Bombax ceiba is a deciduous tree belonging to the Genus Bombax in the Malvaceae family. It holds significant importance as an ornamental plant of economic, medicinal and edible value in southern China. It is the city flower of Guangzhou and Panzhihua. The traditional propagation methods for B. ceiba are seed sowing and grafting. However, the seeds are difficult to preserve and easily deteriorate, leading to decreased germination rates. Grafting results in even lower survival rates. Therefore, this experiment aimed to study the tissue culture techniques for the rapid propagation of B. ceiba. In this study, B. ceiba seeds were used as the experimental materials, and different disinfection methods were tested to determine the most suitable method for seed disinfection, aiming to obtain sterile seedlings. Using the sterile seedlings as the explants, various basic culture media, combinations of shoot-inducing hormones, induction and proliferation media for apical and adventitious buds, as well as rooting media, were tested and optimized. Additionally, seedling acclimatization experiments were conducted to explore the optimal basic culture medium, hormone combination, induction and proliferation medium, rooting medium, and seedling acclimatization methods. The results showed that the optimal basic culture medium for B. ceiba tissue culture was MS medium supplemented with 30 g/L sucrose and 7.5 g/L agar. The best disinfection method for B. ceiba seeds was 75% ethanol treatment for 1 min, followed by five rinses with sterile water, 4-hour sterilization with a 50% H2O2 solution prepared from 30% H2O2, and another five rinses with sterile water. The optimal plant growth regulators for inducing differentiation of apical buds into young shoots were a combination of 6-benzylaminopurine (6-BA) and naphthaleneacetic acid (NAA). The best induction and proliferation medium was MS medium supplemented with 0.5 mg/L 6-BA, 0.5 mg/L indole-3-butyric acid (IBA), 30 g/L sucrose, and 7.5 g/L agar. The optimal rooting medium was half-strength MS medium supplemented with 0.5 mg/L IBA, 30 g/L sucrose, 7.5 g/L agar, and 0.2 g/L activated charcoal. The recommended substrate for seedling acclimatization was a mixture of sand, peat soil, and vermiculite in a ratio of 1∶1∶1. Through the optimization of seed disinfection methods, culture media selection, hormone combinations, and seedling acclimatization experiments, an efficient tissue culture system for B. ceiba was established. This would provide technical support for the in vitro preservation of B. ceiba seedlings, propagation of superior individuals, healthy seedling breeding, and the industrialized production of seedlings, and lay a foundation for the genetic transformation system of B. ceiba.

木棉是木棉科木棉属落叶乔木植物,是我国南方重要的观赏植物,是广州市和攀枝花市的市花,不仅具有较高的观赏价值,还具有有较高的经济价值、药用价值和食用价值。木棉传统的繁殖方式为播种和嫁接繁殖,其种子不易保存、易变质从而降低种子萌发率;而嫁接繁殖成活率更低。因此,本试验开展木棉组织培养快繁技术研究,以木棉种子为试验材料,通过开展对木棉种子不同消毒方法试验,确定最适合木棉种子的消毒方法,获得无菌种苗;并以无菌种苗为外植体,开展不同基础培养基、顶芽诱导激素组合、顶芽和丛生芽诱导和增殖培养基、生根培养基的筛选及炼苗试验,探索最佳基础培养基、激素组合、诱导培养基和增殖培养基、生根培养基及炼苗方法。结果表明:木棉组织培养最佳基础培养基为MS+蔗糖30 g/L+卡拉胶7.5 g/L;木棉种子最佳消毒方法为A2:75%乙醇消毒1 min→无菌水清洗5次→50%H2O2溶液消毒4 h→无菌水清洗5次;最佳木棉种子苗顶芽诱导分化幼芽的植物生长调节剂为6-BA+NAA的组合;最佳诱导及增殖培养基为C2:MS+6-BA 0.5 mg/L+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L;最佳生根培养基为E3:1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L;最佳炼苗基质为沙子∶泥炭土∶蛭石=1∶1∶1。通过对木棉种子消毒方法、培养基的筛选、激素组合及炼苗试验,对木棉组织培养技术进行优化,并建立了高效木棉组织培养技术体系,为木棉种苗离体保存、优良单株扩繁、健康种苗繁育及工厂化育苗提供技术支持,也可为木棉遗传转化体系奠定基础。

