This research aimed to identify a pathogenic fungus of the brown spot disease in the leaves of Cycas and screen for its chemical control agents, which provides a theoretical basis for the protection of Cycas. The pathogenic fungus was isolated from the lesion spots in some Cycas leaves. The pathogenicity of the fungus was confirmed by isolating and purifying the infected leaf tissues and reintroduced to the healthy leaves of Cycas diannanensis following Koch’s postulates. Morphological identification of the pathogenic fungus was conducted. To further confirm the identification, a phylogenetic tree was constructed using eight gene sequences: SSU, LSU, GAPDH, tef1, rpb2, Alt, OPA10-2 and ITS. Effective chemical control agents were screened from seven chemical agents by inhibition assay in vitro. Phylogenetic analyses showed that this pathogen was Alternaria burnsii. Among the tested chemical agents, the most effective inhibition was fluoxastrobin, which displayed 73.48% inhibition rate at a concentration of 0.67 mg/L.

本研究旨在分离并鉴定造成苏铁属植物羽叶褐斑病的病原真菌,筛选出对该病原真菌具有强烈抑制作用的化学防治药剂,为苏铁的保护提供理论依据。首先,通过病斑组织分离、纯化后回接滇南苏铁叶片,根据科赫氏法完成其致病性验证。其次,对其进行形态鉴定;再应用SSULSUGAPDHtef1rpb2AltOPA10-2ITS 8个基因序列,构建多基因进化树进一步鉴定病原真菌。最后,通过平板抑菌试验,从7种化学药剂中筛选出高效的抗菌药剂。本研究发现,形态学鉴定与多基因联合进化分析表明,该病原菌为链格孢属的巴斯基链格孢(Alternaria burnsii);在7种化学药剂平板抑菌试验中,氟酰羟·苯甲唑效果最佳,其浓度在0.67 mg/L时抑菌率达到73.48%。

本研究旨在分离并鉴定造成苏铁属植物羽叶褐斑病的病原真菌,筛选出对该病原真菌具有强烈抑制作用的化学防治药剂,为苏铁的保护提供理论依据。首先,通过病斑组织分离、纯化后回接滇南苏铁叶片,根据科赫氏法完成其致病性验证。其次,对其进行形态鉴定;再应用SSULSUGAPDHtef1rpb2AltOPA10-2ITS 8个基因序列,构建多基因进化树进一步鉴定病原真菌。最后,通过平板抑菌试验,从7种化学药剂中筛选出高效的抗菌药剂。本研究发现,形态学鉴定与多基因联合进化分析表明,该病原菌为链格孢属的巴斯基链格孢(Alternaria burnsii);在7种化学药剂平板抑菌试验中,氟酰羟·苯甲唑效果最佳,其浓度在0.67 mg/L时抑菌率达到73.48%。

This research aimed to identify a pathogenic fungus of the brown spot disease in the leaves of Cycas and screen for its chemical control agents, which provides a theoretical basis for the protection of Cycas. The pathogenic fungus was isolated from the lesion spots in some Cycas leaves. The pathogenicity of the fungus was confirmed by isolating and purifying the infected leaf tissues and reintroduced to the healthy leaves of Cycas diannanensis following Koch’s postulates. Morphological identification of the pathogenic fungus was conducted. To further confirm the identification, a phylogenetic tree was constructed using eight gene sequences: SSU, LSU, GAPDH, tef1, rpb2, Alt, OPA10-2 and ITS. Effective chemical control agents were screened from seven chemical agents by inhibition assay in vitro. Phylogenetic analyses showed that this pathogen was Alternaria burnsii. Among the tested chemical agents, the most effective inhibition was fluoxastrobin, which displayed 73.48% inhibition rate at a concentration of 0.67 mg/L.

