Tea quality is closely related to the growth and ecological environment of tea plant (Camellia sinensis). The difference between day and night temperature (DIF) affects the synthesis and accumulation of tea plant metabolites, which in turn affects the quality of tea. In this study, Fuyun No. 6 was selected as the research object, and four DIF treatments (0 ℃, 5 ℃, 10 ℃ and 15 ℃) were simulated to explore the effects of DIF on photosynthesis and volatile and non-volatilequality components of tea fresh leaves. The results showed that the net photosynthetic rate (Pn), stomata conductance (Gs) and transpiration rate (Tr) of tea plants increased significantly with the increase of DIF (P<0.05). Increasing DIF significantly increased the contents of chlorophyll a (Chl a), chlorophyll b (Chl b), chlorophyll (a+b) [Chl (a+b)] and carotenoids (Car), and reached the maximum when DIF was 10 ℃. In addition, the change of DIF had an effect on the volatile components and relative contents of main components in fresh tea leaves, and the relative contents of alcohols and aldehydes were the largest in the volatile components of fresh tea leaves. The increase of DIF could significantly enhance the relative contents of aldehydes, and the relative contents of volatile components and main aroma components in fresh tea leaves were different with the change of DIF. The content of hexanal increased with the increase of DIF, with the highest value of 39.94% at DIF=15 ℃, while the relative content of cis-3-hexenol decreased with DIF increasing. The change of DIF significantly affected the activities of β-glucosidase, β-primeverosidase and lipoxygenase. The activities of β-glucosidase and β-primeverosidase first increased and then decreased with the increase of DIF, and reached the maximum when the DIF was 5 ℃, while the activity of lipoxygenase was opposite. Furthermore, the contents of free amino acids and caffeine increased significantly with the increase