Dendrobium are famous for the ornamental and medicinal value. In order to explore the volatile components in the flowers of Dendrobium in Sect. Holochrysa and Sect. Chrysotoxae Kraenzl., the volatile components in the flowers of five Dendrobium in Sect. Holochrysa and seven Dendrobium in Sect. Chrysotoxae Kraenzl. were extracted by solid phase micro extraction (SPME) and analyzed via gas chromatography-mass spectrometry (GC-MS). Results showed that 74 volatile components were identified from the flowers of the species, 47 of which from D. hancockii in Sect. Holochrysa were the main volatile components; Secondly, there were 37 and 35 volatile components from D. chrysotoxum and D. harveyanum in the Sect. Chrysotoxae Kraenzl., respectively. The least component species was D. farmeri in the Sect. Chrysotoxae Kraenzl. with only 18 volatile components. Most of the species of volatile substances in Dendrobium were more than 20, and most of which were alkenes, except D. kenkinsii and D. densiflorum in the Sect. Chrysotoxae Kraenzl. mainly contained aldehydes. Cluster analysis of volatile ingredients showed that all the Dendrobium flowers were clustered together except a few Dendrobium plants were not clustered in the original grouping, which indicated that the volatile substances in Dendrobium flowers could be used as a reference for Dendrobium grouping. This study would provide references for the breeding, resource exploration and utilization of Dendrobium, and exert significant value on the grouping of Dendrobium.

石斛是著名的观赏和药用植物,为探讨石斛属寡花组和顶叶组石斛物种花朵中的挥发性成分,采用固相微萃取(SPME)法和气相色谱-质谱联用(GC-MS)技术,对5种寡花组和7种顶叶组石斛植物盛花期花朵的挥发性成分进行测定和分析。结果表明:供试的12种石斛属植物花朵中共鉴定出挥发性成分74种,其中烯类36种、醛类13种、醇类12种、酯类3种、酮类5种、烷烃类3种、芳香族类2种;挥发性物质种类较多的是寡花组中的竹叶石斛,有47种,其次是顶叶组中的鼓槌石斛和苏瓣石斛,分别有37种和35种,最少的是顶叶组中的黄法式石斛,仅有18种,大多石斛的挥发性物质种类均在20种以上,并且大多以烯类化合物为主,仅有顶叶组中的小黄花石斛和密花石斛以醛类化合物为主。对挥发性成分进行聚类分析,发现除了少数石斛未聚到原先的分组外,其余的均聚到一起,说明石斛花朵挥发性物质可以作为石斛分组的依据。研究结果可作为顶叶组石斛植物品种选育、资源开发和利用提供一定的参考依据,对石斛属植物分组具有借鉴意义。

石斛是著名的观赏和药用植物,为探讨石斛属寡花组和顶叶组石斛物种花朵中的挥发性成分,采用固相微萃取(SPME)法和气相色谱-质谱联用(GC-MS)技术,对5种寡花组和7种顶叶组石斛植物盛花期花朵的挥发性成分进行测定和分析。结果表明:供试的12种石斛属植物花朵中共鉴定出挥发性成分74种,其中烯类36种、醛类13种、醇类12种、酯类3种、酮类5种、烷烃类3种、芳香族类2种;挥发性物质种类较多的是寡花组中的竹叶石斛,有47种,其次是顶叶组中的鼓槌石斛和苏瓣石斛,分别有37种和35种,最少的是顶叶组中的黄法式石斛,仅有18种,大多石斛的挥发性物质种类均在20种以上,并且大多以烯类化合物为主,仅有顶叶组中的小黄花石斛和密花石斛以醛类化合物为主。对挥发性成分进行聚类分析,发现除了少数石斛未聚到原先的分组外,其余的均聚到一起,说明石斛花朵挥发性物质可以作为石斛分组的依据。研究结果可作为顶叶组石斛植物品种选育、资源开发和利用提供一定的参考依据,对石斛属植物分组具有借鉴意义。

Dendrobium are famous for the ornamental and medicinal value. In order to explore the volatile components in the flowers of Dendrobium in Sect. Holochrysa and Sect. Chrysotoxae Kraenzl., the volatile components in the flowers of five Dendrobium in Sect. Holochrysa and seven Dendrobium in Sect. Chrysotoxae Kraenzl. were extracted by solid phase micro extraction (SPME) and analyzed via gas chromatography-mass spectrometry (GC-MS). Results showed that 74 volatile components were identified from the flowers of the species, 47 of which from D. hancockii in Sect. Holochrysa were the main volatile components; Secondly, there were 37 and 35 volatile components from D. chrysotoxum and D. harveyanum in the Sect. Chrysotoxae Kraenzl., respectively. The least component species was D. farmeri in the Sect. Chrysotoxae Kraenzl. with only 18 volatile components. Most of the species of volatile substances in Dendrobium were more than 20, and most of which were alkenes, except D. kenkinsii and D. densiflorum in the Sect. Chrysotoxae Kraenzl. mainly contained aldehydes. Cluster analysis of volatile ingredients showed that all the Dendrobium flowers were clustered together except a few Dendrobium plants were not clustered in the original grouping, which indicated that the volatile substances in Dendrobium flowers could be used as a reference for Dendrobium grouping. This study would provide references for the breeding, resource exploration and utilization of Dendrobium, and exert significant value on the grouping of Dendrobium.

