LEC2 is an important member of the LAV subfamily in the plant B3 superfamily and plays a significant regulatory role in embryo development and meristem maintenance. To explore the function of the PvoLEC2 in the cell regeneration of Plukenetia volubilis, the genomic sequence of PvoLEC2 (gPvoLEC2) was cloned through phylogenetic analysis and molecular biology techniques. The function of gPvoLEC2 in promoting plant regeneration was verified in Nicotiana benthamiana, and the function of gPvoLEC2 was studied in P. volubilis hairy roots. Phylogenetic and gene structure analysis indicated that the LEC2-ABI3 subfamily of Euphorbiaceae plants such as P. volubilis, and Arabidopsis thaliana, Oryza sativa, Populus spp. were relatively conserved. Expression analysis showed that PvoLEC2 was only weakly expressed in the late stage of P. volubilis seed development (50-65 DAP), and the motifs and key amino acid sites of the B3 domain were conserved. Overexpression of gPvoLEC2 led to dwarfing of transgenic N. benthamiana plants, difficulty in the growth of the main stem, and abnormal leaf development. At the same time, the regeneration ability of gPvoLEC2 transgenic N. benthamiana organs in vitro was significantly higher than that of the wild type, but lower than that of the positive control-ZmWUS2 & STM transgenic N. benthamiana. Overexpression of gPvoLEC2 in P. volubilis hairy roots led to their shortening and thickening. The results indicate that the genomic sequence of PvoLEC2 is a functional sequence that promotes plant regeneration, but it also causes abnormal development.

LEC2是植物B3超家族中LAV亚家族的重要成员,在胚胎发育以及分生组织维持等过程中起着重要的调控作用。为了探究LEC2基因在星油藤细胞再生过程中的作用,本研究通过系统进化等生物信息学分析和分子生物学技术克隆星油藤PvoLEC2的基因组序列(gPvoLEC2),利用本氏烟草验证gPvoLEC2促进植物再生的功能,并且对星油藤毛状根中的gPvoLEC2的功能进行研究。系统进化和基因结构分析表明,星油藤等大戟科植物与拟南芥、水稻、杨树的LAV亚家族中LEC2-ABI3亚群比较保守。表达分析显示,PvoLEC2仅在星油藤种子发育晚期(50~65 DAP)微弱表达,且B3结构域的基序及关键氨基酸位点保守。过表达gPvoLEC2导致转基因本氏烟草植株矮化、主茎难以长高、叶片发育畸形,同时gPvoLEC2转基因本氏烟草器官离体培养的再生能力明显高于野生型,但低于阳性对照——ZmWUS2 &STM转基因本氏烟草,而在星油藤毛状根中过表达gPvoLEC2导致其缩短、增粗。本研究结果表明星油藤PvoLEC2的基因组序列是一段具有促进植物再生的功能序列,但同时也会导致植物发育畸形。

