Broomcorn millet (Panicum miliaceum L.) is a traditional grain crop in China. Currently, there is still limited research on the genetic transformation methods for P. miliaceum. This study aimed to establish an efficient genetic transformation method for P. miliaceum mediated by Agrobacterium. Mature seeds of the millet variety Jishu 5 were used as the explants to induce embryogenic callus of millet on CIM and MSD induction media, respectively. They were used as the transformation receptor materials and infected with Agrobacterium containing plant expression vectors for 60 minutes and co-cultured for 6 days. The calli were then transferred to a screening medium containing 0.025 g/L of hygromycin to screen for resistant calli. Then, they differentiated on a differentiation medium containing 0.015 g/L of hygromycin. Finally, they rooted into seedlings on a rooting medium containing 0.015 g/L of hygromycin. PCR detection was performed on the regenerated plants using vector specific primers to identify whether they were positive transgenic plants. Based on the multiple batches of transformation experiments, the screening efficiency and transformation efficiency of resistant regenerated plants of broomcorn millet were statistically analyzed. The results showed that both CIM and MSD media could induce embryogenic callus of broomcorn millet, but CIM medium had a better effect on inducing embryogenic callus of broomcorn millet. After being infected by Agrobacterium and co-cultured with Agrobacterium, the multiple resistant regenerated plants were obtained through screening, differentiation, and rooting on a medium containing hygromycin. The PCR identification positive rate of the resistant regenerated plants obtained from three experiments was 100%, and the average transformation efficiency was over 30%. This study successfully established an Agrobacterium mediated genetic transformation method for broomcorn millet, which is simple to operate, efficient, cost-effective, and not limited by seasons. It could be carried out on a large scale, providing an effective technical means for genetic improvement of broomcorn millet.

