Date palm is hermaphroditic and highly heterogeneous in genetics, resulting in severe phenotypic variation. Propagation of seedlings depends on asexual reproduction, and tissue culture technology is the main means of commercializing date seedling production at present. However, the difficulty in inducing embryogenesis and the long induction cycle lead to difficulties in system construction and high tissue culture costs. Therefore, it is urgent to study the effects of hormone regulation on the endogenous metabolic substances of date embryos and optimize the embryogenesis system. This study is based on the selection of date explants, induction of callus and embryogenic callus in the previous studies, and uses orthogonal experimental design to screen and match the plant growth regulators and concentrations in the MS culture medium to induce embryogenesis from callus. The study statistically recorded the embryogenesis rate, induction cycle, and survival rate; date embryos with different levels of hormone were sampled; the morphological changes of date embryos in a regular manner were observed; and the changes in the content of total protein (Cpr), soluble sugar (WSS), catalase (CAT), glutathione reductase (GR), amino acids (AA), auxin, cytokinin (CTK), abscisic acid (ABA), and gibberellin (GA) were measured. The results showed that the embryogenic rate of C9 was higher than that of C5, C4, C6, and the time for embryogenesis was the shortest in the C5 treatment. In the C9 treatment, the highest content of catalase and amino acids was found in the embryos, and the highest protein content was found in the C5 treatment. The coefficient of variation was the largest for glutathione reductase content, and the maximum difference was found in amino acid content. The correlation analysis showed that thiocyanate and gibberellin had the strongest correlation with glutathione reductase, and soluble sugar had the strongest correlation with protein and amino acid. Principal component analysis showed that the first four principal components accounted for 88.714% of the total variance, with an eigenvalue of 3.877 for the first principal component, a contribution rate of 43.076%, and the first principal component was mainly influenced by thiocyanate content, soluble sugar content, protein content, and gibberellin content, with the highest eigenvalue of 0.921 for thiocyanate content, reflecting the impact of thiocyanate on embryogenesis. The study would provide theoretical support for optimizing the embryogenesis system and laying a technical foundation for achieving efficient and rapid propagation of date palm.

椰枣为棕榈科作物,雌雄异株,基因高度杂合,性状分离严重。种苗繁育依赖于无性繁殖,组织培养技术是目前实现椰枣种苗商业化生产的主要手段。但是,体胚发生困难、诱导周期长导致体系构建困难、组培成本高,研究激素调控对椰枣体胚内源代谢物质的影响,优化体胚发生体系成为当务之急。本研究基于前期对椰枣外植体种类的筛选、愈伤组织及胚性愈伤组织诱导的基础上,利用正交试验设计,通过对MS培养基中植物生长调节剂种类和浓度进行筛选配比,从愈伤组织中诱导出体胚,计算体胚发生率、诱导周期及成活率,对不同激素水平下获得的椰枣体胚进行取样,观察体胚的形态变化规律;测定样品中总蛋白(Cpr)、可溶性糖(WSS)、过氧化氢还原酶(CAT)、谷胱甘肽还原酶(GR)、氨基酸(AA)、生长素(auxin)、细胞分裂素(CTK)、脱落酸(ABA)、赤霉素(GA)等指标的含量变化规律。结果表明:C5(MS+30 g/L蔗糖+2 mg/L NAA+2 mg/L 6-BA+3 mg/L 2,4-D+3 mg/L 2-ip)处理的体胚发生时间最短,在C9(MS+30 g/L蔗糖+3 mg/L NAA+3 mg/L 6-BA+2 mg/L 2,4-D+3 mg/L 2-ip)处理下,体胚中过氧化氢酶含量和氨基酸含量最高,在C5处理下,蛋白质含量最高;变异系数最大的为谷胱甘肽还原酶含量,极差最大的是氨基酸含量;相关性分析表明,细胞分裂素和生长素均与谷胱甘肽还原酶相关性最强,可溶性糖与蛋白质和氨基酸的相关性最强;主成分分析显示,前4个主成分累计贡献率达到88.714%,第1个主成分的特征值为3.877,贡献率为43.076%,在第1个主成分中,主要由生长素含量、可溶性糖含量、蛋白质含量及赤霉素含量这4个指标共同影响,其中生长素含量的特征值最大为0.921,主要反映生长素对体胚发生的影响。本研究结果为优化椰枣体胚发生体系提供理论依据,同时为实现椰枣高效快繁奠定技术基础。

