Natural rubber is a key and widely used industrial raw material, which is mainly produced from Hevea brasiliensis. Cis-prenyltransferase (CPT) is an important component of natural rubber biosynthesis in laticiferous cells. It has been reported that HbCPT7 and HbCPT8 are specifically and highly expressed in latex, which is the cytoplasm of laticifers, and HblMYB19 and HblMYB44 can significantly promote the expression of HbCPT8 in tobacco and yeast. However, the transcription factors that regulate the specific expression of HbCPT7 in latex are still not clear. Here, we found there are several MYB- and MYC-binding sites located in promoter region of HbCPT7. We totally identified 226 HbMYBs and 19 HbMYCs in Hevea by using a genome-wide scanning approach. The transcriptome data showed that HbMYB1, HbMYB2, HbMYC1, HbMYC2, HbMYC3, HbMYC4 and HbMYC5 were highly expressed in latex. HbMYB1, HbMYB2, HbMYC1, HbMYC4 and HbMYC5 were successfully cloned from the latex of rubber trees. Through yeast one-hybrid assays, it was discovered that HbMYC5 could bind to the promoter regions of 1–1000 bp and 1501–1971 bp upstream of the start codon of HbCPT7. The results would provide a new insight into the molecular mechanism of HbCPT7 in regulating natural rubber biosynthesis.

天然橡胶是重要的工业原料,广泛应用于生产生活中。在橡胶树(Hevea brasiliensis)的产胶细胞乳管中,顺式异戊烯基转移酶(cis-prenyltransferase,CPT)是催化天然橡胶合成的关键组分。已报道的HbCPT7HbCPT8在胶乳中特异表达,并且在酵母和烟草中的HblMYB19和HblMYB44能够显著促进HbCPT8的表达。然而,调控HbCPT7胶乳特异表达的转录因子尚不清楚。本研究发现HbCPT7的启动子区具有多个MYB和MYC转录因子结合的顺式作用元件,通过生物信息学分析,在橡胶树全基因组水平上鉴定到226个HbMYBs和19个HbMYCs转录因子,其中HbMYB1HbMYB2HbMYC1HbMYC2HbMYC3HbMYC4HbMYC5在胶乳中高表达。本研究成功克隆了HbMYB1HbMYB2HbMYC1HbMYC4HbMYC5,并通过酵母单杂交实验证实HbMYC5能够结合HbCPT7起始密码子上游1~1000 bp和1501~1971 bp的启动子区域。本研究为解析HbCPT7调控天然橡胶合成的分子机制提供新的线索。

天然橡胶是重要的工业原料,广泛应用于生产生活中。在橡胶树(Hevea brasiliensis)的产胶细胞乳管中,顺式异戊烯基转移酶(cis-prenyltransferase,CPT)是催化天然橡胶合成的关键组分。已报道的HbCPT7HbCPT8在胶乳中特异表达,并且在酵母和烟草中的HblMYB19和HblMYB44能够显著促进HbCPT8的表达。然而,调控HbCPT7胶乳特异表达的转录因子尚不清楚。本研究发现HbCPT7的启动子区具有多个MYB和MYC转录因子结合的顺式作用元件,通过生物信息学分析,在橡胶树全基因组水平上鉴定到226个HbMYBs和19个HbMYCs转录因子,其中HbMYB1HbMYB2HbMYC1HbMYC2HbMYC3HbMYC4HbMYC5在胶乳中高表达。本研究成功克隆了HbMYB1HbMYB2HbMYC1HbMYC4HbMYC5,并通过酵母单杂交实验证实HbMYC5能够结合HbCPT7起始密码子上游1~1000 bp和1501~1971 bp的启动子区域。本研究为解析HbCPT7调控天然橡胶合成的分子机制提供新的线索。

Natural rubber is a key and widely used industrial raw material, which is mainly produced from Hevea brasiliensis. Cis-prenyltransferase (CPT) is an important component of natural rubber biosynthesis in laticiferous cells. It has been reported that HbCPT7 and HbCPT8 are specifically and highly expressed in latex, which is the cytoplasm of laticifers, and HblMYB19 and HblMYB44 can significantly promote the expression of HbCPT8 in tobacco and yeast. However, the transcription factors that regulate the specific expression of HbCPT7 in latex are still not clear. Here, we found there are several MYB- and MYC-binding sites located in promoter region of HbCPT7. We totally identified 226 HbMYBs and 19 HbMYCs in Hevea by using a genome-wide scanning approach. The transcriptome data showed that HbMYB1, HbMYB2, HbMYC1, HbMYC2, HbMYC3, HbMYC4 and HbMYC5 were highly expressed in latex. HbMYB1, HbMYB2, HbMYC1, HbMYC4 and HbMYC5 were successfully cloned from the latex of rubber trees. Through yeast one-hybrid assays, it was discovered that HbMYC5 could bind to the promoter regions of 1–1000 bp and 1501–1971 bp upstream of the start codon of HbCPT7. The results would provide a new insight into the molecular mechanism of HbCPT7 in regulating natural rubber biosynthesis.