我国苏铁属植物种类丰富,主要包括:苏铁(Cycas revoluta)、叉孢苏铁(C. segmentifida)、叉叶苏铁(C. bifida)、宽叶苏铁(C. balansae)、长叶苏铁(C. dolichophylla)、单羽苏铁(C. simplicipinna)、篦齿苏铁(C. pectinata)、多歧苏铁(C. multipinnata)、锈毛苏铁(C. ferruginea)、灰干苏铁(C. hongheensis)、六籽苏铁(C. sexseminifera)、陈氏苏铁(C. chenii)、四川苏铁(C. szechuanensis)、德保苏铁(C. debaoensis)、贵州苏铁(C. guizhouensis)、台湾苏铁(C. taiwaniana)、滇南苏铁(C. diannanensis)、攀枝花苏铁(C. panzhihuaensis)、台东苏铁(C. taitungensis)和谭清苏铁(C. tanqingii[1]。由于苏铁属植物化石记录丰富,它们为古生物学、古气候学及古地理研究提供了大量证据,也为研究种子植物的起源和进化提供了重要线索[2]。苏铁属早在1974年就被纳入《濒危野生动植物物种国际贸易公约》(CITES)保护范围。在我国,苏铁属在1996年被列为一级保护植物。目前,在我国多个地区,已专门为苏铁建立自然保护区。其中,迁地保护是一种有效的保护手段。南宁植物园(南宁青秀山)从1998年至今迁入苏铁属、非洲铁属、泽米铁属等苏铁类植物1万多株[3]。苏铁病害不仅破坏其观赏价值,也是目前造成树木死亡的主要原因之一,对苏铁资源的保护利用构成严重威胁。因此,对南宁植物园进行系统性的苏铁病害调查十分有必要。
目前,已有报道表明,苏铁属真菌性病害主要包括由炭疽菌属(Colletotrichum)的两个种C. siamense[4]C. cycadis[5]侵染引起的叶斑病,由苏铁拟茎点霉(Phomopsis cycadis)引起的苏铁叶枯病[6],由苏铁壳二孢菌(Ascochyta cycadia)引起的苏铁白斑病[7],由苏铁间座壳菌(Diaporthe cycasis)引起苏铁叶枯病[8],由茄腐皮镰刀菌(Fusarium solani)引起的苏铁根茎腐烂病[9],由苏铁拟盘多毛孢(Pestalotiopsis cycadis),苏铁壳蠕孢(Stagonospora caricinella),链格孢属(Alternaria)、叶点霉属(Phyllosticta)和色二孢属(Diplodia)引起的叶斑病[10-11],由煤炱菌属(Capnodium)引起的苏铁煤污病[12]以及由壳针孢属(Septoria)引起的苏铁叶枯病[13]。曾报道印度有链格孢属(Alternaria)和胶孢炭疽菌(C. gloeosporioides)引起的苏铁叶部病害。在美国佛罗里达和巴基斯坦有茎点霉属(Phoma sp.)引起的苏铁叶枯病[14-15]
针对Colletotrichum sp.、P. cycadisA. cycadia引起的叶斑病所使用的化学防治药剂主要包括多菌灵、苯甲丙环唑和戊唑醇[3]。放线菌属(Streptomyces)及其次生代谢物对A. alternata的抑制率可达到(65.43±1.63)%[16]。但与大田作物相比,在苏铁上使用的化学药剂种类报道较少。
我们在南宁植物园的苏铁园采集了多种病样标本,其中包括发病羽片及茎秆标本,寄主包含德保苏铁(C. debaoensis)、篦齿苏铁(C. pectinata)、滇南苏铁(C. diannanensis)、叉叶苏铁(C. bifida)、叉孢苏铁(C. segmentifida)等。对所采集病害标本进行单孢分离纯化,得到82个菌株。