Isoamylase (Isoamylase, ISA) catalyzes the hydrolysis of α-1,6 linkages, thereby affecting the branching structure of starch and promoting crystallization and granule formation. The study utilized yeast two-hybrid screening to identify proteins interacting with MeISA2 to explore new proteins that may interact with MeISA2 to influence cassava starch accumulation. A bait protein vector pGBKT7-MeISA2 was constructed, and it was demonstrated that MeISA2 did not possess autoactivation activity and was non-toxic to yeast cells. Through yeast two-hybrid screening of a cDNA library, a total of 7 candidate interacting proteins were obtained. Point-to-point yeast two-hybrid re-validation experiments confirmed the interaction between MeISA2 and MePsbO1. MePsbO1 belongs to the Photosystem II oxygen-evolving enhancer protein (PsbO) family, which facilitates the efficient operation of the photosynthetic electron transfer process. Bioinformatics analysis revealed that MePsbO1 had the closest phylogenetic relationship with HbPsbO1 from Hevea brasiliensis, a member of the Euphorbiaceae family, with a similarity of 90.12%, while its similarity to Arabidopsis thaliana AtPsbO1 was only 77.71%. It is predicted that MePsbO1 is localized in the chloroplast, and the MePsbO1 gene is predominantly expressed in leaves. Plant ISA is involved in the synthesis of transient leaf starch, thus it is hypothesized that MePsbO1 and MeISA2 may interact to jointly regulate the rate of photosynthesis and the accumulation of transient leaf starch. The results study would provide new insights into the mechanism by which the MeISA2 gene participates in the accumulation and distribution of photosynthetic products in cassava source organs.

异淀粉酶(isoamylase,ISA)催化α-1,6连接键水解,影响淀粉的分支结构,促进结晶化和颗粒形成。为了挖掘协同MeISA2影响木薯淀粉积累的新蛋白,本研究利用酵母双杂交筛选MeISA2的互作蛋白。本研究构建了pGBKT7-MeISA2诱饵蛋白载体,证明MeISA2不具有自激活活性,且对酵母细胞无毒害作用。通过酵母双杂交筛选cDNA文库,共获得7个候选互作蛋白。通过点对点酵母双杂交回转验证实验,验证了MeISA2与MePsbO1存在互作关系。MePsbO1属于光系统II产氧增强蛋白(photosystem II oxygen-evolving enhancer protein,PsbO),该类蛋白可促进光合作用电子传递过程的高效运行。生物信息学分析发现,MePsbO1与同属大戟科的橡胶树HbPsbO1的亲缘关系最近,相似性达到90.12%,而与拟南芥AtPsbO1的相似性仅为77.71%。预测MePsbO1定位于叶绿体,且MePsbO1基因主要在叶片中表达。植物ISA参与叶片临时性淀粉的合成,因此推测MePsbO1和MeISA2可能通过互作协同调控光合作用速率以及叶片临时性淀粉的积累。本研究结果为MeISA2基因参与木薯源器官的光合产物积累和分配机制研究提供新线索。