天然橡胶(顺式-1,4-聚异戊二烯)是重要的工业原料,也是不可或缺的战略资源,主要来自巴西橡胶树(Hevea brasiliensis),因其具有优越的化学和物理特性,如良好的弹性、绝缘性、较强的抗撕裂、耐摩擦以及抗冲击等性能,广泛应用于国防军工、交通运输、机械制造以及医疗卫生等领域[1-3]。目前市场上以天然橡胶为原料的产品多达4万余种[4],特别在军用航空轮胎中,天然橡胶是不可替代的原材料。
天然橡胶的生物合成主要由4个阶段组成:合成异戊烯基焦磷酸(isopentenyl pyrophosphate,IPP)及其同分异构体(dimethylallyl diphosphate,DMAPP)、缩合IPP/DMAPP形成线性的类异戊二烯(geranyl diphosphate,GPP;farnesyl diphosphate,FPP;geranylgeranyl diphosphate,GGPP)、进一步缩合IPP/DMAPP和线性的类异戊二烯形成聚异戊二烯链、修饰聚异戊二烯链[5]。IPP主要通过植物细胞质中的甲羟基戊酸途径(mevalonic acid,MVA)合成,异戊烯焦磷酸异构酶(isopentenyl diphosphate isomerase,IPPI)催化IPP形成DMAPP,二者在香叶基二磷酸合酶(diphosphate synthase,GPS)、法尼基焦磷酸合酶(farnesyl diphosphate synthase,FPS)和香叶基香叶基焦磷酸合酶(geranylgeranyl pyrophosphate synthase,GGPS)的催化下产生天然橡胶的起始底物,即香叶基焦磷酸(GPP)、法尼基焦磷酸(FPP)和香叶基香叶基焦磷酸(GGPP)[5]。橡胶粒子是天然橡胶合成和储存的场所,其表面的顺式异戊烯基转移酶(cis-prenyltransferase,CPT)具有橡胶转移酶的催化活性,能聚合IPP/DMAPP和GPP、FPP或GGPP形成天然橡胶的基本骨架,即顺式-1,4-聚异戊二烯[6-8]。研究发现,Hevea brasiliensis CPT7HbCPT7)和CPT8HbCPT8)在胶乳中特异高表达[9]HbCPT7受割胶诱导表达[10]HbCPT8的表达水平与天然橡胶产量显著正相关[11],说明HbCPT7HbCPT8与天然橡胶的生物合成密切相关。
AOKI等[12]分析了HRT1HbCPT8)的启动子区,发现其启动子区含有MYB和bZIP转录因子的结合位点,推测该基因的表达水平可能受转录因子的调控。WANG等[13]发现转录因子HblMYB19和HblMYB44在橡胶树乳管中高丰度表达,并能够在酵母和烟草中激活HbCPT8的表达,认为HblMYB19和HblMYB44可能是调控HbCPT8乳管特异表达的重要因子。然而,调控HbCPT7乳管特异表达的转录因子尚未鉴定。
MYB是真核生物中一类高度保守的转录因子超家族,通过MYB保守结构域结合下游基因的启动子区来调控基因的表达水平[14]。MYB保守结构域含有1~4个由52个氨基酸组成的重复序列(R),每一个R形成3个α螺旋,调控MYB转录因子与顺式作用元件的识别[14]。根据R的数目,MYB转录因子可分为R2R3、1R、R1R2R3和4R四种类型[14]。MYC亦是真核生物中一类高度保守的转录因子家族,其氮端和碳端分别含有1个HLH-MYC_N和bHLH(basic helix-loophelix)保守结构域[10]。本研究通过分析HbCPT7启动子序列的顺式作用元件,发现含有MYB和MYC转录因子的结合位点。为鉴定调控HbCPT7的MYB和MYC转录因子,利用生物信息学分析,从橡胶树基因组中分别鉴定到226个HbMYBs和19个HbMYCs转录因子,其中HbMYB1HbMYB2HbMYC1HbMYC2HbMYC3HbMYC4HbMYC5在胶乳中特异表达;成功克隆了HbMYB1HbMYB2HbMYC1HbMYC4HbMYC5,并采用酵母单杂交的方法证实HbMYC5能够结合HbCPT7起始密码子上游1~1000 bp和1501~1971 bp的启动子区域。本研究结果为解析HbCPT7调控天然橡胶生物合成的分子机制提供新的线索。
