The study was aimed to clarify the changes in moisture content, fiber microstructure, and metabolites during the hot air drying process of areca nut, and to precisely regulate the drying time. This study focuses on areca nut and five models were used to fit the hot air drying curves of areca nut at different temperatures, and a betel nut drying kinetics model was established. At the same time, scanning electron microscopy, LC-MS were used to explore the changes in the microstructure and metabolites of areca nut during the drying process. The results showed that the drying curve of betel nut showed an exponential decrease, and the drying rate curve was divided into two stages: acceleration and deceleration. Temperature had a significant impact on the drying rate. The Page model fit the hot air drying curve of areca nut well, with a coefficient of determination R2 above 0.98 and a RMSE in the range of 0.02-0.06. The difference between the predicted and true values of the model was not significant, and it could better reflect the water changes during the areca nut drying process. Microstructure analysis shows that the fiber layer of areca nut became dense and compact with increasing drying time, and the whitening treatment caused the fiber structure of areca nut to become loose. The texture results indicated that with the increase of drying time, the hardness and chewiness of areca nut increased, while the elasticity decreased. The differential metabolite results showed that the drying process affected organic acids, lipids, and lipid molecules by affecting the metabolism of amino acid, linoleic acid, central carbon, flavonoid biosynthesis, and phenylpropanoid biosynthesis. This study would enrich the basic research on areca nut processing and provide a theoretical basis for the selection of hot air drying time for areca nut.