木棉是木棉科木棉属落叶乔木植物,是我国南方重要的观赏植物,是广州市和攀枝花市的市花,不仅具有较高的观赏价值,还具有有较高的经济价值、药用价值和食用价值。木棉传统的繁殖方式为播种和嫁接繁殖,其种子不易保存、易变质从而降低种子萌发率;而嫁接繁殖成活率更低。因此,本试验开展木棉组织培养快繁技术研究,以木棉种子为试验材料,通过开展对木棉种子不同消毒方法试验,确定最适合木棉种子的消毒方法,获得无菌种苗;并以无菌种苗为外植体,开展不同基础培养基、顶芽诱导激素组合、顶芽和丛生芽诱导和增殖培养基、生根培养基的筛选及炼苗试验,探索最佳基础培养基、激素组合、诱导培养基和增殖培养基、生根培养基及炼苗方法。结果表明:木棉组织培养最佳基础培养基为MS+蔗糖30 g/L+卡拉胶7.5 g/L;木棉种子最佳消毒方法为A2:75%乙醇消毒1 min→无菌水清洗5次→50%H2O2溶液消毒4 h→无菌水清洗5次;最佳木棉种子苗顶芽诱导分化幼芽的植物生长调节剂为6-BA+NAA的组合;最佳诱导及增殖培养基为C2:MS+6-BA 0.5 mg/L+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L;最佳生根培养基为E3:1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L;最佳炼苗基质为沙子∶泥炭土∶蛭石=1∶1∶1。通过对木棉种子消毒方法、培养基的筛选、激素组合及炼苗试验,对木棉组织培养技术进行优化,并建立了高效木棉组织培养技术体系,为木棉种苗离体保存、优良单株扩繁、健康种苗繁育及工厂化育苗提供技术支持,也可为木棉遗传转化体系奠定基础。

Bombax ceiba is a deciduous tree belonging to the Genus Bombax in the Malvaceae family. It holds significant importance as an ornamental plant of economic, medicinal and edible value in southern China. It is the city flower of Guangzhou and Panzhihua. The traditional propagation methods for B. ceiba are seed sowing and grafting. However, the seeds are difficult to preserve and easily deteriorate, leading to decreased germination rates. Grafting results in even lower survival rates. Therefore, this experiment aimed to study the tissue culture techniques for the rapid propagation of B. ceiba. In this study, B. ceiba seeds were used as the experimental materials, and different disinfection methods were tested to determine the most suitable method for seed disinfection, aiming to obtain sterile seedlings. Using the sterile seedlings as the explants, various basic culture media, combinations of shoot-inducing hormones, induction and proliferation media for apical and adventitious buds, as well as rooting media, were tested and optimized. Additionally, seedling acclimatization experiments were conducted to explore the optimal basic culture medium, hormone combination, induction and proliferation medium, rooting medium, and seedling acclimatization methods. The results showed that the optimal basic culture medium for B. ceiba tissue culture was MS medium supplemented with 30 g/L sucrose and 7.5 g/L agar. The best disinfection method for B. ceiba seeds was 75% ethanol treatment for 1 min, followed by five rinses with sterile water, 4-hour sterilization with a 50% H2O2 solution prepared from 30% H2O2, and another five rinses with sterile water. The optimal plant growth regulators for inducing differentiation of apical buds into young shoots were a combination of 6-benzylaminopurine (6-BA) and naphthaleneacetic acid (NAA). The best induction and proliferation medium was MS medium supplemented with 0.5 mg/L 6-BA, 0.5 mg/L indole-3-butyric acid (IBA), 30 g/L sucrose, and 7.5 g/L agar. The optimal rooting medium was half-strength MS medium supplemented with 0.5 mg/L IBA, 30 g/L sucrose, 7.5 g/L agar, and 0.2 g/L activated charcoal. The recommended substrate for seedling acclimatization was a mixture of sand, peat soil, and vermiculite in a ratio of 1∶1∶1. Through the optimization of seed disinfection methods, culture media selection, hormone combinations, and seedling acclimatization experiments, an efficient tissue culture system for B. ceiba was established. This would provide technical support for the in vitro preservation of B. ceiba seedlings, propagation of superior individuals, healthy seedling breeding, and the industrialized production of seedlings, and lay a foundation for the genetic transformation system of B. ceiba.