LEC2是植物B3超家族中LAV亚家族的重要成员,在胚胎发育以及分生组织维持等过程中起着重要的调控作用。为了探究LEC2基因在星油藤细胞再生过程中的作用,本研究通过系统进化等生物信息学分析和分子生物学技术克隆星油藤PvoLEC2的基因组序列(gPvoLEC2),利用本氏烟草验证gPvoLEC2促进植物再生的功能,并且对星油藤毛状根中的gPvoLEC2的功能进行研究。系统进化和基因结构分析表明,星油藤等大戟科植物与拟南芥、水稻、杨树的LAV亚家族中LEC2-ABI3亚群比较保守。表达分析显示,PvoLEC2仅在星油藤种子发育晚期(50~65 DAP)微弱表达,且B3结构域的基序及关键氨基酸位点保守。过表达gPvoLEC2导致转基因本氏烟草植株矮化、主茎难以长高、叶片发育畸形,同时gPvoLEC2转基因本氏烟草器官离体培养的再生能力明显高于野生型,但低于阳性对照——ZmWUS2 &STM转基因本氏烟草,而在星油藤毛状根中过表达gPvoLEC2导致其缩短、增粗。本研究结果表明星油藤PvoLEC2的基因组序列是一段具有促进植物再生的功能序列,但同时也会导致植物发育畸形。

LEC2 is an important member of the LAV subfamily in the plant B3 superfamily and plays a significant regulatory role in embryo development and meristem maintenance. To explore the function of the PvoLEC2 in the cell regeneration of Plukenetia volubilis, the genomic sequence of PvoLEC2 (gPvoLEC2) was cloned through phylogenetic analysis and molecular biology techniques. The function of gPvoLEC2 in promoting plant regeneration was verified in Nicotiana benthamiana, and the function of gPvoLEC2 was studied in P. volubilis hairy roots. Phylogenetic and gene structure analysis indicated that the LEC2-ABI3 subfamily of Euphorbiaceae plants such as P. volubilis, and Arabidopsis thaliana, Oryza sativa, Populus spp. were relatively conserved. Expression analysis showed that PvoLEC2 was only weakly expressed in the late stage of P. volubilis seed development (50-65 DAP), and the motifs and key amino acid sites of the B3 domain were conserved. Overexpression of gPvoLEC2 led to dwarfing of transgenic N. benthamiana plants, difficulty in the growth of the main stem, and abnormal leaf development. At the same time, the regeneration ability of gPvoLEC2 transgenic N. benthamiana