Cibotium barometz, a nationally second-class protected plant in China, exhibits significant medicinal and ornamental value. It is naturally distributed in the southern regions of China and exhibits stringent requirements for its growth environment. Investigating the physiological response mechanisms of C. barometz to low-temperature stress can provide a scientific foundation for its artificial cultivation and the comprehensive development and utilization of its resources. This study utilized artificially cultivated C. barometz as the experimental material and established three temperature levels: 5 ℃, 15 ℃ and 25 ℃. Each temperature level was subjected to treatments lasting 5 days, 10 days, and 15 days, respectively. Following the stress treatment, chlorophyll fluorescence parameters, physiological indicators, and total flavonoid content in the leaves were measured. The growth and physiological characteristics of C. barometz under different temperature conditions were analyzed based on the experimental results. The results indicated that following exposure to 5 ℃ and 15 ℃, the photosynthetic capacity of C. barometz leaves significantly decreased, whereas the activities of superoxide dismutase (SOD) and peroxidase (POD) markedly increased. In contrast, no significant differences were observed in the levels of free proline (Pro) and soluble sugars. After 15 days of treatment at 5 ℃, the malondialdehyde (MDA) content in the leaves of C. barometz significantly increased. Proper low-temperature treatment significantly enhanced the accumulation of total flavonoids in the leaves of C. barometz. On the 10th day of treatment at 5 ℃and 15 ℃, the total flavonoid content reached 688.78% and 511.01% of that in the 25 ℃ treatment group, respectively, representing the highest levels observed. This study demonstrated that low-temperature stress exerts differential effects on various physiological indicators in C. barometz. The activities of SOD and POD, and the total flavonoid content, can serve as key indicators for evaluating the cold tolerance of C. barometz seedlings. Temperatures of 15 ℃ and below can significantly reduce the photosynthetic capacity of C. barometz, leading to abiotic stress and a marked increase in the total flavonoid content in its leaves. This result could provide an important reference for optimizing temperature control conditions in the artificial cultivation of. C. barometz.