根据ITS区域对这些真菌进行初步鉴定,这些菌株主要包括炭疽菌属(Colletotrichum)、镰刀菌属(Fusarium)、链格孢属(Alternaria)、拟盘多毛孢属(Pestalotiopsis)、叶点霉属(Phyllosticta)、黑孢霉属(Nigrospora)、枝孢菌属(Cladosporium)、球毛壳属(Chaetomium)、炭团菌属(Hypoxylon)、炭角菌属(Xylaria)、葡萄座腔菌属(Botryosphaeria)、曲霉属(Aspergilus)、小新壳梭孢属(Neofusicoccum),其中炭疽菌属、叶点霉属、镰刀菌属、链格孢属、拟盘多毛孢属的分离频率最高。在已有文献报道中,引起苏铁叶部病害的链格孢菌仅鉴定到属。因此,需要进一步明确致病的链格孢菌物种,并确认其寄主类型。同时初步筛选出具有防治潜力的化学药剂,为苏铁属植物保护奠定理论基础。
用于分离病原真菌的苏铁品种:德保苏铁、滇南苏铁、叉孢苏铁。
用于致病性测定的苏铁品种:滇南苏铁。
用于室内防效测定杀菌剂见表1
从南宁植物园青秀山苏铁园采集苏铁羽叶典型褐斑病的样品,采用常规组织分离法分离病原菌[4]。待病原菌的菌落长出后,挑取顶端菌丝进行纯化。采用单孢分离法获得纯培养后,再转入PDA斜面培养与保存。
采用离体叶片无伤、刺伤接种和幼苗刺伤接种2种方法测定菌株致病性。从田间摘取健康且长势较为一致的滇南苏铁羽叶,用清水洗净,再用75%乙醇进行表面消毒。用6 mm打孔器取供试菌株的菌饼分别接种于无伤和刺伤的离体羽叶,羽叶另一边于无伤、刺伤处贴上空白PDA圆块为对照,放入培养皿中保湿培养。离体羽叶接种,每个处理重复6次,无伤和刺伤接种共计24个样品。同时接种滇南苏铁幼苗,每个处理接种3盆幼苗,每盆幼苗接种3片羽叶。保湿培养10~ 15 d,观察处理的发病情况。再从发病的部位对病原菌进行分离。
在PDA上培养病原菌,观察菌落形态,培养14 d后,产生孢子。洗脱孢子制成孢子悬浮液,在显微镜(Leica DM2500 Microscope)下观察。观察样本共计30份。
收集培养5~7 d的菌丝,使用Biospin真菌基因组DNA提取试剂盒提取菌株DNA。以DNA为模板,扩增SSU(NS1/NS4)、ITS(ITS1/ITS4)[17]LSU(LSUFd/LR5)[18]GAPDH(GPD1/GPD2)[19]tef1(EF1-728F/EF1-986R)[20]rpb2(RPB2-5F2[21]/fRPB2-7cR[22])、Alt(Alt-for/Alt-rev)[23]OPA10-2(OPA10-2R/OPA10-2L)[24]8个基因序列。扩增所需引物如表2所示。
扩增体系如下:2×Es Taq MasterMix 20 µL,DNA 1 µL,10 mmol/L上游引物与10 mmol/L下游引物各1 µL,ddH2O 17 µL。PCR扩增条件为:94 ℃预变性3 min;94 ℃变性30 s,55 ℃退火30 s,72 ℃延伸1 min,共35个循环;72 ℃终延伸5 min。PCR产物用1%琼脂糖进行凝胶电泳,电压为120 V,时间为30 min。在荧光图像分析系统上成像拍照。将片段大小符合预期的扩增产物送至广州擎科生物技术有限公司进行测序,在NCBI(National Center for Biotechnology Information)数据库中进行测序结果的BLAST分析,并选择同源性相近的真菌种属序列构建系统发育树。并将本研究扩增出来的正确序列提交至NCBI的GenBank数据库。通过MEGA6软件对样品及同源性相近的真菌种属进行进化树构建。采用Neighbor-Joining法进行数据分析,bootstrap值为1000。
测定啶氧菌酯、苯甲·丙环唑、氟酰羟·苯甲唑、苯甲·嘧菌酯、氟菌·肟菌酯、氯氟醚·吡唑酯、吡唑醚菌酯7种杀菌剂对苏铁褐斑病病原菌的抑菌效果。将杀菌剂配制成不同浓度的母液,将母液依次以5倍的比例稀释成4个浓度梯度,制成含药PDA平板,含药平板为田间用量的1/30。加入ddH2O为空白对照,每个浓度设置3个重复。用打孔器沿菌落外围切成6 mm的菌饼,接种于含药平板上,于(28±1)℃培养箱中,黑暗培养7 d。通过菌落生长速率计算其抑菌率。采用十字交叉法测量菌落直径,计算公式:相对抑菌率=(对照菌落平均直径-处理菌落平均直径)/对照菌落直径×100%,以药剂浓度对数值为自变量(x)、相对抑菌率的几率值为因变量(y),建立毒力回归方程,计算相关系数(R)和抑菌有效中浓度(ID50)。