organs in vitro was significantly higher than that of the wild type, but lower than that of the positive control-ZmWUS2 & STM transgenic N. benthamiana. Overexpression of gPvoLEC2 in P. volubilis hairy roots led to their shortening and thickening. The results indicate that the genomic sequence of PvoLEC2 is a functional sequence that promotes plant regeneration, but it also causes abnormal development.

星油藤(Plukenetia volubilis L.)是大戟科(Euphorbiaceaec)多年生木质藤本植物,又名美藤果、南美油藤、印奇果等,原产于南美洲的热带雨林,是一种热带油料作物,与油用牡丹(Paeonia suffruticosa)、元宝枫(Acer truncatum Bunge)并称为三大新兴木本油料作物[1]。冷榨的星油藤油有丰富的亚麻酸、亚油酸、维生素E及多种微量元素,是一种优秀的新资源食品[2-3],在食用和药用方面均有广阔的应用前景[2,4]。同时,星油藤的种植周期短于油茶树(Camellia oleifera Abel)、油橄榄树(Olea europaea L.)等木本油料植物,在种植当年就可收获果实[2,5]。但由于星油藤细胞再生比较困难,限制了该物种的遗传改良。提高星油藤再生效率将有助于利用基因组编辑技术创造高产、多抗、环境友好的新品种,从而保证我国星油藤产业的可持续发展。
LAV [LEAFY COTYLEDON2(LEC2)–ABSCISIC ACID INSENSITIVE3(ABI3)–VAL(VP1/ABI3-LIKE)]亚家族属于B3超家族,由LEC2-ABI3亚群和VAL亚群组成。在拟南芥(Arabidopsis thaliana)中LAV亚家族包含6个基因,LEC2-ABI3亚群的LEAFY COTYLEDON2LEC2)、ABSCISIC ACID INSENSITIVE3ABI3)、FUSCA3FUS3)以及VAL亚群的VAL1VAL2VAL3。LEC2-ABI3亚群的基因在种子发育过程中起重要作用,这些基因在种子中高水平表达,而在其他植物器官中的表达量较低。ABI3在种子发育过程中调控脱落酸响应基因,参与种子萌发过程。LEC2FUS3基因能够促进种子胚胎发育,FUS3参与种子成熟以及种子储存蛋白表达等过程,LEC2通过调控生长素生物合成相关基因诱导体细胞胚发生[6]。研究表明,FUS3LEC2的过表达会使营养器官(或组织)出现胚性特征[7-8],如转基因拟南芥真叶具有子叶特征,无表皮毛等。与LEC2-ABI3亚群不同,VAL亚群的基因在植物生长发育过程中,在多个植物器官中表达,抑制细胞再生。当前对VAL亚群的3个成员中研究较多的是VAL1VAL2[9]val1val2双突变体植株在根和茎上长出胚性愈伤组织,检测发现FUS3LEC2val1val2双突变体中显著上调表达,这表明VAL1VAL2是植物组织再生的负调控因子[10]
LEC2基因是LAV亚家族中被充分研究的基因之一,最初是在模式植物拟南芥中过表达LEC2基因发现其能够促进体细胞向胚性细胞转变,诱导体细胞胚胎发生[8]。研究表明LEC2过表达诱导体细胞胚胎产生的原因可能是LEC2基因能够诱导IPA-YUC生长素合成途径中YUC1YUC4YUC10等关键调控因子的表达[6]。同时,过表达LEC2还可以有效提高种子含油量,在拟南芥中过表达蓖麻(Ricinus communis L.)RcLEC2基因,能激活油体蛋白相关基因的表达,种子含油量增加约2.7倍[11]。近年来发现木本植物的LEC2也具有促进体细胞胚胎发生的功能,在可可(Theobroma cacao L.)子叶中过表达TcLEC2可以显著增加体细胞胚胎发生的频率[12],在木薯(Manihot esculenta Crantz)中过表达MeLEC2,转基因木薯成熟叶的叶肉细胞可通过体细胞胚途径诱导出再生芽[13]