采用CTAB法提取橡胶树叶片的DNA并以其为模板,参考热研7-33-97基因组中CPT7的序列设计特异性引物(表1),在PrimeSTAR® HS DNA Polymerase(R010Q)DNA聚合酶的作用下克隆HbCPT7的启动子序列。将克隆的目的基因片段送至海南楠山生物技术有限公司进行Sanger测序,测序正确的片段即为HbCPT7的启动子序列。利用PlantCARE(https://bioinformatics.psb.ugent.be/webtools/plantcare/html/)在线软件预测其顺式作用元件。
橡胶树MYB转录因子鉴定:分别以拟南芥R2R3型MYB转录因子AtMYB3(At1g22640)、3R型MYB转录因子AtMYB3R3(At3g09370.3)、4R型MYB转录因子ATMYB4R1(At3g18100)以及MYB-related蛋白中CCA1-like亚类的LATE ELONGATED HYPOCOTYL(At1g01060)、CPC-like亚类的AtMYB-like 2(At1g71030.1)、I-box-binding-like亚类的AtRL6(At1g75250)和R-R-type亚类的AtMYBL(At1g49010)的氨基酸序列为query序列[15],利用TBtools软件,采用BLASTp的序列比对方法,从热研7-33-97的基因组数据库中检索同源序列(p-value <E-20[16]。橡胶树MYC转录因子鉴定:以拟南芥MYC转录因子AtMYC2(At1g32640.1)的氨基酸序列为query序列,利用TBtools软件,采用BLASTp的序列比对方法,从热研7-33-97的基因组数据库中检索同源序列。At1g22640、At3g09370.3、At3g18100、At1g01060、At1g71030.1、At1g75250、At1g49010和At1g32640.1的氨基酸序列下载自Tair(https://www.arabidopsis.org/)。利用课题组已发表的热研7-33-97的转录组数据[9]和TBtools软件分析HbMYBsHbMYCs的表达水平。利用NCBI-Conserved domain database(https://www.ncbi.nim.nih.gov/guide/domins-Structures/)在线工具分析HbMYBs和HbMYCs的保守结构域。使用TRIzol(ThermoFisher)试剂提取橡胶树胶乳的RNA,取1 μg总RNA,在Hiscript 1 st Strand cDNA Synthesis kit(+gDNA wiper)(R312)试剂盒的作用下获得胶乳cDNA,并以其为模板,设计特异性引物(表1),利用PrimeSTAR® HS DNA Polymerase(R010Q)DNA聚合酶扩增HbMYB1HbMYB2HbMYC1HbMYC4HbMYC5的编码序列(coding sequence,CDS)。
利用限制性内切酶Sma Ⅰ和Sal Ⅰ线性化pAbAi载体,利用同源重组引物(表1)扩增HbCPT7启动子目标片段,采用ClonExpress Ultra One Step Cloning Kit(C115-02,Vazyme)试剂盒构建pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000、pAbAi-pHbCPT7-1001-1500、pAbAi-pHbCPT7-1501-1971载体。