在对青秀山苏铁病害进行调查的过程中,在德保苏铁、滇南苏铁以及叉孢苏铁中观察到一种褐色病斑,这种病斑中心呈浅褐色,边缘呈深褐色,有的病斑周围还有黄色晕圈(图1A),在症状类似的叶部病斑上均可分离到病原菌。在滇南苏铁的离体叶片上,接种该病原菌,保湿培养10~15 d,羽叶上逐渐出现类似症状。离体叶片上的病斑中心和边缘区别并不明显,病斑周围也有黄色晕圈(图1B),接种该病原菌的12片离体叶片均出现上述症状。在滇南苏铁幼苗羽叶上,接种该病原菌,在30 d左右出现症状(图1C),在接种该病原菌的滇南苏铁幼苗共计6盆中的18片羽叶均出现上述症状。在出现上述症状的羽叶上,可以重新分离到该病原菌,我们将该病原菌编号为GX225。
该病原菌在PDA上培养初期菌落成灰白色,菌落边缘整齐,菌丝绒毛状,气生菌丝发达。在培养10 d后,菌落逐渐变成深棕色(图1D),并在培养基中产生黑色色素(图1E)。分生孢子梗呈棕色至深褐色,大多呈锐角分枝(图1F)。分生孢子呈灰棕色至褐色,大多呈酒瓶状,有纵隔和横隔。有的分生孢子顶部有脐点(图1G),有的分生孢子没有明显的脐点(图1H)。分生孢子一般具有1~4个横隔,0~2个纵隔,隔膜处有缢缩;长23.7~ 31.5 μm,宽7.9~13.2 μm(n=30)。我们根据分生孢子形态初步鉴定该病原菌为Alternaria sp.。
以提取的致病菌株GX225的DNA为模板,采用PCR方法扩增SSULSUGAPDHtef1rpb2AltOPA10-2ITS 8段基因序列。通过凝胶电泳检测,将大小符合预期的片段进行测序分析。将获得的序列提交至NCBI的GenBank数据库获得序列登记号(表2)。将上述8段序列与Alternaria sp.不同种的同源序列进行比对,并进行ClustalW分析,基于上述8个序列用邻接法(Neighbor-Joining)构建系统发育树(图2)。根据系统发育的进化关系,得知GX225菌株与Alternaria burnsii关系最近。进化树的所有分支置信值均大于50%,GX225菌株所在的分枝内的置信值达100%;GX225菌株与A. burnsii的相似性达99.2%,故将GX225菌株鉴定为A. burnsii
为了筛选出具有防治潜力的化学药剂,我们评估了市场上常用的杀菌剂对GX225菌株(A. burnsii)的抑制效果,结果如表3所示。在7种供试药剂中,氟酰羟·苯甲唑的抑菌效果最佳,抑制中量(inhibition dose,ID50)为0.011 mg/L。当浓度为0.67 mg/L时,氟酰羟·苯甲唑的抑菌率可达到73.48%。氟菌·肟菌酯抑菌效果仅次于氟酰羟·苯甲唑,浓度为1.43 mg/L时抑菌率为73.45%,抑制中量为0.021 mg/L。此外,其余5种菌剂对菌株GX225(A. burnsii)均有较强的抑制作用(表3)。
在孟加拉国,有研究报道A. burnsii可以寄生笋瓜(Cucurbita maxima)的种子,但不引起明显的病症[25]。在印度则发现A. burnsii侵染孜然芹(Cuminum cyminum)引起叶斑病,最终造成植株枯萎的情况[26]。而本研究是首次报道在苏铁中A. burnsii侵染的情况,从而丰富了该菌的寄主范围。在形态鉴定中,苏铁上分离的A. burnsii与笋瓜和孜然芹上分离得到的A. burnsii描述基本一致,分生孢子的显微形态也基本相同。