兰科(Orchidaceae)石斛属(Dendrobium SW.)植物具有重要的观赏价值和药用价值,是世界著名的观赏和药用植物,所有中国产石斛属植物均列入《中国国家重点保护野生植物名录》[1]。花香是由植物花朵散发出来具有香气的物质[2]。不同植物的花香都是由不同的化学成分组成,即使化学成分只有细微差别,所产生的气味也是千差万别,浓淡不一。植物的花香不仅对植物本身具有重要意义,如吸引昆虫等传粉者帮助植物繁殖、驱赶天敌自我保护、抵御各种非生物胁迫等,同时也是植物重要观赏性状之一[3]。植物花香具有可调节人的心理、生理机能,改变人的精神状态,杀菌驱虫、净化空气等功效,如茉莉[Jasminum sambac(L.)Aiton]花香可使人凝神静气、理气解郁、平静心态;天竺葵(Pelargonium hortorum Bailey)花香有平复神经、消除疲劳、促进睡眠的功效[4]
香气成分是衡量石斛药用价值和观赏价值的重要标准之一,选育花色和花香俱佳的石斛品种的趋势日益凸显,所以有必要挖掘出石斛中的芳香种质资源。目前,国内已有学者对石斛花的香气成分进行了相关研究。郝瑞杰[5]认为,上千种石斛的花香化合物中,主要花香化合物有12种,其中包含柠檬烯、罗勒烯、月桂烯、芳樟烯、β-蒎烯、α-蒎烯、石竹烯等。李崇晖等[6]采用固相微萃取(SPME)法萃取4种石斛花朵的挥发性成分,结果表明,鼓槌石斛(D. chrysotoxum)和细叶石斛(D. hancockii)的主要挥发性成分是3-蒈烯;罗河石斛(D. lohohense)的主要香气成分为水杨酸甲酯和D-柠檬烯;密花石斛(D. densiflorum)的主要香气成分为2-亚甲基-4,8,8-三甲基-4-乙烯基-双环[5.2.0]壬烷和α-法尼烯。吕素华等[7]研究发现,壬醛、α-蒎烯是铁皮石斛(D. officinale)鲜花的主要香气成分。黄昕蕾等[8]分析了鼓槌石斛(D. chrysotoxum)鲜花的香气,发现影响鼓槌石斛香气的主要物质为β-罗勒烯、α-蒎烯和苯乙醛。袁明焱等[9]分析发现美花石斛(D. loddigesii Rolfe)鲜花的主要成分为乙酸2-乙基己酯(35.29%)、1,3,6-辛三烯(20.51%)、L-芳樟醇(4.31%)、L-柠檬烯(3.00%)。仇硕等[10]分析了细茎石斛(D. moniliforme L. Sw.)鲜花的香气成分,α-蒎烯的相对含量到达27%以上。由此可以看出,萜类物质是石斛属花朵的主要香气物质种类,烷类和醇类化合物也是石斛花朵香气成分的重要组成成分[9-11],常见的有(E)-氧化芳樟醇、芳樟醇、马鞭草烯醇等,其中,芳樟醇普遍存在于大多数石斛花朵中且为主要的赋香成分[11-13]
目前有关寡花组和顶叶组石斛品种花朵挥发性成分描述和收录的文献还较欠缺。本研究以5种寡花组和7种顶叶组石斛植物花朵为研究材料,利用气相色谱-质谱联用(GC-MS)技术对其盛花期花朵的挥发性成分进行分析,确定花朵挥发性成分并计算其相对含量;初步分析这12种石斛花朵挥发性成分的共性和特性,并进行聚类分析。研究结果为石斛植物资源评价、品种选育、花香气代谢产物研究和药效成分分析提供参考。