国家二级保护植物金毛狗(Cibotium barometz)具有巨大的药用和观赏价值,自然分布于我国南方地区,对生长环境的要求较高。研究金毛狗对低温胁迫的生理响应机制,可为其人工栽培及资源的综合开发利用提供科学依据。本研究以人工栽培的金毛狗叶片为试验材料,设置5、15、25 ℃三个温度水平,每个温度水平分别处理5、10、15 d。胁迫处理后,测定其叶片中的叶绿素荧光参数、生理指标及总黄酮含量,分析不同温度条件下金毛狗叶片的生长和生理特性。结果表明:在5、15 ℃处理后,金毛狗叶片光合能力显著下降,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性明显升高,而游离脯氨酸(Pro)和可溶性糖含量差异不显著。在5 ℃条件下处理15 d时,金毛狗叶片内丙二醛(MDA)含量显著增加。适当的低温处理显著促进了金毛狗叶片中总黄酮含量的积累,在5、15 ℃条件下处理至10 d时,总黄酮含量达到最高值,分别为25 ℃处理组的688.78%和511.01%。本研究结果表明,低温胁迫对金毛狗各项生理指标的影响存在差异,SOD、POD活性及总黄酮含量可作为评估金毛狗成苗耐寒性鉴定的主要指标。15 ℃及以下的温度会显著降低金毛狗的光合作用能力,导致逆境胁迫,并显著增加叶片中的总黄酮含量。此结果可为金毛狗人工种植中优化温控条件提供重要参考。

国家二级保护植物金毛狗(Cibotium barometz)具有巨大的药用和观赏价值,自然分布于我国南方地区,对生长环境的要求较高。研究金毛狗对低温胁迫的生理响应机制,可为其人工栽培及资源的综合开发利用提供科学依据。本研究以人工栽培的金毛狗叶片为试验材料,设置5、15、25 ℃三个温度水平,每个温度水平分别处理5、10、15 d。胁迫处理后,测定其叶片中的叶绿素荧光参数、生理指标及总黄酮含量,分析不同温度条件下金毛狗叶片的生长和生理特性。结果表明:在5、15 ℃处理后,金毛狗叶片光合能力显著下降,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性明显升高,而游离脯氨酸(Pro)和可溶性糖含量差异不显著。在5 ℃条件下处理15 d时,金毛狗叶片内丙二醛(MDA)含量显著增加。适当的低温处理显著促进了金毛狗叶片中总黄酮含量的积累,在5、15 ℃条件下处理至10 d时,总黄酮含量达到最高值,分别为25 ℃处理组的688.78%和511.01%。本研究结果表明,低温胁迫对金毛狗各项生理指标的影响存在差异,SOD、POD活性及总黄酮含量可作为评估金毛狗成苗耐寒性鉴定的主要指标。15 ℃及以下的温度会显著降低金毛狗的光合作用能力,导致逆境胁迫,并显著增加叶片中的总黄酮含量。此结果可为金毛狗人工种植中优化温控条件提供重要参考。

Cibotium barometz, a nationally second-class protected plant in China, exhibits significant medicinal and ornamental value. It is naturally distributed in the southern regions of China and exhibits stringent requirements for its growth environment. Investigating the physiological response mechanisms of C. barometz to low-temperature stress can provide a scientific foundation for its artificial cultivation and the comprehensive development and utilization of its resources. This study utilized artificially cultivated C. barometz as the experimental material and established three temperature levels: 5 ℃, 15 ℃ and 25 ℃. Each temperature level was subjected to treatments lasting 5 days, 10 days, and 15 days, respectively. Following the stress treatment, chlorophyll fluorescence parameters, physiological indicators, and total flavonoid content in the leaves were measured. The growth and physiological characteristics