在破解玉米(Zea mays L.)、高粱(Sorghum bicolor L.)、小麦(Triticum aestivum L.)等作物遗传转化障碍的过程中发现,具有再生功能的发育调节基因通常来源于WUSCHEL-RELATED HOMEOBOX(WOX)、APETALA2/ETHYLENERESPONSIVE FACTOR(AP2/ERF)等基因家族,很少关注LAV亚家族。前期研究表明,星油藤的遗传转化可诱导出转基因愈伤,但难以从这些转基因愈伤中诱导出再生芽或体细胞胚胎。本研究从星油藤、蓖麻等大戟科植物中鉴定LAV亚家族,分析该基因家族成员在大戟科植物与拟南芥、水稻(Oryza sativa L.)等模式物种间的保守性,同时利用本氏烟草(Nicotiana benthamiana)验证PvoLEC2的基因组序列(gPvoLEC2)的再生功能,并且在星油藤的毛状根中对gPvoLEC2的功能进行初步研究,为提高星油藤的再生能力以及遗传改良提供优质的候选基因。
本氏烟草种子经75%乙醇和1%次氯酸钠溶液消毒后播种于MS固体培养基上,生长30 d后,叶片可用于转化实验。星油藤种子由云南普洱市宁洱哈尼族彝族自治县农户提供。星油藤种子去壳后经75%乙醇和2.5%次氯酸钠溶液消毒后播种于WPM培养基,培养7~10 d时可用于转基因星油藤毛状根诱导实验。发根农杆菌K599菌株由北京林业大学孟冬教授惠赠,根癌农杆菌GV3101菌株由本实验室保存。
星油藤、蓖麻、橡胶树[Hevea brasiliensis(Willd. ex A. Juss.)Müll. Arg.]、小桐子(Jatropha curcas L.)、木薯等大戟科物种全基因数据、转录组数据均来自EupDB数据库(http://eupdb.liu-lab.com/);拟南芥LAV亚家族序列来自拟南芥数据库TAIR(http://www.arabidopsis.org/);水稻、杨树(Populus spp.)、玉米LAV亚家族序列及数据来源于NCBI数据库。
从TAIR(http://www.arabidopsis.org/)数据库中分别下载拟南芥LAV家族各成员的氨基酸序列,在Geneious prime 2021本地BLAST插件的星油藤、蓖麻、小桐子等物种本地数据库中进行序列比对(参数为E<1e–10),获得与拟南芥相似度较高的LAV序列信息,作为候选基因。从Pfam(http://pfam.xfam.org/)数据库中下载含有B3基因家族的特征结构域PF00847的隐马尔科夫模型,使用HMMER 3.0(http://hmmer.janelia.org)在线软件建立HMM模型进行同源序列的鉴定。利用CDD(https://www.ncbi.nlm.nih.gov/cdd/)和ExPASy Proteomics Server(https://prosite.expasy.org/scanprosite/)在线软件确认候选序列具有完整的B3特征结构域的LAV亚家族基因。使用Mega 11.0软件,采用最大似然法(maximum likelihood,ML)生成系统进化树,Bootstrap重复1000次。
利用大戟科植物EupDB公共数据库中星油藤不同组织器官的RNA-seq数据,对星油藤LAV亚家族基因在不同组织器官[根(root)、茎(stem)、叶(leaf)、雌花序(female inflorescence)、雄花序(male inflorescence)、果(fruit)和种子(seed)]的表达进行分析。提取基因在各组织器官表达的FPKM值,利用GraphPad Prism 8.0[14]软件绘制表达热图。
设计特异性引物(gPvoLEC2-F: ttttctgattaacagggtaccATGGAGAACCTTTTTTCACTTTTGT,gPvoLEC2-R: tcttcatcttcatatggatccCTAAGCGCTGCACAAGTCTTCC;小写字母为Infusion同源臂),利用In-fusion技术将gPvoLEC2克隆至pW502载体获得UBI10pro::gPvoLEC2的过表达载体(图1)。