木棉(Bombax celibate)又名红花木棉、英雄树,为木棉科木棉属植物[1],原产中国南部、亚洲热带和澳大利亚地区[2]。木棉树体高大,树形优美,先花后叶,花朵硕大,花色鲜艳,春季开放,花期较长,具有较高的观赏价值,是我国南方地区重要的绿化树种[3-4]。木棉观赏价值极高,而且在适宜的环境条件下生长速度快,是一种难得的生态林、景观林和经济林,是园林绿化行道树的良好树种,在我国已有2000多年的栽培历史[5]
木棉花和种子都具有较高的经济价值,木棉花和树皮均可入药,木棉花入药清热除湿,树皮可作为滋补类药物入药,木质较轻,木材常加工为蒸笼、船上用品、火柴梗或纸张等使用。木棉种子中含有20%~25%的油分,榨取其中的油分可以制成肥皂,榨油剩余的材料可作为肥料或家畜饲料使用。木棉果实内部充满棉絮,其棉絮可做枕头、棉被的填充物,同时木棉棉絮也是救生圈的优良填充材料[6]
木棉传统的繁殖方式是播种和扦插[7],但由于种子含油量高,易变坏丧失萌发力,萌发率低,一般要求采后当年及时播种,扦插成活率低,且播种繁殖不能保持母本的优良性状,木棉组织培养育苗,相对于传统的种子繁殖或扦插繁殖,组织培养育苗更利于实现木棉的工程化育苗。前人对木棉植物的研究比较少,主要集中在生理生化、木棉种质资源调查与评价方面[8-12],有关组培研究方面的报道不多。本研究以木棉种子为材料,无菌种苗为外植体,通过消毒方法、基础培养基、诱导激素组合、诱导与增殖培养基、生根培养基的筛选及炼苗试验,建立高效木棉组培快繁技术体系,为木棉种苗离体保存、优良单株扩繁及健康种苗繁育及工厂化育苗提供技术支持。
本试验材料所用木棉种子采摘于昌江县王下乡木棉树。
将饱满无破损的木棉种子放置于烧杯中,用洗衣粉水浸泡10 min,流水冲洗30 min,置于无菌操作台用无菌水洗3遍,再用75%乙醇消毒1 min,无菌水冲洗5次,消毒剂选择30%H2O2,配成50%溶液浸泡3、4、5h;4%NaClO溶液浸泡20、40、60 min,最后用无菌水冲洗5次(表1),用H2O2和NaClO不同消毒处理方法进行消毒,最后用无菌水冲洗5次,放在无菌滤纸上吸干种子表面水分,待接种。
以MS为基本培养基,附加蔗糖MS+蔗糖30 g/L+卡拉胶7.5 g/L和B5+蔗糖30 g/L+卡拉胶7.5 g/L中,pH 5.9。每个处理接种20瓶,每瓶3粒种子,重复3次。初始暗培养,露白后进行光照处理,温度25 ℃、光照强度3000 lx条件下培养室中进行培养,接种第3天和第7天观察并记录污染率,第7天记录种子萌发率,筛选最佳消毒方法。
木棉种子苗接种7 d左右开始露白,15 d长成试验材料需要的无菌种苗,将木棉无菌种子苗顶芽为诱导材料,接种到添加不同激素的MS培养基中(表2),MS为对照培养基,每种培养基接种10瓶,每瓶接种5个外植体,置于25~28 ℃条件下暗培养一周,转置LED灯光照10~12 h/d,光照强度1000~1200 lx,25~28 ℃条件下培养,25 d后统计诱导率。
切取木棉顶芽诱导的再生芽带芽茎段,接入到不同增殖培养基中(表3),每种培养基接种10瓶,每瓶接种5个外植体,置于LED照明灯光照10~12 h/d,光照强度1000~1200 lx,温度25~28 ℃;25 d后统计结果。
切取木棉顶芽诱导的不定芽带芽茎段,接入到不同基础培养基中(表4),每种培养基接种10瓶,每瓶接种5个外植体,置于LED照明灯光照10~12 h/d,光照强度1000~1200 lx,温度25~28 ℃;25 d后统计结果。
选择生长一致、健壮的丛生芽苗切取单株接种到不同生根培养基中(表5),每个处理接种10瓶,每瓶接种2株,置于26~28 ℃条件下暗培养7 d,再置于LED照明灯光照16 h/d,光照强度1000~1200 lx,温度26~28 ℃条件下培养,30 d后统计生根率。