of C. barometz under different temperature conditions were analyzed based on the experimental results. The results indicated that following exposure to 5 ℃ and 15 ℃, the photosynthetic capacity of C. barometz leaves significantly decreased, whereas the activities of superoxide dismutase (SOD) and peroxidase (POD) markedly increased. In contrast, no significant differences were observed in the levels of free proline (Pro) and soluble sugars. After 15 days of treatment at 5 ℃, the malondialdehyde (MDA) content in the leaves of C. barometz significantly increased. Proper low-temperature treatment significantly enhanced the accumulation of total flavonoids in the leaves of C. barometz. On the 10th day of treatment at 5 ℃and 15 ℃, the total flavonoid content reached 688.78% and 511.01% of that in the 25 ℃ treatment group, respectively, representing the highest levels observed. This study demonstrated that low-temperature stress exerts differential effects on various physiological indicators in C. barometz. The activities of SOD and POD, and the total flavonoid content, can serve as key indicators for evaluating the cold tolerance of C. barometz seedlings. Temperatures of 15 ℃ and below can significantly reduce the photosynthetic capacity of C. barometz, leading to abiotic stress and a marked increase in the total flavonoid content in its leaves. This result could provide an important reference for optimizing temperature control conditions in the artificial cultivation of. C. barometz.

金毛狗[Cibotium barometz(L.)J. Sm.]作为国家二级保护植物,具有巨大的药用和观赏价值,自然分布于四川、重庆、贵州、云南、广西、广东、福建、江西等地[1]。该物种对生长环境有较高要求,在20~30 ℃的温度范围内已成功实现人工栽培[2]。低温作为一种主要的非生物胁迫因素,对植物的生长发育有着极大的影响。金毛狗的最佳生长及耐受温度尚未明确,这在很大程度上限制了该珍稀植物的规模化种植和开发利用。因此研究金毛狗应对低温胁迫的生理响应机制具有重要意义。金毛狗的根状茎是其主要用药部位,具有悠久的民间及临床应用历史,常用于治疗腰膝酸软、手足麻木等症状[3]。目前,关于金毛狗生理活性成分的分析、提取及应用研究主要集中在其根状茎部分,而对叶片的研究相对较少。傣医学认为金毛狗叶片具有利尿通淋、清热凉血等功效,可用于治疗肠炎、痢疾等症[4]。前人研究已验证金毛狗叶片富含黄酮类化合物[5],这些成分具有抗氧化、抗炎和抗肿瘤等生理活性。SAITO等[6]从金毛狗的地上部分分离出了山柰素(kaempferol),林燕如等[7]则通过大孔树脂工艺成功纯化了金毛狗叶片中的总黄酮。目前,国内外尚未见关于金毛狗在逆境胁迫下诱导黄酮等活性物质的研究报道。超氧化物歧化酶(superoxide dismutase,SOD)、过氧化物酶(peroxidase,POD)是植物体内清除自由基的主要酶类,脂质过氧化物丙二醛(malondialdehyde,MDA)常作为衡量膜质过氧化程度的标志,游离脯氨酸(proline,Pro)、可溶性糖是植物体内重要的渗透调节物质。本研究以金毛狗为试材,通过低温处理后测定并分析其叶片中的叶绿素荧光参数、SOD活性等5项生理指标及总黄酮含量的变化情况。所得结果有助于揭示金毛狗的耐寒生理机制及其药用成分的形成规律,为金毛狗的人工种植及资源的综合开发利用提供科学依据。