椰枣为棕榈科作物,雌雄异株,基因高度杂合,性状分离严重。种苗繁育依赖于无性繁殖,组织培养技术是目前实现椰枣种苗商业化生产的主要手段。但是,体胚发生困难、诱导周期长导致体系构建困难、组培成本高,研究激素调控对椰枣体胚内源代谢物质的影响,优化体胚发生体系成为当务之急。本研究基于前期对椰枣外植体种类的筛选、愈伤组织及胚性愈伤组织诱导的基础上,利用正交试验设计,通过对MS培养基中植物生长调节剂种类和浓度进行筛选配比,从愈伤组织中诱导出体胚,计算体胚发生率、诱导周期及成活率,对不同激素水平下获得的椰枣体胚进行取样,观察体胚的形态变化规律;测定样品中总蛋白(Cpr)、可溶性糖(WSS)、过氧化氢还原酶(CAT)、谷胱甘肽还原酶(GR)、氨基酸(AA)、生长素(auxin)、细胞分裂素(CTK)、脱落酸(ABA)、赤霉素(GA)等指标的含量变化规律。结果表明:C5(MS+30 g/L蔗糖+2 mg/L NAA+2 mg/L 6-BA+3 mg/L 2,4-D+3 mg/L 2-ip)处理的体胚发生时间最短,在C9(MS+30 g/L蔗糖+3 mg/L NAA+3 mg/L 6-BA+2 mg/L 2,4-D+3 mg/L 2-ip)处理下,体胚中过氧化氢酶含量和氨基酸含量最高,在C5处理下,蛋白质含量最高;变异系数最大的为谷胱甘肽还原酶含量,极差最大的是氨基酸含量;相关性分析表明,细胞分裂素和生长素均与谷胱甘肽还原酶相关性最强,可溶性糖与蛋白质和氨基酸的相关性最强;主成分分析显示,前4个主成分累计贡献率达到88.714%,第1个主成分的特征值为3.877,贡献率为43.076%,在第1个主成分中,主要由生长素含量、可溶性糖含量、蛋白质含量及赤霉素含量这4个指标共同影响,其中生长素含量的特征值最大为0.921,主要反映生长素对体胚发生的影响。本研究结果为优化椰枣体胚发生体系提供理论依据,同时为实现椰枣高效快繁奠定技术基础。

Date palm is hermaphroditic and highly heterogeneous in genetics, resulting in severe phenotypic variation. Propagation of seedlings depends on asexual reproduction, and tissue culture technology is the main means of commercializing date seedling production at present. However, the difficulty in inducing embryogenesis and the long induction cycle lead to difficulties in system construction and high tissue culture costs. Therefore, it is urgent to study the effects of hormone regulation on the endogenous metabolic substances of date embryos and optimize the embryogenesis system. This study is based on the selection of date explants, induction of callus and embryogenic callus in the previous studies, and uses orthogonal experimental design to screen and match the plant growth regulators and concentrations in the MS culture medium to induce embryogenesis from callus. The study statistically recorded the embryogenesis rate, induction cycle, and survival rate; date embryos with different levels of hormone were sampled; the morphological changes of date embryos in a regular manner were observed; and the changes in the content of total protein (Cpr), soluble sugar (WSS), catalase (CAT), glutathione reductase (GR), amino acids (AA), auxin, cytokinin (CTK), abscisic acid (ABA), and gibberellin (GA) were measured. The results showed that the embryogenic rate of C9 was higher than that of C5, C4, C6, and the time for embryogenesis was the shortest in the C5 treatment. In the C9 treatment, the highest content of catalase and amino acids was found in the embryos, and the highest protein content was found in the C5 treatment. The coefficient of variation was the largest for glutathione reductase content, and the maximum difference was found in amino acid content. The correlation analysis showed that thiocyanate and gibberellin had the strongest correlation with glutathione reductase, and soluble sugar had the strongest correlation with protein and amino acid. Principal component analysis showed that the first four principal components accounted for 88.714% of the total variance, with an eigenvalue of 3.877 for the first principal component, a contribution rate of 43.076%, and the first principal component was mainly influenced by thiocyanate content, soluble sugar content, protein content, and gibberellin content, with the highest eigenvalue of 0.921 for thiocyanate content, reflecting the impact of thiocyanate on embryogenesis. The study would provide theoretical support for optimizing the embryogenesis system and laying a technical foundation for achieving efficient and rapid propagation of date palm.