干燥是槟榔加工的第1步,干燥过程直接影响槟榔的品质和货架期。为了明确槟榔热风干燥过程的水分变化、纤维微观结构及代谢物的变化规律,以期对槟榔干燥时间进行精准调控。本研究以槟榔鲜果和槟榔杀青果为研究对象,利用5种模型对不同温度下槟榔热风干燥曲线进行拟合,建立槟榔干燥动力学模型,同时利用扫描电子显微镜、LC-MS等探讨干燥过程中槟榔微观结构、代谢物的变化规律。结果表明:槟榔干燥曲线呈指数下降,干燥速率曲线分为加速和降速2个阶段,温度对干燥速率有显著影响;Page模型对槟榔热风干燥曲线拟合较好,模型决定系数(R2)在0.98以上,均方差(mean square deviation,RMSE)在0.02~0.06范围内,模型预测值和真实值差异不显著,可以较好地反映槟榔干燥过程中的水分变化规律。微观结构分析表明,随着干燥时间的增加槟榔纤维层变得致密、紧实,杀青处理导致槟榔纤维结构松散。质构结果表明,随着干燥时间的增加,槟榔硬度和咀嚼度增加,弹性降低。差异代谢物结果显示,干燥过程通过影响氨基酸代谢通路、中心碳代谢通路、亚油酸代谢通路、黄酮类生物合成、苯丙烷类生物合成进而影响有机酸、脂质和类脂分子等发生变化。研究结果为丰富槟榔加工基础研究,以及为槟榔热风干燥时间的选择提供理论依据。

干燥是槟榔加工的第1步,干燥过程直接影响槟榔的品质和货架期。为了明确槟榔热风干燥过程的水分变化、纤维微观结构及代谢物的变化规律,以期对槟榔干燥时间进行精准调控。本研究以槟榔鲜果和槟榔杀青果为研究对象,利用5种模型对不同温度下槟榔热风干燥曲线进行拟合,建立槟榔干燥动力学模型,同时利用扫描电子显微镜、LC-MS等探讨干燥过程中槟榔微观结构、代谢物的变化规律。结果表明:槟榔干燥曲线呈指数下降,干燥速率曲线分为加速和降速2个阶段,温度对干燥速率有显著影响;Page模型对槟榔热风干燥曲线拟合较好,模型决定系数(R2)在0.98以上,均方差(mean square deviation,RMSE)在0.02~0.06范围内,模型预测值和真实值差异不显著,可以较好地反映槟榔干燥过程中的水分变化规律。微观结构分析表明,随着干燥时间的增加槟榔纤维层变得致密、紧实,杀青处理导致槟榔纤维结构松散。质构结果表明,随着干燥时间的增加,槟榔硬度和咀嚼度增加,弹性降低。差异代谢物结果显示,干燥过程通过影响氨基酸代谢通路、中心碳代谢通路、亚油酸代谢通路、黄酮类生物合成、苯丙烷类生物合成进而影响有机酸、脂质和类脂分子等发生变化。研究结果为丰富槟榔加工基础研究,以及为槟榔热风干燥时间的选择提供理论依据。

The study was aimed to clarify the changes in moisture content, fiber microstructure, and metabolites during the hot air drying process of areca nut, and to precisely regulate the drying time. This study focuses on areca nut and five models were used to fit the hot air drying curves of areca nut at different temperatures, and a betel nut drying kinetics model was established. At the same time, scanning electron microscopy, LC-MS were used to explore the changes in the microstructure and metabolites of areca nut during the drying