An antimicrobial peptide (SAMP) derived from Microcitrus australasica can not only reduce the concentration of Candidatus Liberibacter asiaticus (CLas) in the infected trees and reduce the symptoms of the diseased trees, but also induce innate immunity and prevent the infection of Huanglongbing in healthy trees that are not infected with Huanglongbing. MaSAMP was analyzed by functional structure, subcellular localization and phylogeny. The results showed that the open reading frame (ORF) of MaSAMP was 201 bp in length, encoding 67 amino acids. The molecular weight of the corresponding protein was 7.59959 kDa, the theoretical isoelectric point was 5.38, and the instability index (Instability index (II)) was 33.22. The aliphatic index was 87.16%, and the grand average of hydropathicity was -0.09, indicating that the protein was a hydrophilic one. The protein contained one Dabb domain and was predominantly structured by α-helices and random coils. In terms of evolution, MaSAMP was phylogenetically close to the stress-response A/B barrel domain-containing protein CISIN_1g033887mg from Citrus sinensis and the A/B barrel domain protein HS1 from Citrus×clementina, with protein similarity of 97.01% and 91.04%, respectively. Subcellular localization results indicated that MaSAMP was localized in chloroplasts. Furthermore, through homologous recombination, a pCAMBIA1300-35S::MaSAMP-EGFP overexpression vector fused with an EGFP fluorescent tag was successfully constructed, and transgenic citrus plants were obtained using agrobacterium-mediated in planta transformation technology. This study would provide a material basis for subsequent research on the biological functions of MaSAMP.

来源于澳指檬(Microcitrus australasica)的一种抗菌肽(stable antimicrobial peptide,SAMP),不仅可以降低已感染柑橘黄龙病的病树体内的黄龙病菌(Candidatus Liberibacter asiaticus,CLas)浓度、减轻病树症状,而且还可以诱导未感染黄龙病的健康树先天免疫、阻止感染黄龙病。本研究对MaSAMP的功能结构、亚细胞定位、系统发育等进行分析,结果表明,MaSAMP的开放阅读框(open reading frame,ORF)全长为201 bp,编码67个氨基酸,编码的蛋白分子量为7.59959 kDa,理论等电点为5.38,不稳定指数[instability index(II)]为33.22,脂肪族氨基酸系数(aliphatic index)为87.16%,平均亲水系数(grand average of hydropathicity)为–0.091,属于亲水性蛋白,含有1个Dabb结构域,MaSAMP蛋白结构以α-螺旋、无规则卷曲为主。MaSAMP与甜橙胁迫响应A/B桶状结构域蛋白CISIN_1g033887mg、克里曼丁橘A/B桶状结构域蛋白HS1亲缘关系较近,蛋白相似度分别为97.01%、91.04%。亚细胞定位结果显示,MaSAMP定位于叶绿体。进一步通过同源重组成功构建了融合EGFP荧光标签的pCAMBIA1300-35S::MaSAMP-EGFP过表达载体,并借助农杆菌介导的原位转化技术获得柑橘转基因植株。本研究为后续MaSAMP的生物学功能研究提供材料基础。