椰枣树(Phoenix dactylifera L.)是雌雄异株的多年生植物,属于棕榈科刺葵属,具有耐旱、耐碱、耐热的特点,因此能够适应高温干旱、盐碱、沙漠等极端恶劣的生态环境,广泛分布于中东和北非等干热地区,在我国的海南、云南、广东等地均有分布。椰枣树和椰枣在阿拉伯人民日常生活中扮演了极为重要的角色,营养价值极高,是阿拉伯一些国家重要的出口农作物,在阿拉伯国家的生态和经济建设中发挥着重要作用[1]。椰枣树雌雄异株,且基因高度杂合,种子繁殖性状分离严重,无法保持母本的优良特性[2-3]。无性繁殖成为椰枣种苗繁育的主要手段,无性繁殖技术包括2个方面:母株分蘖苗繁殖和组织培养技术。其中分蘖苗繁殖是从母株上切割下分蘖苗,然后复壮生根后田间定植[4-5]。组织培养技术是取椰枣的器官组织通过体胚发生途径获得再生植株,而后壮苗生根,温室炼苗后田间定植[6]。农业生产以蘖苗分株的方式开展,但单株母本全生命周期内分蘖苗数量仅能达到15~25株,其繁殖数量和速度受到极大限制,难以满足栽培种植需求。目前,国际上依赖组培手段实现优良母本椰枣种苗商业化生产,生产技术由发达国家垄断,体胚发生率低,生产成本高,导致组培苗价格高昂,椰枣组培苗国际售价单株高达50美元,也成为全世界椰枣育种的关键技术难题。
植物体细胞胎在离体培养中通过体细胞胚胎发生途径形成再生植株己是极其普遍的现象,并认为该发生途径是植物体细胞在离体培养条件下的一个基本发育途径。体细胞胚胎发生在很多领域都得到了广泛的应用,为植物细胞分化、全能性表达及其机理等理论问题研究提供了理想的实验体系[7]。体细胞胚胎发生途径是细胞全能性表达最完全的一种方式,它不仅表明植物体细胞具有全套遗传信息,而且重演了合子胚形态发生的过程[8-9]。胚状体具有两极性,在适宜的环境里可以直接生成小植株,快速大量产生种苗。细胞突变个体的选择和育种等方面的应用为新品种的培育提供了新的方法。此外,体细胞胚胎发生技术还在杂种合子的挽救、体细胞杂种和单倍体、三倍体等植株的诱导培养等诸多方面均有广阔的应用前景,不仅在科研上有着极为重要的意义,而且在生产实践中也有着巨大的经济价值。在椰枣离体培养的过程中,取样组织材料的污染、褐变、玻璃化、愈伤组织诱导周期长、体胚发生困难、不定芽诱导率低等问题是困扰组织培养的主要难题,也是造成椰枣组培成本高、难以实现商业化的主要原因。随着植物组织培养技术研究的不断深入,大部分植物可通过筛选合适培养基和培养条件对其不同部位组织器官、细胞及原生质体进行培养,通过直接或间接途径,经分裂、分化、脱分化形成完整植株。在前期的试验中发现,椰枣外植体在进行启动培养80~90 d后会出现褐化,在这个阶段,需要重点调控组织褐变,可保证后期的愈伤组织诱导率提高,缩短诱导时间[10]。体胚发生过程中愈伤组织生长状态很难保持一致,影响培养基的优化。生长素与细胞分裂素异戊烯基腺嘌呤的组合,脱落酸和聚乙二醇的组合在椰枣体细胞胚的同步化和成熟化过程中起重要作用[11-12]。萘乙酸可有效促进胚性愈伤组织的增殖,有研究表明胚性愈伤组织在含0.1 mg/L萘乙酸(NAA)的培养基上可以连续生长,并产生大量重复的体细胞胚[13]。经过多次继代培养,胚性愈伤组织可形成起源于愈伤组织表面的原胚[6]。酪蛋白水解物是18种氨基酸的混合物,是钙、磷酸盐、几种微量元素和维生素的来源,有研究发现在愈伤组织培养基中加入酪蛋白水解物可促进愈伤组织的产生[14-15]。在MS培养基中添加0.5 mg/L ABA,持续4周,可促进胚胎成熟,并促进贮藏蛋白质、脂类和淀粉的积累。ABA可以抑制胚异常结构的形成,抑制体胚的早熟萌发[17]。关于椰枣体胚对激素调控的生理响应鲜有报道,愈伤组织在诱导体胚过程,愈伤组织经过继代培养后,体胚发生率会出现逐代下降的趋势,如何精准地调控细胞分裂素和生长素种类和浓度、优化培养基成为提高体胚发生率及体胚质量的关键问题,也是目前国内构建椰枣高效再生体系亟需解决的难题。