of DIF, which were 2.44 and 2.55 times at DIF=15 ℃ than those at DIF=0 ℃, respectively. The total flavonoids content and ratio of tea polyphenols to amino acids decreased significantly with the increase of DIF (P<0.05), with the lowest value of 2.84% and 3.24 at DIF=15 ℃. The contents of free amino acids in fresh tea leaves were significantly positively correlated with Pn, while the ratio of tea polyphenols to amino acids showed significantly negatively correlated with Pn (P<0.01). This study indicated that the increase of DIF could significantly affect the volatile aroma substances of fresh tea leaves and improve the taste and quality of tea, which could provide a certain theoretical basis for tea plantation site selection and tea planting.

茶叶品质与茶树(Camellia sinensis)的生长生态环境密切相关,其中昼夜温差的变化影响茶树代谢产物的合成与积累,进而影响茶叶品质。本研究选取福云六号茶树为研究对象,模拟设置0、5、10、15 ℃ 4个昼夜温差(DIF)处理,通过模拟DIF变化来探究其对茶树光合作用与茶鲜叶挥发性及非挥发性品质成分的影响。研究结果表明:随着DIF增大,茶树净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)显著增大(P<0.05)。DIF增大显著增加叶绿素a(Chl a)、叶绿素b(Chl b)、叶绿素a+b[Chl(a+b)]、类胡萝卜素(Car)含量,并且在DIF为10 ℃时含量达到最大。DIF变化对茶鲜叶挥发性组分和主要组分的相对含量均有影响,茶鲜叶挥发性成分中醇类和醛类相对含量最大。DIF增大提高醛类物质的相对含量,并且茶鲜叶挥发性成分的种类与主要香气成分的相对含量随DIF变化有所差异,其中正己醛的相对含量随DIF的增大而增加,DIF为15 ℃时其相对含量最高,为39.94%,青叶醇的相对含量则随DIF的增大而降低。DIF的变化显著影响β-葡萄糖苷酶、β-樱草糖苷酶和脂氧合酶活性,其中β-葡萄糖苷酶、β-樱草糖苷酶活性随DIF的增大呈先增后降的趋势,在DIF为5 ℃时达到最大,而脂氧合酶活性则相反。游离氨基酸和咖啡碱含量随DIF的增大显著增加,DIF为15 ℃时其含量分别是DIF为0 ℃时的2.44倍和2.55倍;总黄酮含量与酚氨比则随DIF的增大显著降低(P<0.05),在DIF为15 ℃时均达到最低,分别为2.84%和3.24。茶鲜叶中的游离氨基酸含量与Pn呈极显著正相关,而酚氨比与Pn呈极显著负相关(P<0.01)。本研究发现DIF增大可明显影响茶鲜叶的挥发性香气物质,提高茶叶滋味品质,研究结论可为茶园选址和茶树种植提供理论依据。

茶叶品质与茶树(Camellia sinensis)的生长生态环境密切相关,其中昼夜温差的变化影响茶树代谢产物的合成与积累,进而影响茶叶品质。本研究选取福云六号茶树为研究对象,模拟设置0、5、10、15 ℃ 4个昼夜温差(DIF)处理,通过模拟DIF变化来探究其对茶树光合作用与茶鲜叶挥发性及非挥发性品质成分的影响。研究结果表明:随着DIF增大,茶树净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)显著增大(P<0.05)。DIF增大显著增加叶绿素a(Chl a)、叶绿素b(Chl b)、叶绿素a+b[Chl(a+b)]、类胡萝卜素(Car)含量,并且在DIF为10 ℃时含量达到最大。DIF变化对茶鲜叶挥发性组分和主要组分的相对含量均有影响,茶鲜叶挥发性成分中醇类和醛类相对含量最大。