分别挑取携带目的基因UBI10pro::gPvoLEC2(简称gPvoLEC2)、NOSpro::ZmWUS2 & CmYLCVpro::STM(简称ZmWUS2 & STM图1)的根癌农杆菌GV3101单菌落接种于含50 mg/L卡那霉素(kanamycin)和20 mg/L利福平(rifampicin)的LB液体培养基中,在28 ℃、220 r/min的条件下振荡培养至OD=0.6~1.0。将菌液转移至50 mL离心管,4000 r/min离心10 min,弃上清液,用含100 μmol/L乙酰丁香酮的1/2MS液体培养基(pH 5.8)重悬农杆菌至OD=0.3~0.4。本氏烟草的侵染流程参照JANSING等[15]的方法实施。
收集T0gPvoLEC2ZmWUS2 & STM转基因烟草植株种子(T1代),经75%乙醇和1%次氯酸钠溶液消毒后播种于MS固体培养基上。待转基因本氏烟草幼苗生长至12 d,将其切分为3种外植体(子叶、下胚轴和根),并以相同生长条件下的野生型本氏烟草作为对照。将3种外植体分别放入芽诱导培养基(含1 mg/L 6-BA、0.1 mg/L NAA、200 mg/L头孢噻肟钠、100 mg/L特美汀的MS固体培养基)中诱导,培养25 d时统计再生芽诱导情况。
利用携带目的基因35S::DsRed2(简称DsRed2图1)、35S::gPvoLEC2的发根农杆菌K599侵染星油藤幼苗,参考俞璟菁等[16]的方法诱导转基因毛状根,并稍作修改。星油藤幼苗裹菌后放入含2 mg/L 6-BA、0.2 mg/L IBA、400 mg/L头孢噻肟钠、100 mg/L特美汀的WPM固体培养基,在26 ℃暗培养5 d后,转入26 ℃、16 h光照/8 h黑暗的光周期条件下培养诱导转基因星油藤毛状根。
系统进化显示,星油藤LEC2-ABI3亚群中LEC2PvoB3-69)、ABI3PvoB3-03)、FUS3PvoB3-18)与蓖麻、小桐子、橡胶树等大戟科物种的LEC2-ABI3亚群的基因均为单拷贝(图2)。玉米LEC2ABI3均有2个成员,不存在FUS3,但在水稻中存在OsFUS3aOsFUS3b两个成员。VAL亚群变化较大,供试的5种大戟科物种中的VAL3数量较少,仅有蓖麻中有1个RcVAL3;但存在多个VAL1VAL2成员,如橡胶树、木薯。星油藤VAL亚群中仅含有PvoB3-06VAL2)和PvoB3-27VAL1)2个成员。从基因结构(图2)来看,LEC2-ABI3亚群比较保守,星油藤等大戟科植物以及拟南芥、杨树等木本植物均有6个外显子,但玉米ZmLEC2aZmLEC2b以及水稻OsLEC2均含有9个外显子。VAL亚群变异幅度较大,VAL1VAL2两个进化支含有12~13个外显子。在VAL3进化枝中,拟南芥VAL3含有8个外显子,水稻和玉米的VAL3成员含有12个外显子,而蓖麻的RcVAL3有14个外显子。由此可知,星油藤LAV亚家族中LEC2-ABI3亚群的数量和基因结构与其他双子叶植物相同,功能可能比较保守。
为了研究星油藤LAV亚家族成员的表达特异性,对鉴定得到的LAV亚家族基因在根、茎、茎尖、叶、雌花序、雄花序和种子等不同组织器官中的表达特征进行分析。结果(图3)表明,PvoABI3PvoFUS3在开花后50~65 d的种子内特异性表达,且PvoABI3的表达量很高,PvoLEC2在各个组织部位的表达量均较低。PvoVAL1在果中特异性表达,在种子中也有少量低丰度的表达;PvoVAL2在根系以及雄花中表达,其中在根中的表达量较高,在茎和果中也有低丰度表达。研究表明,拟南芥LEC2在球形胚等胚胎发育早期表达,调控胚胎的形态建成。本研究选取星油藤果实发育早期的样品提取RNA,仍未检测到PvoLEC2表达,推测该基因可能在星油藤胚胎发育更早期表达。因此,克隆该基因的基因组序列gPvoLEC2用于后期功能研究。gPvoLEC2基因全长序列为2896 bp,基因结构如图2PvoB3-69所示,包含6个外显子,5个内含子(GenBank accession: PP454233)。
以拟南芥RAV1的B3结构域为模板,对星油藤PvoLEC2与橡胶、木薯、蓖麻、小桐子等物种的同源蛋白B3结构域进行多重序列比对,发现该结构域由7个β-折叠和2个α-螺旋组成(图4)。β-折叠内部的疏水链核心氨基酸Leu267、Ile282、Leu284、Val307、Trp318、Leu320、Phe322、Leu335、Leu354、Leu356、Leu366、Phe368在星油藤PvoLEC2与其他物种同源蛋白B3结构域中是保守的,α-螺旋核心氨基酸Thr339、Phe342在星油藤PvoLEC2与其他物种同源蛋白B3结构域中也是保守的。这表明不同物种LEC2同源蛋白的B3结构域的功能可能相同。