本研究通过调配培养基植物生长调节剂种类和浓度,筛选出体胚发生的最佳培养基,并测定不同激素水平下获得的椰枣体胚中总蛋白、可溶性糖、还原酶类(过氧化氢还原酶、谷胱甘肽还原酶)、氨基酸、内源激素(生长素、细胞分裂素、脱落酸、赤霉素)等含量的变化规律,筛选出不同激素配比在诱导体胚形成过程中具有差异性变化的内源代谢物质。本研究结果为优化体胚发生体系提供理论依据,同时为实现椰枣高效快繁奠定技术基础。
供试品种为埃及椰枣品种Hainay,试验地点为中国热带农业科学院椰子研究所(108°21′E,19°20′N)。
首先取椰枣的茎尖生长组织作为外植体,通过调控培养基配方及培养条件,诱导产生愈伤组织,具体操作参考张宁等[10]的方法。利用正交试验设计,体胚发生的培养基处理编号为C1~C9,通过对MS培养基中植物生长调节剂种类和浓度进行筛选配比,从愈伤组织中分化出体胚,体胚萌发后获得不定芽,并生根获得脱毒苗。其中选择体胚发生率≥30%处理下的体胚作为供试材料(图1)。
基础培养基:MS盐、30 g/L蔗糖、3.5 g/L植物凝胶、1 g/L活性炭、植物生长调节剂[(萘乙酸(NAA)、6-苄氨基嘌呤(6-BA)、2,4-二氯苯氧乙酸(2,4-D)、异戊烯腺嘌呤(2-ip)]。调整培养基pH 5.7,于121 ℃高温高压下灭菌20 min。
体胚发生试验采用正交设计法,按照正交表L9(43)进行试验设计,以NAA、6-BA、2,4-D和2-ip为试验因素,每个因素设3个水平(表1)。如表2所示,共设9个处理,每个处理10瓶,每瓶接种10个愈伤组织,重复3次。
体胚发生试验:选择胚性愈伤组织,转接至体胚发生培养基中,每隔45 d继代1次,28 ℃黑暗下培养,继代3~5次后统计体胚发生率及诱导周期。体胚发生率=(体胚发生的愈伤组织数/供试愈伤组织数)×100%
取样不同激素水平下获得的椰枣体胚,测定样品中总蛋白(Cpr)、可溶性糖(WSS)、还原酶类[过氧化氢还原酶(CAT)、谷胱甘肽还原酶(GR)]、氨基酸(AA)、内源激素[生长素(auxin)、细胞分裂素(CTK)、脱落酸(ABA)、赤霉素(GA)]等指标的含量。
测定生理指标:Cpr、WSS、CAT、GR、AA、POD、SOD、HP指标检测所使用的试剂盒为96样微板法,试剂盒购自苏州格锐思生物科技有限公司,所用设备为赛默飞的酶联免疫检测仪;auxin、CTK、ABA、GA指标检测所使用的试剂盒为双抗体一步夹心法酶联免疫吸附试验(ELISA)检测试剂盒,96样微板法,试剂盒购自Mlbio公司(酶联生物科技有限公司),所用设备为赛默飞的酶联免疫检测仪。
测定原理:用纯化的植物激素抗体包被微孔板,制成固相抗体,在包被单抗的微孔中依次加入植物激素,再与辣根过氧化物酶(horseradish peroxidase,HRP)标记的植物激素抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后底物TMB显色。TMB在HRP的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的植物激素呈正相关。用酶标仪在波长450 nm下测定吸光度(OD值),通过标准曲线计算样品中的激素浓度。