糜子(Panicum miliaceum L.)是我国传统的粮食作物,截至目前,针对糜子遗传转化方法的研究仍较少,本研究旨在建立农杆菌介导的糜子高效遗传转化方法。以糜子品种冀黍5号的成熟种子为外植体,在CIM和MSD诱导培养基上分别诱导糜子的胚性愈伤,以其作为转化受体材料,用含植物表达载体的农杆菌侵染60 min并共培养6 d,转接至含0.025 g/L潮霉素的筛选培养基上筛选出抗性愈伤,接着在含0.015 g/L潮霉素的分化培养基上分化,最后在含0.015 g/L潮霉素的生根培养基上生根成苗,用载体特异性引物对再生植株进行PCR检测,鉴定其是否为阳性转基因植株。根据多个批次的转化实验统计糜子的抗性再生植株的筛选效率和转化效率。结果表明:CIM和MSD诱导培养基均能诱导出糜子胚性愈伤,但CIM培养基诱导的糜子胚性愈伤效果更好;糜子胚性愈伤在被农杆菌侵染及与农杆菌共培养后,经在含潮霉素的培养基上筛选、分化和生根后获得多株抗性再生糜子植株,3次试验获得的糜子抗性再生植株的PCR鉴定阳性率为100%,平均转化效率为30%以上。本研究成功建立了农杆菌介导的糜子遗传转化方法,操作简单,转化效率高,成本低廉,且转化不受季节限制,能规模化开展,为糜子的遗传改良提供有效的技术手段。

糜子(Panicum miliaceum L.)是我国传统的粮食作物,截至目前,针对糜子遗传转化方法的研究仍较少,本研究旨在建立农杆菌介导的糜子高效遗传转化方法。以糜子品种冀黍5号的成熟种子为外植体,在CIM和MSD诱导培养基上分别诱导糜子的胚性愈伤,以其作为转化受体材料,用含植物表达载体的农杆菌侵染60 min并共培养6 d,转接至含0.025 g/L潮霉素的筛选培养基上筛选出抗性愈伤,接着在含0.015 g/L潮霉素的分化培养基上分化,最后在含0.015 g/L潮霉素的生根培养基上生根成苗,用载体特异性引物对再生植株进行PCR检测,鉴定其是否为阳性转基因植株。根据多个批次的转化实验统计糜子的抗性再生植株的筛选效率和转化效率。结果表明:CIM和MSD诱导培养基均能诱导出糜子胚性愈伤,但CIM培养基诱导的糜子胚性愈伤效果更好;糜子胚性愈伤在被农杆菌侵染及与农杆菌共培养后,经在含潮霉素的培养基上筛选、分化和生根后获得多株抗性再生糜子植株,3次试验获得的糜子抗性再生植株的PCR鉴定阳性率为100%,平均转化效率为30%以上。本研究成功建立了农杆菌介导的糜子遗传转化方法,操作简单,转化效率高,成本低廉,且转化不受季节限制,能规模化开展,为糜子的遗传改良提供有效的技术手段。

Broomcorn millet (Panicum miliaceum L.) is a traditional grain crop in China. Currently, there is still limited research on the genetic transformation methods for P. miliaceum. This study aimed to establish an efficient genetic transformation method for P. miliaceum mediated by Agrobacterium. Mature seeds of the millet variety Jishu 5 were used as the explants to induce embryogenic callus of millet on CIM and MSD induction media, respectively. They were used as the transformation receptor materials and infected with Agrobacterium containing plant expression vectors for 60 minutes and co-cultured for 6 days. The calli were then transferred to a screening medium containing 0.025 g/L of hygromycin to screen for resistant calli. Then, they differentiated on a differentiation medium containing 0.015 g/L of hygromycin. Finally, they rooted into seedlings on a rooting medium containing 0.015 g/L of hygromycin. PCR detection was performed on the regenerated plants using vector specific primers to identify whether they were positive transgenic plants. Based on the multiple batches of transformation experiments, the screening efficiency and transformation efficiency of resistant regenerated plants of broomcorn millet were statistically analyzed. The