process. The results showed that the drying curve of betel nut showed an exponential decrease, and the drying rate curve was divided into two stages: acceleration and deceleration. Temperature had a significant impact on the drying rate. The Page model fit the hot air drying curve of areca nut well, with a coefficient of determination R2 above 0.98 and a RMSE in the range of 0.02-0.06. The difference between the predicted and true values of the model was not significant, and it could better reflect the water changes during the areca nut drying process. Microstructure analysis shows that the fiber layer of areca nut became dense and compact with increasing drying time, and the whitening treatment caused the fiber structure of areca nut to become loose. The texture results indicated that with the increase of drying time, the hardness and chewiness of areca nut increased, while the elasticity decreased. The differential metabolite results showed that the drying process affected organic acids, lipids, and lipid molecules by affecting the metabolism of amino acid, linoleic acid, central carbon, flavonoid biosynthesis, and phenylpropanoid biosynthesis. This study would enrich the basic research on areca nut processing and provide a theoretical basis for the selection of hot air drying time for areca nut.

槟榔(Areca catechu L.)为棕榈科常绿乔木,广泛种植在南亚、东南亚等国家以及我国南部的热带和亚热带地区[1]。槟榔是我国的四大南药之首,具有杀虫消积、下气、行水等功效。此外,槟榔含有多酚、生物碱、黄酮化合物、矿物质等活性成分,具有抑菌、抗氧化、抗抑郁等功能[2]。海南作为槟榔的最大种植区,2021年种植面积达26.7多万公顷。槟榔采摘后,除小部分用于鲜食外,90%以上的槟榔需要干燥后存储、运输,作进一步加工。