DIF增大提高醛类物质的相对含量,并且茶鲜叶挥发性成分的种类与主要香气成分的相对含量随DIF变化有所差异,其中正己醛的相对含量随DIF的增大而增加,DIF为15 ℃时其相对含量最高,为39.94%,青叶醇的相对含量则随DIF的增大而降低。DIF的变化显著影响β-葡萄糖苷酶、β-樱草糖苷酶和脂氧合酶活性,其中β-葡萄糖苷酶、β-樱草糖苷酶活性随DIF的增大呈先增后降的趋势,在DIF为5 ℃时达到最大,而脂氧合酶活性则相反。游离氨基酸和咖啡碱含量随DIF的增大显著增加,DIF为15 ℃时其含量分别是DIF为0 ℃时的2.44倍和2.55倍;总黄酮含量与酚氨比则随DIF的增大显著降低(P<0.05),在DIF为15 ℃时均达到最低,分别为2.84%和3.24。茶鲜叶中的游离氨基酸含量与Pn呈极显著正相关,而酚氨比与Pn呈极显著负相关(P<0.01)。本研究发现DIF增大可明显影响茶鲜叶的挥发性香气物质,提高茶叶滋味品质,研究结论可为茶园选址和茶树种植提供理论依据。

Tea quality is closely related to the growth and ecological environment of tea plant (Camellia sinensis). The difference between day and night temperature (DIF) affects the synthesis and accumulation of tea plant metabolites, which in turn affects the quality of tea. In this study, Fuyun No. 6 was selected as the research object, and four DIF treatments (0 ℃, 5 ℃, 10 ℃ and 15 ℃) were simulated to explore the effects of DIF on photosynthesis and volatile and non-volatilequality components of tea fresh leaves. The results showed that the net photosynthetic rate (Pn), stomata conductance (Gs) and transpiration rate (Tr) of tea plants increased significantly with the increase of DIF (P<0.05). Increasing DIF significantly increased the contents of chlorophyll a (Chl a), chlorophyll b (Chl b), chlorophyll (a+b) [Chl (a+b)] and carotenoids (Car), and reached the maximum when DIF was 10 ℃. In addition, the change of DIF had an effect on the volatile components and relative contents of main components in fresh tea leaves, and the relative contents of alcohols and aldehydes were the largest in the volatile components of fresh tea leaves. The increase of DIF could significantly enhance the relative contents of aldehydes, and the relative contents of volatile components and main aroma components in fresh tea leaves were different with the change of DIF. The content of hexanal increased with the increase of DIF, with the highest value of 39.94% at DIF=15 ℃, while the relative content of cis-3-hexenol decreased with DIF increasing. The