选取生长状况基本一致的三年生金毛狗叶片为试验材料,植株均为福建省亚热带植物研究所林志楷副研究员人工培育。栽植土基本养分为有机质20.64 g/kg,pH 5.83,水解氮164 mg/kg,有效磷190 mg/kg,速效钾109 mg/kg。
参考金毛狗自然生境冬季气温,本研究设计了5 ℃和15 ℃作为低温处理,并以25 ℃为对照处理。于2023年3—4月在福建省亚热带植物研究所气候室进行试验,分别调节3个气候室温度为5、15、25 ℃,共3个温度水平,每个温度水平分别处理5、10、15 d。每个处理3个重复,每个重复金毛狗植株5株。在处理时间到达5、10、15 d分别采集长势大致相同的金毛狗叶片,叶片采集后立即用锡箔纸包好,分别编号并做好标记,根据不同的采集时间和处理温度对样品进行分组,放入液氮中速冻,–80 ℃超低温冰箱保存待测。在处理时间15 d时测量叶绿素荧光参数。
取叶龄相同、大小相近的二回羽状叶,经暗处理后采用IMAGING-PAM MINI叶绿素荧光仪(德国WALZ)读取荧光参数,测量光合电子传递速率(photosynthetic electron transport rate,ETR)、光化学效率Y(Ⅱ)、光化学淬灭系数(photochemical quenching,qP)、非光化学淬灭系数(non-photochemical quenching,qN)。取新鲜叶片去除叶脉,叶片SOD活性采用氮蓝四唑比色法测定,POD活性使用南京建成生物工程研究所试剂盒测定,MDA、Pro、可溶性糖、总黄酮含量使用北京Solarbio试剂盒测定。
采用Microsoft Excel 2016软件进行数据处理和作图,采用SPSS 26.0统计分析软件进行方差分析。数值为3次生物学重复的平均值±标准差,不同小写字母表示LSD分析方法下同一处理天数不同温度之间差异显著(P<0.05)。
图1所示,金毛狗叶片在不同温度下ETR值随着光合有效辐射(photosynthetically active radiation,PAR)增加表现出不同的趋势。金毛狗为喜荫植物,3个温度的ETR均值在PAR为11 μmol/(m2·s)时相当。25 ℃处理的ETR均值在PAR为43 μmol/(m2·s)时达到最高,之后随着PRA的增加仍维持平稳。5、15 ℃两个处理在PAR为23 μmol/(m2·s)时出现拐点,之后随着PRA的继续增加ETR逐渐降低。3个温度处理15 d后差异明显,表明15 ℃及以下温度对金毛狗的光保护能力造成了不利影响。
图2可知,暗适应后各温度处理叶的中部Y(Ⅱ)变化较大,叶中部大约同一位置在同一辐射下PSⅡ的实际光合效率温度处理表现为25 ℃>15 ℃>5 ℃。25 ℃处理数值最高,为0.815,高于5、15 ℃处理,随着光照强度和时间的增加,5、15 ℃两个处理的Y(Ⅱ)更快降至0,而25 ℃处理的Y(Ⅱ)还能保持在0.061,表明金毛狗在15 ℃及以下温度的光合效率有所降低。
金毛狗叶片qP值随辐射变化过程如图3所示,qP值从1(白色)下降到0。相较于25 ℃处理,5、15 ℃处理的金毛狗叶片qP值下降更快,但图像中肉眼可见的变化并不明显,判断植株光合能力是否减弱还需要结合其他叶绿素荧光参数指标加以分析。
高qN值在叶绿素荧光成像系统下呈现为紫色。如图4所示,处理温度越低,金毛狗叶片的qN值越高,5 ℃处理在颜色变化上最明显,光合作用效率显著降低。可见,金毛狗叶片qN与外部环境温度的关联较为密切,5 ℃较15 ℃对植株低温胁迫更明显。
图5可知,在5、15 ℃处理下,金毛狗叶片的SOD活性随着时间的延长总体上升,在5~10 d时相对平稳,在10~15 d时提升较多;25 ℃处理下,在5~10 d之间有所增加,分析其原因可能是由于植物在生长中抗氧化酶的正常积累;3个温度处理下同一时间段比较,5、15 d时低温处理的SOD活性比25 ℃均显著升高20%以上(P<0.05)。
图6可知,3个温度处理POD活性均随着时间的延长总体升高,但是提升幅度不大。3个温度处理下同一时间段比较,2个低温处理与25 ℃处理的POD活性均显著升高49%以上(P<0.05)。在5、10 d时,5 ℃与15 ℃处理相比,POD活性均升高12%以上(P<0.05),温度越低对植株的胁迫越大。在15 d时,5 ℃与15 ℃处理的POD活性趋近,分析其原因可能是植株的POD保护能力已经达到饱和。
图7所示,MDA含量随着处理时间的延长发生不同的变化。在5、10 d时,3个温度处理的MDA含量变化差异均不显著;15 d时,15、25 ℃处理的MDA含量分别降低10.15%、7.41%,而5 ℃处理的MDA含量升高22.08%,表明此处理条件下的叶片细胞已经实质受损。
图8所示,3个温度处理的Pro含量均随着时间的延长有所提高,但增幅不大。Pro含量在植物体内正常情况下不高,只有发生胁迫特别是冻害时才会大量积累。结果显示,同一时间段3个温度处理之间金毛狗叶片的Pro含量差异均不显著。但15、25 ℃处理的Pro含量始终较5 ℃处理更低,表明5 ℃对植株造成了轻微的胁迫。