槟榔干燥是槟榔加工的第1步,干燥过程的水分含量直接影响槟榔的微观结构、质构特性、品质、货架期等。干燥是解决贮藏问题和延长保质期最常用的手段,通过降低物料中的水分含量,延缓腐烂,进而延长保质期[3]。针对干燥过程的水分动态变化进行研究,才能准确预测其水分的扩散规律、控制槟榔的水分含量,提高槟榔品质和贮藏安全性。干燥动力学通过探讨物料在干燥过程中的水分变化规律,反映物料的脱水特性[4]。目前,干燥方式主要有热风干燥、冷冻干燥、热泵干燥、微波干燥等[5],其中热风干燥技术是最为常见的干燥方法,通过控制温度和风速等参数达到理想的干燥环境,具有操作简单、生产成本低、设备投资小等优点[6-7]。目前很多学者探讨了热风干燥下不同材料,如青椒[7]、延胡索[8]、云南栘依[9]、牛大力[10]等的干燥特性,并利用Page、Weibull等函数对干燥曲线进行拟合,建立了干燥动力学模型。在槟榔干燥研究方面,袁源等[11]探讨了不同干燥温度和时间对槟榔果皮挥发性成分的影响;李梁等[12]研究了不同含水量对槟榔质构特性的影响。然而有关槟榔干燥动力学及干燥过程中槟榔微观结构等的变化尚无报道。本研究对不同干燥温度下槟榔鲜果及槟榔杀青果(沸水30 min)的水分变化情况、微观结构、质构特性及代谢物变化进行分析,建立槟榔鲜果、槟榔杀青果的干燥动力学模型,探讨干燥过程槟榔纤维结构及代谢物的变化规律,为槟榔干燥过程整体判断及水分含量的精准调控提供理论参考。
槟榔鲜果购自海南省三亚市崖州区崖州湾科技城槟榔种植户。
为了探讨干燥温度对槟榔干燥特性的影响,首先将槟榔鲜果在沸水中煮沸30 min(杀青),随后在50、55、60 ℃干燥36 h,分别在干燥2、5、8、12、16、20、23、26、30、36 h取出,测定水分含量。每个处理温度设置3次重复,取平均值。
为了探讨预处理方式对槟榔干燥特性的影响,根据槟榔杀青果在不同温度下的干燥情况,选择55 ℃对槟榔鲜果进行干燥,分别在干燥2、5、8、12、16、20、23、26、30、36 h取出,测定水分含量。
槟榔鲜果和槟榔杀青果分别在不同干燥时间取出后,测定其重量,记为wt,继续干燥,直至恒重,记为w。按照以下公式计算含水量(W):
式中,wt表示干燥至t时的槟榔重量,w表示槟榔干重。
根据以下公式计算槟榔干燥过程的干燥速率(DR):
式中,DR为干燥速率[g/(g·min)],Δt表示相邻2次测定的时间间隔(min),Xt表示干燥至t时的槟榔干基含水量(g/g),Xt+Δt表示干燥至t+Δt时的槟榔干基含水量(g/g)。
根据以下公式计算水分比(MR):
式中,Xt表示干燥至t时的槟榔干基含水量(g/g),Xe表示槟榔干燥达到平衡时的干基含水量(g/g),X0表示槟榔初始干基含水量(g/g),其中Xe远小于X0Xt,通常忽略不计,因此公式可以简化为:
利用Newton、Lagarithmic、Page等5种动力学模型方程(表1)对槟榔鲜果(50、55、60 ℃)、槟榔杀青果的干燥曲线进行拟合,并以均方差(mean square deviation,RMSE)、决定系数(R2)作为数学模型拟合结果优劣的标准,RMSE、R2的计算公式参照王海丽等[8]的报道。其中R2越接近于1,RMSE越小,表示模型对实际过程拟合得越好。
取出不同干燥时间的槟榔,于–80 ℃预冻,后冷冻干燥。取槟榔果中部位置,将其切成1 cm×1 cm大小的正方形,利用扫描电子显微镜测定槟榔纤维横切面、纵切面及表面的微观结构。测定方法:将双面导电胶粘于载物台上,放置槟榔纤维块,喷金处理后观察拍照,加速电压为15 kV。
利用质构仪测定不同干燥时间槟榔的质构特性。槟榔取出后,在室温下平衡1 h。质构测定采用TPA模式,P36探头,压缩比为20%,测定前探头的下降速度为1.0 mm/s,测试速度为1.0 mm/s,测定后的上升速度为1.0 mm/s,测定高度为5 mm,触发力为100 N,2次测定时间间隔为3 s。
利用高效液相色谱-质谱联用仪对槟榔鲜果和槟榔杀青果在55 ℃干燥过程中的代谢物进行分析。槟榔鲜果在干燥0、26、72 h取样,槟榔杀青果在干燥0、5、36 h取样。取50 mg固体样本于1.5 mL离心管中,加入400 μL提取液(乙腈∶甲醇=1∶1),涡旋混匀30 s后,低温超声提取30 min(5 ℃,40 KHz);随后将样品在–20 ℃静置30 min,13 000×g离心15 min后取上清液,氮气吹干;120 µL复溶液(乙腈∶水=1∶1)复溶,低温超声萃取5 min(5 ℃,40 KHz),13 000×g离心5 min后取上清液上机分析。取等体积的所有样本代谢物混合制备成质控样本(quality control,QC),在仪器分析过程中,每6个样本中插入1个QC样本,以考察整个分析过程的重复性。利用Progenesis QI软件对测定数据进行分析,同时将MS和MS/MS质谱信息与代谢公共数据库HMDB(http://www.hmdb.ca/)和Metlin(https://metlin.scripps.edu/)数据库进行匹配,得到代谢物信息。