利用限制性内切酶Bbs I线性化pAbAi(空载体)、p53-AbAi(阳性对照)、重组载体pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000、pAbAi-pHbCPT7-1001-1500和pAbAi-pHbCPT7-1501-1971的质粒,具体操作参照TaKaRa Bio公司的Matchmaker® GoldYeast One-Hybrid Library Screening System User Manual说明书手册,随后转化Y1H酵母感受态细胞。取100 μL转化后的酵母菌液涂布于SD/-Ura平板上,30 ℃暗培养3~5 d,直至长出菌斑。取适量酵母菌斑,加入配置好的酵母破壁酶(25 μL酵母破壁酶,470 μL山梨醇缓冲液,5 μL巯基还原剂),30 ℃处理1~2 h。以处理后的菌液为模板进行PCR,参考酵母基因组DNA提取试剂盒(DP307)说明书操作,进而检测启动子片段是否整合至酵母基因组中。分别配制含有0、100、200、300、400、500、700、800 ng/mL金担子素(aureobasidin A,AbA)的SD/-Ura培养基。每个载体选取1个阳性克隆进行活化,将活化后的菌液(OD600=0.2)分别稀释10、100、1000倍。分别取5 μL稀释后的菌液滴至含有不同浓度AbA的SD/-Ura平板上,30 ℃暗培养3~5 d,观察长斑情况。
从橡胶树胶乳的cDNA中克隆HbMYB1、HbMYB2、HbMYC1HbMYC4HbMYC5目的基因片段,使用带酶切位点的引物扩增插入的目的基因片段,酶切载体(载体为pGADT7,使用EcoR Ⅰ和BamH Ⅰ进行双酶切)后,利用ClonExpress Ultra One Step Cloning Kit(C115-01)同源重组酶连接插入片段和酶切后的载体,产物转入DH5α感受态细胞。一端使用载体引物,另一端使用插入片段引物对产物进行PCR检测,筛选阳性克隆送至海南楠山生物技术有限公司进行Sanger测序,选取序列正确的阳性单克隆摇菌,提取重组质粒AD-HbMYB1、AD-HbMYB2、AD-HbMYC1、AD-HbMYC4和AD-HbMYC5,同时,提取载体pGADT7的质粒,具体操作参照TaKaRa Bio公司的Matchmaker® GoldYeast One-Hybrid Library Screening System User Manual手册。将pAbAi-pHbCPT7-1-500、pAbAi- pHbCPT7-501-1000、pAbAi-pHbCPT7-1501-1971分别转化至酵母菌株中,将转化后的酵母菌液点板至SD/-Leu和SD/-Leu/AbA(500 ng/mL)的平板上,30 ℃暗培养3~5 d,观察长斑情况。
以橡胶树叶片DNA为模板,扩增出HbCPT7基因起始密码子上游1971 bp的启动子区(图1A)。使用PlantCARE软件预测该区域的顺式作用元件发现,123个核心顺式作用元件(TATA-box和CAAT-box)、6个光响应元件(AE-box、chs-CMA2a、G-box、GT1-motif)、2个蛋白质代谢调控元件(O2-site)、1个细胞周期调节相关元件(MSA-like)、6个激素响应相关元件(GARE-motif、STRE、ABRE、ERE)、4个胁迫相关响应元件(TC-rich、WUN-motif)、12个MYB结合位点和5个MYC结合位点(图1B)。
从热研7-33-97的基因组数据库中共检索到258条含有MYB保守结构域的序列,去除重复的和氨基酸数目小于100的序列,共得228个预测蛋白。通过保守结构域分析,发现226个预测蛋白均具有MYB转录因子的保守结构域,将上述蛋白命名为HbMYB1~HbMYB226。
