Fenjiaren is a new Dendrobium variety bred by Sanming Academy of Agricultural Sciences for both ornamental and edible purposes. In factory seedling cultivation, it was found that Fenjiaren had slow cluster bud proliferation, low proliferation coefficient, underdeveloped root system of tissue culture seedlings and other problems, which affecting the large-scale production of this variety. To improve the efficiency of factory seedling cultivation of the variety, the sterile seedlings were used as the explants, the effects of plant growth regulators including 6-BA, NAA, and additives such as banana puree and AC on the proliferation of stem segments and the rooting of seedlings were studied using L9(33) orthogonal experiments. The suitable culture medium for the proliferation of stem segments of Fenjiaren was 1/2 MS+6-BA 3 mg/L, banana puree 100 g/L, white sugar 30 g/L, activated carbon AC 1 g/L, and agar powder 5.6 g/L. After 4 months of cultivation, the proliferation coefficient of the stem segment was 5.08. Range analysis indicated that the primary and secondary relationship of the factors was 6-BA>NAA>banana puree. The inter subject effect test showed that 6-BA had a significant impact on the growth rate coefficient (P<0.05). The most suitable culture medium for rooting was 1/2 MS+NAA 1 mg/L+banana puree 100 g/L+white sugar 15 g/L+agar powder 5.6 g/L. After 2 months of cultivation, the growth rate coefficient of the rooting was 12.82, the average plant height of the seedling was 12.66 cm, the average leaf length was 4.02 cm, the average number of roots was 11.80, and the average plant weight was 1.67 g. Range analysis indicated that the primary and secondary relationship of the factors was NAA>AC>banana puree. This study optimized the proliferation and rooting conditions of a new variety of Fenjiaren, which could provide technical reference for the seedling breeding of new varieties of Dendrobium and other Orchid plants.

粉佳人是三明市农业科学研究院选育的观赏石斛新品种,在工厂化育苗中,发现粉佳人丛芽增殖慢,增殖系数低,组培苗根系不够发达等问题,影响该品种大规模生产。为提高石斛新品种粉佳人工厂化育苗效率,以粉佳人无菌苗为外植体,采用L9(33)正交试验探讨植物生长调节剂6-BA、NAA及添加物香蕉泥、活性炭AC对粉佳人茎段不定芽增殖及组培苗生根的影响。结果表明,粉佳人茎段不定芽增殖适宜的培养基为1/2 MS+6-BA 3 mg/L+香蕉泥100 g/L+白糖30 g/L+AC 1 g/L+琼脂粉5.6 g/L。培养4个月后,茎段不定芽增殖系数为5.08。极差分析结果表明,3种因素(6-BA、NAA、香蕉泥)的主次关系为6-BA>NAA>香蕉泥,主体间效应检验表明6-BA对生长速度系数的影响达显著水平(P<0.05)。粉佳人生根壮苗最适宜的培养基为1/2 MS+NAA 1 mg/L+香蕉泥100 g/L+白糖15 g/L+琼脂粉5.6 g/L,培养2个月后,组培苗生根的生长速度系数为12.82,苗平均株高12.66 cm,平均叶长4.02 cm,平均根数11.80条,平均单株重1.67 g。极差分析表明3种因素(NAA、AC、香蕉泥)的主次关系为NAA>AC>香蕉泥,主体间效应检验表明,3种因素(NAA、AC、香蕉泥)对粉佳人生根的影响均不显著。表型性状相关性分析结果表明,苗单株重与株高、叶长、叶宽、根数呈极显著正相关(P<0.01)。本研究对石斛新品种粉佳人的增殖及生根条件进行了优化,可为石斛新品种及其他兰科植物的种苗繁育提供技术参考。