所有试验均设置3次生物学重复,利用SPSS 20.0软件进行显著性分析(P<0.05);采用Origin 2021软件作图并对动力学模型进行拟合。
槟榔鲜果及槟榔杀青果在不同温度下的干燥曲线和干燥速率曲线如图1所示。从图1可以看出,槟榔干基含水量随着干燥时间的延长逐渐下降,不同温度下的干燥曲线变化趋势也相似,均呈现指数下降。干燥速率曲线显示,槟榔在干燥过程中主要有明显的加速和降速2个阶段,这与王安娜等[9]、胡希婷等[13]的研究结果一致。在干燥初期,干燥速率迅速升高,干基含水量显著下降,说明此时水分去除迅速,这可能是由于干燥初期,槟榔表面的水分首先排出,此时槟榔组织细胞中自由水丰富,这部分自由水迅速从松散的纤维层排出,干燥速率迅速增大至峰值,干基含水量显著下降。随着干燥时间的延长,干燥速率逐渐下降,干基含水量下降变缓,说明水分越来越难以去除。这是由于随着干燥时间的延长,槟榔纤维结构变得紧密,同时槟榔的水分扩散从表面转向内部,水分子的排出由最初的自由水排出逐渐转变为结合水排出,结合水与一些大分子如蛋白质、纤维素等结合紧密,导致水分排出缓慢,干燥速率下降。继续干燥,干燥速率趋近于0,干基含水量趋于稳定,说明此时槟榔达到恒重状态[14-15]。此外,干燥温度越高,干燥曲线越陡(图1A),干燥速率越大(图1B),干燥过程中水分去除速度越快,槟榔达到恒重时间越短。当干燥温度为50、55、60 ℃时,槟榔达到恒重的时间分别为48、30、26 h,说明干燥温度显著影响槟榔干燥前期的干燥速率,槟榔中水分子的扩散速度随着温度的升高显著升高,水分子去除加快,干基含水量下降更快[16]
对比槟榔鲜果和槟榔杀青果(沸水30 min)在55 ℃下的干燥曲线(图1C)和干燥速率曲线(图1D),可以看出,经过杀青处理后槟榔干燥速率更大,干基含水量下降更明显,槟榔达到恒重时间由60 h缩短为30 h,说明杀青处理使槟榔中的水分子更容易排出,缩短了干燥时间。这可能是由于杀青过程槟榔纤维结构被破坏,纤维层变得松散,水分子更容易排出。此外,杀青过程槟榔中结合水与大分子物质之间的化学键被破坏,结合水更容易排出。
利用表1中的5种动力学模型对槟榔干燥曲线进行拟合,其中模型Newton、Lagarithmic和Henderson and Pabis的线性方程显示,lnMR与干燥时间t呈线性关系,模型Wang and Singh线性方程中MR与干燥时间t呈线性关系,模型Page的ln(–lnMR)与lnt呈线性关系。
不同预处理及不同干燥温度条件下,槟榔干燥过程的MR与干燥时间t呈非线性关系(图2A图2B),因此Wang and Singh模型不适用于槟榔干燥曲线的拟合;槟榔干燥过程的lnMR与t呈非线性关系(图3A图3B),因此模型Newton、Lagarithmic和Henderson and Pabis也不适用于槟榔干燥曲线的拟合;槟榔干燥过程的ln(–lnMR)与lnt呈线性关系(图4A图4B),因此Page模型适用于槟榔干燥曲线的拟合,并用该模型对槟榔热风干燥过程中水分变化规律进行预测。
将不同温度及不同预处理的槟榔干燥曲线通过Page模型ln(–lnMR)=lnk+nlnt进行拟合,建立热风干燥槟榔动力学模型(表2)。不同温度、不同预处理下,槟榔热风干燥Page模型的Pearson相关系数(r)和决定系数(R2)分别在0.99和0.98以上,RMSE在0.02~0.06范围内,说明Page模型对不同温度和不同预处理条件下的槟榔干燥过程拟合较好。