异淀粉酶(isoamylase,ISA)催化α-1,6连接键水解,影响淀粉的分支结构,促进结晶化和颗粒形成。为了挖掘协同MeISA2影响木薯淀粉积累的新蛋白,本研究利用酵母双杂交筛选MeISA2的互作蛋白。本研究构建了pGBKT7-MeISA2诱饵蛋白载体,证明MeISA2不具有自激活活性,且对酵母细胞无毒害作用。通过酵母双杂交筛选cDNA文库,共获得7个候选互作蛋白。通过点对点酵母双杂交回转验证实验,验证了MeISA2与MePsbO1存在互作关系。MePsbO1属于光系统II产氧增强蛋白(photosystem II oxygen-evolving enhancer protein,PsbO),该类蛋白可促进光合作用电子传递过程的高效运行。生物信息学分析发现,MePsbO1与同属大戟科的橡胶树HbPsbO1的亲缘关系最近,相似性达到90.12%,而与拟南芥AtPsbO1的相似性仅为77.71%。预测MePsbO1定位于叶绿体,且MePsbO1基因主要在叶片中表达。植物ISA参与叶片临时性淀粉的合成,因此推测MePsbO1和MeISA2可能通过互作协同调控光合作用速率以及叶片临时性淀粉的积累。本研究结果为MeISA2基因参与木薯源器官的光合产物积累和分配机制研究提供新线索。

Isoamylase (Isoamylase, ISA) catalyzes the hydrolysis of α-1,6 linkages, thereby affecting the branching structure of starch and promoting crystallization and granule formation. The study utilized yeast two-hybrid screening to identify proteins interacting with MeISA2 to explore new proteins that may interact with MeISA2 to influence cassava starch accumulation. A bait protein vector pGBKT7-MeISA2 was constructed, and it was demonstrated that MeISA2 did not possess autoactivation activity and was non-toxic to yeast cells. Through yeast two-hybrid screening of a cDNA library, a total of 7 candidate interacting proteins were obtained. Point-to-point yeast two-hybrid re-validation experiments confirmed the interaction between MeISA2 and MePsbO1. MePsbO1 belongs to the Photosystem II oxygen-evolving enhancer protein (PsbO) family, which facilitates the efficient operation of the photosynthetic electron transfer process. Bioinformatics analysis revealed that MePsbO1 had the closest phylogenetic relationship with HbPsbO1 from Hevea brasiliensis, a member of the Euphorbiaceae family, with a similarity of 90.12%, while its similarity to Arabidopsis thaliana AtPsbO1 was only 77.71%. It is predicted that MePsbO1 is localized in the chloroplast, and the MePsbO1 gene is predominantly expressed in leaves. Plant ISA is involved in the synthesis of transient leaf starch, thus it is hypothesized that MePsbO1 and MeISA2 may interact to jointly regulate the rate of photosynthesis and the accumulation of transient leaf starch. The results study would provide new insights into the mechanism by which the MeISA2 gene participates in the accumulation and distribution of photosynthetic products in cassava source organs.