来源于澳指檬(Microcitrus australasica)的一种抗菌肽(stable antimicrobial peptide,SAMP),不仅可以降低已感染柑橘黄龙病的病树体内的黄龙病菌(Candidatus Liberibacter asiaticus,CLas)浓度、减轻病树症状,而且还可以诱导未感染黄龙病的健康树先天免疫、阻止感染黄龙病。本研究对MaSAMP的功能结构、亚细胞定位、系统发育等进行分析,结果表明,MaSAMP的开放阅读框(open reading frame,ORF)全长为201 bp,编码67个氨基酸,编码的蛋白分子量为7.59959 kDa,理论等电点为5.38,不稳定指数[instability index(II)]为33.22,脂肪族氨基酸系数(aliphatic index)为87.16%,平均亲水系数(grand average of hydropathicity)为–0.091,属于亲水性蛋白,含有1个Dabb结构域,MaSAMP蛋白结构以α-螺旋、无规则卷曲为主。MaSAMP与甜橙胁迫响应A/B桶状结构域蛋白CISIN_1g033887mg、克里曼丁橘A/B桶状结构域蛋白HS1亲缘关系较近,蛋白相似度分别为97.01%、91.04%。亚细胞定位结果显示,MaSAMP定位于叶绿体。进一步通过同源重组成功构建了融合EGFP荧光标签的pCAMBIA1300-35S::MaSAMP-EGFP过表达载体,并借助农杆菌介导的原位转化技术获得柑橘转基因植株。本研究为后续MaSAMP的生物学功能研究提供材料基础。

An antimicrobial peptide (SAMP) derived from Microcitrus australasica can not only reduce the concentration of Candidatus Liberibacter asiaticus (CLas) in the infected trees and reduce the symptoms of the diseased trees, but also induce innate immunity and prevent the infection of Huanglongbing in healthy trees that are not infected with Huanglongbing. MaSAMP was analyzed by functional structure, subcellular localization and phylogeny. The results showed that the open reading frame (ORF) of MaSAMP was 201 bp in length, encoding 67 amino acids. The molecular weight of the corresponding protein was 7.59959 kDa, the theoretical isoelectric point was 5.38, and the instability index (Instability index (II)) was 33.22. The aliphatic index was 87.16%, and the grand average of hydropathicity was -0.09, indicating that the protein was a hydrophilic one. The protein contained one Dabb domain and was predominantly structured by α-helices and random coils. In terms of evolution, MaSAMP was phylogenetically close to the stress-response A/B barrel domain-containing protein CISIN_1g033887mg from Citrus sinensis and the A/B barrel domain protein HS1 from Citrus×clementina, with protein similarity of 