为了验证模型的准确性,利用建立的模型预测槟榔在50、55、60 ℃热风干燥不同时间下的水分比(MR),并与真实测定值进行对比。由图5可知,热风干燥下槟榔水分比预测值和实际测定值相近,说明Page模型拟合度高,可以准确地反映槟榔热风干燥过程的水分变化规律。
分别对热风干燥不同温度下的水分比预测值和真实值进行T检验,结果见表3。根据T检验的双边检验结果得出50、55、60 ℃和槟榔鲜果的P值分别为0.78、0.20、0.15、0.88,P值均大于0.05,说明不同热风干燥温度槟榔的水分比预测值和真实值差异不显著。建立的槟榔热风干燥动力学Page模型可以很好地预测槟榔干燥过程中的水分比,进而对干燥时间进行筛选调控。
为了观察干燥过程中槟榔纤维微观结构的变化规律,根据槟榔干燥曲线和干燥速率曲线的研究结果,选择55 ℃作为槟榔干燥温度进行干燥。槟榔鲜果和槟榔杀青果干燥过程的SEM结果见图6。从图6可看出,不论是槟榔鲜果还是槟榔杀青果,随着干燥时间的延长,槟榔纤维横截面变得更加致密(图6A~图6C图6G~图6I),从纤维层纵截面可以观察到每条纤维内部随着干燥时间的延长密度增大,变得更加紧实(图6D~图6F图6L~图6N)。这是由于干燥过程中存在于纤维结构中的水分子排出。此外,随着干燥时间的延长纤维层变得致密,这也导致水分子越来越难排出,这与2.1.1中随着干燥时间的延长,干燥速率逐渐下降,干基含水量下降变缓的研究结果一致。对比槟榔鲜果和槟榔杀青果,杀青处理使槟榔纤维结构变得松散(图6A图6G),导致其在干燥过程中水分子更容易排出,验证了2.1.1的推测。此外,在同一干燥时间下,槟榔杀青果的纤维横截面和纵截面更加紧实、致密,说明槟榔杀青果在干燥过程中水分排出较快,很快达到恒重状态,这与2.1.1的研究结果一致。
利用质构仪测定55 ℃槟榔干燥过程中硬度、弹性和咀嚼性的变化。从图7可以看出,随着干燥时间的延长,槟榔杀青果硬度、咀嚼度呈上升趋势,弹性呈下降趋势(图7A)。这是由于槟榔经30 min沸水处理后,纤维结构变松散,柔软度较高,随着干燥时间的延长,水分散失,纤维结构变得致密、紧实,这与SEM结果一致。而槟榔鲜果本身水分含量较高,纤维排列有序、硬度较大,导致干燥初期硬度、咀嚼度及弹性最大,随着干燥时间的延长,水分子丧失,纤维结构被破坏,硬度、咀嚼性及弹性呈降低趋势(图7B)。
为了了解干燥过程对槟榔代谢物的影响,利用LC-MS对干燥过程槟榔的代谢物进行检测。根据QC样本PCA图以及潜在的特征峰在QC样本中的相对标准偏差(relative standard deviation,RSD),当RSD值为30%时,样本正离子和负离子模式下峰值累计百分比达到79%,大于70%,质控样品在PCA图中聚类明显,不同干燥时间样本组内集中,组间存在明显的分离(图8),说明设备、色谱条件及测定数据稳定可靠,可用于后续分析。
LC-MS正离子模式下共检测出9740个质谱峰,负离子模式下共检测出8887个质谱峰,在数据库中进行对比,一级模式下共注释到5052个代谢物,其中正离子模式下共注释到2841个代谢物,负离子模式下共注释到2211个代谢物。二级模式下共鉴定到425种代谢物,其中正离子模式下共注释到298个代谢物,负离子模式下共注释到127个代谢物。这些代谢物包括2种生物碱及其衍生物,35种苯环类化合物,66种脂质和类脂分子,18种核苷、核苷酸及其类似物,69种有机酸及其衍生物,11种有机氮化合物,36种有机氧化合物,48种有机杂环化合物,37种苯丙烷和聚酮类化合物。
根据OPLS-DA模型获得的变量权重值进行差异代谢物筛选,其中VIP>1且P<0.05的代谢物作为2组之间的显著差异代谢物,分别筛选槟榔鲜果和槟榔杀青果在55 ℃干燥过程中以及槟榔杀青果和槟榔鲜果之间的差异代谢物。
PCA结果显示,槟榔鲜果与干燥26 h的槟榔鲜果(图9A)、干燥26 h和72 h的槟榔鲜果(图9B)、槟榔杀青果与干燥5 h的槟榔杀青果(图9C)、干燥5 h和干燥36 h的槟榔杀青果(图9D)、槟榔鲜果和槟榔杀青果(图9E)、槟榔鲜果绝干状态和槟榔杀青果绝干状态(图9F)分别分布在PCA图中的不同区域,说明干燥过程以及杀青处理对槟榔代谢物有显著影响。