木薯(Manihot esculenta Crantz)是大戟科双子叶植物,广泛种植于非洲、美洲、亚洲等热带和亚热带地区[1]。木薯的主要营养器官是块根,淀粉是块根的主要成分,同时也是木薯生长发育主要的营养来源[2]。木薯有耐贫瘠、抗旱和高产等种植特性,其块根中的淀粉是理想的工业原料,且是全球10亿人口的主要粮食作物[3]。木薯淀粉常用作增稠剂、稳定剂和凝胶剂,适用于糕点、面包和无麸质食品。纺织与造纸工业中,木薯淀粉用于提高纤维的强度和抗磨损能力,改进纸张的表面光滑度和强度。
淀粉由直链淀粉和支链淀粉组成,木薯块根中直链淀粉和支链淀粉的含量比例对木薯淀粉的品质和食用口感具有重要影响[4]。淀粉脱支酶(debranching enzyme,DBE)、可溶性淀粉合成酶(starch synthases,SS)和淀粉分支酶(starchbranching enzymes,SBE)对支链淀粉的合成具有催化作用[5]。可溶性淀粉合成酶主要由可溶性淀粉合成酶Ⅰ(SSⅠ)、可溶性淀粉合成酶Ⅱ(SSⅡ-1、SSⅡ-2)、可溶性淀粉合成酶Ⅲ(SSⅢ-1、SSⅢ-2)、可溶性淀粉合成酶Ⅳ(SSⅣ)和可溶性淀粉合成酶Ⅴ(SSⅤ)组成,在生物淀粉合成中的支链淀粉合成中发挥关键作用,SS活性影响淀粉颗粒形态、结构和理化特性;淀粉分支酶具有SBEⅠ、SBEⅡ和SBEⅢ三种类型,在淀粉生物合成中催化外部和内部的α-1,6键转化成α-1,4-线性葡聚糖,也是唯一作用于葡聚糖并通过α-1,6-糖苷键连接产生分支的酶[6]。淀粉脱支酶包括异淀粉酶(isoamylase,ISA)和普鲁兰酶(pullulanase),ISA在植物淀粉结构的合成和数量上具有重要的作用,影响木薯支链淀粉和直链淀粉的比例[7]
ISA专门催化支链淀粉、β-极限糊精和糖原中α-1,6连接键的水解,但无法水解普鲁兰[8]。植物有3种异淀粉酶亚型(ISA1、ISA2和ISA3),其中ISA1和ISA2在植物淀粉合成中具有重要影响。目前,在水稻、大麦、马铃薯、玉米作物中均被证明当ISA基因缺失时,均会影响其淀粉颗粒的形成[9-11]。ISA1和ISA2负责去除支链淀粉合成中产生的支链,而ISA3则专门降解支链淀粉中的支链α-葡聚糖[12]。ISA1既能与自身形成同源复合物,又能与ISA2结合形成异源复合物,在异源复合物中ISA1起催化作用,ISA2由于其活性位点失去了所有3个催化残基而成为非催化蛋白,发挥调节作用[13]。异淀粉酶处理木薯淀粉,可水解淀粉中的1,6-α-D-糖苷键,降解支链淀粉并释放线性短链糖,这导致直链淀粉比率提高,同时生成更多的还原糖[14]。PANPETCH等[15]克隆了KU50木薯品种的MeISA1MeISA2基因,原核表达其重组蛋白,证明MeISA1和MeISA2蛋白同时存在时才能具有出脱分支酶活性,并分析了其酶学特性。本研究克隆了我国木薯主栽品种SC8的MeISA2基因,通过酵母双杂交实验筛选MeISA2的互作蛋白,以期为深入探讨MeISA2在淀粉合成过程中的功能提供新的信息。
植物材料为华南8号木薯(SC8)。酵母感受态AH109和大肠感受态DH5α均购自上海唯地生物技术有限公司;RNA反转录试剂盒(MonSCRIPTTM RTⅢ All-in-One-Mix With dsDNase)和qRT-PCR试剂盒(ChemoHS qPCR Mix)购自武汉莫纳公司;酵母双杂交载体pGADT7、pGADT7-T、pGBKT7、pGBKT7-p53均由本实验室保存。;PrimeSTAR HS(Premix)DNA PolyMeras酶购自TaKaRa公司;质粒提取试剂盒和纯化试剂盒购自OMEGA生物试剂公司;RNA提取试剂盒购自北京聚合美生物科技有限公司;X-α-gal酵母显色底物购自北京索莱宝科技有限公司;酵母缺陷培养基和2×Rapid Tap MasterMix酶分别购自clontech公司和南京诺唯赞生物科技股份有限公司;限制性核酸内切酶EcoRⅠ、BamHⅠ购自赛默飞世尔科技公司。
根据木薯基因组数据库信息,设计MeISA2基因克隆引物(表1),以SC8木薯cDNA模板进行PCR扩增,通过1%凝胶电泳检测PCR扩增产物,纯化备用。将纯化的MeISA2片段与酶切纯化pGBKT7空载体(酶切位点EcoR Ⅰ和BamH Ⅰ)进行同源重组。将同源重组产物转化到DH5α大肠杆菌中,利用pGBKT7通用检测引物(表1)进行菌液PCR鉴定,筛选阳性菌液进行Sanger测序,提取测序结果比对正确的菌液中的重组质粒,于–20 ℃冰箱保存备用。