为了探究过表达gPvoLEC2是否具有促进再生的功能,对10 d的gPvoLEC2转基因无菌苗进行器官离体培养,以先前已经证明具有再生能力的ZmWUS2 & STM转基因本氏烟草作为阳性对照。离体培养25 d后发现(表1图5),WT的72片子叶中8片子叶能够诱导出再生芽,诱导率为11.1%。gPvoLEC2的126片子叶中66片子叶能够诱导出再生芽,诱导率为52.4%,且诱导出的再生芽多集中在叶基部。ZmWUS2 & STM的84片子叶中78片子叶能够诱导出再生芽,诱导率为92.9%,再生芽诱导率远高于野生型本氏烟草和gPvoLEC2转基因本氏烟草子叶。ZmWUS2 & STM转基因烟草子叶上诱导出的转基因芽更为密集,子叶的各个部位均能诱导出再生芽,并且多集中在子叶的近轴面。
对胚轴的再生情况统计显示,gPvoLEC2的63个胚轴中31个胚轴能够诱导出再生芽,诱导率为49.2%;WT的36个胚轴中5个胚轴能够诱导出再生芽,诱导率为13.9%;ZmWUS2 & STM的42个胚轴中21个胚轴能够诱导出再生芽,诱导率为50.0%。野生型本氏烟草胚轴膨大不明显,再生芽的诱导数也极少,gPvoLEC2转基因本氏烟草和ZmWUS2 & STM转基因烟草胚轴的转基因芽诱导率相当,且远高于野生型本氏烟草胚轴的再生芽诱导率。gPvoLEC2转基因本氏烟草胚轴和ZmWUS2 & STM转基因烟草胚轴诱导的再生芽主要集中在两端的切口处。
对主根的再生情况进行统计发现,野生型本氏烟草根愈伤增殖速度最慢,36条根中2条根能够诱导出再生芽,诱导率为5.6%,再生芽诱导效率最低。gPvoLEC2转基因本氏烟草根愈伤增殖速度最快,63条根中10条根能够诱导出再生芽,诱导率为15.9%,再生芽诱导率高于ZmWUS2 & STM和WT。ZmWUS2 & STM的42条根中3条根能够诱导出再生芽,诱导率为7.1%。以上结果表明,相较于野生型本氏烟草,gPvoLEC2转基因本氏烟草子叶、胚轴以及主根的再生芽诱导能力均有所提升,即gPvoLEC2具有促进植物再生的功能。
为探究过表达gPvoLEC2以及ZmWUS2 & STM对本氏烟草生长发育的影响,将ZmWUS2 & STMgPvoLEC2转基因本氏烟草的种子播种至土壤中,38 d后对T1代植株进行表型分析。结果发现,gPvoLEC2转基因本氏烟草体型与野生型存在较大差别,地上部分主茎缩短,很难伸长,植株明显矮化。gPvoLEC2转基因本氏烟草T1代植株叶片圆肥,形态不规则,部分畸形且叶脉偏心、不突出,革质化。T1ZmWUS2 & STM转基因本氏烟草呈侏儒状,体型大小趋近于WT幼年期,茎不伸长,叶片簇生,叶小而密,植株地上部分高度通常为3~5 cm,并且随着培养时间的延长也无明显生长趋势,叶片极小且密,叶片无明显卷曲(图6)。以上结果表明,过表达gPvoLEC2ZmWUS2 & STM都会导致转基因本氏烟草发育畸形,但表型变异并不一致。
为探究过表达gPvoLEC2对星油藤生长发育的影响,使用携带目的基因DsRed2gPvoLEC2ZmWUS2 & STM的发根农杆菌K599诱导星油藤转基因毛状根。最终成功诱导出DsRed2gPvoLEC2的星油藤转基因毛状根,未能诱导出ZmWUS2 & STM的星油藤转基因毛状根。由图7可知,gPvoLEC2转基因星油藤毛状根与DsRed2转基因毛状根存在较为明显的区别,相较于3条独立的DsRed2转基因星油藤毛状根,3条独立的gPvoLEC2转基因星油藤毛状根均出现了较为明显的膨大和缩短,部分毛状根出现卷曲。随着培养时间的延长,gPvoLEC2转基因毛状根很缓慢的伸长,与DsRed2转基因毛状根的细长表型呈现出明显的区别。可见,过表达gPvoLEC2抑制星油藤毛状根伸长,这与过表达抑制本氏烟草茎伸长的表型类似,但是否促进毛状根再生还有待研究。
