析筛选出占据主成分权重较大的花部位数量性状和5个主要类群,为建兰的品种改良和杂交育种提供有力的理论支持。后续工作将持续进行建兰种质资源的表型性状多样性研究,深入开展表型相关性、种质综合评价体系研究。下一步以分子基因标记技术和建兰表型性状多样性研究结合起来,为培育优质种质资源、发掘优良基因提供科学的理论依据。