results showed that both CIM and MSD media could induce embryogenic callus of broomcorn millet, but CIM medium had a better effect on inducing embryogenic callus of broomcorn millet. After being infected by Agrobacterium and co-cultured with Agrobacterium, the multiple resistant regenerated plants were obtained through screening, differentiation, and rooting on a medium containing hygromycin. The PCR identification positive rate of the resistant regenerated plants obtained from three experiments was 100%, and the average transformation efficiency was over 30%. This study successfully established an Agrobacterium mediated genetic transformation method for broomcorn millet, which is simple to operate, efficient, cost-effective, and not limited by seasons. It could be carried out on a large scale, providing an effective technical means for genetic improvement of broomcorn millet.

糜子(Panicum miliaceum L.),属禾本科黍属,起源于我国的黄河流域,又名黍稷,一年生小粒禾谷类作物,是人类最早驯化的作物之一,具有生育期短、抗旱耐瘠等特点,在干旱环境中具生产优势,主要分布在亚洲和欧洲的半干旱地区[1-3]。在中国,糜子曾是华北地区的主粮,如今仅在山区和边远地带栽培,年种植面积约32万hm2,产量约30万t[4]。目前,全球糜子种质资源有29 000余份,我国糜子种质资源库有9885份糜子资源[5-6]。与玉米、水稻等作物相比,糜子驯化水平低,具有许多野生性状,这些性状严重阻碍了糜子的育种进程及品种改良,成为现代糜子遗传改良的重点。目前我国糜子育种方法落后,主要为杂交育种和诱变育种等常规育种方法,分子标记辅助选择育种也很少[7-8]。现代基因工程技术的发展,为糜子的育种及品种改良提供了一种快速有效的途径。但由于糜子的遗传转化研究发展比较缓慢,严重影响了糜子的转基因育种速度。因此,建立糜子的高效遗传转化方法,对糜子的育种及品种改良尤为重要。
目前常见的植物遗传转化方法为基因枪法和农杆菌法。基因枪法是将包裹了带目的基因的DNA溶液的高速微弹直接送入完整的植物组织中,通过细胞和组织培养技术,可再生出植株,选出其中转基因阳性植株即为转基因植株[9]。其主要优点是不受受体植物范围的限制,但该仪器不易获得,且耗材成本高昂,操作复杂,外源基因插入拷贝数高,影响了基因枪法的广泛应用。农杆菌法是将目的基因插入到经过改造的T-DNA区,借助农杆菌的感染实现外源基因向植物细胞的转移与整合,然后通过组织培养技术,再生出转基因植株,具有操作简单,成本低,遗传稳定,转化效率高且外源基因插入拷贝数低等优点,目前在多种植物中广泛应用[10-16]。本研究以糜子品种冀黍5号的成熟种子为外植体,建立农杆菌介导的糜子高效遗传转化方法,为糜子的遗传改良提供了有效的技术手段。
糜子品种为冀黍5号。含潮霉素抗性基因(HPT)筛选标记的植物表达载体pRLG103_Predicted由Daniel Voytas实验室提供。大肠杆菌DH5α感受态和农杆菌AGL1感受态分别购自北京全式金生物技术股份有限公司和上海唯地生物技术有限公司。植物基因组DNA提取试剂盒(DP305)购自天根生化科技(北京)有限公司;Taq酶MIX购自北京康为世纪生物科技有限公司;MS购自Duchefa Biochemie公司;植物凝胶购自Caisson Labs公司;潮霉素购自Gold Biotechnology公司;蛋白胨和酵母粉购自Oxoid公司;琼脂糖购自Biowest公司;硫酸软骨素钠(软骨素)购自北京璞棠生物科技有限公司生产。其它试剂均为国产分析纯。