粉佳人是三明市农业科学研究院选育的观赏石斛新品种,在工厂化育苗中,发现粉佳人丛芽增殖慢,增殖系数低,组培苗根系不够发达等问题,影响该品种大规模生产。为提高石斛新品种粉佳人工厂化育苗效率,以粉佳人无菌苗为外植体,采用L9(33)正交试验探讨植物生长调节剂6-BA、NAA及添加物香蕉泥、活性炭AC对粉佳人茎段不定芽增殖及组培苗生根的影响。结果表明,粉佳人茎段不定芽增殖适宜的培养基为1/2 MS+6-BA 3 mg/L+香蕉泥100 g/L+白糖30 g/L+AC 1 g/L+琼脂粉5.6 g/L。培养4个月后,茎段不定芽增殖系数为5.08。极差分析结果表明,3种因素(6-BA、NAA、香蕉泥)的主次关系为6-BA>NAA>香蕉泥,主体间效应检验表明6-BA对生长速度系数的影响达显著水平(P<0.05)。粉佳人生根壮苗最适宜的培养基为1/2 MS+NAA 1 mg/L+香蕉泥100 g/L+白糖15 g/L+琼脂粉5.6 g/L,培养2个月后,组培苗生根的生长速度系数为12.82,苗平均株高12.66 cm,平均叶长4.02 cm,平均根数11.80条,平均单株重1.67 g。极差分析表明3种因素(NAA、AC、香蕉泥)的主次关系为NAA>AC>香蕉泥,主体间效应检验表明,3种因素(NAA、AC、香蕉泥)对粉佳人生根的影响均不显著。表型性状相关性分析结果表明,苗单株重与株高、叶长、叶宽、根数呈极显著正相关(P<0.01)。本研究对石斛新品种粉佳人的增殖及生根条件进行了优化,可为石斛新品种及其他兰科植物的种苗繁育提供技术参考。

Fenjiaren is a new Dendrobium variety bred by Sanming Academy of Agricultural Sciences for both ornamental and edible purposes. In factory seedling cultivation, it was found that Fenjiaren had slow cluster bud proliferation, low proliferation coefficient, underdeveloped root system of tissue culture seedlings and other problems, which affecting the large-scale production of this variety. To improve the efficiency of factory seedling cultivation of the variety, the sterile seedlings were used as the explants, the effects of plant growth regulators including 6-BA, NAA, and additives such as banana puree and AC on the proliferation of stem segments and the rooting of seedlings were studied using L9(33) orthogonal experiments. The suitable culture medium for the proliferation of stem segments of Fenjiaren was 1/2 MS+6-BA 3 mg/L, banana puree 100 g/L, white sugar 30 g/L, activated carbon AC 1 g/L, and agar powder 5.6 g/L. After 4 months of cultivation, the proliferation coefficient of the stem segment was 5.08. Range analysis indicated that the primary and secondary relationship of the factors was 6-BA>NAA>banana puree. The inter subject effect test showed that 6-BA had a significant impact on the growth rate coefficient (P<0.05). The most suitable culture medium for rooting was 1/2 MS+NAA 1 mg/L+banana puree 100 g/L+white sugar 15 g/L+agar powder 5.6 g/L. After 2 months of cultivation, the growth rate coefficient of the rooting was 12.82, the average plant height of the