change of DIF significantly affected the activities of β-glucosidase, β-primeverosidase and lipoxygenase. The activities of β-glucosidase and β-primeverosidase first increased and then decreased with the increase of DIF, and reached the maximum when the DIF was 5 ℃, while the activity of lipoxygenase was opposite. Furthermore, the contents of free amino acids and caffeine increased significantly with the increase of DIF, which were 2.44 and 2.55 times at DIF=15 ℃ than those at DIF=0 ℃, respectively. The total flavonoids content and ratio of tea polyphenols to amino acids decreased significantly with the increase of DIF (P<0.05), with the lowest value of 2.84% and 3.24 at DIF=15 ℃. The contents of free amino acids in fresh tea leaves were significantly positively correlated with Pn, while the ratio of tea polyphenols to amino acids showed significantly negatively correlated with Pn (P<0.01). This study indicated that the increase of DIF could significantly affect the volatile aroma substances of fresh tea leaves and improve the taste and quality of tea, which could provide a certain theoretical basis for tea plantation site selection and tea planting.

茶树起源于我国云贵高原的深山密林之中[1],是极具中国特色的经济作物,在21世纪初期,茶类饮品的消费量已成为世界上第二大无酒精饮料。随着人们生活水平的提高,高品质茶越来越受到消费者的青睐。茶叶品质通常指茶叶的色、香、味、形[2]。茶叶的色泽、香气和滋味均与茶树新陈代谢所产生的生物化学物质息息相关。其中茶叶挥发性香气化合物、氨基酸类次生代谢物、黄酮类次生代谢物质以及生物碱类次生代谢物质为茶树主要的次级代谢产物,其含量决定着茶叶品质,其中,挥发性香气化合物影响茶叶香气,氨基酸类次生代谢物、黄酮类次生代谢物质以及生物碱类次生代谢物质则影响茶汤滋味[3-4]。茶树次生代谢是其长期进化过程中与环境相互作用的结果,茶树所处的生态环境影响茶树的生长与茶叶品质[5]
在影响茶树生长及品质的众多因子中,温度是影响茶树生长和光合作用的主要因素之一。温度影响茶树的酶促反应,决定茶树中的酶活性,从而影响茶树代谢,进而影响茶叶化学物质的形成、转化和积累。有大量研究指出,昼夜温差(difference between day and night temperature,DIF)对作物品质有显著影响[6-8],白天温度较高有利于植物进行光合作用,促进光合产物形成;夜晚温度较低,植物进行呼吸作用,有利于干物质积累[9-10]。研究指出,高山茶香气高与高山茶园夜晚温度较低有关,短期的低温可诱导茶叶中脂肪氧化酶基因表达,促进茶叶芳香物质醇和醛的合成与释放[11-12]。许晓青[13]在葡萄果实发育过程中绿叶气味物质的研究中发现青叶醛和青叶醇与昼夜温差有显著正相关关系。李莉等[14]研究发现在番茄花果期适当增大昼夜温差可提高果实品质。李永菊等[15]指出在合适的昼夜温差下,有利于增加茶叶的蛋白质、氨基酸、维生素等含量。BOO等[16]通过研究指出,莴苣的总酚和花青素含量在较低昼夜温度(13 ℃/10 ℃、20 ℃/13 ℃)下有所提高。郑卫杰等[17]研究表明,最有利于文心兰干物质积累和叶绿素合成的昼夜温差是15 ℃。也有研究发现,昼夜温差较大会刺激脱落酸产生,从而提高茎的伸长,有助于植物应对环境条件的变化[18-19]