选取经休眠处理的冀黍5号的干燥成熟种子,去除种皮,将约2 g种子装入50 mL无菌离心管中,用20 mL有效浓度为3%的次氯酸钠溶液和50 μL吐温20溶液,消毒灭菌处理20 min,用无菌水清洗多次后将种子接种于胚性愈伤诱导培养基上。接种时注意种子芽点朝上,芽点不直接接触培养基,每皿培养基接种子约20个,24 ℃恒温培养,湿度为55%,培养过程中先以12 h/12 h的光/暗培养模式进行光照发芽,发芽后转为暗培养,暗培养28 d后从芽点处挑取诱导的胚性愈伤组织,继续暗培养7 d。使用2种胚性愈伤诱导培养基CIM和MSD进行糜子胚性愈伤诱导。CIM中含有MS干粉4.4 g/L、蔗糖30.0 g/L、七水合硫酸锌0.035 g/L、五水合硫酸铜0.0006 g/L、激动素0.0005 g/L、2,4-二氯苯氧乙酸0.002 g/L和植物凝胶9.0 g/L,pH 5.8。MSD中含有MS干粉4.4 g/L、蔗糖30.0 g/L、2,4-二氯苯氧乙酸0.002 g/L和植物凝胶9.0 g/L,pH 5.8。
从–80 ℃冰箱取出含载体pRLG103_Predicted(图1)的AGL1农杆菌菌株,置于LB培养基(蛋白胨10.0 g/L、酵母粉5.0 g/L、氯化钠10.0 g/L和琼脂粉15.0 g/L)上涂板活化,24 ℃暗培养3 d,长出单菌落;挑取3个以上单菌落,同时置于含0.05 g/L的羧苄青霉素和0.05 g/L卡那霉素的LB培养液中,于20 ℃以200 r/min进行震荡培养24 h,离心得到新鲜的农杆菌菌液。将新鲜的农杆菌菌液加至侵染液中,摇晃进行悬浮处理,得农杆菌菌悬液,并用侵染液将农杆菌菌悬液浓度调整到OD600值为0.6,然后于20 ℃以100 r/min低速振荡培养2~3 h进行活化,得活化的农杆菌菌液。侵染液中含有MS干粉2.2 g/L、蔗糖30 g/L、吗啉乙磺酸1.0 g/L、乙酰丁香酮0.04 g/L和质量体积比为0.1%的泊洛沙姆,pH 5.4。
将糜子的胚性愈伤诱导组织加至活化农杆菌菌液中,按每30 mL活化农杆菌菌液中加入60~80块直径约为0.5 cm糜子的胚性愈伤诱导组织的比例添加,于20 ℃以120 r/min轻摇振荡侵染60 min(振荡侵染采用暗培养),侵染完成后,倒掉菌液,得经侵染的糜子胚性愈伤组织。将经侵染的糜子胚性愈伤组织倒在无菌滤纸上,吸干多余的菌液,并在超净台上适当吹干,约30 min后转接到放有无菌滤纸的共培养培养基上,20 ℃暗培养6 d,得共培养后糜子胚性愈伤组织。共培养培养基中含有MS干粉2.2 g/L、蔗糖30.0 g/L、吗啉乙磺酸1.0 g/L、2,4-二氯苯氧乙酸0.002 g/L、激动素0.0005 g/L、乙酰丁香酮0.04 g/L和植物凝胶4.0 g/L,pH 5.4。
将共培养后糜子胚性愈伤组织转移至愈伤筛选培养基上,每皿约20~30粒共培养后糜子胚性愈伤组织,24 ℃暗培养2周,完成一次继代,转至光照培养2周(光照培养是于24 ℃采用12 h/12 h光/暗培养),完成第二次继代,整个共培养筛选过程继代2次,每2周继代1次,共培养筛选总用时1个月,得抗性愈伤组织。愈伤筛选培养基中含有MS干粉4.4 g/L、蔗糖30.0 g/L、七水合硫酸锌0.035 g/L、五水合硫酸铜0.0006 g/L、激动素0.0005 g/L、2,4-二氯苯氧乙酸0.002 g/L、植物凝胶9.0 g/L、特美汀0.5 g/L和潮霉素0.025 g/L,pH 5.8。
先从愈伤筛选培养基中将抗性愈伤组织转移至第一阶段分化培养基中,于24 ℃采用12 h/12 h的光/暗培养方式进行光培养2周,再转接至第二阶段分化培养基中,24 ℃采用12 h/12 h的光/暗培养方式进行光培养2周,得苗高约2 cm的分化幼苗。将分化幼苗接入生根培养基中,于24 ℃采用12 h/12 h的光/暗培养方式进行生根培养1~2周得糜子的抗性再生植株。第一阶段分化培养基中含有MS干粉4.4 g/L、蔗糖30.0 g/L、MS微量元素母液(1000×)1.0 mL/L、MS维生素(1000×)1.0 mL/L、6-苄氨基嘌呤0.003 g/L、激动素0.003 g/L、2,4-二氯苯氧乙酸0.001 g/L、椰子水5%(V/V)、特美汀0.5 g/L、潮霉素0.015 g/L、稀释5000倍的软骨素1.0 mL/L和植物凝胶9.0 g/L,pH 5.8。第二阶段分化培养基中含有MS干粉4.4 g/L、蔗糖20.0 g/L、MS微量元素母液(1000×)1 mL/L、MS维生素(1000×)1 mL/L、6-苄氨基嘌呤0.001 g/L、激动素(KT)0.001 g/L、2,4-二氯苯氧乙酸0.0005 g/L、椰子水5%(V/V)、特美汀0.5 g/L、潮霉素0.015 g/L、稀释5000倍的软骨素1.0 mL/L和植物凝胶9.0 g/L,pH 5.8。生根培养基中含有MS干粉2.2 g/L、蔗糖15.0 g/L、MS微量元素母液(1000×)1.0 mL/L、MS维生素(1000×)1.0 mL/L、3-吲哚丁酸0.0005 g/L、特美汀0.5 g/L、潮霉素0.015 g/L和植物凝胶9.0 g/L,pH 5.8。
糜子的抗性再生植株用水洗掉培养基,移栽到植物培养箱中利用营养土进行栽培,栽培过程中待转基因植株长到20 cm高后,剪取叶片提取其基因组DNA。以上述DNA为模板,对抗性再生植株进行了潮霉素抗性基因HPT和载体特异序列M1的PCR检测,使用的检测引物见表1。PCR检测使用的酶为Taq Mix,反应体系为:无菌水9.5 μL、引物(10 μmol/L)各1.0 μL、Taq Mix 12.5 μL、DNA模板1.0 μL。PCR扩增程序为:95 ℃预变性5 min;95 ℃变性30 s,60 ℃退火30 s,72 ℃延伸30 s,35个循环;72 ℃延伸7 min。PCR扩增产物经2%的琼脂糖凝胶电泳分离检测。根据不同批次的糜子转化实验统计糜子的抗性再生植株的筛选效率和转化效率。其中,筛选效率=PCR阳性的抗性再生植株数量/抗性再生植株数量×100%;转化效率=PCR阳性的抗性再生植株数量/胚性愈伤诱导组织数量×100%。