对226个HbMYB蛋白进行进化分析,发现橡胶树MYB转录因子可分为3个亚家族,其中Group 1和Group 2中的MYB转录因子属于1R类型,除HbMYB211和HbMYB225为R1R2R3型MYB以及HbMYB217为4R型MYB外,其余Group 3的HbMYBs均是R2R3型的MYB转录因子(图2A)。利用热研7-33-97树皮、叶片和胶乳的转录组数据[9],分析226个HbMYB基因的表达水平,发现163个HbMYB基因在3个组织中的FPKM值均小于5。分析其余基因的表达量,发现HbMYB1(EVM0021518.1)和HbMYB2(EVM0023669.1)在胶乳中高表达(图2B)。从橡胶树胶乳的cDNA中成功克隆了HbMYB1HbMYB2的编码序列,长度分别为995、1058 bp,与基因组注释的编码序列一致(图2C)。
从热研7-33-97的基因组数据库中共检索到19条编码MYC蛋白的序列,通过保守结构域分析,发现上述序列均含有bHLH-MYC_N和bHLH两个保守结构域,将其分别命名为HbMYC1-HbMYC19(图3A)。对HbMYCs的氨基酸序列进行进化分析,发现HbMYC转录因子家族可分为3个亚家族。利用热研7-33-97树皮、叶片和胶乳的转录组数据[9]分析HbMYCs基因的表达模式,结果发现,HbMYC1(EVM0032728.1)、HbMYC2(EVM0015982.1)、HbMYC3EVM0021390.1)、HbMYC4EVM0029927.1)及HbMYC5EVM0033326.1)在胶乳中特异性高表达(图3B)。随后,从橡胶树胶乳中成功克隆得到HbMYC1HbMYC4HbMYC5的编码序列,长度分别为1280、1314、1469 bp,均与基因组注释的编码序列长度一致(图3C)。
为了鉴定调控HbCPT7胶乳特异表达的HbMYBs和HbMYCs转录因子,将克隆得到的HbCPT7的启动子序列分为4部分,即pHbCPT7-1-500、pHbCPT7-501-1000、pHbCPT7-1001-1500和pHbCPT7-1501-1971(图4A),分别构建至pAbAi载体,获得诱饵载体pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000、pAbAi-pHbCPT7-1001-1500、pAbAi-pHbCPT7-1501-1971。使用限制性内切酶Bbs I线性化诱饵载体、阳性对照(p53-AbAi)以及空载体(pAbAi)并转化Y1H酵母感受态细胞。取SD/-Ura平板上的菌斑加入配置好的酵母破壁酶,30 ℃处理1~2 h,以处理后的菌液为模板进行PCR,提取转化后的酵母菌液,利用酵母基因组DNA提取试剂盒检测启动子片段是否整合至酵母基因组上,结果如图4B所示,目的片段均整合至诱饵菌株的基因组上。
将诱饵菌液(OD600=0.2)分别稀释10、100、1000倍,涂布至含有0、100、200、300、400、500、700和800 ng/mL AbA的SD/-Ura的培养基上,检测诱饵菌株的自激活活性。30 ℃暗培养3 d后发现,pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000和pAbAi-pHbCPT7-1501-1971在含有400 ng/mL AbA的SD/-Ura培养基上不生长,说明400 ng/mL的AbA能够抑制pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000和pAbAi-pHbCPT7-1501-1971的自激活活性(图4C),pAbAi-pHbCPT7-1001-1500在AbA浓度为100、200、300、400、500、700、800 ng/L的培养基上均能生长,说明上述不同浓度梯度的AbA均不能抑制pAbAi-pHbCPT7-1001-1500的自激活活性。