供试石斛样品均采自中国热带农业科学院热带作物品种资源研究所热带花卉资源圃,经黄明忠副研究员鉴定,具体信息见表1,花的形态见图1
每种石斛选取3株长势良好、无病虫害的植株作为采集对象,于上午11:00—11:30采集盛花期开放2~3 d,具有香气的生长良好、无病虫害的新鲜花朵,混样,液氮速冻保存于–80 ℃超低温冰箱中。将混样剪碎置于20 mL螺纹口透明顶空瓶中,用聚四氟乙烯硅胶隔垫密封。先将65 μm PDMS-DVB(SupelcoCo.)于250 ℃老化25 min,样品置于45 ℃平衡15 min,吸附15 min,萃取头取出立即插入GC进样口,解析附1 min,进行GC-MS测定。
色谱条件:利用Agilent 7810B-5977BMSD GC-MS联用仪(美国安捷伦公司),色谱柱为DB-5毛细管柱(30 m×0.25 mm×0.25 μm);载气为高纯度氦气,流速为1.0 mL/min。升温程序从40 ℃开始,3 ℃/min升温至100 ℃,然后在5 ℃/min升温至245 ℃,保持1 min,然后280 ℃运行3 min。
质谱条件:电离模式EI,离子源温度为230 ℃,电子能量为70 eV,传输线温度为250 ℃。质谱以1 s的间隔在30~350 amu离子质荷比(m/z)范围内扫描。
通过NIST2.0标准谱库对GC-MS原始数据进行检索,鉴定成分并按峰面积归一化计算各成分百分含量。质谱图库检索匹配度≥90%的化合物,通过CAS号查询化合物名称,并结合相关文献确定石斛属花朵的挥发性成分。采用Microsoft Excel 2020软件对试验数据进行整理分析,采用Origin 2023软件进行聚类分析。
依据GC-MS条件对供试石斛品种进行挥发性成分测定,依次得到12个石斛品种挥发性成分的总离子流图(图2)。对总离子流图中各信号峰经质谱数据系统检索比对,确定挥发性组分中的化合物名称,并用峰面积归一化法进行定量分析,计算出化合物相对含量。
对石斛属5个寡花组和7个顶叶组不同品种的盛花期花朵挥发性成分进行鉴定,共检测出74种挥发性物质,其中,烯类36种、醛类13种、醇类12种、酯类3种、酮类5种、烷烃类3种、芳香族类2种(表2)。研究结果表明,石斛属花朵中的挥发性物质组成丰富,并且不同品种的石斛花朵挥发性物质组成存在较大差异。供试的12种石斛属花朵挥发性物质种类较多的是寡花组中的竹叶石斛,有47种;其次是顶叶组中的鼓槌石斛和苏瓣石斛,分别有37种和35种,最少的是顶叶组中的黄法式石斛,仅有18种;其余石斛的挥发性物质种类均在20种以上。
供试石斛属花朵中的挥发性物质大多以烯类化合物为主,仅有顶叶组中的小黄花石斛和密花石斛以醛类化合物为主。其中竹叶石斛中的烯类化合物最多,有24种,苏瓣石斛次之,有21种,短棒石斛和鼓槌石斛均达16种,有4种石斛的烯类化合物少于10种,最少的是小黄花石斛和密花石斛,仅有7种。供试石斛属花朵的挥发性物质中,醛类化合物的种类仅次于烯类,不同种间差异不大,大多均有10种左右,其中黄法式石斛的醛类化合物最少,仅有5种。醇类、酯类和酮类化合物在12种石斛属花朵中普遍存在,醇类化合物在竹叶石斛和流苏石斛中种类最多,达到7种,黄法式石斛和密花石斛最少,仅有1种;酯类和酮类化合物在供试12种石斛中的种类均较少,均少于5种;此外,还在少数石斛中检测到烷烃类和芳香族类化合物,其中竹叶石斛中含有这2类物质(表2)。