对拟南芥、水稻、杨树、玉米以及星油藤、蓖麻、橡胶树、小桐子、木薯等大戟科物种LAV基因家族的基因进行基因结构和系统进化分析,结果显示星油藤等大戟科植物的LEC2-ABI3亚群比较保守。前期研究表明ABI3的基因结构在陆生植物进化过程中相当保守,几乎所有陆生植物ABI3基因的内含子相位都是一致的,FUS3LEC2在进化的过程中也具有较高的保守性[17],与本研究结果一致。虽然已报道LEC2-ABI3亚群的ABI3FUS3LEC2与胚胎发育有关,但ABI3参与调控植物激素ABA信号传导[18],是种子萌发的关键负调控基因[19-20]FUS3基因是调控油脂合成的植物特异性转录因子之一,在拟南芥中FUS3基因在油脂积累方面发挥重要调控作用[21]LEC2在多个物种中的同源基因均能促进体细胞向胚性细胞转变、诱导体细胞胚胎发生,如萝卜(Raphanus sativus L.)[22]、可可、木薯等。因此,本研究推测LEC2在星油藤中的同源基因PvoLEC2可能也具有促进再生的功能。
本研究通过本氏烟草验证了星油藤gPvoLEC2的再生功能,对于挖掘林木中与再生相关的发育调节基因提供了新思路。近年来利用BABY BOOMBBM)和WUSCHELWUS)等促进细胞再生功能的发育调节基因显著提高了玉米、高粱等作物的遗传转化效率[23],这些作物在以往的研究中是难以转化的。这些研究案例具备2个特点:一是使用的发育调节基因大多是已知功能的同源基因,如BBMWUS;二是被转化的物种具备基本的遗传转化条件。但是,在林木中使用发育调节基因促进遗传转化难以获得成功,可能是已知功能的发育调节基因在林木中并不一定可行。如植物细胞全能性的关键调控因子BBM基因,在甘蓝型油菜(Brassica napus)中过表达BnBBM可诱导体细胞胚胎的发生[24],在拟南芥和毛白杨(Populus tomentosa Carrière)中过表达BnBBM均能诱导体细胞胚胎的形成[24-25],在再生困难的辣椒中过表达BnBBM也能有效地促进再生[25],但林木中目前仅有苹果的遗传转化借助MdBBM取得了成功[26]。另外,林木中普遍缺乏基本的遗传转化体系,直接使用发育调节基因破解林木遗传转化障碍需要逐一尝试,工作量过大且难以成功。
据报道,常用的发育调节基因有SOMATIC EMBRYOGENESIS RECEPTOR-LIKE KINASESERK)、LEC2BBMWUSSHOOT-MERISTEMLESSSTM)等[8,23,27-29]。星油藤中这些发育调节基因的功能还无法确定,并且遗传转化步骤繁杂,转化周期长,将这些发育调节基因不经筛选直接应用的可行性较差。考虑到本氏烟草在基础生物学中的诸多优势,使用具备再生功能的ZmWUS2 & STM转基因烟草作为阳性对照,验证gPvoLEC2的再生功能,结果表明gPvoLEC2转基因本氏烟草的子叶、胚轴和主根离体培养的再生芽诱导率相较于野生型本氏烟草均有所提高。
在本氏烟草和星油藤的毛状根中过表达gPvoLEC2均导致植物(器官)发育异常,转基因本氏烟草茎缩短、明显矮化、叶片畸形,转基因星油藤毛状根缩短、增粗,二者表型类似。此前有研究显示在烟草(Nicotiana tabacum L.)中过表达LEC2基因,转基因植株表现出胚轴缩短,子叶缩小甚至畸形的现象[30-31]。在拟南芥中过表达大豆GmLEC2基因也表现出相似的表型,植株矮化,莲座叶变小[32]。在可可中过表达TcLEC2基因,虽然能够提高体细胞胚胎诱导的效率,但这些体细胞胚胎生长发育会受到影响,难以发育成完整植株[12]。在可可中过表达TcBBM可以诱导体细胞胚胎的发生,但是持续的过表达TcBBM影响可可的正常生长发育,诱导出的体细胞胚胎也无法发育成完整植株[33]。在苹果(Malus pumila Mill)中过表达MdBBM也产生了多效性影响,一方面MdBBM显著提高了苹果的遗传转化效率,另一方面转基因苹果幼苗难以生根,根系的形态产生变化,生长发育受到影响[26]。为获得正常发育的植株,在玉米中过表达BBMWUS2,有效地促进了再生的同时对启动子进行更换,获得了健康可育的植株[34]。还有研究表明,可以通过雌二醇诱导的系统来控制发育调节基因的表达,获得完整的转基因植株后关闭发育调节基因的表达,从而避免发育调节基因负面效应,该策略在不同的物种中得到了有效的应用[26,35]。因此,如何削减gPvoLEC2负面效应是该基因在星油藤基因工程的应用过程中需要攻克的难点。
本研究通过系统进化、氨基酸序列多重比对,结合本氏烟草功能验证,从星油藤LAV亚家族中鉴定了1个促进细胞再生的发育调节基因gPvoLEC2,这表明在缺乏基本遗传转化的林木中,利用本氏烟草验证发育调节基因的再生功能行之有效。在后续研究中,探讨PvoLEC2的编码区序列以及LAV亚家族其他成员在星油藤再生过程中的作用也非常必要。