以槟榔鲜果为对照,干燥26 h后共鉴定到197种差异代谢物,其中127种表达上调,70种表达下调(图10A)。这些差异代谢物主要包括35种有机酸及其衍生物,34种脂质和类脂分子,20种有机杂环化合物,19种苯丙烷类和聚酮类,15种有机氧化合物,9种苯环型化合物,10种核苷、核苷酸和类似物,5种有机氮化合物,1种生物碱及其衍生物,49种其他类型的代谢物。以槟榔鲜果干燥26 h为对照,干燥72 h后共鉴定到211种差异代谢物,其中165种代谢物表达上调,46种代谢物表达下调(图10B),这些差异代谢物主要包括35种有机酸及其衍生物,26种脂质和类脂分子,25种有机杂环化合物,22种苯丙烷类和聚酮类,14种有机氧化合物,14种苯环型化合物,10种核苷、核苷酸和类似物,6种有机氮化合物,1种生物碱及其衍生物,58种其他类型的代谢物(图11A)。
以槟榔杀青果为对照,干燥5 h后共鉴定到179种差异代谢物,其中129种表达上调,50种表达下调(图10C)。这些差异代谢物主要包括30种有机酸及其衍生物,27种脂质和类脂分子,18种有机杂环化合物,14种苯丙烷类和聚酮类,16种有机氧化合物,16种苯环型化合物,11种核苷、核苷酸和类似物,3种有机氮化合物,44种其他类型的代谢物。以槟榔杀青果干燥5 h为对照,干燥36 h后共鉴定到211种差异代谢物,其中163种表达上调,48种表达下调(图10D),这些差异代谢物主要包括32种有机酸及其衍生物,37种脂质和类脂分子,29种有机杂环化合物,20种苯丙烷类和聚酮类,12种有机氧化合物,15种苯环型化合物,8种核苷、核苷酸和类似物,5种有机氮化合物,1种生物碱及其衍生物,52种其他类型的代谢物(图11B)。
以槟榔鲜果为对照,经杀青处理后,共鉴定到153种差异代谢物,其中74种表达上调,79种表达下调(图10E)。这些差异代谢物主要包括21种有机酸及其衍生物,22种脂质和类脂分子,21种有机杂环化合物,23种苯丙烷类和聚酮类,11种有机氧化合物,10种苯环型化合物,5种核苷、核苷酸和类似物,5种有机氮化合物,1种生物碱及其衍生物,34种其他类型的代谢物。以干燥后槟榔鲜果为对照,干燥后槟榔杀青果共鉴定到233种差异代谢物,其中114种表达上调,119种表达下调(图10F)。这些差异代谢物主要包括34种有机酸及其衍生物,36种脂质和类脂分子,28种有机杂环化合物,28种苯丙烷类和聚酮类,15种有机氧化合物,14种苯环型化合物,8种核苷、核苷酸和类似物,6种有机氮化合物,2种生物碱及其衍生物,62种其他类型的代谢物(图11C)。杀青过程主要对苯丙烷类和聚酮类、脂质和类脂分子、有机酸及其衍生物、有机杂环化合物产生影响。
不论是槟榔鲜果还是槟榔杀青果,干燥时间越长,其差异代谢物数量越多;干燥过程主要影响有机酸及其衍生物、脂质和类脂分子、有机杂环化合物。
为了进一步研究干燥过程中槟榔代谢物的变化,对差异代谢物进行代谢途径富集分析(图12)。代谢物途径富集分析是指分析选定的代谢集,并使用超几何分布算法获得该代谢集中代谢物显著富集的途径。对于槟榔鲜果,对干燥过程影响较大的通路有中心碳代谢通路、蛋白质消化和吸收通路、ABC转运通路、氨基酸代谢通路、黄酮类生物合成、苯丙烷类生物合成。对于槟榔杀青果,干燥过程影响较大的通路有亚油酸代谢通路、中心碳代谢通路、ABC转运通路、氨基酸代谢通路、蛋白质消化和吸收通路。对比杀青果和槟榔鲜果,杀青处理主要影响槟榔的苯丙烷类生物合成、苯丙氨酸、酪氨酸和色氨酸生物合成、精氨酸和脯氨酸代谢通路、黄酮类生物合成。说明杀青过程对槟榔黄酮类生物合成、苯丙烷类生物合成影响较大,即这2条通路在煮沸的过程中已经发生了很大的变化,导致经热水煮沸后的杀青果在干燥过程中这2条通路并无明显变化,而未经杀青处理的槟榔鲜果在干燥过程中这2条通路变化较大。
总的来说,干燥和杀青过程通过影响中心碳代谢通路、蛋白质消化和吸收通路、氨基酸代谢通路(苯丙氨酸、酪氨酸、色氨酸、精氨酸、脯氨酸)、亚油酸代谢通路、黄酮类生物合成、苯丙烷类生物合成进而影响有机酸、脂质和类脂分子等发生变化。