将pGBKT7空载和pGBKT7-MeISA2重组质粒分别转化到AH109酵母感受态,涂布于SD/-Trp固体培养基上,30 ℃倒置培养2~3 d。挑取菌斑至含有2×Rapid Master Mix的PCR管中,使用BD检测引物进行菌斑PCR鉴定,筛选酵母阳性菌株。将筛选出的阳性pGBKT7空载酵母菌株和pGBKT7-MeISA2酵母菌株分别接种到SD/-Trp液体培养基中,30 ℃培养至OD600=0.6。将酵母菌液按照1∶1、1∶10、1∶100、1∶1000、1∶10 000稀释后,分别吸取5 μL点板于SD/-Trp固体培养基上,30 ℃倒置培养2~3 d,观察菌落生长情况。
将pGADT7空载和pGBKT7-MeISA2重组质粒共转到AH109酵母感受中,均匀涂布于SD/-Trp-Leu固体培养基上,30 ℃培养2~3 d,挑取单菌落分别使用AD检测-F/R、BD检测-F/R进行PCR检测,筛选阳性菌株。将筛选的阳性菌株接种到SD/-Trp-Leu液体培养基中扩大培养,分别吸取菌液5 μL点到SD/-Trp-Leu、SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal固体培养基上,将本实验室保存的pGBKT7-p53+pGADT7-T(阳性对照)和pGBKT7-lam+pGADT7-T(阴性对照)点板在上述固体培养基上,30 ℃培养2~3 d,观察酵母菌落生长情况。
将30 μg SC8木薯cDNA文库质粒与pGBKT7-MeISA2重组质粒共转到AH109酵母感受态,并涂布于SD/-Trp-Leu- His固体培养基,30 ℃倒置培养2~3 d。挑取酵母菌斑在20 μL 0.9% NaCl溶液中吸打混匀,吸取5 μL点板于SD/-Trp-Leu-His-Ade+X-α-gal培养基上,倒置培养2~3 d,观察酵母菌斑生长情况。挑取培养基上变蓝的菌斑,使用AD检测-F/R进行PCR检测,Sanger测序,获得候选互作蛋白信息。
根据候选互作蛋白信息搜索木薯基因库数据,获得候选蛋白的CDS序列。设计带有同源重组接头的特异性引物(表1),以SC8木薯cDNA为模板扩增候选蛋白基因编码区。通过同源重组技术将候选互作蛋白克隆到pGADT7空载载体上。将pGBKT7-MeISA2诱饵蛋白载体与含有候选蛋白基因的pGADT7载体分别共转到AH109酵母感受态,并点板到SD/-Trp-Leu、SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal固体培养基上,30 ℃倒置培养2~3 d,观察酵母生长情况。
利用本实验室已经完成的SC8木薯的体胚、愈伤、韧皮部、木质部、须根、茎、嫩叶、成熟叶的转录组信息分析互作蛋白基因在木薯各组织或器官的表达情况。
以SC8木薯的cDNA为模板,PCR扩增MeISA2基因的CDS区,凝胶电泳检测显示获得约2600 bp的条带,且条带明亮单一(图1),符合预期的MeISA2基因长度(2652 bp)。纯化PCR产物,通过同源重组技术将MeISA2基因克隆到pGBKT7载体上。PCR筛选阳性菌落,测序,提取序列比对正确的pGBKT7-MeISA2诱饵载体质粒备用。
将构建成功的pGBKT7-MeISA2诱饵载体与pGBKT7空载分别转到AH109酵母感受态中,PCR鉴定阳性菌株。梯度稀释后点板于SD/-Trp固体培养基培养。结果显示,含有pGBKT7-MeISA2和pGBKT7的酵母菌株生长情况类似(图2),表明pGBKT7-MeISA2表达蛋白不具有毒性,酵母菌株能正常生长。
为了验证诱饵蛋白MeISA2是否具有自激活能力,将pGBKT7-MeISA2诱饵载体与pGADT7空载共转AH109酵母菌株进行自激活检测。结果如图3所示,pGBKT7-p53+pGADT7-T(阳性对照)和pGBKT7-lam+pGADT7-T(阴性对照)酵母菌株和阳性菌株均能在SD/-Trp-Leu培养基上正常生长;而在SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal培养基上只有阳性对照酵母菌株能正常生长,且在SD/-Trp-Leu-His-Ade+X-α-gal培养基上变蓝;含有pGBKT7-MeISA2质粒的菌株和阴性对照菌株均不能生长(图3)。此结果表明MeISA2诱饵蛋白无自激活能力。
使用实验室保存的SC8 cDNA文库质粒筛选MeISA2的互作蛋白,在SD/-Trp-Leu-His-Ade+X-α-gal培养基上共获得7个变蓝的酵母菌斑,表明7个变蓝的酵母菌斑所表达的蛋白可能与MeISA2蛋白存在互作关系。对变蓝菌斑进行PCR检测并进行Sanger测序,通过NCBI网站序列比对共获得7个候选蛋白信息(表2)。