目前,人们对茶叶的需求量逐渐增加,对茶叶品质的要求也越来越高。福云六号是福建省特色茶树,在全国主要产茶区均有分布,在生态和经济方面具有极高价值。本研究以福云六号茶树为研究对象,通过模拟温差变化对茶树光合作用、茶鲜叶挥发性香气物质以及茶叶非挥发性品质成分含量的影响进行研究,从而初步明确昼夜温差、光合作用与茶叶品质之间的联系,为茶叶品质提升提供一定的科学理论依据,并且为茶园选址提供一定的参考,有利于茶产业更好地发展。
选用2年生福建省特色茶树福云六号为研究对象。将茶树种植于外口径29.6 cm,内口径25.4 cm,高19.7 cm,底部直径17.8 cm的花盆中,每个花盆中栽培3株茶树。茶树盆栽缓苗3个月后,选取3盆长势一致的植株,利用人工气候箱进行控温处理。
试验设置4个昼夜温差(DIF)处理,分别为0 ℃(15 ℃/15 ℃)、5 ℃(20 ℃/15 ℃)、10 ℃(25 ℃/15 ℃)、15 ℃(30 ℃/15 ℃),4个处理均每天光照12 h(6:00—18:00),光合有效辐射为200 μmol/(m2·s)(距离LED灯板20 cm处),空气相对湿度为70%±5%。在人工气候箱培养60 d后进行样品采集与分析。
培养结束后,利用LI-CORLI-6400光合仪(LI-COR Biosciences,USA)测定福云六号茶树冠层处新展开叶片的净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)及蒸腾速率(Tr)等光合参数。具体测量方法参照文献[20]。
光合参数测定结束后,摘取一芽四叶放干冰中暂存,随后置于–80 ℃冰箱中保存,一部分用于茶鲜叶叶绿素含量和茶鲜叶挥发性香气相关酶活性的分析,另一部分用冷冻干燥机进行冻干磨粉保存。
气体样品使用吹扫捕集仪(CDS7000E+7400,USA)-气质联用仪(7890B-5977A,Agilent Technologies,USA)分析。称取经冷冻干燥粉碎后的茶样0.5 g于40 mL吹扫瓶中,加入5 mL沸纯水,迅速封口,放入自动进样器(CDS7000E)进样分析。吹扫捕集条件:吹扫流量为40 mL/min;吹扫时间为11 min;脱附温度为200 ℃;脱附时间为5 min;烘烤温度为230 ℃;烘烤时间为5 min。GC条件:选用DB-624色谱柱(60 m×250 μm×1.4 μm);氦气为载气;柱流量为1.5 mL/min;设置初始温度为40 ℃,保持2.5 min,然后以3 ℃/min升温到180 ℃,保持1 min,再以10 ℃/min升温至230 ℃,保持3 min;离子源为电子轰击源(EI);电压为70 eV。数据采集使用全扫描模式(Scan),采用标准的质谱图库(NIST14)进行检索,香气物质的定性和定量方法具体参照文献[21]。
茶多酚的测定参照GB/T 8313—2018《茶叶中茶多酚和儿茶素类含量的检测方法》;游离氨基酸的测定参照GB/T 8314—2013《茶游离氨基酸总量的测定》;咖啡碱的测定参照GB/T 8312—2013《茶咖啡碱测定》;水浸出物的测定参照GB/T 8305—2013《茶水浸出物测定》;总黄酮含量的测定采用分光光度法,具体方法参照文献[22];可溶性糖含量的测定采用蒽酮法。
叶绿素指标测定参照《植物生理学实验教程》[23];鲜叶的β-葡萄糖苷酶活性、β-樱草糖苷酶活性和脂氧合酶活性测定均使用试剂盒(上海优选生物科技有限公司),试剂盒采用双抗体一步夹心法酶联免疫吸附试验(ELISA)。
使用SPSS 18软件单因素方差分析(one-way ANOVA)对茶叶品质指标、茶树光合参数及茶鲜叶挥发性香气相关酶活性进行显著性分析,利用LSD法比较组间差异,对茶树光合作用参数及茶叶非挥发性品质成分含量进行Pearson相关性分析;采用Origin 2022软件绘图。
对不同温差处理的茶鲜叶样品的挥发物成分进行分析鉴定,经NIST标准谱库检索,对各峰加以确认后用峰面积归一法进行定量分析,以各组分的峰面积占总峰面积的百分比表示组分的相对含量。将各物质进行分类,茶鲜叶样品的挥发物成分分为醇类、酯类、醛类、烯烃类、酸类、酮类、杂氧化合物、碳氢化合物和其他。在不同温差处理下,检测出的挥发性香气种类及相对含量有所差异(表1图1)。其中,在昼夜温差为0 ℃时,醇类物质占比最高(39.56%),其次为醛类(35.41%);而在其他温差处理下,醛类物质占比最高,其次为醇类。香气成分方面,正己醛、青叶醛、青叶醇、2-乙基己醇的相对含量在各温差处理下较高,最高相对含量分别为39.94%(15 ℃)、47.21%(5 ℃)、7.26%(0 ℃)、25.60%(0 ℃)。