97.01% and 91.04%, respectively. Subcellular localization results indicated that MaSAMP was localized in chloroplasts. Furthermore, through homologous recombination, a pCAMBIA1300-35S::MaSAMP-EGFP overexpression vector fused with an EGFP fluorescent tag was successfully constructed, and transgenic citrus plants were obtained using agrobacterium-mediated in planta transformation technology. This study would provide a material basis for subsequent research on the biological functions of MaSAMP.

我国柑橘产量和栽培面积均居世界首位[1],柑橘产业在水果贸易、乡村振兴中发挥着重要的作用。我国有19个柑橘种植省(市),其中有11个省遭受柑橘黄龙病(Citrus Huanglongbing,HLB)的危害,损失巨大,仅广东年损失达50亿元[2]。柑橘黄龙病是一种由寄生于韧皮部的革兰氏阴性细菌(Candidatus Liberibacter asiaticus,CLas)引起的系统性病害,表现为叶片斑驳黄化,植株枯萎,果实小,结果率低,产量锐减,并具有传播快的特征,故被称为柑橘中的癌症。该病害严重影响了我国柑橘产业的发展[3-6]。近百年来的农业实践和研究表明,尽管柑橘黄龙病可防可控,但不可根治[7],而其根本有效的解决途径仍有赖于抗性育种。
澳指檬(Microcitrus australasica)是一种原产于澳大利亚的澳指檬属的柑橘资源,最近因其对柑橘黄龙病有极高的耐受性而受到广泛关注[8-11]。有研究者对澳指檬及其杂交种进行不同组合的嫁接接毒实验,经过2 a的接毒评价,发现嫁接后,在砧木发病的情况下,澳指檬及后代叶片中均未检测到黄龙病菌[8]。通过体细胞融合技术,将OLL8甜橙或Page橘子愈伤组织来源的原生质体与澳指檬叶肉分离的原生质体进行细胞融合,培育的细胞融合植株在黄龙病的环境中生长6 a仍保持黄龙病阴性[9]
抗菌肽可替代抗生素,靶向细胞膜或特定的细胞内成分,通过多种特殊机制抑制多种微生物,是作为前瞻性抗真菌和抗细菌疗法的新兴候选药物[12],并且其产生耐药性的可能很小[13]。植物中具有许多独特的防御机制,抗菌肽是植物免疫系统最突出的部分,对病原体具有直接和持久的抗性,且抗菌肽是植物天然合成,对植物和哺乳动物细胞无毒害[14]。抗菌肽在整个植物体内的表达可用于增强抗病性或作物改良。MOULIN等[15]研究表明,富含胰蛋白酶抑制剂(10~14 kDa)的叶提取物通过抑制病原来源蛋白酶的酶活性,实现对辣椒黄花叶病毒(Pepper yellow mosaic virus,Pep YMV)的抗病毒活性。刘琦琦等[16]发现转入抗菌肽CecropinB能提高锦橙对柑橘溃疡病的抗性。随后有学者通过组学分析从澳指檬中鉴定出1种热稳定蛋白MaSAMP,体外实验表明该蛋白可有效降低黄龙病菌浓度及黄龙病阳性树的发病症状,同时,喷施和注射后亦可激活植株的先天免疫系统,抑制黄龙病感染[10],但关于该功能蛋白对柑橘属植株的具体生物学功能未见报道。本研究通过生物信息学分析进一步明确MaSAMP的生化功能及系统发育关系,同时借助农杆菌介导的原位转化,构建柑橘过表达遗传材料,为后续探究MaSAMP在黄龙病抗性中的生理功能及抗性育种等提供遗传材料基础。
2023年12月于广东省农业科学院果树研究所以经评价具高黄龙病耐受性的澳指檬为材料,使用天根(TIANGEN)多糖多酚植物总RNA提取试剂盒提取澳指檬成熟叶片RNA,再通过TaKaRa反转录试剂盒合成cDNA。遗传转化所用柑橘种子由广东省农业科学院果树研究所提供。
以澳指檬cDNA为模板,参考HUANG等[10]的方法设计引物MaSAMP-F/R(表1),通过PCR扩增获得MaSAMP基因。PCR反应体系为:无菌水20 μL、cDNA 2 μL、上/下游引物各1.5 μL、PrimeSTAR Max Premix(2×)25 μL。反应程序为:98 ℃ 3 min;98 ℃ 10 s,55 ℃ 5 s,72 ℃ 12 s;72 ℃ 5 min。扩增后进行凝胶电泳检测产物,使用美基(Magen)试剂盒回收目的条带。利用全式金Blunt Simple Cloning Kit进行TA克隆,胶回收产物2 μL,无菌水2 μL,pEASY Blunt Simple Cloning Vector 1 μL,37 ℃反应10 min,将连接产物转入Trans1-T1 Phage Resistant感受态后再超净工作台上加入250 μL无抗LB液体到上述产物中,200 r/min、37 ℃培养1 h。将菌液涂于50 μg/mL氨苄青霉素的平板上,37 ℃倒置培养12 h。挑取单菌落进行菌液PCR,体系为Premix Taq 10 μL、M13F/R各1 μL、模板2 μL、无菌水6 μL,通过电泳检测后挑选阳性菌液测序。