通过比较分析发现,供试的12种石斛属种质资源花朵中检测到的挥发性物质组成差异较大,共有成分有7种,分别为月桂烯、3-乙基-2-甲基-1,3-己二烯、反式-2-戊烯醛、反式-2-己烯醛、反式-2-辛烯醛、丙烯酸异冰片酯和1-戊烯-3-酮。除了2组石斛所含有的共有成分外,寡花组石斛中的共有成分有(+)-α-蒎烯、桧烯、3-乙基-2-甲基-1,3-己二烯、β-石竹烯、正己醛和(E)-2-庚烯醛;顶叶组石斛中的共有成分有正己醇(表3)。
进一步分析发现,月桂烯、(+)-3-蒈烯、D-柠檬烯、罗勒烯、3-乙基-2-甲基-1,3-己二烯、β-石竹烯和α-芹子烯是主要的烯类化合物,大多香气阈值均较低,也是典型的花香成分。正己醛、庚醛和反式-2-己烯醛是主要的醛类化合物,而醇类化合物以正己醇为主(表3)。
挥发性物质组成及含量的差异造成了香气的差异。研究结果表明,供试的12种石斛中烯类和醛类含量均占绝对优势,2类成分的总含量均超过60%。在总挥发性物质的占比中,除小黄花石斛和密花石斛的醛类化合物的占比高于烯类化合物外,其他石斛都是烯类化合物占比较高,苏瓣石斛的烯类化合物占比显著高于其他石斛,占60%,其次为竹叶石斛、短棒石斛和黄法式石斛,分别占51%、50%、50%,小黄花石斛最少,仅占28%。醛类化合物在小黄花石斛和密花石斛中占比最高,为其主要的花香成分,分别占40%和48%,在竹叶石斛中最少,只占21%。醇类化合物是导致香气类型差异的重要部分,在供试的12种石斛中普遍存在,在流苏石斛中占比最大,占25%,在黄法式石斛、聚石斛和密花石斛中占比最少,分别占6%、7%和5%。酯类化合物和酮类化合物在石斛中的占比均不高,酯类化合物还具有宜人的清香,是石斛属花朵挥发性物质的组成部分,二者中的占比均不超过10%。烷烃类化合物只在竹叶石斛、唐氏石斛、红灯笼石斛和小黄花石斛4种石斛中检测到,在唐氏石斛中占比最高,达10%,其他石斛中均只有4%。芳香族类化合物在竹叶石斛、黄法式石斛、鼓槌石斛和聚石斛4种石斛中检测到。竹叶石斛是唯一同时检测到烷烃类和芳香族类化合物的石斛(图3)。
利用Origin软件对12种石斛属植物的花朵香气成分进行聚类分析,热图可对GC-MS分析得到的挥发性物质成分数据进行可视化处理,颜色由蓝色到红色代表化合物含量依次增加,颜色越深,物质含量越高。采用Z分数[标准化为N(0,1)]方法对数据进行归一化处理,从聚类分析热图可以看出,12种石斛属植物聚成3个类群,竹叶石斛、流苏石斛和苏瓣石斛为1个类群,短棒石斛、黄花石斛、苏瓣石斛、红灯笼石斛、黄法式石斛、聚石斛、小黄花石斛和密花石斛聚为1个类群,该类群又分为2个小类群,唐氏石斛、短棒石斛、黄花石斛和红灯笼石斛为1个小类群,黄法式石斛、聚石斛、小黄花石斛和密花石斛聚为1个小类群,而鼓槌石斛单独成1个类群。可以看出寡花组的5种石斛全聚为一类,顶叶组石斛中的5种石斛又聚为一类,说明根据挥发性物质的种类也可作为石斛分组的参考依据(图4)。