在昼夜温差为15 ℃时,显著增加了茶鲜叶的游离氨基酸和咖啡碱含量,并且显著降低了总黄酮含量与酚氨比(P<0.05)(表2)。在昼夜温差为0 ℃处理下,茶多酚、游离氨基酸和咖啡碱含量最低,分别为9.81%、1.39%、1.43%,水浸出物、总黄酮、可溶性糖含量与酚氨比最高。在昼夜温差为15 ℃时,茶多酚、游离氨基酸和咖啡碱含量最高,分别为11.00%、3.39%、3.65%。与昼夜温差为0 ℃处理相比,随着昼夜温差的增大,茶多酚、游离氨基酸、咖啡碱含量也随之增大,其中昼夜温差为15 ℃处理下的游离氨基酸含量与咖啡碱含量显著高于其他处理(P<0.05),而水浸出物、总黄酮、可溶性糖含量和酚氨比随昼夜温差的增大而降低,其中昼夜温差为15 ℃处理下的总黄酮含量与酚氨比显著低于其他处理(P<0.05)。
茶树生长离不开光合作用,光合作用是绿色植物的基本功能,而其中温度是影响光合速率的重要因素之一。由图2可知,昼夜温差变化影响茶树的光合参数。净光合速率(Pn)随昼夜温差的增大而增加,表现为昼夜温差15 ℃>10 ℃>5 ℃>0 ℃,且10 ℃与15 ℃昼夜温差处理下的Pn显著大于其他处理(P<0.05)。在不同昼夜温差处理下,昼夜温差增大使福云六号叶片气孔导度(Gs)增加,在昼夜温差为15 ℃时最高,达到显著水平(P<0.05)。胞间二氧化碳浓度(Ci)在昼夜温差为10 ℃时最低,其他处理间无显著差异(P>0.05)。蒸腾速率(Tr)则随昼夜温差的增大而增加,在昼夜温差为15 ℃时达到显著水平(P<0.05)。
表3可知,叶绿素a与叶绿素b随昼夜温差的增大而出现先增加后下降的趋势,在昼夜温差为10 ℃时叶绿素含量最高,与昼夜温差为0 ℃处理相比,10 ℃昼夜温差处理的叶绿素a与叶绿素b含量分别增加了57.0%和54.3%,且效果显著(P<0.05)。类胡萝卜素则在昼夜温差为10 ℃时含量最高,在昼夜温差为15 ℃时含量最低,与0 ℃昼夜温差处理相比昼夜温差为15 ℃时类胡萝卜素含量显著降低44.4%(P<0.05),其他处理均无显著差异(P>0.05)。
图3所示,温度显著影响β-葡萄糖苷酶、β-樱草糖苷酶和脂氧合酶活性(P<0.05)。β-葡萄糖苷酶和β-樱草糖苷酶活性随昼夜温差的增大呈现先增后降的趋势,并在昼夜温差为5 ℃时该酶活性最高。而脂氧合酶活性则随昼夜温差的增大呈现先降后增的趋势,与昼夜温差为0 ℃时相比,昼夜温差为5 ℃时,脂氧合酶活性下降35.08%,差异达显著水平(P<0.05)。
将不同温差处理下福云六号茶树光合参数与茶鲜叶非挥发性品质成分含量进行相关性分析。结果表明,茶树光合参数与茶鲜叶非挥发性品质成分含量具有一定的相关性。其中,Pn与游离氨基酸(FAA)含量呈极显著正相关,与咖啡碱(Caf)含量呈显著正相关,相关性系数(r)分别为0.848和0.679,与可溶性糖(SS)含量呈显著负相关,与酚氨比(P/A)呈极显著负相关(r=–0.724)。Gs与FAA含量呈显著正相关,Tr与FAA、Caf含量呈极显著正相关,与总黄酮(TF)含量呈显著负相关,与P/A呈极显著负相关(r=–0.718)。Chl(a/b)与FAA、Caf含量呈极显著负相关,r分别为–0.728和–0.816,而与TF含量呈显著正相关。Car与FAA含量呈显著负相关,与Caf含量呈极显著负相关(r=–0.724),而与可溶性糖(SS)呈显著正相关(图4)。
茶叶香气是由茶叶中的挥发性香气化合物所形成,是茶叶重要的感官品质之一,也是决定茶叶品质的重要因子之一。茶叶的香型与香气很大程度上取决于茶树体内合成的不同香气化合物及香气化合物含量。目前在茶叶中发现700多种芳香物质,将其分为醇类、醛类、酮类、酯类、碳氢化合物、杂环化合物等,其中醇类、醛类、酯类和碳水化合物含量相对较高[24-25],本研究也证实了这一观点。本研究发现茶鲜叶挥发性成分中的醇类和醛类相对含量最大,其中2-乙基己醇、青叶醛、青叶醇和正己醛的相对含量较高,2-乙基己醇、青叶醛和青叶醇呈现青草气,正己醛则具有果香。茶鲜叶的挥发性成分种类与主要香气成分的相对含量随温差变化有所差异,本研究中的青叶醇相对含量随昼夜温差的增大而降低,这与WANG等[26]的研究结果类似。香气化合物的合成与释放离不开酶活性,温度变化是影响酶活性的因素之一。