形态指标观测:选择愈伤组织、胚性愈伤组织、体胚等作为供试材料,用石蜡切片技术对组织样本进行切片,番红固绿染色,观察细胞组织的形态结构;试验流程:脱蜡至水-固绿染色-番红染色-脱水封片-显微镜镜检。
试验数据用Excel软件整理,用SPSS 22软件进行单因素、多因素方差分析、LSD法多重比较检验及显著性分析,用R语言(3.6.2)进行相关性分析。
表3可以看出,不同试验因素与水平的处理对体胚发生率及发生时间产生不同程度的影响。在C9处理下,体胚发生数最多,达67.33%,显著高于其他处理(P<0.05),体胚发生时间为41.33 d。C5与C6体胚发生时间分别为35.33 d和35.00 d,差异不显著,其中C5的体胚发生率达到45.67%,显著高于C6的34.33%(P<0.05)。C2和C8的体胚发生时间较长,分别达到56.33 d和63.00 d,且体胚发生率仅为18.00%和13.00%,显著低于其他处理(P<0.05)。由此可见C9处理可以提高体胚发生率,C5处理可以缩短体胚发生时间。
图2可以看出,经过番红固绿染色后,细胞呈现幼嫩的绿色,具备分生的活力。C6、C4的细胞排列紧密,形状不规则,边缘锯齿状,内部液泡丰富,分生区细胞排列致密,愈伤组织表皮细胞分化,C4的分生区胚性细胞更丰富,进入分裂状态的更多;C5和C9的分生区诱导出具备子叶的体胚,C5的子叶液泡较大,细胞排列稀疏,边缘形状不规则,C9的体胚细胞具有典型的线形结构,中间有一个明显的核区域,周围是细胞质,这种结构表明它可能是一个正在分裂或分化的细胞。
表4可以看出,体胚发生率最高的处理组C9,其体胚中过氧化氢酶活性和氨基酸含量显著高于C6处理(P<0.05),体胚发生率仅次于C9的C5处理组,其体胚中氨基酸含量与C9处理下并无显著差异;细胞分裂素和赤霉素含量在4组处理中均无显著差异;C4和C5处理体胚脱落酸含量均无显著差异,C6和C9处理体胚脱落酸含量均无显著差异;细胞生长素含量在C4和C5处理下差异并不显著,C9和C6处理下的差异不显著;C6处理下的体胚可溶性糖含量显著低于C4、C5和C9,C4、C5和C9处理的体胚可溶性糖含量均无显著差异性;C6处理的体胚蛋白质含量显著低于C4、C5和C9,C5处理的体胚蛋白质含量最高。由此可见,体胚内的氨基酸含量、过氧化氢酶活性、蛋白质含量对体胚发生影响较大。
表5可以看出,内源代谢物含量指标均存在不同程度的变异。氨基酸含量的极差最大,达742.66,生长素含量的极差最小,仅有1.19。在各指标中,变异系数最大的为谷胱甘肽还原酶活性,为10.62%,变异系数最小的为赤霉素含量仅有0.36%。在9个指标中,生长素含量、细胞分裂素含量、脱落酸含量、赤霉素含量的变异系数均小于1%,且只有谷胱甘肽还原酶活性的变异系数高于10%。
表6可知,9个指标间均存在不同程度的相关性,其中谷胱甘肽还原酶活性除了与过氧化氢酶活性呈负相关外,与其他指标均呈正相关,与生长素含量的相关性最强,相关系数达0.730;过氧化氢酶活性除了与谷胱甘肽还原酶活性呈负相关外,与其他指标均呈正相关,其中相关性最强的为可溶性糖含量,相关系数达0.603;可溶性糖与所有指标均呈正相关,相关性最强为蛋白质含量,相关系数为0.844;氨基酸含量与所有指标均呈正相关,其中相关性最强为可溶性糖含量,相关系数为0.788;细胞分裂素与所有指标均呈正相关,相关性最强为谷胱甘肽还原酶活性,相关系数为0.451,其次为可溶性糖含量,其相关系数为0.488。脱落酸含量与其他指标呈正相关,其中相关性最强为细胞分裂素含量,相关系数为0.210,除细胞分裂素含量外,与蛋白含量、可溶性糖含量和谷胱甘肽还原酶活性的相关系数均相近。