为考察添加细胞分裂素和生长素对愈伤组织形成的影响,在愈伤诱导阶段设置了2种培养基CIM和MSD。结果表明,CIM和MSD培养基均能诱导糜子胚性愈伤,但CIM诱导效率更高(图2),表明KT的添加对糜子愈伤的诱导有促进作用。糜子的胚性愈伤组织致密,非水渍状,其在CIM培养基上诱导的不同阶段的状态见图3
糜子胚性愈伤在筛选培养基上暗培养2周后,糜子胚性愈伤会发生褐化,将糜子愈伤转至光培养2周,期间会有抗性愈伤的生长,抗性愈伤会分化出绿色的组织(图4A),转至分化培养基上继续培养会分化出糜子再生苗。将糜子再生苗幼苗接入生根培养基中进行生根培养1~2周,即为抗性苗(图4B)。具体生根培养时间根据幼苗大小决定,待生根完成后,得糜子的抗性再生植株,整个转化周期约为12~16周。
将糜子的抗性再生植株移栽到植物培养箱培养至约20 cm高时进行PCR检测,HPTM1的检测结果为阳性,即为抗性转基因苗。多个糜子抗性再生植株的HPTM1的PCR检测结果如图5所示,糜子的抗性再生植株都能扩增出目的基因HPTM1,而野生型对照(CK-)、DNA提取对照(CK1)和空白对照(CK2)均未扩增出目的条带,说明这些抗性再生植株均为转基因阳性植株,目的载体已经成功导入到糜子基因组中。根据3个批次的糜子转化实验结果统计糜子的抗性再生植株的筛选效率和转化效率(表2),结果表明,本方法获得的糜子抗性再生植株的筛选效率为100%,平均转化效率约为30.4%。
糜子是起源于我国的最古老农作物之一,曾经是我国北方重要栽培作物,但随着农业生产的发展,逐渐退出主栽作物地位成为区域特色作物,主要原因是糜子产量没有大的突破,品质没有根本提升[9]。近年来,膳食结构多样化成为人们追求健康生活的需求,杂粮倍受重视,糜子作为药食同源作物展现了新的种植前景[17]。因此,改良糜子品质和提高糜子产量对糜子的产业发展具有重要意义。现代基因工程技术的发展为糜子提供了快速有效的转基因育种的途径。
有效的遗传转化对于转基因育种至关重要,但目前针对糜子的遗传转化方法仅在近3年开始有少量报道。2021年,申请号为202111531566.3的中国发明专利申请公开了一种黍属植物的遗传转化方法,其采用基因枪轰击法获得了糜子的转基因植株[18]。但该方法可能引起突变、丢失和基因沉默等问题,不利于外源基因在糜子中稳定表达,同时还具有操作复杂、命中率低、转化效率低、育种成本高等缺点。2024年,研究者成功利用农杆菌介导法获得了糜子的基因编辑植株。LIU等[19]利用农杆菌介导法将基因编辑载体导入糜子品种Hongmi的基因组中,以潮霉素抗性基因为筛选标记获得了八氢番茄红素脱氢酶(PDS)基因敲除的转基因植株,转基因植株出现白化症状。此外,LU等[20]也利用农杆菌介导法将基因编辑载体导入糜子品种Longmi4的基因组中,以潮霉素抗性基因为筛选标记获得了Pmsd1敲除的转基因植株,转基因植株出现明显的矮化症状。虽然上述研究已经通过基因枪法或农杆菌介导法获得了糜子的转基因株系,但由于农杆菌介导的转化法受基因型的限制,不同品系的胚性愈伤诱导能力、农杆菌侵染能力、愈伤脱分化能力均不同,导致不同品系的转化效率也存在较大差异。因此,针对不同品种的糜子的遗传转化方法仍需进行各项条件的摸索。
本研究以糜子品种冀黍5号的成熟种子为外植体,采用CIM培养基诱导糜子胚性愈伤,以潮霉素抗性基因为筛选标记经过抗性愈伤的筛选、分化和生根获得了糜子的转基因植株。本研究建立的糜子遗传转化方法与上述糜子遗传转化研究[19-20]中的方法有诸多不同:上述研究中愈伤诱导使用的激素为2,4-二氯苯氧乙酸或2,4-二氯苯氧乙酸和6-苄氨基嘌呤,而本研究中使用的激素为2,4-二氯苯氧乙酸和激动素,有较高的愈伤诱导效率;上述研究中使用的农杆菌均为EHA105,本研究使用的农杆菌为AGL1,具有良好的侵染糜子的效果;本研究在侵染培养液中添加了吗啉乙磺酸和泊洛沙姆,起到稳定侵染液pH的作用;LIU等[19]在糜子胚性愈伤分化时使用的激素是6-苄氨基嘌呤,而本研究经过对分化培养基中的激素种类及比例的摸索,发现在分化阶段使用2种分化培养基能获得良好的分化效率,阶段一和阶段二培养基均含激素2,4-二氯苯氧乙酸、激动素和6-苄氨基嘌呤,只是含量上有变化,同时在分化培养基中添加了椰子水和软骨素,也起到促进愈伤分化的作用。LIU等[19]在糜子筛选抗性胚性愈伤时使用的潮霉素浓度为0.05 g/L,本研究中使用的潮霉素浓度为0.025 g/L,这可能是由于品种差异导致的。本研究建立的冀黍5号的遗传转化方法获得的抗性再生植株PCR鉴定的阳性率为100%,平均转化效率30%以上,而LIU等[19]建立Hongmi的遗传转化方法的平均转化效率仅为16.7%。
本研究建立了糜子品种冀黍5号的农杆菌介导的高效遗传转化方法,转化周期为12~16周,操作简单,转化效率高,成本低廉,且转化不受季节限制,能规模化开展,为糜子品种的遗传改良提供有效的技术手段。