β-葡萄糖苷酶、β-樱草糖苷酶和脂氧合酶是产生茶鲜叶挥发性香气成分的重要酶。在茶鲜叶中许多香气成分以糖苷形式存在,主要以葡萄糖苷和樱草糖苷为主,分别被β-葡萄糖苷酶和β-樱草糖苷酶水解释放,而脂氧合酶则是产生醇、醛、酯类等小分子物质的关键酶。在本研究中,β-葡萄糖苷酶和β-樱草糖苷酶活性随昼夜温差的增大呈先增后降的趋势,并在昼夜温度为20 ℃/15 ℃时酶活性最高,这与赵芹等[27]的研究结果类似。赵芹等[27]研究发现温度在15~20 ℃时茶叶酶活性最高,酶活性随温度的升高而降低。本研究发现温度变化显著影响β-葡萄糖苷酶、β-樱草糖苷酶和脂氧合酶活性,这可能是茶鲜叶挥发性香气成分的相对含量与气味存在差异的原因。茶鲜叶香气成分的相关性酶与其基因的表达在茶树次生代谢过程中发挥着重要作用,昼夜温差变化如何影响茶鲜叶挥发性品质相关代谢物有待深入研究。
温度变化对茶叶非挥发性品质成分产生了显著影响。茶多酚是茶叶品质成分中重要的化学成分,是决定茶汤浓度的重要物质,茶多酚含量适量增加有利于茶叶品质的提升。在本研究中,昼夜温差增大,茶多酚含量增加,这可能是温度升高促进茶树进行光合碳代谢,使多酚类物质合成积累较多。XIANG等[28]在儿茶素相关基因表达调控儿茶素生物合成对温度变化的响应研究中发现,温度升高使茶树新梢生长加快,总儿茶素、总酯化儿茶素等含量增加,与本研究结果类似。游离氨基酸含量不仅作为茶树氮库的数量表征还直接影响茶汤鲜爽度[29-30]。本研究发现,昼夜温差增大,游离氨基酸含量显著增加,并在昼夜温差为15 ℃时其含量达到最大,这可能是昼温升高加快了蛋白质和多肽在酶的催化下水解[31],使茶树叶片中的氨基酸含量增加,同时说明昼夜温差的增大能提高茶汤的鲜爽度。酚氨比则反映了茶叶的适制性,也是衡量茶汤鲜爽度的重要指标之一,酚氨比越小,滋味越醇,越鲜爽,茶叶品质越好[32]。本研究发现昼夜温差增大,酚氨比显著降低,并在昼夜温差为15 ℃时酚氨比达到最低,这说明昼夜温差增大有利于低品质茶叶原料改良,有利于提高茶叶品质。并且本研究相关性分析发现,游离氨基酸、咖啡碱、可溶性糖含量和酚氨比与茶树光合参数之间有一定相关性,说明昼夜温差的变化可能影响茶树的光合作用,进而对茶叶次生代谢产物产生一定影响,茶叶次生代谢产物对光合作用的响应机制是值得深入研究的科学问题。
温度是影响茶园生态的重要环境因子之一,温度变化对茶树光合作用有重要影响[33-34]。白天温度高可促进光合作用,而夜晚温度低可以减少呼吸作用的消耗[35]。本研究中,昼温升高,昼夜温差增大,净光合速率显著升高,这与袁小康[36]、BUNCE[37]、毛丽萍等[38]的研究结果一致,这可能是因为昼夜温差增大使光合作用关键酶rubisco的活性被激化,提高了叶肉细胞的光合活性,从而促进光合速率的增加。植物光合作用不仅受温度、光照等环境因子的影响,还与植物自身的生理生态息息相关,其中叶绿素含量对植物光合速率的影响更为直接。杨再强等[39]研究表明增加叶绿素含量可以有效提高植物对光能的捕获能力,促进光能传递,从而促进光合速率的提高。在本研究中,叶绿素含量随温差增大而升高,并且在昼夜温差为10 ℃(25 ℃/15 ℃)时叶绿素与类胡萝卜素含量达到最大,这与杨再强等[39]的研究结果一致,说明在昼夜温差为10 ℃(25 ℃/15 ℃)处理下有利于光合色素的合成,光合活性更好,促进了光合速率的提高。而在昼夜温差为0 ℃(15 ℃/15 ℃)与15 ℃(30 ℃/15 ℃)处理下的叶绿素与类胡萝卜素含量较低,这可能是茶树处于低温或高温的状态下使叶绿体结构遭到破坏,影响了光合色素的合成[40-41]。由此发现温度的高低均会对茶树叶绿素含量产生影响,进而影响茶树净光合速率。茶树光合作用是重要且复杂的化学过程,是茶树生产的物质基础,其中温度对茶树光合作用的影响需要进一步探究。
昼夜温差变化影响茶树光合作用,进而对茶鲜叶挥发性与非挥发性品质成分产生一定影响。昼夜温差增大,醛类物质相对含量提高,其中正己醛的相对含量随昼夜温差的增大而增大,并在昼夜温差为15 ℃时达到最大,但青叶醇的相对含量则随昼夜温差的增大而降低。昼夜温差的变化显著影响茶鲜叶挥发性香气化合物相关酶活性,其中β-葡萄糖苷酶与β-樱草糖苷酶活性在昼夜温差为5 ℃时最大,脂氧合酶活性则相反。就非挥发性品质而言,昼夜温差的增大显著增加游离氨基酸含量,显著降低酚氨比,使茶汤更加鲜爽。茶树净光合速率与游离氨基酸含量呈极显著正相关关系,而与酚氨比呈极显著负相关关系。开展昼夜温差等环境因子对茶鲜叶挥发性及非挥发性品质成分影响的研究,不仅能为高山出好茶提供理论依据,还可为茶园选择和茶树种植提供实际参考。