本研究不仅在滇南苏铁(C. diannanensis)上发现褐斑病,在具有相类似症状的德保苏铁(C. debaoensis)、叉孢苏铁(C. segmentifida)和2种苏铁属的植物羽叶上均分离到A. burnsii。在滇南苏铁的离体叶片接种出现相似的症状,这表明A. burnsii可寄生苏铁属植物的多个种。据前期研究报道,Alternaria sp.可引起苏铁属苏铁(C. revoluta)叶斑病,其病状表现为:发病初期病斑为不规则形或长形病斑,后期扩展,病症部位变为黑褐色,病健交界处为红褐色波浪状,病叶上有黑色霉层[10]。但上述研究并未将Alternaria鉴定到种,且根据其对苏铁羽叶上症状的描述,与本研究在德保苏铁、滇南苏铁、叉孢苏铁观察到的症状有相似之处。故推测A. burnsii在苏铁属植物的寄主范围可能并不局限于上述3个种。关于A. burnsii在苏铁属植物是否存在其他寄主,有待进一步研究。
根据WOUDENBERG等[27]的报道,链格孢属(Alternaria)内种的鉴定可基于SSULSUGAPDHtef1rpb2AltOPA10-2ITSendoPG 9个序列构建系统发育树。而本研究中根据上述文献推荐的endoPG扩增引物扩增出的片段与其预期大小不符,这可能是其推荐的endoPG引物序列在Alternaria spp.不够保守导致的。而根据WOUDENBERG等[28]早期的文献报道,GAPDHtef1rpb2AltITS也可以较好地区分链格孢属内不同的种。因此,本研究基于SSULSUGAPDHtef1rpb2AltOPA10-2ITS构建的系统发育树分析,可以较好地将Alternaria spp.包括A. alstroemeriaeA. arborescensA. burnsiiA. eichhorniaeA. gaisenA. gossypinaA. iridiaustralisA. jacinthicolaA. longipesA. tenuissimaA. alternata区分开。
多菌灵、苯甲丙环唑和戊唑醇可防治Colletotrichum sp.、P. cycadisA. cycadia引起的叶斑病[3]。本研究在啶氧菌酯,苯甲·丙环唑、氟酰羟·苯甲唑、苯甲·嘧菌酯、氟菌·肟菌酯、氯氟醚·吡唑酯和吡唑醚菌酯7种药剂中,筛选到抑菌效果最好的是氟酰羟·苯甲唑。然而,众所周知,长期使用单一化学药剂极易使病原菌产生抗药性。所以,后续研究可以扩大药剂的筛选范围,同时也可以筛选具有生物防治潜力的微生物,用于防治A.burnsii引起的苏铁属植物叶斑病。根据报道,针对115株A. alternata菌株的抗药性测定显示,A. alternata对嘧菌酯类药物尚未产生抗药性,而对戊唑醇类药物抗药性的频率达100%;同时,虽然A. alternata对啶酰菌胺产生的抗药性弱,但抗药性发生频率达98.26%;此外,即使A. alternata对腐霉利抗药性发生频率仅为12.17%,但所产生的抗药性较为强烈[29]。因此,我们在使用化学药剂防治时需要警惕菌株抗药性的产生。据报道,在室内平板抑菌试验中,浓度为100 μg/mL的ε-聚赖氨酸盐酸盐对A. dumosa的抑制率可达80.10%,浓度为200 μg/mL的肉桂酸钾对A. dumosa的抑制率可达71.80%[30]。放线菌属(Streptomyces sp.)NEAU-ZC16菌株在平板对峙试验中对A. alternata的抑菌率可达到(69.77±1.20)%。其次生代谢物分离出的活性成分Ⅲ-2-1对A. alternata抑菌率可达到(65.43±1.63)%,其ID50为1.89 pg/mL,毒力回归方程为y=4.8357+ 0.5949x,r=0.9820[16]。由此可见,通过筛选拮抗菌及其次生代谢产物来防治A. burnsii引起的苏铁属植物褐斑病是一种极具潜力的方式。
A. burnsii能引起德保苏铁(C. debaoensis)、滇南苏铁(C. diannanensis)、叉孢苏铁(C. segmentifida)褐斑病。通过SSULSUGAPDHtef1、rpb2、AltOPA10-2ITS 8个基因序列构建系统发育树,可以有效鉴定Alternaria sp.。氟酰羟·苯甲唑能有效抑制A. burnsii的生长,因此,氟酰羟·苯甲唑是一种具有防治A. burnsii引起的苏铁属植物褐斑病潜力的化学药剂。