采用SPSS 22.0软件将数据进行标准化处理,对处理后的数据进行主成分分析,得到体胚发生阶段内源代谢物质的主成分的特征值、贡献率、累计贡献率和特征向量,结果如表7表8所示。
有研究认为,特征值≥1或者累计贡献率>85%的主成分具有一定代表性[16]。由表7可知,前4个主成分累计贡献率达到88.714%,根据累计贡献率>85%的标准,表明这4个主成分可以反映全部指标的大部分信息,而其他成分的特征值均<1,且贡献率趋于平缓,代表性不强,因此可以用前4个主成分对椰枣内源代谢物进行综合评价。
表7表8可知,第1主成分的特征值为3.877,贡献率为43.076%;在第1主成分中,主要包含生长素含量、可溶性糖含量、蛋白质含量及赤霉素含量这4个指标,其中生长素含量的特征值最大为0.921,其次为可溶性糖含量、蛋白质含量及赤霉素含量,特征值分别为0.850、0.804、0.677;第2主成分的特征值为2.325,贡献率为25.835%,在第2主成分中,主要包含过氧化氢酶活性与谷胱甘肽还原酶活性2个指标的影响,特征值分别为0.953与0.820;第3主成分的特征值为1.011,贡献率为11.235%,第3主成分主要是氨基酸含量、脱落酸含量及谷胱甘肽还原酶活性3个指标影响,特征值分别为–0.502、0.463及0.426;第4主成分的特征值为0.771,贡献率为8.568%,第4主成分主要是细胞分裂素含量、脱落酸含量及赤霉素含量的影响,其特征值分别为0.495、–0.483、0.437。
由于植物组织或细胞在离体培养条件下往往缺乏合成生长素与细胞分裂素的能力,细胞的分化和分裂以及形态构成过程中必须依靠生长素和细胞分裂素2种激素的共同作用,为此在培养基中添加不同种类或不同浓度的外源激素诱导形态发生已受到广泛的重视。生长素和细胞分裂素不同浓度、比例不但可诱导细胞分裂和生长,而且能控制细胞分化和形态建成,诱导基因的差别表达。有研究表明胚的发育主要是生长素与细胞分裂素之间的平衡改变,导致了细胞中基因表达调节控制系统改变,最终导致细胞发育途径的改变[17]。本研究通过调配培养基植物生长调节剂种类和浓度,筛选出体胚发生的最佳培养基,并测定不同激素水平下获得的椰枣体胚中总蛋白、可溶性糖、还原酶类(过氧化氢还原酶、谷胱甘肽还原酶)、氨基酸、内源激素(生长素、细胞分裂素、脱落酸、赤霉素)等含量的变化规律,发现不同激素水平的处理对椰枣体胚的内源代谢物含量有显著影响。
生长素含量、谷胱甘肽还原酶活性和可溶性糖含量与体胚的发生关系密切。2,4-D作为生长素是诱导多种植物离体细胞转变为胚性细胞的重要激素,有以下作用机制:通过激活一些基因表达特异蛋白以促进体细胞胚胎发育,也可以抑制这些基因表达以抑制体细胞胚胎发育;2,4-D浓度与DNA甲基化有关;可能与培养细胞内部因子有关(通过改变细胞内源IAA而起作用)[18]。陈以峰等[19]研究表明内源IAA在体细胞胚胎发生的诱导过程中起关键作用,并认为内源IAA含量上升或维持在较高水平是胚性细胞出现的一个共同标志团。在本研究中,生长素含量最高的处理组C9显示了最高的体胚发生率和较快的体胚发生时间,这表明生长素的高含量有助于提高体胚的形成效率。此外,谷胱甘肽还原酶活性也在C9处理中达到最高,这与过氧化氢酶活性的增加一致,表明高水平的生长素可能促进了体内抗氧化防御系统的活性,从而有利于维持细胞的正常生理功能并促进体胚的形成。在椰枣体胚发生过程中,激素调控对其生理响应的影响显著。生长素作为一种关键的植物激素,其含量的变化直接影响体胚的发生率和生长状态。同时,本研究也揭示了可溶性糖含量在不同处理中的变化趋势。可溶性糖作为提供能量的主要来源,其在C9处理中的较高水平可能为体胚提供了足够的能量支持,从而有助于体胚的快速发育。