挑选高于3~5 cm、叶色较绿、根系发达的木棉组培苗,从组织培养室移至实验室或温室大棚,瓶内炼苗7 d;然后将瓶盖揭开1/3,放置3 d;最后将瓶口完全揭开放置3 d后,用镊子将苗轻轻取出,洗净培养基,将洗好的苗移栽至不同炼苗基质种植(表6),移至光照培养箱培养,光照时间12 h/d,炼苗过程中,保证基质水分充足,每天喷水2~3次。
表7可知,接种第3天对照(CK)污染率达到80%,第7天污染率为100%,未污染萌发率为0;处理A1第3天污染率为10%,第7天污染率为30%,未污染萌发率为87%;处理A2第3天污染率为8%,第7天污染率为20%,未污染萌发率为85%;处理A3第3天污染率为0,第7天污染率为19%,未污染萌发率为70%;处理A4第3天污染率为75%,第7天污染率为95%,未污染萌发率为60%;处理A5第3天污染率为60%,第7天污染率为75%,未污染萌发率为50%;处理A6第3天污染率为60%,第7天污染率为70%,未污染萌发率为20%;综合污染率和种子萌发率最适合作为木棉种子的消毒剂是H2O2,NaClO作为消毒剂不但污染率高,同时也影响种子的萌发,CK的污染率高达100%,因此,经过多种试验结果表明,木棉种子消毒最佳方法为A2:75%污水乙醇消毒1 min→无菌水清洗5次→H2O2∶水=1∶1消毒4 h→无菌水清洗5次,第7天污染率为20%,未污染萌发率为85%。
表8可知,培养基B3:MS+6-BA 1.0 mg/L+ IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L植物生长调节剂6-BA+IBA较适合作为木棉种子苗顶芽诱导的激素,分化幼芽3个以上,幼芽生长健壮、长势好,植株和叶片颜色显绿(图1D);对照MS不分化幼芽(图1A);培养基B1:MS+6-BA 1.0 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L中只添加6-BA一种植物生长调节剂分化芽少,发黄,芽短(图1B);培养基B2:MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L中能分化1~3个芽,芽长势弱(图1C);培养基B4:MS+6-BA 1.0 mg/L+IAA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L有愈伤,分化幼芽少,难形成苗(图1E);培养基B5:MS+6-BA 1.0 mg/L+2,4-D 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L形成大量愈伤,不分化芽,适合作为诱导愈伤培养基(图1F)。因此,较适合作为木棉种子苗顶芽诱导分化幼芽的植物生长调节剂为6-BA+NAA的组合。
表9可知,木棉幼芽诱导率随激素6-BA浓度的增加而提高,增殖系数也提高,但当浓度过高愈伤增加影响了幼芽的生长,诱导率和增殖系数反而下降,最适合木棉幼芽增殖的培养基为C2:MS+6-BA 1.0 mg/L+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L,诱导率为97%,增殖系数为5.5。
表10可知,MS为培养基更适合木棉幼芽的诱导,诱导幼芽数量多,丛芽健壮、偏绿、叶片展开(图2A);木棉幼芽在B5培养基单芽多、2个芽较少,植株高叶片展开,但叶片枯黄容易脱落(图2B),不适合作为木棉诱导基础培养基。