利用在线网站NCBI的Conserved Domain(https://www.ncbi.nlm.nih.gov/Structure/cdd/wrpsb.cgi)在线工具查找目的基因的保守结构域。使用ExPASy(https://web.expasy.org/protparam/)在线软件中的ProtParam工具分析MaSAMP蛋白理化性质。使用SOPMA(http://npsa-prabi.ibcp.fr)在线软件预测其二级结构。使用SignalP-5.0(https://services.healthtech.dtu.dk/services/SignalP-5.0/)在线软件预测该蛋白的信号肽,使用TMHMM(http://www.cbs.dtu.dk/services/TMHMM)在线软件预测蛋白质跨膜序列。在NCBI数据库中进行BLAST,选取一致性较高的序列通过DNAMAN 9.0软件进行蛋白多重序列比对。使用MEGA 11软件,采用最大简约法构建系统发育树。
选择SacⅠ、SalⅠ酶对pCAMBIA1300-EGFP质粒进行双酶切,条件为金属浴37 ℃ 1.5 h。以测序验证成功的pEASY-T1-MaSAMP质粒为模板进行目的片段扩增,体系为:无菌水21 μL、质粒DNA 1 μL、上/下游引物各1.5 μL、PrimeSTAR Max Premix(2×)25 μL,反应条件为:98 ℃ 3 min;98 ℃10 s,57 ℃ 10 s,72 ℃ 12 s;72 ℃ 5 min。通过凝胶电泳回收目的片段。利用同源重组反应,将目的片段和酶切产物进行连接,通过热激法将连接产物转入DH5α大肠杆菌,均匀涂布在含卡那霉素的LB固体平板上,37 ℃过夜培养。挑取单菌落后进行菌液PCR验证,挑取阳性菌液测序。
于2024年8月选取饱满完好的种子播种于塑料花盆,于26 ℃种植室培养4~6周,取幼苗用于原位转化,转化方法参照谢幸男等[17]的方法。待切口长出新芽后,通过LUYOR-3415激发光源进行检测。
选择培养60 d左右的柑橘实生苗嫩叶,参照酶解分离法[18-19]对柑橘原生质体进行分离,运用PEG介导法[19]将质粒pCAMBIA 1300-35S::MaSAMP-EGFP转化到柑橘原生质体中,培养12 h后于荧光显微镜下观察荧光。
通过ExPASy在线软件中的ProtParam工具对MaSAMP蛋白理化性质分析,结果显示,MaSAMP蛋白由67个氨基酸组成,其相对分子量为7.59959 kDa,理论等电点为5.38;其不稳定指数(instability index(II))为33.22,GASTEIGER等[20]将不稳定指数小于40的蛋白预测为稳定,反之则可能是不稳定蛋白,表明MaSAMP为稳定蛋白;脂肪族氨基酸系数(aliphatic index)为87.16%,表明该蛋白脂肪族氨基酸含量高;平均亲水系数(grand average of hydropathicity)为–0.091,说明该蛋白具有一定的亲水性。使用ExPASy在线软件中的ProtScale工具对其疏水性进行分析(图1A)发现,第8、9位氨基酸均为最低分值-1.267,第54位氨基酸具有最高分值1.267,且整体分布偏于负值,故预测其具有亲水性。
MaSAMP的二级结构预测(图1B)显示,该序列含有34.33% α-螺旋(alpha helix)、32.84%无规则卷曲(random coil)、25.37%延伸链(extended strand)、7.46% β-转角(beta turn)。MaSAMP蛋白三级结构预测(图1C)显示,其QMQE值为0.67,QMQEN值为0.61±0.07,一致性为77.42%,该模型准确度较高,为α-螺旋、β-转角、无规则卷曲和延伸链构成空间构象。
SignalP-5.0软件预测结果显示该蛋白不存在信号肽(图1D),表明其不是分泌蛋白。TMHMM软件进行蛋白质跨膜序列预测显示该蛋白是非跨膜蛋白(图1E)。
利用NCBI的CDD数据库对MaSAMP蛋白保守结构域进行查找(图1F),发现其具有1个Dabb(stress responsive A/B Barrel Domain)保守结构域,属于ABM(antibiotic biosynthesis monooxygenase)超家族。