从变异系数分析来看,虽然所有指标均表现出一定程度的变异,但生长素、谷胱甘肽还原酶和可溶性糖是影响椰枣体胚形成的主要因素。对于不同处理下椰枣体胚内源代谢物质含量的变化,可以看到谷胱甘肽还原酶活性的变异系数最大,达到10.62%,而生长素含量的极差最小,仅为1.19。这表明谷胱甘肽还原酶活性在试验中表现出较大的变动,可能是由于不同培养条件下细胞应激反应或代谢途径的差异所致。同时,谷胱甘肽还原酶、过氧化氢酶和可溶性糖等内源代谢物之间存在较强的相关性,这与它们在植物应对环境压力中的共同调控作用相符合。体胚发生率最高的处理组C9显示了较高的过氧化氢酶活性和氨基酸含量,这可能暗示着在这些培养条件下,椰枣体胚的生长素调节作用最为显著,从而促进了细胞分裂和蛋白质合成。此外,C5处理虽然体胚发生率较C9低,但其氨基酸含量与C9无显著差异,说明在某些激素配比下,椰枣体胚发育的生理生化指标达到一个较为理想的状态。这提示在实际应用中可以通过调节这些关键激素的水平来优化体胚发生体系。崔凯荣等[20]认为外源激素通过细胞信号转导诱导基因差别表达,而引起体细胞胚胎发生。生长素与其结合蛋白(SABP)结合形成的“生长素-受体”复合物、ABA的胞内受体和胞外受体、细胞分裂素结合蛋白(cBPs)等均影响特定基因的表达[21]。6-BA在苹果体细胞胚胎诱导中诱导蛋白质的调控主要是在翻译水平上而不依赖于新RNA的合成[22]。除生长素和细胞分裂素外,ABA在植物体细胞胚胎的诱导中也起到很重要的作用。
通过主成分分析和综合贡献率来看,本研究发现前4个主成分累计贡献率达到88.714%,表明这些指标可以综合反映所有变量的大部分信息。其中生长素、可溶性糖、蛋白质和赤霉素这4种内源代谢物在第1主成分中的特征值较高,显示了它们在椰枣体胚发生过程中的关键作用。这进一步验证了在试验设计时考虑激素种类和浓度的重要性,以及其对椰枣体胚发育影响的复杂性和多因素控制的特点。ABA在蔗糖调控胡萝卜体细胞胚胎发育的信号传递网络中可能是一个重要的中介信号因子[23]。ABA有可能直接作为反式因子调节基因表达但更可能是先与靶细胞效应部位结合,再通过细胞的第二信使分子传递信号,激活反式作用与顺式作用元件结合,从而在转录水平上调控基因表达。体细胞胚胎发生过程中各种内源激素的代谢和动态平衡起着重要而关键的作用。赤霉素、乙烯、多胺等在体细胞胚胎发生中也有重要作用。在皇冠草体细胞胚胎发生中,GA3的变化趋势和细胞分裂素基本相同[24]。内源激素平衡受外源生长调节物质的影响,生长调节剂的使用在调节植物形态发生中具有重要作用[22]。一般认为外源ABA有利于体细胞胚胎发育和成熟。ABA处理促进针叶树体细胞分化、抑制早期体细胞胚胎的发生、提高体细胞胚胎分化的同步化水平[25]。GA3抑制许多植物体细胞胚胎发生 但对生根与成株有促进作用。有报道指出GA3抑制胡萝卜体细胞胚胎的发育,GA3对体细胞胚胎发生有副作用[26]。但GA3对红江橙幼果体细胞胚胎后期生长、水母雪莲体细胞胚胎的诱导有利[27]
本研究结果不仅为椰枣体胚发生的机理提供了新的见解,而且为实际生产中如何通过调整培养基中激素配比来提高椰枣组织培养的效率提供了理论依据和技术指导。在未来的研究中,可以进一步探索其他环境因子如光照、温度等对椰枣体胚发生的影响,以全面优化椰枣的组织培养体系。综上所述,通过对椰枣体胚发生及其内源代谢物的系统研究,不仅揭示了激素调控在椰枣体胚发育中的关键作用,也为优化椰枣组织培养技术和快速繁殖体系提供了重要的理论依据和实践指导。