seedling was 12.66 cm, the average leaf length was 4.02 cm, the average number of roots was 11.80, and the average plant weight was 1.67 g. Range analysis indicated that the primary and secondary relationship of the factors was NAA>AC>banana puree. This study optimized the proliferation and rooting conditions of a new variety of Fenjiaren, which could provide technical reference for the seedling breeding of new varieties of Dendrobium and other Orchid plants.

石斛属(Dendrobium)植物是兰科(Orchidaceae)多年生附生草本植物[1],不仅具有药用价值,还具有很高的观赏价值,是兰科植物中观赏价值较高的四大名兰之一[2]。目前,石斛兰已被广泛栽培于世界各地,经过育种家开展的新品种选育工作,已有不少经典石斛兰品种流行于市场。我国开展石斛兰新品种选育较晚,目前市场流行品种大都为国外引进品种[3]
三明市农业科学研究院自2012年起开展石斛种质资源收集、保存等工作[4-16]。粉佳人是本课题组选育的观赏石斛新品种,2020年获得农业农村部植物新品种权(CNA20182196.6)。粉佳人结合了父母本的优良性状,生长速度快、抗病虫性好、花色彩明丽,茎秆粗壮、生长量大,具有良好的开发潜力。
与传统石斛种苗生产以种子播种繁殖不同,为稳定石斛新品种特性,在生产中需要通过茎段不定芽繁殖途径进行种苗繁育。相对种子播种的原球茎途径,茎段不定芽繁殖途径因增殖系数低[17],培养时间长[18],产量有限[19],难以实现规模化生产。本研究针对石斛新品种粉佳人茎段增殖特点,通过优化生长调节物质及添加物等条件对茎段不定芽增殖、小苗生根的影响,以提高增殖系数,获得健壮的组培苗。本研究将为石斛新品种及其他兰科植物的种苗繁育提供技术参考。
以石斛新品种粉佳人无菌苗为供试材料,试验于2022年3~9月在三明市农业科学研究院药用植物研究所组培室进行。
采用L9(33)正交试验设计(表1),优化粉佳人茎段不定芽增殖培养基配方,各处理以1/2 MS+白糖30 g/L+AC 1 g/L+琼脂粉5.6 g/L为基本培养基,pH 5.6~5.8,每瓶接种20条单节茎段为一个重复,每个处理5个生物学重复,接种4个月后统计茎段不定芽生长速度系数、长芽率、增殖系数。培养条件为光照14 h/d,光照强度1500~2000 lx,培养温度(25±2)℃。K1=(Q2Q1)/Q1K2=(N2/N1)×100%,K3=N3/N1。式中,K1表示生长速度系数,Q1表示接种的质量,Q2表示增殖后的质量;K2表示长芽率,N2表示长芽的茎段数,N1表示接种的茎段数;K3表示增殖系数,N3表示增殖后的茎段数。
采用L9(33)正交试验设计(表2),优化粉佳人生根壮苗培养基配方,各处理以1/2 MS+白糖15 g/L+琼脂粉5.6 g/L为基本培养基,pH 5.6~5.8,每瓶接种20株2~3 cm无根苗为一个重复,每个处理5个生物学重复,接种2个月后统计生根苗生长速度系数(同上)、合格苗比率(种苗分级标准参照T⁄ZNZ 168.3—2023《丽水市铁皮石斛标准综合体第3部分:种苗繁育》[20])、折干率及表型性状。培养条件同上。H=N4/N5×100%,F=W1/W2×100%。H表示合格苗比率,N4表示合格苗株数,N5表示生根苗总数;F表示折干率,W1表示整株苗干重,W2表示整株苗鲜重。
采用Excel 2007软件进行数据统计,SPSS 19.0软件进行方差分析及相关性分析。
表3可知,处理7的生长速度系数为12.38,极显著高于其他处理;长芽率前3位分别为处理7(88.49%)、处理1(85.00%)、处理4(84.09%),且三者间差异不显著;增殖系数位于前2位的是处理7(5.08)、处理4(4.75),且二者间差异不显著。综合考虑生长速度系数、长芽率和增殖系数,最适宜粉佳人茎段增殖的为处理7:6-BA 3 mg/L+香蕉泥100 g/L。
极差分析结果表明,3个因素对生长速度系数的影响依次为:6-BA>NAA>香蕉泥,对长芽率的影响依次为:NAA>6-BA>香蕉泥;对增殖系数的影响依次为:6-BA>NAA>香蕉泥。根据各列K值大小可知,生长速度系数、长芽率和增殖系数的最优组合为A3B1C3:6-BA 3 mg/L+香蕉泥100 g/L。
主体间效应检验(表4)表明,NAA和香蕉泥对粉佳人茎段增殖影响不显著,6-BA对生长速度系数的影响达显著水平(P<0.05),对长芽率的影响达极显著水平(P<0.01),三者对增殖系数影响的主次关系为:6-BA>NAA>香蕉泥。不同处理的茎段增殖情况见图1,生长速度系数、长芽率和增殖系数最高的处理7(图1G1、G2)茎段萌出的芽多、壮、长势均匀,增殖程度最佳。
表5可知,生长速度系数位于前3位的是处理7(12.82)、处理1(6.69)、处理8(4.94),处理7极显著高于其他处理;合格苗比率位于前3位的分别为处理9(93.79%)、处理7(93.33%)、处理2(93.04%),三者间差异不显著;折干率位于前3位的是处理7(9.42)、处理1(8.82)、处理3(8.53),处理7、处理1二者间差异不显著。综合考虑生长速度系数、合格苗比率和折干率,最适宜粉佳人生根的为处理7:NAA 1 mg/L+香蕉泥100 g/L。