蛋白序列比对(图2)发现,MaSAMP与甜橙(Citrus sinensis)胁迫响应A/B桶状结构域蛋白CISIN_1g033887mg(KDO72075.1)、克里曼丁橘(Citrus×clementina)A/B桶状结构域蛋白HS1(XP_006419297.1)、甜橙胁迫响应A/B桶状结构域蛋白HS1(KAH9683996.1)、酸橙(Citrus × changshan-huyou)WN943_028324(KAK9179 126.1)、苦楝(Melia azedarach)胁迫响应A/B桶状结构域蛋白(KAJ4721944.1)、欧洲桤木(Alnus glutinosa)胁迫响应A/B桶状结构域蛋白HS1(XP_062178246.1)、藜麦(Chenopodium quinoa)胁迫响应A/B桶状结构域HS1样蛋白(XP_021762050.1)相似度分别为97.01%、91.04%、89.55%、86.57%、85.07%、82.09%、82.09%。系统发育分析(图3)表明多个品种的柑橘以及楝科、桦木科、苋科中均含有MaSAMP同源蛋白,该蛋白与甜橙胁迫响应A/B桶状结构域蛋白CISIN_1g033887mg(KDO72075.1)的亲缘关系最近。
以澳指檬叶片cDNA为模板进行PCR扩增,获得一条201 bp的条带(图4)。通过菌液PCR、测序验证,测序结果显示获得一段长度为201 bp,编码67个氨基酸的目的序列(图5),表明基因克隆成功。
Sal I和Sac I酶对pCAMBIA1300-EGFP进行双酶切,获得1条酶切后的线性条带(图6A)。目的基因与载体进行连接后转入大肠杆菌DH5α,利用载体上的引物(1300-35S-F/1300-GFP-R)进行菌液PCR获得1条402 bp的条带(图6B),再对菌液进行测序验证,结果显示载体中插入了1段长度为201 bp的序列,表明pCAMBIA1300-35S::MaSAMP-EGFP过表达载体构建成功。
利用农杆菌介导的原位转化法,通过农杆菌EHA105,将构建成功的重组载体转化至柑橘幼苗。农杆菌侵染后培养4~6周对新芽进行检测。利用表达载体上携带的EGFP绿色荧光蛋白表达元件,在440~460 nm激发光源下筛选到转化成功的植株(图7)。
将融合绿色荧光蛋白(EGFP)的pCAMBIA 1300-35S::MaSAMP-EGFP质粒导入柑橘原生质体进行亚细胞定位分析,利用荧光显微镜观察。结果显示,MaSAMP-EGFP在原生质体中成功表达,原生质体的叶绿体中检测到了清晰的EGFP荧光信号(图8),表明MaSAMP蛋白定位于叶绿体中。
HUANG等[10]通过组学分析在澳指檬中获得抗菌肽基因MaSAMP,并通过原核表达获得MaSAMP。在对感染黄龙病柑橘的喷洒和注射实验中,长达14个月的黄龙病菌浓度检测结果表明,MaSAMP对黄龙病菌具有极好的灭杀能力。罗澍[21]进一步克隆出Long-SAMP不同特异转录本类型,并对其氨基酸序列进行分析比较。Long-SAMP是一段含有109个氨基酸的长肽,而根据HUANG等[10]的报道,抗菌肽的功能区段仅存在于Long-SAMP的后67个氨基酸中。因此,本研究从HUANG等的研究中获取引物,克隆出MaSAMP基因,并对该序列进行分析。
本研究结果显示,MaSAMP蛋白的相对分子量为7.59959 kDa,理论等电点为5.38,不稳定指数为33.22,根据GASTEIGER等[20]的注释判定MaSAMP为稳定蛋白。脂肪族氨基酸系数为87.16%,说明该蛋白属于脂肪族氨基酸。平均亲水系数为负数,表明该蛋白具一定的亲水性,同时Kyte and Doolittle算法亦表明其具有亲水性。该蛋白二级结构中有34.33%为α-螺旋结构,32.84%为无规则卷曲,25.37%为延伸链,7.46%为β-转角,该蛋白三级结构与二级结构预测结果一致。天然抗菌肽长约10~100个氨基酸,且大多两端均含有亲水和疏水残基,其α-螺旋、β-折叠等结构有重要功能特征[22]。该蛋白不存在信号肽,说明该蛋白为非分泌蛋白,且与TMHMM软件的预测结果一致,即为非跨膜蛋白。MaSAMP蛋白保守结构域为Dabb(stress responsive A/B barrel domain),GU等[23]从胡杨(Populus euphratica)的热休克蛋白(heatshock protein 90,Hsp90)和羟脯氨酸糖蛋白(hydroxyproline-rich glycoprotein,HRGP)中发现Dabb结构域,2个蛋白均参与了植物耐盐胁迫反应。Dabb类蛋白被发现显著抑制各种病原真菌的细胞生长,参与诱导植物对多种病原真菌的防御[24-25]。MaSAMP含有Dabb结构域,推测该蛋白可能参与植物耐盐胁迫反应和诱导植物对病原菌的防御过程。Blast发现,甜橙、克里曼丁橘、酸橙、苦楝、欧洲桤木、藜麦存在与MaSAMP相似度大于80%的序列,系统发育分析表明澳指檬MaSAMP与上述物种相似序列蛋白有一定亲缘关系,与甜橙胁迫响应A/B桶状结构域蛋白CISIN_1g033887mg的亲缘关系更近。根据亚细胞定位结果,MaSAMP蛋白定位于叶绿体中。腾海艳[26]研究发现水稻Ossp1基因定位于叶绿体,且其表达具有干旱响应性,推测其功能可能与叶绿体内相关代谢过程或植物的逆境响应有关。