干燥通过降低物料中水分含量,延缓腐烂,进而延长保质期。针对干燥过程水分动态变化进行研究,才能准确预测其水分扩散规律,进而对干燥过程进行调控。水分在物料中的存在方式主要有3种:自由水、结合水以及流动性介于自由水和结合水之间的水分,其中自由水流动性最强,结合水流动性最弱。槟榔是我国的四大南药之首,具有杀虫消积、下气、行水等功效。目前对槟榔干燥动力学的研究较少,通过槟榔干燥特性的研究,有助于掌握其干燥规律,并进一步对干燥过程进行调控。
通过研究槟榔热风干燥特性表明,槟榔在整个干燥过程中的干燥曲线呈现指数下降,干燥速率曲线主要有明显的加速和降速2个阶段。干燥初期,槟榔中含有大量的自由水,槟榔内外水分含量均匀,槟榔表面的水分通过松散的纤维层迅速排出,干燥速率迅速增大。随着干燥时间的延长,槟榔水分扩散从表面转向内部,与大分子物质紧密结合的结合水开始排出,由于槟榔表面与内部的水分分布不均匀,出现表皮皱缩,导致表面积骤减,SEM显示,随着干燥时间的延长,槟榔纤维层变得致密、有序,这些都阻止了水分子的迁移排出,进而导致干燥速率下降。这种趋势与牛大力[10]、皂荚籽[17]的热风干燥特性一致。通过研究干燥温度和预处理对槟榔热风干燥的影响,发现温度越高,干燥速率越大,干燥曲线越陡,说明升高干燥温度可以缩短干燥时间,温度是影响槟榔干燥前期干燥速率的主要因素,这与王安娜等[9]、方良材等[10]、谢永康等[18]的研究结果一致。这是因为温度越高,槟榔表面和内部的水分子扩散速度越快,水分子去除加快。预处理方式也会影响干燥特性,本研究发现,杀青预处理通过在沸水中煮沸槟榔后再进行干燥,其干燥速率更大,槟榔达到恒重时间由60 h缩短到30 h。结合SEM结果,杀青处理使槟榔纤维结构变得松散,槟榔组织的细胞受到破坏,破坏了结合水与大分子物质之间的化学键,水分子更容易排出。
利用5种干燥模型对槟榔干燥曲线进行拟合,由于槟榔干燥过程的ln(-lnMR)与lnt呈线性关系,Page模型可以用于槟榔干燥曲线的拟合。该模型的相关系数(r)和决定系数(R2)分别在0.99和0.98以上,RMSE较小,模型的预测值和真实值差异不显著,拟合较好,可以很好地预测槟榔干燥过程中的水分比。马田田等[19]采用5种干燥模型对南极磷虾热风干燥特性进行拟合并对建立的模型进行验证,结果表明,Page模型的R2最大,模型的预测值与试验值重合度高,可以准确地预测南极磷虾热风干燥过程中水分比的变化规律。陈家鼎等[4]研究发现,Page模型可以准确地描述烟叶叶片和主脉的干燥动力学。王安娜等[9]研究表明,Page模型可以较好地拟合云南栘依的热干燥特性,该模型的R2在0.98以上,卡方、误差平方和(SSE)较低,能够较好地反映热风干燥过程中云南栘依水分比随干燥温度的变化规律。本研究建立的槟榔热风干燥过程动力学模型可以准确地预测槟榔干燥过程中的水分比,进而对干燥时间进行筛选调控。
干燥过程是水分子动态迁移的过程,在此过程中,对热敏感的代谢物也会发生相应的变化。通过研究干燥过程的差异代谢物,发现干燥过程会影响氨基酸代谢通路,包括苯丙氨酸、酪氨酸和色氨酸生物合成,精氨酸和脯氨酸代谢通路。荔淑楠等[20]研究发现,干燥主要通过影响甘氨酸、丝氨酸、苏氨酸代谢,氨基酸的生物合成等代谢路径影响当归代谢物的变化。与本研究结果具有相似之处。
本研究以槟榔鲜果和槟榔杀青果为研究对象,建立了不同热风干燥温度下槟榔的干燥动力学模型,同时探讨了干燥过程对槟榔微观结构、质构特性及代谢物的影响。结果表明:槟榔干燥曲线呈指数下降,干燥速率曲线分为加速和降速2个阶段,干燥温度显著影响槟榔的干燥速率;Page模型对槟榔热风干燥过程拟合较好,决定系数(R2)均在0.98以上,RMSE在0.02~0.06范围内,模型预测值和真实值差异不显著,可以较好地反映槟榔干燥过程的水分变化规律;槟榔纤维微观结构随着干燥时间的延长变得致密、紧实,杀青处理导致槟榔纤维结构松散,水分子更容易排出;随着干燥时间的延长,槟榔硬度、咀嚼度增加,弹性降低;干燥过程通过影响丙氨酸、天冬氨酸、谷氨酸、亚油酸及中心碳代谢进而影响有机酸、脂质和类脂分子等发生变化。研究结果可以为槟榔热风干燥时间的选择提供理论依据。