花朵中的香气成分是植物花朵释放的次生代谢产物,是由各种挥发性香气组分共同作用形成的,具有较高香气值的成分可作为花的特征香气[14]。已有研究表明,双色茉莉不同原生种及品种之间花香成分的类型一致,萜烯类化合物是最主要的赋香成分[15-16]。本研究的12种石斛花朵中烯类化合物在其总挥发性物质中均占有较高比例,主要的烯类化合物有D-柠檬烯、桧烯、(+)-α-蒎烯、α-侧柏烯、β-石竹烯、罗勒烯、γ-松油烯、α-律草烯、月桂烯、毕澄茄烯等。本研究首次测得唐氏石斛、短棒石斛、黄花石斛、苏瓣石斛和小黄花石斛花朵中的挥发性物质,这5种石斛的主要花香成分均含有(+)-α-蒎烯、月桂烯、反式-2-己烯醛和1-戊烯-3-酮。
夏科等[17]研究发现,流苏石斛的香气成分含量最高的是罗勒烯,杨晓蓓等[18]研究发现,流苏石斛香气成分含量最高的是α-蒎烯和β-蒎烯,以上研究的供试材料分别来自广西和云南。本研究检测出流苏石斛中香气成分含量最高的是(+)-α-蒎烯、正己醛和桧烯,未检测到罗勒烯,本研究的流苏石斛来自云南,推测同种石斛的主要赋香成分可能由于石斛花朵的采集时间、地点及生长气候等条件的不同而有差异。王元成等[19]发现黄法式石斛的主要香气成分为芳樟醇、罗勒烯、β-石竹烯,而本研究发现黄法式石斛的主要赋香成分是(+)-α-蒎烯、月桂烯和(+)-3-蒈烯,未检测到罗勒烯和芳樟醇,其成分和相对含量差异较大。丁灵等[20]研究分析了7种不同花色密花石斛的挥发性成分,与本研究中密花石斛花朵的主要挥发性成分的组成和含量明显不同。仇硕等[10]分析测定了3种不同花色聚石斛花朵的挥发性成分,与本研究中的黄法式石斛和密花石斛花朵的主要挥发性化合物种类数量接近,但在含量上均有差异。红灯笼石斛的主要赋香成分是月桂烯、β-石竹烯、α-芹子烯,颜沛沛等[21]研究发现,红灯笼石斛中香气成分含量最高的成分是乙酸辛酯,其余成分也极其相似,而本研究中未检测出乙酸辛酯。李崇晖等[6]在引种于云南的鼓槌石斛中未检测到顺式- β-罗勒烯,含量最高的为3-蒈烯,而夏科等[17]通过检测发现,广西的鼓槌石斛中顺式-β-罗勒烯含量较高,未检测到3-蒈烯,本研究种也未检测到3-蒈烯,推测是不同地域、不同环境等因素造成石斛挥发性成分的不同。
王文静等[22]研究发现,不同种类石斛含有的赋香成分的组成和含量不同,从而导致香气的差异性。蒎烯具有松木香味,蒎烯中含量最高的α-蒎烯是合成香料的重要原料[22]。蒎烯、(-)-α-蒎烯和β-蒎烯具有松萜特有的强烈松木、针叶及树脂样气息[23]。本研究中,(+)-α-蒎烯在除红灯笼石斛和密花石斛外,在其余10种石斛花朵中均测得其成分,而(-)-α-蒎烯在小黄花石斛以及黄法式石斛中的相对含量较高,2-蒎烯则为鼓槌石斛的重要赋香成分。月桂烯,又称香叶烯,具有令人愉悦的甜橘味和清淡的香脂气味,是合成香茅醛、薄荷醇和紫丁香醇等香料香精的重要原料和有机中间体[24],本研究中的12种石斛均含有相对含量较高的月桂烯,说明月桂烯是石斛属花朵的主要香气成分,可用于提取精油或制成香水。具有类似甜杏、坚果香气的庚醛[25]在除黄花石斛和黄法式石斛外的其余石斛中均检测到。正己醛呈青草清香及苹果香味[26],在供试所有石斛中均存在,相对含量最高达46.35%。研究结果可为培育石斛属芳香品种及其精油的开发应用提供一定的理论参考。