为进一步验证候选蛋白是否与MeISA2蛋白存在互作关系,将上述7个含有候选蛋白基因的pGADT7载体分别与pGBKT7-MeISA2诱饵载体共转到AH109酵母菌株。将酵母菌株点到SD/-Trp-Leu、SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal固体培养基上培养。结果显示,所有酵母菌株均能在SD/-Trp-Leu培养基上正常生长,而含pGBKT7-MeISA2诱饵载体、pGADT7-MePsbO1载体的酵母菌和pGBKT7-p53+pGADT7-T(阳性对照)在SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal固体培养基上均能正常生长,且在SD/-Trp-Leu-His-Ade+X-α-gal固体培养基上变蓝(图4)。此结果表明MePsbO1与MeISA2存在互作关系。
生物信息学分析结果显示,MePsbO1基因开放阅读框长度为1005 bp,由2个外显子和1个内含子组成(图5),编码334个氨基酸残基,分子质量为35 397.84 Da。其中,亮氨酸丙氨酸(Ala)和苏氨酸(Thr)的数量最多,均为33个氨基酸,占总量的9.3%;色氨酸(Trp)的含量最低,只有1个,仅占0.3%。有38个带负电荷的Asp和Glu残基,以及36个带正电荷的Arg和Lys残基,理论等电点(pI)为5.57,表明MePsbO1是一种酸性蛋白。亚细胞定位预测MePsbO1定位于叶绿体。系统进化树分析发现,MePsbO1与橡胶树的HbPsbO1亲缘关系最近,与拟南芥AtPsbO1的亲缘关系最远(图6)。蛋白的多序列结果比对显示,MePsbO1和HbPsbO1的相似性为90.12%,与AtPsbO1的相似性仅为77.71%(图7)。
分析SC8木薯品种各组织器官转录组数据,探究MePsbO1基因表达的模式,结果发现MePsbO1基因在木薯嫩叶中的表达量最高,其次是成熟叶,在茎中也具有较高的表达量,但是在体胚、愈伤、块根韧皮部、块根木质部、须根中的表达量极低(图8)。此表达模式符合植物PsbO1基因参与叶片光合作用的功能特征。
异淀粉酶参与淀粉颗粒的形成和降解[16]。在马铃薯的研究中发现,ISA1和ISA2结合形成异二聚酶参与淀粉的合成[17]。ISA1和ISA2除了能结合形成异多聚体,ISA1还能形成有活性的同源多聚酶复合物[13],ISA2则是非催化亚基,多种植物证明ISA2蛋白活性位点有氨基酸的替换[11]。研究发现ISA3与ISA1、ISA2形成的多聚体未存在密切关联,认为ISA3是以单体或与其他蛋白结合发挥作用[17]。与野生型相比,拟南芥AtISA1、AtISA2和双突变体淀粉含量的积累减少,但可溶性葡萄糖增加[18]。单子叶植物玉米中发现在ISA1ISA2突变体中,ISA1突变导致支链淀粉的合成产生了显著变化,而在ISA2突变体对支链淀粉的合成影响不大[19]。由此推测ISA1和ISA2是双子叶植物中ISA活性所需要的,但在单子植物中,ISA2不是ISA活性所必需的。
植物光系统II(PSII)的外周蛋白是与PSII膜内核心复合体相连的蛋白质,主要位于类囊体腔的一侧,在氧气进化复合物(oxygen evolving complex,OEC)的功能和PSII的稳定性中发挥重要作用,主要的外周蛋白包括PsbO、PsbP、PsbQ,其中PsbO是33 kDa锰稳定蛋白(manganesestabilizing protein,MSP),是PSII中最主要的外周蛋白[20]。PsbO在光系统Ⅱ捕捉并利用光能将水分子分解成氧气、质子和电子的过程中起辅助和强化作用,促进光合作用电子传递过程的高效运行[21]。PsbO还具有多种辅助功能,包括钙结合、锰稳定结合,以及与GTP结合相关的PSII反应中心D1亚基的GTP依赖性降解,PsbO可结合GTP,并催化GTP水解[22]。拟南芥存在2种PsbO蛋白:AtPsbO1和AtPsbO2[23]。AtPsbO1缺失突变体(psbo1)的光系统Ⅱ功能受损,表现为生长迟缓、叶片浅绿、对光失活更敏感以及D1蛋白周转速率加快[24]
本研究通过酵母双杂交筛选获得MeISA2的互作蛋白MePsbO1,该蛋白与同属大戟科的橡胶树HbPsbO1的亲缘关系最近,相似性达到90.12%,而与拟南芥AtPsbO1的相似性仅为77.71%。预测MePsbO1定位于叶绿体,且MePsbO1基因主要在叶片中表达,推测该基因在木薯光合作用中具有重要作用。叶绿体是临时性淀粉合成的主要场所,光合作用固定的碳水化合物可以淀粉的形式贮存在叶绿体中[25]。ISA在植物叶片临时性淀粉的合成中起到关键作用,通过调节淀粉的分支结构,促进结晶化和颗粒形成[26]。因此推测,MePsbO和MeISA2可能通过互作协同调控光合作用速率以及叶片临时性淀粉的积累,影响木薯源器官的光合产物积累和分配。本课题组在后续研究中将通过双分子荧光互补实验、荧光素酶互补实验进一步研究MePsbO1和MeISA2的互作机理,采用过表达和基因编辑明确它们在木薯临时性淀粉合成过程的关系及作用。