表11可知,木棉组培苗在对照CK:1/2 MS生根培养基中不长根;在E1:1/2 MS+NAA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+0.2 g/L活性炭培养基中生根率65%,生根条数极少,平均条数0.53条、叶片黄、苗弱不易成活;在E2:1/2 MS+ NAA 1.0 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L中生根率85%,根数少,平均根数0.63条,叶片黄、苗弱不易成活;在E3:1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L培养基中生根率达95%;平均根数3.15,叶片偏绿、苗健壮;在E4:1/2 MS+IBA 1.0 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L培养基中生根率达85%;平均根数2.05,叶片偏绿、苗健壮。因此,较适合作为木棉组培苗生根的激素为IBA,生根率高,平均根条数也多,叶片偏绿,苗健壮;而NAA对木棉组培苗也有根长出,但是极少,且叶片黄苗弱。较合适作为木棉组培苗的生根培养基为E3:1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L。
表12可知,沙子保水性不好、营养不足,不利于木棉组培苗炼苗;泥炭土湿度较高、易积水、苗易烂根,影响木棉组培苗成活率;蛭石太轻,组培苗飘浮起来、苗不易固定、泡水死亡;基质沙子∶泥炭土∶蛭石=1∶1∶1配比炼苗成活率达到87%,保水性好、不易积水、组培苗不烂根、苗健壮。因此,较适合作为木棉组培苗炼苗基质为沙子∶泥炭土∶蛭石=1∶1∶1。
在讨论不同消毒剂及消毒时间对木棉种子消毒及萌发的影响时,CK采用75%乙醇消毒1 min,用无菌水清洗5次接种到MS培养基中,第7天污染率达到100%,污染率高;采用H2O2作为消毒剂,消毒不同时间第7天污染率为19%~30%,未污染萌发率为70%~87%;采用NaClO作为消毒剂,消毒不同时间第7天污染率为70%~95%,未污染萌发率为20%~60%。可知,消毒剂H2O2较适合作为木棉种子消毒剂,以H2O2为消毒剂,开展3个不同时间处理试验,处理A1和A2的污染率差别不大,但是A2消毒方法未污染种子的萌发率较高,因此,筛选出较适合的消毒方法为A2:75%污水乙醇消毒1 min→无菌水清洗5次→H2O2∶水=1∶1消毒4 h→无菌水清洗5次,第7天污染率为20%,未污染萌发率为85%。
在筛选适合作为木棉组培基础培养基的过程中,发现B5培养基不合适作为木棉组培基础培养基,B5作为基础培养基芽分化少,芽弱,顶端发黄死亡;MS作为基础培养基木棉分化幼芽多、健壮偏绿,因此,MS适合作为木棉组培基础培养基。
在筛选适合木棉幼芽诱导和增殖培养基激素的过程中,综合彭洪杏等[13]和刘娟旭等[14]的研究成果,制定不同的激素及浓度相结合的培养基,筛选出较适合作为木棉幼芽分化和增殖的激素是6-BA和IBA相结合,较合适的培养基为C2:MS+6-BA 1.0 mg/L+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L,诱导率97%,增殖系数5.5。
组培苗生根是植物组织培养技术的重要环节之一,是决定植物组织培养是否成功的关键因素,也是植物组培快繁体系中的关键环节[15],特别是木本植物,组培苗的生根难度大,根容易膨化,影响根的形成。本试验以1/2 MS为生根基础培养基,开展NAA和IBA两种激素不同浓度的4种生根培养基,结果可知,E3:1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+卡拉胶7.5 g/L+活性炭0.2 g/L较合适作为木棉组培苗生根培养基,但是木棉组培苗的生根数量并不多,生根率可达到95%,但是平均根数量3.15,在今后的实验过程中可以考虑添加椰子水、香蕉等附加物结合IBA设置更多处理组,更加细化不同组合培养基对木棉组培苗生根的影响。
生根苗炼苗后开展不同基质及配比对组培苗炼苗成活率的影响,由于木棉组培苗生根数量不多,有可能影响了炼苗的成活率,成活率最高只达到87%,较佳的炼苗基质配比为沙子∶泥炭土∶蛭石=1∶1∶1,成活率为87%。