利用同源重组技术将HbMYB1HbMYB2HbMYC1HbMYC4HbMYC5构建至载体pGADT7上获得prey载体,即AD-HbMYB1、AD-HbMYB2、AD-HbMYC1、AD-HbMYC4和AD-HbMYC5。采用酵母单杂交的方法检测诱饵菌株pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000和pAbAi-pHbCPT7-1501-1971是否与prey载体互作。如图5所示,AD-HbMYC5与pAbAi-pHbCPT7-1-500、pAbAi-pHbCPT7-501-1000、pAbAi-pHbCPT7-1501-1971的组合均能在含有500 ng/mL AbA的SD/-Ura平板上长出菌斑,说明HbMYC5能够结合HbCPT7起始密码子上游的1~1000 bp和1501~1971 bp的启动子区域。
本研究从热研7-33-97中鉴定到226个HbMYBs和19个HbMYCs转录因子,利用热研7-33-97树皮、叶片和胶乳的转录组数据分析上述基因的表达模式,发现HbMYB1HbMYB2HbMYC1HbMYC2HbMYC3HbMYC4HbMYC5在胶乳中高丰度表达。与已发表的结果相比,HbMYB1、HbMYB2与HbMYC1、HbMYC4分别是橡胶树中新鉴定的MYB和MYC类转录因子。此外,HbMYC2HblMYC2)、HbMYC3HblMYC6)和HbMYC5HblMYC3)的表达模式与ZHANG等[10]报道的结果一致。
天然橡胶存在于橡胶树韧皮部的乳管细胞中,通过割开树皮(割胶)来收集胶乳。已有研究报道,开割树中的茉莉酸含量显著高于未开割树的,说明割胶促进内源茉莉酸含量的积累。DENG等[17]发现HbCPT7在开割树中的表达水平显著高于未开割树,并且HbCPT7在茉莉酸处理后显著上调表达,表明HbCPT7响应茉莉酸信号,也说明HbCPT7在胶乳中特异高表达可能是由于茉莉酸的诱导。MYC转录因子可以与E3泛素连接酶CORONATINE INSENSITIVE1(COI1)、茉莉酸ZIM结构域抑制因子Jasmonate-ZIM(JAZ)形成COI1/JAZ/MYC复合体,参与茉莉酸信号途径。当植物体内的茉莉酸积累时,茉莉酸活性物质(JA-Ile)结合JAZ-COI1复合体使JAZ被泛素化和解离,进而被26S蛋白酶体降解,最终释放出MYC,随后MYC在转录水平调控响应茉莉酸信号的基因,因此MYC是调节茉莉酸信号的重要蛋白[16]。ZHANG等[10]用茉莉酸替代物(冠菌素,Coronatine)处理橡胶树0.5~4 h[18-20],显著提高了形成层中HblMYC3HbMYC5)的表达水平,说明HblMYC3HbMYC5)能够响应茉莉酸信号[10]。本研究发现HbMYC5在酵母中能够与HbCPT7的启动子序列结合,进而推测橡胶树割胶后,内源茉莉酸可能通过诱导HbMYC5的表达调控胶乳中HbCPT7的表达水平。此外,HbMYC2[18]和HbMYC2b[21]分别显著促进HbFPS和小橡胶粒子基因(Hevea brasiliensis small rubber particle proteinHbSRPP)的表达,其中HbSRPP是胶乳中丰度最高的蛋白,在抑制胶乳凝固及维持橡胶粒子的稳定性等方面发挥重要作用,进一步说明MYC是调控橡胶树产胶相关基因表达的重要转录因子。
AOKI等[12]分析了橡胶树乳管特异表达基因HbREF1HbSRPP1HRT1HbCPT8)的启动子区,发现启动子区含有MYB和bZIP转录因子的结合位点,推测上述基因的表达水平可能受MYB转录因子的调控。HbWRKY1、HbWRKY3、HbWRKY14、HbWRKY55、HblMYB19、HblMYB44、HbMYC2b以及HbMADS4等转录因子均能与HbSRPP1的启动子序列结合,促进或抑制HbSRPP1的表达,表明上述转录因子协同调控HbSRPP在胶乳中高丰度表达[8]。本研究利用酵母单杂交的方法,未发现HbMYB1、HbMYB2与HbCPT7启动子直接结合,表明在天然橡胶的合成过程中HbMYB1和HbMYB2可能不参与调控HbCPT7的表达。综上所述,本研究克隆分析了HbCPT7的启动子序列,鉴定了在橡胶树胶乳中高丰度表达的MYB和MYC类转录因子,并发现HbMYC5能与HbCPT7的启动子序列结合,研究结果为解析HbCPT7调控天然橡胶合成的分子机理提供新的思路。