极差分析结果表明,3个因素对生长速度系数的影响依次为:NAA>AC>香蕉泥,对合格苗比率的影响依次为:香蕉泥>NAA>AC;对折干率的影响依次为:香蕉泥>NAA>AC。根据各列K值大小可知,生长速度系数、合格苗比率和折干率的最优组合为A3B1C3:NAA 1 mg/L +香蕉泥100 g/L。
主体间效应检验(表6)表明,NAA、AC和香蕉泥对粉佳人生根的影响均不显著,三者对生长速度系数的影响的主次关系为NAA>AC>香蕉泥。不同处理的小苗生根情况见图2,处理7(图2G1、G2),芽苗的长势优于其他处理。
表7可知,处理7(表7图3G)生根苗的株高(12.66)、叶长(4.02)、根数(11.80)、单株重(1.67)均高于其他处理,其中株高与处理1(9.10)的差异显著(P<0.05);叶长与其他处理差异不显著;根数与处理1(4.60)、处理2(2.80)、处理3(6.20)、处理4(5.60)、处理6(3.00)、处理8(5.80)的差异极显著(P<0.01),与处理5(7.20)、处理9(6.60)的差异显著(P<0.05);单株重与处理1(0.55)、处理2(0.63)、处理3(0.68)、处理6(0.78)、处理8(0.82)、处理9(0.76)差异极显著水平(P<0.01),与处理4(1.29)、处理5(1.30)差异显著(P<0.05)。处理7的茎粗(2.54)、叶宽(0.84)、叶数(7.60)和根长(5.30)位于第二,其中茎粗与最高的处理8(2.72)差异不显著,叶宽与其他处理差异不显著,叶数与最高的处理5(8.40)差异不显著,根长与最高的处理2(5.52)差异不显著。
相关分析(表8)表明,单株重与株高、叶长、叶宽、根数呈极显著正相关(P<0.01),与叶数、根长无相关性。
石斛组织培养中,与原球茎增殖途径相比,茎段不定芽诱导途径增殖系数普遍更低,因此,不定芽诱导增殖培养是决定石斛茎段快繁的重要环节。当粉佳人的茎段接种于1/2 MS+白糖30 g/L+AC 1 g/L+琼脂粉5.6 g/L+6-BA 3 mg/L+NAA 0 mg/L+香蕉泥100 g/L培养基时,增殖系数可达5.08,芽诱导率达88.49%。6-BA是主要影响因子,6-BA对茎段增殖生长速度系数及长芽率的影响呈显著或极显著水平。随着6-BA浓度升高,茎段不定芽增殖的平均生长速度、长芽率、增殖速度也随之升高。当6-BA浓度不变,增殖系数随着NAA浓度升高而降低,表明较高浓度的NAA会抑制芽的增殖。这一点在其他种石斛的培养中也有类似的发现,姜燕燕等[21]用茎段不定芽诱导、增殖及生根3步法实现铁皮石斛培养,在不定芽诱导中同样发现,6-BA对不定芽的诱导和增殖的影响作用显著,当6-BA的浓度达到2 mg/L时,诱导率就可达到92.9%,但当6-BA的浓度为1 mg/L与3 mg/L时,增殖系数分别下降了43.5%~47.9%与31.8%~35.2%。郭国业等[22]同样在铁皮石斛研究中发现,当使用MS+0.8%琼脂+3%蔗糖+0.1 mg/L NAA+1 mg/L 6-BA培养基,可使铁皮石斛芽增殖系数达到2.13,过高的6-BA虽能诱导较多的芽苗,但易导致诱导芽苗生长势变弱。
而本试验使用3 mg/L 6-BA仍可使芽诱导数及增殖系数保持较高水平,除了种质特性不同,还可能与本试验中在芽诱导阶段添加较高浓度香蕉泥有关。香蕉泥对粉佳人茎段芽增殖有促进作用,当香蕉泥浓度为100 g/L时,茎段芽增殖的平均生长速度系数、折干率最高。蔡正旺[23]在铁皮石斛的茎段培养中也得到类似结果。因此,较高浓度的6-BA,低浓度的NAA及较高浓度香蕉泥是保证茎段芽苗诱导与芽体质量的重要因素。
石斛组织培养中,与原球茎途径分化的芽相比,虽然茎段诱导的芽的生长势更强,根系更壮[24],但为了提高移栽成活率,仍需进行生根壮苗培养。在本试验中,不同水平的NAA、AC及香蕉泥对生根苗的合格率及折干率影响均不显著,可能是茎段诱导的芽对环境具备较强的适应能力。3个因素中,NAA虽然对粉佳人的增殖效果不明显,但在生根阶段,NAA是主要的影响因子,随着NAA浓度的升高,平均生根速度、平均合格苗的比率也升高。当NAA的浓度为1 mg/L,生根效果最优,植株健壮,根系发达。这与李媛等[25]在肿节石斛、尖刀唇石斛、报春石斛3种石斛的生根培养的结果略有不同,李媛等[25]的研究结果表明,NAA浓度为0.5~0.75 mg/L时,3种石斛的茎段生根苗可以达到最佳的生根诱导率与生长势,NAA的浓度为1 mg/L时,3种石斛的生根率、生根条数和根生长情况均有所下降。本研究中,粉佳人生根苗在NAA浓度为1 mg/L的处理7中,其生长速度系数达到12.82,折干率达到9.42%,远高于NAA为0.5 mg/L的3个处理。这可能与培养基中添加的AC有关。AC常用于多种植物组培苗生根培养,但本试验中,AC对粉佳人的生根及长势产生明显的抑制作用。这在其他兰科植物中也有类似的发现,刘丹[26]研究表明,低浓度或不添加AC的处理更适合金线莲生根。本研究也表明,不添加活性炭AC时,芽苗长势更优,生根苗的平均株高、平均叶长、平均根数、平均单株重高于AC 0.5 g/L和1 g/L浓度。
本研究利用三因素三水平正交试验,探讨植物生长调节剂(6-BA、NAA)和添加物(香蕉泥、AC)对石斛新品种粉佳人茎段不定芽增殖及小苗生根的影响,建立了高效的粉佳人种苗生产技术体系,可为石斛及其他兰科植物新品种工厂化育苗提供理论支持和技术参考。