图9可知,3个温度处理下同一时间段比较,可溶性糖含量的差异均不显著。3个温度处理的可溶性糖含量均随时间的延长而增加,15 d与5 d相比,5、15、25 ℃处理分别提高了28.68%、22.24%、12.34%。同其余指标一样,可溶性糖含量适度变化也是植物生长中的正常代谢所导致,但5 ℃处理增幅最大,再次表明该温度对植株产生了更严重的胁迫。
图10所示,金毛狗叶片中的总黄酮含量在3个时间段温度处理均表现为5 ℃>15 ℃>25 ℃,而且同一时间段内3组处理差异显著(P<0.05)。25 ℃处理的总黄酮含量一直保持在较低水平。5、15 ℃处理的总黄酮含量在10 d时达到最高,分别是25 ℃处理的688.78%和511.01%,之后在15 d时出现明显下降。
为更直观地了解低温胁迫对金毛狗光合作用能力的影响,本研究进行了叶绿素荧光测定。光响应曲线反映植物叶片对光强瞬间变化的适应机制,是研究植物光合生理特性及其适应性的重要手段[8]。ETR代表PSⅡ电子传递速率,能反映植物有效耗散过剩光能的光保护作用的大小[9]。本研究结果显示,15 ℃和5 ℃处理的ETR低于25 ℃处理,表明这2个温度对金毛狗植株已经产生了逆境胁迫,植株电子传递速率与实际光合效率降低,无法将多余的光能用于CO2同化或者无法将大量的电子分配到光呼吸中,导致植物有效耗散过剩光能的光保护作用减弱。光化学效率Y(Ⅱ)是光照条件下PSⅡ的实际光合效率,能够间接反映线性电子传递的活性,较高的Y(Ⅱ)能促进碳同化的高效运转和有机物的积累[10]。qP数值越低,表示植物在光适应状态下,更少的光能被用于光化学反应,从而反映出植物的光合作用能力更弱。qN值升高,虽然有助于保护植物免受损伤,但同时也意味着光合作用效率的暂时降低。Y(Ⅱ)、qN、qP测定结果表明,5 ℃和15 ℃均显著抑制了金毛狗的光合能力,且5 ℃条件下的抑制作用更明显。低温胁迫能对蕨类植物的光合作用产生不利影响,这与毕敏慧[11]、毛林鲜等[12]的研究结果一致。
进一步分析金毛狗叶片的生理特性,各项指标对低温胁迫表现出不同的响应。SOD、POD是植物体内清除自由基的主要酶类,其活性大小反映了植物对逆境的适应能力[13]。在15 ℃和5 ℃两个温度下,金毛狗叶片中SOD与POD两项指标在较短时间内显著增加,表明其保护酶系统对低温的反应十分敏感,15 ℃及以下温度已迫使植株通过清除有害ROS来保护自身,SOD与POD在金毛狗耐冷机制中发挥重要作用。MDA含量是鉴定细胞膜过氧化的重要指标[14]。25 ℃处理下MDA含量减少可能是因为植株快速生长导致其他物质迅速增加;15 ℃处理下MDA含量减少可能与植株具有较高SOD与POD活性有关。在对苜蓿根和藜麦幼苗叶片的胁迫研究中均也检测到MDA含量降低的现象[15-16]。5 ℃处理15 d后,活性氧造成的破坏已超过金毛狗抗氧化酶系统的保护能力,细胞膜过氧化加剧引起导致MDA含量增加。Pro能够维持生物膜和酶蛋白的结构稳定性,作为非酶促抗氧化物质,它能清除逆境中产生的大量ROS,增强植物的抗低温能力[17]。可溶性糖的积累可以增强细胞膜的稳定性,并参与清除细胞中的ROS[18]。本研究中金毛狗叶的MDA(5 ℃处理15 d测得的结果除外)、Pro、可溶性糖含量在同一时间段低温处理组与对照组整体差异不明显,说明叶片内部调节功能较强。综合5项指标结果,金毛狗主要通过提高POD和SOD活性来增强其抗冷性,对5 ℃及以上较低温度具备一定的耐冷能力。
本研究结果表明,低温胁迫能显著影响金毛狗叶片中黄酮物质的积累。黄酮物质虽然不是植物生长所必需的物质,但在植物生长发育和逆境适应中发挥着重要作用,其可作为ROS的直接或间接清除剂,充当信号分子激活与植物防御相关的信号传导途径和调控机制,从而增强植物抗性[19]。15 ℃及以下温度可以引导金毛狗叶片中黄酮物质的合成和积累增加,温度越低叶片中总黄酮含量越高,温度与总黄酮含量之间呈现负相关关系,此研究结果与前人在桃子[20]、佛手瓜[21]上的研究结果相似;总黄酮含量随着低温处理时间的延长呈先升高后降低的趋势,在长时间胁迫后总黄酮含量显著下降,这一现象与董新纯等[22]研究苦荞遭受UV-B伤害后期总黄酮含量下降的研究结果一致。因此,低温胁迫时间也是影响金毛狗调节黄酮物质积累的重要因素之一。本研究结果解释了传统中药金毛狗在秋末至冬季采挖时药材质量较高的原因。
厉广辉等[23]对花生的研究表明,总黄酮含量与SOD活性、MDA含量、植物株高和叶面积高度相关。钟楚等[24]通过对烤烟的逆境研究发现,总黄酮与POD和SOD表现出协同效应。金毛狗在遭受低温胁迫后,光合能力显著下降,但通过快速积累总黄酮并同时提高SOD和POD活性来抵御逆境,最大限度地延缓了MDA的积累损害。此外,金毛狗无需过早启动Pro和可溶性糖等渗透调节机制,从而确保植株体内物质和能量的高效利用。
综上所述,15 ℃及以下的低温显著抑制了金毛狗的光合作用能力,并对其各项生理指标产生了不同程度的影响。具体表现为:SOD和POD活性显著升高,MDA含量在5 ℃条件下后期明显增加,而Pro和可溶性糖含量变化不显著。此外,低温胁迫显著提高了金毛狗叶片中总黄酮的含量。因此,SOD、POD和总黄酮含量可作为评价金毛狗耐寒性的关键指标。本研究为揭示金毛狗对低温胁迫的应答调控机制提供了基础数据,有助于提高人工栽培的质量和效率。然而,本研究存在一定的局限性,如温度处理范围较广,未来的研究应进一步细化温度梯度设置,延长处理时间,并结合多组学分析以获得更全面的数据。
